Die erfindungsgemäss erhältlichen Verbindungen sind Phosphodiesterase-Inhibitoren und Herz- stimulantien, von denen eine bevorzugte Klasse die Myokard-Kontraktionskraft ohne wesentliche i Erhöhung der Herzgeschwindigkeit selektiv steigert. Die Verbindungen sind für die heilende und prophylaktische Behandlung von Herzzuständen, wie z. B. Die erfindungsgemäss erhältlichen Verbindungen mit einem oder mehreren asymmetrischen Zen- tren kommen als ein oder mehrere Paare von Enantiomeren vor, und solche Paare oder einzelne Isomeren können durch physikalische Methoden trennbar sein, z. B. durch fraktioniertes Kristallisie- ren der freien Basen oder geeigneter Salze oder durch Chromatographie der freien Basen. Erfin- dungsgemäss können die getrennten Paare sowie deren Gemische, wie racemische Gemische oder ge- trennte D- und L-optisch aktive isomere Formen erhalten werden. Die pharmazeutisch annehmbaren Säureadditionssalze der erfindungsgemäss erhältlichen Verbin- dungen sind solche, die sich aus Säuren mit pharmazeutisch annehmbaren Anionen bilden, wie das Hydrochlorid, Hydrobromid, Hydrojodid, Sulfat oder Bisulfat, Phosphat oder saures Phosphat, Acetat, Maleat, Fumarat, Lactat, Tartrat, Citrat, Gluconat, Saccharat und p-Toluolsulfonat. Im folgenden soll der Begriff "pharmazeutisch annehmbare Biovorstufe" erläutert werden. Es ist natürlich in der pharmazeutischen Chemie übliche Praxis, unerwünschte physikalische oder chemische Eigenschaften eines Arzneimittels durch dessen Überführung in ein chemisches Derivat zu überwinden, das diese unerwünschte Eigenschaft nicht aufweist, das aber nach Verabreichung an ein Tier oder einen Menschen in das Stamm- oder Ausgangs-Arzneimittel rückumgewandelt wird. Wird z. B. das Arzneimittel bzw. der Wirkstoff nach Verabreichung an ein Tier oder einen Patienten auf oralem Wege nicht gut absorbiert, kann die Möglichkeit bestehen, den Wirkstoff in ein chemi- sches Derivat zu überführen, das gut absorbiert wird und im Serum oder in den Geweben in das Stamm-Arzneimittel rück'gewandelt wird. Ist der Wirkstoff wieder in Lösung instabil, kann es möglich sein, ein chemisches Derivat des Wirkstoffes herzustellen, das stabil ist und in Lösung verabreicht werden kann, aber im Körper zum Stammwirkstoff rückumgewandelt wird. Der pharmazeutische Chemiker ist sich der Möglichkeit der Überwindung grundlegender Mängel eines Arzneimittels durch chemische Veränderungen, die nur vorübergehend sind und nach Verabreichung an ein Tier oder einen Patienten reversibel sind, wohl bewusst. Im vorliegenden Zusammenhang bedeutet der Ausdruck "pharmazeutisch annehmbare Biovorstufe einer Verbindung der Formel (I) " eine Verbindung mit einer von den Verbindungen der Formel (I) verschiedenen Strukturformel, die aber dennoch nach Verabreichung an ein Tier oder einen Menschen im Körper des Patienten in eine Verbindung der Formel (I) umgewandelt wird. Die herzstimulierende Wirksamkeit der erfindungsgemäss erhältlichen Verbindungen zeigt sich durch ihre Wirksamkeit in einem oder mehreren der folgenden Tests : (a) Steigerung der Kontraktionskraft im isolierten, spontan schlagenden Meerschweinchen-Doppelvorhof-Präparat ; (b) Steigerung der Herzmuskel-Kontraktilität (linksventrikulär dP/dt max.) am betäubten Hund mit einem in die linke Kammer implantierten Katheter ; (c) Steigerung der Herzmuskel-Kontraktilität des Hundes bei Bewusstsein mit einem in die linke Kammer implantierten Messwertgeber. Beim Test (a) werden die positiven inotropen und chronotropen Reaktionen der Herzvorhöfe auf die Testverbindung bei verschiedenen Dosen gemessen und mit den durch Isoprenalin hervorgerufenen Reaktionen verglichen. Der Vergleich der erhaltenen Dosis-Ansprechkurven liefert ein Mass <Desc/Clms Page number 3> für die Stärke gegen die Geschwindigkeitsselektivität der Testverbindung. Beim Test (b) wird die positive inotrope Wirkung der Testverbindung nach intravenöser Verabreichung beim anästhesierten Hund gemessen. Die Stärke des inotropen Mittels, die Selektivität zur Steigerung der Stärke gegen die Kontraktionsfrequenz und die Wirkungsdauer des positiven inotropen Effekts der Testverbindung werden erhalten, wie auch die peripheren Effekte, z. B. der Effekt des Blutdrucks. Beim Test (c) wird die positive inotrope Wirkung der Testverbindung nach intravenöser oder oraler Verabreichung bei einem Hund bei Bewusstsein mit einem in die linke Kammer implantierten Messwertgeber gemessen. Die Stärke des inotropen Mittels, die Selektivität der Steigerung der Stärke gegen die Kontraktionsfrequenz und die Wirkungsdauer des inotropen Effekts der Testverbindung werden wie die peripheren Effekte erhalten, z. B. der Einfluss auf den Blutdruck. Auf Grund der Leistungen der erfindungsgemäss erhältlichen Verbindungen bei den obigen Tests wurden folgende bevorzugte Gruppen gefunden : R'ist vorzugsweise Wasserstoff. (R) bedeutet vorzugsweise 6, 7-Di (C,-C,-alkoxy), insbesondere bevorzugt 6, 7-Dimethoxy. Y steht vorzugsweise in 4-Stellung der Piperidinogruppe. EMI3.1 EMI3.2 EMI3.3 EMI3.4 EMI3.5 am- N (CH3) S02. Die erfindungsgemäss erhältlichen Verbindungen können allein verabreicht werden, werden aber im allgemeinen im Gemisch mit einem pharmazeutischen Träger verabreicht, der im Hinblick auf die beabsichtigte Verabreichungsweise und die pharmazeutische Standard-Praxis gewählt wird. Beispielsweise können sie oral in Form von Tabletten mit Exzipienten, wie Stärke oder Lactose, oder in Kapseln, entweder allein oder im Gemisch mit Exzipienten, oder in Form von Elixieren oder Suspensionen mit Aroma- oder Farbstoffen verabreicht werden. Sie können parenteral, z. B. intravenös, intramuskulär oder subkutan injiziert werden. Für parenterale Verabreichung werden sie am besten in Form einer sterilen wässerigen Lösung verwendet, die andere gelöste Stoffe enthalten kann, z. B. genügend Salze oder Glucose, um die Lösung isoton zu machen. <Desc/Clms Page number 4> Für die Verabreichung beim Menschen zur heilenden oder prophylaktischen Behandlung von Herzzuständen, wie Herzversagen bei Blutandrang, werden die oralen Dosismengen der aktivsten neuen Verbindungen, die im Bereich von 20 mg bis 1 g täglich liegen, eingenommen in 2 bis 4 Teilmengen pro Tag, bezogen auf einen durchschnittlichen erwachsenen Patienten (70 kg). Dosierungen für intravenöse Verabreichung liegen im Bereich von 1 bis 300 mg pro Einzeldosis je nach Erfordernis, z. B. bei der Behandlung von akutem Herzversagen. So könnten für einen typischen erwachsenen Patienten Einzeltabletten oder-kapseln 5 bis 500 mg aktiver Verbindung in einem geeigneten pharmazeutisch annehmbaren Träger enthalten. Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man eine Verbindung der allgemeinen Formel EMI4.1 worin RI, RI, R'und n die obige Bedeutung haben, a) mit einem Isocyanat R 6 NeO, wenn R6 eine andere Bedeutung als Wasserstoff hat, oder für Verbindungen, in denen R 6 Wasserstoff ist, mit Natrium- oder Kaliumcyanat in Gegen- wart von Säure umsetzt oder b) einem Halogenformiat oder Acylhalogenid der Formel QCOR". worin Q2 Chlor oder Brom ist und R3 die obige Bedeutung hat, oder c) einem Halogensulfonat, Sulfonylhalogenid oder Sulfamylhalogenid der Formel QSO ; , R , worin Q2 Cl oder Br ist und RI die obige Bedeutung hat, oder d) einem Carbamylhalogenid der Formel R6 R'NCOQ 2, worin Q 2 Chlor oder Brom bedeutet und R 6 und R'die oben angegebene Bedeutung haben, jedoch eine andere Bedeutung als Wasserstoff haben ; oder e) einem Anhydrid oder Pyrocarbonat der Formel (RCOjO, worin R3 die obige Bedeutung hat, umsetzt, wobei beliebige Hydroxy- oder primäre oder sek. Aminogruppen in R 2 bis R', wenn gewünscht, vor der Reaktion geschützt werden und die Schutzgruppen nach der Reaktion entfernt werden ; worauf gegebenenfalls das so erhaltene Produkt der For- mel (I) durch Reaktion mit einer pharmazeutisch verwendbaren Säure in ein pharmazeu- tisch verwendbares Säureadditionssalz übergeführt wird. Bei einer typischen Arbeitsweise gemäss a) werden das Phthalazin und das Isocyanat zusammen, z. B. unter Rückfluss in einem geeigneten organischen Lösungsmittel, z. B. Chloroform, während 12 bis 24 h erwärmt. Das Produkt kann nach herkömmlichen Verfahren isoliert und gereinigt werden. Gruppen, die mit den Isocyanatgruppen bzw. dem Cyanat reagieren können, aber natürlich andere als die -NH-Gruppe von -NHR4 im Piperidinoring, sollten im allgemeinen durch herkömmliche Schutzgruppen vor der Umsetzung geschützt werden, wobei die Schutzgruppe nach der Umsetzung in bekannter Weise entfernt wird. Gruppen, die möglicherweise geschützt werden müssen, sind z. B. EMI4.2 sek. Aminogruppen,Typischerweise lässt man bei den Arbeitsweisen b), c), d) und e) die Reaktionskomponenten zusammen bei Raumtemperatur etwa 72 h in einem inerten organischen Lösungsmittel, z. B. Säureadditionssalze der Verbindungen der Formel (I) können aus dem Roh- oder Reinprodukt der freien Base in bekannter Weise durch Umsetzen der freien Base mit der Säure in einem inerten 'Lösungsmittel hergestellt werden, z. B. durch Mischen alkoholischer Lösungen und Gewinnen der erhaltenen Fällung durch Filtrieren. Das Produkt kann dann bis zur Reinheit umkristallisiert werden. Die Ausgangsmaterialien für das erfindungsgemässe Verfahren sind entweder bekannte Verbin- dungen oder können nach bekannten Verfahren hergestellt werden. Die Lösung wurde dann bei Raumtemperatur 18 h gerührt, im Vakuum eingeengt, das Konzentrat mit 50 ml Wasser geschüttelt und dann zweimal mit je 50 ml Chloroform extrahiert. Die gereinigten organischen Extrakte wurden mit 50 ml verdünntem Natriumhydroxyd geschüttelt, dann über Magnesiumsulfat getrocknet und im Vakuum zur Trockne eingeengt, wobei ein braunes Öl anfiel, das mit 70 ml 30 bis 400-Petroläther verrieben wurde, worauf der Äther dekantiert wurde. Zur Herstellung des Oxalats wurde der Rückstand in warmem Äthylacetat gelöst, filtriert und das Filtrat im Vakuum zur Trockne eingeengt und erneut in einer kleinen Menge Äthylacetat gelöst. Das Gemisch wurde dann im Vakuum eingeengt und der Rückstand in 25 ml Wasser suspendiert, worauf zweimal mit je 25 ml Chloroform extrahiert wurde. Die vereinigten Chloroformextrakte wurden mit 20 ml Wasser gewaschen und im Vakuum zu einem öligen Rohprodukt eingeengt, das sich beim Verreiben mit 20 ml Äther EMI7.6 7-Dimethoxy-1- [4- (2-hydroxypropyl) -Analyse : EMI7.7 <tb> <tb> Berechnet <SEP> für <SEP> Ct. <SEP> H <SEP> N, <SEP> 0, <SEP> : <SEP> C <SEP> 65, <SEP> 2 <SEP> ; <SEP> H <SEP> 7, <SEP> 6 <SEP> ; <SEP> N <SEP> 12, <SEP> 7% <tb> gefunden <SEP> : <SEP> C <SEP> 65,2; <SEP> H <SEP> 7,6; <SEP> N <SEP> 12,6%. <tb> <Desc/Clms Page number 8> Beispiel 8 : Herstellung von 6, 7-Dimethoxy-1- [4-Acetonylpiperidino] phthalazin-hydrochlorid EMI8.1 1, 7 g 6, 7-Dimethoxy-1- [4- (2-hydroxypropyl) piperidino ] phthalazin wurden zu einem gerührten Gemisch von 3, 5 g Chromtrioxyd (0, 297 mm), 90 ml trockenem Methylenchlorid und 5, 5 g trockenem S Pyridin gegeben. Suspendierte Chrom- salze wurden durch Hindurchführen der organischen Schicht durch eine kurze Siliziumdioxydsäule und dann durch Filtrieren durch"Hyflo", bedeckt mit einer Aktivkohleschicht (0, 5 cm), entfernt. Eindampfen der klaren Lösung im Vakuum ergab das Oxydationsprodukt als strohfarbenes Öl, das 'in der Mindestmenge Äthylacetat erneut gelöst und mit überschüssigem ätherischem Chlorwasserstoff behandelt, wurde, um das Hydrochlorid auszufällen. Umkristallisieren des Salzes aus Äthanol ergab 0, 5 g 6, 7-Dimethoxy-1- [4-acetonylpiperidino], phthalazin-hydrochlorid Fp. 213 bis 214 C. Claims (1)
- PATENTANSPRÜCHE : 1. Verfahren zur Herstellung von neuen Phthalazinen der allgemeinen Formel EMI8.3 worin (i) R'Wasserstoff oder nied. Alkyl ist, (ii) (R") l bis 3 Substituenten bedeutet, wobei jeder Rest R'Hydroxy oder nied. Alkoxy darstellt und n 1 bis 3 ist, oder zwei der Reste R'eine in Nachbarstellungen zum Benzolring befindliche Methylendioxy- oder Äthylendioxygruppe bedeuten, und (iii) Y in 3-oder 4-Stellung des Piperidinorings sitzt und entweder a) eine Gruppe -NR'-COR'ist, wobei R3 gegebenenfalls amino-, hydroxy-, nied. alkoxy-, aryl-oder heteroarylsubstituiertes nied. Alkyl, nied. Alkenyl- oder nied.Alkinylmethyl, gegebenenfalls amino-, aryl-, heteroaryl-, nied. alkoxy- oder hydroxysubstituiertes <Desc/Clms Page number 9> EMI9.1 EMI9.2 EMI9.3 EMI9.4 EMI9.5 zeutisch verwendbare Säureadditionssalze, dadurch gekennzeichnet, dass man eine Verbindung der allgemeinen Formel EMI9.6 worin R1, R2, R4 und n die obige Bedeutung haben, a) mit einem Isocyanat UNCO, wenn R6 eine andere Bedeutung als Wasserstoff hat, oder für Verbindungen, in denen RI Wasserstoff ist, mit Natrium-oder Kaliumcyanat in Gegenwart von Säure umsetzt oder b) einem Halogenformiat oder Acylhalogenid der Formel Q2COR3, worin Q2 Chlor oder Brom ist und R3 die obige Bedeutung hat, oder c) einem Halogensulfonat, Sulfonylhalogenid oder Sulfamylhalogenid der Formel QSOR, EMI9.7 als Wasserstoff haben ;oder e) einem Anhydrid oder Pyrocarbonat der Formel (R3CO2)O, worin R3 die obige Bedeutung hat, umsetzt, wobei beliebige Hydroxy- oder primäre oder sek. Aminogruppen in R2 bis R', wenn gewünscht, vor der Reaktion geschützt werden und die Schutzgruppen nach der Reaktion entfernt werden ; worauf gegebenenfalls das so erhaltene Produkt der Formel (I) durch Reaktion mit einer pharmazeutisch verwendbaren Säure in ein pharmazeutisch verwendbares Säureadditionssalz übergeführt wird.2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass Ausgangsverbindungen eingesetzt werden, worin R2 Wasserstoff, (R2)n 6,7-Dimethoxy bedeuten und die Gruppe -NHR'in 4-Stellung des Piperidinrings der Verbindung der Formel (III) steht.3.
