背景技术
由人或小鼠早期胚胎确立,胚胎干细胞(ES细胞)能够长期培养,同时维持其分化成在活生物中发现的所有类型细胞的潜力。具有这个特征,人ES细胞预期充当用于许多疾病的细胞移植疗法,包括帕金森氏病、青少年糖尿病和白血病。然而,ES细胞移植造成与器官移植一样引起排斥的问题。此外,从伦理观点来看,不少人反对由破坏人胚胎确立的ES细胞的使用。

发明内容
技术问题
本发明的目的是提供无需使用病毒载体例如逆转录病毒通过使体细胞重编程序产生iPS细胞的方法。
解决问题的技术方案
本发明人广泛研究以解决上文描述的问题,并且发现通过借助于非病毒表达载体例如质粒载体将编码重编程序因子的基因引入体细胞内可以产生iPS细胞,并且通过该方法可以由体细胞获得安全的iPS细胞。本发明已基于这些发现而发展。
因此,本发明提供了产生诱导的多潜能干细胞的方法,其包括将至少一种非病毒表达载体引入体细胞内的步骤,所述非病毒表达载体整合了编码重编程序因子的至少一种基因。
在优选实施方案中,本发明提供了其中载体是在染色体外自主可复制的非病毒表达载体的上述方法;和其中载体是质粒载体的上述方法。
在另一个优选实施方案中,本发明提供了其中编码重编程序因子的基因是通过WO 2005/80598中描述的筛选核重编程序因子的方法选择的基因之一或多种此种基因的组合的上述方法;和其中编码重编程序因子的基因是选自下述的一种或多种基因、优选2种基因的组合、更优选3种基因的组合、特别优选4种或更多种基因的组合的上述方法:Oct家族基因、Klf家族基因、Sox家族基因、Myc家族基因、Lin家族基因和Nanog基因。
更优选的组合是(a)由Oct家族基因和Sox家族基因组成的2种基因的组合;(b)由Oct家族基因、Klf家族基因和Sox家族基因组成的3种基因的组合;(c)由Oct家族基因、Klf家族基因、Sox家族基因和Myc家族基因组成的4种基因的组合;(d)由Oct家族基因、Sox家族基因、Lin家族基因和Nanog基因组成的4种基因的组合;(e)由Oct家族基因、Klf家族基因、Sox家族基因、Myc家族基因、Lin家族基因和Nanog基因组成的6种基因的组合;等等。此外,还优选在组合中包括TERT基因和/或SV40 Large T抗原基因。视情况而定,优选排除Klf家族基因。
其特别优选的组合是由Oct3/4和Sox2组成的2种基因的组合;由Oct3/4、Klf4和Sox2组成的3种基因的组合;由Oct3/4、Klf4、Sox2和c-Myc组成的4种基因的组合;由Oct3/4、Sox2、Lin28和Nanog组成的4种基因的组合;以及由Oct3/4、Klf4、Sox2、c-Myc、Lin28和Nanog组成的6种基因的组合。还优选在这些组合中包括TERT基因和/或SV40 Large T抗原基因。视情况而定,优选排除Klf4。

本发明还提供了由上述诱导的多潜能干细胞诱导和分化的体细胞。
本发明还提供了干细胞疗法,其包括给患者移植通过诱导的多潜能干细胞的分化诱导获得的体细胞,所述诱导的多潜能干细胞使用从患者分离的体细胞通过上述方法获得。
本发明进一步提供了使用各种细胞评估化合物、药物、有毒物质等的生理活性和毒性的方法,所述各种细胞通过经由上述方法获得的诱导的多潜能干细胞的分化诱导获得。
发明的有益效果
无需使用整合到染色体内的载体例如逆转录病毒产生,由本发明提供的诱导的多潜能干细胞是有利的,因为肿瘤发生和其他问题在通过使诱导的多潜能干细胞分化获得的体细胞和组织中不出现。在本发明的优选实施方案中,在通过本发明的方法产生的诱导的多潜能干细胞中,引入的至少一种非病毒表达载体的全部或一些附加型地存在,基本上不整合到染色体中。因此,本发明的方法使得可能由例如患者的体细胞制备高度安全的诱导的多潜能干细胞,并且通过使这种细胞分化获得的细胞(例如,心肌细胞、胰岛素生产细胞或神经细胞等)可以安全地用于干细胞移植疗法,以用于广泛多样的疾病,包括心力衰竭、胰岛素依赖性糖尿病、帕金森氏病和脊髓损伤。

本发明的方法预期产生诱导的多潜能干细胞,包括将至少一种非病毒表达载体引入体细胞内的步骤,所述非病毒表达载体整合了编码重编程序因子的至少一种基因。非病毒表达载体优选是在染色体外自主可复制的表达载体,更优选质粒表达载体。
作为用于鉴定核重编程序因子的方法的例子,可以利用WO2005/80598中描述的核重编程序因子筛选法。其中的全部公开内容引入本文作为参考。通过参考上述出版物,本领域技术人员能够筛选核重编程序因子,并且利用它们用于本发明的方法。还可能使用由上述筛选法修饰或改变的方法鉴定核重编程序因子。
编码重编程序因子的基因组合的一些例子公开于WO2007/69666中。其中的全部公开内容引入本文作为参考。适当时通过参考上述出版物,本领域技术人员能够选择适合于在本发明的方法中使用的基因。编码重编程序因子的基因组合的其他例子在Science,318,第1917-1920页,2007、WO2008/118820等中给出。因此,本领域技术人员能够理解编码核重编程序因子的基因组合的多样性;通过利用WO 2005/80598中描述的核重编程序因子筛选法,除WO2007/69666和Science,2007(同上)中描述的组合外的合适基因组合可以在本发明的方法中使用。
编码重编程序因子的优选基因包括选自下述的一种或多种基因、优选2种基因的组合、更优选3种基因的组合、并且特别优选4种基因的组合:Oct家族基因、Klf家族基因、Sox家族基因、Myc家族基因、Lin家族基因和Nanog基因。
Oct家族基因、Klf家族基因、Sox家族基因和Myc家族基因的例子在WO2007/69666中给出。同样,对于Lin家族基因,本领域技术人员同样能够提取家族基因。例如作为Lin家族基因的例子,可以包括Lin28和Lin28B。
更优选的组合包括但不限于,
(a)由Oct家族基因和Sox家族基因组成的2种基因的组合;
(b)由Oct家族基因、Klf家族基因和Sox家族基因组成的3种基因的组合;

所有这些基因在哺乳动物包括人中共同存在。得自任选选择的哺乳动物(例如,人、小鼠、大鼠、牛、羊、马、猴)的基因可以在本发明中使用。除野生型基因外,也可以使用这样的突变基因,其翻译产物具有置换、插入和/或缺失的几个(例如,1-10个、更优选1-6个、更优选1-4个、更优选1-3个、特别优选1或2个)氨基酸,且具有与野生型基因产物的那种相似的功能。例如,作为c-Myc基因,可以使用野生型,编码稳定型突变体(T58A)的基因等。这同样适用于其他基因产物。

需要时,Klf家族基因可以从上述组合中排除。
此外,除上述基因外,可以组合选自Fbx15、ERas、ECAT15-2、Tcl1和β-联蛋白的一种或多种基因,和/或也可以组合选自ECAT1、Esg1、Dnmt3L、ECAT8、Gdf3、Sox15、ECAT15-1、Fthl17、Sall4、Rex1、UTF1、Stella、Stat3和Grb2的一种或多种基因。这些组合在WO2007/69666中具体描述。
如果一种或多种这些基因已在待重编程序的体细胞中表达,那么一种或多种基因可以从待引入的基因中排除。当使用待整合到染色体内的载体将一种或多种这些基因引入待重编程序的体细胞内时,剩余的一种或多种基因可以根据本发明的方法使用非病毒表达载体引入。可替代地,当这些基因的一种或多种基因产物借助于融合蛋白或核显微注射引入核内时,剩余的一种或多种基因可以根据本发明的方法使用非病毒表达载体引入。
特别优选的基因组合是,
(1)由Oct3/4和Sox2组成的2种基因的组合;
(2)由Oct3/4、Klf4和Sox2组成的3种基因的组合;
(3)由Oct3/4、Klf4、Sox2和c-Myc组成的4种基因的组合;
(4)由Oct3/4、Sox2、Lin28和Nanog组成的4种基因的组合;
(5)由Oct3/4、Sox2、c-Myc、TERT和SV40 Large T抗原组成的5种基因的组合;
(6)由Oct3/4、Klf4、Sox2、c-Myc、TERT和SV40 Large T抗原组成的6种基因的组合;
(7)由Oct3/4、Klf4、c-Myc、Sox2、Lin28和Nanog组成的6种基因的组合;
(8)由Oct3/4、Klf4、c-Myc、Sox2、Lin28、Nanog和TERT或SV40 Large T抗原组成的7种基因的组合,
等等。
除上述基因外,可以进一步组合编码诱导细胞无限增殖化的因子的基因。如WO2007/69666中公开的,例如,适当时,可以单独或组合使用选自TERT基因和下述基因的一种或多种基因:HPV16E6、HPV16E7和Bmil。
当重编程序使用内源表达Sox2和c-Myc神经干细胞等作为体细胞来源执行时,还可以提及由Oct3/4和Klf4组成的2种基因的组合、或由Oct3/4和c-Myc组成的2种基因的组合(参见2008年6月29日在线公开的Nature,第1-5页(doi:10.1038/nature07061))。
在上文组合(3)、(5)、(6)和(7)中,可以使用L-Myc代替c-Myc。
应当指出基因组合并不限于其。此外,本发明的范围包括其中使用非病毒表达载体将选自上述基因的一种或多种基因引入体细胞内,并且通过另一种方式将剩余基因或基因产物引入体细胞内的方法。例如,还可能使用非病毒表达载体将选自上述基因的一种或多种基因引入体细胞内,并且使用病毒载体例如逆转录病毒载体、慢病毒载体、腺病毒载体、腺伴随病毒载体、仙台病毒载体,将剩余基因引入体细胞内。
当使用非病毒表达载体将编码重编程序因子的2种或更多种基因引入体细胞内时,待引入的2种或更多种基因中的一些可以在与其他基因的那种不同的时间引入体细胞内,或待引入的所有种类的基因可以同时引入体细胞内;然而,优选待引入的所有基因同时引入体细胞内。当2种或更多种不同非病毒表达载体用于引入2种或更多种基因时,所有种类的非病毒表达载体可以同时引入体细胞内;这代表本发明的优选实施方案。
在本发明的方法中,作为编码重编程序因子的基因,例如,可以使用由Oct家族基因、Klf家族基因、Sox家族基因和Myc家族基因组成的4种基因的组合。还可以使用由Oct家族基因、Klf家族基因和Sox家族基因组成的3种基因的组合,或选自上述3种基因的2种基因的组合。
在本发明的方法中,优选上述4种、3种或2种基因同时引入体细胞内。为了引入上述4种、3种或2种基因,可以使用整合了所有这些基因的一种非病毒表达载体。可替代地,适当时,可以组合使用几种非病毒表达载体,以便涵盖这些基因的所有组合。当使用几种非病毒表达载体时,优选的是优选使用2种或3种、更优选2种非病毒表达载体。优选这些非病毒表达载体同时引入体细胞内。
如果引入的基因数目超过4种,那么适当时,可以组合几种非病毒表达载体,以便涵盖这些基因的所有组合。当使用几种非病毒表达载体时,优选的是优选使用2种至5种、更优选2种至4种、更优选3种或4种非病毒表达载体。这些非病毒表达载体优选同时引入体细胞内。
优选方法的例子是其中具有Oct家族基因、Klf家族基因和Sox家族基因的一种非病毒表达载体,以及具有Myc家族基因的一种非病毒表达载体同时或在不同时间引入体细胞内的方法;在这种方法中,优选2种非病毒表达载体同时引入体细胞内。在另一个优选实施方案中,还可能使用其中具有Oct家族基因、Klf家族基因、Sox家族基因和Myc家族基因的一种非病毒表达载体引入体细胞内的方法。
在本发明的优选实施方案中,可以使用由Oct3/4、Klf4、Sox2和c-Myc组成的4种基因的组合,或选自这4种基因的3种或2种的任选选择组合,优选3种或2种基因的组合,其中所述组合不包含c-Myc。这个优选实施方案在下文中具体描述,本发明的范围决不限于其。

在上文背景中,当将多种基因(例如,Oct家族基因、Klf家族基因和Sox家族基因)整合入一种非病毒表达载体中时,这些基因可以优选与使得能够进行多顺反子表达的间插序列一起插入非病毒表达载体内。通过使用使得能够进行多顺反子表达的间插序列,可能更有效表达在一种非病毒表达载体中整合的多种基因。使得能够进行多顺反子表达的有用序列包括例如,口蹄疫病毒的2A序列(SEQ ID NO:61,有时称为FMDV 2A自加工序列)(PLoS ONE 3,e2532,2008;Stem Cells 25,1707,2007)、IRES序列等,优选2A序列。更具体而言,当构建这样的非病毒表达载体时,所述非病毒表达载体具有以这个顺序以从5′到3′末端方向的(i)Oct3/4、Klf4和Sox2、(ii)Oct3/4和Klf4、(iii)Klf4和Sox2、(iv)Oct3/4和Sox2、(v)Sox2和Klf4或(vi)c-Myc、Lin28和Nanog,优选将2A序列插入这些基因之间。因此,本发明还提供了2A序列用于制备用于iPS细胞诱导的非病毒表达载体的用途,所述非病毒表达载体具有2种或更多种重编程序因子。
将非病毒表达载体引入体细胞内的操作的重复次数并无具体限制,只要可以完成本发明使体细胞重编程序以产生诱导的多潜能干细胞的作用,转染可以执行一次或更多任选选择次数(例如,1次至10次、1次至5次等)。当将2种或更多种非病毒表达载体引入体细胞内时,优选这些所有种类的非病毒表达载体同时引入体细胞内;然而,即使在这种情况下,转染也可以执行一次或更多任选选择次数(例如,1次至10次、1次至5次等),优选地转染可以重复执行2次或更多次(例如,3次或4次)。
当转染重复2次或更多次时,时间间隔由下述例示但不限于下述:12小时至1周、优选12小时至4天,例如1天至3天。
如本文所使用的,术语“诱导的多潜能干细胞(iPS细胞)”指具有与ES细胞的那种相似的性质的细胞,更具体而言包括由具有多潜能性和增殖(自我更新)能力的体细胞重编程序的未分化细胞。然而,应当指出,这个术语不得在各种意义上解释为限制性的,并且必须以最广泛的含义解释。在WO2005/80598(在这个出版物中,使用术语ES样细胞)中描述了借助于假定核重编程序因子制备诱导的多潜能干细胞的方法,并且还特别描述了分离诱导的多潜能干细胞的方法。WO2007/69666公开了重编程序因子的具体例子和使用其使体细胞重编程序的方法。因此,希望在实现本发明中,本领域技术人员参考这些出版物。
除编码重编程序因子的基因外,转录所需的调节序列(例如,启动子、增强子和/或终止子等)优选与非病毒表达载体中的基因可操作地连接。
作为启动子,可以使用在体细胞中显示转录活性的DNA序列,并且适当时可以根据动物物种和体细胞种类选择启动子。可以在哺乳动物细胞中表达的有用启动子的例子包括巨细胞病毒(人CMV)的IE(立即早期)基因启动子、SV40起始启动子、逆转录病毒启动子、金属硫蛋白启动子、热激启动子、SRα启动子等。人CMV的IE基因增强子可以与启动子一起使用。有用的启动子是CAG启动子(包含巨细胞病毒增强子、鸡β-肌动蛋白启动子和β-珠蛋白基因多腺苷酸化信号位点)。
非病毒表达载体可以整合允许表达载体在哺乳动物体细胞中自主复制的DNA序列。DNA序列的例子是SV40复制起点。
非病毒表达载体优选是在染色体外自主可复制的表达载体,并且非病毒表达载体优选是不整合到染色体中的那种。更优选的例子包括质粒载体。质粒载体的例子包括但不限于,大肠杆菌(Escherichia coli)衍生的质粒(ColE系列质粒例如pBR322、pUC18、pUC19、pUC118、pUC119和pBluescript等)、放线菌属(Actinomyces)衍生的质粒(pIJ486等)、枯草芽孢杆菌(Bacillus subtilis)衍生的质粒(例如,pUB110、pSH19及其他)、酵母衍生的质粒(YEp13、YEp 24、Ycp50等)等,以及人工质粒载体等。
容易获得的非病毒表达载体的例子包括但不限于pCMV6-XL3(OriGene Technologies Inc.)、EGFP-C1(Clontech)、pGBT-9(Clontech)、pcDNAI(FUNAKOSHI)、pcDM8(FUNAKOSHI)、pAGE107(Cytotechnology,3,133,1990)、pCDM8(Nature,329,840,1987)、pcDNAI/AmP(Invitrogen)、pREP4(Invitrogen)、pAGE103(J.Blochem.,101,1307,1987)、pAGE210等。
需要时,非病毒表达载体可以可以整合选择标记。选择标记的例子包括在宿主细胞中缺陷的基因,例如二氢叶酸还原酶(DHFR)基因或粟酒裂殖糖酵母(Schizosaccaromyces pombe)TPI基因,以及关于对药物例如氨苄青霉素、卡那霉素、四环素、氯霉素、新霉素或潮霉素的抗性的基因。
虽然引入体细胞内的非病毒表达载体例如质粒载体一般不整合到细胞的基因组内,但在关于iPS细胞诱导的选择压力下,由于重编程序因子的稳定表达的必要性,可能观察到非病毒表达载体增加的整合效率。因此,当目的iPS细胞预期用于再生医学等时,非病毒表达载体可以优选包含使得能够切除转基因的序列,例如loxP序列(Chang等人,在2009年2月12日在线公开的STEM CELLS(doi:10.1002/stem.39))、piggyback转座子(Kaji等人,在2009年3月1日提前在线公开的Nature(doi:10.1038/nature07864);Woltjen等人,在2009年3月1日提前在线公开的Nature(doi:10.1038/nature07863))和在启动子区中的四环素应答元件(Tet-OnR & Tet-Off R Gene Expression Systems,Clontech)。
使在本发明中使用的编码重编程序因子的基因、启动子、增强子和/或终止子等以合适顺序连接,以构建能够在体细胞中表达重编程序因子的非病毒表达载体的方法,对于本领域技术人员是显而易见的。
当使用编码重编程序因子的2种或更多种基因时,基因可以整合入一种非病毒表达载体中。可替代地,可以使用整合了不同基因的2种或更多种非病毒表达载体。在后面一种情况下,适当时,可以组合整合了2种或更多种基因的一种非病毒表达载体和整合了与其不同的一种或多种基因的非病毒表达载体。
本领域技术人员可获得的将表达载体引入动物细胞内的任何方法都可以用于将非病毒表达载体引入体细胞内。有用方法的例子包括转染试剂例如FuGENE 6转染试剂(Roche)的使用、microporator的使用、电穿孔法、磷酸钙法、脂转染法、DEAE-葡聚糖介导的转染法、转染法、显微注射法、阳离子脂质介导的转染法等。核转染(nucleofection)也可以用于引入基因。这些方法可以组合使用。
在将非病毒表达载体引入体细胞内中,表达载体可以引入在饲养细胞上培养的体细胞内,并且可以仅引入体细胞内。为了增加表达载体引入效率,后面一种方法有时是合适的。使用的饲养细胞可以是用于培养胚胎干细胞的那些;例如,可以使用用化学试剂例如丝裂霉素C处理或暴露于辐射等的来自14至15天小鼠胚胎的原代培养成纤维细胞、STO(成纤维细胞衍生的细胞系)等。
通过在合适条件下培养整合了非病毒表达载体的体细胞,可能允许核重编程序自主进展,并且由体细胞产生诱导的多潜能干细胞。培养整合了非病毒表达载体的体细胞以获得诱导的多潜能干细胞的步骤可以以与使用逆转录病毒的常规方法相同的方式进行;例如,这可以如出版物例如Cell,126,第1-14页,2006;Cell,131,第1-12页,2007;和Science,318,第1917-1920页,2007中所述来达到。在产生人诱导的多潜能干细胞中,有时希望在表达载体引入后的细胞培养密度设定为比关于普通动物细胞培养的那种更低的水平。例如,优选以10,000至100,000细胞、优选约50,000细胞/细胞培养皿的细胞密度继续培养。任何培养基都可以用于培养,适当时由本领域技术人员选择;例如,在产生人诱导的多潜能干细胞中,有时优选使用适合于人ES细胞培养的培养基。关于培养基和培养条件的选择,上述出版物充当参考。
所得到的诱导的多潜能干细胞可以使用未分化细胞特有的各种标记进行鉴定;用于这种鉴定的方法也在上述出版物中具体且详细地描述。允许维持ES细胞的未分化状态和多潜能性的各种培养基或不允许维持这些性质的培养基是本领域已知的;通过组合使用合适的培养基,可以有效分离诱导的多潜能干细胞。通过利用用于ES细胞的共同鉴定方法,分离的诱导的多潜能干细胞的分化潜力和增殖潜力对于本领域技术人员可容易证实。当所得到的诱导的多潜能干细胞在合适条件下增殖时,获得诱导的多潜能干细胞集落;可能基于集落形状鉴定诱导的多潜能干细胞的存在。例如,已知小鼠诱导的多潜能干细胞形成隆起集落,而人诱导的多潜能干细胞形成扁平集落,并且这些集落的形状分别与小鼠ES细胞和人ES细胞集落的那些极其类似;因此,本领域技术人员可能基于集落形状鉴定所得到的诱导的多潜能干细胞。当使用体细胞进行重编程序时,所述体细胞具有在ES细胞中特异性表达的基因启动子下游整合标记基因例如GFP的基因,如果细胞变得对于标记(GFP)阳性,则可能鉴定诱导的多潜能干细胞。
待通过本发明的方法重编程序的“体细胞”指任何细胞,除全能和多潜能细胞例如早期胚胎和ES细胞外,并且它的选择并无限制。例如,还可以使用处于胎儿期中的体细胞、新生儿体细胞和成熟体细胞。优选地,使用得自哺乳动物包括人的体细胞;更优选使用人或小鼠衍生的体细胞。具体地,可以提及(1)组织干细胞(体干细胞(somatic stem cells))例如神经干细胞、造血干细胞、间充质干细胞和牙髓干细胞,(2)组织祖细胞,或(3)分化的细胞例如淋巴细胞、上皮细胞、肌细胞、成纤维细胞(真皮细胞等)、毛细胞、肝细胞和胃粘膜细胞。当诱导的多潜能干细胞用于治疗疾病时,希望使用从待治疗的患者或共享与患者的那种相同类型HLA的另一个人分离的体细胞;例如,可以使用牵涉于疾病的体细胞和牵涉于疾病治疗的体细胞。
在本发明中,为了增加诱导的多潜能干细胞确立的效率,除引入本发明的非病毒表达载体外,还可以引入或添加各种确立效率改进剂。iPS细胞确立效率改进剂的例子包括但不限于,组蛋白脱乙酰酶(HDAC)抑制剂[例如,丙戊酸(VPA)(Nat.Biotechnol.,26(7):795-797(2008)),低分子抑制剂例如制滴菌素A、丁酸钠、MC 1293和M344,基于核酸的表达抑制剂例如针对HDAC的siRNA和shRNA(例如,HDAC1 siRNA(Millipore)、针对HDAC1的HuSH 29聚体shRNA构建体(OriGene)等),等],G9a组蛋白甲基转移酶抑制剂[例如,低分子抑制剂例如BIX-01294(Cell Stem Cell,2:525-528(2008)),基于核酸的表达抑制剂例如针对G9a的siRNA和shRNA(例如,G9a siRNA(人)(Santa Cruz Biotechnology)等),等],L-通道钙激动剂(例如,Bayk8644)(Cell Stem Cell,3,568-574(2008)),UTF1(Cell Stem Cell,3,475-479(2008)),Wnt信号传导(例如,可溶性Wnt3a)(Cell StemCell,3,132-135(2008)),2i/LIF(2i是促分裂原活化蛋白激酶信号传导和糖原合酶激酶-3的抑制剂;PloS Biology,6(10),2237-2247(2008)),p53抑制剂(例如,针对p53的siRNA和shRNA(Cell StemCell,3,475-479(2008))等。基于核酸的表达抑制剂可以为具有编码siRNA或shRNA的DNA的表达载体形式。在这种情况下,编码siRNA或shRNA的DNA可以连同重编程序因子一起插入本发明的非病毒表达载体内。
对通过本发明的方法产生的诱导的多潜能干细胞不实施关于其用途的限制,并且其可以用于使用ES细胞的所有类型的研究和调查,以及用于使用ES细胞的疾病治疗,以代替ES细胞。例如,通过用视黄酸、生长因子例如EGF或糖皮质激素等处理经由本发明的方法由从患者收集的体细胞获得的诱导的多潜能干细胞,可以诱导所需分化细胞(例如,神经细胞、心肌细胞、血细胞等)以形成合适组织。通过将因此获得的分化细胞或组织送回患者,可以完成通过自体细胞移植的干细胞疗法。应当指出本发明的诱导的多潜能干细胞的用途并不限于上述具体实施方案。
本发明还提供了用于在产生诱导的多潜能干细胞的上述方法中使用的非病毒表达载体,即整合了编码重编程序因子的至少一种基因的非病毒表达载体(优选质粒载体)。载体的结构如本发明的产生诱导的多潜能干细胞方法部分中详细描述的。
例子是整合了Oct家族基因、Klf家族基因和Sox家族基因的非病毒表达载体,优选以这个顺序以从5′到3′末端方向整合。更优选的例子是这些基因与使得能够进行多顺反子表达的间插序列一起整合的非病毒表达载体,特别优选其中OCT3/4、Klf4和Sox 2与使得能够进行多顺反子表达的间插序列一起,以这个顺序以从5′到3′末端方向整合的非病毒表达载体,所述间插序列优选FMDV 2A自加工序列。
因为引入体细胞内的非病毒表达载体例如质粒载体一般不整合到细胞的基因组内,所以在优选实施方案中,本发明提供了其中转基因不整合到基因组内的诱导的多潜能干细胞。因为此种iPS细胞减少在由其分化的组织或器官中引起肿瘤发生和/或干扰(例如,破坏或激活)内源基因的危险,所以它可以优选用于再生医学例如细胞移植疗法。
然而,在关于iPS细胞诱导的选择压力下,由于重编程序因子的稳定表达的必要性,可以观察到非病毒表达载体增加的整合效率。因此,在另一个优选实施方案中,本发明提供了其中转基因以质粒形式整合到基因组内的诱导的多潜能干细胞。与通过逆转录病毒感染诱导的iPS细胞相比较,此种iPS细胞可以减少在由其分化的组织或器官中引起肿瘤发生的危险。此外,需要时,可以使用Cre/loxP系统(Chang等人,2009(同上))或piggyback转座子载体和piggyback转座子(Kaji等人,2009(同上);Woltjen等人,2009(同上))或四环素依赖性基因诱导,从基因组中切除转基因。使用质粒载体或腺病毒载体,可以将用于切除的Cre重组酶或转座酶引入iPS细胞内且在iPS细胞中表达。在使用四环素依赖性基因诱导的情况下,同时表达Tet阻抑蛋白或突变的Tet阻抑蛋白。
实施例
本发明在下文中借助于下述实施例更详细地描述,然而,所述实施例不应解释为限制本发明的范围。
实施例1
具有Nanog报道分子的小鼠用作实验系统(Okita等人Nature,第448卷,第313-317页,2007)。通过将EGFP和嘌呤霉素抗性基因整合入购自BACPAC Resources的BAC(细菌人工染色体)的Nanog基因座内制备这些小鼠。小鼠Nanog基因在多潜能细胞例如ES细胞和早期胚胎中特异性表达。已显示对这种报道分子阳性的小鼠iPS细胞具有几乎等价于ES细胞的那种的分化潜能。使这些Nanog报道小鼠与Fbx 15报道小鼠交配(Tokuzawa等人Mol Cell Biol,第23卷,2699-2708(2003)),由此产生具有Nanog报道分子和Fbx 15报道分子的突变小鼠。
用于重编程序的质粒这样进行制备:用EcoRI处理pCX-EGFP(由在Osaka University的Dr.Masaru Okabe提供的质粒:FEBS Letters,407,313-319,1997),并且插入其中Oct3/4、Sox2和Klf4(全是小鼠衍生的基因)的编码区经由口蹄疫病毒的2A序列以Oct3/4、Klf4和Sox2的顺序连接的构建体代替EGFP(pCX-2A-mOKS;图2)。同样,制备具有在其中插入的c-Myc编码区的质粒(pCX-c-Myc;图2)。
在制备2A序列以及Oct3/4、Klf4和Sox2连接在一起的构建体中,首先,使有义和反义寡核苷酸退火且插入用XbaI和PstI消化的pBluescript II KS(-)载体内,所述寡核苷酸包含口蹄疫病毒的2A序列(SEQ ID NO:61)、上游限制性内切核酸酶位点(XbaI和BglII)、和下游限制性内切核酸酶位点(BspHI、Mfel和PstI)(pBS-2A)。随后,通过PCR扩增编码Oct3/4或Klf4的小鼠cDNA,用BamHI位点替换翻译终止密码子,并且将每种cDNA克隆到pCR2.1内。随后,使用合适的限制性内切核酸酶,使Oct3/4和Klf4的cDNAs与pBS-2A连接在一起,以产生pBS-Oct3/4-2A和pBS-Klf4-2A。随后,使用合适的限制性内切核酸酶,将Klf4-2A框内插入pBS-Oct3/4-2A,由此产生pBS-Oct3/4-2A-Klf4-2A。随后,使用合适的限制性内切核酸酶,使所得到的Oct3/4-2A-Klf4-2A构建体框内与具有翻译终止密码子的Sox2cDNA连接在一起。最后,使所得到的其中2A序列与Oct3/4、Klf4和Sox2连接在一起的Oct3/4-2A-Klf4-2A-Sox2-STOP构建体插入pCX-EGFP的EcoRI位点内,从而制备pCX-2A-mOKS。
从上述突变小鼠胎儿(受精后13.5天)中分离成纤维细胞(MEFs)。不表达Nanog基因,MEFs不表达产生绿色荧光的EGFP。同样,将MEFs以1.3x105细胞/孔播种至先前用0.1%明胶(Sigma)包被的6孔培养板(Falcon)。使用的培养基是DMEM/10%FCS(补加有10%胎牛血清的DMEM(Nacalai Tesque),MEFs在37℃、5%CO2下进行培养。第二天,将4.5μL FuGene6转染试剂(Roche)加入100μL Opti-MEM IReduced-Serum Medium(Invitrogen)中,并且允许培养基在室温下静置5分钟。其后,加入1.5μg表达载体(pCX-2A-mOKS),并且允许培养基在室温下静置15分钟,这之后将培养基加入MEF培养基中。第二天,取出培养基,并且如上所述用FuGene6转染试剂引入1.5μg另一种表达载体(pCX-c-Myc)。
第二天,用新鲜供应(DMEM/10%FCS)替换培养基,并且如上所述引入表达载体(pCX-2A-mOKS);第二天,用ES细胞培养基(补加有15%胎牛血清、2mM L-谷氨酰胺(Invitrogen)、100μM非必需氨基酸(Invitrogen)、100μM 2-巯基乙醇(Invitrogen)、50U/mL青霉素(Invitrogen)和50mg/mL链霉素(Invitrogen)的DMEM(NacalaiTesque))替换培养基,并且如上所述使用FuGene6转染试剂引入表达载体(pCX-c-Myc)。
第二天,用ES细胞培养基替换培养基。在播种后第9天时,取出MEF培养基,并且通过添加PBS 2mL洗涤细胞。取出PBS后,加入0.25%胰蛋白酶(Trypsin)/1mM EDTA(Invitrogen),并且使反应在37℃下执行约5分钟。细胞增加后,加入ES细胞培养基,使细胞悬浮,并且将1x106(Exp432A)或2x105(Exp432B)细胞播种到先前在其上已播种饲养细胞的100mm皿上。使用的饲养细胞是已用丝裂霉素C处理以终止其细胞分裂的SNL细胞。
随后,每2天用新鲜供应更换ES细胞培养基,直至出现可见集落;约第17天开始建群,并且约第24天观察到完全建群(图1)。上文时间表概括于图1和3中的Exp432中。
获得的细胞逐渐变得GFP阳性,显示与小鼠ES细胞的那种不能区分的形态学(图4中的432A-1),对于各种ES细胞标记在与ES细胞相似的水平下测试阳性(图5中的iPS-432A-1),并且产生成年嵌合小鼠。基于小鼠iPS细胞的集落形状特征以及GFP阳性结果和对其他非分化标记阳性的结果,得出结论为,通过将上述表达载体引入MEF细胞内,核重编程序完全进展以产生iPS细胞,并且iPS细胞增殖且形成可见集落。因此,这些结果显示无需使用逆转录病毒或慢病毒即可制备iPS细胞。PCR分析检测出上述表达载体整合到宿主基因组内(图6中的iPS-432A-1)。
实施例2
为了避免pCX-2A-mOKS和pCX-c-Myc整合到宿主基因组内,修改转染规程。
在实验开始后第1、3、5和7天时,一起转染pCX-2A-mOKS和pCX-c-Myc(图3中的Exp440)。结果,获得许多GFP阳性集落,并且产生在形态上与ES细胞不能区分的细胞(图4中的440A-3)。获得的细胞在与ES细胞相同的水平表达ES细胞标记(图5中的iPS-440A)。为了检查质粒DNA到基因组内的整合,设计了能够扩增质粒的每个部分的16组PCR引物(图2、13和14)。在通过修改规程获得的11个GFP阳性克隆的9个中,未观察到外源DNA的扩增(图6)。此外,在DNA印迹分析中,在这些克隆中未检测到外源基因的整合(图11)。尽管未明确排除小质粒片段的可能存在,但上文结果显示这些iPS细胞的确不具有整合到宿主基因组内的pCX-2A-mOKS和pCX-c-Myc质粒。
为了排除无整合的iPS细胞得自可能的污染Nanog-GFP ES细胞的可能性,执行SSLP分析。在图3中的Exp440中,使用来自5个胎儿的MEF细胞。在SSLP分析中,这5个胎儿是可区分的,并且鉴定无整合的iPS细胞的衍生(图12)。这个分析还显示无整合的iPS细胞与得自129S4品系的ES细胞不同(图12)。
实施例3
为了证实无整合的iPS细胞的多潜能性,将如实施例2中所述获得的iPS细胞皮下移植给裸鼠。测试的所有克隆(440A-3、-4、-8和-10)都产生肿瘤,所述肿瘤包括广泛多样的细胞类型,包括得自所有3个胚层的细胞(图7)。此外,将无整合的iPS细胞注射到ICR小鼠胚泡内。根据毛色判断,由注射的所有克隆(440A-3、-4、-6、-8、-9和-10)获得成年嵌合体(图8)。在这些嵌合小鼠中,PCR分析未检测出任何转基因的整合(图9)。PCR分析检测出在嵌合体中的Nanog和Fbx15报道分子(图9)。与无整合的iPS细胞自双重报道小鼠出现,以及本发明人的实验室未保存双重报道ES细胞的事实组合,这些结果显示嵌合体得自无整合的iPS细胞,而不是污染ES细胞。因此,这些结果证实无整合的iPS细胞具有多潜能性。
获得的71只嵌合小鼠及其后代的长期检查显示,在得自通过使用逆转录病毒引入4种基因(Oct3/4、Klf4、Sox2、c-Myc)制备的iPS细胞的嵌合小鼠及其后代中,与正常小鼠比较,死亡率较早开始增加,而得自无4种基因整合的iPS细胞的嵌合小鼠及其后代显示类似于正常小鼠的那种的存活曲线。
当获得的嵌合小鼠和野生小鼠交配时,获得F1小鼠;因此,证实无整合的iPS细胞对种系产生影响(种系传递)。
实施例4
人牙髓干细胞(克隆名称;DP31,PCT/JP2008/068320、J.Dent.Res.,87(7):676-681(2008))用作实验系统。使用如Cell,131,861-872(2007)中所述的慢病毒,允许DP31表达小鼠亲嗜性病毒受体Slc7a1基因。这些细胞使用MSCGM bullet试剂盒(Lonza)进行培养。
以与实施例1相同的方式,由pCX-EGFP(由在Osaka University的Dr.Masaru Okabe提供,FEBS Letters,407,313-319,1997)制备用于重编程序的质粒。具体地,用EcoRI处理pCX-EGFP,并且插入其中SOX2和KLF4的编码区经由口蹄疫病毒的2A序列连接的构建体代替EGFP,由此制备质粒pCX-hSK。同样,制备具有在其中c-Myc、Lin28和Nanog经由2A序列连接的质粒(pCX-hMLN)、具有在其中插入OCT3/4编码区的质粒(pCX-hOCT3/4)、和具有在其中插入SV40LargeT抗原的质粒(pCX-SV40LT)。
用PBS洗涤在100mm皿中培养的DP31,加入0.25%胰蛋白酶/1mM EDTA(Invitrogen),并且使反应在37℃下执行约5分钟。细胞增加后,加入MSCGM,使细胞悬浮,并且将6x105细胞回收在15mL管中。使细胞在800rpm下离心5分钟;取出上清液后,并且使用HumanDermal Fibroblast Nucleofector Kit(Amaxa)引入表达质粒。使用的质粒量对于pCX-hOCT3/4是0.5μg,对于pCX-hSK是1.0μg,对于pCX-hMLN是1.5μg,并且对于pCX-SV40LT是0.5μg。处理后,将细胞播种至6孔板。用MSCGM培养10天后,细胞再次用PBS洗涤,加入0.25%胰蛋白酶/1mM EDTA(Invitrogen),并且使反应在37℃下执行约5分钟。细胞增加后,加入MSCGM,使细胞悬浮,并且将1x106细胞播种在先前在其上已播种饲养细胞的100mm皿上。使用的饲养细胞是已用丝裂霉素C处理以终止其细胞分裂的SNL细胞。其后,直至开始观察到集落,每2天用新鲜供应替换培养基。通过使分别补加有MSCGM和bFGF(4ng/mL)的等体积灵长类动物ES细胞培养基(ReproCELL)混合来制备使用的培养基。约第19天开始建群,从而证实人iPS细胞的确立(图15)。
接下来,用与上文所述相同的7种基因转染胎儿人HDF(Cellapplications,INC)。转染后,使用补加有4ng/ml重组人bFGF(WAKO)的灵长类动物ES细胞培养基(ReproCELL)培养细胞。MSTO细胞充当饲养细胞。在转染后第31天时的细胞照片(5个克隆:203A-1至203A-5,其中挑取203A-4作为阴性对照)显示于图16中,并且在第二代传代培养物中的细胞照片显示于图17中。203A-1至203A-3和203A-5克隆显示一般的ES细胞样形态学,从而证实人iPS细胞的确立。
对这些细胞实施基因组PCR分析,并且检查转基因到基因组内的整合。结果显示于图18中。在所有克隆中,检测出Oct3/4(pCX-hOCT3/4)和c-Myc(pCX-hMLN)的整合。在除203A-4外的克隆中检测出Klf4(pCX-hSK)的整合。在任何一种克隆中都未检测出SV40LT(pCX-SV40LT)的整合。
实施例5
以与实施例4中相同的方式,用排除SV40Large T抗原的6种基因(pCX-hSK、pCX-hMLN、pCX-hOCT3/4)转染实施例4中使用的牙髓干细胞DP31。在转染后第35天时的细胞照片(5个克隆:217A-1至-4和-6)显示于图19中。在第二代传代培养物中的细胞照片显示于图20中。所有克隆都显示一般的ES细胞样形态学,从而证实人iPS细胞的确立。
对确立的这些人iPS细胞克隆(217A-1至217A-4、217A-6)实施基因组PCR分析。结果显示于图21中。在所有这些克隆中,证实转基因的整合。
实施例6
允许得自6岁大的日本女性的HDF细胞系(HDF-120;JCRB)表达Slc7a1基因。用上述6种基因和针对p53的shRNA(shRNA2:SEQ IDNO:62)(引入的载体:pCX-hOCT3/4、pCX-hSK、pCX-hMLN-shp53)转染所得到的细胞(HDF-120-Slc)。
使用Microporator(100μL尖端,1600V,10ms,3次),将pCX-hOCT3/4(0.5μg)、pCX-hSK(1.0μg)、和pCX-hMLN-shp53(1.5μg)的每一个电引入6.0x105HDF-120-Slc细胞内。10天后,每种载体再次在相同条件下电引入,并且将细胞播种在MSTO上(100mm皿)。这些细胞使用DMEM/10%FCS培养直至第10天时,其后使用补加有4ng/ml重组人bFGF(WAKO)的灵长类动物ES细胞培养基(ReproCELL)。在第一次电穿孔后第35天时的细胞照片显示于图22中。在传代培养后的细胞照片显示于图23中。显示一般的ES细胞样形态学,从而证实人iPS细胞的确立。基因组PCR分析证实转基因的整合(图24中的泳道279A-2)。
实施例7
根据实施例2中的规程,将分别整合了4种基因Oct3/4、Klf4、Sox2和c-Myc的表达载体(pCX-Oct4、pCX-Sox2、pCX-Klf4,pCX-c-Myc)引入得自Nanog报道小鼠(Okita等人Nature,第448卷,第313-317页,2007)的MEF细胞内。
首先,将Nanog报道MEF细胞播种到明胶包被的6孔板上(1.3x105细胞/孔),并且在第1、3、5和7天时,使用FuGene6由pCX-Oct4(0.37μg)、pCX-Sox2(0.36μg)、pCX-Klf4(0.39μg)和pCX-c-Myc(0.38μg)的每一个转染。在第9天时,将1x106细胞(1.0)或0.2x106细胞(0.2)播种到MSTO-PH或明胶(100-mm皿)上,并且在第25天时选择集落。选择后的细胞照片显示于图25中。获得小鼠iPS细胞特有的集落形状和GFP阳性结果,从而证实小鼠iPS细胞的确立。对确立的小鼠iPS细胞克隆(497A-1至A-5)实施基因组PCR分析。结果显示于图26中。497A-2和497A-5两者都显示是无任何外源基因整合的iPS细胞。
工业适用性
根据本发明的方法,可能例如由患者的体细胞制备高度安全的诱导的多潜能干细胞。通过使诱导的多潜能干细胞分化获得的细胞(例如,心肌细胞、胰岛素生产细胞、神经细胞等)可以安全地用于干细胞移植疗法,以用于用于广泛多样的疾病,包括心力衰竭、胰岛素依赖性糖尿病、帕金森氏病和脊髓损伤。

Claims (10)
1.一种产生诱导的多潜能干细胞的方法,其包括将至少一种质粒载体引入体细胞内的步骤,所述质粒载体整合了编码重编程序因子的至少一种基因,
其中将Oct3/4基因、K1f家族基因、Sox家族基因和Myc家族基因引入体细胞内,
其中所述K1f家族由K1f1、K1f2、K1f4和K1f5组成,
其中所述Sox家族由Sox1、Sox2、Sox3、Sox15和Sox17组成,且
其中所述Myc家族由c-Myc、L-Myc和N-Myc组成。
2.权利要求1的方法,其中所述质粒载体是在染色体外自主可复制的。
3.权利要求1的方法,其中引入的基因进一步包含一种或多种选自Lin家族基因、Nanog基因、TERT基因和SV40 Large T抗原基因的基因,
其中所述Lin家族由Lin28和Lin28b组成。
