enthalten.
Erfindungsgemäss gelangt man zu Cyclosporin G, Dihydrocyclosporin G und Isocyclosporin G, indem man

a) CyclOEporin G erhält, ir.dcrr. man einen Cyclosporin G
produzierenden Staria, beispielsweise von Tolypocladiurt,
z.B. von Tolypocladiuin inflatuir. Gc.r.is , in
Gegenwart eines Nahrmediurr.s züchtet und Cyelosporin
G isoliert, oder irden man
b) Dihydrocyclosporin G erhält, indem ir.an Cyclosporin
G hydriert, oder indem man
c) Isocyclosporin G erhält, indem man Cyclosporin G einer Umlagerung unterwirft.
Die Züchtung nach Verfahren a) lässt sich nach an sich bekannten Methoden für die Züchtung von analogen Stämmen,
z.B. wie in Beispiel 1 beschrieben, durchfuhren.
Ein. bevorzugter Cyclosporin G produzierender Stairm ist
der Stamm NRRL 8044 der Pilzspecies Tolypocladium inflatum Garns. Eine Kultur davon wurde beim United
States Department of Agriculture (Northern Research and Development Division), Peoria, 111., USA unter der
Kulturnumrr.er NRRL 804 4 deponiert und steht der Oeffentlichkeit
zur Verfügung. Eine andere Kultur davon wurde beim Fermentation Research Institute, Inage, Chiba City,
Japan, unter der Kulturnumir.er FRI FERi-I-P No. 2796
deponiert. Dieser Stairjn wurde vormals der Piizspecies
Trichoderma polysporum (Link ex. Pers.) zugeordnet und
ist z.B. in der englischen Patentschrift 1,491,509 beschrieben.
Für die Herstellung von Cycloi.porin G lassen sich
auch Stämme der Pilzspecies Tolypocladium inflatum
Gams verwenden, wie sie z.B. durch Selektion oder
Mutation des Pilzptar.K'.es NRRL 80 4 4 unter der Ein-Wirkung
von Ultravio? ett- oder Röntgenstrahlen ock;r
durch Anwendung anderer .Massnc.hmen, z.B. durch Eg-handlung
von Labcratoriuraskulturen reit geeigneten

Cyclosporin G kann auch an sich bekannte Weise isoliert vorden, z.B. wie in Beispiel 1 beschrieben.
Hierbei kann Cyclor;porin G abgetrennt werden von
gleichzeitig vorhandenen Naturprodukten, z.B. des weniger
polarere Cyclosporin D, das etwas polarere Cyclcsporin Λ
(auch bekannt als S 7481/F-l), das polarere Cyclcsporin
B (auch bekannt als S 7481/F-2) und das noch polarere Cyclosporin C.
Das erfindungsgernässe Verfahren b) lässt sich nach an
sich bekannten Methoden ausführen, z.B. durch katalytische Hydrierung.
Als Lösungsmittel kommen vorzugsweise Aethylacetat oder niedere aliphatische Alkohole, wie z.B. Methanol,
Aethanol, Isopropanol, in Frage. Die Hydrierung erfolgt zweckmässigerweise im neutralen Bereich bei
Temperaturen zwischen 20 und 30 ° und bei Atmosphärendx"uck
oder wenig erhöhtem Druck. Als Katalysator komrat Platin, vorzugsweise Palladium, z.B. Palladium auf
Kohle, in Frage.
Die Umlagerung nach dem Verfahren c) lässt sich nach
an sich bekannten Methoden ausführen, vorzugsweise durch Säurebehandlung, beispielsweise mit starken
organischen Säuren, z.B. mit Trifluoressigsäure, vorzugsweise Methansulfonsäure oder p-Toluolsulfcnsäure.
Die: Menge der starken Säure, die zur Isomerisierung
angewendet wird, liegt vorzugsweise zwischen 1 und A Mol pro Mol Cyclor.porin G.

Kothanoi, halocenierte Kohlenwasserstoffe, z.B.
Chloroform oder Aether, z.B. Dioxan, in Frage.
Die Reakt.ionstemperatur kann zwischen 20 und G5 ° C,
vorzugsweise von 45 bis 55 ° C variieren.
Cyclosporin G, Dihydrocyclospcrin G und Isocyclosporin
G zeichnen sich durch interessante chemotherapeutische und phariTiakologische Eigenschaften aus
und können daher als Heilmittel verwendet werden.
Insbesondere zeichnen sich Cyclosporin G, Dihydrocyclosporin G und Isocyclosporin G durch eine
immunosuppressive und entzündungshemmende Wirkung aus.
Als Heilmittel können Cyclosporin G, Dihydrocyclosporin G und Isocyclosporin G allein oder in geeigneter

Hilfsstoffen verabreicht werden.
In den nachfolgenden Beispielen erfolgen alle Temperaturangcxben in Celsiusgraden.

500 Liter einer Nährlösung, die pro Liter 40 g Glucose, 2,0 g Natriumcaseinat, 2,5 g Airjnoniumphosphat,
5 g i:gSO. .7H O, 2 g KH0PO , 3 g NaNO ,
0,5 g KCl, 0,0.1 g FeSO, und entraineralisiertes Kasnv.i:
enthält, werde; mit 50 Liter einer Vorkultur des Stammes NRRL 8C 44 angeimpft und in einem Stahlfermenter
unter Rühren (170 UPM) und Belüftung (1 Liter Luft//>
in. /Liter Nährlösung) 13 Tage bei 27 inkubiert (siele DOS 2,455,859).
Die Kulturbrühc wird mit der gleichen Menge n-Butylacetat
ausgerüT.rt, nach Abtrennung der organischen
Phase wird diese im Vakuum konzentriert und der Rohextrakt durch 2-stufige Verteilung zwischen Methanol-Wasser
(9 : 1) und Petroläther entfettet. Die methanolische Fiiace wird abgetrennt, im Vakuum
konzentriert und das Rohprodukt durch Zugabe von Wasser ausgefä]lt. E'as nach der Filtration gewonnene
Material wird ει der 5- bis 7-fachen Menge Sephaäcx
LH-20 mit Methanol als Elutionsmittel chromatographiert.
Die Spitzenfraktionen werden anscnliessend an Kiesolgel 60, Korngrösse 0,063 - 0,2 mm (Merck)
mit Ilexan-Aceton (2:1) chromatographiert, wobei die zuerst eluierten Fraktionen vorwiegend Cyclosporin A
und Cycloyporin D entlialten, die später eluierten
Anteile vorwiegend Cyclosporin C. Zur weiteren Reinigung werden die Cyclosporin A- und D-haltigen
Fraktionen aus dor 2- bis 2,5-fachen Menge Aceton bei - 15 ° kristallisiert und anschliessend durch zweimalige
Chromatographie an Kieselgel 60, Korngrosso
0,063 - 0,2 mm (Merck) 'weiter aufgetrennt, wobei die

»nit Hexan-Aceton (2 : 1) zuerst oluiorten Fraktionen
Cyclosporin D in star): angereicherter Form enthalten. Diese v/erden in der doppelten Menge Aceton gelöst
und bei ·- 15 ° kristallisieren lassen. Das dabei erhaltene Rohkristalliaat besteht aus sehr stark ai.■ bereichertem
Cyclosporin D, die Mutterlauge enthält neben Cyelosporin D noch weitere Komponenten/ so
Cyclosporin G. Zur Gewinnung von Cyclosporin G wird die zur Trockne verdampfte Mutterlauge an Kieselgel
60, Komgrösse 0,063 - 0,2 nun (Merck) mit wassergesättigtem
Essigester chromatographiert, wobei die zuerst eluierten Fraktionen Cyclosporin D enthalten,
und die später eluierten Fraktionen Cyclosporin G im
Gemisch neben weiteren Komponenten. Zur weiteren Anreicherung wird das Gemisch zuerst an Kieselgel 60,
Korngrösse 0,063 - 0,2 mm (Merck) mit Chloroform-Methanol
(98 : 2) und anschlicscend zur Auftrennung
mit Ilexan-Aceton (2 : 1) chro:.iatographiert; dabei
enthalten die späteren eluierten Fraktionen Cyclosporin G in sehr stark angereicherter Form. Zur Reingewinnung
v/erden die Cyclosporin G-Anteile zweimal aus Aether-Petroläther (1 : 1) bei Raumtemperatur
kristallisiert; sie liefern dünnschichtchromatographischeinheitliches,
reines (Reinheitsgrad >■ 95%) Cyclospcrin G als
2.'3 farblose Polyeder vom Smp. 193 - 194 ° (nach Trocknen
der Kristalle im Hochvakuum bei 80 ° während 2 Stunden).

700 mg Palladium-Kohle (10 % Palladium) werden in
20 ml Aethanol während 30 Minuten vorhydriert. Zu dieser Suspension des Palladiumkatalysators wird die
Lösung von 4,55 g Cyclosporin G in 60 ml Aethanol zugegeben
und darauf bei 21 ° und einem Druck von 742 ir.m
Quecksilbersäule bis zur beendeten Wasserstoffaufnahme
hydriert. Anschliessend filtriert man vom Katalysator ab und dampft das Filtrat im Vakuum bei 20 bis 40 °
zur Trockene ein. Der Rückstand besteht aus dünnschichtchromatographisch einheitlichem Dihydrocyclcsporin
G in Form eines weissen amorphen Pulvers vom Smp. 150 - 153 ° (nach Trocknen im Hochvakuum während
4 Stunden bei 80 °).

Beispiel 3: Isocyclosporin G
Zur Lösung von 6,0 8 g Cyclosporin G in 40 ml absolutem Dioxan gibt man die Lösung von 1,20 g Methansulfonsäure
in 20 ml Dioxan und hält das Gemisch bei 50 ° unter Feuchtigkeitsausschluss. Der Fortgang der Reaktion
wird im Dünnschichtchromatogramm verfolgt [Polygi-sm
SIL G-FolJen, Chloroform-Methanol-Eisessig (90:6:4),
Joddair.pf zur Sichtbarmachung] . Nach 16 Stunden wird auf Raumtemperatur gekühlt. Durch Zufügen von 1,13 σ
wasserfreiem Natriumacetat wird die Säure abgestumpft,
nach Rühren während 45 Minuten das ausgeschiedene Salz

abgenutscht und das Filtra.t im Vakuum boi 4 5 eingeöerapft.
Dir. 8,1 g Rückstand werden an 500 g Kiesel gel,
Merck, Korngrösse 0,06 3 - 0,2 run, chromatographicrt,
wobei zur Elution Chlorofom-Mcthnncl (98 : 2) dient.
Ii Die praktisch aus reinem Isocyclospcrin G bectehc.ndcn
Fraktionen werden vereinigt, in Vakuum bei 50 ° eingedampft
und der Rückstand aus Aether bei + 7 ° kristallisiert, wobei Isocyclosporin G vom Smp. 143 146
° anfällt.

5. Fermentationsbrühen, die bei der Züchtung eines Cyclosporin G produzierenden Stammes gewonnen
werden.
6. Fermentationsbrühen, die bei der Züchtung des Stammes NRRL 80 44 oder FRI FERM-P 2796 gewonnen
werden.
