Abstract
Die Erfindung betrifft Verfahren zur Herstellung von gewissen Enol-Ethern und Estern der Formel, worin X und Y je Wasserstoff, Fluor oder Chlor; R1 2-Thienyl oder Benzyl und R Alkanoyl, Cycloalkylcarbonyl, Phenylalkanoyl, Chlorbenzoyl, Methoxybenzoyl, Thenoyl, v-Alkoxycarbonylalkanoyl, Alkoxycarbonyl, Phenoxycarbonyl, 1-(Acyloxy)alkyl, 1-(Alkoxycarbonyloxy)alkyl, Alkyl, Alkylsulfonyl, Methylphenylsulfonyl oder Dialkylphosphonat bedeuten, die nuetzlich als Arzneimittelvorstufen der bekannten entzuendungshemmenden und analgetischen Mittel 3-Acyl-2-oxindol-1-carboxamide sind. Description
Die Erfindung betrifft entzündungshemmende (antiinflammatorischti) Mittel und insbesondere Enolester- und -ether-Arzneimittelvorstufen von 3-Acyl-2-oxindol-l-carboxamiden, einer Klasse von bekannten nicht-steroiden entzündungshemmenden Mitteln.
Ober die Verwendung von Oxindolen als entzündungshemmende Mittel wurde zuerst in U.S. 3 634 453 berichtet, die aus l-substituierten-2-oxindol-3-carboxamiden bestanden. Kürzlich wurde eine Reihe von .3-Acyl-2-oxindol-l-carboxamiden in U.S. 4 556 672 als Hemmer der Enzyme Cyclooxygenase (CO) und Lipoxygenase (LO) und als analgetisch und entzündungshemmende nützliche Mittel bei Säugern offenbart.

Besonders bevorzugt sind Verbindungen der Formel (I), worin R1 2-Thienyl, X Chlor, Y Wasserstoff und R Alkanoyl mit zwei bis zehn Kohlenstoffatomen sind. Bevorzugt innerhalb dieser Gruppe sind Verbindungen, worin R Acetyl, Propionyl und i-Butyryl ist.
Fine zweite bevorzugte Gruppe von Verbindungen der Formel (I) sind solche, worin R1 2-Thienyl, X Chlor, Y Wasserstoff und R Phenylalkanoyl mit sieben bis zehn Kohlenstoffatomen sind. Besonders bevorzugt innerhalb dieser Gruppe ist die Verbindung, worin R Phenylacetyl ist.
Eine dritte bevorzugte Gruppe von Verbindungen sind solche der Formel (I), worin R1 2-Thi;»nyI, X Chlor, Y Wasserstoff und R lü-Alkoxycarbonylalkanoyl sind, wobei dieses Alkoxy ein bis drei Kohlenstoffatome und dieses Alkanoyl drei bis fünf Kohlenstoffatome aufweisen. Besonders bevorzugt

innerhalb dieser Gruppe ist die Verbindung, worin R ω-Ethoxycarbonylpropionyl ist.
Eine vierte Gruppe von bevorzugten Verbindungen der Formel (I) sind solche, worin R1 2-Thienyl, X Chlor, Y Wasserstoff und R Alkoxycarbonyl mit zwei bis zehn Kohlenstoffatomen sind. Besonders bevorzugt innerhalb dieser Gruppe sind Verbindungen, worin R Methoxycarbonyl, Ethoxycarbonyl und n-Hexoxycarbonyl ist.
Eine fünfte Gruppe von bevorzugten Verbindungen der Formel (I) sind 3olche, worin R1 2-Thienyl, X Chlor, Y Wasserstoff und R 1-(Alkoxycarbonyloxy)alkyl sind, wobei dieses Alkoxy zwei bis fünf Kohlenstoffatome und dieses Alkyl ein bis vier Kohlenstoffatome aufweisen. Besonders bevorzugt innerhalb dieser Gruppe ist die Verbindung, worin R 1-(Ethoxycarbonyloxy)ethyl ist.
Eine sechste Gruppe von bevorzugten Verbindungen der Formel (I) sind solche, worin R1 2-Thienyl, X Chlor, Y Wasserstoff und R Alkylsulfonyl mit ein bis drei Kohlenstoffatomen sind. Besonders bevorzugt innerhalb dieser Gruppe ist die Verbindung, worin R Methylsulfonyl ist.
Eine siebente Gruppe von bevorzugten Verbindungen der Formel (I) sind solche, worin R1 2-Thienyl, X Chlor, Y Chlor und R Alkanoyl mit zwei bis zehn Kohlenstoffatomen sind. Besonders bevorzugt innerhalb dieser Gruppe sind Verbindungen, worin R Acetyl, Propionyl und i-Butyryl ist.
Eine achte Gruppe von bevorzugten Verbindungen der Formel (I) sind solche, worin R1 2-Thienyl, X Fluor, Y Chlor und R Alkoxycarbonyl mit zwei bis zehn Kohlenstoffatomen sind. Besonders bevorzugt innerhalb dieser Gruppe sind Verbindungen, worin R Methoxycarbonyl, Ethoxycarbonyl und n-Hexoxycarbonyl ist.
Eine neunte Gruppe von bevorzugten Verbindungen der Formel (I) sind solche, worin ?.* Benzyl, X Wasserstoff, Y Fluor und R Alkanoyl mit zwei bis zehn Kohlenstoffatomen sind. Besonders bevorzugt innerhalb dieser Gruppe ist die Verbindung, worin R Acetyl ist.
Eine zehnte Gruppe von bevorzugten Verbindungen der Formel (I) sind solche, worin R1 Benzyl, X Wasserstoff, Y Fluor und R Alkoxycarbonyl mit,zwei bis zehn Kohlenstoffatomen sind. Besonders bevorzugt innerhalb dieser Gruppe ist die Verbindung, worin R Methoxycarbonyl ist.
Die vorliegende Erfindung umfaßt auch eine Methode zur Behandlung von Entzündungen in Säugern, gekennzeichnet durch die Verabreichung einer entzündungshemmend wirksamen Menge einer Verbindung, ausgewählt aus solchen der Formel (I), an diese Sauger.
Die Enolether und -ester der vorliegenden Erfindung sind nicht Enolsäuren wie die Stammverbindungen und zeilen im Vergleich zu diesen Stammverbindungen verminderte gastrische Reizungen.
Der Ausdruck "Arzneimittelvorstufe" (Pr-x.rug) bezieht sich auf Verbindungen, die Arzneimittelvorläufer sind, d:;.e nach Verabreichung und Absorption das Arzneimittel in vivo durch irgendeinen metabolischen Prozeß abgeben.
Obwohl alle üblichen Verabreichungsarten bei den Verbindungen der Erfindung nützlich sind, ist die bevorzugte Verabreichungsart der orale Weg. Nach gastrointestinaler Absorption werden die vorliegenden Verbindungen in vivo zu den entsprechenden Verbindungen der Formel (I), worin R Wasserstoff ist, oder einem Salz davon hydrolysiert. Da die Arzneimittelvorstufen der Erfindung nicht Enolsäuren sind, wird daher die Beeinträchtigung des gastrointestinalen Bereichs durch die saure Stammverbindung auf ein Minimum reduziert. Da

gastrointestinale Schwierigkeiten als die hauptsächlichste ungünstige Reaktion von nicht-steroiden entzündungshemmenden Arzneimittel festgestellt wurde [siehe z.B. DelFavero in "Side Effects of Drugs Annual 7", Dukes und Elis, Eds. Exerpta Madica, Amsterdam, 1983, S. 104-115], besitzen die Verbindungen (I) der Erfindung einen deutlichen Vorteil gegenüber den enolischen Stammverbindungen.
Bei der Umwandlung der 3-Acyl-2-oxindol-l-carboxamide in Verbindungen der Formel (I) können die Substituenten an der exocyclischen Doppelbindung in 3-Stellung syn, anti oder eine Mischung von beiden sein. Die Verbindungen der Strukturen

oder Mischungen davon werden dfo.ialb als
dargestellt. Alle Formen dieser Isomeren werden als Teil der vorliegenden Erfindung betrachtet.
Es werden zwei Methoden verwendet, um die Verbindungen der vorliegenden Erfindung zu synthetisieren; die erste Methode umfaßt die Behandlung einer Lösung des geeigneten 3-Acyl-2-oxindol-l-carboxamids und einer äquimolaren Menge Triethylamin in einem reaktionsinerten Lösungsmittel, wie Chloroform, bei O0C mit einer äquimolaren Menge plus einem geringen Oberschuß des erforderlichen Säurechlorids, Chlor-

formiats, Oxoniumsalzes oder Alkylierungsmittels. Man läßt die Reaktionsmischung sich auf Raumtemperatur erwärmen und 2-3 Stunden stehen. Falls das Oxindol-Ausgangsmaterial sich nicht vollständig umgesetzt hat, wird die Mischung auf O0C gekühlt und zusätzliches Acylierungs- oder Alkylierungsmittel zugesetzt und das Verfahren wiederholt, bis das gesamte Oxindol-Ausgangsmaterial verbraucht ist.
Das Produkt wird aus dem Reaktionslösungsmittel isoliert, nachdem es mit IN Salzsäure gewaschen wurde, gefolgt von Extraktion mit einer gesättigten Natriumbicarbonatlösung. Das nach Entfernung des Lösungsmittels im Vakuum zurückbleibende Produkt wird durch Umkristallisieren oder Chromatographie gereinigt.
Das zweite zur Herstellung von Verbindungen der vorliegenden Erfindung nützliche Verfahren besteht darin, daß man in einem wasserfreien reaktionsinerten Lösungsmittel, wie Aceton, das geeignete 3-Acyl-2-oxindol-l-carboxamid, einen dreifachen molaren Oberschuß des erforderlichen a-Chloralkylcarbonats, einen fünffachen molaren Oberschuß Natriumjodid und einen zweifachen molaren Überschuß wasserfreies Kaliumcarbonat zusammenbringt und diese Reaktionsmischung Stunden unter Rückfluß erhitzt.
Die Reaktionsmischung wird mit Wasser verdünnt und das Produkt mit einem wasserunmischbaren Lösungsmittel, wie Diethylether oder Chloroform, extrahiert. Einengen des das Produkt enthaltenden Lösungsmittels liefert das rohe Material, das durch Umkristallisieren und/oder Chromatographie gereinigt werden kann.
Die als Ausgangsmaterialien benötigten 3-Acyl-2-oxindol-1-carboxamide sind durch an sich bekannte Verfahren erhältlich, siehe zum Beispiel die oben zu diesen Verbindungen angeführte Referenz. Die anderen oben angegebenen Ausgangsre-

agentien sind im Handel erhältlich oder können durch an sich bekannte Methoden hergestellt werden.
Die Arzneimittelvorstufen der Formel (I) werden durch bekannte Methoden auf ihre entzündungshemmende und analgetische Wirksamkeit geprüft, wie dem Fußödem-Test bei Ratten, dem Adjuvans-induzierten Arthritis-Test bei Ratten oder dem Phenylbenzochinon-induzierten "Writhing"-Test bei Mäusen, wie sie vorgängig bei der Evaluierung der Stammverbindungen verwendet und in den oben zitierten Referenzen und auch anderswo in der Literatur beschrieben wurden; siehe z.B. C. A. Winter, in "Progress in Drug Research" herausgegeben von E. Jucker, Birkhauser Verlag, Basel, Bd. 10, 1966, S. 139-192.
Im Vergleich mit S-Acyl^-oxindol-l-carboxamid-Stammverbindungen wurde bei den neuen Arzneimittelvorstufen der Formel (I) festgestellt, daß sie in Tests, die durch eine Modifikation der Methode von T. J. Carty et al., Prostaglandins, 19., 51-59 (1980) durchgeführt wurden, eine verminderte Fähigkeit zur Hemmung der Prostaglandin-Synthese aus Arachidonsäure besitzen. Im modifizierten Verfahren werden Kulturen von basophilen Leukämiezellen von Ratten (RBL-I), hergestellt durch die Methode von Jakschik et al., ibid., 16, 733 (1978), an Stelle von Fibroblasten von iusen (MC5-5) und Synovial- Zellkuituren von Kaninchen verwendet.
Die Verbindungen der Erfindung sind also selbst relativ unwirksam als entzündungshemmende Mittel, ergeben aber eine entzündungshemmend wirksame Verbindung nach Hydrolyse in vivo. Da die Verbindungen (I) nicht Enolsäuren sind und es bekannt ist, daß die Hydrolyse stattfindet, nachdem die Arzneimittelvorstufe den Magen verläßt, vermindern sie in signifikanter Weise die durch orale Verabreichung der enolischen Stammverbindungen verursachten gastrischen Reizungen.
Auf molarer Basis werden die vorliegenden Arzneimittelvorstufen im allgemeinen in der gleichen Menge und Häufigkeit

verabreicht wie die bekannten S-Acyl-^-oxindol-l-carboxamide, von denen sie abgeleitet sind. Die nicht-enolische Natur der vorliegenden Verbindungen gestattet aber im allgemeinen eine höhere tolerierte Dosis, falls eine solche höhere Dosis zur Bekämpfung von Schmerz und Entzündung erforoerlich ist.
Die vorliegenden Arzneii ".ttelvorstuf en werden auch in der gleichen Weise formuliert und auf den gleichen Wegen verabreicht wie die bekannten Stammverbindungen, wie es in der oben angegebenen Referenz beschrieben wurde. Der bevorzugte Verabreichungsweg ist der orale, wodurch aus der nicht-enolischen Natur der vorliegenden Verbindungen besonderer Vorteil gezogen wird.
Die vorliegende Erfindung wird durch die folgenden Beispiele veranschaulicht, wird aber durch die spezifischen Details dieser Beispiele nicht beschränkt.
Allgemeine Verfahren Methode A
Zu einer Aufschlämmung von 3-Acyl-2-oxindol-l-carboxamid in Chloroform wird eine äqui^olare Menge Triethylamin zugesetzt. Die erhaltene Lösung wird auf O0C gekühlt und ein geringer Oberschuß des geeigneten Säurechlorids, Chlorformiats, Oxoniumsalzes oder Alkylierungsmittels zugesetzt. Nach Rühren bei O0C während 2 Stunden und dann bei Raumtemperatur während 2 Stunden wird dann die Mischung, falls das 3-Acyloxindol-lcarboxamid nicht vollständig verbraucht ist, abermals auf O0C gekühlt und zusätzliches Säurechlorid, Chlorformiat oder Oxoniumsalz zugesetzt und die Mischung 2 Stunden bei O0C und dann 2 Stunden bei Raumtemperatur gerührt. Dieses Verfahren kann wiederholt werden, um eine vollständige Umsetzung des 3-Acyloxindol-1-carboxamidssicherzustellen. Nach vollständiger Umsetzung wird die Mischung filtriert und das Filtrat mit IN

Salzsäure (2X) und gesättigter tyatriumbicarbonatlösung (2X) gewaschen. Die organische Schicht wird über MgSOi getrocknet, filtriert und im Vakuum eingeengt. Das erhaltene Produkt wird durch Umkristallisieren oder Chromatographie gereinigt.
Methode B
Eine Mischung von 3-Acyl-2-oxindol-l-carboxamid, einem dreifachen molaren Oberschuß des geeigneten a-Chloralkyl- oder α-Chlor(aralkyl)carbonate, einem fünffachen molaren Oberschuß Natriumiodid und einem zweifachen molaren Überschuß wasserfreies Kaliumcarbonat (getrocknet 1 Stunde im Hochvakuum bei 1650C) in Aceton (getrocknet über Molekularsieben) wird 16 Stunden unter Rückfluß erhitzt.
Die gekühlte Mischung wird dann mit Wauser verdünnt und mit Ether extrahiert. Die vereinigten Etherextrakte werden über MgSO« getrocknet, filtriert, und das Filtrat wird im Vakuum eingeengt. Das erhaltene rohe Produkt wird durch Chromatographie und/oder Umkristallisation gereinigt.
Indem man dem angegebenen Verfahren folgt und von den erforderlichen Reagentien ausgeht, wurden die angegebenen Arzneimittelvorstufen hergestellt:

Claims (3)

Verfahren zur Herstellung einer Verbindung der Formel (I)(Dworin X und Y je Wasserstoff, Fluor oder Chlor bedeuten; R1 2-Thienyl oder Benzyl ist; und R Alkanoyl mit zwei bis zehn Kohlenstoffatomen, Cycloalkylcarbonyl mit fünf bis sieben Kohlenstoffatomen, Phenylalkanoyl mit sieben bis zehn Kohlenstoffatomen, Chlorbenzo^l, Methoxybenzoyl, Thenoyl, ω-Alkoxycarbonylalkanoyl, wobei dieses Alkoxy ein bis drei Kohlenstoffatome und dieses Alkanoyl drei bis fünf Kohlenstoffatome aufweisen; Alkoxycarbonyl mit zwei bis zehn Kohlenstoffatomen; Phenoxycarbonyl; l-(Acyloxy)alkyl, wobei dieses Acyl zwei bis vier Kohlenstoffatome und dieses Alkyl ein bis vier Kohienstöffatome;Alkylsulfonyl mit ein bis drei Kohlenstoffatomen; Methylphenylsulfonyl oder Dialkylphosphonat, wobei dieses Alkyl ein bis drei Kohlenstoffatome aufweist, darstellt, dadurch gekennzeichnet, daß manA) "bei etwa 25°C eine Verbindung der Formel- 28 -2850 04C-Rmit einer äquimoleren Menge einer Verbindung der FormelR-Clin einem reaktionsinerten Lösungsmittel, das eine äquioolare Menge eines tertiären Amins enthält, umsetzt, bis dieReaktion in wesentlichen vollständig ist oder, daß man B) eine Verbindung der Formel (I)worin X und Y je ausgewählt sind aus der Gruppe, bestehend aus Hasserstoff, Fluor und Chlor; R1 ausgewählt ist aus der Gruppe, bestehend aus 2-Thienyl und Benzyl; und R l-(Alkoxycarbonyloxy)alkyl ist, wobei dieses Alkoxy zwei bis fünf Kohlenstoffatome und dieses Alkyl ein bis vier Kohlenstoff atome aufweisen, dadurch ^er stellt ' da^ man be* 25~ 750C eine Verbindung der Formel ^~^C-RJ5 0 0 4- 29 -rait etwa einem 3-4-fachen molaren Oberschuß einer Verbindung der FormelR-Clin einem wassermischbaren reaktionsinerten Lösungsmittel, das etwa einen fünffachen molaren Oberschuß Natriumiodid und etwa einen zweifachen molaren Oberschuß eines Alkalimetallcarbonate enthält, umsetzt, bis die Reaktion im wesentlichen vollständig ist.
- 2. Verfahren nach Punkt 1, Variante A, dadurch gekennzeichnet, daß das Lösungsmittel Chloroform und das tertiäre Amin Triethylamin sind.
- 3. Verfahren nach Punkt 1, Variante B, dadurch gekennzeichnet, daß das reaktionsinerte Lösungsmittel Aceton und das Alkaliinetallcarbonat Kaliumcarbonat sind.
