Description
Die Erfindung betrifft neue Malonato-platinkoordinationsverbindungen
und ein diese Verbindungen enthaltendes Antitumormittel.
Platinkoordkiationsverbindungen und Verfahren zu
ihrer Herstellung werden von J. C. Bailar, Jr., The

Gegenstand der Erfindung sind Platinkoordinationsverbindungen
der Formel
Pt(II)A1(OOC)2CHR
entsprechend dem vorstehenden Patentanspruch 1.
Gegenstand der Erfindung ist außerdem ein Antitumormittel, welches dadurch gekennzeichnet ist, daß es
als Wirkstoff eine der im Anspruch 1 definierten Platinkoordinationsverbindungen enthält.
Die erfindungsgemäßen PIatin(II)-Koordinationsverbindungen
enthalten einen zweizähnigen Malonatoliganden, der substituiert oder unsubstituiert sein kann.
Als Substituenten R können die OH-Gruppe oder Methyl- oder Äthylgruppen vorliegen oder der Rest
CHR der Malonsäure kann zu einem Cyclobutanrest geschlossen sein.
Die erfindungsgemäßen Koordinationsverbindungen
enthalten außerdem zwei einzähnige Liganden, die Ammoniak oder primäre Amine sind, wenn χ in der
vorstehenden Formel 2 bedeutet, oder einen zweizähnigen Aminliganden, nämlich Äthylendiamine oder
Propylendiamine, wenn .v 1 bedeutet.
Geeignete einzähnige Aminliganden sind niedere Alkylamine, wie Methyl-, Äthyl-, n-Propvl-, Isopropyl-
oder n-Butylamin.
Die erfindungsgemäßen Koordinationsverbindungen weisen zwei identische oder verschiedene einzähnige
Liganden auf.
Geeignete zweizähnige Aminliganden sind Äthylendiamin
und Propylendiamin-
Bevorzugt werden Verbindungen, wie Malonatoäthylendiamin-platin(ll),
Methylmalonato-äthylendiaminplatin(ll) und Äthylmalonato-äthylendiamin-plalin(ll),
und Verbindungen, wie Malonato-diammin-platin(ll), Methylmalonato-diammin-platin(ll) und Äthylmalonato-diammin-platin(II).
Die erfindungsgemäßen Koordina'.ionsvcrbindiingen
können nach einer beliebigen einer Vielzahl bekannter Methoden hergestellt werden. Eine allgemeine Methode
zur Herstellung der Pt(ll)-Koordinationsverbindungert
wird nachstehend beschrieben: Ausgangsverbindungen der Formel
cis-[Pt A(HaI)3],
in der Hai für J, Cl oder Br steht und A einen zweizähnigen oder zwei einzähnige Aminliganden
bedeutet (erhalten nach der Methode von S. C. Dhara,
Indian J. Chem, Band 8, S. 193 [1970]) werden mit Silbernitrat unter Bildung des Diaquokomplexes umgesetzt.
Dieser wird dann mit dem Malonation unter Bildung der erfindungsgemäßen Koordinationsverbindungen
umgesetzt. Diese Methode wird durch das folgende Reaktionsschema dargestellt:

worin A einen zweizähnigen Aminliganden oder zwei einzähnige Aminliganden bedeutet.
Geeignete Methoden zur Herstellung der erfindungsgemäßen Verbindungen werden in den nachstehenden
Beispielen beschrieben.
Malonato-diamin-platin(II)
[Pt(NHj)2(C3H2O4)]

22,55 g Silbernitrat (geringfügig weniger als die stöchiometrische Menge, um eine Verunreinigung mit
Silber zu vermeiden) wurden in 50 ml Wasser gelöst und in einem konischen 250-ml-Kolben zu 20 g
[Pt(NHi)2CI2]
-,ο gegeben. Der Kolbeninhalt wurde unter raschem
Pühren auf einer Heizplatte auf 600C erwärmt, bis die Silbcrchlorid Ausfällung vollständig war und die Mutterlauge
fast farblos war. Das Silberchlorid wurde mit Hilfe einer feinporigen Glasfritte abfiltriert und der
,-, Niederschlag wurde mehrmals mit heißem Wasser gewaschen, wobei ein Gesamtvolumen des Filtrats von
IOO bis 200 ml erhallen wurde.
Ug Malonsäure (2facher Überschuß) wurden in 30 ml Wasser gelöst und mit einer Lösung von KOH
bo (etwa 13 g in JO ml) bis auf einen pH-Wert von 5 bis 6
neutralisiert. Die erhaltene Kaliummalonatlösung wurde zu dem Platin enthaltenden Filtrat gegeben und das
Gemisch wurde auf der Heizplatte vorsichtig erwärmt (um das Stoßen zu vermeiden) bis sich in großer Menge
b, weiße Kristalle des Produkts zu bilden begannen. Das
Gemisch wurde dann auf Raumtemperatur abgekühlt und das Produkt wurde abfiltriert. Das Filtrat wurde
erneut 5 bis IO Minuten erhitzt und auf O0C abgekühlt.
wobei ein weiterer Anteil gewonnen wurde. Die Rohausbeute in dieser Stufe betrug 20,5 g (93%).
Das Produkt wurde umkristallisiert, indem es in siedendem oder fast siedendem Wasser gelöst wurde.
Für die vorstehende Ausbeute (20,5 g) waren etwa 3 Liter siedenden Wassers zur völligen Auflösung
erforderlich. Malonsäure wurde in einer Menge von 1 g/I in dem Wasser gelöst, um Hydrolyse zu
unterdrücken (UV-spektrometrische Untersuchungen und Leitfähigkeitsmessungen haben gezeigt, daß die
Hydrolyse vernachlässigbar ist). Die filtrierte Lösung wurde auf 0°C abgekühlt, wobei weiße flockige Nadeln
(18,25 g; 83%) erhalten wurden.
Die Kristalle zersetzen sich zwischen 185 und 1900C.
Die Struktur des Produkts wurde durch das IR-Spektrum
bestätigt.
Die Löslichkeit des Produkts ist niedrig in kaltem Wasser und beträgt 20 mg/100 ml bei 20° C und
43 mg/100 ml bei 37°C, ist jedoch höher in fast siedendem Wasser (90 bis 100"C) und beträgt bei dieser
Temperatur etwa 0,65 g/100 ml.
Die empirische Zusammensetzung wurde durch Elementaranalyse bestätigt:

3,64 g Silbernitrat wurden in 20 mi Wasser gelöst und
zu 3,5 g [Pt(NH2);!(CH2)2CI2] gegeben, das in einem
konischen Kolben in 30 ml Wasser suspendiert war. Das Gemisch wurde 5 bis 10 Minuten auf einer warmen
Heizplatte gerührt, bis sich der gesamte gelbe Platinkomplex unter Bildung einer gelben Mutterlauge
und eines reichlichen weißen Silberchlorid-Niederschlags gelöst hatte. Das Gemisch wurde durch ein
feinporiges Filter filtriert und der Niederschlag wurde zweimal mit kleinen Volumenanteilen an heißem
Wasser gewaschen. Das klare Filtrat, das mit den Waschflüssigkeiten vereinigt worden war, wurde zu
einer wäßrigen Lösung von 2 g Methylmalonsäure in 20 ml Wasser gegeben, die auf einen pH-Wert von 5 bis
6 eingestellt worden war. Das Gemisch wurde 5 Minuten auf etwa 80°C erhitzt und dann auf O0C
abgekühlt. Es bildeten sich glänzende weiße Kristalle, die abfiltriert und mit kaltem Wasser und Aceton
gewaschen wurden (Ausbeute 2,65 g).
Die Mutterlauge und die Waschwässer wurden auf etwa die Hälfte ihres ursprünglichen Volumens
eingedampft (etwa 30 ml) wobei nach dem Abkühlen auf 0" C ein zweiter Anteil erhalten wurde (Ausbeute 0,85 g).
Die gesamte Rohausbeute betrug 3,50 g (88%). Der Komplex wurde aus einem möglichst geringen VoIunenanteil
an siedendem Wasser (etwa 250ml) umkrislallisiert,
wobei vor dem Abkühlen auf O0C durch ein feinporiges Filter filtriert wurde. Die Ausbeute an
glänzenden weißen Plättchen betrug 2.96 g (74%).

Eine zweite Ausbeute (0,33 g; 8%) wurde durch Volumenverminderung der Mutterlauge erhalten.
Allgemeine Strukturbestätigung
Aufgrund der beobachteten Veränderung in den Elektronenspektren, beim Übergang von der Aquo- zu
der Malonato-Verbindung zeigt sich, daß die Malonatgruppe mit dem Platin koordiniert ist Daher sind
Strukturen, wie
[Pt(NH3HH2OKl2(H2C3O4)
ausgeschlossen, wodurch die analytischen Daten bestätigt werden. Gleichzeitig zeigen die Messungen der
Nullpunkts-Leitfähigkeit eine neutrale Verbindung an. Die IR-Spektren zeigen das Vorliegen von koordinierten
Carboxylgruppen (1600 bis 1650 cm-' und 1400 cm-1) und die Abwesenheit von CO2-H-Gruppen
(die bei 1700 bis 1750 cm-' auftreten würden). Die Schwingungen der Carboxylgruppe sind schließlich in
Übereinstimmung mit einer Chelatstruktur, wie der Vergleich mit Oxalaikornplexen bekannter Struktur zeigt.
Die erfindungsgemäßen Verbindungen wurden auf ihre Antitumoraktivität unter Verwendung eines Standard-Screening-Tumors
(Testtumors), dem festen Sarkorn 180-Tumor in der weiblichen Schweizer Maus
geprüft, wobei die Standardvorschriften eingehalten wurden, die für diese Prüfung durch »The National
Cancer Institute« festgelegt wurden (Cancer Chemotherapy Rep. 25 [1962]).
Für diese Tests wurde ein S 180-Tumor, der aus einer getöteten Maus entnommen wurde, von überflüssigem
Gewebe befreit und unter sterilen Bedingungen in Stücke einer Größe von etwa 10 mg zerschnitten. Diese
Gewebestücke wurden dann mit Hilfe eines Trokars neuen Mäusen subkutan in dem linken Achselbereich
implantiert. Die Mäuse waren im Durchschnitt etwa 4 Wochen alt und wogen 18 bis 20 g. Wenn Tag 0 ab Tag
der Implantation angeommen wurde, wurden die Tiere am Tag 10 getötet. Die Tumoren vi-rden ausgeschält
und gewogen und das Verhältnis des Gewichts der Tumoren der Mäuse bei den behandelten Tieren zu der
Vergleichsgruppe von Tieren wurde festgestellt. Dieses
Verhältnis, multipliziert mit 100, wird als = -Verhältnis
in Tabelle 1 angegeben.
Bei der ersten Versuchsgruppe wurde die Koordinationsverbindung frisch in sterilem destilliertem Wasser
gelöst und am Tag 1 jeder der Testmäuse intraperitoneal injiziert. Das Injektionsvolumen betrug gewöhnlich
'/2 ml. In manchen Fällen wurde eine feine Dispersion
der für die Prüfung der erforderlichen Dosis bereitet, w«nn die chemische Substanz in dieser Lösungsmittelmenge
nicht löslich war. Einige der angegebenen Testergebnisse wurden daher an Tieren erhalten, denen
eine Aufschlämmung der Verbindung injiziert wurde. Diese Versuche wurden in der nachstehenden Tabelle I
so bezeichnet. Außerdem wurde bei einigen der Verbindungen etwa 1 ml der Lösung entweder in einer
einzigen Injektion oder in zwei im Abstand von einigen Stunden durchgeführten Injektionen von je '/>ml
injiziert. Diese Injektionen wurden anfänglich in 4 unterschiedlichen Dosiswerten für jede neue Verbindungen
gegeben, wobei für jeden Dosiswert 6 Mause verwendet wurden. Die Tests umfassen einen Dosierungsbereich
von einer niederen unwirksamen Dosis bis zu einem oberen Dosiswert, der innerhalb der
Versuchsdauer den Tod einiger Tiere verursachte. Die Ergebnisse sind in Tabelle I aufgeführt.

Außer den vorstehend beschriebenen Injektionen am Tag 1 wurden in mehreren Fällen die Injektionen bis
zum Tag 8 des Tumorwaehstums verschoben. In diesen Fällen war der Tumor gewöhnlich wenigstens größer als
'/2 g. wie durch Abtasten bestimmt wurde. Die Tiere erhielten dann eine Injektion und wurden während einer

Dauer von etwa 60 Tagen beobachtet. Die Aktivität wurde durch die Anzahl der Tiere gemessen, deren
Tumoren sich bis zum Punkt des Verschwindens zurückgebildet hatten, wobei das Tier während dieser
Dauer überlebte. Die dabei erhaltenen Prüfergebnisse sind in der nachstehenden Tabelle 2 aufgeführt.

Die in Tabellen 1 und 2 beschriebenen Ergebnisse korn in Ratten und den ADJ/PC6A-Tumor in Mäusen
zeigen an, daß die erfindiingsgemäßen Verbindungen durchgeführt. Die anfänglichen bzw. unmittelbaren
sehr wirksame Antitumormittel gegen den S 180-Tumor Testergebnisse sind in Tabelle 3 gezeigt und bestätigen

Die erfindungsgemäßen Malonato-platinkoordinationsverbindungen
werden vorzugsweise in Wasser oder anderen pharmazeutisch geeignete Trägerflüssigkeit
suspendiert oder gelöst Das parenteral verabrdchbare
Mittel sollte vorzugsweise etwa 0,5 mg ins etwa
mg pro ml enthalten, wobei natürlich diese Menge weitgehend in Abhängigkeit von der verwendeten
speziellen Verbindung und dem zu behandelnden Tier variieren kann.
Die erfindungsgemäßen Platinkoordinationsverbin-(hingen
werden vorzugsweise parenteral einem Tier verabreicht, das an einem malignen Tumor leidet Die
Behandlungsdauer und die Dosierung hängen natürlich in jedem Fall von der Größe des Wirtstiers, der Art und

Äthylendiamin oder Propylendiamin ist und R ein Wasserstoffatom, eine Hydroxygruppe, eine Methyl- ι ο
oder Äthylgruppe bedeutet oder die Gruppe CHR einen Cyclobutanrest darstellt
2. Antitumormittel, dadurch gekennzeichnet, daß es als Wirkstoff eine Verbindung nach Anspruch 1
enthält.
