Abstract

背景技术
本发明涉及一种用于营养补充的组合物;该组合物的制备方法;该组合物在制备功能食品或药剂方面的用途,所述的功能食品或药剂用于下列人的营养、预防和治疗:从疾病或外科手术中渐渐康复的病人,具有有限食欲的人,例如老年人或食欲减退的患者,或者消化其它蛋白质源的能力减退的人,这样的人例如是慢性胃炎患者,其胃蛋白酶消化能力降低;本发明还涉及一种提供营养或治疗的方法,其包括服用有效量的该组合物。
许多人并不充分摄入营养素来完善营养膳食。为了帮助这些人,营养补充剂得到了发展。营养补充剂通常并不是用来提供用于完全营养膳食的所有必需营养素;相反,它们一般是用来补充膳食,使其具有更完全的营养性。然而,在一些情况下,它们提供了完全营养。
对于营养补充剂有许多对象;例如病人、渐渐康复的病人、食欲不佳的病人和老年人。对于病人和渐渐康复的病人,食品的自然摄取通常低于正常水平,不能充分满足营养所需。体力的康复和恢复能力由此被损害。另一方面,相当数量的老年人吃得太少,不能满足所有的营养需要。这通常是由于降低了能量需要,随之降低了体重,并使身体活动减少。食欲不佳的病人被定义为食欲减退,不能摄入足够的营养素。在所有的这些情况中,补充缺失的营养素可以带来好处。
多种营养补充剂是可用的。通常在北美市场发现的一族补充剂是被Ross Laboratories以商品名ENSURE出售的。主要使用的的蛋白质源是酪蛋白酸盐和大豆分离蛋白。另一族补充剂是以商品名RESOURCE出售的Novartis Nutrition Ltd的产品。在这种情况下,蛋白质源基于酪蛋白酸盐。还有的一族补充剂是以商品名NUBASICS出售的Nestle Clinical Nutrition的产品。一般,该族产品中使用的蛋白质源是酪蛋白酸盐。但是发现这些产品会带来一些问题,这就是这些产品并不必然使消费者摄取充足的营养素;要么是因为没有服用足够量的产品,要么是没有食用足够量的其它食品。特别是对于渐渐康复中的病人,老年患者和其它由于食欲减退而食欲不佳的病人更是如此,这些情况中,导致了营养素摄取不充分。
基于其它蛋白质源例如乳清蛋白质的营养补充剂也是可用的,或者它们已经记载于文献中。一般,基于乳清蛋白质的营养补充剂是以果汁的形式提供的;例如EP专利申请0486425和US 5,641,531中所记载的。但是,这些产品也会带来问题,即它们一般不提供脂源,尽管脂类对于足够的营养是必需的。

发明概述
引人注目的是,解决上述问题并能够使这样的人群保持或恢复体力的组合物已经被发现。
本发明的第一个方面是提供了一种用于营养补充的组合物,其包含(i)蛋白质源,该蛋白质源提供组合物总热量的至少约8%,且其包括至少约50wt.%的乳清蛋白质,该乳清蛋白质包含一部分至少是部分水解的乳清蛋白质;(ii)脂源,具有的ω3∶6脂肪酸比例是约5∶1至约10∶1,且提供组合物总热量的至少约18%;(iii)碳水化合物源,其提供组合物的其余热量和(iv)平衡的大量营养剂构成(profile),其至少包含维生素E和维生素C。

令人意想不到的是,现已发现,根据本发明用于营养补充的组合物因为它含有乳清蛋白质因此易于消化并且很少会导致饱腹感。因此病人不能充分摄取补充剂的问题会减少。同样,病人不能食用足够的其它食品的问题可以被减少。而且,该组合物具有很好平衡的脂类构成,这些脂类构成提供易于利用能源。
优选该组合物的实施方案包括作为主要蛋白质(氨基酸)源的乳清蛋白质。该乳清蛋白质可以是甜乳清或酸乳清或其组合。优选是通过酶处理、优选用胰蛋白酶、Alcalase或Novozyme处理乳清蛋白质产生的乳清蛋白质部分水解产物的形式,且因此很少粘性、较轻,并提供了比公知的用于强化饮料和营养补充的组合物更易于消化的优点。
应该理解本发明并不仅仅限于乳清蛋白质水解产物。例如,可以使用非水解乳清蛋白质,优选当组合物制成粉状时。
乳清蛋白质用一种或多种上述酶的部分水解可以在pH范围是约6.6至8.8(优选约8.5)和温度是约40至约70℃(优选约65℃),酶浓度是蛋白质的约0.5至约2.5(优选约1.0)%条件下进行。优选进行酶处理约5至约120分钟(优选15分钟),以实现充分的水解。
优选乳清蛋白质水解产物占配方中蛋白质含量的至少50%。优选是单独的蛋白质源,但可以与完整的乳清蛋白质或其它蛋白质或肽源结合,所述的肽包括在乳清或牛奶中天然发现的肽,例如酪蛋白甘大肽(caseino glycomacropeptide)(CGMP)。令人意想不到的是,发现尽管部分水解蛋白质在组合物中具有高含量,但在物理上却很稳定,并具有非常满意的口味,这是由于用于制备水解产物的方法和风味体系的选择,提供了可接受的感官特性。

显然,由于乳清蛋白质的性质和其能够易于消化的事实,该组合物在对需要营养补充的人们具有有益的效果,例如需要营养补充的人是从疾病或手术恢复的那些人,具有有限食欲的那些人例如老年人,或消化其它蛋白质源有问题的人,例如患有慢性胃炎的人,公知这些人胃蛋白酶的消化能力降低。显然,该组合物能够使这样的人保持或很快恢复其体力,并由此有助于渐渐康复病人的恢复。
优选地,脂源包含约40%至约65wt.%的单不饱和脂肪酸;和约15%至约30wt.%的多不饱和脂肪酸。饱和脂肪酸含量优选小于约30wt.%。在该脂肪混合物中至多可混入20wt.%的中链甘油三酸酯,以便于消化。就每100g的脂源而言,脂源可以含有至少约30mg的维生素E。
优选该脂源提供组合物总热量的约25%至约35%,更优选约30%。
优选碳水化合物源包括蔗糖、玉米糖浆、麦芽糖糊精或其组合。优选,碳水化合物源提供该组合物总能量的约50%至60%。

优选该组合物的实施方案是粉状的形式,以用水稀释后使用或准备以液体形式用于强化饮料;或是具有奶油蛋糕或果馅饼样结构的布丁形式,以供吞咽困难或具有其它的吞咽问题的那些人的食用;或是棒状形式,以提供各种不同的令人感兴趣的选择。
优选,该组合物的实施方案被配制成用于人的食用和/或服用。优选,另一种实施方案是配制成用于伴生(companion)动物食用。
优选地,本发明的组合物的实施方案包括添加至少一种益生微生物。该益生微生物有助于使抗生素治疗之后的肠内菌群恢复自然平衡。
更优选地,粉状形式的组合物的实施方案含有乳酸菌和/或其发酵代谢产物。优选地,乳酸菌选自詹生氏乳杆菌、副双歧乳杆菌或其组合。该产品有助于抑制胃中幽门螺杆菌的生长,该幽门螺杆菌与特别是患有胃炎人的溃疡的发展有联系。最优选地,益生菌包括以NCC 2461保藏号保藏的副双歧乳杆菌株。
本发明的优点是提供的组合物可以是功能食品,且因此不需要特殊的服用。
本发明另一个优点是具有稀薄的奶油蛋糕或果馅饼样结构的布丁形式,该产品可以为吞咽困难的人所食用。
本发明的另一个优点是具有富含维生素E和维生素C的优选实施方案,这样可以用来补充与感染、脓毒症或其它氧化性紧张状态有关的消耗之后的血液中的这些营养素的水平。优选,实施方案还包括牛磺酸,这样可以用来补充与感染、脓毒症或其它氧化性紧张状态有关的消耗之后的血液中的牛磺酸的水平。
本发明的另一个优点是,优选实施方案特别富含在患有胃病或肠道手术之后的病人中吸收差的维生素B-12和叶酸。

发明详述
本发明提供了一种营养补充剂,其特别适合于向老年患者或其它食欲不佳的病人提供补充营养同时降低过饱水平。由于其成分,所述的补充剂更易于消化,且因此使病人更可能摄取有效治疗量的补充剂或其它食品来充分补充营养。
蛋白质源包括至少约50wt.%的乳清蛋白质,该乳清蛋白质优选至少部分水解。用于产生水解产物的乳清蛋白质可以是市售的乳清蛋白质源,其基于甜乳清或酸乳清或其组合。优选地,乳清蛋白质是含超过80wt.%乳清蛋白质的乳清蛋白质源。合适的乳清蛋白质浓缩物是LACPRODAN 9087,而合适的乳清蛋白质分离体源包括ALACEN 895(NewZealand Milk Products Inc),BiPRO(Le Sueur Isolates of LeSueuf,Minnesota),PROVON-190(Avonmore Ingredients Inc ofMonroe Wisconsin)和LACPRODAN 9212(Royal Proteins,Inc ofRosemont Illinois)。
如果需要,蛋白质源可以包括一定量的其它合适种类的蛋白质。例如,蛋白质源还可以包括少量酪蛋白、大豆蛋白、米蛋白、豌豆蛋白、角豆胶蛋白、燕麦蛋白、牛奶蛋白、酪蛋白-甘-大肽(caseino-glyco-macropeptide)或这些蛋白质的混合物。而且,如果需要,蛋白质源还可以进一步包括少量的自由氨基酸。其它合适种类的蛋白质优选构成小于约50wt.%的蛋白质源,更优选小于约30wt.%。
该蛋白质源优选提供约8%至约20%的营养补充剂的能量。例如,在一个适合于成人的实施方案中,该蛋白质源可提供营养补充剂能量的约15%至约18%,或用于儿科的实施方案中提供营养补充剂能量的约8%至约14%。
该营养组合物包括脂源。优选该脂源提供营养补充剂能量的约18%至约40%;更优选营养补充剂能量的25%至约35%。例如,脂源可以提供营养补充剂能量的约30%。
该脂源可以包括中链甘油三酸酯(MCT),至多占总脂类的20wt.%。该脂源富含单不饱和脂肪酸。特别是,该脂源含至少约40wt.%的单不饱和脂肪酸。优选,该脂源含约45wt.%至约65wt.%的单不饱和脂肪酸;例如约55wt.%。
该脂源也可含有多不饱和脂肪酸。优选该脂源含约15wt.%至约30wt.%的多不饱和脂肪酸;例如约20wt.%的多不饱和脂肪酸。补充剂的脂类构成优选设计成具有的多不饱和脂肪酸ω-6(n-6)与ω-3(n-3)的比例是约1∶1至约10∶1。优选n-6与n-3脂肪酸比例是约5∶1至约9∶1;例如约7∶1。
该脂源具有的饱和脂肪酸含量小于约35wt.%;包括中链甘油三酸酯。更优选,脂源含小于约30wt.%的饱和脂肪酸。
合适的脂源包括高油酸向日葵油、高油酸红花油、向日葵油、红花油、菜籽油、豆油、橄榄油、加拿大菜籽油、玉米油、花生油、米糠油、奶油、榛实油和结构化脂类。分馏的椰子油是中链甘油三酸酯的合适来源。
营养补充剂也包括碳水化合物源。该碳水化合物源优选提供营养补充剂能量的约40%至约65%;特别是营养补充剂能量的约50%至约60%。例如,碳水化合物源可以提供补充剂能量的约54%。可以使用几种碳水化合物,包括麦芽糖糊精、玉米糖浆、玉米淀粉、改性淀粉、或蔗糖、或果糖、或其混合物。如果需要,该补充剂可以是无乳糖的。
该营养补充剂优选包括完全的维生素和矿物质构成。例如,可以提供足够充足的维生素和矿物质,以供应就每1000卡的营养补充剂而言约50%至约500%的维生素和矿物质推荐每日允许量。该营养补充剂优选富含维生素E。例如,该营养补充剂可以含有80至120国际单位的维生素E/1000kcal。更优选地,该营养补充剂含约30国际单位的维生素E/250ml份的补充剂。而且,该营养补充剂还富含维生素C,提供量是约150至约250mg/1000kcal或优选约60mg/份。该补充剂也优选含有200g叶酸和3g的维生素B-12/份。用于儿科的补充剂的实施方案具有改进的维生素和矿物质的构成,是针对这一年龄组的特殊需要而特制的。
该营养补充剂还包括可溶益生纤维源。益生纤维是通过选择性的刺激结肠中细菌的生长和/或活性从而有益于宿主的纤维,所述细菌能改善宿主健康。合适的可溶益生纤维包括果寡糖(FOS)和菊粉。合适的菊粉提取物可以由比利时的Orafti SA of Tirlemont 3300获得,商标是″Raftiline″。同样地,合适的果寡糖可以由比利时的Orafti SAof Tirlemont 3300获得,商标是″Raftilose″。
优选地,提供的FOS和菊粉的比例是约60∶约40至约80∶约20,最优选约70∶约30。其它可能的纤维包括树胶,例如瓜尔豆胶、黄原胶、木寡糖、阿拉伯树胶、果胶、金合欢胶、稳定淀粉、葡聚糖或这些物质的混合物。所述纤维不应导致饱腹感。
可溶的益生纤维据报道有助于胃肠道中双岐乳杆菌的生长和在某种情况下预防或减少病原体例如梭状芽胞杆菌的生长。而且,促进双歧乳杆菌生长据报道还有多种其它的有益效果。另外,在结肠中纤维发酵期间,产生短链脂肪酸。这些脂肪酸是肠细胞的燃料。
该可溶的益生纤维优选含量是每天足以向病人提供约4至约9g的可溶的可发酵的纤维。因此,益生纤维可以以约6g至约12g/1000kcal的量存在。另一种实施方案包含9g或更少的益生纤维混合物,例如4g的混合物。
如果需要,该营养补充剂也可以含有不溶的膳食纤维源。合适的不溶膳食纤维源是豆和谷物的壳纤维;例如豌豆壳纤维、燕麦壳纤维、大麦壳纤维和大豆壳纤维。
该营养补充剂优选的能量含量是约800kcal/l至约2000kcal/l;例如约1000kcal/l或约1500kcal/l的能量含量。
该营养补充剂可以是适合于口服或肠内给药的可溶粉、浓缩液体、布丁、棒/点心或即用制剂的形式。即饮饮料制剂是特别优选的。也可以含有各种风味剂、甜味剂和其它添加剂。人工甜味剂例如双氧噁噻嗪和基于L-门冬氨酰基的甜味剂可以使用,例如双氧噁噻嗪钾或阿斯巴甜或其混合物。
该营养补充剂可以例如通过将悬浮于水中的、优选是悬浮于已经经过了反渗透的水中的蛋白质源与脂源一起混合来制备。可以使用市售的液化器形成液体混合物。如果使用,在混合物中可以包括乳化剂。维生素和矿物质可以在此时添加,但通常在这之后添加以避免热降解。任何添加的亲油维生素、乳化剂等在混合之前可以被溶进脂源中。该液体混合物接着被均质;例如分两阶段,在第一阶段是约7MPa至约40MPa,而第二阶段是约2MPa至约14MPa。如前所述进行蛋白质水解。或者,乳清蛋白质可以在水中进行复水,在乳液形成之前水解。如果水解的乳清蛋白质和其它完整的蛋白质的混合物是所需的,则这是优选工艺。如果这是实践中被采用的,则接着完整蛋白质和脂类在水解步骤之后添加到水解的乳清蛋白质中,且该混合物接着被均质化。优选通过加热或通过调节pH或结合的方式使酶变性,从而实现水解的终止。通过采用所设计的使加热对蛋白质稳定性和产品味道和品质损害最小化的条件,实现对酶活性的钝化。例如,通过在约90℃加热5分钟至约110℃加热约15秒,实现酶的钝化。这可以通过喷射蒸汽或热交换器例如板式热交换器来实现。
该液体混合物可以接着被逐渐冷却至约20℃至约30℃;例如通过闪冷却和热交换器,优选板式热交换器。碳水化合物源可以在此时或之后,以干粉的形式或液体浆液的形式加入。该混合物可以接着被进一步冷却,以添加任何热敏成分;例如维生素和矿物质。水,优选已经经过了反渗透的水,可以接着被混合进来,以形成液体混合物。该均质混合物的pH和固体含量在此处被方便地进行标准化。
该液体混合物可以接着被热处理,例如使用无菌工艺,以降低细菌量和对该产品灭菌。例如,该液体混合物可以被快速地加热至约110℃持续5分钟至约150℃持续约5秒。这可以通过喷射蒸汽或通过热交换器,例如板式热交换器进行。该液体混合物接着被均质化;例如在两阶段中,在第一阶段是7MPa至约40MPa,而第二阶段是约2MPa至约14MPa。
如果需要制备液体营养补充剂,则将均质的混合物注入合适的容器,例如罐中。填料可以是无菌的,或容器可以被蒸煮。进行填料的合适的设备是市售的。
并不想被理论所束缚,相信乳清蛋白质被快速地从胃中排空,且在肠内被快速地水解和吸收。这导致了较短的病人有饱腹感的餐后期,且由此导致食欲的快速恢复。相反,蛋白质,例如酪蛋白,缓慢地从胃中排空,导致稳定的、延长的、病人有饱腹感的餐后期。因此,营养补充剂可以用于向食欲不佳和/或蛋白质-能量营养不良的老年的和患病的和渐渐康复的病人提供补充营养。
还发现所述氨基酸构成很适合于促进谷氨酸盐的内源性产生。因此该营养补充剂可以被用作动物或人的谷氨酸盐间接来源。特别是该营养补充剂可以向耗尽谷氨酸盐的紧张状态的病人提供营养;例如针对处于危险病情的患者、或患有脓毒症、受伤、烧伤、有炎症的患者;或由手术恢复中的病人。而且,该营养补充剂可以用于促进受伤或有肠道疾病的患者谷氨酸盐的合成,或保持肠道的生理机能。此外,该营养补充剂可以用来保持或提高人和动物的血浆谷氨酸盐水平,并提高免疫能力。
而且,发现乳清蛋白质含高水平的苏氨酸,这是一种重要的粘蛋白的构造块。因此该营养补充剂的优点是可以用于向处于粘蛋白产生受损或降低或有此危险的患者提供补充营养,例如这样的患者是有炎症反应的病人、营养不良、膀胱纤维化、恶性肿瘤、慢性肠炎、溃疡性结肠炎和克罗恩氏病的患者,服用了非甾族消炎药治疗的患者等,并与总的非肠道营养相一致。另外,发现乳清蛋白质含高水平的半胱氨酸,这是一种重要的抗氧化剂和谷胱甘肽的直接前体。因此,该营养补充剂的优点是可以用于向耗尽谷胱甘肽和低抗氧化剂的患者提供补充营养。例如,该营养补充剂可以向老年人或患有急性或慢性炎症,或从急性或慢性炎症中恢复的病人供应营养。
病人所需服用的营养补充剂的量根据各种因素是不同的,例如要根据病人的病况、病人的体重、病人的年龄和其它营养源。然而,所需的量对于医务工作者来说是易于掌握的。该营养补充剂可以以多剂量服用,例如采用2至5次来满足每日供应量,或可以只服用一次。
采用实施例的方式,并非进行限制,本发明的实施例用于进一步的举例说明。实施例1
制备一种立即可饮的营养补充剂。该营养补充剂包括以下成分:

该补充剂的能量密度是1000kcal/l。实施例2
制备一种立即可饮的营养补充剂。该营养补充剂中16%的热量来自蛋白质,该蛋白质的70%是水解乳清,其余的是完整蛋白质。其余的成分如实施例1所述。
该补充剂的能量密度是1000kcal/l。实施例3
制备一种立即可饮的营养补充剂。该营养补充剂中16%的热量来自蛋白质,该蛋白质的50%至100%可以源自乳清或水解乳清蛋白质。
该补充剂的能量密度是1500kcal/l。实施例4
制备一种粉状的营养补充剂。该营养补充剂中16%的热量来自蛋白质,该蛋白质的50%至100%可以是乳清或水解乳清蛋白质。该补充剂含有益生菌,优选詹生氏乳杆菌。实施例5-儿科RTD
制备一种立即可饮的营养补充剂,该补充剂是针对儿童生长所特制的。例如,该营养补充剂中含有较低的热量百分含量作为蛋白质,优选10-12%的热量是蛋白质,而较高的热量百分含量是碳水化合物。其余的成分如实施例1中所述。
该补充剂的能量密度是1000kcal/l。实施例6-儿科粉
制备一种粉状的营养补充剂。该营养补充剂10-12%的热量来自蛋白质,该蛋白质的50%至100%可以是乳清或水解乳清蛋白质。该补充剂含有益生菌,优选双歧芽胞杆菌和嗜热链球菌。实施例7-棒和布丁
制备补充剂的可替代形式,例如布丁和点心棒。该布丁和果子或果酱露馅饼的形式适合于吞咽困难的对象。所有的所制备的可替代形式其10%-16%的热量来自蛋白质,该蛋白质的50%至100%可以是乳清或水解乳清蛋白质。
应该理解对本文所述的优选实施方案的多种变化和改进对本领域普通技术人员是显而易见的。实施这样的变化和改进并不脱离本发明的实质和范围,并不会有损于其所带来的优点。因此,这样的变化和改进为所附的权利要求所覆盖。
Claims (25)
1.一种组合物,其包含
蛋白质源,该蛋白质源提供组合物总热量的至少约8%,且其包括蛋白质源至少约50wt.%的乳清蛋白质,
脂源,具有的ω3至6脂肪酸比例是约5∶1至约10∶1,且提供组合物总热量的至少约18%;
碳水化合物源和大量营养剂构成,该大量营养剂构成至少包含维生素E和维生素C。
2.根据权利要求1的组合物,其中乳清蛋白质包括部分或基本上全部水解的乳清蛋白质。
3.根据权利要求1或2的组合物,其中乳清蛋白质包括乳清蛋白质水解产物,其构成蛋白质源的至少50wt.%。
4.根据前述权利要求任一项的组合物,其中至少50wt.%的乳清蛋白质是水解的。
5.根据前述权利要求任一项的组合物,其中该组合物包括酪蛋白-甘大肽。
6.根据前述权利要求任一项的组合物,其中蛋白质源提供组合物总能量的至多约20%,优选约10%至约20%。
7.根据前述权利要求任一项的组合物,其中脂源包含约40wt.%至约65wt.%的单不饱和脂肪酸。
8.根据前述权利要求任一项的组合物,其中饱和脂肪酸含量小于约30wt.%。
9.根据前述权利要求任一项的组合物,其中脂源提供组合物总能量的约25%至约35%。
10.根据前述权利要求任一项的组合物,其中碳水化含物源包括蔗糖、玉米糖浆、麦芽糖糊精和/或其组合。
11.根据前述权利要求任一项的组合物,其中碳水化合物源提供组合物总能量的约50%至约60%。
12.根据前述权利要求任一项的组合物,包括选自维生素E、维生素C、牛磺酸、叶酸和维生素B-12中的至少一种微量营养素。
13.根据前述权利要求任一项的组合物,其中该组合物包含选自菊粉、阿拉伯树胶、稳定淀粉、葡聚糖、木寡糖(XOS)、果寡糖(FOS)和/或其组合的至少一种益生纤维。
14.根据前述权利要求任一项的组合物,其中该组合物包括至少一种益生微生物。
15.前述权利要求任一项的组合物用作营养补充剂的用途。
16.权利要求1-14任一项的组合物用于宠物食品的用途。
17.权利要求1-14任一项的组合物用于制备可摄取的载体以向消化蛋白质能力受损的人提供营养的用途。
18.权利要求1-14任一项的组合物用于制备可摄取的载体以向未接受足够量营养的人提供营养的用途。
19.权利要求1-14任一项的组合物用于制备可摄取的载体以向患病的人补充营养的用途。
20.权利要求1-14任一项的组合物用于制备可摄取的载体以向消化蛋白质能力降低的人提供营养的用途。
21.根据权利要求18的用途,其中所述的人是食欲不佳的人、老年人、由病中恢复的人,由经过了手术恢复的人、患有慢性胃炎的人、胃蛋白酶消化能力降低和/或渐渐康复的人。
22.制备权利要求1-14任一项的组合物的方法,其包括将蛋白质源、脂源、碳水化合物源和微量营养素混合的步骤。
23.根据权利要求21的方法,包括将蛋白质源用一种或多种酶在pH范围是约6.6至约8.8、温度是约40至约70℃和酶浓度是蛋白质的约0.5至约2.5%条件下进行水解的步骤。
24.根据权利要求23的方法,其中蛋白质的水解通过酶处理进行约5至约120分钟。
25.根据权利要求22至24任一项的方法,包括向产品中添加至少一种益生菌或益生元的步骤。
