2. Verfahren zur Herstellung von Verbindungen gemäß Anspruch 1, dadurch gekennzeichnet, daU man in
an sich bekannter Weise 7-a-Thienylacetamidocephalosporansäure mit einer Verbindung der allgemeinen

worin M ein Alkalimetallatom ist, umsetzt und gewünschtenfalls die erhaltene Verbindung mit einem Alkylhalogenid

überführt.

annehmbaren Salze dieser Verbindungen.
Diese neuen Verbindungen sind Antibiotica, die beständig sind gegenüber Penicillinase und gut wirksam
gegenüber einem breiten Spektrum grampositiver und gramnegativer phatogener Organismen.
Die erfindungsgemäßen Verbindungen der obengenannten allgemeinen Formeln 1 und II lassen sich
herstellen, indem man in an sich bekannter Weise 7-a-Thienylacetamidocephalosporansäure mit einer
Verbindung der allgemeinen Formel

worin M ein Alkalimetallatom ist, umsetzt und gewünschtenfalls die erhaltene Verbindung mit einem
Alkylhalogenid mit 1 bis 6 Kohlenstoffatomen oder mit Allylbromid in eine Verbindung der allgemeinen
Formel II überführt.
Unter der Angabe »Alkyl« werden primäre, sekundäre und tertiäre geradkettige oder verzweigte Alkylreste
verstanden, wie Methyl, Äthyl, n-Propyl, Isopropyl, η-Butyl, Isobutyl, sec.-Butyl, tert.- Butyl,
n-Amyl, Isoamyl, 2-Amyl, 3-Amyl, neo-Pentyl, n-Hexyl
oder Isohexyl.
Die Angabe »pharmazeutisch annehmbare Salze« bezieht sich auf Salze mit Natrium, Kalium, Lithium,
Calcium, Barium, Magnesium, Aluminium und anderen Metallen mit annehmbar niedrigem Toxizitätsspiegel,
sowie mit Ammonium und eine Reihe organischer Stickstoffbasen, wie Methylammonium, Dimethylammonium,
Trimethylammonium, Tetramethylammonium, Cholin, die entsprechend substituierten Äthylammoniumionen, Procain, Chinin und Dibenzyläthylendiamin.
Zur Herstellung der Verbindungen der Formel I löst man ein Salz der 7-a-Thienylacetamidocephalosporansäure
in Wasser, versetzt mit einer wäßrigen Lösung eines Alkalipiperazinodithiocarboxylats, vorzugsweise
in wenigstens geringem Molüberschuß, und rührt und erwärmt mehrere Stunden bei gewöhnlicher
oder etwas erhöhter Temperatur. Der pH-Wert der Piperazinodithiocarboxylatlösung soll etwa 7,5 bis 8
nicht überschreiten und kann gegebenenfalls mit Salzsäure oder Schwefelsäure entsprechend eingestellt
werden. Die Umsetzung kann bei Temperaturen zwischen etwa 25 und 100° C, vorzugsweise bei etwa
40 bis 600C, und für eine Zeit von etwa 1 bis zu etwa 24 Stunden oder mehr durchgeführt werden. Die so
erhaltenen Produkte sind in vielen Fällen wasserunlöslich und können zweckmäßig abfiltriert und aus
Chloroform/Äther umkristallisiert werden. Wenn sie wasserlöslich sind, lassen sie sich durch Zugabe von
Natriumchlorid bis zu etwa dem halben Sättigungswert als gelbe, glasartige Körper aussalzen. Die Ausgangsstoffe
und Nebenprodukte bleiben dabei weitgehend in Lösung. Die gelben, glasartigen Körper lassen sich
!sieht durch Auflösen in Chloroform, Waschen mit halbgesättigter wäßriger Natriumchloridlösung zur
Entfernung von Verunreinigungen, Verdünnen mü Äther und Kristallisation reinigen.
Die oben erhaltenen Verbindungen der allgemeinen Forme! 1 können dann zur Herstellung der Verbindungen
der allgemeinen Formel II mit einem entsprechenden Alkylhalogenid, wie Methyljodid oder
-bromid oder Allylbromid in üblicher Weise umgesetzt werden.
ίο Die als Ausgangsstoff verwendete 7-u-Thienylacetamidocephalosporansäure
läßt sich leicht nach seit einiger Zeit wohlbekannten Methoden herstellen. Die zweckmäßigste und einfachste Arbeitsweise besteht in
der Acylierung von 7-Aminocephalosporansäure mit einem Acyliermittel der gewünschten Struktur unter
üblichen Bedingungen.
Dieals Ausgangsprodukte ferner verwendeten Piperazinodithiocarboxylate
lassen sich am besten nach dem Verfahren von Bögemann, Methoden der
Organischen Chemie (H ο u b e η — Wey 1), Stuttgart:
Georg Thieme Verlag, 1955, Bd. 9, S. 826. herstellen.
Die Erfindung wird an Hand der folgenden Beispiele erläutert. Mit nur geringen Abweichungen geht
man zur Herstellung der Verbindungen derallgemeinen Formel I wie folgt vor:
0,0012 Mol Natrium - 7 - « - thienylacetamidocephalosporanat
und eine äquimolare Menge des entsprechenden Natriumpiperazinodithiocarboxylats
werden in 10 ml Wasser gelöst und in einem thermostatisierten ülbad 24 Stunden auf 40 bis 45° C erwärmt.
Nach dieser Zeit ist die Lösung im allgemeinen klar. Das Produkt wird als gelber, glasartiger Körper
durch Zugabe eines gleichen Volumens wäßriger gesättijiter Natriumchloridlösung gefällt und mehrere
Stunden lang gekühlt. Die überstehende Lösung wird von der festen Phase dekantiert, und der Feststoff
wird in 25 bis 50 ml Chloroform gelöst. Die Chloroformlösung wird etwa lOmal nacheinander mit jeweils
12 bis 15 ml einer halbgesättigten wäßrigen Natriumchloridlösung gewaschen. In manchen Fällen, besonders
gegen Ende der Waschbehandlung, bilden sich störende Emulsionen, die sich jedoch durch
Zentrifugieren leicht brechen lassen. Zweckmäßiger-
45. weise werden während des Waschens qualitative Prüfungen der Ultraviolettspektren der Waschlösungen
vorgenommen. Ein Verschwinden des Spektrums der Ausgangsstoffe und ein Auftreten des Spektrums
des Produkts zeigt an, wann das Waschen beendet ist.
Die gewaschene Chloroformlösung wird auf das halbe Volumen oder weniger eingedampft, dann mit Äther
verdünnt und gekühlt. Das sich hierbei als feines Pulver abscheidende Natriumsalz des gewünschten
Produkts wird zentrifugiert und im Vakuum getrocknet.
Die Schmelzpunkte der Produkte sind infolge einer Zersetzungsneigung nicht scharf und schwanken daher
in Abhängigkeit von der Temperatur des Schmelzpunktblocks, bei der die Verbindungen aufgebracht
werden. Sämtliche Produkte weisen jedoch mit der erwarteten Struktur übereinstimmende Infrarotspektreri
auf und ergeben gegen Bacillus subtilis einen Fleck in Bioautogrammen von Papierchromatogrammen.

den Ultraviolettspektren, Titrationen und Analysen zur vollständigen Charakterisierung der Substanzen
aus.
7 - α - Thienylacetamido - 3 - (4' - methyipiperazinothiucarbonylthiomethyl)
- 3 - cephem -4 - carbonsäure · natriumsalz wird aus 7-u-Thienylacetamidocephalosporansäure
und 4-MethyIpiperazinodithiocarboxylat erhalten. Ausbeute: 23,5% der Theorie. pK^ 4,6 und
6,1. Ultraviolettabsorptionsmaximum bei 272 ηΐμ

1 - u- Thienylacetamido - 3 - (4' - äthylpiperazinothiocarbonylthiomethyl)-3-cephem-4-carbonsäure-
natriumsalz wird aus 7-a-Thienylacetamidocephalosporansäure
und 4-Äthylpiperazinodithiocarboxylat erhalten. "Ausbeute 6,1% der Theorie. Ultraviolettabsorptionsmaxima
bei 242 und 270 mu U■ = 16 550
bzw. 19 500).
7 - α - Thienylacetamido - 3 - (4' - η - propylpiperazinothiocarbonylthiomethyl)
- 3 - cephem - 4 - carbonsäurenatriumsalz wird aus 7-u-Thienylacetamidocephalosporansäure
und 4 - η - Propylpiperazinodithiocarboxylat erhalten. Ausbeute: 25,2% der Theorie. pK.;, 4.4
und 6,2. Ultraviolettabsorptionsmaximum bei 270 ηΐμ

- 3 - cephem -4 - carbonsäurenatriumsalz wird aus 7-a-Thienylacetamidocephalosporansäure
und 4-Isopropylpiperazinodithiocarboxylat.

7 - α - Thienylacetamido - 3 - (4' - η - butylpiperazinothiocarbonylthiomethyl)
- 3 - cephem - 4 - carbonsäurenatriumsalz wird aus 7-u-Thienylacetamidocephalosporansäure
und 4 - η - Butylpiperazinodithiocarboxylat erhalten. Ausbeute 23,2% der Theorie. pK;, 4,5
und 6,25. Ultraviolettabsorptionsmaximum bei 271 πΐμ
(f· = 21 600).
7 - α - Thienylacetamido - 3 - (4' - η -amylpiperazino-{hiocarbonylthiomethy!)
- 3 - cephem - 4 - carbonsäurenatriumsalz wird aus 7-«-Thienylacetamidocephalosporansäure
und 4 - η - Amylpiperazincdithiocarboxylat erhalten. Ausbeute: 14,2% der Theorie. ρ K^, 4,4
und 6,2. Ultraviolettabsorptionsmaximum bei 270 ηΐμ
(f = 19 700).
7 -«-Thienylacetamido - 3 -(4'-(ß- hydroxyäthyl)-piperazinothiocarbonylthiomethyl)-
3 -cephem - 4 -carbonsäure - natriumsalz wird aus 7 - α - Thienylacetamidocephalosporansäure
und 4-(/i-Hydroxyäthyl)-piperazinodithiocarboxylat erhalten. Das chloroformunlösliche
Rohprodukt wird als zwitterionische Verbindung bei pH 5,5 gefallt. Der Feststoff wird
abfiltriert, mit Wassei gewaschen und mit wäßriger 1 n-Natriumhydroxidlösung bei pH 7,2 wieder gelöst.
Die Lösung wird im Vakuum zur Trockne eingedampft und der Rückstand aus Methanol/Isopropanol
umkristallisiert. Ausbeute: 9,6% der Theorie. pK^, 4,4
und 5 8 Ultraviolettabsorptionsmaxima bei 235 und 270 rau U = 16 500 bzw. 20 500).
DieHerstellung der quaternären Piperazinium verbindungen
der allgemeinen Forme) II wird folgendermaßen durchgerührt:
Das 4-Alkylpiperazinodithiocarboxylatderivat des
Cephalosporins wird wie in den vorhergehenden Beispielen beschrieben hergestellt mit der Ausnahme,
daß man die Chloroformlösung nur 2mal mit der Natriumchloridlösung wäscht, dann eindampft und
den Rückstand trocknet. Das so erhaltene rohe Natriumsalz (etwa 0,01 Mol) wird in 100 ml trockenem
Chloroform gelöst und mit einer Lösung von 0,0/05 Mol von Alkylhalogenid oder Allylbromid in
10 ml Chloroform vermischt. Die Mischung wird bei Zimmertemperatur 4 bis 7 Tage unter gelegentlichem
Schütteln in einem verschlossenen Kolben aufbewahrt.
Während dieser Zeit erfolgt eine allmähliche Ausfallung von Feststoff. Der cremefarbene Niederschlag
wird abfiltriert und luftgetrocknet, und dann ;ur Entfernung von mitgefälltem Natriumiodid mit Wasser
verrieben. Das Produkt, ein gelbes wasserunlösliches Rohkristallisat, wird in Dimethylformamid
(25 bis 35 ml/g) unter leichtem Erwärmen und vorsichtiger Zugabe von Wasser bis zum Verschwinden
der Trübung aufgelöst. Dann werden 5 bis 10 Volumteile Tetrahydrofuran zugegeben, und die trübe Mischung
wird abgekühlt. Der gebildete Niederschlag wird zentrifugiert, mit Äther gewaschen und vakuumgetrocknet.
7-«-Thienylacetamido-3-(4',4'-dimelhylpiperazinothiocarbonylthiomethyl)
- 3 - cephem - 4 - carbonsäurebetain wird aus 7-a-Thieny!acetamido-3-(4'-methylpiperazinothiocarbonyl
thiomethyl) - 3 - cephem - 4 - carbonsäure und Methyljodid erhalten. Das gebildete
Betain ist im Gegensatz zu den anderen 4'.4'-quaternierten Produkten in Dimethylformamid unlöslich.
Es wird daher in warmem Dimethylformamid verrieben, abfiltriert, mit Wasser und dann mit Äther
gewaschen und getrocknet. Ausbeute: 48,0% der Theorie. pK^ 4,58. Ultraviolettabsorptionsmaximum
bei 269 ηΐμ ύ = 20 400).
7- u -Thienylacetamido- 3-(4'- methyl -4'-n- propylpiperazinothiocarbonylthiomethyl)-3-cephem-4-car-
bonsäure- betain wird aus 7 - α - Thienylacelamido-3 ..(4'. η - propylpiperazinothiocarbonylthiometbyl)-3-cephem-4-carbonsäure
und Methyljodid erhalten. Ausbeute 26,4% der Theorie. pK^, 4,6. Ultraviolettabsorptionsmaximum
bei 273 ιτίμ (>
= 23 000).
7-a-Thienylacetamido-3-(4'-methyl-4'-allylpiperazinothiocarbonylthiomethyl)
- 3 - cephem - 4 - carbonsäure - betain wird aus 7 - α - Thienylacetamido-3
- (4' ·■ methylpiperazinothiocarbonyllhiomethyl)-3-cephem-4-carbonsäure
und Allylbromid erhalten. Ausbeute: 66,3% der Theorie. pK^, 4,6. Ultraviolettabsorptionsmaximum
bei 271 πΐμ U — 21 650).
Beispiel 11
7 - α - Thienylacetamido - 3 -(4' - methyl - 4' - η - butylpi
perazinolhiocarbonylthiomethyl)- 3 -cephem -4 -carbonsäure -betain wird aus 7-K-Thienvlacetamido-
3-(4' - η - butylpiperazinothiocarbonylthiomethyl)-3-cephem-4-carbonsäure
und Methyljodid erhalten. Ausbeute: 25% der Theorie. pK.;, 4,6. Ultraviolettabsorptionsmaximum
bei 271 ma (> = 22 900).
Die Überlegenheit der erfindungsgemäßen Verbindungen
gegenüber den bekannten Amibiotica Cephalothin und Penicillin G geht aus einer Arbeit
in »Journal of Medicinal Chemistry«, 8, S. 174 bis
181. mit dem Titel »The Chemistry of Cephalosporins
IV« hervor, in der die zur Ermittlung dieser Daten herangezogenen Arbeitsweisen auch näher beschrieben
sind. Die Versuchsergebnisse sind dabei insbesondere in Tabelle I zusammengefaßt, wobei die
Zuordnung für die vorliegenden Verbindungen aus den Tabellen IV und Vl hervorgeht. Im einzelnen
wurden dabei folgende Ergebnisse erhalten:
