一般在多种其它物质存在下制备乳液,以获得乳化作用、粘度、稳定性和外观各方面期望的平衡。例如,乳液制剂常常需要至少一种乳化剂(往往为多种乳化剂的组合)。这些乳化剂促进一种不混溶相分散入另一种不混溶相中,且有助于稳定乳液。乳化剂包括多种合成的和天然的成分。例如,在食品如人造奶油或焙烤产品中广泛使用甘油单酯及其化学衍生物作为乳化物。天然乳化物的例子是卵磷脂(一种存在于蛋黄中的磷脂),其通常用于制备蛋黄酱。也可在乳液中包含活性成分。这在含有难溶于水溶液的活性剂如某些维生素和核苷酸之组合物中尤其希望,。活性成分也常常配制成乳液以增加其稳定性。一种包含药剂的乳液系统的例子见于美国专利5,602,183,其公开了一种含有抗炎症剂的伤口愈合组合物。前述范例仅仅为包括在本领域已知乳液制剂中的庞大数目的成分中的几个。乳化物及其应用的综述可见于Becher,P.,乳液工业百科全书,Dekker编,1983。
在油料作物的种子中,包括经济上重要的作物如大豆、油菜籽、向日葵和棕榈,水不溶的油组分储存在多种本领域已知的独立亚细胞结构中,如油体、含油体(oleosome)、脂体或球形体中(Huang,1992,植物分子生物学年鉴,43:177-200)。除了化学上称为脂肪酸的甘油脂的油类混合物(三酰基甘油酯),油体还包含磷脂和一系列相关蛋白质,统称为油体蛋白。从结构上看,油体可被认为是由其中包埋了油体蛋白的磷脂单层包被的三酰基甘油脂基质(Huang,1992,植物分子生物学年鉴,43:177-200)。存在于植物种类的油体部分中的种子油是多种三酰基甘油酯的混合物,其确切的成分取决于油所来源的植物种类。通过经典育种和遗传工程的结合可操纵种子的油类特性,拓宽天然可得的植物油组分的种类。关于该领域所作努力的综述参见Designer油料公司/育种、加工和生物技术学,D.J.Murphy编,1994,VCH Verlagsgesellschaft,Weinheim,德国。
植物种子油用于多种工业应用,尤其是在食品工业、洗涤剂工业和化妆品工业。为了获得用于这些方面的植物油,压榨种子且随后采用诸如有机萃取、脱胶、中和、漂白和过滤的工艺精炼。已公开了植物油料种子的水抽提方法(如,Embong和Jelen,1977,加拿大工业食品科技杂志,10:239-243)。由于先有技术所教导的方法之目的在于获得纯油,在这些产品加工过程中油体丧失了其结构完整性。因此,先有技术中从植物油配制的乳液一般不含有完整的油体。
虽然已有许多基于矿物油的产品之应用占了市场的主流,在其它的应用中来源于植物和化石的油构成了直接竞争。例如,已广泛用于洗涤剂制造的月桂油获自矿物油以及椰子油,且最近又可获自遗传工程化的油菜籽(Knanf,V.C.,1994,脂科学技术,96:408)。然而,目前日益需求原料的生物可降解来源。基于植物油体的本发明的乳液优于类似的基于矿物油的制剂,其优点在于其油部分来自可再生的有利于环境的来源。
美国专利5,683,710和5,613,583公开了包含来自产油植物脂囊泡的乳液。这些专利中公开的乳液从相对较粗的种子提取物中制备,包含多种种子成分,如糖基化和非糖基化蛋白质。这些专利涉及的乳液的缺点在于它们含有赋予乳液多种不期望性质的杂质种子成分,这些性质包括致敏性以及不期望的气味、香味、色泽以及感官特性。由于种子杂质的存在,公开于这些专利中的脂囊泡制剂的应用范围有限。
发明概述
本发明涉及含有油体的新型乳液制剂。本发明的乳液制剂可以非毒性和食品级的形式获得。此外,乳液制剂可有利地从一种奶油状质地油体制品中制备,因而可方便地用于多种化妆品和工业用途。本发明人发现,活细胞的油体可用于多种新型乳液基食品、化妆品、药物和工业产品的制剂。广义上说,本发明提供了一种含有源自细胞的经洗涤的油体的乳液制剂。
本发明也提供了用于制备乳液制剂的方法以及乳液制剂在多种工业和化妆品组合物中的用途。
相应地,本发明提供了一种用于制备乳液制剂的方法,包括:1)从细胞获得油体;2)洗涤油体;3)将经洗涤的油体配制成乳液。
在本发明的一个优选实施方案中,经洗涤的油体制品获自植物种子,包括获自油菜籽、大豆、玉米和向日葵的种子。相应地,本发明提供了一种用于从植物种子制备乳液制剂的方法,包括:
a)研磨植物种子;
b)从研磨过的种子中去除固体;
c)从水相中分离油体相;
d)洗涤油体相以得到经洗涤的油体制品;
e)将经洗涤的油体制品配制入乳液。
在本发明的一个优选实施方案中,在种子研磨之前或之后加入液体相。
在本发明的另一个优选实施方案中,乳液的配制e)还包括加入液体相到经洗涤的油体制品中。
本发明乳液可用于多种用途,包括食品和饲料产品、药品、个人护理用品和工业产品。本发明乳液制剂特别适用于食品级产品的制备,因为它无毒,结构呈奶油状,且可生物降解。
在结合附图及下面的发明详述之后,本发明的其它目的、优点和特征是显而可见的。

发明详述
如上述,本发明涉及包含来源于细胞的乳液制剂。在一个实施方案中,本发明提供了一种包含经洗涤的油体的乳液制剂。
在另一实施方案中,本发明提供了一种用于制备乳液制剂的方法,包括1)从细胞获得油体;2)洗涤油体;3)配制经洗涤的油体进入乳液。
细胞可以是含有油体的(或油体样结构的)任何细胞,包括植物细胞、动物细胞、真菌细胞和细菌细胞。在本发明的一个优选实施方案中,油体可获自植物细胞。可采用下述方法通过破裂植物细胞膜和细胞壁从植物细胞中获得油体,所述方法是那些释放细胞组成物而基本不破坏油体的结构完整性的方法。更优选的,油体获自植物种子。相应地,本发明还提供了用于制备乳液制剂的方法,包括:
1)通过如下方法从植物种子中获得油体;
(a)研磨植物种子;
(b)从研磨的种子除去固形物;及
(c)从水相中分离油体相;
2)洗涤油体相以产生经洗涤的油体制品;以及
3)将经洗涤的油体制品配制成乳液。
在本发明的一个优选实施方案中,在种子研磨之前或之后向种子加入液体相。
在本发明的另一个优选实施方案中,配制乳液(e)还包括将一种液体相加入到经洗涤的油体制品中。
术语“研磨”如此处所用是指碾磨、压榨、劈剁或压碎种子,在本发明中这些术语可互换使用。在加工中,种子细胞破碎。
术语“固形物”如此处所用,是指在水相或油体相中不溶的任何物质,如种子壳。
术语“洗涤油体”如此处所用是指将细胞杂质从油体相中除去的任何方法,特别是除去赋予乳液制剂不期望性质的那些杂质,如致敏性、不希望的颜色、气味、口味或感官性质,或其它任何不期望的性质。洗涤方法的例子包括基于重力作用的分离方法(诸如离心)和基于体积排阻的分离技术(诸如膜超滤和错流微滤)。洗涤方法及条件相应于经洗涤的油体制品的期望纯度。
术语“经洗涤的油体制品”如此处所用,指一种大量细胞物质被除去的油体制品,包括除去赋予乳液制剂不期望性质的杂质,如致敏性、不希望的颜色、气味、口感或感官性质,或其它任何不期望的性质。优选地,经洗涤的油体制品含有低于10%的其它种子蛋白质。
“将油体配制入乳液”,这是指如果需要,混合或匀浆经洗涤的油体制品直到形成乳液。在一个优选实施方案中,加入了其它成分,如液体相,混合经洗涤的油体制品和该液体相直到获得均质混合物。
由于下述优点,经洗涤的油体制品特别适于乳液的配制。油体的性质
本发明乳液制剂包含大小、形状和密度大体上一致的完整的经洗涤的油体。在电子显微镜下观察时,可见油体基本上为球形结构(见例子Murphy,D.J和Cummins I.,1989,植物化学(Phytochemistry),28:2063-2069;Jacks,T.J.等人,1990,JAOCS,67:35-361)。油体的大小一般从0.4μm至1.5μm(Murphy,D.J.和Cummins I.,1989,植物化学,28:2063-2069)。用Malvern体积分析仪分析,发现在从油菜籽分离的经洗涤的油体制品中,油体的直径1μm,为对称的单峰型分布。利用Malvern体积分析仪,可将经洗涤的油体制品与市售水包油乳液(包括豆奶、蛋黄酱(Kraft真蛋黄酱)和两种椰奶制品(Tosca,Aroy-D)明显区分开来。油体的确切大小和密度至少部分取决于存在的蛋白质/磷脂/三酰基甘油酯成分的确切比例。在电子显微镜下观察时,制备本发明的经洗涤的油体不会导致油体的形状发生实质性改变(与完整种子中存在的那些相比)。
一旦破裂了含有油体的细胞,由于利用了离心力水溶液中的油体级分会漂浮,可将油体级分方便快速地从水溶液中分离出。溶液中,当油体密度高于溶剂(如95%的乙醇)密度时,在同样条件下油体会沉淀。也可利用基于体积排阻的分离技术如膜过滤从水级分中分离油体级分,这种方法的优点在于可获得大小更均一的油体。
在本发明经洗涤的油体制品中存在的油体可耐受强酸和强碱,包括在至少低至pH2的酸性条件和至少高于pH10的碱性条件下长时间暴露。当暴露于pH12时,可见油少量损失,表明油体结构完整性的丧失。此外,用多种有机溶剂(包括甲醇、乙醇、己烷、异丙醇和乙酸乙酯)萃取也不会或略微影响存在于经洗涤的油体制品中油体的完整性。存在于经洗涤的油体制品中的油体也可经受与阴离子的洗涤剂(十二烷基硫酸钠(SDS))、阳离子洗涤剂(十六烷三甲基溴)和非离子洗涤剂(Tween-80)混合。在SDS存在下经洗涤的油体制品的煮沸可导致肉眼可见的油体结构的一部分丧失完整性。在高达至少100℃下维持2小时,存在于经洗涤的油体制品中的油体保持稳定。经洗涤的油体制品经缓慢冻融导致其物理外观的改变,特征为相对于均质的乳液而言形成了团块。通过如下方法可防止在冻融后油体形成更大程度的团块:a)在液氮中速冻而不是在-20℃缓慢冷冻;或b)在冷冻前向油体制品中加入高于5%(V/V)的甘油。可耐受相对剧烈的化学和物理条件是本发明经洗涤的油体制品中存在的油体的独特性质。
本发明提供了包含已去除显著量的种子杂质之油体的乳液制剂。这些杂质包括蛋白质、易挥发物和赋予不期望的颜色、气味、香味、感官特性或其它不期望特性的其它化合物。已报道许多种子蛋白质可引起致敏反应。例如,Ogawa等人,(1993,生物科学生物技术与生物化学(Biosci.Biotechnol.Biochem.),57:1030-1033)报道大豆糖蛋白P34(也称为Gly m Bd 30K)的致敏性。也报道了针对油菜籽、小麦和大麦种子蛋白质的致敏反应(Armetia等人,1993,临床实验变态反应(Clin.Exp.Allergy.)23:410-415;Monsalve等人,1993,临床实验变态反应27:833-841)。因此杂质种子蛋白质的去除是有利的。可选择洗涤条件以获得基本上纯的油体制品。在这种情况中,制品中主要仅有油体存在。
对于多种应用而言,也希望可获得越纯越好的油体制品,因为这样可更好地控制最终乳液的配制加工。为使经洗涤的油体制品可被包含在不同的乳液中,期望将挥发性保持在最低,且颜色优选为浅色或白色。油体制品的洗涤可得到颜色较浅的制品。此外,除去了大多数易挥发物。通过洗涤也除去了那些可促进微生物生长的化合物,因为观察到与未经洗涤的制品相比,经洗涤的油体制品有较长的贮存期。通过洗涤去除的其它化合物包括抗营养型芥子油甙(glucosinilate)和/或其降解产物和纤维材料。当加热至60℃至80℃时,可观察到与未经洗涤的制品相比,被经洗涤的油体品吸收的大量残留的水。经冷却至室温并离心后,观察到经洗涤的油品保持稳定,而在未经洗涤的制品中出现相分离。考虑到经洗涤的油体稳定性的增加,当配制工艺中涉及到加热时这种油体是优选的。加热至40℃时,经洗涤的油体制品可吸收大量外部添加的水而不会导致相分离。于是在配制水乳液时,经洗涤的油体是优选的。也在经洗涤的油体制品与未经洗涤的制品之间比较了吸收外部加入的油的能力。在形成不稳定的乳液之前可向经洗涤的油体制品中加入大量外部的油。在配制工艺中加入外源油或蜡时(如制备润滑剂或化妆品时)这一特点在配制中有利。当比较经洗涤的油体制品与未经洗涤的制品之间的粘度时,发现经洗涤的制品更粘稠。油体制品更加粘稠是期望的,因为这可避免在配制加工中加入增稠剂。
因此,此处提供的经洗涤的油体制品在许多方面优于未经洗涤的制品。本发明的经洗涤的油体制品是较好的制品,贮存期长且有优选的颜色、气味和粘稠度等特性。经洗涤的油体制品也有较好的水吸收及油吸收特性。最后,由于大量种子蛋白质的去除很少出现致敏反应。这些特性使得经洗涤的油品可用于多种化妆品用和工业用乳液的配制中。
用如本申请实施例2中详述的方法制备的获自油菜籽的经洗涤的油体和未经洗涤的油体完成上述观察。无论油体的来源,据信存在于本发明经洗涤的油体制品中的油体性质之一为可耐受剧烈的化学和物理条件。然而,在此之前所记载的油菜籽油体的一种或多种性质将会或多或少取决于获得所述经洗涤的油体制品的细胞。不仅如此,还要清楚地知晓,本发明涉及了一种可从任何包含油体的细胞中获得的油体制品。
在本发明的一个优选实施方案中,油体制品获自植物种子。乳液中完整油体的存在与这些油体所述的特性将本发明乳液制剂与其它可从植物种子制备的物质明确区分开来。油体的来源与制备
本发明的经洗涤的油体制品可获自任何含有油体或油体样结构的细胞。这包括动物细胞、植物细胞、真菌细胞、酵母细胞(Leber,R.等人,1994,酵母10:1421:1428)、细菌细胞(Piepar-Furst等人,1994,细菌学杂志,176:4328-4337)以及藻类细胞(Rossler,P.G.,1988,生理学杂志(J.Physiol.)(London),24:394-400)。在本发明的一个优选实施方案中,油体获自植物细胞,包括其中存在油体或油体样结构的来自花粉、孢子、种子和植物营养性器官的细胞(Huang,1992,植物生理学年鉴,43:177-200)。更优选地,本发明的经洗涤的油体制品获自植物种子,最优选地,获自下列植物种类,包括油菜籽(Brassicaspp.)大豆(Glycine max)、向日葵(Helianthus annuus)、油棕榈(Elaeis guineeis)、棉籽(Gossypium spp.)、花生(Arachis hypogaea)、椰子(Cocus nucifera)、蓖麻(Ricinus communis)、红花(Carthamustinctorius)、芥末(Brassica spp.和Sinapis alba)、芫荽(Coriandrumsativum)、南瓜(Cucurbita maxima)、亚麻子/亚麻(Linumusitatissimum)、巴西果(Bertholletia excelsa)、希蒙得木(Simmondsiachinensis)和玉米(Zea mays)。用本领域普通技术人员周知的农业载培方法培养植物且使其结籽。收获种子后(如需要,除去如石头、种子壳(脱壳)等物料)通过例如筛选或洗涤,以及任选地干燥种子,随后将种子用机械压迫、研磨或碾压进行加工。在本发明的一个优选实施方案中,在研磨种子之前加入一种液体相。这被称为湿法研磨。优选的液体为水,虽然如乙醇等有机溶剂也可使用。已报道在油萃取加工中湿法研磨用于来自多种植物种类的种子,包括:芥末(Aguilar等人,1990,织物研究杂志(J.Texture Studies.)22:59-84)、大豆(美国专利3,971,856;Carter等人,1974,美国油化学协会杂志(J.Am.OilChem.Soc.)51:137-141)、花生(美国专利4,025,658;美国专利4,362,759)、棉籽(Lawhon等人,1977,美国油化学协会杂志,63:533-534)和椰子(Kumar等人,1995,INFORM 6(11):1217-1240)。在研磨前将种子在液体相中浸泡约15分钟至约2天的一段时间也是有利的。浸泡可软化细胞壁且便于研磨加工。较长时间的浸泡可模拟种子萌发过程,导致种子组成的成分中某种有利的转变。在另一实施方案中,液体相在种子研磨后加入。这称为干法研磨。优选的所加入的液体相为水。
优选的利用胶体磨如MZ130(Fryma Inc.)研磨种子。除了胶体磨,能够加工工业规模量的种子的其它研磨和压榨设备也可用于此处所述的发明,包括:薄片磨、盘式磨粉机、胶体磨、针磨(pin mill)、轨道磨(orbital mill)、IKA磨和工业规模的匀浆机。磨的选择取决于种子通量要求以及所用种子的来源。关键之处在于在研磨过程中种子油体保持完整。因此,任何通常用于油籽加工中意在破坏油体的操作条件均不适用于本发明的方法中。研磨温度优选在10℃至90℃之间,更优选在26℃至30℃之间,pH优选保持在2.0至10之间。
固体杂质如种子壳、纤维性物料、不溶的碳水化合物和蛋白质及其它不溶杂质均被从压榨的种子级分中去除。固体杂质的分离可利用倾析式离心机如HASCO 200两相倾析式离心机或NX310B(AlphaLaval)来完成。根据种子通量的要求,可通过利用其它型号的倾析式离心机如三相倾析式离心机来改变倾析式离心机的能力。可根据所用特定的离心机改变操作条件,且必须调整以使不溶性杂质物料沉淀下来,且经过倾倒仍保持沉淀。在这些条件下可观察到油体相与液体相的部分分离。
不溶性杂质去除后可从水相中分离油体相。在本发明的一个优选实施方案中,利用了管式离心机。在其它实施方案中,可采用旋液分离器、层碟式离心机或自然重力作用下的相分层或者任何其它基于重力的分离方法。也可利用体积排阻法从水相中分离油体级分,如膜超滤和错流微滤。在优选实施方案中,管式离心机为Sharples型AS-16(Alpha Laval)或AS-46 Sharples(Alpha Laval)。关键参数是用于操作离心机的环形挡料圈(dam)的尺寸。环形挡料圈是带有中央环形开口的可拆卸环,在AS-心部6中开口从28-36mm,调节水相从油体相中的分离,从而控制所获油体级分的纯度。在优选的实施方案中,当使用AS-16时采用29或30mm的环形挡料圈。所用确切挡料圈尺寸取决于所用油籽的类型以及所期望的油体制品的最终的一致性。分离的效率还受到流速的影响。使用AS-16时,流速一般在750-1000ml/分钟(环形挡料圈尺寸29)或在400-600ml/分钟(环形挡料圈30),温度优选地保持在26℃至30℃。依据所用离心机型号,必须调节流速和环形挡料圈尺寸以实现从水相中的油体级分最优分离。这些调节为本领域普通技术人员显而易见。
从油体级分中分离固体和水相也可利用基于重力的分离方法(如三相管式离心机、或倾析式离心机、或旋液分离器)或基于体积排阻分离方法同时进行。
在本方法的这个阶段所获组分一般相对较粗,且含有大量种子蛋白质,其中包括糖基化和非糖基化蛋白质,以及其它杂质如淀粉或芥子油甙或其降解产物。本发明包含去除大量种子杂质的步骤。为实现杂质种子物料的去除,一经从水相中分离的油体制品至少洗涤一次,方法是重悬浮油体级分,并离心重悬浮的级分。这种方法产生用于本申请的称为经洗涤的油体制品的产物。洗涤次数一般取决于油体级分的期望纯度。依据所用洗涤条件,可获得基本上纯的油体制品。在这种制品中仅存的蛋白质为油体蛋白。为了洗涤油体级分,可使用管式离心机或其它离心机,如旋液分离器或层碟离心机。油体的洗涤可用水、缓冲系统(如浓度在0.01M至最少2M之间的氯化钠、高pH(11-12)处的0.1M硼酸钠)、低盐缓冲液(如50mM Tris-HCl(pH7.5))、有机溶剂、洗涤剂或任何其它液体相。在优选的实施方案中,优选在高pH(11-12)进行洗涤。可依所用种子类型的不同采用相应的用于洗涤的液体相以及洗涤条件(如pH和温度)。在pH2至pH11-12之间的一系列不同pH下洗涤可能有益,因为这样可以逐步除去杂质特别是蛋白质。选择洗涤条件以使洗涤步骤去除大量的杂质而不影响油体结构的完整性。在实施方案中如进行多于一步的洗涤,各不同洗涤步骤的条件可以不同。可方便地利用SDS凝胶电泳或其它分析技术监测经过油体的洗涤后种子蛋白质和其它杂质的去除。在各个洗涤步骤之间不必除去全部的水相,最终经洗涤的油体制品可悬浮在水、缓冲体系(如50mM Tris-HCl(pH7.5))或任何其它液体相中,且如果需要,可将pH调至pH2至pH10之间的任何pH值。
制备经洗涤的油体制品的方法可以分批操作或连续方式进行。特别是当使用管式离心机时,产生一种连续流量的加工系统,该系统为步骤(a)和(b)、(b)和(c)以及(c)和(d)之间的泵操作。在优选的实施方案中,泵为1英寸M2 Wilden气动双隔膜泵。在其它实施方案中,可使用液压泵或蠕动泵。为了维持向倾析式离心机和向管式离心机均匀一致的供应,可在分离步骤之间加入如IKA匀浆机的匀浆机。串联的匀浆机也可加在多种用于洗涤油体制品的离心机或基于体积排阻的分离装置之间。在每一洗涤步骤中的环形挡料圈、缓冲液组成、温度和pH可以与第一个分离步骤中所用的环形挡料圈尺寸不同。
在本发明的一个优选实施方案中,当从软组织例如橄榄的中果皮组织中分离油体时,用于破裂细胞的技术可与那些用于破碎坚硬种子的技术略有不同。例如,基于压力的技术优于碾压技术。小规模分离油体的方法也已有报道,如从橄榄(Olea europaea)及鳄梨(Perseaamericana)中果皮组织中分离油体,(Ross等人,植物科学,1993,93:203-210),以及从油菜籽(Brassica napus)微孢子衍生的胚中分离油体(Holbrook等人,植物生理学,1991,97:1051-1058)。
在本发明的一个优选实施方案中,当从非植物细胞中获得油体时,按照与上述相似的方法分离经洗涤的油体制品。已有记载关于从酵母中分离油体的方法(Ting等人,1997,生物化学杂志,272:3699-3706)。
油级分的化学及物理性质可在至少两方面加以改变。首先,不同植物种类含有的油体带不同的油组成成分。例如,椰子中富含月桂酸油(C12),而芥酸油(C22)大量存在于某些芸苔属(Brassica)的种中。其次可通过应用本领域普通技术人员已知的育种和遗传工程技术在特定植物种群中改变油的相对量。这些技术均意在改变控制参与油合成的代谢途径中的酶的相对活性。通过应用这些技术,可获得带有复杂的不同油的种子。例如,通过育种已产生了带有低芥酸含量的油菜籽(Canola)(Bestor,T.H.,1994,Dev.Genet.15:458);以及带有改变了的油的植物品系,其中油的双键的位置和数量发生了改变;改变了脂肪酸的链长度,且已通过遗传工程引入了期望的官能团(Topfer等人,1995,科学,268:681-685)。利用类似方法本领域普通技术人员可进一步拓展油体的可得来源。不同的油组成成分会产生油体的不同物理化学性质。因此,通过从作为油体来源的不同的种类或植物品类中筛选油籽或其混合物,可得到带有不同质地和粘稠度的宽组成的乳液。配制乳液
可利用本领域已知的方法将经洗涤的油体制品配成乳液。优选的,至少将一种添加成分加入经洗涤的油体制品。添加成分可作为溶液、悬浮液、胶体或固体加入,添加成分的量取决于制剂。经过配制后添加成分可与油体结合、保持悬浮在溶液中或形成油体分散于其中的悬浮液。添加成分也可穿透包围油体的磷脂单层或三酰基甘油脂基质。可穿透油体的成分包括油、蜡和颜料尼尔红(Nile Red)。在本发明的一个优选实施方案中,添加成分为液体相。另在本发明的一个优选实施方案中,液体相是水。水可直接加入也可通过与其它成分结合的水分形式加入。水的最终量并不关键,只要经过对成分的混合可形成稳定的乳液。一般地,组合物中含有至少1%到至多99%的水。通常需要混合以提供足够的乳液,这也可能需要加热或加压。
在另一优选实施方案中,添加的成分是油或蜡。油或蜡可掺入油体的三酰基甘油酯基质,以此方式,脂溶性成分如脂溶性维生素可递送到油体基质。当油或蜡包含有添加的成分时,油体可保持悬浮在亲脂相中,或形成双重乳液(double emulsion)。
最终的组合物可以为固体或液体形式,或者有任意期望的粘稠度。乳液也可用下列胶凝剂增稠,如纤维素或其衍生物、Carbopol和衍生物、角豆(carob)、角叉菜胶(carregeenans)及衍生物、黄原胶、sclerane胶、长链烷醇胺和苯酮及其衍生物,一般它们的浓度小于2%(重量百分比)。
乳液还可包含表面活性剂,以湿润、发泡、渗透、乳化、溶解和/或分散所选定的物料。如需要加入阴离子表面活性剂,例如椰油单甘油酯硫酸钠;阳离子表面活性剂,如氯化月桂基三甲基铵、氯化十六烷基吡啶和溴化三甲基铵;非离子表面活性剂包括Pluronic、烷基苯酚的环氧乙烷缩合物;以及诸如脂肪族季铵盐的衍生物、磷鎓和硫鎓化合物的两性离子表面活性剂。
如需要也可在乳液制剂中包括可结合金属离子的鳌合剂,如酒石酸、EDTA、柠檬酸、碱金属柠檬酸盐、吡咯磷酸盐或阴离子聚合物聚羧酸酯。
通常,乳液制剂可经处理以防止被细菌、真菌、菌质、病毒等污染或发生如氧化反应的不期望的化学反应。在优选实施方案中,可以通过加入防腐剂来实现,例如,连二硫酸钠或其它化学添加剂;或通过辐照,例如通过诸如钴60或铯37辐照的离子化辐照或通过紫外线辐照。
此外,可将活性成分加入经洗涤的油体制品中。例如,可利用目前的乳液制剂将化妆品组合物配制成稳定悬液,在护肤膏中可包括维生素和保湿剂。将一种活性成分加入本发明的乳液中的特别有益的方法是通过oleosin基因融合体的构建,详见WO96/21029。简言之,WO96/21029公开了一种以oleosin融合蛋白质方法产生蛋白质和肽的方法。这些融合蛋白质的产生是通过遗传工程将oleosin的编码基因与目标蛋白质或肽的编码基因相连接。在例如油籽植物中这种融合基因的表达导致定向于油体的融合蛋白的合成。油体级分的分离导致融合蛋白的回收。原则上任何期望的蛋白质或肽均可用这种技术生产。例如,已构思了极地鱼抗冻肽(Davies,P.L.等人,1990,FASEB J.4:2460-2468)以oleosin融合蛋白质生产。经洗涤的油体制品还可用于制备冰淇淋、高营养奶或其它带有改善的冰冻性质的冰冻食品物料,方法是抑制或防止冰晶形成。在另一个实施例中,可以oleosin融合体生产治疗用途的蛋白质。油体可用于配制一种期望的悬浮液以用于口腔用组合物或用于外部皮肤用途。本发明的这一实施方案还在本发明的11个实施例中进一步阐述,其中制备了一种包含有油体的鱼用食物,该油体中含有oleosin-锂鱼生长激素融合体。
也可配制一种带有可形成薄膜性质的乳液。这种乳液当应用于某种表面后可干燥形成一种包被层(coating)。应用了包被的油体薄膜的乳液例子是在鱼用食物中,其中将油体用于鱼用食物以增加食物值。一种可形成薄膜的乳液在本发明的实施方案中特别有用,如在递送药物或香味的易挥发物过程中需要缓慢释放一种活性成分时。在干燥过程中从乳液膜中释放活性成分的时间除了其它因素外,取决于膜的厚度。当使用较厚包被层时,干燥时间较长因而可较缓慢地释放活性成分。在多种构思的制剂中,仅当膜干燥后才释放活性成分。其它因素如乳液的组成成分以及活性成分的类型与浓度也决定了释放的特性。例如潜溶剂(如乙醇)也可包含于制剂中且影响释放时间。在食品工业中活性成分的释放也是期望的,如在消费过程中包裹在乳液中之香味的释放。依所用的具体乳液制剂,香味的释放可引起突然加强的感觉或更微妙的香气与味道的混合。
乳液制剂也可用于喷雾和气溶胶中。优选地,体积小的油体如直径1μm或1μm以下见于B.napus的那些可用于此目的。在这些喷雾中还可包括如乙醇和香气等易挥发物。这类乳液也可喷到干燥的食物制品表面,如土豆片和干汤。乳液可包括一种香料,且以可防腐的量加入,或可有助于维持食物的水分含量。乳液制剂的应用
本发明涉及用于工业用和日用组合物的乳液的制备。已注意到乳液可以用于在物理性质上和用途上有很大变化的多种组合物。这些特定的实施方案包括如在食品或饲料产品、药物产品、个人护理用品和工业用产品等应用。
食物和饲料包括非奶制代用食品(如非奶制奶酪或酸乳酪)、人造奶油、蛋黄酱、酸辣酱油、糖霜、冰淇淋、色拉调料、合成芥末、糖果、口香糖、布丁、焙烤食品、调味剂、果汁混浊剂、婴儿配方食品、香味剂、结构改进剂(起酥油)、宠物食品、鱼用食物和畜用饲料。
个人护理用品包括肥皂、化妆品、护肤膏、面膏、牙膏、口红、香水、美容品、粉底、胭脂、睫毛油、眼影、防晒乳、头发护理产品及头发染料。
药物产品可用本发明的经洗涤的油体制品配制,包括治疗剂、诊断剂和递送剂。作为治疗剂或诊断剂,乳液还含有活性成分。活性成分可以是希望递送至宿主的任何物质。在一个实施方案中,活性成分可以是有治疗或诊断价值的蛋白质或肽。这种肽包括抗原(用于疫苗制剂)、抗体、细胞因子、血液凝集因子和生长激素。本发明的乳液之工业性用途包括涂料、包被层、润滑剂、薄膜、凝胶、钻井液、纸填料、胶乳、建筑和道路建设材料、墨水、染料、蜡、擦亮剂和农用化学制剂。在优选实施方案中,本发明涉及可被动物和人摄食的组合物。因此,这些可被摄食的食物必定为食品级。应用乳液的特定产品或特定形式并不关键,且可以为期望的。显然,用经洗涤的油体制品配制的乳液可应用于任何日用或工业用产品。
本发明乳液制剂在低pH的稳定性可用于酸性乳液的配制。例如,乳液制剂可用于蛋黄酱样食品的制备,该食品中除了经洗涤的油体制品之外,如需要还含有植物油、芥末、醋和蛋黄。可浇灌的乳液如色拉调料可用增加醋的量和/或加入水来制备。
一个采用加热但无明显破坏作用的应用例子是制备例如贝夏美调味酱的香味酱或制备如巧克力调味酱的甜味酱。在这些应用中,经洗涤的油体制品被用作烹饪替代物。为制备贝夏美调味酱,向1份加热的经洗涤的油体制品中加入1份(W/W)面粉,搅拌直至形成稠的悬浊液。在中等加热下逐步加入牛奶,直到获得具有期望粘稠度的调味酱。
乳液制剂也可用于黄油替代品。在此应用中,将小量水加入经洗涤的油体制品中,例如小于10%,直至获得期望的粘度。如需要也可加入天然黄油香料和增稠剂。黄油替代品可用于甜玉米、面包、饼混合物或面包制造。提供风味及作为防腐剂的盐一般加至约2.5%(重量/体积)的水平。如需要,可加入如胭脂种子提取物或胡萝卜素的颜色以加深色泽。此用途的优点在于基于油体的黄油不含氢化脂肪酸,这种脂肪酸用于配制人造奶油且易于获得期望的一致性,但也与心血管疾病相关。
可制备多种硬度的从泡沫至可浇灌的起酥。在此用途中,将空气赶入乳液制剂中,且乳液制剂可认为被分散到连续相空气中。可将起酥用于需要奶油状和膨松状的混合物中。这些混合物包括糖霜、合成奶油、冰淇淋和面饼糊。
可从人工或天然香料和营养物制备果汁仿制品。这种仿制果汁不含有正常外观,且由于透明度看上去软或稀。通过加入例如0.1至1%(V/V)的经洗涤的油体制品或其乳液,可产生悬浊以赋予果汁丰富的外观。因而本发明油体制品可用作悬浊剂。
在另一项涉及果汁的应用中,经洗涤的油体制品或其乳液可加入带有可沉淀固体的果汁中,如蕃茄汁。加入少量经洗涤的油体制品,例如0.1至1%(V/V),可减低果汁中固体的沉降速率,有助于维持丰富的外观。
本发明的经洗涤的油体制品的局部应用也已预期。在本实施方案中,将乳液配制成皮肤学可接受的乳液,其可用于例如面部和/或身体皮肤(包括指甲和唇)的保湿,或具有抵抗皮肤衰老、痤疮、色素沉积、脱发作用,或促进毛发去除,促进伤口愈合和/或皮肤组织的重构。经洗涤的油体制品优选地占最终组合物的1-99%(重量百分比)。
本发明的化妆品组合物可含有其它烃类化合物,如植物油、动物油、矿物油或合成油、或蜡或其混合物。它们包括石蜡、凡士林油、全氢化角鲨烯、arara油、杏仁油、calphyllum油、鳄梨油、芝麻油、蓖麻油、希蒙得木油、橄榄油或谷物胚油。酯可包括如羊毛酸酯、油酸酯、月桂酸酯、硬脂酸酯、十四烷酸酯。也可包括多种醇,例如油酰乙醇、亚油酰乙醇或亚麻酰乙醇、异硬脂酰乙醇或辛酰十二烷醇、乙醇或多醇。还可包括的烃类有辛酸酯、癸酸酯、蓖麻酸酯、辛酸甘油三酯或癸酸甘油三酯,或C10至C22脂肪酸甘油三酯。这些试剂的加入可导致双重乳液的形成。
在25℃固化的氢化油也可包含在本发明的化妆品制剂中,诸如氢化蓖麻油、棕榈油或椰子油或氢化牛油;甘油单酯、甘油二酯、甘油三酯或甘油蔗糖酯;羊毛脂;以及在25℃固化的脂肪酸。可被包括的蜡有动物蜡,如蜂蜡;植物蜡,如巴西棕榈蜡、小烛树蜡、小冠巴西棕蜡、木蜡或来自软木纤维或甘蔗的蜡;矿物蜡,如石蜡、褐煤蜡、微晶蜡或地蜡及合成蜡。
可以包括颜料,可为白色或有颜色的,无机的或有机的,和/或珠光颜料。这些颜料包括二氧化钛、氧化锌、二氧化锆、黑铁氧化物、黄铁氧化物、红铁氧化物和褐铁氧化物、二氧化铈、氧化铬、铁蓝、碳黑、钡、锶、钙和铝沉淀色料,以及包被有氧化钛或氧化铋的云母。
通常用于护肤膏中的活性成分有如维生素,例如维生素A或C;和α-羟酸,如柠檬酸、乙醇酸、乳酸和酒石酸均可包含于化妆品及/或皮肤学组合物中。例如,美国专利5,602,183教导维生素C或抗坏血酸促进结缔组织生长,特别是用于加强皮肤强度以及用于使皮肤抵抗外部侵袭,诸如烟雾和紫外线照射。可包括在护肤膏和化妆品中的保湿剂有例如矿物油和尿素。也可添加抗氧化剂如天然存在的生育酚和多酚,或丁基化羟基甲苯及羟基甲苯醚。诸如辛基甲氧基肉桂酸(Parsol MCX),3-二苯铜(Uvinul M40)和丁基甲氧基二苯甲酰甲烷(Parsol 1789)的防晒剂也可用于制备防晒液。可用于配制化妆品组合物的药物活性成分包括例如抗生素、抗真菌剂和抗炎症剂。
最终化妆品的形式可以为游离的、可浇灌的或密实的粉末(粉底、胭脂或眼影),相对较油的产品如口红、睫毛油或用于身体或面部的油或乳液。
经洗涤的油体制品也可用于作为牙膏中的一种口腔可接受的载体,其还可包含硅、表面活性剂、鳌合剂、氟化物、增稠剂、增甜剂、食用香料如薄荷油、酶和杀虫剂。
可配制的工业用产品的例子有涂料,其中主要的树脂如基于硅类的化合物、丙烯酸基化合物、聚酯、醇酸树脂、氟、环氧化合物、聚氨基甲酸乙酯可部分或全部被本发明的经洗涤的油体制品替代。此外,添加剂,如颜料、染料、玻璃薄片和铝片、颜料分散剂、增稠剂、匀染剂、硬化催化剂、硬化剂如异氰酸酯(dioisocyanate)、硬化催化剂、凝胶抑制剂、紫外线吸收剂、游离基淬灭剂等如需要也可配制入涂料组合物。
经洗涤的油体制品也可被配制成润滑剂。例如,经洗涤的油体制品可用于部分或完全替代润滑油,如动物油、植物油、石油润滑油、合成润滑油或者润滑膏,如锂脂膏、尿脂膏和钙脂膏。其它用于润滑制剂的组合物包含抗氧化剂、去污剂分散剂、油性剂、磨擦调节剂、粘度系数增强剂、倾点下降剂、固体润滑材料、防锈剂和消泡剂。
也可用本发明的经洗涤的油体制品制备蜡。这些蜡包括清洗蜡型,如那些向汽车上提供稳定疏水性薄膜涂饰剂的蜡和其它保护性包被层。其它用于蜡制品的组合物包括表面活性剂、矿物油如混合石蜡和芳香油和/或环烷烃(naphtenic)油、香水、杀虫剂、着色剂,其可依需要加入适当的量。
当配制工业用产品如涂料或润滑剂时,油体相的纯度并不关键,可以不必洗涤油体。工业用乳液可通过如下方式制备:1)从细胞中获得油体;和2)将油体配制入工业用乳液。油体可由以下方式获得:a)研磨植物种子;b)从经研磨的种子中除去固形物;和c)从水相中分离油体相。本发明也包括一种工业用乳液,其含有依本发明制备的油体。
下列非限制性实施例用于阐明本发明。实施例实施例1从油籽油菜、大豆、向日葵、白芥末、花生、南瓜、亚麻、红花和玉米中获得经洗涤的油体制品(实验室规模)
获自油籽油菜(Brassica napus cv Wester)、大豆、向日葵、白芥末、花生、南瓜、亚麻、红花和玉米的干燥成熟的种子在5倍体积的冷研磨缓冲液(50mM Tris-HCl,pH7.5,0.4M蔗糖和0.5M NaCl)中匀浆,使用polytron在高速进行。以10Xg将匀浆物离心30分钟,以除去颗粒物且将油体从含有大量可溶性种子蛋白质的水相中分离出来。用金属刮刀将油体级分从上清液的表面撇出,加入到1倍体积的研磨缓冲液中。为了在随后步骤中实现高效洗涤,发现需要将油体在研磨缓冲液中充分的重新分散。这可以通过利用低速的polytron在研磨缓冲液中温和匀浆油体来实现。利用注射器,将重新分散的油体小心地置于5倍体积的冷缓冲液下层(50mM Tris-HCl,pH7.5),如上述离心。离心后,移走油体,将洗涤步骤重复两次。最终经洗涤的油体制品重悬于1倍体积的冷缓冲液(Tris-HCl,pH7.5)中,用polytron重新分散。
油体样品溶于SDS样品缓冲液,经SDS凝胶电泳分析。结果示于图1。
如此获得的物料可方便地用于多种制剂中。实施例2大规模从油籽油菜、向日葵和玉米中获得经洗涤的油体制品
本实施例描述了从canola、向日葵和玉米种子中大规模回收油体级分。所得制品含有完整的油体,且其纯度类似于用实验室规模获得的制品。种子研磨
通过胶体磨(Colloid Mill,MZ-130(Fryma);加工能力:500kg/小时)的料斗加入总量为10-15kg的干燥油籽油菜(canola)种子(Brasscia napus cv Wester)、向日葵(Helianthus annuus)或玉米(Zeamay),该磨装备有MZ-120交叉齿形转动盘/固定盘研磨装置和顶部加料料斗。在研磨前由外接水管加入约50-70升水。磨操作设在间距为1R,用于在18℃至30℃下获得小于100微米的颗粒。种子研磨后,向种子浆中加入自来水至终体积90升。固形物去除
所得种子浆液泵入操作速度达到3,500转/分以上的倾析式离心机(Hasco 200两相倾析式离心机,最大操作速度6,000转/分),用1英寸MZ Wilden气动双隔膜泵使浆液以360升/小时的流速从磨转移到倾析式离心机。在15-20分钟内约15公斤种子被沉淀分取处理。油体分离
油体级分的分离利用管式离心机型号AS-16(Alpha Laval)完成,该离心机装配有三相分离碟及可拆卸环形挡料圈系列;加工能力:150升/小时;环形挡料圈:30mm。操作速度为15,000转/分(13,200Xg)。在离心机达到操作速度后,使用Watson-Marlow(704型)蠕动泵将倾倒出的液体相(DL)泵入管式离心机。这使倾倒出的液体相被分成含有水和可溶性种子蛋白质的重相(HP)以及含有油体的轻相(LP)。在经过一次离心机后所获的油体级分称为未经洗涤的油体制品。然后将油体级分再通过离心机处理三次。每次通过离心机处理之前,浓缩的油体与约5倍体积的新鲜水混合。全部过程在室温下进行。在第二次分离后获得的制品均称为经洗涤的油体制品。经三次洗涤后除去了大部分杂质可溶性蛋白质,经SDS凝胶电泳获得的油体蛋白质图样与用实验室规模的方法获得那些有相似的图案。实施例3通过洗涤油体相除去种子蛋白质
本实施例描述从油籽油菜、玉米和向日葵种子中回收经洗涤的油体级分。利用不同的洗涤条件,可见洗涤从油体制品中除去了大量的种子蛋白质。这些蛋白质包括可能致敏的蛋白质。

完成4次洗涤后取0.9ml油体制品样本在0.1ml 1M Na2CO3中匀浆,室温下搅拌30分钟。经洗涤的油体级分经过离心后回收,用水洗一次,用于SDS凝胶电泳。
全部样品溶于SDS样品缓冲液,用SDS凝胶电泳分析样品。结果示于图2。实施例4洗涤油体相对于水保持特性的影响
从油菜籽中如实施例2所述制备经洗涤的油体制品和未经洗涤的油体相。为了测定未经洗涤的油体相和经洗涤的油体制品之间水保持能力的差异,用搅拌器彻底混合30ml油体制品。然后分别在40℃、60℃或80℃水浴中温育制品两小时,将样品在1,500Xg下离心20分钟(未稀释的样品)。通过15g经洗涤的或未经洗涤的油体制品与15ml水的混合制备另一套样品。将样品在搅拌器上混合后分别于40℃、60℃或80℃温育两小时,于1,500g离心20分钟后测定样品中的水含量(稀释的样品)。由于蒸发损失的质量在80℃和60℃情况下测量。
80℃时,含油体的未稀释制品通过蒸发损失了大量水。未经洗涤的油体制品的质量损失了26%,而经洗涤的制品损失了16%。经离心,未经洗涤的制品释放了约2.5ml水相,而经洗涤的油体保持在同一相中。两种稀释的制品均吸收水。在两种情况中油体体积增加到18.5±1ml。
60℃时,未稀释的制品通过蒸发损失了约10%的水。离心后,未经洗涤的制品释放了约0.5ml的水相,而经洗涤的油体制品仍保持同一相。两种稀释的制品均吸收水。60℃时,两种情况中油体的体积增加到18±1ml。
40℃时,未稀释的样品均释放了约2ml水相。当比较稀释的样品时,未经洗涤的制品吸收了约3ml水,类似于在60℃或80℃时的情况。然而,在40℃时经洗涤的制品吸收了8ml水。
这些实验显示,在加热到60℃或80℃的经洗涤的油体制品中,与未经洗涤的制品相比,水与油体制品保持更紧密的结合。当冷却后,经洗涤的油体制品似乎比未经洗涤的乳液更为稳定。当加热到40℃,经洗涤的油体制品可吸收较多量的外源加入的水,而不会导致相分离。实施例5洗涤油体对于油吸收特性的影响
如实施例2所述,从油菜籽制备经洗涤的油体制品和未经洗涤的油体相。为测定未经洗涤的油体相与经洗涤的油体制品之间油吸收能力的差异,将2克油体制品分散到50ml试管中的12ml精炼、漂白及去臭的油菜籽(canola)油中。将混合物每隔5分钟搅拌30秒,共进行30分钟。然后将试管以4,400转/分离心25分钟。倾倒游离的油,通过重量差异确定吸收的油之百分比。检测了3种经洗涤的油体制品,和3种未经洗涤的油体制品。
发现未经洗涤的油体之油吸收能力在3个批次中变化很大,从18.7%到28%。而经洗涤的油体制品具有可重复的油吸收,为32±1%。因此,认为经洗涤的油体制品较好,因为1)可提供更大的油吸收量;且2)以更为可重复的方式进行吸收。实施例6包含经洗涤的油体制品的蛋黄酱样乳液的制备
如实施例2所述从油菜籽中制备经洗涤的油体制品,运用家用电动搅拌机混合下列组分制备蛋黄酱样乳液:向日葵油 78克蛋黄 8克醋 9克盐 0.5克经洗涤的油体 5克
获得了具有蛋黄酱样质地的产品。该蛋黄酱样产品在4℃下至少稳定1天。实施例7不含胆固醇的蛋黄酱样乳液的制备
如实施例2从油菜籽中制备经洗涤的油体制品,通过混合下列成分制备蛋黄酱样乳液:向日葵油 200克经洗涤的油体 100克醋 30ml
获得了具有蛋黄酱样质地的产品。由于制备的蛋黄酱不含蛋黄(在市售蛋黄酱中通常采用的一种成分),用经洗涤的油体制备的产品不含胆固醇。当利用离心鉴定稳定性时此蛋黄酱与市售蛋黄酱一样稳定。实施例8包含经洗涤的油体制品的酸辣酱油样产品的制备
如实施例2从油菜籽制备经洗涤的油体制品,通过手工混合下列成分产生酸辣酱油样乳液:向日葵油 17.5克芥末 0.4克醋 0.5克经洗涤的油体 7.7克

获得贝夏美调味酱样调味汁。此调味汁的一致性可如加入的牛奶量所定。也可在需要时加入其它香料。此产品中无氢化脂肪酸使其优于从市售家用麦吉林(人造奶油)制备的调味汁。实施例11用于包被鱼用食物的药物乳液的制备
按以下方法获得来自表达融合于oleosin的鲤鱼生长激素(cGH)的转基因油籽油菜(B.napus)植物之经洗涤的油体制品,其中融合蛋白定向于油体。利用聚合酶链式反应结合两个cGH特异性引物,从含有普通鲤鱼(Cyprinus carpio)生长激素cDNA的插入片段之质粒中扩增DNA片段,该DNA片段中含有缺少其22个氨基酸信号序列的cGH编码区(Koren等人,1989,基因,67:309-315)。使用pOThromb(VanRooijen,1993,博士论文,Calgary大学)作为亲本质粒,采用本领域技术人员已知的克隆策略将扩增的cGH片段以正确的读框融合,位于拟南芥oleosin的3’端。在pOTthromb中一个凝血酶切割位点构建于oleosin编码序列的3′侧。将oleosin-cGH融合基因导入二元载体pCGN1559(Mc Bride和Summerfelt,1990,植物分子生物学,14:269-276),所得构建体用于转化根瘤土壤杆菌(A.tumefaciens)。利用土壤杆菌菌株转化B.napus cv Wester的幼苗。获得了来自转基因植物的种子,如实施例1所述从转基因种子分离油体。
然后将油体吸入注射器中,用大约每毫克鱼用食物2.5微克油体蛋白的量喷到鱼用食品上。用油体包被的鱼用食品过夜晾干。然后将总量为50mg的鱼用食品与10ml水混合,温育0、30、45或60分钟。然后收集食物,重悬于0.2毫升50mM Tris-HCl(pH7.5)中,在2.5%SDS中煮沸后用于SDS凝胶电泳分析。在鱼用食品中存在的油体利用Western杂交和针对cGH的单克隆抗体来评测。
通过在Western杂交的单一泳道上观察的单一条带信号之强度判断,油体在水中温育后,含有cGH的油体仍与鱼用食品保持紧密结合。在水中温育了30、45或60分钟的鱼用食品显示出与未在水中温育的对照鱼用食品相比基本上相同的cGH的量。
本实施例显示,可制备赋予乳液特定期望性质的转基因植物变种。实施例还显示,可以从可用作包被层或薄膜的经洗涤的油体制品制备乳液。最后,本实施例还显示,经洗涤的油体制品可用于配制药用组合物。
虽然已描述了某些特别优选的实施例以阐述本发明,对于本领域普通技术人员而言,落在本发明的范围之中的其它应用是显而易见的。
Claims (42)
1.一种用于制备乳液制剂的方法,包括:
(1)从细胞中获得油体;
(2)洗涤油体以获得经洗涤的油体制品;和
(3)将经洗涤的油体制品配制成乳液。
2.根据权利要求1的方法,其中所述细胞是植物细胞。
3.根据权利要求2的方法,其中所述植物细胞获自孢子、花粉、种子或营养性植物器官。
4.根据权利要求2的方法,其中油体获自植物种子。
5.一种用于制备乳液制剂的方法,包括:
(1)通过如下方法从植物种子中获得油体:
(a)研磨植物种子;
(b)从经研磨的种子中除去固形物;以及
(c)从水相中分离油体相;
(2)洗涤油体相以获得经洗涤的油体制品;和
(3)将经洗涤的油体制品配制成乳液。
6.根据权利要求5的方法,其中在研磨步骤之前或之后加入液体相。
7.根据权利要求1-6中任一项的方法,其中所述配制包括混合经洗涤的油体直至形成乳液。
8.根据权利要求7的方法,其中在经洗涤的油体中加入液体相,且将液体相与经洗涤的油体混合直至形成乳液。
9.根据权利要求6或8的方法,其中所述液体相是水。
10.根据权利要求9的方法,其中所述液体相在乳液中存在的量从约1%至约99%v/v。
11.根据权利要求4或5的方法,其中所述植物为油料种子植物。
12.根据权利要求4或5的方法,其中所述植物选自油菜籽(芸苔属)、大豆、向日葵、油棕榈、棉籽(棉属)、花生、椰子、蓖麻、红花、芥末(芸苔属和欧白芥)、芫荽、南瓜、亚麻、巴西果、希蒙得木和玉米。
13.根据权利要求5的方法,其中所述固形物通过离心或过滤所述经研磨的种子而除去。
14.根据权利要求5的方法,其中通过基于重力的方法或基于体积排阻的方法将所述油体相从所述水相中分离。
15.根据权利要求5的方法,其中经洗涤的油体制品基本上不含非油体种子蛋白质、抗营养性化合物、淀粉、芥子油甙或其降解产物和纤维。
16.根据权利要求15的方法,其中所述种子蛋白质是糖基化的。
17.根据权利要求1-16中任一项的方法,其中所述配制包括加入防腐剂。
18.根据权利要求1-16中任一项的方法,其中所述乳液是食品或饲料产品、个人护理用品、药物产品或工业用产品。
19.根据权利要求5的方法,其中在研磨前种子在一种液体相中浸泡约15分钟至约2天。
20.一种包含经洗涤的油体制品的乳液。
21.根据权利要求20的乳液,其还包含一种液体相。
22.根据权利要求21的乳液,其中所述液体相是水。
23.根据权利要求20的乳液,其还包含一种防腐剂。
24.根据权利要求20的乳液,其中油体获自植物。
25.根据权利要求24的乳液,其中油体获自下列植物:油菜籽(芸苔属)、大豆、向日葵、油棕榈、棉籽(棉属)、花生、椰子、蓖麻、红花、芥末(芸苔属和欧白芥)、芫荽、南瓜、亚麻、巴西果、希蒙得木和玉米。
26.根据权利要求20-25中任一项的乳液,其中所述乳液为食品、个人护理用品、药物产品或工业用产品。
27.根据权利要求26的乳液,其中所述食品或饲料产品选自非奶制代用食品、非奶制奶酪、非奶制酸乳酪、人造奶油、蛋黄酱、酸辣酱油、糖霜、冰淇淋、色拉调料、合成芥末、糖果、口香糖、布丁、焙烤食品、调味剂、果汁、婴儿配方食品、香味剂、结构改进剂、宠物食品和畜用饲料。
28.根据权利要求26的乳液,其中所述食品或饲料产品是蛋黄酱、无胆固醇的蛋黄酱、贝夏美调味汁、酸辣酱油、芥末或鱼用食物。
29.根据权利要求26的乳液,其中所述个人护理用品选自肥皂、化妆品、护肤膏、面膏、牙膏、口红、香水、美容品、粉底、胭脂、睫毛油、眼影、防晒乳和头发护理产品。
30.根据权利要求26的乳液,其中所述药物产品选自治疗剂、诊断剂和递送剂。
31.根据权利要求30的乳液,其中所述治疗剂中包括生长激素。
32.根据权利要求26的乳液,其中所述工业用产品选自涂料、包被层、润滑剂、薄膜、凝胶、钻井液、纸填料、胶乳、建筑或道路建设材料、墨水、染料、蜡、擦亮剂和农用化学制剂。
33.根据权利要求1-19中任一项制备的乳液的用途,其用于制备个人护理用品、食品或饲料产品、工业用产品或药物产品。
34.根据权利要求33的乳液的用途,其中所述食品或饲料产品包括非奶制代用食品、非奶制奶酪、非奶制酸乳酪、人造奶油、蛋黄酱、酸辣酱油、糖霜、冰淇淋、色拉调料、合成芥末、糖果、口香糖、布丁、焙烤食品、调味剂、果汁、婴儿配方食品、香味剂、结构改进剂、宠物食品和畜用饲料。
35.根据权利要求33的乳液的用途,其中所述食品或饲料产品是蛋黄酱、无胆固醇的蛋黄酱、贝夏美调味汁、酸辣酱油、芥末或鱼用食物。
36.根据权利要求33的乳液的用途,其中所述个人护理用品包括肥皂、化妆品、护肤膏、面膏、牙膏、口红、香水、美容品、粉底、胭脂、睫毛油、眼影、防晒乳和头发护理产品。
37.根据权利要求33的乳液的用途,其中所述工业用产品包括涂料、包被层、润滑剂、薄膜、凝胶、钻井液、纸填料、胶乳、建筑或道路建设材料、墨水、染料、蜡、擦亮剂和农用化学制剂。
37.根据权利要求33的乳液的用途,其中所述药物产品包括治疗剂、诊断剂和递送剂。
39.根据权利要求1-19中任一项制备的乳液的用途,其用作包被层、薄膜或气溶胶。
40.一种用根据权利要求20-25中任一项的乳液包被的鱼用食物制品。
41.根据权利要求40的鱼用食物制品,其中所述乳液还额外含有治疗性的肽。
42.根据权利要求41的鱼用食物制品,其中所述治疗性肽为激素。
