Abstract
Description
免疫系の特定の重要な側面を刺激することによって、そして特定の他の側面を抑制することによって作用する新しい薬剤化合物を発見するために、近年大きな努力がなされており、著しい成功が収められている(例えば、米国特許第6,039,969号および第6,200,592号を参照)。本明細書中で免疫応答修飾物質(immune response modifier、IRM)と呼ばれるこれらの化合物は、トール様受容体(TLR)として知られる基本の免疫系メカニズムにより作用して、選択されるサイトカイン生合成を誘発すると思われる。これらは、様々な種類の疾患および状態を処置するために有用であり得る。例えば、特定のIRMは、ウィルス性疾患(例えば、ヒトパピローマウィルス、肝炎、ヘルペス)、新形成(例えば、基底細胞癌、扁平上皮癌、光線性角化症、メラノーマ)、およびTH2媒介性疾患(例えば、喘息、アレルギー性鼻炎、アトピー性皮膚炎)を処置するために有用であり、またワクチンアジュバントとしても有用である(米国特許第6,083,505号および米国特許出願公開第2004/0076633号)。
特定のIRM化合物は、DNAワクチンのためのアジュバントとして有用であり得ることが分かっている。
特定のIRM化合物は、臨床的に適切な癌関連抗原ペプチドを標的とするDNAワクチンのためのアジュバントとして有用であると確認されている。さらに、特定のIRM化合物は可能性のあるDNAワクチンアジュバントとして提案されているが、これは、自然発生的な(すなわち、非形質移入の)腫瘍特異性抗原を標的とするDNAワクチンのためのアジュバントとしてIRM化合物が有効であり得るという最初の証明である。
「アゴニスト」は、受容体(例えば、TLR)と組み合わされて細胞の活性を誘発することができる化合物を指す。アゴニストは、受容体に直接結合するリガンドでもよい。あるいは、アゴニストは、例えば(a)受容体に直接結合する別の分子と複合体を形成することによって、あるいは(b)別の方法で別の化合物の修飾をもたらし、他の化合物を受容体に直接結合させることによって、受容体と間接的に組み合わされてもよい。アゴニストは、特定のTLRのアゴニスト(例えば、TLR8アゴニスト)またはTLRの特定の組み合わせ(例えば、TLR7/8アゴニスト−TLR7およびTLR8の両方のアゴニスト)と見なされ得る。
「HER−2」、「neu」、および「HER−2/neu」は、ラットneu癌原遺伝子およびそのヒト相同体、HER−2、またはそのマウス相同体、neuによりコードされる185kDタンパク質を交換可能に指す。
アルファフェトプロテイン(AFP)は、肝細胞癌(HCC)に関連する臨床的に適切な抗原ペプチドである。HCC、原発性の肝臓癌は、癌死亡の主要な原因である。アジアに特有の疾患は、B型肝炎感染の結果として肝硬変を患っている個人において顕著である。毎年およそ120万人の新たな症例が発生し、ほとんど全ての患者は診断の6ヶ月以内に死亡する。AFPの抗原性部分をコードする発現ベクターで免疫化されたマウスは、腫瘍の成長の遅延を経験した。このような発現ベクターは、例えば米国特許出願公開第2003/0143237号に記載されている。
以下の実施例は、単に、本発明の特徴、利点、および他の詳細をさらに説明するために選択されている。しかしながら、実施例はこの目的を果たすが、使用される特定の材料および量、ならびに他の条件および詳細が、本発明の範囲を不当に限定し得る状況で解釈されてはならないことは明白に理解されるはずである。
実施例において使用されるIRM化合物は、表1に示される。
活性化ラットneu遺伝子を含有するメスのFVB/Nマウス(カリフォルニア州ホリスターのチャールズ・リバー・ラボラトリーズ(Charles River Laboratories,Hollister,CA))を、標準的な明/暗療法(12時間明:12時間暗)の下で、特定の病原体がない状態に保持した。マウスをプラスチック製のメッキしていないケージ(1つのケージにつき4〜6匹のマウス)内に収容し、標準的なペレットフードおよび水道水を随意に与えた。
指示される最終濃度が得られるまでIRM化合物を0.2%のDMSOおよび水に溶解することによって、IRM溶液を調製した。
実施例1に記載したように、動物を分類し、免疫化および/または処置した。Ca2+およびMg2+を含まないリン酸緩衝食塩水(PBS、メリーランド州ゲイサーズバーグのギブコ(GIBCO,Gaithersburg,MD))溶液中で60ミクロンメッシュのふるいによって脾臓を収集して処置した。脾臓細胞をリンパ球M(カナダ国オンタリオ州ホーンビイのシダーレーン・ラボラトリーズ社(Cedarlane Laboratories,Ltd.,Hornby,Ontario,Canada)において分画し、密度勾配遠心分離(500g、20分)によって単核細胞を分離した。勾配の界面からの細胞をPBSで2回洗浄し、ペニシリン(100U/mL)およびストレプトマイシン(100μg/mL)を含有するRPMI1640(メリーランド州ゲイサーズバーグのライフ・テクノロジーズ社(Life Technologies,Inc.))中に再懸濁した。
Fmed=媒体のみの中でインキュベートした標的からのFであり、
Fexp=エフェクター細胞と共にインキュベートした標的からのFである。
実施例1に記載したように、動物を分類し、免疫化および/または処置した。免疫化期間の完了の2週間後に、コントロールおよび実験動物から血清を収集した。血清を−80℃で貯蔵し、フローサイトメトリーにより連続的に分析した。高レベルの腫瘍特異性の抗原p185neuを発現する2×105N202.1A細胞を、2%BSAおよび0.5%ナトリウムアジドが補充された冷PBS(PBS−アジド−BSA)で2回洗浄した。次に、PBS−アジド−BSA中に1:10で希釈した50μLのコントロールまたは免疫血清を用いて、標準的な間接免疫蛍光法手順で細胞を染色した。フルオレセイン結合ウサギ抗マウスIg(カリフォルニア州サンディエゴのEMDバイオサイエンシーズ社(EMD Biosciences,Inc.,San Diego,CA))を二次抗体として使用した。細胞をアイソトン(Isoton)II中に再懸濁し、コールター・エピックス(COULTER EPICS)XL(カリフォルニア州フラートンのベックマン・コールター社(Beckman Coulter,Inc.,Fullerton,CA))フローサイトメーターにより評価した。血清のN202.1A結合能(Sbp)、抗原特異性の体液性免疫応答の尺度を以下のように計算した。

脾細胞を実施例2に記載したように入手し、刺激物質としてN202.1A腫瘍細胞(20:1比の刺激物質:リンパ球)の存在下、10%FCSを含有するRPMI媒体中で、37℃および5%CO2で一晩インキュベートした。細胞を収集し、PE結合抗CD4または抗CD8モノクローナル抗体(カリフォルニア州サンノゼのBDバイオサイエンシーズ、ベクトン、ディッキンソン社(BD Biosciences,Becton,Dickinson and Co.,San Jose,CA))により5%FCSおよび0.01%NaN3を含有するPBS緩衝液中で染色した。次に、細胞を0.2%ホルマリン(formaline)中で固定し、FITC結合抗IL10、抗IL−12、または抗IFN−γ(BDバイオサイエンシーズ)により5%FCSおよび0.05%ホルマリンを含有するPBS緩衝液中で連続的に染色した。コールター・エピックスXLフローサイトメーター(カリフォルニア州フラートンのベックマン・コールター社)によって染色を評価した。
結果は図5に要約される。
上記のように調製および送達した50μgまたは100μgのpCMV−ECD−TMワクチンにより、2、30、および58日目にアカゲザルを左上腕で免疫化する。免疫化部位において、IRM化合物で処置される動物は、PBS中に溶解した0.5mg/kgのIRM1、もしくは0.05mg/kg、0.5mg/kg、または5mg/kgのIRM3、IRM4、IRM5、またはIRM6、もしくは50mg/kgのIRM5を含有する皮内注射を受ける。これらのIRM化合物の受け取りは、0日目から開始して期間中3日ごとに行われる。16、45、および72日目に血液を採取し、ELISPOTによりIFN−γ産生細胞の数を測定する。IFN−γ産生細胞の数は、IRM用量に依存するように変動し得る。
実施例5に記載したように、アカゲザルを分類し、免疫化および/または処置する。免疫化期間の完了の2週間後に、コントロールおよび実験動物から血清を収集する。血清を−80℃で貯蔵し、フローサイトメトリーにより連続的に分析する。高レベルの腫瘍特異性抗原Her−2を発現する2×105SK−BR−3細胞(バージニア州マナサス(Mannasas)のATCC)を、2%BSAおよび0.5%ナトリウムアジドが補充された冷PBS(PBS−アジド−BSA)で2回洗浄する。次に、PBS−アジド−BSA中に1:10で希釈した50μLのコントロールまたは免疫血清を用いて、標準的な間接免疫蛍光法手順で細胞を染色する。フルオレセインイストチオシアネート(isthothiocyanate)結合ロバ抗ヒトIgG(H+L)(ペンシルベニア州ウェストグローブのジャクソン・イムノリサーチ・ラブズ社(Jackson ImmunoResearch Labs,Inc.,West Grove,PA))を二次抗体として使用する。細胞をフローサイトメトリー染色緩衝液(カリフォルニア州カマリロのバイオソース・インターナショナル(Biosource International,Carmarillo,CA)中に再懸濁し、FACSCalibur(カリフォルニア州サンノゼのBDバイオサイエンシーズ(BD Biosciences))フローサイトメーターにより評価する。血清のSK−BR−3結合能(Sbp)、抗原特異性の体液性免疫応答の尺度は以下のように計算される。

以下のグループ:(1)pCMV−ECD−TMにより免疫化、IRMによる処置なし(HER−2/neu)、(2)pCMV−ECD−TMにより免疫化、IRM2により処置(IRM+HER−2/neu)、または(3)未処置(コントロール)のそれぞれに対して、実施例1と同様にして動物を処置した。免疫化期間の完了の2週間後に、動物から血清を収集し、同じ処置を受ける動物の間でプールした。

- 癌を処置するためのDNAワクチンの製造におけるIRM化合物の使用であって、前記DNAワクチンが、
臨床的に適切な癌関連抗原ペプチドをコードする発現ベクター;および
TLR8選択性アゴニストである化合物
を含む使用。 - 前記化合物が、イミダゾキノリンアミン、テトラヒドロイミダゾキノリンアミン、イミダゾピリジンアミン、1,2−架橋イミダゾキノリンアミン、6,7−縮合シクロアルキルイミダゾピリジンアミン、イミダゾナフチリジンアミン、テトラヒドロイミダゾナフチリジンアミン、オキサゾロキノリンアミン、チアゾロキノリンアミン、オキサゾロピリジンアミン、チアゾロピリジンアミン、オキサゾロナフチリジンアミン、チアゾロナフチリジンアミン、ピラゾロピリジンアミン、ピラゾロキノリンアミン、テトラヒドロピラゾロキノリンアミン、ピラゾロナフチリジンアミン、またはテトラヒドロピラゾロナフチリジンアミンを含む、請求項33〜35のいずれか一項に記載の使用。
- 前記化合物が、チアゾロキノリンアミンを含む、請求項36に記載の使用。
- 癌処置用組成物を調製する方法あって、
臨床的に適切な癌関連抗原ペプチドに対する免疫応答を発生させるために有効な量の、臨床的に適切な癌関連抗原ペプチドをコードする発現ベクターを被検者に投与すること;
臨床的に適切な癌関連抗原ペプチドへの免疫応答を増強するために有効な量のIRM化合物を被検者に投与すること;
臨床的に適切な癌関連抗原ペプチドへの血清免疫応答を被検者に発生させること;および
被検者の血清の少なくとも一部を採取すること
を含む方法。 - 前記癌が、肝細胞癌、子宮頸癌、メラノーマ、肺癌、結腸癌、乳癌、膵癌、または卵巣癌を含む、請求項38に記載の方法。
- 前記癌関連抗原ペプチドが、Her−2/neu、マンマグロブリン−A、MUC1、アルファフェトプロテイン、HPV E6、HPV E7、TRP−1、またはVEGF2を含む、請求項38に記載の方法。
- 被検者において癌を処置する方法であって、
臨床的に適切な癌関連抗原ペプチドに対する免疫応答を発生させるために有効な量の、臨床的に適切な癌関連抗原ペプチドをコードする発現ベクターを哺乳類に投与すること;
臨床的に適切な癌関連抗原ペプチドへの免疫応答を増強するために有効な量のIRM化合物を哺乳類に投与すること;
臨床的に適切な癌関連抗原ペプチドへの血清免疫応答を哺乳類に発生させること;
哺乳類の血清の少なくとも一部を採取すること;および
癌を処置するために有効な量で、前記哺乳類の血清の少なくとも一部を被検者に投与すること
を含む方法。 - 前記哺乳類の血清の一部が、臨床的に適切な癌関連抗原に対する抗体を含む、請求項41に記載の方法。
