ist R = Äthyl oder Phenyl.
Die Erfindung betrifft auch pharmazeutische Präparate enthaltend obige Verbindungen oder Salze davon als Wirkstoff.
Der Ausdruck "Alkyl" betrifft Gruppen wie Methyl, Äthyl, Propyl, Butyl; "Isoalkyl" solche wie Isopropyl, Isobutyl;
"Cycloalkyl" solche wie Cyclopentyi, Cyclohexyl. Repräsentative
Aryl- oder Heteroaryl-Gruppen sind Phenyl oder Thienyl? substituiertes Aryl ist z.B. Chlorphenyl, Methoxyphenyl,
Trxfluormethylphenyl und Amino-phenyl oder N, N-substituiertes Aminophenyl, wie Dinieder-alkylarainophenyl,
worin "Nieder" insbesondere C, -C, bedeutet.

Die erfindungsgemäßen Salze sind insbesondere pharmazeutisch
und Veterinär geeignete, sowie solche, die zur Reinigung und Kristallisation der neuen Verbindungen nützlich sind.
Unter solchen Salzen sind zu nennen die Hydrochloride, Sulfate, Methan-sulfonate und Salze mit organischen Säuren.
N-Alkyl-Verbindungen sind z.B. N-Methyl-, N-Äthyl-,
N-Benzyl-Verbindungen, insbesondere in Form der Iodide, Bromide oder Chloride.
Ausgangsverbindungen, die zur Herstellung von Verbindungen (II) der Erfindungen des 4-'Alkyl-6-oxa-1-azatricyclo-

(4.2.2.0 ' )-dodecan-5-on-Typs geeignet sind, sind monosubstituierte Ester der Malonsäure, wie Diäthyl-methylmalonat, Diäthyl-äthylmalonat, Diäthyl-propylmalonat, Diäthyl-isopropylmalonat, Diäthyl-n-butylmalonat, Diäthylbenzylmalonat und dergl.. Ausgangsstoffe zur Herstellung von Verbindungen vom 4-Aryl-6-oxa-1-azatricyclo-(4.2.2.0 ' ) dodecan-5-on-Typ sind ortho-, meta- und para-substituierte Ester der Phenyl-Essigsäure, wie Äthyl-p-methylphenylacetat, Äthyl-m-methoxyphenylacetat oder substituierte Phenylacetonitrile, wie p-Chlorphenyl-acetonitril.
(4.2.2.0 ' )-dodecan-5-on-Typs geeignet sind, sind monosubstituierte Ester der Malonsäure, wie Diäthyl-methylmalonat, Diäthyl-äthylmalonat, Diäthyl-propylmalonat, Diäthyl-isopropylmalonat, Diäthyl-n-butylmalonat, Diäthylbenzylmalonat und dergl.. Ausgangsstoffe zur Herstellung von Verbindungen vom 4-Aryl-6-oxa-1-azatricyclo-(4.2.2.0 ' ) dodecan-5-on-Typ sind ortho-, meta- und para-substituierte Ester der Phenyl-Essigsäure, wie Äthyl-p-methylphenylacetat, Äthyl-m-methoxyphenylacetat oder substituierte Phenylacetonitrile, wie p-Chlorphenyl-acetonitril.
Verbindungen der Formel (III) werden in zwei Stufen hergestellt durch Umsetzen von 2-Methylen-quinuklidin-3-on
mit Methyl-benzylketon gefolgt von selektiven katalytischen Reduktionen.

Die erfindungsgemäßen Verbindungen sind wertvolle Wirkstoffe in pharmazeutischen Präparaten. Sie sind als Medikamente
wertvoll, in der Behandlung und Linderung der
Auswirkungen der Parkinson'sehen Krankheit. Sie sind wirksame allgemeine psycho-motorische Stimulantien und als
solche von besonderem Wert in der Geriatrie. Die Verbindungen der vorliegenden Erfindung können als Medikamente bei Antimotion-Krankheit verwendet werden, sie sind wertvoll in der Opthalmologie als Mydriatica. Sie können bei der
Behandlung des Rauchen-Abgewöhnens verwendet werden, da sie übertriebene Tabakeffekte induzieren. Die erfindungsgemäßen Verbindungen sind wirksame "Wachmacher" und können als
solche mit Barbituraten verwendet werden. Sie können verwendet werden, um der Müdigkeit durch Antihistaminica und ähnliche Drogen entgegenzuwirken.

Sie sind auch in der Behandlung von Hyperkinese bei Kindern, von Narkolepsie, von geistigen Depressionen organischen
Ursprungs und Fettleibigkeit nützlich.
Die Dosierung beträgt im allgemeinen etwa 10 mg bis etwa 100 mg für einen Erwachsenen pro Tag. Die erfindungsgemäßen
Medikamente können durch Injektion in einem geeigneten Verdünnungsmittel oder Träger verabfolgt werden, oder per os
in Form von Suppositorien und auf jede andere herkömmliche Weise, wie durch Infusion o. dgl.
Die erfindungsgemäßen Verbindungen sind von besonderem Wert
in Kombination mit Antihistminica, wenn tagsüber verwendet,
da die Mischung bei dem behandelten Patienten keine Müdigkeit verursacht.
Für eine opthalmologische Verwendung werden die erfindungsgemäßen Medikamente vorteilhaft in herkömmlichen Trägern
oder Puffern verwendet. Die Konzentrationen können gemäß dem gewünschten Effekt variiert werden und liegen im allgemeinen
bei etwa 2 bis 10% (Gewicht) Wirkstoff.
Die vorliegende Erfindung betrifft auch pharmazeutische und Veterinäre Präparate, enthaltend irgendeine der neuen
Verbindungen oder Kombinationen davon als Wirkstoff.
Die Erfindung betrifft auch Stereoisomere und optische Isomere der obigen Verbindungen. Diese Isomeren existieren
wegen der Asymmetrie an den C-Atomen in 2-, 4- und 7-Stellung obiger Verbindungen (II) bzw. an den Stellungen 4a, 8a und
7 in Verbindungen III.
Die folgenden Beispiele illustrieren die Erfindung, ohne sie einzuschränken.

Beispiel 1 :
Äthanolisches Natriumäthoxid, zuvor hergestellt aus 2.9 g
metallischem Natrium,und 100 ml Äthanol werden zu Diäthylmethyl-malonat,
50 g, gegeben. Die Mischung wird 30 Minuten unter Rückfluß erhitzt, dann auf 5° abgekühlt. Eine Lösung
von 2-Methyl-enequinuklidin-3-on, 39.4 g, in Äthanol, 50 ml, wird dann tropfenweise unter Rühren zugegeben, IJaeh 16 Stunden
bei Raumtemperatur wird die erhaltene Lösung mit Essigsäure neutralisiert und dann unter vermindertem Druck eingedampft.
100 ml Wasser werden zugegeben und die Mischung wird mit Chloroform extrahiert. Abdampfen dieses Lösungsmittels
ergibt Diäthyl-(3-oxo-quinuklidin-2-yl)-methylmethyliaalonat,
72 g (81%), F. 58.4° (aus Petroläther) .
Eine Lösung dieser Verbindung, 72 g, in Äthanol, 50 ml, wird bei 5° unter Rühren mit einer Lösung von Natriumborhydrid,
3.55 g, in Äthanol, 600 ml, versetzt. Die Borhydrid-Lösung wird in kleinen Anteilen innerhalb von
6 Stunden zugegeben. Nach 20 Stunden wird die Mischung mit konzentrierter Salzsäure neutralisiert und dann unter
vermindertem Druck eingedampft. Der Rückstand wird in 200 ml Wasser aufgenommen und mit Chloroform extrahiert. Abdampfen
dieses Lösungsmittels ergibt einen sirupösen Rückstand, 55f/ der das Diäthyl-(3-hydroxyquinuklidin-2-yl)-methylmethylmalonat
ist. Sein Methiodid-Salz, hergestellt mit Methyl-iodid in Aceton, schmilzt bei 228.2°.
Dieses Produkt, 54.8 g, wird mit 200 ml konzentrierter Salzsäure und 100 ml Wasser 20 Stunden unter Rückfluß erhitzt.
Teilweises Abdampfen des Lösungsmittels führt zur Kristallisation von 2-(trans-3-Hydroxyquinuklidin-2-yl)-methylpropansäure-Hydrochlorid,
8 g (18%), als Nebenprodukt, F. 268.8-269.4°. Weitere Konzentration der Mutterlauge und Kühlen
bewirkt die Kristallisation des gewünschten 4-Methyl-6-oxa-1-azatricyclo-(4.2.2.0
' )-dodecan-5-on-Hydrochlorids, 27.5 g, (66%), F. 267.2° unter Zersetzung.

Äthyl-phenylacetat, 49.2 g wird zu einer Lösung von Natriumäthoxid
gegeben, die zuvor aus metallischem Natrium, 2.9 g und Äthanol, 100 ml, hergestellt wurde. Die Mischung wird
auf 5° gekühlt. Dann wird eine Lösung von 2-Methylenquinuklidin-3-on,
39.4 g, in Äthanol, 50 ml, tropfenweise unter Rühren zugegeben. Nach 16 Stunden bei Raumtemperatur
wurde die Lösung mit Essigsäure neutralisiert und dann unter vermindertem Druck eingedampft. 100 ml Wasser wurden
zu dem Rückstand gegeben. Die Mischung wurde mit Chloroform extrahiert. Abdampfen dieses Lösungsmittels ergab Äthyl-(3-oxoquinuklidin-2-yl)-methyl-phenylacetat,
54 g (60%), Sdp. 195-200° bei 1 min Hg, F. des Methiodid-Salzes
194.7-195.6°
22.4 g dieser Verbindung wurden in Äthanol, 100 ml, gelöst. Die Lösung wurde bei 5° mit einer Lösung von Natriumborhydrid,
1.2 g in Äthanol, 200 ml, behandelt. Die Borhydrid-Lösung wurde in kleinen Anteilen während 6 Stunden zugegeben.
Nach 20 Stunden wurde die Mischung mit konzentrierter Salzsäure neutralisiert, dann bei vermindertem Druck eingedampft.
Der Rückstand wurde in 200 ml Wasser aufgenommen und mit Chloroform extrahiert. Abdampfen des Lösungsmittels
ergab Äthyl-(3-hydroxyquinuklidin-2-yl)-methyl-phenylacetat,
19.5 g, (80%), F. 147.7-148.2° nach ümkristallisation aus Aceton.
Die obige Verbindung, 16.9 g, wird in einer Mischung mit
konzentrierter Salzsäure, 100 ml, und Wasser, 40 ml, 20 Stunden unter Rückfluß erhitzt. Abdampfen des Lösungsmittels
unter vermindertem Druck ergibt einen glasigen Rückstand, der wieder in 50 ml Wasser gelöst wird. Es wird mit Natriumbicarbonat
neutralisiert und mit Chloroform extrahiert. Abdampfen des Lösungsmittels ergibt einen Rückstand, der
mit Petroläther trituriert wird und kristallines 4-Phenyl-

1 ^-Äthano^-oxo^-phenyl-perhydroquinf. lin-Hydrochlorid.
Das 1,4'-£thano-6-oxo-7-phenyl-1,2, S^^ö^^^a-octahydroquinolin
wurde durch eine Modifikation des Verfahrens von Oppenheimer und Bergmann, Synthesis 269 (1972) wie
folgt hergestellt:
Zu einer Lösung hergestellt aus 3 g Natriummetall in 100 ml absolutem Methanol wurden 45 g Methyl-benzyl-keton gegeben.
Die erhaltene Lösung wurde 30 Minuten unter Rückfluß erhitzt, dann auf 0°C abgekühlt und 39 g 2-Methylen-3-quinuclidinon
zugegeben. Die Lösung wurde gerührt, 48 Stunden bei Raumtemperatur, dann durch Zugabe von 3 ml Essigsäure neutralisiert.
Die Lösungsmittel wurden unter vermindertem Druck

abgedampft. Es wurde ein rohes öl erhalten, das mit Chloroform
extrahiert wurde. Nach Abdampfen des Lösungsmittels wurden 52 g (Ausbeute 60 %) des gewünschten Produkts erhalten,
F., aus Aceton, 125.7°C. Behandeln mit Salzsäure ergab das entsprechende Hydrochlorid-Salz, F. über 3000C
(Zers.) .
Die Doppelbindung in obiger Verbind;.ng wurde selektiv reduziert
durch Lösen von 15 g der freien Base in 150 ml Äthanol
und Reduzieren mit Wasserstoff bei 3 Atmosphären in Gegenwart von 0.5 g 10% Palladium auf Kohle in einem Parr-Gerät.
Nach 48 Stunden wurde die Lösung filtriert und eingedampft. Der Rückstand wurde aus Aceton umkristallisiert, F. 122.6°C.
Behandeln mit gasförmigem Chlorwasserstoff in Aceton ergab das Hydrochlorid von 1,4-Äthano-6-oxo-7-phenylperhydroquinolin
in fast quantitativer Ausbeute, F. über 3000C (Zers.).
1,4-Äthano-6-oxo-7-äthyl-1,2,3,4,6,7,3,8a-octahydroquinolin-Hydrochlorid.
1,4-Äthano-6-oxo-7-Äthylperhydroquinolin-Hydrochlorid wurde
hergestellt durch katalytische Hydrierung der Doppelbindung in 1,4-Äthano-6-oxo-7-äthyl-1,2,3,4,6,7,8,8a-octahydroquinolin
durch eine Modifikation der Methode von Oppenheimer et al (siehe Beispiel 3). Zu einer Lösung von 1 g Natriummetall in
100 ml Äthanol gab man 6.3 g Äthylaceto-acetat. Die erhaltene Lösung wurde 30 Minuten unter Rückfluß erhitzt, dann auf 00C
gekühlt. 5·5 g 2-Methylen-3-quinuclidinon wurden zugegeben.
Die Lösung wurde über Nacht bei Raumtemperatur gerührt, mit Essigsäure neutralisiert und die Lösungsmittel abgedampft.
Der Rückstand wurde mit Toluol extrahiert, das dann verworfen wurde. Der Extrakt wurde mit 17 ml konzentrierter Salzsäure
und 5 ml Wasser 7 Stunden unter Rückfluß erhitzt. Abdampfen des Lösungsmittels und Trituration mit Äthanol ergab Kristallisation
des Hydrochlorid-Salzes von 1,4-Äthano-e-oxo-

7-äthyl-1,2,3,4,6,7,8,8a-octahydroquinolin, F. über 300°C
(Zers.). Die freie Base, F. 63-5°C, kann durch Destillation
unter vermindertem Druck, Sdp. 3mm Hg: 1480C, gereinigt
werden, Ausbeute 60 %.
Die erfindungsgemäßen V rbindungen der Formel III, worin
X CH„ ist, haben pharmakologische Eigenschaften, analog
den Verbindungen der Formel II, worin X =0 ist,- Sie sind
von speziellem Wert als psychomotrische Stimulantien. Sie sind wirksame Antagonisten zu den drpressiven Effekten von
Barbituraten und verhindern Zittern bei Tieren, die mit Oxotremorin in einer Dosis von 0.3 g bis 3.0 g pro kg
vorbehandelt sind. Da ihr Metabolismus auf anderen Wegen verläuft als der ähnlicher Verbindungen mit Valerolacton-Struktur,
können sie in pharmazeutischen Präparaten verwendet werden, die Verbindungen dieser zwei Typen enthalten.
Die mittlere lethale Toxizitätsdosis von erfindungsgemäßen
Verbindungen wurde gemäß Litchfield et al., J.Pharmacol. Exp. Ther. 96, 39 (1949) bestimmt. In jedem Experiment wurden
mindestens 5 Gruppen von 6 Mäusen für jede Dosis verwendet. Die LD^0 wurde mit 95 % Zuverlässigkeit bestimmt.
Meerschweinchen-Ileum-Test:
Periphere Antimuscarin-Aktivität wurde mittels der kumulativen
Dosisrespons gemäß Kuhnen-Clausen, Toxicol. App. Pharmacol. 23, 443 (1972) bestimmt.
Dieser wurde gemäß Brinclecombe et al., J. Pharm. Pharmacol. 23, 745-757 (1971) durchgeführt.
Eine Prüfung der erfindungsgemäßen Verbindungen zeigte,
daß die mittlere Dosis lethalis der Hydrochlorid-Salze in

Mäusen nach subcutaner Injektion in Salzlösung bei 75 mg
bis 273 mg/kg Körpergewicht liegt. Die toxischsten Verbindungen sind solche, worin R=Äthyl und die wenigst toxischen
solche, worin R=Phenyl ist.
Die Verbindungen sind wirksam in der Verhinderung von Zittern, induziert in Mäusen durch Gabe von 200 Microgramm Oxotremorin
pro Maus (intraperitoneal). Die mittlere Dosis für Verbindungen II und III mit R=A"thyl ist 3 mg/kg bei subcutaner
Injektion und für Verbindung II mit R=Phenyl 6 mg/kg und für III mit R=Phenyl 0.6 mg/kg. Dam.i c ist der therapeutische
Index für diesen Effekt 25 bis 50, und 86 für die letztgenannte Verbindung. Das äquipotente molare Verhältnis zu
Atropin ist 10 zu 8. Verglichen mit Atropin ist der peripherale anti-cholinergische Effekt der erfindungsgemäßen
Verbindungen vernachlässigbar. Z.B. ist das äquipotente molare Verhältnis in der Vermeidung von Kontraktion des
Meerschweinchen-Ileums: Atropin: 1, Verbindung II mit
R=Äthyl: 7200, Verbindung II mit R=Phenyl: 9000, Verbindung
III mit R=Äthyl: 1300, R=Phenyl: 1700. So ist der zentrale Effekt der erfindungsgemäßen Verbindungen wesentlich
ausgeprägter als ihre periphären Effekte. Das Verhältnis von zentraler zu periphärer Aktivität für Atropin ist 1:35,
während das Verhältnis für erfindungsgemäße Verbindungen 3:1 gemäß Inch et al, J. Phar. Pharmacol. 25, 359 (1973) ist.
Neben anderen Effekten der erfindungsgemäßen Verbindungen
ist zu erwähnen schnelle Mydriasis bei topicaler Anwendung von Lösungen im Auge. Z.B. ergibt eine 2%ige Lösung am Kaninchenauge
einen Anfang von Mydriasis nach 8 bis 10 Minuten. Der Effekt dauerte etwa 1 Stunde. Injektion in Mäuse ergab
eine geringe Hyperthermie (nicht über +0.80C). Diese erreichte
ein· Maximum nach 20 Minuten. Erhöhte psychomotorische Aktivität
wurde in Mäusen gefunden, wie sich aus der Aufrichtungsrate ergibt. Eine Potentierung des Effekts von Nicotin ergab

sich bei Applikation in die obere Nackenganglien der Katze. Fasciculation der gestreiften Muskulatur wird
nur bei sehr hoher Dodis induziert, die sich der lethalen Dosis nähert.

- 18. Präparat nach Anspruch 17 in Form von Dosiseinheiten.
- 19. Präparat nach Anspruch 17, in Form einer mydriatischen Lösung.
- 20. Verfahren zur Herstellung von Verbindungen gemäß Anspruch 1, worin X Sauerstoff und R Alkyl ist, dadurch gekennzeichnet , daß man 2-Methylen-quinuclidin-3-on mit einem Alkyl-substituierten Dialkylraalonat in Gegenwart von Natriumalkoxid zu Dialkyl-(3-oxoquinuclidin-2-yl)-methyl-alkylmalonat umsetzt, letzteres mit Natriumborhydrid zu dem entsprechenden Dialkyl-(3-hydroxyquinuclidin-2-yl)-methyl-alkylmalonat reduziert, letzteres mit einer konzentrierten Mineralsäure zu dem gewünschten Produkt, einem 'J-Alkyl-ö-oxa-i-azatricyclo-(4.2.2.02/7)-dodecan-5-on-Salz, behandelt.
- 21. Verfahren zur Herstellung von Verbindungen der Formel I nach Anspruch 1, worin X Sauerstoff und R Aryl oder Heteroaryl ist, dadurch gekennzeichnet, daß man eine Reaktion gemäß Anspruch 20 durchführt, wobei Alkyl-aryl-acetat oder Alkylheteroaryl-acetat anstelle von Dialkyl-alkylmalonat verwendet wird.
- 22. Verfahren zur Herstellung von Verbindungen der Formel I nach Anspruch 1, worin X =CH2 und R Alkyl ist, dadurch ge-709820/1059-VS-kennzeichnet, daß man 2-Methylen-3-quinuclidinon mit einem alkyl-substituierten Alkyl-acetoacetat in Gegenwart von Natriumalkoxid umsetzt, das so erhaltene Zwischenprodukt mit einer Mineralsäure und Wasser zu 1, 4-A"thano-6-oxo-7-alkyl-1,2,3,4,6,7,8,8a-octahydroquinolin behandelt, welch letzteres mit Wasserstoff in Gegenwart katalytischer Mengen Palladium auf Kohle zu dem gewünschten Produkt reduziert wird.
- 23. Verfahren zur Herstellung von Verbindungen der Formel I nach Anspruch 1, worin X =CH2 und R Aryl oder Heteroaryl ist, dadurch gekennzeichnet^ daß man 2-Methylen-3-quinuclidinon mit einer Verbindung der Formel CH3-CO-CH2-Aryl oder CHg-CO-CH2-Heteroaryl in Gegenwart von Natriumalkoxid zu 1,4-Äthano-6-oxo-7-aryl- (oder -heteroaryl) -1,2,3,4,6,7,8,8aoctahydroquinolin umsetzt, welches mit Wasserstoff in Gegenwart katalytischer Mengen Palladium auf Kohle zu dem gewünschten Produkt reduziert wird.709820/1059
