在一个实施方式中,在泪点塞中给予γ-晶状体球蛋白电荷掩蔽剂。如在本文中使用的,术语泪点塞是指具有适用于分别通过下或上泪点(inferior or superior lacrimalpunctum)插入到眼睛的下或上泪小管(inferior or superior lacrimal canaliculus)中的尺寸和形状的设备。
在一个实施方式中,通过离子电渗给予γ-晶状体球蛋白电荷掩蔽剂。离子电渗(Iontophoresis)是使用小的电荷来通过皮肤递送药物或其他化学品的技术。

实施例
方法
γD-和γS-晶状体球蛋白DNA序列在通过麻省理工学院(剑桥,MA)的King实验室提供的pQe1质粒中。γ-晶状体球蛋白蛋白质序列包含6x N-端组氨酸标签(His标签)用于纯化目的。将质粒转化(transform)到克隆感受态细胞系中以生产另外的质粒DNA。随后将质粒转化到用于蛋白合成的表达感受态细胞系(TAM 1大肠杆菌细胞(ActiveMotif.Carlsbad,CA))中。
将γ-晶状体球蛋白质粒DNA化学转化到用于蛋白合成的M15pRep大肠杆菌细胞中。生长1L培养物用于蛋白质纯化。通过Ni亲和色谱法纯化γ-晶状体球蛋白蛋白质。包含在γ-晶状体球蛋白蛋白质上的N端His标签优先结合至Ni柱。可以用咪唑梯度洗脱结合蛋白质,该咪唑梯度竞争性地结合至Ni,释放纯化的蛋白质。用于纯化流程简要地是(如前所述):将蛋白质负载在柱上,以及首先用20mM(0%B)、然后35mM(10%B)、然后60mM(20%B)咪唑洗涤。然后在150mM(57%B)咪唑下洗脱蛋白质。通过SDS PAGE凝胶电泳和快速蛋白质液相层析(FPLC)确认纯度。
化学修饰方案:
1)使用HiTrapTM脱盐柱将一批纯化蛋白质缓冲交换至PBS pH=6.8。
(2)在添加CA(PEG)n之前测量浓度(A280)。
(3)将100mg的CA(PEG)n添加到3-5ml批量的以上蛋白质中。
(4)在250RPM下在4℃下将混合物放置在振荡器上过夜。
实施例1:未修饰的γD-晶状体球蛋白蛋白质的动态光散射
首先,通过DLS研究0.1mg/mL浓度下的未修饰的γD-晶状体球蛋白蛋白质的溶液。针对延迟时间(滞后时间)绘制测量的相关函数g2-1,在图2中给出(散射角30°)。在图3中给出各种松弛时间的重量的CONTIN分析(散射角在30°至60°的范围)。可以清楚的是溶液中存在两个群体,一个对应于聚集体,以及另一个对应于未聚集的蛋白质。如图4中给出的使用动态光散射的理论分析这些数据显示相应的回转的流体动力学半径(hydrodynamicradius of gyration)分别是64nm和1.2nm。
实施例2:用CA(PEG)4修饰的γD-晶状体球蛋白蛋白质的动态光散射
图5-7中给出了如实施例1通过CA(PEG)4修饰的相应结果,结果是聚集体不存在以及未聚集的蛋白质的尺寸是2nm。浓度是0.9mg/mL蛋白质以及33mg/ml CA(PEG)4。CA(PEG)4是

共价连接至分子刚毛的取代的或未取代的胺、琥珀酰亚胺、羧酸、异氰酸酯、异硫氰酸酯、磺酰氯、醛、碳二亚胺、酰基叠氮化物、酸酐、氟苯、碳酸酯、N-羟基琥珀酰亚胺酯、亚氨酯、环氧化物或氟苯基酯;
其中,所述分子刚毛是具有1至3个氧乙烯基基团的聚乙二醇、具有1至3个烷氧基乙烯基基团的烷氧基-聚乙二醇、或具有1至3个芳氧基乙烯基基团的芳氧基聚乙二醇。
2.根据权利要求1所述的眼科组合物,其中,所述双官能分子包含NH2、N-羟基琥珀酰亚胺或COOH。
3.根据权利要求1所述的眼科组合物,其中,所述双官能分子是

其中,所述第二分子刚毛是聚乙二醇、烷氧基-聚乙二醇、或烷氧基聚乙二醇,具有4至200个氧乙烯基、烷氧基乙烯基或芳氧基乙烯基基团;聚(2-羟基丙基)甲基丙烯酰胺(HPMA);聚(2-羟基乙基)甲基丙烯酸酯(HEMA);聚(2-噁唑啉)、聚(间-胆碱磷酸)、聚赖氨酸、或聚谷氨酸,所述第二分子刚毛具有150至8000的分子量。
5.根据权利要求1所述的眼科组合物,其中,所述眼科组合物是滴眼剂。
6.根据权利要求1所述的眼科组合物,其中,所述眼科组合物为眼科溶液、眼科悬浮液、眼科软膏、喷雾、或眼科凝胶的形式。
7.根据权利要求6所述的眼科组合物,其中,所述眼科组合物包含缓冲剂、等渗剂、增溶剂、防腐剂、增粘剂、螯合剂、防氧化剂、抗生素、糖、pH调节剂、或它们的组合。
8.根据权利要求1所述的眼科组合物,其中,所述眼科组合物是眼科设备。
9.根据权利要求8所述的眼科组合物,其中,所述眼科设备为隐形眼镜或泪点塞的形式。
10.根据权利要求1所述的眼科组合物,其中,所述眼科组合物是缓释组合物。
11.根据权利要求4所述的眼科组合物,其中,所述眼科组合物是缓释组合物。
12.一种抑制或逆转眼睛中的白内障形成、老花眼、或年龄有关的晶状体退化的进展的方法,包括
