Die Erfindung betrifft neue Chinolin-, Isochinolin- und Phthalazinderivate als Antagonisten des Gonadotropin freisetzenden Hormons.
Description
Das Gonadotropin freisetzende Hormon (GnRH) ist ein Hormon, welches
überwiegend, aber nicht ausschließlich, in Säugern von Nervenzellen des
Hypothalamus synthetisiert, über die Portalvene in die Hypophyse
transportiert und reguliert an die gonadotrophen Zellen abgegeben wird.

aufweisenden Rezeptor stimuliert GnRH die Produktion und die Freisetzung
gonadotroper Hormone mittels der Second Messenger Inositol-1,4,5-
trisphosphat und Ca2+-Ionen. Die durch GnRH ausgeschütteten
Gonadotropine Luteinisierendes Hormon (LH) und Follikelstimulierendes
Hormon (FSH) stimulieren die Produktion von Sexualsteroiden und die
Keimzellreifung in beiden Geschlechtern. Zusätzlich zu GnRH (auch GnRH1
bezeichnet) gibt es zwei weitere Formen von GnRH, nämlich GnRH2 und 3.
Der GnRH-Rezeptor wird als pharmakologisches Target bei einer Reihe von
Erkrankungen verwendet, die von einer funktionierenden
Sexualhormonproduktion abhängig sind, beispielsweise Prostatakrebs,
prämenopausalem Brustkrebs, Endometriose und uterinen Fibroiden. Bei
diesen Erkrankungen können GnRH-Superagonisten oder -Antagonisten
erfolgreich eingesetzt werden. Eine mögliche weitere Indikation bildet
insbesondere die männliche Fertilitätskontrolle in Kombination mit einer
Substitutionsdosis von Androgenen.
Ein Vorteil von GnRH-Antagonisten im Vergleich zu Superagonisten ist ihre
unmittelbare Wirksamkeit bei der Blockierung der Gonadotropinsekretion.
Superagonisten bewirken anfänglich eine Überstimulation der Hypophyse,
die zu erhöhten Gonadotropin- und Sexualsteroidausschüttungen führt.
Diese hormonelle Reaktion wird aufgrund von Desensibilisierung und
Herabregulierung der GnRH-Rezeptorkonzentrationen erst nach einer
gewissen Verzögerung beendet. Möglicherweise können deshalb GnRH-
Superagonisten, sowohl alleine als auch in Kombination mit Testosteron,
die Spermaproduktion bei Männern nicht wirksam unterdrücken und sind
somit nicht für die männliche Fertilitätskontrolle geeignet. Im Gegensatz
dazu sind peptidische GnRH-Antagonisten, insbesondere in Kombination
mit einer Substitutionsdosis von Androgen in der Lage, eine signifikante
Oligozoospermie im Menschen hervorzurufen.
Peptidische GnRH-Antagonisten haben jedoch eine Reihe von Nachteilen.
So weisen sie eine erheblich geringere Wirksamkeit als Superagonisten auf
und müssen folglich in erheblich höheren Dosierungen verabreicht werden.
Auch ist ihre orale Bioverfügbarkeit gering, sodass sie durch Injektion
verabreicht werden müssen. Wiederholte Injektionen führen wiederum zu
einer Verringerung der Compliance. Darüber hinaus ist die Synthese von
peptidischen GnRH-Antagonisten im Vergleich zu nichtpeptidischen
Verbindungen aufwendig und kostspielig.
Chinolin-Derivate als nichtpeptidische GnRH-Antagonisten werden
beispielsweise in WO 97/14682 offenbart. Bisher konnten jedoch keine
nichtpeptidischen GnRH-Antagonisten auf den Markt gebracht werden.
Die der vorliegenden Erfindung zugrundeliegende Aufgabe bestand darin,
neue GnRH-Antagonisten bereitzustellen, die bekannten peptidischen
Verbindungen überlegen sind und eine wirksame Alternative zu bekannten
nichtpeptidischen Verbindungen darstellen. Die neuen GnRH-Antagonisten
sollen sowohl eine hohe Wirksamkeit als auch eine hohe orale
Bioverfügbarkeit besitzen. Weiterhin sollten sie einfach und mit möglichst
geringen Kosten synthetisiert werden können.
Diese Aufgabe wird gelöst durch Verbindungen der allgemeinen Formel (1):

aromatischen Ring durch ein oder mehrere Halogenatome, insbesondere

Verbindungen, bei denen mindestens einer von R3 und R4, insbesondere
beide, Wasserstoffatome sind.
Eine bevorzugte Bedeutung von Z ist eine direkte Bindung oder ein
Sauerstoffatom, während G vorzugsweise eine -C=C-Gruppe bedeutet. L
ist bevorzugt eine NH-Gruppe, während Q vorzugsweise eine
Carbonylgruppe und R51 eine C1-C6-Alkylgruppe ist. Besonders bevorzugte
Bedeutungen für R61 sind Wasserstoffatome oder C1-C3-Alkylgruppen,
insbesondere Methylgruppen und für R62 ein Aralkylrest, z. B. eine
Benzylgruppe.

Die erfindungsgemäßen Verbindungen (1) können als Antagonisten des
Gonadotropin-freisetzenden Hormons beispielsweise zur männlichen
Fertilitätskontrolle, zur Hormontherapie, zur Behandlung weiblicher Sub-
und Infertilität, zur weiblichen Empfängnisverhütung und zur
Tumorbekämpfung eingesetzt werden.
In der männlichen Fertilitätskontrolle bewirken die erfindungsgemäßen
Verbindungen eine Verringerung der Spermatogenese. Vorzugsweise erfolgt
eine kombinierte Verabreichung mit Androgenen, z. B. Testosteron oder
Testosteron-Derivaten, wie etwa Testosteronestern. Die Verabreichung der
Testosteron-Derivate kann beispielsweise durch Injektion, z. B. durch
intramuskuläre Depotinjektion erfolgen.
Auch in der Hormontherapie, beispielsweise zur Behandlung von
Endometriose, Uterus-Leiomyomen und uterinen Fibroiden können die
Verbindungen (1) gegebenenfalls in Kombination mit anderen Hormonen,
z. B. Östrogenen oder/und Progestinen, eingesetzt werden. Besonders
bevorzugt sind Kombinationen der erfindungsgemäßen GnRH-Antagonisten
und gewebeselektiven partiellen Östrogenagonisten wie Raloxifen®.
Darüber hinaus können die erfindungsgemäßen Verbindungen (1) zur
Erhöhung der weiblichen Fertilität, beispielsweise durch Induzierung der
Ovulation, und der Behandlung von Sterilität eingesetzt werden.
Andererseits sind die Verbindungen (1) auch für die Empfängnisverhütung
bei Frauen geeignet. So kann der GnRH-Antagonist an den Tagen 1 bis 15
des Zyklus zusammen mit Östrogen, vorzugsweise mit sehr geringen
Östrogendosierungen, verabreicht werden. An den Tagen 16 bis 21 des
Einnahmezyklus wird Progestagen der Östrogen-GnRH-Antagonist-
Kombination zugesetzt. Der GnRH-Antagonist kann kontinuierlich über die
gesamte Zyklusdauer verabreicht werden. Auf diese Weise kann eine
Verringerung der Hormondosierungen und somit eine Verringerung der
Nebenwirkungen von unphysiologischen Hormonspiegeln erreicht werden.
Weiterhin können vorteilhafte Wirkungen bei Frauen erzielt werden, die an
polyzystischem Ovariensyndrom und Androgen-abhängigen Erkrankungen
wie Akne, Seborrhoe und Hirsutismus leiden. Auch ist eine verbesserte
Zykluskontrolle gegenüber bisherigen Verabreichungsmethoden zu
erwarten. Weitere Indikationen sind benigne Prostatahyperplasie,
Gonadenprotektion bei Chemotherapie, kontrollierte Ovarienstimulation/
künstliche Reproduktionstechniken, frühkindliche Entwicklungsstörungen,
z. B. Pubertas praecox und polyzystische Ovarien.
Schließlich können die erfindungsgemäßen GnRH-Agonisten auch für die
Behandlung von hormonabhängigen Tumorerkrankungen, wie
prämenopausalem Brustkrebs, Prostatakrebs, Eierstockkrebs und
Endometriumkrebs eingesetzt werden, indem sie die endogenen
Sexualsteroidhormone unterdrücken.
Die erfindungsgemäßen Verbindungen (1) sind als GnRH-Antagonisten zur
Verabreichung für den Menschen, jedoch auch für veterinärmedizinische
Zwecke, z. B. bei Haus- und Nutztieren, aber auch bei Wildtieren geeignet.
Die Verabreichung kann auf bekannte Art und Weise beispielsweise oral,
topisch, rektal, intravaginal, nasal oder durch Injektionen erfolgen. Die orale
Verabreichung ist bevorzugt. Die Verbindungen (1) werden in eine
verabreichungsfähige Form gebracht und gegebenenfalls mit
pharmazeutisch akzeptablen Träger- bzw. Verdünnungsmitteln vermischt
werden. Die orale Verabreichung kann beispielsweise in fester Form als
Tablette, Kapsel, Dragee oder Pulver, jedoch auch in Form einer trinkbaren
Lösung erfolgen. Die nicht orale Verabreichung kann beispielsweise durch
intravenöse, subkutane oder intramuskuläre Injektion oder durch Salben,
Cremes oder Suppositoren erfolgen. Gegebenenfalls kann auch eine
Verabreichung als Retardform erfolgen. Die Dosierung kann je nach Art der
Indikation, der Schwere der Erkrankung, dem Alter, Geschlecht,
Körpergewicht und der Sensitivität des zu behandelnden Subjekts variieren.
Vorzugsweise werden Dosierungen von 0,01 bis 30 mg, besonders
bevorzugt von 0,1 bis 3 mg und am meisten bevorzugt von 0,1 bis 1 mg
pro kg Körpergewicht und Tag verabreicht. Die Verabreichung kann in einer
einzigen Dosis oder mehreren getrennten Dosierungen erfolgen.
Im Folgenden ist eine Reihe besonders bevorzugter Verbindungen (1)
aufgelistet:
Weiterhin soll die Erfindung durch die folgenden Beispiele erläutert werden.
78 mg 6-(4-Acetamidophenoxy)-5-(chlormethyl)-1-(2',6'-difluorbenzyl)-
1,4-dihydro-4-oxo-chinolin-3-carbonsäure-etylester, gelöst in 3 ml
Dimethylformamid, wurden mit 84 µl N-Methylbenzylamin und 84 µl N,N-
Diisopropyl-ethylamin bei 0°C versetzt und bei Raumtemperatur 20
Stunden gerührt. Nach Zugabe von 20 ml wässriger
Natriumbicarbonatlösung wurde der Niederschlag abgesaugt, mit Wasser
und danach mit n-Hexan gewaschen und bei Raumtemperatur im Vakuum
getrocknet. Man erhält 70 mg der Titelverbindung.

Das Ausgangsmaterial, 6-(4-Acetamidophenoxy)-5-(chlormethyl)-1-(2',6
difluorbenzyl)-1,4-dihydro-4-oxo-chinolin-3-carbonsäure-etylester, wurde
auf folgendem Weg erhalten:
50 g 4-Fluor-3-nitroanilin und 69 g Ethoxymethylenmalonsäure-diethylester
wurden 3 Stunden auf 120°C erwärmt. Das Gemisch wurde auf n-Hexan
gegeben, 2 Stunden gerührt und das kristalline Material abgesaugt und im
Vakuum bei Raumtemperatur getrocknet. Man erhält 93 g N-(4-Fluor-3-
nitrophenyl)-aminomethylenmalonsäure-diethylester. In 3 Portionen zu 31 g
wird dieser zu jeweils 150 ml eines auf 260°C vorgeheizten Gemisches
aus 26,5% Diphenyl und 73,5% Diphenylether (DOWTHERM A®) gegeben
und 30 Minuten bei dieser Temperatur gerührt. Nach Abkühlen wird mit
500 ml n-Hexan verdünnt und der Niederschlag abgesaugt. Man erhält
insgesamt 64 g eines Gemisches von 1,4-Dihydro-6-fluor-5-nitro-4-oxo-
chinolin-3-carbonsäure-ethylester und von 1,4-Dihydro-6-fluor-7-nitro-4-
oxo-chinolin-3-carbonsäure-ethylester.

22 g des vorstehend beschriebenen Gemisches werden in 500 ml
Dimethylformamid mit 16 g Kaliumcarbonat und 23,7 g 2,6-
Difluorbenzylbromid 5 Stunden bei Raumtemperatur gerührt. Das
Reaktionsgemisch wird auf 1 l wässerige Ammoniumchloridlösung gegeben
und mit Ethylacetat dreimal extrahiert. Nach Trocknen mit Natriumsulfat
und Eindampfen im Vakuum werden 500 ml n-Hexan zugegeben und es
wird 15 Minuten gerührt. Nach Dekantieren der Hexanphase wird der
Rückstand aus Ethylacetat umkristallisiert. Man erhält 10,4 g der
Titelverbindung.

3,74 g 4-Acetamidophenol in 40 ml Dimethylformamid werden mit 733 mg
Natriumhydrid (80% in Mineralöl) versetzt und 15 Minuten bei
Raumtemperatur gerührt. Dann wird diese Lösung zu 5 g der vorstehend
beschriebenen Verbindung, gelöst in 40 ml Dimethylformamid, gegeben.
Nach 5 Stunden bei Raumtemperatur wird das Gemisch auf Eiswasser
gegeben und der Niederschlag abgesaugt. Nach Chromatographie an
Kieselgel (Eluent Dichlormethan/2-Propanol 95 : 5) erhält man 5,17 g der
Titelverbindung.

5,1 g der vorgenannten Verbindung werden in 650 ml Methanol mit 510 mg

2 g der vorgenannten Verbindung werden im Gemisch von 24 ml
konzentrierter Schwefelsäure und 12 ml Wasser gelöst und bei 0°C mit
325 mg Natriumnitrit in 4 ml Wasser versetzt. Nach 15 Minuten wird mit
Natriumbicarbonatlösung ein pH von 3 eingestellt und 100 mg Harnstoff
zugegeben. Dann werden 723 mg Kaliumjodid in 0,5 ml Wasser zugesetzt
und eine Stunde bei Raumtemperatur gerührt. Nach Extraktion mit
Dichlormethan/Methanol (95 : 5, v/v) wird die organische Phase mit
wässriger Natriumthiosulfatlösung gewaschen, getrocknet und
eingedampft. Nach Chromatographie an Kieselgel (Eluent Dichlormethan
mit 0-15% Isopropanol) erhält man 697 mg der Titelverbindung
zugegeben. Dann werden 723 mg Kaliumjodid in 0,5 ml Wasser zugesetzt
und eine Stunde bei Raumtemperatur gerührt. Nach Extraktion mit
Dichlormethan/Methanol (95 : 5, v/v) wird die organische Phase mit
wässriger Natriumthiosulfatlösung gewaschen, getrocknet und
eingedampft. Nach Chromatographie an Kieselgel (Eluent Dichlormethan
mit 0-15% Isopropanol) erhält man 697 mg der Titelverbindung.

450 mg der oben genannten Styrylverbindung werden in 25 ml
Tetrahydrofuran und 7 ml Wasser gelöst und mit 0,11 ml einer 2,5
prozentigen Lösung von Osmiumtetroxid in tert.Butanol versetzt. Nach 1,5
Minuten Rühren bei Raumtemperatur werden 482 mg, nach 20 und 22
Stunden 100 mg und nach 24 Stunden weitere 100 mg Natriumperjodat
zugesetzt. Nach 26 Stunden wird mit Wasser verdünnt und mit Ethylacetat
extrahiert. Nach Trocknen der organischen Phase mit Natriumsulfat wird
eingedampft. Man erhält 352 mg der Titelverbindung als Schaum.

300 mg des oben genannten Aldehyds werden in 13,8 ml Essigsäure
gelöst und im Abstand von 10 Minuten mit 3 Portionen von je 10 mg
Natriumborhydrid versetzt. Nach Verdünnen mit Wasser wird mit
Ethylacetat extrahiert und die organische Phase mit

80 mg des vorstehend genannten Alkohols werden in 2 ml Dichlormethan
gelöst und mit 0,1 ml Thionylchlorid versetzt. Nach 20 Minuten bei
Raumtemperatur wird Wasser zugegeben, mit Dichlormethan extrahiert und
die organische Phase mit Kochsalzlösung gewaschen. Nach Trocknen mit
Natriumsulfat wird eingedampft. Man erhält 80 mg der Titelverbindung als
Schaum.

Diese Verbindung entsteht aus 6-(4-Acetamidophenoxy)-5-(N-benzyl-N-
methylaminomethyl)-1-(2',6'-difluorbenzyl)-1,4-dihydro-4-oxo-chinolin-3-
carbonsäure-ethylester durch Erhitzen mit Titan-tetraisopropylat in
Isopropanol.
Diese Verbindung entsteht analog Beispiel 1 aus 5-(Chlormethyl)-1-(2',6'-
difluorbenzyl)-1,4-dihydro-6-(4-isobutyramidophenoxy)-4-oxo-chinolin-3-
carbonsäure-etylester und N-Methylbenzylamin als Schaum.
Diese Verbindung wird erhalten, wenn man in Beispiel 1/c. 4-
Isobutyramidophenol anstelle 4-Acetamidophenol verwendet und analog
Beispiel 1/d. bis Beispiel 1/i. weiter umsetzt.
Man erhält die Titelverbindung, indem man 6-(4-Acetamidophenyl)-1-
benzyl-5-(N-benzyl-N-methylaminomethyl)-phthalazin-4-on in Gegenwart
einer Base wie Natriumcarbonat oder Natriumhydroxid mit Acetylchlorid
oder Acetanhydrid umsetzt.
6-(4-Acetamidophenyl)-1-benzyl-5-(N-benzyl-N-methylaminomethyl)-
phthalazin-4-on erhält man auf folgendem Wege:
1-Benzyl-5,6-dijod-phthalazin-4-on (Indian J. Chem. 16B, 1978, 301-304)
werden analog Beisp. 1/f. mit 1 Äquivalent 4-Acetamidophenyl-boronsäure
umgesetzt. Die Titelverbindung erhält man rein durch Chromatographie an
Kieselgel.
130 mg (0,226 mmol) 6-(4-Acetamidophenoxy)-5-(chlormethyl)-1,4-
dihydro-4-oxo-1-(2'-trifluormethylbenzyl)-chinolin-3-carbonsäure-ethylester,
gelöst in 5 ml DMF, werden bei -5°C mit 125 µl N,N-Diisopropyl-ethylamin
und 126 µl (0,97 mmol) N-Benzyl-methylamin versetzt. Nach dem
Erwärmen auf Raumtemperatur läßt man 20 Stunden nachrühren und gibt
anschließend das Reaktionsgemisch zu 50 ml gesättigter
Natriumbicarbonatlösung. Der Feststoff wird abgesaugt, mit Wasser
gewaschen und im Vakuum getrocknet. Die weitere Reinigung erfolgt
durch Chromatographie an Kieselgel mit einem Eluationsmittel aus
90 Teilen Dichlormethan, 10 Teilen Ethanol und 1 Teil konz. Ammoniak
(Rf: 0,38).2
Es werden 45 mg der Titelverbindung als Schaum erhalten.

Das Ausgangsmaterial 6-(4-Acetamidophenoxy)-5-(chlormethyl)-1,4-
dihydro-4-oxo-1-(2'-trifluormethylbenzyl)-chinolin-3-carbonsäure-ethylester
wird analog dem in den Beispielen 1a bis 1i beschriebenen Weg unter
Verwendung von 2'-Trifluormethyl-benzylbromid an Stelle von 2,6-
Difluormethylbenzylbromid hergestellt
35 mg (0.061 mmol) 6-(4-Methylaminocarbonyl-phenoxy)-5-(chlormethyl)-
1-(2',6'-difluorbenzyl)-1,4-dihydro-4-oxo-chinolin-3-carbonsäure-ethylester,
gelöst in 1.3 µl DMF, werden bei 0°C mit 35 µl N,N-Diisopropyl-ethylamin
und 35 µl (0.25 mmol) N-Methylbenzylamin versetzt. Nach dem Erwärmen
auf Raumtemperatur läßt man 20 Stunden nachrühren und gibt dann das
Reaktionsgemisch zu 10 ml gesättigter Natriumbicarbonatlösung. Der
angefallene Feststoff wird abgesaugt, mit Wasser und Hexan gewaschen
und über Phosphorpentoxid im Vakuum getrocknet.
Es werden 27 mg der Titelverbindung als Schaum erhalten.

Das Ausgangsmaterial 6-(4-Methylaminocarbonyl-phenoxy)-5-(chlormethyl)-
1-(2',6'-difluorbenzyl)-1,4-dihydro-4-oxo-chinolin-3-carbonsäure-ethylester
wird analog dem in den Beispielen 1a bis 1i beschriebenen Weg unter
Verwendung von 4-Hydroxy-N-methylbenzoesäureamid an Stelle von 4-
Acetamidophenol hergestellt.
45 mg (0.081 mmol) 6-(4-Acetamidophenoxy)-5-(chlormethyl)-1-(1-
naphthyl-methyl)-1,4-dihydro-4-oxo-chinolin-3-carbonsäure-ethylester,
gelöst in 1.7 ml DMF, werden bei 0°C mit 46 µl N,N-Diisopropyl-ethylamin
und 46 µl (0.33 mmol) N-Methylbenzylamin versetzt. Nach dem Erwärmen
auf Raumtemperatur läßt man 20 Stunden nachrühren und gibt dann das
Reaktionsgemisch zu 15 ml gesättigter Natriumbicarbonatlösung. Der
angefallene Feststoff wird abgesaugt, mit Wasser und Hexan gewaschen
und über Phosphorpentoxid im Vakuum getrocknet.
Es werden 32 mg der Titelverbindung als Schaum erhalten.

Das Ausgangsmaterial 6-(4-Acetamidophenoxy)-5-(chlormethyl)-1-(1-
naphthyl-methyl)-1,4-dihydro-4-oxo-chinolin-3-carbonsäure-ethylester
wird analog dem in den Beispielen 1a bis 1i beschriebenen Weg unter
Verwendung von 1-Chlormethylnaphthalin an Stelle von 2,6-
Difluormethylbenzylbromid hergestellt.
Buserelin wurde von Welding (Frankfurt/Main, Deutschland) bezogen. Die
Verbindung wurde mit 125I durch Verwendung der Chloramin T-Methode
und Na125I (4000 Ci/mmol; Amersham-Buchler, Braunschweig,
Deutschland) markiert. Die markierte Substanz wurde durch Reverse Phase
HPLC auf einer Spherisorb ODS II-Säule (250 × 4 mm, Teilchengröße 3 µm)
durch Elution mit 50% Acetonitril/0,15% Trifluoressigsäure bei einer
Fließrate von 0,5 ml/min aufgereinigt. Die spezifische Aktivität war 2000
Ci/mmol.
Alle anderen Chemikalien wurden aus kommerziellen Quelle im höchsten
verfügbaren Reinheitsgrad bezogen.
Alpha T3-1-Zellen (Bilezikjian et. al., Mol. Endocinol 5 (1991), 347-355)
wurden in DMEM-Medium (Gibco-BRL, Eggenstein-Leopoldshafen,
Deutschland) mit Penicillin (100 I.U./ml), Streptomycin (0,1 mg/ml) und
Glutamin (0,01 mol/l) und 10% fötalem Kälberserum (FCS; PAA
Laboratories, Coelbe, Deutschland) auf Plastikgewebekulturplatten (Nung,
245 × 245 × 20 mm) kultiviert. CHO-3-Zellen (Schmid et. al., J. Biol.
Chem. 275 (2000), 9193-9200) wurden unter identischen Bedingungen
kultiviert, abgesehen davon, dass Ham's F12-Medium (Gibco-BRL)
verwendet wurde.
10 konfluente Zellkulturplatten wurden zweimal mit 50 ml Phosphat
gepufferter Salzlösung (PBS) gespült. Die Zellen wurden durch Abschaben
mit einem Gummischaber in 5 ml PBS geerntet und durch Zentrifugation bei
800 Upm für 10 min in einer Laborzentrifuge (Heraeus) sedimentiert. Das
Zellpellet wurde in 5 ml 0,25 mol/l Saccharose/0,01 mol/l Triethanolamin,
pH 7,4 resuspendiert. Die Zellen wurden durch drei Zyklen Einfrieren in
Trockeneis/Ethanolbad und Auftauen bei Raumtemperatur lysiert. Das Lysat
wurde bei 900 Upm für 10 min zentrifugiert und das Sediment verworfen.
Der Überstand wurde bei 18.000 Upm in einem Sorvall SS34 Rotor für 30 min
zentrifugiert. Das Pellet (Zellmembranen) wurde durch Pottern in 5 ml
Assaypuffer (0,25 mol/l Saccharose, 0,01 mol/l Triethanolamin, pH 7,5, 1 mg/ml
Ovalbumin) suspendiert und in 200 µl Aliquots bei -20°C
aufbewahrt. Die Proteinbestimmung erfolgte nach der Methode von
Bradford (Anal. Biochem. 72 (1976), 248-254).
Bindungsuntersuchungen für Kompetitionskurven wurden als Triplikate
durchgeführt. Eine Testprobe enthielt 60 µl Zellmembransuspension (10 µg
Protein für αT3-1-Zellen oder 40 µg Protein für CHO3-Zellen), 20 µl 125I
markiertes Buserelin (100.000 lpm pro Probe für Kompetitionskurven und
zwischen 1.500 und 200.000 lpm für Sättigungsexperimente) und 20 µl
Testpuffer oder Testverbindungslösung. Die Testverbindungen wurden in
destilliertem Wasser oder 50% Ethanol gelöst. Serielle Verdünnungen (5 ×
10-6 mol/l bis 5 × 10-12 mol/l) wurden in Testpuffer hergestellt. Die
unspezifische Bindung wurde in Gegenwart eines Überschusses an
unmarkiertem Buserelin (10-6 mol/l) bestimmt. Die Testproben wurden für
30 min bei Raumtemperatur inkubiert. Gebundener und freier Ligand
wurden durch Filtration (Whatman GF/C-Filter 2,5 cm Durchmesser) unter
Verwendung einer Amicon 10× Sammelvorrichtung getrennt und zweimal
mit 5 ml 0,02 mol/l Tris/HCl, pH 7,4 gewaschen. Die Filter wurden mit 0,3%
Polyethylenimin (Serva; Heidelberg, Deutschland) für 30 min befeuchtet,
um die unspezifische Bindung zu verringern. Die durch die Filter
zurückgehaltene Radioaktivität wurde in einem 5 Kanal-Gamma-Zähler
(Wallac-LKB 1470 Wizard) bestimmt.

oder - c) die Gruppe -A-NR14-CO-NR15R16 ist, worin A eine ggf. durch eine C1-C6-Alkylgruppe substituierte Alkylengruppe mit 1-4 C-Atomen, insbesondere mit 1 C-Atom, eine Carbonylgruppe, ein Sauerstoffatom oder die Gruppe -SOx- mit X = 0, 1 oder 2 ist, R14 und R15 jeweils unabhängig ein Wasserstoffatom oder eine gerade oder verzweigte Alkylkette mit 1-10 C-Atomen sind und R16 eine gerade oder verzweigte Alkylkette mit 1-10 C-Atomen, eine Cycloalkylgruppe mit 3- 10 C-Atomen, eine Cycloalkylalkylgruppe mit 7-20 C-Atomen, eine Aralkylgruppe mit 7-20 C-Atomen, wobei der Arylrest ggf. durch Alkylgruppen oder Halogenatome substituiert sein kann, eine ggf. durch Alkylgruppen oder Halogenatome substituierte Phenylgruppe oder ein ggf. durch Alkylgruppen oder Halogenatome substituierter heterocyclischer Ring ist,
R3 und R4 jeweils unabhängig ein Wasserstoffatom oder eine Alkylgruppe mit 1-10 C-Atomen sind und R3 auch ein Halogenamtom sein kann,
R5 eine über den Rest Z verknüpfte Gruppe ist,
worin G-C=C-, -C=N-, -N=C-, ein Sauerstoff- oder Schwefelatom ist, Z eine Direktbindung, ein Sauerstoff- oder ein Schwefelatom, die Gruppe CH-R52 oder -CHR52-CH-R53-,
wobei R52 und R53 unabhängig von einander die Bedeutung eines Wasserstoffatoms oder einer Alkylgruppe und n der Zahlen 1 und 2 haben, eine -C∼C-Dreifachbindung oder eine E- oder Z-konfigurierte Gruppe -CR52=CR53- oder C=CR52R53 ist, wobei R52 und R53 unabhängig von einander die Bedeutung eines Wasserstoffatoms oder einer Alkylgruppe haben, L eine CH2- oder eine NH-Gruppe ist, Q eine Carbonyl- oder -SOx-Gruppe mit X = 0, 1 oder 2 ist und R51 eine ggf. durch eine Alkylgruppe substituierte Amino- oder eine gerade oder verzweigte, ggf. durch Halogenatome, Hydroxyl- oder Alkoxygruppen substituierte Alkylgruppe oder eine ggf. durch Halogenatome, Hydroxyl- oder Alkoxygruppen substituierte Cycloalkylgruppe mit 3-7 Ringgliedern ist,
R6 die Gruppe CH2-N(R61)R62 ist, wobei R61 jeweils unabhängig ein Wasserstoffatom oder eine Alkylgruppe und R62 eine Alkylgruppe oder eine ggf. substituierte Aralkylgruppe oder Heteroarylalkylgruppe mit 7-20 C-Atomen sind und
in jeder Orientierung bedeuten kann, ferner alle Stereoisomeren der genannten Strukturen sowie Salze derselben mit physiologisch verträglichen Säuren oder Basen.

ausgewählt ist.
5. Verbindungen nach Anspruch 1 oder 2,
dadurch gekennzeichnet,
dass R1 die Gruppe -CO-OR12 ist.
6. Verbindungen nach Anspruch 5,
dadurch gekennzeichnet,
dass R12 aus Methyl, Ethyl oder i-Propyl ausgewählt ist.
7. Verbindungen nach einem der Ansprüche 1 bis 6,
dadurch gekennzeichnet,
dass R2 eine 2',5'-Difluorbenzylgruppe ist.
8. Verbindungen nach einem der Ansprüche 1 bis 7,
dadurch gekennzeichnet,
dass R3 und R4 Wasserstoffatome sind.
9. Verbindungen nach einem der Ansprüche 1 bis 8,
dadurch gekennzeichnet,
dass Z eine direkte Bindung oder ein Sauerstoffatom ist.
10. Verbindungen nach einem der Ansprüche 1 bis 9,
dadurch gekennzeichnet,
dass G -C=C- bedeutet.
11. Verbindungen nach einem der Ansprüche 1 bis 10,
dadurch gekennzeichnet,
dass L eine NH-Gruppe ist.
12. Verbindungen nach einem der Ansprüche 1 bis 11,
dadurch gekennzeichnet,
dass Q eine Carbonylgruppe ist und R51 eine C1-C6-Alkylgruppe ist.
13. Verbindungen nach einem der Ansprüche 1 bis 12,
dadurch gekennzeichnet,
dass R61 ein Wasserstoffatom oder eine Methylgruppe ist oder/und
R62 eine Benzylgruppe ist.
14. Verwendung von Verbindungen nach einem der Ansprüche 1 bis 13
als Antagonisten des Gonadotropin-freisetzenden Hormons (GnRH).
15. Verwendung nach Anspruch 14 zur männlichen Fertilitätskontrolle,
zur Hormontherapie, zur Behandlung weiblicher Sub- und Infertilität,
zur weiblichen Empfängnisverhütung und zur Tumorbekämpfung.
16. Verfahren zur Herstellung von Verbindungen der allgemeinen Formel
(1) durch
- a) Umsetzung einer Verbindung der allgemeinen Formel (2)
wobei R7 eine Abgangsgruppe bedeutet und alle anderen Reste die bei Verbindung (1) angegebene Bedeutung haben, mit einer Verbindung der allgemeinen Formel (3)
R8-N(R61)R62 (3)
wobei R8 ein Wasserstoffatom oder ein Metallatom bedeutet und R61 und R62 die bei Verbindung (1) angegebenen Bedeutungen haben, - b) Umsetzung einer Verbindung der allgemeinen Formel (4)
worin R9 die Gruppe -OSO2CnF2n+1, ein Halogenatom, besonders ein Brom- oder Jodatom oder eine andere Abgangsgruppe ist und alle anderen Reste die bei Verbindung (1) angegebene Bedeutung haben, mit einer Verbindung der allgemeinen Formel (5)
wobei R10 eine ein Metall oder ein Nichtmetall enthaltende Gruppe, eine ggf. in ein Metallsalz überführte Hydroxy- oder Mercapto-Gruppe, die Gruppe -C∼C-R31 oder eine E- oder Z- konfigurierte Gruppe -CR52=CR53R31 oder -CR31=CR52R53 ist, worin R31 eine ein Metall oder ein Nichtmetall enthaltende Gruppe ist und alle anderen Reste die bei Verbindung (1) angegebene Bedeutung haben, mit oder ohne Mitwirkung eines Katalysators; - c) wenn Y in Verbindung (1) ein Stickstoffatom ist, durch
Umsetzung einer Verbindung der allgemeinen Formel (6)
wobei R32 ein Wasserstoffatom oder ein Metallatom bedeutet und alle anderen Reste die bei Verbindung (1) angegebene Bedeutung haben, mit einer Verbindung der allgemeinen Formel (7)
R33-R2 (7)
wobei R33 eine Abgangsgruppe bedeutet und R2 die bei Verbindung (1) angegebene Bedeutung hat, oder - d) wenn W in Verbindung (1) ein Stickstoffatom ist, durch
Umsetzung einer Verbindung der allgemeinen Formel (8)
wobei R32 ein Wasserstoffatom oder ein Metallatom bedeutet und alle anderen Reste die bei Verbindung (1) angegebene Bedeutung haben, mit einer Verbindung der allgemeinen Formel (9)
R33-R1 (9)
wobei R33 eine Abgangsgruppe bedeutet und R1 die bei Verbindung (1) angegebene Bedeutung hat.
