背景技术
近几年来,已具有显著成功的努力,以发现通过刺激免疫系统的某些关键方面以及通过抑制某些其它方面(参见例如,美国专利号6,039,969(Tomai等人)和6,200,592(Tomai等人)而起作用的新药物化合物。这些化合物,在本文中被称为免疫应答调节剂(IRM),似乎通过称为Toll样受体(TLR)的基本免疫系统机理诱导选定的细胞因子生物合成、共刺激分子的诱导以及增加抗原递呈能力而起作用。
许多IRM可用于治疗多种疾病和病症。例如,某些IRM可用于治疗病毒性疾病(如,人乳头状瘤病毒、肝炎、疱疹)、肿瘤(如,基底细胞癌、鳞状上皮细胞癌、光化性角化症、黑素瘤)、TH2-介导的疾病(如哮喘、过敏性鼻炎、特应性皮炎)、以及自体免疫疾病。

发明内容

在反应方案I的步骤(2)中,使式V的化合物在环境温度下与N-羟基琥珀酰亚胺和标准偶联试剂反应,所述标准偶联试剂例如合适溶剂例如二氯甲烷或吡啶中的1,3-二环己基碳二亚胺(DCC)或1-[3-(二甲基氨基)丙基]-3-乙基碳二亚胺(EDC)。式VI的产物可使用常规方法分离。
在反应方案I的步骤(3)中,使式VII的肼基苯甲酸或肼基烟酸化合物在环境温度下与丙酮反应,以提供式VIII的腙取代的化合物。起始的式VII的肼取代的化合物为3-肼基苯甲酸(其中A=CH的VII)和5-肼基烟酸(其中A=N的VII)。这些化合物可根据提供的参考文献中的工序制备,以制备式IV的化合物。
在反应方案I的步骤(4)中,使用例如上文对于步骤(2)描述的条件,使式VIII的化合物在环境温度下与N-羟基琥珀酰亚胺反应。
反应方案I
本发明的化合物可根据反应方案II制备,其中R、R2、A和n如上定义,并且X′为具有至多8个碳原子的亚烷基。
在反应方案II的步骤(1)中,将式X的4-氯-3-硝基喹啉还原,以提供式XI的3-氨基-4-氯喹啉。还原可使用常规异质氢化催化剂例如碳载铂或碳载钯进行。对于式X的一些化合物,例如,其中R为卤素的化合物,铂催化剂是优选的。该反应可使用Parr设备在合适的溶剂例如甲苯和/或异丙醇中进行。该产物可通过常规方法分离。式X的许多化合物是已知的,或者可使用已知合成方法制备,参见例如美国专利号4,689,338(Gerster);5,175,296(Gerster);5,367,076(Gerster);和5,389,640(Gerster等人);以及其中引用的文件。式XI的一些化合物是已知的。例如,3-氨基-4-氯喹啉、3-氨基-4,5-二氯喹啉和3-氨基-4,7-二氯喹啉已通过Surrey等人Journal of theAmerican Chemical Society《美国化学会志》,73,第2413-2416页(1951)制备。
作为另外一种选择,还原步骤(1)可使用单相或二相连二硫酸钠还原进行。该反应使用由Park,K.K.;Oh,C.H.;和Joung,W.K.Tetrahedron Letters《四面体通讯》,34,第7445-7446页(1993)描述的条件进行,在存在碳酸钾和乙基紫精二溴化物、乙基紫精二碘化物、或1,1’-二-正辛基-4,4’-联吡啶二溴化物的情况下,在环境温度下将连二硫酸钠添加到在二氯甲烷和水的混合物中的式X的化合物中。该产物可使用常规方法分离。
在反应方案II的步骤(2)中,使式XI的3-氨基-4-氯喹啉与式R2C(O)Cl或R2C(O)Br的酰基卤反应,以提供式XII的N-(4-氯喹啉-3-基)酰胺。在存在碱例如三乙胺、吡啶、或4-二甲基氨基吡啶的情况下,将酰基卤添加到合适溶剂例如二氯甲烷中的式XI化合物溶液中。该反应可在低温例如0℃下或在环境温度下运行。该产物可通过常规方法例如重结晶分离。
在反应方案II的步骤(3)中,使式XII的N-(4-氯喹啉-3-基)酰胺与式CBZ-NH-X’-ONH2的羟胺反应并环化,以提供式XIII的1H-咪唑并[4,5-c]喹啉。CBZ为对于羧基苄基基团的通用化学缩写。将CBZ-NH-X’-ONH2添加到醇溶剂中的式XII化合物溶液中。该反应可在高温下例如在回流温度下在异丙醇中进行。还可将碱例如三乙胺添加到反应中。该产物可通过常规方法例如重结晶分离。
在反应方案II的步骤(3)中使用的CBZ-NH-X’-ONH2化合物可通过使式H2N-X’-OH的氨基烷基醇与氯甲酸苄酯反应来制备,以提供式CBZ-NH-X’-OH的化合物。在存在碱例如吡啶的情况下,将氯甲酸苄酯添加到合适溶剂例如二氯甲烷中的式H2N-X’-OH溶液中。该反应初始可在低温例如0℃下运行,随后缓慢升温至环境温度。该产物可通过常规方法例如重结晶分离。式CBZ-NH-X’-OH的化合物还使用光延反应(Mitsunobu Reaction)条件进行反应,以提供式CBZ-NH-X’-ONH2的化合物。将N-羟基邻苯二甲酰亚胺、式CBZ-NH-X’-OH的化合物和三苯基膦在合适的溶剂例如二氯甲烷中混合,并且冷却至0℃。缓慢添加偶氮二甲酸二异丙酯(D1AD),并且使反应升温至环境温度。如果需要的话,反应可在高温例如60℃下进行。在减压下浓缩后,通过在合适溶剂例如乙醇中用肼(水溶液)处理来去除邻苯二甲酰亚胺保护基团。式CBZ-NH-X’-ONH2的产物可通过常规方法例如重结晶分离。
在反应方案II的步骤(4)中,使式XIII的1H-咪唑并[4,5-c]喹啉进行三次转化,以提供式XIV的1H-咪唑并[4,5-c]喹啉-4-胺。首先,使用常规氧化剂将式XIII的化合物氧化,所述常规氧化剂能够形成吡啶环中的氮原子的N-氧化物。该反应通过用3-氯过氧苯甲酸处理合适溶剂例如氯仿或二氯甲烷中的式XIII化合物溶液进行。可任选地分离N-氧化物反应产物,或可在相同反应容器中用原位形成的N-氧化物进行二次转化。在二次转化中,通过用合适的活化剂活化N-氧化物,随后用合适的胺化剂胺化,将N-氧化物产物胺化。合适的活化剂包括烷基或芳基磺酰氯(例如苯磺酰氯、甲磺酰氯和对甲苯磺酰氯)。芳基磺酰氯(例如对甲苯磺酰氯)在一些实施例中是有用的。合适的胺化剂包括氨(例如以氢氧化铵的形式)和铵盐(例如碳酸铵、碳酸氢铵、磷酸铵)。该反应可通过以下进行:将式XIII的化合物的N-氧化物溶解于合适的溶剂例如二氯甲烷、1,2-二氯乙烷、或氯仿中,并且添加氢氧化铵,随后添加芳基磺酰氯。该反应可任选地伴随加热进行。任选地,该反应可在密封的压力容器中在高温(85-100℃)下进行。在三次转化中,CBZ保护基团在酸性条件(例如在高温下的浓盐酸)下去除,以提供式XIV的化合物。式XIV的化合物可作为游离碱或作为酸盐(例如马来酸盐或延胡索酸盐)通过常规方法分离。
在反应方案II的步骤(5)中,使式XIV的化合物与式V的化合物(来自其中A=CH或N的反应方案I)反应,以提供式XV的化合物。该反应可在环境温度下在溶剂例如二氯甲烷、吡啶、或1-丁醇中用标准偶联试剂进行,所述标准偶联试剂例如1,3-二环己基碳二亚胺(DCC)或1-[3-(二甲基氨基)丙基]-3-乙基碳二亚胺(EDC)。式XV的化合物可使用常规方法分离。作为反应方案II的步骤(5)的替代方法,使式XIV的化合物与式VI的化合物(来自其中A=CH或N的反应方案I)反应,以提供式XV的化合物。式XIV的化合物可溶解于合适的醇溶剂例如1-丁醇中,并且式VI的化合物可在环境温度下缓慢添加。
在反应方案II的步骤(6)中,乙酰胺(acetamine)保护基团在酸性条件下去除,以提供式XVI的化合物。该反应可在盐酸中在环境温度或高温(例如60℃)下进行。式XVI的产物可例如作为盐酸盐通过冻干分离。
在反应方案II的步骤(7)中,根据步骤(5)中所述的相应工序,使式XIV的化合物与式VIII或式IX的化合物(来自其中A=CH或N的反应方案I)反应,以提供式XVII的化合物。
在反应方案II的步骤(8)中,乙酰胺保护基团在酸性条件下去除,以提供式XVIII的化合物。该反应可根据步骤(6)中所述的工序进行。
反应方案II
本发明的化合物可根据反应方案III制备,其中R、R2、A和n如上定义,并且X′为具有至多8个碳原子的亚烷基。
在反应方案III的步骤(1)中,用式HO-X’-NH2的胺处理式X的4-氯-3-硝基喹啉,以提供式XIX的化合物。多种式HO-X’-NH2的胺是可商购获得的,并且其它的可通过已知的合成方法制备。在存在叔胺例如三乙胺的情况下,通过将式HO-X’-NH2的胺添加到合适溶剂例如二氯甲烷中的式X的4-氯-3-硝基喹啉溶液中,进行该反应。该反应可在环境温度下或在亚环境温度例如0℃下进行。反应产物可使用常规方法分离。
在反应方案III的步骤(2)中,将式XIX的化合物还原,以提供式XX的二胺。该反应可例如使用反应方案II的步骤(1)中所述的方法进行。
在反应方案III的步骤(3)中,使式XX的二胺与羧酸等同物反应,以提供式XXI的1H-咪唑并[4,5-c]喹啉。合适的羧酸等同物包括式R2C(O-烷基)3的原酸酯、式R2C(O-烷基)2(O-C(O)-烷基)的1,1-二烷氧基烷基链烷酸酯、以及式R2C(O)Cl的酰氯。羧酸等同物的选择通过在R2处的所需取代基确定。例如,原甲酸三乙酯将提供其中R2为氢的化合物,并且原丁酸三甲酯将提供其中R2为丙基基团的化合物。
步骤(3)可通过将羧酸等同物添加到合适溶剂例如甲苯或二甲苯中的式XX的二胺中进行。任选地,可添加催化盐酸吡啶。该反应通常在足够高的温度下进行,以驱除在该反应期间形成的醇或水。便利地,Dean-Stark捕集器可用于收集挥发物。式XXI的1H-咪唑并[4,5-c]喹啉产物可使用常规技术分离并任选地纯化。作为另外一种选择,当式R2C(O)Cl的酰氯用作羧酸等同物时,反应方案III的步骤(3)可在两个步骤中进行。步骤(3)的部分(i)可通过将酰氯添加到合适溶剂例如二氯甲烷或乙腈中的式XX的二胺溶液中进行。任选地,可添加叔胺例如三乙胺、吡啶、或4-二甲基氨基吡啶。该反应可在环境温度下进行。酰胺产物可使用常规技术分离并任选地纯化。步骤(3)的部分(ii)涉及在存在碱的情况下加热部分(i)中制备的酰胺,以提供式XXI的1H-咪唑并[4,5-c]喹啉。在存在碱例如氢氧化钠或含水碳酸钾的情况下,该反应可在合适的溶剂例如乙醇中在高温下进行。式XXI的产物可使用常规方法分离。
其中n为0的式XXI的多种化合物是已知的,并且已通过其它相关途径制备;参见例如美国专利号4,689,338(Gerster)、5,605,899(Gerster等人)和5,175,296(Gerster)。
在反应方案III的步骤(4)中,在光延反应条件下用N-羟基邻苯二甲酰亚胺处理式XXI的羟基取代的化合物,以提供式XXII的受N-邻苯二甲酰亚胺保护的羟胺。该反应可通过以下进行:将三苯基膦和N-羟基邻苯二甲酰亚胺添加到合适溶剂例如四氢呋喃或N,N-二甲基甲酰胺中的式XXI的醇溶液中,然后缓慢添加偶氮二甲酸二异丙酯(DIAD)。该反应可在环境温度下或在高温例如60℃下进行。该产物可使用常规方法分离。
在反应方案III的步骤(5)和(6)中,使用能够形成N-氧化物的常规氧化剂,将式XXII的受N-邻苯二甲酰亚胺保护的羟胺氧化,以提供式XXIII的1H-咪唑并[4,5-c]喹啉-5N-氧化物,然后将式XXIII的1H-咪唑并[4,5-c]喹啉-5N-氧化物胺化,以提供式XXIV的1H-咪唑并[4,5-c]喹啉-4-胺。该反应可使用反应方案II的步骤(4)中的一次和二次转化中所述的条件进行。在这些条件下,去除N-邻苯二甲酰亚胺保护基团,以提供式XXIV的1H-咪唑并[4,5-c]喹啉-4-胺。式XXIV的化合物是已知的,并且它们的其它制备方法已有所描述;参见例如美国专利号7,648,997(Kshirsagar等人)。
在反应方案III的步骤(7)中,使式XXIV的化合物与式V或VI的化合物(来自其中A=CH或N的反应方案I)反应,以提供式XXV的化合物。该反应可根据反应方案II的步骤(5)中所述的相应方法进行。
在反应方案III的步骤(9)中,根据反应方案II的步骤(5)中所述的相应工序,使式XXIV的化合物与式VIII或式IX的化合物(来自其中A=CH或N的反应方案I)反应,以提供式XXVII的化合物。
在反应方案III的步骤(8)和(10)中,如反应方案II的步骤(6)中所述,在酸性条件下去除乙酰胺保护基团,以分别提供式XXVI和XXVIII的化合物。
反应方案III
本发明的化合物可根据反应方案IV制备,其中R、R2、A和n如上定义,BOC为叔丁氧羰基,并且Y和Z为具有总计至多8个碳原子的亚烷基基团。
在反应方案IV的步骤(1)中,用叔丁氧羰基基团(BOC)保护式XXIX的氨基醇的氨基基团,以提供式XXX的化合物。在存在碱例如氢氧化钠的情况下,四氢呋喃中的氨基醇溶液可用二碳酸二叔丁酯处理。许多式XXIX的氨基醇是可商购获得的,并且其它的可使用已知的合成方法制备。
在反应方案IV的步骤(2)中,将式XXX的受保护的氨基醇转变为式XXXI的碘化物。碘可添加到二氯甲烷中的三苯基膦和咪唑溶液中,然后可添加二氯甲烷中的受保护的氨基醇XXX溶液。该反应可在环境温度下进行。式XXXI的化合物可使用常规方法分离。
在反应方案IV的步骤(3)中,式XXXII的1H-咪唑并[4,5-c]喹啉-1-基醇用式XXXI的碘化物烷基化,以提供式XXXIII的1H-咪唑并[4,5-c]喹啉-1-基醚。式XXXII的1H-咪唑并[4,5-c]喹啉-1-基醇表示在反应方案III中由式XXI表示的化合物子集。式XXXII的化合物可使用对于反应方案III中的式XXI化合物的合成描述的工序制备。在步骤(3)中,式XXXII的醇可与合适溶剂例如N,N-二甲基甲酰胺中的氢化钠反应,以形成醇盐。碘化物在环境温度下添加到醇盐溶液中,然后在高温(大约100℃)下搅拌。
在反应方案IV的步骤(4)中,将式XXXIII的1H-咪唑并[4,5-c]喹啉-1-基醚氧化,以提供式XXXIV的1H-咪唑并[4,5-c]喹啉-5N-氧化物。在反应方案IV的步骤(5)中,将式XXXIV的1H-咪唑并[4,5-c]喹啉-5N-氧化物胺化,以提供式XXXV的1H-咪唑并[4,5-c]喹啉-4-胺。该反应可使用反应方案II的步骤(4)中的一次和二次转化中所述的条件进行。
在反应方案IV的步骤(6)中,通过在酸性条件下的水解去除BOC保护基团,以提供式XXXVI的1H-咪唑并[4,5-c]喹啉-4-胺。式XXXV的化合物可在环境温度下或伴随微热在乙醇中用盐酸处理。其中n为0的式XXXVI的化合物是已知的,并且制备和分离方法已有所描述;参见例如美国专利号6,660,747(Crooks等人)。
在反应方案IV的步骤(7)中,使式XXXVI的化合物与式V或VI的化合物(来自其中A=CH或N的反应方案I)反应,以提供式XXXVII的化合物。该反应可根据反应方案II的步骤(5)中所述的相应方法进行。
在反应方案IV的步骤(9)中,根据反应方案II的步骤(5)中所述的相应工序,使式XXXVI的化合物与式VIII或式IX的化合物(来自其中A=CH或N的反应方案I)反应,以提供式XXXIX的化合物。
在反应方案IV的步骤(8)和(10)中,在酸性条件下去除乙酰胺保护基团,以分别提供式XXXVIII或XL的化合物。该反应可在盐酸中在环境温度或高温(例如60℃)下进行。式XXXVIII或XL的产物可通过冻干作为盐酸盐分离。
反应方案IV
本发明的化合物还可使用反应方案I至IV的修改形式制成,所述反应方案I至IV的修改形式对于本领域技术人员将是显而易见的。例如,具有包含至多8个碳原子的X基团的式I化合物可使用反应方案IV的修改形式制成,以其中用C1-8亚烷基替换-Y-O-Z-的式XXXVI的化合物起始。此类起始化合物可使用美国专利号6,069,149(Nanba)中所述的方法制成。在另一个实例中,具有烷基氨基亚烷基基团的式I的化合物可使用例如美国专利号5,389,640(Gerster等人)中所述的方法使用反应方案II至IV的修改形式制备。本发明的化合物还可使用下文实例中所述的合成途径制备。
缀合物的制备
式I或I-A的化合物可与含醛抗原反应,以提供本发明的缀合物。因此,本发明的缀合物通常为式I或I-A的化合物或盐与含醛抗原的反应产物。这些反应产物通常为亚肼基苯甲酰胺或亚肼基烟酰胺1H-咪唑并[4,5-c]喹啉-4-胺。
含醛抗原可根据多种方法制备。在一些情况下,该抗原可与含有醛官能基团和第二反应性官能基团的异双官能连接化合物反应。在此类情况下,该抗原通常具有允许与异双官能连接化合物反应(例如在第二反应性官能基团处)的反应性官能基团。例如,抗原可具有来自赖氨酸残基的一个或多个(例如通常为多个)末端氨基基团,所述氨基基团可例如与异双官能连接化合物上的羧酸或其衍生物反应。本领域技术人员将理解,在包含多个氨基基团的生物分子例如蛋白质(即赖氨酸)中,与根据需要一样多的氨基基团可与异双官能连接化合物反应。改性程度可受使用的连接化合物的摩尔当量数控制。
在一些实施例中,异双官能连接化合物是芳族的。作为具体实例,氨基官能抗原可与琥珀酰亚胺4-甲酰苯甲酸酯(SFB)反应,以形成酰胺键,并且从而提供通过芳族环共价连接至抗原的醛官能基团。该反应可在适当的缓冲溶液中(例如在pH范围为7.2至7.5的磷酸盐缓冲液中)进行。SFB可溶解于适当的极性溶剂(例如DMSO或DMF)中,并且与含抗原的缓冲溶液混合。该反应可便利地在室温下进行。
在一些实施例中,异双官能连接化合物是芳族的,并且包括聚(乙烯氧基)链段。在这些实施例的一些中,异双官能连接化合物为其中p如上文其实施例的任一个中所定义。此类化合物可例如通过使羧基-PEG-胺化合物与N-琥珀酰亚胺-4-甲酰苯甲酸酯反应制备,所述羧基-PEG-胺化合物为例如可购自依利诺斯州洛克福德的赛默飞科技公司(Thermo Scientific,Rockford,IL)的那些。随后,例如通过与N,N,N’,N’-四甲基-O-(N-琥珀酰亚胺)脲四氟硼酸盐(TSTU)反应,羧酸基团可转变为活化酯。异双官能连接化合物与抗原的反应可在适当的缓冲溶液中(例如在范围为7.2至7.5的pH下的磷酸盐缓冲液中)进行。异双官能连接化合物可溶解于适当的极性溶剂(例如DMSO或DMF)中,并且与含抗原的缓冲溶液混合。该反应可便利地在室温下进行。关于此类异双官能连接化合物的另外信息,参见于2011年6月3日提交的共同待审的美国专利申请序列号61/493,143,该专利的公开内容以引用方式全文并入本文。
在其它实施例中,该抗原可能不需要异双官能连接化合物。例如,该抗原可具有可容易地转化为醛基团的官能基团。例如,在糖蛋白或糖脂的碳水化合物上的伯羟基基团可容易地氧化为醛。
在一些实施例中,本发明提供包含式I的化合物或盐、含醛抗原和药学上可接受的载体的组合物。本发明的缀合物可在此类组合物中制备。在制备根据本发明的缀合物的一些实施例中,式I或I-A的化合物或盐可溶解于适当的极性溶剂(例如DMSO、DMF)中,并且与含醛抗原的适当缓冲溶液混合。如果使用其中P为酸不稳定的受保护氨基基团的式I化合物,则例如酸性缓冲溶液(例如具有在4.7至6.2范围内的pH)可影响氨基基团的脱保护,并且同时允许与含醛抗原反应。该反应通常在室温下进行。因此,在一些实施例中,本发明提供制备缀合物的方法,所述方法包括在其中使受保护的氨基基团脱保护且形成缀合物的条件下,将其中P为受保护的氨基基团的式I的化合物或盐、含醛抗原和载体组合。
当芳族、含醛异双官能连接化合物用于制备含醛抗原时(例如当氨基官能抗原与琥珀酰亚胺4-甲酰苯甲酸酯(SFB)反应,以形成酰胺键,并且从而提供共价连接至抗原的醛官能基团时),芳族醛基团与式I的化合物的反应可便利地使用UV分光光度曲线测定进行跟踪。形成的双芳族腙键提供鉴别性发色团,其具有在354nm处的最大吸光度和等于29,000的摩尔消光系数。如下文实例中所证实,通过将在354nm处测量的缀合物吸光度除以29,000的摩尔消光系数,可计算掺入抗原中的式I化合物的摩尔数。
为了促进反应中的溶解度和稳定性以提供本文所公开的缀合物,根据所选抗原或蛋白质的性质,多种添加剂可用于反应混合物中。例如,甘油和/或表面活性剂(例如聚山梨醇酯80)可用于促进溶解度和稳定性。另外,在缀合物中提供聚(乙烯氧基)链段可用于促进溶解度和稳定性。在这些实施例的一些中,Z为-C(O)-NH-(CH2CH2O)p-CH2CH2-,其中p如上文实施例中的任一个中所定义。为了促进反应效率,催化剂(例如苯胺)可以有效量(例如至多200mM)添加。当甘油和/或表面活性剂添加到反应混合物中时,催化剂可用于例如促进反应。
在一些实施例中,该抗原为蛋白质。在本发明的缀合物中可为有用的抗原的示例性蛋白质包括来自H1N1PR8的血凝素、乙型肝炎表面抗原、利什曼原虫属(Leishmania)抗原、呼吸道合胞病毒分泌蛋白F、疟疾表面抗原、前列腺碱性磷酸酶前列腺癌抗原和M期磷蛋白1膀胱癌抗原。
最佳反应条件可根据不同蛋白质特性而改变,所述蛋白质特性包括等电点、总平均亲水性、不稳定性指数(试管中的蛋白质稳定性估计)、由脂族侧链(丙氨酸、缬氨酸、异亮氨酸和亮氨酸)占据的相对体积(其视为增加球状蛋白质热稳定性的正面因素)、阴离子残基数和阳离子残基数。此类特性对于多种蛋白质是已知的。
蛋白质的稳定性及其天然构象的维护受到在其内部域内的疏水相互作用以及在其结构的外表面上的氢键和电荷相互作用的组合影响。因为这些表面相互作用通过用试剂例如含醛异双官能化合物和式I的化合物改性而改变,因此可改变蛋白质的天然构象。为了提供缀合物(即式I的化合物或盐与含醛蛋白质的反应产物),化合物或盐与含醛蛋白质的比率可改变,使得蛋白质的稳定性及其天然构象得到维护。在一些实施例中,化合物或盐与含醛蛋白质的比率在30:1至1:3的范围内。在一些实施例中,化合物或盐与含醛蛋白质的比率在20:1至1:2的范围内。在一些实施例中,化合物或盐与含醛蛋白质的比率在10:1至1:1的范围内。式I的化合物或盐的当量数例如可与相同实施例中使用的异双官能连接化合物的当量数相同或相似。在缀合物的一些实施例中,式II的缀合链段与含醛蛋白质的比率在30:1至1:6的范围内。在一些实施例中,式II的缀合链段与含醛蛋白质的比率在20:1至1:5的范围内。在一些实施例中,式II的缀合链段与含醛蛋白质的比率在10:1至1:1的范围内。
药物组合物和方法
由式I或I-A的化合物和含醛抗原制成的缀合物(在一些实施例中为式II的缀合物)可以任何合适方式(例如肠道或非肠道)在本文公开的药物组合物中施用。如本文所用,肠道是指通过消化道(包括通过口服摄取)施用。非肠道是指给不是通过消化道的施用,其将包括鼻腔(如,通过吸入透粘膜)、局部、眼科和颊面施用,但是在实践中通常是指使用例如常规的针注射、使用微针阵列注射、或任何其它已知的注射方法的注射(如,静脉注射、肌内注射、皮下注射、瘤内注射或透皮注射)。
由式I或I-A的化合物和含醛抗原制成的缀合物(在一些实施例中为式II的缀合物)可在适于向受试者施用的任何药物组合物中提供,并且可以任何合适形式存在于药物组合物中(例如溶液、悬浮液、乳状液、或任何形式的混合物)。药物组合物可由任何药学上可接受的赋形剂、载体、或媒介物配制。该药物组合物还可包含一种或多种添加剂,包括皮肤渗透促进剂、着色剂、芳香剂、调味剂、增湿剂、增稠剂、悬浮剂、表面活性剂和分散剂。
除了上文和下文具体描述的抗原,本发明的药物组合物和方法可包括例如混合或单独施用的其它另外的活性剂。此类另外的试剂可包括化学治疗剂、细胞类毒素剂、抗体、抗病毒素剂、细胞因子、肿瘤坏死因子受体(TNFR)激动剂、或另外的免疫应答调节剂。可与本发明的缀合物(在一些实施例中为式II的缀合物)结合递送的TNFR激动剂包括CD40受体激动剂,例如美国专利申请公布号2004/0141950(Noelle等人)中所公开。用于与本发明的IRM制剂组合使用的其它活性成分包括在例如美国专利申请公布号2003/0139364(Krieg等人)中公开的那些。
由式I或I-A的化合物和含醛抗原制成的缀合物(在一些实施例中为式II的缀合物)已显示在人细胞中诱导INF-α和TNF-α的产生,如下文实例中所述。诱导INF-α和TNF-α产生的能力表明本发明的化合物和缀合物可以多种不同方式调节免疫应答,从而使它可用于多种疾病的治疗中。可通过本文公开的化合物和缀合物的施用诱导产生的其它细胞因子通常包括I型干扰素(例如INF-α)、IL-1、IL-6、IL-8、IL-10、IL-12、MIP-1、MCP-1和多种其它细胞因子。在其它效应中,这些及其它细胞因子抑制病毒产生和肿瘤细胞生长,使得式I的化合物以及由其制成的缀合物可用于病毒性疾病和赘生性疾病的治疗中。例如,肿瘤坏死因子、干扰素、或白介素已显示刺激某些单核细胞/巨噬细胞衍生的细胞因子的迅速释放,并且还能够刺激B细胞分泌在抗病毒素和抗肿瘤活性方面起重要作用的抗体。
除了诱导细胞因子产生的能力,本文所述的缀合物还可影响先天性免疫应答的其它方面。例如,可以刺激自然杀伤细胞活性,这可能是由于细胞因子的诱导的效应。本发明的缀合物的IRM活性还可包括活化巨噬细胞,这继而刺激了一氧化氮的分泌和另外的细胞因子的产生。本发明的缀合物的IRM活性还可包括通过T细胞诱导细胞因子产生、活化对抗原特异性的T细胞、和/或活化树突状细胞。另外,缀合物的IRM活性可包括B-淋巴细胞的增殖和分化。缀合物的IRM活性还可影响获得性免疫应答。例如,IRM活性可包括诱导1型T辅助细胞(TH1)细胞因子IFN-γ的产生和/或抑制2型T辅助细胞(TH2)细胞因子IL-4、IL-5和/或IL-13的产生。

因此,本发明提供在动物中诱导细胞因子生物合成的方法,所述方法包括向所述动物施用有效量的由式I或I-A的化合物和含醛抗原制成的缀合物(在一些实施例中为式II的缀合物)(例如在药物组合物中)。
在由式I或I-A的化合物和含醛抗原制成的缀合物(在一些实施例中为式II的缀合物)的一些实施例中,该抗原为疫苗,并且根据本发明的方法包括给动物接种疫苗的方法,所述方法包括向所述动物施用由式I的化合物和抗原制备的缀合物(在一些实施例中为式II的缀合物)。疫苗包括施用于提高或者体液的和/或细胞介导的免疫应答的任何物质,诸如活的或减毒的病毒和细菌免疫原和进行灭活的病毒、肿瘤来源的、原生动物的、生物体来源的、真菌的和细菌的免疫原、类毒素、毒素、多糖、蛋白质、糖蛋白、肽、细胞疫苗(如,使用树枝状的细胞)、DNA疫苗、重组蛋白、糖蛋白和肽。示例性的疫苗包括用于癌症、BCG、霍乱、瘟疫、伤寒、肝炎A,B和C、流行性感冒A和B、变异流行性感冒病毒、脑灰质炎、狂犬病、麻疹、腮腺炎、风疹、黄热病、破伤风、白喉、流感嗜血杆菌b、肺结核、流行性脊髓膜炎和肺炎球菌的疫苗、腺病毒、HIV、水痘、细胞巨化病毒、登革热、猫白血、禽类瘟疫、HSV-1和HSV-2、猪瘟、日本脑炎、呼吸道合胞病毒、轮状病毒、乳头状瘤病毒、严重急性呼吸系统综合症(SARS)、炭疽热和黄热病的疫苗。还参见例如公开于国际公开号WO02/24225(Thomsen等人)中的疫苗。
本发明的方法可在任何合适的受试者上执行。合适的受试者包括动物诸如人类、非人灵长类、啮齿类、狗、猫、马、猪、绵羊、山羊或牛。
施用该缀合物用于诱导细胞因子生物合成或用于接种疫苗的动物可具有疾病(如,病毒性疾病或赘生性疾病),并且化合物的施用可提供治疗处理。另外,该缀合物可在动物获得疾病之前施用于动物,使得该缀合物的施用可提供预防处理。例如,缀合物可由式I或I-A的化合物和HIV抗原制成,并且可提供对于HIV的治疗和/或预防处理。在另一个实例中,缀合物可由式I或I-A的化合物和肿瘤相关的抗原制成,并且可提供针对与抗原相关的肿瘤的治疗和/或预防处理。
可通过施用IRM缀合物治疗的示例性病症包括:

(f)某些自体免疫疾病诸如系统性红斑狼疮、原发性血小板增多症、多发性硬化症、盘状狼疮和斑秃症;以及
(g)与伤口修复相关的疾病,诸如抑制瘢痕疙瘩形成和其它类型的结疤(如,增强伤口愈合,包括慢性伤口)。
IRM缀合物还可用于具有失能的免疫功能的个体。例如,某些缀合物可用于治疗在例如移植患者、癌症患者和HIV患者中的机会性感染和肿瘤,其在细胞介导的免疫的抑制后出现。
应当理解,在上文所提及的疾病的治疗中,例如,本文公开的缀合物可与其它疗法诸如上文提及的活性剂和其它工序(如,化学消融术、激光烧蚀、冷冻疗法和外科切除)组合使用。
有效诱导细胞因子生物合成的缀合物的量为足以引起一种或多种细胞类型,诸如单核细胞、巨噬细胞、树突状细胞和B-细胞产生一种或多种细胞因子(例如IFN-α、TNF-α、IL-1、IL-6、IL-IO和IL-12)的量,该量增加超过此类细胞因子的背景水平。精确的量将根据本领域中已知的因素改变,但是预计剂量为约100纳克/千克(ng/kg)至约50毫克/千克(mg/kg),在一些实施例中为约10微克/千克(μg/kg)至约5mg/kg、约100μg/kg至约1mg/kg、或约0.01mg/m2至约10mg/m2。作为另外一种选择,可使用刚好在治疗过程开始之前获得的实际体重计算剂量。对于以该方式计算的剂量,在治疗过程开始之前计算体表面积(m2),使用杜博瓦(Dubois)方法:m2=(重量kg0.425×高度cm0.725)×0.007184。有效治疗或抑制病毒感染的量例如为与未经治疗的对照动物相比较,引起病毒感染临床表现中的一种或多种减少的量,所述临床表现例如病毒损害、病毒负荷、病毒产生率和死亡率,并且可包括前述剂量中的任一种。有效治疗赘生性病症的化合物或药物组合物的量为将引起肿瘤尺寸或肿瘤病灶数目减少的量,并且可包括前述剂量中的任一种。

A部分
将戊酸酐(6.03g)和含吡啶盐酸盐(0.198g)的吡啶(8.28g)溶液添加至含3-氨基-4-氯喹啉(2.94g)的吡啶(5.0g)溶液中,并且将反应在室温下搅拌16小时,随后在60℃下加热3小时。将反应在减压下浓缩并添加碳酸钠(15mL的10%水溶液)。将反应搅拌30分钟,然后过滤。将所得的固体用水(60mL)洗涤并在真空下干燥4小时,以提供4.59g作为棕色薄片的粗制N-(4-氯喹啉-3-基)戊酰胺。将粗产物从庚烷(10mL)中重结晶并使用回流庚烷通过索氏萃取16小时进一步纯化回收产物。将来自索氏萃取装置的收集烧瓶在冷冻机中冷却2小时。通过过滤收集所得的固体并在真空下干燥,以得到2.00g作为白色固体的N-(4-氯喹啉-3-基)戊酰胺。
B部分
4-氨基-1-丁醇(7.68g)和含吡啶(7.00g)的二氯甲烷(100mL)溶液在冰浴中冷却并在30分钟的时间内在搅拌下缓慢添加含氯甲酸苄酯(14.37g)的二氯甲烷(100mL)溶液。移除冰浴并将反应搅拌另外的16小时。添加盐酸(1.2M,200mL)并分离相。在减压下将有机相干燥(MgSO4),过滤并浓缩。将所得的残余物从甲苯中重结晶并在真空下干燥,以提供5.15g的苄基(4-羟基丁基)氨基甲酸酯。

D部分
将苄基4{-[2-丁基-1H-咪唑并[4,5-c]喹啉-1-基]氧基}氨基甲酸丁酯盐酸盐(10.05g)溶解在二氯甲烷(80mL)中,并且用碳酸钠(2.02g)的30mL H2O溶液萃取。将有机层在冰浴中冷却,并且缓慢添加溶解于二氯甲烷(30mL)中的间氯过苯甲酸(5.93g,1.24当量)溶液。6小时后,向反应中添加氢氧化铵(10mL的28-30%水溶液)。将溶解在10mL二氯甲烷中的苯磺酰氯(6.96g)溶液在大力搅动下缓慢添加。移除冷却浴并将反应搅拌另外的12小时。将反应用水(100mL)稀释并分离有机馏分和含水馏分。将含水馏分用二氯甲烷(30mL)萃取。将混合的有机馏分用两份90mL的5%碳酸钠洗涤。

将1-(4-氨基丁氧基)-2-丁基-1H-咪唑并[4,5-c]喹啉-4-胺双马来酸盐(0.2g)悬浮于1-丁醇(5mL)中,并且用2×5mL份的5%碳酸钠溶液随后用5mL的饱和氯化钠溶液依次洗涤。添加琥珀酰亚胺4-肼基烟酸盐丙酮腙(SANH,0.0216g);可购自依利诺斯州洛克福德的赛默飞科技公司;并且将溶液在环境温度下搅拌17.5小时。通过薄层色谱法(硅胶,1:1甲基-叔丁醚:乙醇的洗脱液)的反应分析显示仅存在1-(4-氨基丁氧基)-2-丁基-1H-咪唑并[4,5-c]喹啉-4-胺(Rf<0.05)和所需产物N-(4-{[4-氨基-2-丁基-1H-咪唑并[4,5-c]喹啉-1-基]氧基}丁基)-6-(N’-异亚丙基肼基)烟酰胺(Rf0.30)。该反应在减压下浓缩,并将5mL的二氯甲烷添加到残余物中。通过过滤去除少量不溶解的物质,并且通过柱色谱法(硅胶,1:1甲基-叔丁醚:乙醇的洗脱液)纯化。将含产物的馏分混合,并且在减压下去除溶剂,以提供作为淡黄色固体的N-(4-{[4-氨基-2-丁基-1H-咪唑并[4,5-c]喹啉-1-基]氧基}丁基)-6-(N’-异亚丙基肼基)烟酰胺(化合物1)。

实例2
将来自H1N1PR8的重组血凝素1(HA)克隆,在大肠杆菌中表达,并且使用标准工序纯化。将分子量为32083.11道尔顿、在C末端处含有6个组氨酸的HA置于含有0.15M NaCl的pH7.5,0.1M磷酸盐缓冲液中。基于HA的分子量和蛋白质的质量,确定HA溶液的摩尔浓度。将溶解于二甲基亚砜(DMSO)中的琥珀酰亚胺4-甲酰苯甲酸酯(SFB)(依利诺斯州洛克福德的赛默飞科技公司)以10倍摩尔过量添加到HA中。随后将溶液在室温下温育2小时。使HA的对照样品在等同体积的DMSO中以类似方式温育。通过使用由含有0.15M NaCl的pH6.0,0.1M磷酸盐缓冲液预平衡的ZEBA旋转柱(依利诺斯州洛克福德的赛默飞科技公司),使SFB改性的HA(表示为HA-SFB)与游离SFB分离。该步骤在用于缀合反应的制备中使HA-SFB溶液变成pH6.0。
为了测定共价缀合的效率,将N-(4-{[4-氨基-2-丁基-1H-咪唑并[4,5-c]喹啉-1-基]氧基}丁基)-6-(N’-异亚丙基肼基)烟酰胺(实例1的化合物1)溶解于DMSO中,并且以30、10、3和1倍摩尔过量的量添加到缓冲的HA-SFB溶液中。反应介质的酸性条件导致化合物1的乙酰胺(acetimine)保护基团的脱保护,以原位形成化合物2。四个测试样品中的每一个在室温下温育2小时。使对照HA-SFB样品在等同体积的DMSO中以类似方式温育。通过使用由达尔贝科磷酸盐缓冲盐水(PBS)预平衡的ZEBA旋转柱(密苏里州圣路易斯的西格玛奥德里奇公司(Sigma-Aldrich,St.Louis,MO)),使共价缀合至实例1的化合物2的HA-SFB(表示为HA-SFB-化合物2)与未缀合的组分分离。
通过使用UV分光光度曲线测定来确定化合物2通过共价缀合掺入HA中的效率,并且结果记录在表1中。通过HA-SFB与化合物2的共价缀合形成的双芳族腙键提供鉴别性发色团。该发色团具有在354nm处的最大吸光度和等于29,000的摩尔消光系数。通过将在354nm处测量的缀合的HA-SFP-化合物2的吸光度除以29,000的摩尔消光系数,计算掺入HA蛋白质中的化合物1的摩尔数。结果显示于下表1中。

实例3
根据实例2中所述的工序,通过将10倍摩尔过量的化合物1添加到HA-SFB中,制备共价缀合的产物(HA-SFB-化合物2)。测定通过缀合产物在人外周单核细胞(PBMC)中的干扰素-α(IFN)和肿瘤坏死因子(TNF)生产的体外诱导。从人类志愿者中制备PBMC并置于96孔微量滴定板中的培养物中。分别将HA、HA-SFB和HA-SFB-化合物2缀合物以1μ蛋白质的最终浓度添加到孔中。随后使细胞在37℃下温育过夜。去除培养基,并且通过ELISA测定来测量IFN浓度(pg/mL)和TNF浓度(ng/mL)。测定的结果报告于下表2中。

实例4
在Balb/C雄性小鼠(马萨诸塞州威尔明顿的查尔斯河实验室国际公司(Charles River Laboratories,International,Wilmington,MA))中评估共价缀合至重组血凝素1(HA)的化合物2(HA-SFB-化合物2)的疫苗佐剂活性。各5只小鼠的组用10微克PBS中的HA抗原(对照)、10微克缀合至SFB的HA抗原(对照),或用SFB改性,然后缀合至实例1的化合物2的HA抗原进行皮下免疫。根据实例2中所述的工序,通过将10倍摩尔过量的化合物1添加到HA-SFB中,制备缀合的产物。在初始免疫后的2周和4周用相同的组合对小鼠进行追加。三周和再次在最终加强后的12周时,将小鼠放血并测定HA特异性抗体滴度。由标准血清ELISA在HA-涂覆的微量滴定板上通过血清样品的系列稀释执行测定。数据呈现为实现终点(2X基线)的血清稀释度且为每组5只小鼠的几何平均值。作为免疫应答的TH1偏差的标记,除了HA-特异性总IgG之外,还测量HA-特异性IgG1和IgG2a亚型。计算结果报告于表3中。
通过腙连接基团共价缀合至HA的化合物2证实有力的疫苗佐剂效应,其中强TH1偏向的免疫应答由增加的HA特异性IgG2a与HA特异性IgG1抗体比率表明。

制备
A部分
将溶解于干燥二氯甲烷(5mL)中的CA(PEG)12(式H2N-CH2CH2-(OCH2CH2)12-CO2H;MW=617.7;可购自依利诺斯州洛克福德的赛默飞科技公司,115mg)、N-琥珀酰亚胺-4-甲酰苯甲酸酯(溶解于干燥二氯甲烷(0.5mL)中的52mg;可购自纽泽西州吉比斯镇的EMD Chemicals公司(EMDChemicals,Gibbstown,NJ))、干燥三乙胺(52μL)和催化量的DMAP在氮气氛下混合。将反应搅拌3小时,然后用二氯甲烷(25mL)稀释。将有机馏分用0.1M磷酸钠(2×10mL)洗涤,随后用盐水洗涤。将有机馏分在硫酸钠之上干燥,过滤,并在减压下浓缩。混合含水洗涤馏分且用多份二氯甲烷萃取。随后用稀盐酸将含水馏分酸化至pH~2,并且用另外两份二氯甲烷萃取。将有机萃取物混合,在硫酸钠之上干燥,过滤,并在减压下浓缩。将所得物质与得自第一次萃取的物质混合,并且使用小硅胶柱纯化。用10-25%甲醇/氯仿洗脱,用水饱和,得到58mg作为无色固体的酰胺产物。1H NMR(氯仿-d,500MHz)δ10.08(s,1H),8.00(d,J=8.2Hz,2H),7.95(d,J=8.4Hz,2H),7.19(m,1H),3.77(t,J=6.1Hz,2H),3.70-3.60(m,48H),2.60(t,J=6.1Hz,2H)。
B部分
将来自A部分的物质溶解于干燥N,N-二甲基甲酰胺(0.5mL)和干燥吡啶(0.5mL)中。添加O-(N-琥珀酰亚胺)-1,1,3,3-四甲基脲四氟硼酸盐(TSTU;46mg;可购自密苏里州圣路易斯的西格玛奥德里奇公司),并且将反应在氮气氛下搅拌3小时。在减压下去除大多数溶剂。将所得物质溶解于氯仿(25mL)和甲醇(5mL)中,并且置于分液漏斗中。添加缓冲溶液(10mL用氢氧化钠调节至pH7.5的0.10M氯化钠、0.05M磷酸钠、1.0mM EDTA溶液),并且将混合物振荡2分钟。将有机馏分收集且用另外份的缓冲溶液(10mL)、水(3×10mL)和盐水依次洗涤。将有机馏分在硫酸钠之上干燥,过滤,并在减压下浓缩,以提供55mg作为无色糖浆的化合物3。1H NMR(氯仿-d,500MHz)δ10.08(s,1H),7.99(d,J=8.2Hz,2H),7.95(d,J=8.1Hz,2H),7.10(m,1H),3.85(t,J=6.5Hz,2H),3.70-3.60(m,48H),2.90(t,J=6.9Hz,2H)2.84(br s,4H)。
实例5
实例5根据实例2的方法制备,伴随改性:将溶解于二甲基亚砜(DMSO)中的化合物3以10倍摩尔过量添加到HA中,以提供化合物3-改性的HA(表示为HA-化合物3)。
将N-(4-{[4-氨基-2-丁基-1H-咪唑并[4,5-c]喹啉-1-基]氧基}丁基)-6-(N’-异亚丙基肼基)烟酰胺(实例1的化合物1)溶解于DMSO中,并且以10倍摩尔过量添加到缓冲的HA-化合物3溶液中。反应介质的酸性条件导致化合物1的乙酰胺保护基团的脱保护,以原位形成化合物2。样品在室温下温育2小时。通过使用由达尔贝科磷酸盐缓冲盐水(PBS)预平衡的ZEBA旋转柱(密苏里州圣路易斯的西格玛奥德里奇公司),使共价缀合至实例1的化合物2的HA-化合物3(表示为HA-化合物3-化合物2)与未缀合的组分分离。

与SFB相比较,使用在共价缀合的产物中的化合物3对最终蛋白质溶解度和回收百分比的作用显示于表4中。可溶蛋白质测量测定为在100K×g离心样品的上清液中回收的HA-SFB-化合物2或HA-化合物3-化合物2的量。总蛋白质测量测定为在离心之前的样品中的HA-SFB-化合物2或HA-化合物3-化合物2的量。可溶蛋白质和总蛋白质测量使用二辛可宁酸(BCA)蛋白质测定(得自依利诺斯州洛克福德的赛默飞科技公司)进行。
