Neue Chinazoline, Verfahren zu deren Herstellung und daraus hergestellte Arzneimittel
Die Erfindung betrifft Chinazolinverbindungen und deren pharmazeutisch verträgliche Säureadditionssalze,Verfahren
zur Herstellung dieser neuen Verbindungen und daraus hergestellte Arzneimittel. Die erfindungsgemäßen Verbindungen
sind geeignet zur Anwendung als antihypertensive Mittel. Die Klasse der erfindungsgemäßen Chinazolinverbindungen
ähnelt hinsichtlich der Struktur den 2,4,6,7-tetrasubstituierten antihypertensiven Chinazolinmitteln, die in der
US-PS 3 511 836 beschrieben sind. Die als Prazosin bekannte Verbindung n2-£4-(2-Furoyl)-1-piperazin-1-ylJ7~4-amino-6,7-dimethoxychinazolin" ist repräsentativ für die Chinazoline
der US-PS 3 511 836 und wurde klinisch erprobt. Wenngleich Prazosin offensichtlich ein wirksames antihypertensives

Mittel ist, so ist es nicht ganz frei von Alpha-Adrenorezeptoren blockierender Wirkung und von unerwünschten Nebenwirkungen wie posturalem Schwindel und orthostatischer

Seiten 117 - 127 (November 1975).
Im allgemeinen sind die erfindungsgemäßen Verbindungen als antihypertensive Mittel wesentlich wirksamer als Prazosin,
zeigen jedoch bemerkenswert geringere Alpha-Adrenorezeptoren blockierende Wirkung. Verglichen mit Prazosin
sind die erfindungsgemäßen Verbindungen auch wirksamere Phosphodiesterase-Inhibitoren und zeigen eine größere
Selektivitiät für das Enzym das auf das cyclische Guanosinmonophosphat-Substrat (cyclisches GMP) einwirkt.
Die erfindungsgemäßen Chinazoline weisen wertvolle pharmakologischeEigenschaften auf, die neuen Verbindungen wirken auch
antihypertensiv. Die Erfindung betrifft insbesondere 2-Piperazinyl-6,7-dimethoxychinazoline der Formel I:

"Cycloalkyl" steht hier für Cycloalkylreste mit 3 bis einschließlich 8 Kohlenstoffatomen im Ring, dazu gehören Gruppen,
wie Cyclopropyl, Cyclobutyl, Cyclopentyl, Cyclohexyl, Cycloheptyl und Cyclooctyl. "Methylcycloalkyl" steht hier für
die oben erwähnten Cycloalkylreste mit 3 bis einschließlich Kohlenstoffatomen im Ring mit einem Methylsubstituenten,
dazu gehören Gruppen wie 1-Methylcyclopropyl, 1-Methylcyclopentyl, 2-Methylcyclopentyl, 3-Methylcyclopentyl, 1-Methylcyclohexyl, 2-Methylcycloheptyl, 4-Methylcyclohexyl und
dergleichen. "Cycloalkenyl" steht hier für Reste mit 4 bis
einschließlich 8 Kohlenstoffatomen im Ring welche eine einzige Kohlenstoff-Kohlenstoff-Doppelbindung im Ring aufweisen.
Dazu gehören Gruppen wie 1-Cyclobutenyl, 1-Cyclopentenyl,
2-Cyclopentenyl, 3-Cyclopentenyl, 1-Cyclohexenyl, 2-Cyclohexenyl und dergleichen.
Die hier verwendete Bezeichnung "pharmazeutisch verträglich" bezieht sich auf Säureadditionssalze der Formel I welche
anionische Komponenten einer großen Anzahl relativ nicht toxischer anorganischer oder organischer Säuren aufweisen.
Das Anion trägt nicht wesentlich zur Toxizität des Salzes oder zur pharmakologisehen Wirksamkeit bei. Solche Salze
werden beispielsweise mit Essigsäure, Milchsäure, Bernsteinsäure, Maleinsäure, Weinsäure, Citronensäure, Gluconsäure,
Ascorbinsäure, Benzoesäure, Zimtsäure, Fumarsäure, Schwefelsäur·, Phosphorsäure, Chlorwasserstoffsäure, Bromwasserstoffsäure, Jodwasserstoffsäure, SuIfamidsäure, Sulfonsäuren,
wie Methansulfonsäure, Benzolsulfonsäuren p-Toluolsulfonsäure und verwandten Säuren gebildet.

Die Herstellung der Monosäureadditionssalze kann auf herkömmliche
Weise erfolgen, indem man eine Lösung oder Suspension der freien Base in einem reaktionsinerten
organischen Lösungsmittel mit einem chemischen Äquivalent i der Säure, oder, falls das Disäureadditionssalz gewünscht j
ist, mit mindestens zwei chemischen Säureäquivalenten behandelt! Bei der Isolierung der Salze verwendet man übliche Konzen- j
trations- oder Kristallisationstechniken.
Eine bevorzugte Gruppe von Chinazolinen sind die Verbindungen der Formel I, worin R auf Amino beschränkt ist und
R1 für Cycloalkyl steht.
Erfindungsgemäß werden Verbindungen der Formel I hergestellt, indem man ein Chinazolinderivat der Formel II

molare Mengen der Reaktionsteilnehmer der Formel II und

bei Verwendung von Ammoniak ein wesentlicher Überschuß bevorzugt. "Reaktionsinertes Lösungsmittel" steht für
organische Lösungsmittel, die selbst nicht in die Reaktion eingreifen, beispielsweise polare Lösungsmittel, wie
aliphatische Alkanole (z.B. Methanol, Xthanol, Isopropanol,
Isoamylalkohol), Tetrahydrofuran, Dioxan, Dimethylacetamid, Dimethylformamid und dergleichen. Wenn die Reaktion vollständig
ist, wird das Lösungsmittel eingedampft. Besteht der sich ergebende Rückstand aus einem Salz der Formel I,
so ergibt Rühren mit einer Base, wie Ammoniumhydroxyd, Natriumhydroxyd., Kaliumhydroxyd, Natriumcarbonat oder
j Kaliumcarbonat und anschließende Reinigung nach herkömmlichen j Kristallisationsverfahren das Produkt der Formel I in Form
der freien Base'.

Die Reaktion ist im allgemeinen nach 16 Stunden bei einer
Temperatur im Bereich von 150 bis 180 0C vollständig, kann
jedoch auch während einer Zeitdauer von 1 bis 48 Stunden bei Temperaturen von 100 bis 180 0C durchgeführt werden.

Gleichung 2 zeigt die Acylierung von 2-Plperazinyl-4-amino-6,7-dimethoxychinazolin der Formel V mit einem Carbonylhalogenid Reagens der Formel VI, worin R- die oben angegebenen Bedeutungen besitzt und "X" Halogen, (bevorzugt Chlor
oder Brom) bedeutet. Die Raktion schreitet bei Raumtemperatur in einem reaktionsinerten organischen Lösungsmittel leicht
voran und ist nach der Zugabe des Halogenid-Reaktionsteilnehmers im wesentlichen vollständig. Repräsentative Beispiele
für verwendete inerte organische Lösungsmittel sind Äther, Benzol, Toluol, Acetonitril, halogenierte Kohlenwasserstoffe,
wie Chloroform, Alkenole, wie Methanol oder Äthanol, Dimethylformamid, DimethylsuIfoxyd und dergleichen. Man verwendet
mindestens ein Moläquivalent des Carbonylhalogenid-Reaktionsteilnehmers (VI) und rührt die Reaktionsmischung 1 bis 6
Stunden nach der Zugabe. Die Produkte der Formel I werden aus der Reaktionsmischung abfiltriert oder durch übliche
Methoden, wie Eindampfen des Lösungsmittel· und Umkristallisatlpn
des Rückstands aus Alkanol-Lösungsmitteln, wie Methanol,
Äthanol, Isopropanol, Chloroform und dergleichen, isoliert.

Die Verbindungen der Formel I können auch gemäß dem in der DOS 2 457 911 beschriebenen Verfahren hergestellt werden,
was in Gleichung 5 dargestellt ist. Bei diesem Verfahren wird ein 2-Aminobenzonitril oder Benzaraidin der Formel IX
mit einem Piperazin-1-carboxamldo-Ester oder Nitril der
Formel X in einem reaktionsinerten Lösungsmittel umgesetzt. Die Reaktion erfolgt bei einer Temperatur von 50 bis 180 C
in Gegenwart von Natriumhydrid. In Formel IX steht "Y" für
den Rest -CN oder -C(=NH)NH2 und in Formel X steht 11Z" für
den Rest -CN, -Ci=NH)-OR3 oder -C(=NH)SR, worin R3 für einen
Alkylrest mit 1 bis einschließlich 4 Kohlenstoffatomen steht.
Die 2-Piperazinyl-6,7*-dimethoxychinazoline der Formel I
stellen wirksame antihyptertensive Mittel dar, die eine wesentlich geringere Alpha-adrenergische Blockierungswirkung
haben als Prazosin. Sie sind auch Phosphodiesterase-Inhibitoren mit Selektivität für das cyclische GMP Enzym.
Diese wünschenswerten Wirkungen können durch pharmakologische Standardtests in vitro und in vivo leicht aufgezeigt werden.
Beispielsweise bewirkt bei der spontan hypertensiven Ratte die orale Verabreichung von 0,4 mg/kg Körpergewicht
4-Amino-2-/i-(eyelopentylcarbonyl)-piperazinyl7~6,7-dimethoxychinazolin
eine Blutdrucksenkung um 50 mm Hg. Um eine ähnliche BlutdruckSenkung mit Prazosin zu erreichen, ist eine Dosis
von 1,8 mg/kg Körpergewicht erforderlich.

Die nachstehende Tabelle gibt die pharmakologische Aktivität

Die obige Tabelle zeigt, daß 4-Amino-2-^~4-(cyclopentylcarbonyl) ··
l-piperaziny^7~6,7-dilnethoxychinazolin ein Verhältnis von
antihypertensiver zu in vivo alphablockierender Wirkung im Vergleich zu Prazosin von 9,0 hat. Was die antihypertensive
Eignung anbetrifft, so ist dies ein sehr günstiges Verhältnis, da es eine Reduktion des Blutdrucks ohne bedeutenswerte
Beeinflussung des alpha-adrenergischen Systems anzeigt.
Eine der unerwünschtesten Nebenwirkungen, die bei der antihypertensiven Therapie auftritt, ist posturale Hypotension
aufgrund von hämodynamisehen Veränderungen die durch Veränderung von supiner in aufrechte Lage eintritt. Posturale
Hypotension wird klinisch meist durch Schwindel oder Ohnmacht
; manifest.
ι Ein geeigneter Tierversuch der auf posturale Hypotension beim
Menschen schließen läßt, ist der "dog-tilt-test" (Kipptest mit Hunden) vgl. J.W. Constantine et al., Am. J. Physiol. 221,
Nr. 6, 1681-1685 (1971). Bei diesem Versuch werden anästhetisierte Hunde, deren Blutdruck überwacht wird, auf einem
speziell konstruierten Tisch befestigt, mit dem sie rasch in eine Stellung von 80 . bis 90 von der horizontalen Lage
in eine Lage mit dem Kopf nach oben während 1 bis 2 Minuten gekippt werden. Jedes Tier wird zum Drehpunkt des Tisches
hin so gelagert, daß das Herz sowohl in supiner als auch in gekippten Lagen in etwa in der gleichen horizontalen Ebene
liegt. Vor der Verabreichung des Mittels (Kontrolle) verursacht das Kippen einen anfänglichen Blutdruckabfall, wonach
ein ausgleichendes Ansteigen des Drucks auf oder geringfügig über die Höhe vor dem Umkippen erfolgt. 10 Minuten nach dem
Kontroll-Kippen wird das Mittel während 3 Minuten intravenös verabreicht und dann wird erneut gekippt. Eine Beinträchtigung
der Reflex-Kompensation zur aufrechten Lage wird als drogenindiziert betrachtet und deutet auf eine mögliche posturale
Hypotension beim Menschen hin.

Die aus dem vorstehend beschriebenen Kipptest mit Hunden erhaltenen Daten zeigen, daß verglichen mit Prazosin
4-Amino-2-/4-(cyclopentylcarbonyl)-1-piperazinyl/-6,7-dimethoxychinazolin nur das 0,36 fache der Wirkung für posturale Hypotension bei gleichen hypertensiven Dosen hat.

Erfindungsgemäß werden bei der Behandlung von Hypertension
von Lebewesen, die unter Hypertension leiden oder dafür
prädestiniert s"in<3 antihypertensir wirkende Mengen an
2-Piperazinyl-6,7-dimethoxychinazolin der Formel I oder
pharmazeutisch verträgliche Salze verabreicht. Unter
systemischer Verabreichung versteht man sowohl orale als auch parenterale Verabreichungsarten. Parenterale Verabreichungsarten sind beispielsweise intramuskuläre, intraperitoneale, rectale und subcutane Verabreichung. Die
Dosierung variiert abhängig von der Verabreichungsform
und der speziellen verabreichten Verbindung. Im allgemeinen genügt die Verabreichung von etwa 0,001 bis 50 mg pro kg Körpergewicht des Lebewesens eines 2-Piperazinyl-6,7-dimethoxychinazolins der Formel I in wirksamen Einzel- oder Mehrfachdosen. Entsprechend der herkömmlichen klinischen Praxis wird ein antihypertensives Mittel der Formel I in einer wesentlich geringeren Dosis als der Dosis von der
man erwartet daß sie wirksam ist, verabreicht. Danach wird die Dosis um geringe Mengen erhöht, bis unter den gegebenen Umständen die optimale antihypertensive Wirkung erreicht ist. Bei wirksamer antihypertensiver Dosierung sind die erfindungsgemäßen Verbindungen im wesentlichen frei von posturaler Hypotension und den schädlichen oder lästigen Nebenwirkungen, wie sie im allgemeinen mit alpha-adrenergischer Blockierung verbunden sind.

Die aktiven Bestandteile der Formel I und deren pharmazeutisch verträgliche Säureadditionssalze können oral in Form von
Tabletten, dispergierbaren Pulvern, Granulaten, Kapseln, als Syrup oder Elixiere, und parenteral als Lösungen, Suspen-

sionen, Dispersionen, Emulsionen und dergleichen verabreicht I werden. Die oral anzuwendenden Mittel können ein oder j
mehrere herkömmliche Adjuvantien, wie Süßstoffe, Geschmacks- [
stoffe, Färbemittel und Konservierungsmittel enthalten, I
damit man ein pharmazeutisch ansprechendes Mittel erhält. '<
Tabletten können den aktiven Bestandteil vermischt mit her- | kömmlichen pharmazeutisch verträglichen Trägern, einschließ- ι
lieh inerter Verdünnungsmittel, wie Calciumcarbonat, Natriumcarbonat,
Lactose und Talkum, Granulierungs- und Disintegrierungsmitteln, wie Stärke und Alginsäure; Bindemitteln,
wie Stärke, Gelatine und Akaziengummi, und Schmiermitteln, I wie Magnesiumstearat, Stearinsäure und Talkum enthalten. ' Die Tabletten können nicht ummantelt oder nach herkömmlichen ι Methoden ummantelt sein, um die Zersetzung und Absorption im : Gastrointestinaltrakt zu verzögern und damit eine anhaltende i Wirkung über eine längere Zeitdauer zu bewirken. Entsprechend
können Suspensionen, Sirup und Elixiere den aktiven Bestandteil in Mischung mit herkömmlichen, für die Herstellung
solcher Mittel verwendeter Träger, wie Suspendiermitteln,
(beipsielsweise Methylcellulose, Tragacanth und Natrium- | alginat), Befeuchtungsmitteln (beispielsweise Lecithin,
Polyoxyäthylenstearat) und Konservierungsmitteln, wie
fithyl-p-hydroxybenzoat, enthalten. Kapseln können den aktiven ' Bestandteil allein oder in Mischung mit einem inerten, festen
Verdünnungsmittel, wie Calciumcarbonat, Calciumphosphat oder
Kaolin enthalten. Die Injektionslösungen werden wie aus dem
Stand der Technik bekannt hergestellt und können geeignete
Dispergier- oder Befeuchtungsmittel und Suspensionsmittel i enthalten, die den oben beschriebenen gleich oder identisch j sind. j

(a) Eine Mischung von 3,6 g (0,02 Mol) N-(Cyclopentylcarbonyl) -piperaz in und 4,79 g (0,02 Mol) 2-Chlor-4-amino-6,7-dimethoxychinazolin
in 50 ml absolutem Äthanol erhitzt man in einem geschlossenen Reaktor 16 Stunden bei
170 0C. Man kühlt die Reaktionsmischung, filtriert ab und verreibt die unlöslichen Bestandteile mit 100 ml
konzentriertem Ammoniumhydroxyd, wobei sich die freie Base ergibt. Das unlösliche Produkt wird gesammelt und
aus Methanol umkristallisiert, wobei man analytisch reines 4-Amino-2-/3-(cyclopentylcarbonyl)-l-piperazinyl7~6,7-dimethoxychinazolin
erhält, Fp. 256,0 bis 259,0 0C (Korr.).

Beispiel 1 (a) beschreibt das Verfahren gemäß der oben erwähnten Gleichung 1, während die folgenden Beispiele 1 (b - e)
die Verfahren gemäß den Gleichungen 2 bis 5 zur Herstellung von 4-Amino-2-/4-(cyclopentylcarbonyl)-l-piperazinyl7-6,7-dimethoxychinazolin
erläutern.
(b) Man gibt 0,1 Mol Cyclopentylcarbonyfchlorid zu 0,1 Mol 4-Amino-2-(1-piperazinyl)-6,7-dimethoxychinazolin
in 300ml Methanol, wobei bei Raumtemperatur kräftig gerührt wird.
Man rührt 2 bis 6 Stunden weiter und isoliert dann das Produkt gemäß dem Verfahren von Beispiel 1 (a), wobei
man 4-Amino-2-/4-(cyclopentylcarbonyl)-1-piperazinyl7-

wünscht, läßt man die basische Behandlung weg und nach Eindampfen des Lösungsmittels erhält man dann
4-Amino-2-/4-(cyclopentylcarbonyl)-piperazinyl7~6,7-dimethoxychinazolin-Hydrochlorid,
Fp. 279 - 280 °C (Zers.) (Korr.) kristallisiert aus Methanol-Isopropanol.

(c) Eine Mischung von wasserfreiem Ammoniak und 4-Chlor-2-/ji-(cyclopentylcarbonyl)
-1 -piperazinyl7~6,7-dimethoxychinazolin in 100 ml Tetrahydrofuran erhitzt man 16 bis
24 Stunden auf 100 0C und isoliert das Produkt gemäß
dem Verfahren von Beispiel 1 (a), wobei man 4-Amino-2-/4-(cyclopentylcarbonyl)-l-piperazinyl7-6,7-dimethoxychinazolin
erhält.
(d) Eine Mischung von 0,1 Mol 4-Amino-2-methylmercapto-6,7-dimethoxychinazolin
und von 0,1 bis 0,15 Mol N-(Cyclopentylcarbonyl) -piperazin in 150 ml Isoamylalkohol erhitzt
man 8 bis 24 Stunden zum Rückfluß. Das Lösungsmittel wird eingedampft und das zurückbleibende Material
wird gemäß dem Verfahren von Beispiel 1(a) behandelt, I
wobei man 4-Amino-2-/4-(cyclopentylcarbonyl)-l-piperazinyl/f
6,7-dimethoxychinazolin erhält.
(e) 0,01 Mol Äthyl-4-(cyclopentylcarbonyl)-piperazin-1-ylformamidat-Hydrochlorid
gibt man zu einer Lösung von 0,01 Mol 4,5-Dimethoxy-2-amlnobenzonitril in 30 ml

Ν,Ν-Dimethylformamid. Man gibt Natriumhydrid (0,02 Mol
einer 56 %-igen Suspension in Mineralöl) zu und rührt die Mischung 0,5 Stunden bei Raumtemperatur, und dann
12 Stunden lang bei 1OO °C. Wenn die Reaktionszeit vollständig ist, gibt man Wasser zu, wodurch man 4-Amino-2-/4-cyclopentylcarbonyl)-1-piperazinyl7~6,7-dimethoxychinazolin
erhält.

3,03 g (0,02 Mol) N-(Cyclopropylcarbonyl)-piperazin und 4,79 g (0,02 Mol) 2-Chlor-4-amino-6,7-dimethoxychinazolin
werden gemäß dem Verfahren von Beispiel 1(a) umgesetzt. Das rohe Produkt ergibt nach Kristallisation aus Äthanol
analytisch reines 4-Amino-2-/4-(cyclopropylcarbonyl)-1 piperazinyl7-6,7-dimethoxychinazolin,
Fp. 283,5 - 285,5 0C (korr.).

Die folgenden Bestandteile werden in den angegebenen Gewichts teilen nach üblichen pharmazeutischen Verfahren vermischt,
um eine Tablettengrundlage zu erhalten.

Diese Tablettengrundlage wird mit ausreichend 4-Amino-2-/4-(cyclopentylcarbony1)

oder einem pharmazeutisch verträglichen Salz davon gemischt, damit sich Tabletten ergeben, die 0,1, 0,25, O,5, 1, 2,5, 5,
7,5, 10, 25 und 50 mg aktiven Bestandteil enthalten, und die dann in einer herkömmlichen Tablettenpresse zu Tabletten der
gewünschten Größe geformt werden.

Claims (11)
1. Verbindungen der Formel I
oder pharmazeutisch verträgliche Säureadditionssalze davon, worin
R Amino oder Hydrazino bedeutet und
R1 für Cycloalkyl oder Methylcycloalkyl mit 3 bis einschließlich
8 Kohlenstoffatomen im Ring und Cycloalkenyl mit 4 bis einschließlich 8 Kohlenstoffatomen
im Ring steht.
2. 4-Amino-2-/4-(cyclopentylcarbonyl)-1-piperazinyl7~6,7-dimethoxychinazolin
oder pharmazeutisch verträgliche Säureadditionssalze davon.

9. Pharmazeutisches Mittel in Form von Dosiseinheiten zur systemischen Verabreichung einer Dosis von 0,001 bis
50 mg/kg Körpergewicht, gekennzeichnet durch mindestens eine Verbindung nach Anspruch 1 in üblichen pharmazeutischen
Trägern und/oder Formulierungsmitteln.
10. Mittel gemäß Anspruch 9, enthaltend 4-Amino-2-[4-(Cyclopentylcarbonyl)-1-piperazinyl]-6,7-dimethoxychinazolin
oder ein pharmazeutisch verträgliches Säureadditionssalz davon.
