Description
Die Erfindung betrifft neue synthetische Impfstoffe und
ein Verfahren zu ihrer Herstellung. Gemäß einer speziellen Ausführungsform betrifft die Erfindung einen neuen synthetischen
Influenza-Impfstoff und ein Verfahren zu seiner Herstellung. Die Erfindung betrifft ferner die Herstellung
relevanter antigener Fragmente des Virus, die Bindung dieser Fragmente an makromolekulare Träger, die erhaltenen
Kon jugate sowie deren Verwendung als Impfstoffe bei der
Bekämpfung von Virusinfektionen.
Die Erfindung wird nachstehend anhand der Herstellung eines
synthetischen Influenza-Impfstoffes erläutert. Die Erfindung ist nicht auf diese Ausführungsform beschränkt, denn das
erfindungsgemäße Prinzip der Herstellung synthetischer antigener Fragmente läßt sich auch auf andere Viren übertragen.
Die dabei erhaltenen Produkte können dann zur Herstellung von Impfstoffen gegen andere Virusinfektionen eingesetzt
werden.
Man kennt heute drei serologisch trennbare Influenza-Virus-
^ typen des Menschen, nämlich den Typ A, B und C. Für die schweren Pandemien, die in größeren Zeitabständen über die
ganze Erde hinwegziehen, sind bisher stets Stämme des Typs A verantwortlich gewesen. Dieser Typ zeigt das Phänomen des
Antigenwandeis. Dies hat zur Folge, daß die bei einer Infektion oder Impfung durch einen bestimmten Subtyp oder Stamm erzeugte
Immunität nur einen geringen Schutz gegen neue Antigenvarianten hervorruft.
Die gegenwärtig bekannten Influenza-Impfstoffe sind entweder attenuierte Lebendimpfstoffe oder Totimpfstoffe. Die
Lebendimpfstoffe haben eine bessere Wirkung als die Totimpf-
Stoffe, sie werden jedoch nicht als genügend unschädlich angesehen.
Die Schutzdauer der beiden Impfstoffe ist verhältnismäßig kurz.
Die beiden wesentlichen antigenen Bestandteile des Influenzavirus sind
. a) die Neuraminidase, die zahlreichen Influenza-Virusstämmen
gemeinsam ist. Antikörper gegen die Neuraminidase sind nahezu nicht virusneutralisierend und nicht schützend.
b) Das Hämagglutinin (HA), das allmählichen Änderungen
(drifts) unterliegt.. Es stellt ein starkes Itnmunogen dar und ist für die serologische Spezifität der verschiedenen
Virusstämme verantwortlich. Antikörper gegen das HA machen den immunen Wirt weniger empfänglich gegenüber einer
Infektion durch ein Virus, welches das gleiche Hämagglutinin enthält. Komplexe des Hämagglutinins und
der Neuraminidase wurden ebenfalls als potentielle Impfstoffe verwendet. Diese Untereinheits-Impfstoffe haben,
sich jedoch als nur sehr schwach immunogen erwiesen, bei Tieren und beim Menschen, die vom gleichen Antigen noch
nie antigen stimuliert wurden.
Das HA ist ein Molekül mit einem Molekulargewicht von 75 OOO bis 80 000 Dalton. Es wurde festgestellt, daß ein bei der
Spaltung mit Bromcyan erhaltenes Fragment verantwortlich ist für die immunologische Aktivität des intakten Proteins.
Es wurde ferner festgestellt, daß es die Bildung von Antikörpern stimuliert, welche seine Hämagglutinin-Aktivität
hemmen. Es ist ferner bekannt, daß ein nicht identifiziertes,
aus 16 Aminosäuren bestehendes tryptisches Peptid des
Hämagglutinins antigene Spezifität besitzt. Weder diese Fragmente noch irgend eine andere ähnliche natürliche oder
synthetische Substanz wurden bisher zur Erzeugung einer Schutzwirkung in vivo eingesetzt.
In anderen Systemen konnte nachgewiesen werden, daß kurze synthetische Peptide, die Fragmenten spezieller Proteine

Ι entsprechen, antigene Eigenschaften des intakten Proteins
tragen. So wurden synthetische Antigene zur Stimulierung der Bildung von Antikörpern gegen Lysozym oder das carcinoembryonale
Antigen (CEA) verwendet. Das gleiche Prinzip wurde zur Stimulierung einer antiviralen Reaktion eingesetzt.
Ein synthetisches, aus 20 Aminosäureresten bestehendes Peptid, das einem Fragment des Hüllproteins des MS-2 Coliphagen entspricht,
wurde durch Totalsynthese hergestellt und an einen synthetischen Träger gebunden. Dieses Konjugat mit komplettem
Freund'schem Adjuvans stimuliert in Kaninchen und Meerschweinchen
die Bildung von Antikörpern, die in der Lage sind, die Vermehrungsfähigkeit des Phagen zu neutralisieren.
Der Erfindung liegt die Aufgabe zugrunde, synthetische Impfstoffe
gegen Virusinfektionen, insbesondere einen synthetischen
Influenza-Impfstoff,sowie ein Verfahren zu ihrer Herstellung
zu schaffen. Dieses Verfahren ist dadurch gekennzeichnet, daß man das relevante antigene Fragment des Virus
herstellt, es an einen geeigneten makromolekularen Träger bindet und die so erhaltene Verbindung als Impfstoff ein- .
setzt. In einer bevorzugten Ausführungsform betrifft die Erfindung einen neuen synthetischen Influenza-Impfstoff und
ein Verfahren zu seiner Herstellung. Die erste Stufe dieses Verfahrens besteht in der Herstellung eines synthetischen
Peptids, das einem antigenen Fragment von Influenza-Hämagglutinin entspricht. Dieses Polypeptid wird an einen geeigneten
Träger gebunden. Das auf diese Weise erhaltene Produkt wird als Impfstoff verwendet. Die Impfung führt zur
Bildung von antiviralen Antikörpern (in vitro) und zum Schutz gegen Infektion (in vivo).
Aufgrund seiner Größe (etwa 550 Aminosäurereste) enthält das Hämagglutinin eine Vielfalt von antigenen Determinanten,
von denen mehrere in Bereichen liegen, die bei unterschiedlichen Stämmen Sequenzänderungen unterliegen . (Stamm-spezifische
Determinanten), und andere in Bereichen liegen, die verschiedenen bekannten Hämagglutininen gemeinsam sind (gemeinsame
Determinanten). Synthetische Peptide, die beiden Typen von Determinanten entsprechen, können zur Herstellung der
synthetischen Impfstoffe verwendet werden.
Es wurden folgende spezifische Determinanten verschiedener Influenza-Virusstamme hergestellt, nämlich

Alle diese Peptide und deren Kombinationen, gebunden an makromolekulare Träger, sind wirksame synthetische Impfstof-
fe. Spezielle Beispiele für geeignete synthetische Träger sind Multi-poly-DL-alanyl-poly-L-rlysin und Multi-poly-L-prolyl-poly-L-lysin
sowie natives Tetanustoxoid.

Sequenz des Hämagglutinins zahlreicher verschiedener Influenza-Virusstämme bilden, erzeugen eine gleichwertige
Immunität gegen sämtliche derartigen Stämme, während die spezifischen Determinanten eine Immunantwort hauptsächlich
gegenüber den Einzelstämmen induzieren mit geringer oder keiner Kreuzreaktivität und Immunisierung gegen andere
Stämme.
Es ist ersichtlich, daß sämtliche hier angegebenen Sequenzen
nur beispielhaft aufzufassen sind und daß andere Substanzen, die relevanten antigenen Determinanten verwandt sind, oder
Sequenzen, in die begrenzt konservative Aminosäure-Änderungen eingefügt wurden, ebenfalls verwendet werden können.
Das Peptid der Sequenz Ser-Lys-Ala-Phe-Ser-Asn-Ala-Tyr-Pro-Tyr-Asp-Val-Pro-Asp-Tyr-Ala-Ser-Leu
wird durch Peptidsynthese in fester Phase nach der Methode von Merrifield unter Verwendung der entsprechenden tert.-Butyloxycarbonyl-Derivate
der verschiedenen Aminosäuren hergestellt. Das Peptid wurde durch Säulenchromatographie gereinigt and auf seine
Reinheit durch Dünnschichtchromatographie und auf seine Zusammensetzung durch Aminosäuren-Analyse charakterisiert.
Das Peptid wurde mittels 1 -Äthyl-3- (3 '-dirnethylaminopropyl) carbodiimid-hydrochlorid
(EDCI) an drei verschiedene Träger gebunden:
a) Multi-poly-DL-alanyl-poly-L-lysin (A—L),
b) Multi-poly-L-propyl-poly-L-lysin (Pro-L) und
c) gereinigtes Tetanustoxoid

Die drei mit A—L erhaltenen Konjugate enthalten 3,7, 4 bis
5 und 7 bis 8 Mol Peptid gebunden an 1 Mol Träger (70 000 Dalton·). Das Konjugat mit Pro—L enthält 24 Mol
Peptid pro Mol Träger (110 000 Dalton). Die beiden Konjugate mit dem Tetanustoxoid enthalten 36,8 und 29,3 Mol
Peptid pro Mol Toxoid (etwa 150 000 Dalton). Die Konjugate mit dem Tetanustoxoid wurden zur Immunisierung von Kaninchen
verwendet. Den Kaninchen wird 1 mg Konjugat in 0,5 ml Phosphat-gepufferte Kochsalzlösung emulgiert mit 0,5 ml
komplettem Freund'schem Adjuvans subkutan injiziert.
1 Monat später folgen zwei Wiederholungs-Impfungen mit der halben verabreichten Menge im Abstand von 2 bis 3 Wochen.
Ferner werden Mäuse intrapertitoneal mit 50 pg Konjugat in
0,5 ml Phosphag-gepufferter Kochsalzlösung emulgiert mit 0,1 ml komplettem Freundschem Adjuvans geimpft. 1 Monat spä.^·
ter wird den Tieren eine Wiederholungsimpfung mit der halben
Menge gegeben. Den Tieren wird wöchentlich Blut entnommen, und zwar 1 Woche nach der letzten Wiederholungs-Impfung.
Weitere Auffrischungs-Impfungen werden erfordeflichenfalls
gegeben.
Die Reaktivität und Spezifität des Kaninchen-Antiserums
wird nach verschiedenen Methoden bestimmt:
T) Doppeldiffusion im Agargel (Figur 1).
Dies zeigt die Gegenwart von spezifischen Antikörpern gegen das Peptid sowie Antikörpern gegen das Tetanustoxoid
an.
2) Radioimmuntest in fester Phase in Mikrotiterplatten
(Figgr 2).
Dies zeigt Spezifität der Antikörper für das Peptid an. ·
3) Radioimmuntest.
Dieser Test zeigt Kreuzreaktivität der Antikörper mit den intakten Hämagglutininen an, welche die vorstehend
genannte Sequenz enthalten.

Die entstehenden Antikörper, die sowohl in Kaninchen als auch in Mäusen verschiedener Inzuchtstamme gebildet wurden, reagieren
mit dem intakten Hämagglutininmolekul. Dies zeigt sich an ihrer Fähigkeit zur Hemmung der Hämagglutination von
Hühnererythrozyten (Hämagglutinations-Hemmungs-Test (HI-Test) ) .
Ein Unterschied im HI-Titer von drei log2 wird zwischen dem
Immunserum und dem entsprechenden vor der Immunisierung ge-IQ
wonnenen Normalserums beobachtet (Figur 3). Dies zeigt, daß die Antikörper in der Lage sind, sich an das intakte
Hämagglutinin anzulagern und mit seiner biologische Funktion zu interferieren.
Die Kaninchen-Antikörper gegen das Peptid sind ebenfalls in der Lage, die Infektiosität des Virus zu hemmen. Dies wurde
in vitro mit Hilfe des Plaque-Testes in Zellkulturen unter Verwendung der Maden Darby Hundenierenzellinie (MDCK) festgestellt.
Aus der Tabelle I geht hervor, daß eine bis zu
2Q 83prozentige Hemmung der Virusplaques durch das Immunserum

Die Immunisierung von Mäusen des Inzuchtstammes C3H/Disn mit dem Peptid-Tetanustoxoid-Konjugat nach dem vorstehend beschriebenen
Impfschema ergab einen wirksamen Schutz gegen eine Infektion durch das Texas-Influenza-Virus. Dies zeigte sich
im geringeren Virusgehalt in den Lungen der immunisierten Mäuse; vgl. Tabelle II. Die Schutzwirkung wird durch den ausgeprägten
Unterschied in der Häufigkeit der Infektion bei den immunisierten Mäusen sowie durch die niedrigere Infektisität
für Bruteiar der Lungenextrakte von immunisierten Mäuse im Vergleich zu Kontrollgruppen, die entweder unbehandelt
oder vor der Infektion lediglich mit dem Tetanustoxoid immunisiert worden waren, ersichtlich

Der vorstehend erläuterte synthetische Impfstoff enthält ein Peptid, das im Hämagglutinin von mindestens neun verschiedenen
Influenza-Virusstämmen mit Subtyp A vorliegt. Es führt zu einem Schutz gegen alle diese Subtypen, der
gleich stark pro Subtyp ausgeprägt ist. ,

Der verwendete Träger, nämlich das Tetanustoxoid, wird zur
Impfung von Menschen gegen Wundstarrkrampf eingesetzt. Der synthetische Impfstoff der Erfindung ist somit nicht nur
unschädlich beim Einsatz in der Hummanmedizin, er kann
auch mit Vorteil zugleich als Auffrischungs-Impfung gegen Tetanus und zum Schutz gegen Influenza angewandt werden.

1. Synthetischer Impfstoff gegen Virusinfektionen,
gekennzeichnet durch einen Gehalt an einem synthetischen Peptid, das einem relevanten
antigenen Fragment des Virus entspricht, gebunden an einen geeigneten Träger.
2. Synthetischer Influenza-Impfstoff, gekennzeichnet durch
einen Gehalt an einem synthetischen Peptid, das einem relevanten antigenen Fragment des Influenzavirus entspricht,
gebunden an einen geeigneten Träger.
3. Impfstoff nach Anspruch 2, dadurch gekennzeichnet, daß das antigene Fragment einer Mehrzahl von Stämmen
des Influenzavirus gemeinsam ist.
4. Impfstoff nach Anspruch 1 oder 2, dadurch gekennzeichnet,
daß der Träger ein Makromolekül ist.
5. Impfstoff nach Anspruch 2, dadurch gekennzeichnet, daß das antigene Fragment das Peptid

7. Impfstoff nach Anspruch 1 oder 2, dadurch gekennzeichnet, daß das Peptid an den Träger über 1-Äthyl-3-(3'-dimethylaminopropyl)-carbodiimid-hydrochlorid
(EDCI) gebunden ist.
8. Verfahren zur Herstellung von synthetischen Impfstoffen gegen Virusinfektionen, dadurch gekennzeichnet, daß man
ein Peptid herstellt, das einem relevanten antigenen' Fragment des Virus entspricht, und das Peptid an einen geeigneten
Träger bindet.
9. Verfahren nach Anspruch 8, dadurch gekennzeichnet, daß das·
Virus ein Influenzavirus und das antigene Fragment ein Peptid gemäß Anspruch 5 ist.
10.Verfahren nach Anspruch 8, dadurch gekennzeichnet; daß der
Träger ein Makromolekül ist.
