Die Erfindung betrifft neue Trijod-isophthalsäure-monoaminosäureamide
der allgemeinen Formel E

worin A einen Aminoacylrest, R einen Alkylrest, der auch'durch eine
Hydroxy- oder. Alkoxygruppe substituiert sein kann, darstellt und X
'und Y verschieden sind und eine Hydroxygruppe oder die Gruppe /Rl "
bedeuten, wobei R1 und R0-unabhängig voneinander Wasser-
stoffatome, gegebenenfalls durch Hydroxygruppen substituierte
Alkylgruppen oder R-, und Rp zusammen mit dem Stickstoffatom
einen heterocyclischen Rest, der ein weiteres Heteroatom enthalten kann, bedeuten,
sowie deren Salze mit physiologisch verträglichen Basen. Als physiologisch verträgliche Basen kommen beispielsweise in
Betracht: Natronlauge, Glucamin, N-Methylglueamin, N,N-Dimethylglucamin,
Äthanolamin, Diäthanoiamin, Morpholin usw. In gewissem

Umfang kann man auch Alkali durch Erdalkali, wie Calcium- und
Magnesiumhydroxid, ersetzen. Es können auch Mischungen der Salze
verwendet werden.
Der Aminoacylrest leitet sich von einer üblichen Amihocarbonsäure
ab. Die Aminocarbonsäure kann eine beliebige Konfiguration einnehmen.
Es können ein oder zwei Aminogruppen in beliebiger Position stehen, wenn auch die a-Aminocarbonsäuren bevorzugt sind.
Weiterhin können die Aminocarbonsäuren auch ungesättigt, verzweigt, mehrbasisch und in üblicher Weise substituiert sein,
z.B. durch Hydroxy-, Mercapto-, gegebenenfalls substituierte Aryl-, Cycloalkyl- oder heterocyclische Gruppen. Ferner kann die
Aminogruppe durch aliphatische, aromatische oder gemischt aromatisch-aliphatische
Reste substituiert sein. Als bevorzugte Aminocarbonsäuren seien beispielsweise genannt:
Glycin, Sarkosin, Alanin, N-Phenylalanin, N-Benzylalanin, Valin,
Leucin, Isoleucin, Serin, Threonin, Cystein, Methionin, Ornithin, Lysin, Asparaginsäure, Glutaminsäure, Asparagin, Glutamin, Arginin,
Phenylalanin, Tyrosin, Prolin, Oxyprolin, Tryptophan, Histidin, ferner auch ß-Aminosäuren, wie ß-Alanin, oder Oligopeptide,
wie Glycyl-glycin, Glycyl-1-Leucin usw.
R, und R^ zusammen mit dem Stickstoffatom sollen vorzugsweise
den Pyrrolidin-, Morpholin- oder Piperazinrest bedeuten. Unter Alkyl und Alkoxy sollen vorzugsweise niedere Reste mit
1-6 Kohlenstoffatomen verstanden werden.
Die neuen Verbindungen stellen wertvolle Röntgenkontrastmittel ;

Endprodukte der allgemeinen Formel E, in der X die Gruppe

stellt werden, indem man Verbindungen der allgemeinen Formel Z1

mit Aminosäuren in Gegenwart organischer oder anorganischer Basen oder mit Aminosäureestern umsetzt und. gegebenenfalls anschließend
die erhaltenen Ester verseift.
Die Umsetzung mit Aminosäuren oder Aminosäureestern wird vorzugs
weise in cyclischen Äthern, wie Dioxan oder Tetrahydrofuran, vorgenommen.
Die als Ausgangsmaterialien verwendeten Verbindungen der allgemeinen
Formel Z1 sind in der deutschen Offenlegungsschrift Nr. 2 031 724 beschrieben.

Die Verbindungen der allgemeinen Formel Z, eine besonders wichtige
Gruppe von Ausgangsverbindungen, können nach an sich bekannten
Verfahren hergestellt werden.
Zur Herstellung der Verbindungen Z, in denen X die Hydroxygruppe

und Y die Gruppe N^" darstellt, wird z.B. 3-Methoxycarbonyl-

5-nitro-benzoylchlorid (J.Med.Chem. 6 (1963)24-) mit einem Aminosäureamid zu dem entsprechenden N-(3-Methoxycarbonyl-5-nitrobenzoyl)-aminosäureamid (I) umgesetzt. Dieses wird in üblicher Weise zur Säure II verseift, durch Reduktion (Druckhydrierung mit Raney-Mckel als Katalysator) in das Amino-isophthalsäure-monoaminosäureamid (III) überführt, das dann beispielsweise mit KJCl^ zu der gewünschten Verbindung IV jodiert werden kann.
5-nitro-benzoylchlorid (J.Med.Chem. 6 (1963)24-) mit einem Aminosäureamid zu dem entsprechenden N-(3-Methoxycarbonyl-5-nitrobenzoyl)-aminosäureamid (I) umgesetzt. Die Verbindungen der allgemeinen Formel Z, in denen X die Gruppe N und Y die Hydroxygruppe bedeutet, können hergestellt
werden, indem man z.B. 3-Methoxycarbonyl-5-nitro-benzoylchlorid
(J.Med.Chem.6(1963)24·) mit einer Aminosäure zur entsprechenden
N-(3-Methoxycarbonyl-5-nitro-benzoyl)-aminosäure (VI) umsetzt;
anschließend die Methoxycarbonylgruppe mit einem entsprechend substituierten Amin oder Ammoniak zu VII amidiert, die Nitrogruppe
hydriert und die Aminoverbindung (VIII) zu IX jodiert.

Verfahren 1:
Zu 193,3 g (1,55 Mol) Glycin-methylamid-hydrochlorid in 3»75 Liter
Wasser werden 378,0 g (1,5 Mol) Natriumbicarbonat zugefügt unter
Eiskühlung. 365,4- g 3-Methoxycarbonyl-5-nitro-benzoylchlorid in
/werden
1,3 1 Aceton/innerhalb von 3 Stunden zugetropft. Danach wird noch 1 1/2 Stunden bei Raumtemperatur nachgerührt, das Produkt abgesaugt, mit Wasser salzfrei gewaschen und im Vakuum bei 7O0C getrocknet.

8»5 g (0»03 Mol) N-(3-Methoxycarbonyl-5-nitro-benzoyl)-glycin
(VI a) in 130 ml absolutem Tetrahydrofuran gelöst, werden nach Zusatz von 4,2 ml Triäthylamin auf -15 C gekühlt und unter
Rühren mit 3 ml Chlorkohlensäureäthylester versetzt. Man rührt ca. 10 Minuten bei -10 bis -50C und fügt eine auf -15°C gekühlte
Lösung von 2,8 ml Methylamin in 20 ml absolutem Tetrahydrofuran tropfenweise unter Rühren langsam hinzu. Danach wird noch
30 Minuten bei-150C und anschließend über Nacht bei Raumtemperatur
gerührt, das TriäthylaminhydroChlorid abfiltriert, das Filtrat im Vakuum eingeengt und der Rückstand aus Acetonitril
umkristallisiert. Schmelzpunkt: 173-1740C.

443,0 g (1,5 Hol) N-{3- Methoxyearbonyl -5-nitro-benzoyl)-glycinmethylamid
in 5,5 1 Dioxan werden unter Zusatz von 3,3 1 O,5n
Natronlauge 2 Stunden bei Raumtemperatur gerührt. Danach werden noch 400 ml Wasser zugefügt, das Dioxan wird im Vakuum abdestilliert,
die Säure mit konzentrierter Salzsäure ausgefällt, abgesaugt, mit Wasser salzfrei gewaschen und im Vakuum bei
7O°C getrocknet.
Ausbeute: 408 g.

44-9,9 g (1,6 Mol) N-(3-Carboxy-5-nitrobenzoyl)-glycin-methylamid
in 2 1 Wasser werden unter Zusatz von 880 ml 2n Ammoniak gelöst und bei Raumtemperatur mit 10%-Raney-Nickel als Katalysator
bei etwa 120 atü hydriert. Nach Entfernen des Katalysators wird die Hydrierlösung als solche zur Jodierung weiterverwendet.
Die freie Verbindung kann aus dem in Methanol gelösten schaumigen
Ammoniumsalz durch Ausfällen mit Trifluoressigsäure erhalten werden.
N-(3-Carboxy-5-amino-2,4,6-tri,iod-benzoyl)-p;lycin-methylamid (IVa)
Eine Lösung des Ammoniumsalzes von N-(3-Carboxy-5-amino-benzoyl)-glycin-methylamid,
hergestellt durch Hydrierung von l,ß Mol
der entsprechenden Nitroverbindung wird auf 140 1 Wasser aufgefüllt, und unter Rühren werden 3,6 1 konzentrierte Salzsäure

wird vom Niederschlag abgesaugt, mit Wasser sorgfältig nachgewaschen»

Ausbeute: 910,5 g·
N-(3-Methoxycarbonyl-5-nitro--benzoyl)-p;lycin ( VI a)
Zu einer Lösung von 62,0 g (0,825 Mol) Glycin und 189,0 g
Natriumbicarbonat in 3 1 Wasser werden unter Rühren im Verlaufe
von 2 1/2 Stunden 183,0 g (0,75 Mol) 3-Methoxycarbonyl-5-nitrobenzoyl-chlorid in 750 ml Aceton tropfenweise zugefügt. Der
zunächst ausfallende Niederschlag geht nach weiterem 3-stündigem Verrühren allmählich wieder in Lösung. Danach wird 2 mal mit
Äther ausgeschüttelt, die wäßrige Phase mit konzentrierter Salzsäure angesäuert, das ausgefallene öl mehrmals mit Essigester
extrahiert und die vereinigten Essigesterauszüge nach Waschen mit Wasser und Trocknen mit Natriumsulfat mit der berechneten
Menge an Dicyclohexylamin versetzt. Nach einigem Stehen wird vom ausgefallenem Dicyclohexylammoniumsalz (Schmelzpunkt: 204-2O5°C
aus Chloroform/Äther)abgesaugt,mehrmals mit Äther gewaschen
und die Säure durch Verteilen zwischen Essigester und 2n Schwefelsäure wieder freigesetzt. Nach Waschen der Essigesterphasen mit
Wasser, Trocknen mit Natriumsulfat und Einengen im Vakuum erhält man 181,0 g der oben angegebenen Verbindung.

Zu 42,3 S (0,15 Mol) N-(3-Methoxycarbonyl-5-nitro-benzoyl)-glycin
(VI a) in I50 ml Methanol werden 15 ml flüssiges Methylamin
bei O0C zugefügt und 3 Tage bei Raumtemperatur
aufbewahrt. Danach wird die Reaktionsmischung im Vakuum bis zur Trockne eingeengt und der Rückstand mit absolutem Alkohol
unter Rückfluß behandelt. Nach Erkalten wird abgesaugt, mit Alkohol nachgewaschen und im Vakuum getrocknet.
Ausbeute: 43»2 g.
N-(3-Methylaminocarbonyl-5-amino-trit1odbenzoyl)-glycin (IX a)
43»7 g (0,14 Mol) N-(3-Methylaminocarbonyl-5-nitro-benzoyl)-glycin-methylammoniumsalz
(VII a) in 0,6 1 Wasser werden mit Raney-Nickel als Katalysator bei ca. 140 atü und Raumtemperatur
hydriert. Nach Entfernen des Katalysators werden in das Piltrat noch 15»4 1 Wasser, 280 ml konzentrierte Salzsäure und 280 ml
2 η KJClp-Lösung zugefügt und 48 Stunden bei Raumtemperatur
gerührt. Danach wird vom Niederschlag abgesaugt, mit Wasser sorgfältig nachgewaschen, das Produkt einige Zeit mit Wasser
verrührt und im Vakuum bei VO0G getrocknet.
Ausbeute: 74,2 g.
In analoger V/eise werden auch die anderen Ausgangsverbindungen hergestellt. In der folgenden Tabelle sind die Ausbeuten und

a) 629,0 g (1 Mol) N-(3-Carboxy-5-amino-2,4-,6-triood^benzoyl)-glycin-methylamid
(IV a), Schmelzpunkt 252-253°C (Zersetzung), werden in 1,2 1 Dimethylacetamid gelöst, und unter Kühlen
und Rühren werden 170 ml Acetylchlorid zugetropft. Man rührt bei Raumtemperatur über Nacht, fügt etwas Wasser hinzu und
engt im .Vakuum ein. Das zurückbleibende öl wird unter Rühren
mit 600 ml Wasser behandelt, wobei ein fester Niederschlag ausfällt. Man läßt einige Zeit stehen, saugt die Verbindung
ab, wäscht mit Wasser sorgfältig nach, löst dann in 2,5 1
Wasser unter Zusatz der berechneten Menge an 2n Ammoniak, behandelt die Lösung 2 Stunden mit Aktiv-Kohle und säuert
schließlich nach Entfernen der Kohle das Filtrat mit konzentrierter
Salzsäure an. Nach Stehen über Nacht wird das Produkt abgesaugt, mit Wasser gewaschen, danach mit Wasser
einige Zeit verrührt, wiederum abgesaugt und im Vakuum bei 700C getrocknet.
Ausbeute: 611 g (91 %) vom Schmelzpunkt 284~285°C (unter
Zersetzung).

(IV b), Schmelzpunkt 259-26O°C (Zersetzung), werden in 200 ml Dimethylacetamid gelöst, und unter Rühren werden
33»4- ml Acetylchlorid zugetropft. Man rührt noch 20 Stunden,
versetzt mit 40 ml Wasser und engt nach weiteren 30 Minuten
Rühren im Vakuum ein. Der Rückstand wird über Nacht mit 150 ml
Wasser verrührt, der Niederschlag wird abgesaugt, in I50 ml
In Natronlauge unter Zusatz von I5 ml 37%iger Natronlauge und

13,5 ml Acetylchlorid tropfenweise zugefügt. Danach wird noch
1 l/2 Stunden bei Raumtemperatur gerührt, weitere 2 ml Acetylchlorid werden zugefügt, und es wird eine weitere Stunde gerührt.
Nach Zugabe von 10 ml Wasser wird im Vakuum eingeengt, zum Rückstand 70 ml Wasser hinzugefügt und 16 Stunden gerührt. Der

methylamid
Zu 28,2 Mol Methoxyessigsäure in 100 ml Dimethylacetaraid läßt
man bei1 maximal 100C 29,0 ml Thionylchlorid unter Rühren innerhalb
von 40 Minuten zutropfen. Nachdem man noch weitere 2 Stunden bei O0C gerührt hat, werden in die Lösung 63,0 g (0,1 Mol)
N_(3_Carboxy-5-amino-2,4,6-trijod-benzoyl)-glycin-methylamid
(IV a), Schmelzpunkt 252-253°C (Zersetzung), in 100 ml Dimethylacetamid im Verlaufe von 1 Stunde bei maximal 8 C unter Rühren
tropfenweise hinzugegeben und dann über Nachtgerührt· Nach Zugabe von 5 ml Wasser wird im Vakuum eingeengt, der Rückstand
mit 700 ml Wasser gerührt, der Niederschlag abgesaugt, mit Wasser sorgfältig gewaschen und noch feucht aus verdünntem
Ammoniak mit konzentrierter Salzsäure umgefällt. Nach einigem Stehen wird die Säure abgesaugt, mit Wasser gut nachgewaschen,
mit frischem Wasser verrührt und nach nochmaligem Absaugen im Vakuum bei 700C getrocknet.
Ausbeute 54,0 g (77 %) dünnschichtchromatographisch einheitliches
Produkt vom Schmelzpunkt 226-2280C (unter Zersetzung).

Zu einer Lösung von 63 g (0,1 Mol) N-(5-Carboxy-5-ainino-2,4,
6-tri3od-benzoyl)-glycin-methylamid (IV a), Schmelzpunkt
252-253°C (unter Zersetzung)in 100 ml Dimethylacetamia werden
bei Temperaturen nicht höher als 1O0C 44 ml Acetoxy-acetylchlorid
langsam unter Rühren zugetropft. Nach 8 stündigem Rühren bei Raumtemperatur fügt man 5OO ml Wasser hinzu, saugt
den ausgefallenen Niederschlag nach einigem Stehen bei O0C ab,
wäscht gut nach und erwärmt dies(en nach Lösen in I50 ml 2 η
Natronlauge und 500 ml Wasser unter Verrühren 1 Stunde auf dem
Dampfbad. Nach Erkalten werden nochmals 5OO ml Wasser zugefügt
und das Produkt mit konzentrierter Salzsäure ausgefällt, Nach Abnutschen, sorgfältigem ITachwaschen und Trocknen im
Vakuum erhält man 51 g (74,2 %) Rohprodukt. Zur \*eiteren
Reinigung wird das aus Methanol isolierte Ammoniumsalz in wäßriger Lösung mit konzentrierter Salzsäure in die freie
Säure vom Schmelzpunkt 227-229°C (unter Zersetzung) überführt.
