5. Pharmazeutische Zubereitung, gekennzeichnet durch den Gehalt eines t-Butylaminopropiophenons
oder dessen pharmazeutisch verträglichen Säureadditionssalzes gemäß Anspruch 1 zusammen mit
einem pharmazeutisch verträglichen Träger.
Es wurde festgestellt, daß die Verbindungen der J5
allgemeinen Formel (I)

und ihre Säureadditionssalze, worin X Chlor oder Fluor 4 >
ist, wertvolle Eigenschaften als Antidepressive aufweiseii,
wenn sie nach Standardversuchen, wie sie allgemein zur Bestimmung der antidepressiven Wirksamkeit
verwendet werden, beispielsweise nach dem mil Tctrabenazin induzierten Sedationstest bei Nagern, vi
untersucht werden. Es wurde festgestellt, daß die Verbindungen der Formel (I) viel größere Dosen zur
Stimulierungswirkung als zur antidepressiven Wirkung erfordern. Die Verbindungen sind ebenso weder
Inhibitoren der Monoaminoxidase, noch haben sie blutdruckerhöhende Wirkung.
Eng verwandte Alkylaminopropiophenone sind bereits bekannt und wurden für verschiedene pharmazeutische
Zwecke eingesetzt; siehe beispielsweise die britischen Patentschriften 7 68 772, 10 11289 und
10 69 797. Die beiden Verbindungen der vorliegenden Erfindung weisen hervorragende Wirksamkeit auf, die
verwandte bekannte Verbindungen nicht besitzen.
Die Verbindungen können nach Verfahren, die dem Fachmann für Verbindungen dieser Art bekannt sind,
hergestellt werden.
Ein besonders geeigneter Weg wird in dem
nachfolgenden Reaktionsschema aufgezeigt:

Die Propiophenone (JV) sind nicht im Handel
erhältlich, und es wurde festgestellt, daß die Stufe 1) ein
geeignetes Herstellungsverfahren hierfür ist Die Bromierung der Stufe 2) verläuft nicht sehr schnell und
kann Erhitzen erforderlich machen. Es ist nicht notwendig, das Keton (V) als reine Substanz zu isolieren,
vorausgesetzt, daß der in Stufe 2) hergestellte Bromwasserstoff entfernt wird.
Es versteht sich, daß m der vorausgehenden Stufe 3) das m-X-«-Brompropiophenon (V) durch die entsprechenden
«-Chlor- oder a-Jodverbindungen ersetzt werden kann. m-X-a-Chlorpropiophenone können
zweckmäßig dadurch hergestellt werden, daß man m-X-Propiophenon mit Sulfurylchlorid umsetzt Ebenso
kann m-a-Dichlorpropiophenon zweckmäßigerweise is
aus oc-Chlorpropiophenon dadurch hergestellt'werden,
daß man dieses mit Sulfurylchlorid in Gegenwart von Aluminiumchlorid umsetzt. Die Reaktion der Stufe 3)
unterliegt sterischer Behinderung, und t-Butylamin reagiert normalerweise sehr langsam mit a-Brompropiophenon.
Es ist wünschenswert, dem Reaktionsgemisch ein organisches Lösungsmittel zuzugeben, und für
diesen Zweck bietet Acetonitril bemerkenswerte Vorteile. Es ist ein »schnelles« Lösungsmittel, ist unter
den Bedingungen nicht reaktionsfähig und weist einen relativ niederen Siedepunkt auf. Andere polare Lösungsmittel,
protonisch oder nicht protonisch, können verwendet werden, wie beispielsweise niedere aliphati
sehe Ketone oder Äther, jedoch verläuft die Reaktion in diesen Lösungsmitteln langsam. Zu weiteren Lösungsmitteln,
die verwendet werden können, gehören Dimethylformamid, Nitromethan, Dimethylsulfoxid und
Hexamethylphospi.oramid.
Es ist wünschenswert, die Reaktrrnspartner der Stufe
3) beispielsweise auf Rücknußtemperatur des Reaktionsgemischs zu erhitzen. Amin -st vorzugsweise
gegenüber dem Keton im Überschuß vorhanden, und es kann bis zur fünffachen Menge gegenüber der
äquimolaren Menge verwendet werden. Wenn das Keton ein m-X-<x-ChIorpropiophenon ist, kann eine
katalytische Menge eines Jodidsalzes, beispielsweise Natriumjodid, dem Reaktionsgemisch zugeführt werden.
Einmal isoliert, sind die m-X-«-t-Butylaminopropiophenone
der Formel (I) stabil und können unter Vakuum, obgleich dies normalerweise nicht notwendig
ist, destilliert werden. Es sind mäßig schwache Basen (pKa etwa 8,5 — 9), und sie werden wünschenswerterweise
als pharmazeutisch verträgliche Salze, geeigneterweise mit einer Mineralsäure, wie Salzsäure,
gelagert und verabfolgt. Unter physiologischen Bedingungen können sie vorherrschend, jedoch nicht
ausschließlich, kationisch sein. In jedem Falle können sie, wenn man sie als Salz verabfolgt, mit den
verschiedenen Anionen entsprechend den anderen, in dem Körper vorhandenen Säuren ein Gleichgewicht
bilden, und ihre Salze mit verschiedenen Säuren können nur im Hinblick auf die dem physiologischen Verhalten
nicht innewohnende Isolierung oder Löslichkeit Vorteile aufweisen. Es ist demgemäß festzustellen, daß alle t>o
Salze der Basen der Formel (I) mit nicht toxischen Säuren untereinander und gegenüber den Basen
äquivalent sind.
Es isl leicht zu erkennen, daß Salze von Säuren, clic
pharmazeutisch nicht verträglich sind, ebenso als hi
Zwischenprodukte zur Herstellung von verträglichen Salzen mittels doppeller Umsetzung, Basenaustausch
oder nach anderen bekannien Verfahren wertvoll sind.
Ein weiteres Verfahren, das zur Herstellung von
Verbindungen der Formel (I) und ihren Saureadditionssalzen
verwendet werden kann, besteht in der Entfernung einer Schutzgruppe Z von einer Verbindung
R-CO-CHMe-NZCMe3 [worin R die substituierte
Phenylgruppe der Formel (I), Z eine Aralkylgruppe (z. B.
eine Benzylgruppe) oder eine Acylgruppe (z. B. eine Acetyl- oder eine Benzyloxycarbonylgruppe) ist], die
nach bekannten chemischen Verfahren entfernt werden kann. So kann beispielsweise die Benzylgruppe durch
Hydrogenolyse entfernt werden. Ebenso kann die Benzyloxycarbonylgruppe durch Hydrogenolyse oder,
wie eine Acetylgruppe, durch Hydrolyse vorzugsweise mit einer Mineralsäure entfernt werden.
Ein weiteres Verfahren ist die Grignard-Kupplung eines Amids oder Nitrils der Formel (VII) mit einem

(worin Y eine Carbamoyl-, wie Dimethylcarbamoyl- oder Cyanogruppe und Hai ein Halogenatom ist), mit
nachfolgender Hydrolyse. Es ist allgemein bekannt, daß eine solche Reaktion unter vasserfreien Bedingungen,
vorzugsweise mit einem Molarüberschuß des Grignard-Reagens, beispielsweise in Äther oder Tetrahydrofuran
durchgeführt werden kann. Die Hydrolyse kann mit Wasser oder einer verdünnten wäßrigen Mineralsäure
vorzugsweise unter 100C durchgeführt werden.
Ein weiteres Verfahren besteht in der Oxidation des entsprechenden Amino-sekundären Alkohols der Formel
(VIII). Die Oxidation kann einfach nach bekannten Verfahren und besonders unter Anwendung der
Oppenauer-Oxidationsreaktion unter Verwendung eines Ketons, beispielsweise Acctons oder Cyclohexanone,
in Gegenwart eines Aluniiniunralk.oxids, beispielsweise
Aluminiumisopropoxids, bewirkt werden.

Die Verbindungen der Formel (I) und ihre Säureadditionssalze können ebenso durch die Reaktion eines
m-X-«-Alkoxypropiophenons der Formel (IX)

worin OR ein Niedrigalkoxyrest mit I bis 3 Kohlenstoffatomen, vorzugsweise Äthoxy ist und X die vorausgehend
erläuterte Bedeutung hat, mit t-Butylamin erhalten werden. Diese Reaktion wird vorzugsweise in einem
polaren Lösungsmittel, wie vorausgehend beschrieben, durchgeführt, wobei die Reaktionspartner auf die
Rüekflußtemperatur des Reaktionsgemischs erhitzt
werden. Die Verbindung der Forme] (IX) kann durch Umsetzung eines Alkalimetallalkoxids mit dem entsprechenden
n>«-Dihalogenpropiophenon erhalten werden.
Die Verbindungen der Formel (I) können weiterhin durch Entfernen einer /J-Keto-Schutzgruppe, beispielsweise
einer Oximgruppe, unter Bildung der gewünschten Carbonylverbindungen der Formel (I) hergestellt
werden. So kann beispielsweise das entsprechende Oxim der Formel (X) umgewandelt werden in die
gewünschten Endprodukte durch Äquilibrieren mit einer reaktionsfähigen Carbonylverbindung,

oder mittels Hydrolyse unter Verwendung solcher Reagenzien wie Mineralsäuren. Das Oxim (X) kann
durch Umsetzen des «-Halogen-JJ-nitroso-m-X-propiophenons
mit t-Butylamin hergestellt werden; die Nitrosoverbindung als solche kann durch die Reaktion
von m-X-Propylenbenzol mit Nitrosylchlorid hergestellt
werden.
Weiterhin betrifft die vorliegende Erfindung eine pharmazeutische Zubereitung (vorzugsweise in Form
von Dosierungseinheiten), die eine Verbindung der Formel (I) (oder ein pharmazeutisch verträgliches Salz
derselben) mit einem pharmazeutisch verträglichen Träger enthält. Geeigneterweise bildet die Verbindung
der Formel (I) oder ihr Säureadditionssalz 5 bis 95 Gew.-% der Zubereitung.
Zur Behandlung eines depressiven Zustands bei Säugern, wie Menschen, Mäusen oder Ratten verabfolgt
man eine antidepressiv wirksame, nicht toxische Menge (Dosis), vorzugsweise in Form von Dosierungseinheit, 4η
einer Verbindung der Formel (I) oder einer pharmazeutisch verträgliches Salz hiervon, und zwar oral,
parenteral oder rektal.
Eine erfindungsgemäße pharmazeutische Zubereitung kann in getrennten Einheiten, wie Tabletten, 4-,
Kapseln, Ampullen oder Suppositorien, dargeboten werden.
Die bevorzugte Dosis zur parenteralen Verabfolgung einer Verbindung der Formel (I) (berechnet als Base)
beträgt ungefähr 5 bis 50 mg/kg Körpergewicht, und die
insbesondere bevorzugte Dosierung beträgt 15 bis 35 mg/kg Körpergewicht des Säugers.
Bei der oralen oder rektalen Verabfolgung beträgt die bevorzugte Dosierung einer Verbindung der Formel (I)
(errechnet als Base) ungefähr 10 bis 100ing/kg -,-Körpergewicht,
während die insbesondere bevorzugte Dosierung (errechnet als Base) ungefähr 30 bis
70 mg/kg beträgt. Eine Verbindung der Formel (I) oder ein Säureadditionssalz derselben wird vorzugsweise
viermal täglich verabfolgt, obgleich sich die Anzahl der täglichen Verabfolgungen nach der Verordnung im
Hinblick auf den zu behandelnden Patienten (Sauger) und nach der Erfahrung des Arztes ändern kann.
Zur Behandlung von Menschen ist die bevorzugte Dosicrungseinhcit einer Verbindung der Formel (I) oder ·,->
deren Säurcadditionssalz (errechnet als Base) zur oralen
Verabfolgung oc'cr Verabfolgting als Suppositorium
ungefähr I^ bis 3(11) mg. wobei die bevorzugtere
Dosierungseinheit im Bereich von ungefähr 100 bis 300 mg und insbesondere von etwa 125 bis 250 mg liegt.
Bei der Konditionierung der erfindungsgemäOen
Verbindungen finden die üblichen Zusätze und Methoden Anwendung.
Beispiele einiger pharmazeutisch verträglicher Salze, die verwendet werden können, sind Salze der folgenden
Säuren: Salzsäure, Schwefelsäure, Phosphorsäure und Toluolsulfonsäure.
Die Erfindung wird durch die nachfolgenden Beispiele erläutert:
Herstellung von m-Chlor-«-t-butylaminopropiophenon a) m-Chlorpropiophenon
Zu Äthylmagnesiumbromid (21:3 molar) wurden
während 45 Min. unter Rühren und Kühlen m-Chlorbenzonitril
(688 g; 5 Mol) in Äther (2,5!) zugegeben. Die erhaltene Lösung wurde 5 Std. unter milden Rückflußbedingungen
erhitzt.
Das Reaktionsgemisch wurde mit kalter verdünnter Salzsäure hydrolysiert, der Äther a! (destilliert und die
wäßrige Lösung 1 Std. bei 90°C erhitzt. Der Kolben wurde dann gekühlt und geimpft Das sich abtrennende
feste Keton wurde mit kaltem Wasser gewaschen und aus Methanol umkristallisiert. Das umkristallisierte
Keton hatte einen Schmelzpunkt von 39 bis 400C und wog 750 g.
b) m-Chlor-Ä-brompropiophenon
In Methylenchlorid (3 1) wurde m-Chlorpropiophenon
(698 g; 4,15 Mol) gelöst. Die Lösung wurde mit Holzkohle und Magnesiumsulfat 2 Std. gerührt und
filtriert. Hierzu wurden unter Rühren 662 g (4,15 Mol weniger 2 g) Brom in Methylenchlorid (1,1 I) zugegeben.
Wenn die Bromfarbe vollkommen verschwunden war, wurde das Lösungsmittel unter Vakuum verdampft.
cJm-Chlor-a-t-butylaminopropiophenonhydrochlorid
Der gemäß b) erhaltene ölige Rückstand wurde in Acetonitril (1300 ml) gelöst. Hierzu wurde t-Butylamin
(733 g) in Acetonitril (1300 ml) zugegeben, wobei die Temperatur unter 32°C gehalten wurde. Man ließ das
Reaktionsgemisch über Nacht stehen. Es wurde dann zwischen Wasser (4200 ml) und Äther (2700 ml) verteilt.
Die wäßrige Schicht wurde mit einem weiteren Anteil Äther (1300 ml) extrahiert. Die kombinierten Ätherschichten
wurden dann mit Wasser (4200 ml), dem Salzsäure zugegeben wurde, gewaschen, bis der
pH-Wert der wäßrigen Schicht 9 war. Die wäßrige Schicht wurde abgetrennt, mit Äther (500 ml) gewasc'ii
η und dann verworfen. Die kombinierten Ätherschichten wurden dann mit Eis (560 g) und konzentrierter
Salzsäure (324 g) gerührt. Die Ätherschicht wurde abgetrennt und erneut mit Wasser (200 ml) und
konzentrierte,· Salzsäure (50 ml) gewaschen. Die beiden letzten Säursschichten wurden kombiniert und unter
Vakuum konzentriert, bis Kristalle erschienen. Die Lösung wurde dann auf 5°C abgeschreckt und filtriert,
Das Produkt wurde dann trockengesaugt, :nit Aceton gewaschen und aus einem Gemisch von Isopropanol
(3 I) und absolutem Äthanol (800 ml) umkristallmert.
Das so erhaltene Dl.-m-Chlor-'Vt-butylaminopropiophcnonhydrochlorid
war analytisch und spcktmgraphisch
rein; Schmelzpunkt 233 — 254' C.

Das NMR-Spektrum entsprach der angegebenen Strukturformel.
Herstellung von m-Fluor-«-t-butylaminopropiophenon
a) m-Fluorpropiophenon
Äthylmagnesiumbromid wurde mit m-Fluorbenzonitril nach dem Verfahren von Beispiel l(a) umgesetzt.
Das Keton hat einen Siedepunkt von 89°C/8 mm Druck und einen Schmelzpunkt von 29—300C.
bJm-Fluor-A-t-butylaminopropiophenonhydrochlorid
Die Bromierung des in Absatz 2(a) beschriebenen Produkts wurde, wie in Beispiel l(b) beschrieben,
durchgeführt, und die Reaktion des a-Bromketons mit t-Butylamin erfolgte wie in Beispiel l(c) beschrieben.
Das DL-Aminoketonhydrochlorid hatte einen Schmelzpunkt von 225-226°C.
Während in den Verfahren der Beispiele I und 2 die Hydrochloride unmittelbar ohne Isolierung der Basen
erhalten wurden, ist es ebenso möglich, die Basen durch Vakuumdestillation abzutrennen. m-Chlor-*-t-butylaminopropiophenon wurde bei 52° C und 0.005 mm Hg
Druck destilliert. Das m-Fluor-Analoge wurde bei 44—480C und 0.04 mm Hg Druck destilliert.
Beispiel 3 (a) <x-Chlorpropiophenon
Propiophenon (26 g: 0,2 Mol) wurde in Tetrachloräthan unter Bildung einer Lösung (100 ml) gelöst, in die
Sulfurylchlorid (27 g; 0,2 MoI) bei Zimmertemperatur gegeben wurde. Es erfolgte sofortige Wärmebildung
unter 5 Min. langer, kräftiger Bildung von sauren Dämpfen. Nach halbstündigem Stehenlassen wurde das
Gemisch 20 Minuten auf 90° C erwärmt, dann verdampft
und das hinterbleibende Öl abdestilliert unter Bildung von oc-Chlorpropiophenon in drei Fraktionen mit einem
Siedepunkt von 76° C bei 05 mm Hg.
(b) Λ-m-Dichlorpropiophenon, hergestellt aus
Propiophenon über «-Chlorpropiophenon
Propiophenon (67,1 g; 05 MoI) wurde in Tetrachloräthan (200 ml) gelöst und auf ungefähr 0°C gekühlt
Sulfurylchlorid (74,2 g; 055 Mol) wurde zugegeben und
die Temperatur unter 300C gehalten. Wenn die Bildung
von sauren Dämpfen aufgehört hatte, wurde die Lösung 10 Minuten auf 90° C erhitzt und dann auf 15° C gekohlt
während wasserfreies Aluminiumchlorid (146,7 g; 1,1 Mol) zugegeben wurde. Die Suspension wurde 1 Std. bei
70° C erhitzt wenn ungefähr die Hälfte des Aluminiüüichiorids gelöst war. In die Suspension wurde unter
Rühren Sulfurylchlorid (74,2 g; 055 MoI) zugegeben.
und das Gemisch wurde 7,5 Std. bei ungefähr 80"C
erhitzt. Weiteres Sulfurylchlorid (74 g; 0.5 Mol) wurde zugegeben und das Gemisch bei 85"C 4.75 Std. erhitzt.
Das Reaktionsgemisch wurde in Eis (I kg) gegossen. Ί abgetrennt und die wäßrige Schicht mit Chloroform
extrahiert. Die gesamte organische Schicht (700 ml) wurde mit Wasser (2 χ ). IO%iger wäßriger Natriumhydroxidlösung (3 χ 150 ml). Wasser (2 χ 400 ml) gewaschen und über wasserfreiem Magnesiumsulfat/Holz-
i" kohle getrocknet. Filtrieren und Verdampfen lieferte ein
braunes öl, das etwas Tetrachloräthan enthielt und eine Reinheit von ungefähr 80%, ermittelt durch Dampfphasenchromatographie, hatte. Siedepunkt 98—99°C/
1 mm Hg.
r> Das IR-Spektrum des Λ-m-Dichlorpropiophenon
enthält eine starke Bande bei 1700 cm1 pC = 0), und die
übrigen Banden wiesen auf einen m-substituierten Benzolring hin. Das NMR-Spektrum entsprach der
angegebenen Strukturformel.
(c) m-Chlor-A-t-butylaminopropiophenon
Das Öl (24.8 g; ca. 0,1 Mol) von Stufe (b) wurde in
Acetonitril (60 ml) und t-Butylamin (36,6 g; 0,5 Mol) gelöst und Natriumjodid (5 g: 0,033 Mol) zugegeben. Die
2~> Lösung wurde 10 Std. am Rückfluß gehalten und 56 Std.
bei Zimmertemperatur stehengelassen. Der größte Teil des überschüssigen t-Butylamins wurde unter reduziertem Druck entfernt und Wasser (200 ml) zugegeben.
Dann wurd~ mit Äther (2 χ 110 ml) extrahiert und die
Ätherschicht mit Wasser, das etwas konzentrierte Salzsäure enthielt, so gewaschen, daß das; Waschprodukt einen pH-Wert von 9 aufwies. Dieses wurde
seinerseits mit Äther (100 ml) gewaschen. Die Gesamtätherphase wurde mit Eis (35 g) und konzentrierter
π Salzsäure (15 ml) behandelt und abgetrennt, und der Äther wurde mit Wasser und konzentrierter Salzsäure
(2 : 1, 27 ml) gewaschen. Die gesamte wäßrige Salzsäureschicht wurde mit Äther (15 ml) gewaschen, mit
Holzkohle behandelt, nitriert und zu einem dicken Harz
4fi verdampft, dem Aceton zugegeben wurde. Die dicke
weiße kristalline Ausfällung wurde filtriert mit Aceton (2 χ) gewaschen und unter Bildung von m-Chlor-*-tbutylaminopropiophenonhydrochlorid getrocknet.
Herstellung von m-Chlor-a-t-butylaminopropiophenon
Stufe (a) — Λ-m-Dichlorpropiophenon
m-Ch!orpropiophenon (8,4 g; 0,05 Mol) wurde in Tetrachloräthan (20 ml) gelöst und Sulfurylchlorid
(7,4 g; 0,055 MoI) zugegeben. Nach Erwärmen auf 50° C trat eine heftige Gasbildung ein, die von selbst be; η
Stehenlassen bei Zimmertemperatur 1 Std. fortdauerte. Nach Zugabe von Sulfurylchlorid (3 g) wurde die
Lösung 05 Std. auf ungefähr 90° C erhitzt Das Gemisch
wurde Ober Nacht bei Zimmertemperatur stehengelassen und zu einem Öl verdampft, das in wenig Benzol
gelöst und zweimal mit Wasser gewaschedn, über wasserfreiem Magnesiumsulfat getrocknet und zu
einem braunen öl (11 g) verdampft wurde, das eine sehr
geringe Menge Tetrachloräthan enthielt Das Produkt hatte etwa 90%ige Reinheit, festgestellt mittels
Dampfphasenchromatographie.
Stufe (b)-
m-Chlor-Ä-t-butylaminopropiophenohydrochlorid
cX-ns-Dichlorpropiophenon (55 g; ca. 0,025 Mo!) des
Öls von Stufe (a) wurde in Acetonitril (15 ml) gelöst und
t-Butylamin (9,1 g; 0.125 Mol zugegeben. Der Ansät/
wurde 22 Std. am Rückfluß gehalten und wie in Beispiel 3(c) weitergearbeitet unter Bildung von m-Chlor-A-t-butylaminopropiophenonhydrochlorid.

Tablettenansatz für 300 mg Tabletten-Bestandteile

50 mg Maisstärke
10 mg fein zerkleinertes Silicagel
5 mg Polyvinylpyrrolidon

5 mg Polyvinylpyrrolidon
Milchzucker, Maisstärke und Salz wurden gemischt und mit einem Bindemittel, Polyvinylpyrrolidon in
alkoholischer Lösung, granuliert. Die Granulate ließ man durch ein Sieb mit einer lichten Maschenweite von
0,991 bis 0,833 mm laufen, dann wurden sie luftgetrocknet, mit fein verteiltem Silicagel als Gleitmittel
behandelt und in Tabletten gepreßt. Es konnte dann, wenn gewünscht, eine Filmbeschichtung aufgebracht
werden.
Beispiel 6
Kapseiherstellung — 400 mg Füllgewicht
Kapseiherstellung — 400 mg Füllgewicht
150 mg Hydrochloridsalz der Verbindung der Formel (I) wurden mit 125 mg Milchzucker und 125 mg
Maisstärke gemischt. Das Gemisch wurde in eine zweiteilige harte Gelatinekapsel gefüllt.

Parenteral Lösung
150 mg Hydrochloridsalz der Formel (I) wurden in sterilem Wasser unter Bildung von 1 ml gelöst. Eine
Mehrfachdosen-Zubereitung kann Bakteriostatika. wie 0,2 bis 0,5 Gewichts-% Volumen Phenol, enthalten.
Beispiel 8
Suppositorien — 400 mg Gewicht
150 g Hydrochloridsalz der Formel (I) wurden mit 250 mg weichgemachter oder geschmolzener Kakaobutter gemischt, und Suppositorien wurden durch
Abschrecken und Formen hergestellt.

4. Verfahren zur Herstellung einer Verbindung gemäß Anspruch 1, dadurch gekennzeichnet, daß
man jeweils in an sich bekannter Weise
