Description
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Polyenverbindungen.
Aus den Schweizerischen Patentschriften No. 544 057, 564 504 und 565 735 sind Anhydroretinonsäurederivate mit tumorhemmenden Eigenschaften bekannt.
Ferner sind aus dem J. Chem. Soc. 1958,1855—1861, 65 Retinoide bekannt, welche als cyclische Endgruppe eine Tri-methylphenylgruppe aufweisen und welchen eine wachstumsfördernde, jedoch keine tumorhemmende Wirkung zugeschrieben wird.

Die Aufgabe der Erfindung besteht darin, neue tumorhemmende Wirkstoffe bereitzustellen, welche bezüglich der obengenannten Anhydroretinonsäurederivate fortschrittliche Eigenschaften, insbesondere ein besseres therapeutisches Vér in der Ri und R2 niederes Alkyl bedeuten, R3, R4 und R5 15 Wasserstoff, niederes Alkyl, niederes Alkoxy, niederes Al-kenoxy, Nitro, Amino, mono- oder di-nieder-Alkylamino, niederes Alkanoylamido oder einen N-heterocyclischen Rest darstellen, R3 und R5 ferner Halogen bedeuten, wobei mindestens einer der Reste R3 und Rs Halogen bedeutet und R4 20 von Alkoxy verschieden ist; und R6 Formyl, Alkoxymethyl, Alkanoyloxymethyl, Carboxyl, Alkoxycarbonyl, di-nieder Alkylcarbamoyl oder N-Heterocyclylcarbonyl bezeichnet.
Die vorstehend genannten niederen Alkylgruppen enthalten vornehmlich bis zu 6 Kohlenstoffatome, wie die Methyl-, 25 Äthyl-, Propyl-, Isopropyl- oder 2-Methylpropylgruppe. Die niederen Alkoxy- und niederen Alkenoxygruppen enthalten ebenfalls vornehmlich bis zu 6 Kohlenstoffatome wie die Methoxy-, Äthoxy- oder Isopropoxygruppe und die Vinyloxy-oder Allyloxygruppe. 30
Von den Halogenatomen sind Fluor und Chlor bevorzugt.
Die Aminogruppe kann durch verzweigtes oder unverzweigtes niederes Alkyl, z. B. durch Methyl, Äthyl, Isopropyl, mono- oder di-substituiert sein.
Die niederen Alkanoylamidogruppen enthalten Reste, die 35 sich von niederen Alkancarbonsäuren mit bis zu 6 Kohlenstoffatomen, z. B. von der Essig-, Propion- oder Pivalinsäure, ableiten.
Die N-Heterocyclylreste sind vornehmlich 5- oder 6glied-rige Reste, die gegebenenfalls neben dem Stickstoffatom als 40 weiteres Heteroatom Sauerstoff, Stickstoff oder Schwefel enthalten. Beispiele hierfür sind die Pyrrolidino-, Piperidino-, Morpholino- oder Thiomorpholinorest.
Die weiterhin genannten Alkoxymethyl- und Alkoxycar-bonylgruppen enthalten vornehmlich Alkoxyrest mit bis zu 45 6 Kohlenstoffatomen. Diese können verzweigt oder unverzweigt sein, wie beispielsweise der Methoxy-, Äthoxy- oder Isoprop-
hältnis (die Definition des Begriffes «therapeutisches Verhältnis» ist in J. Cancer 10, 731-733 [1974] gegeben), zeigen.
Die Erfindung betrifft daher ein Verfahren zur Herstellung von Polyenverbindungen der Formel

oxyrest. Darüber hinaus kommen aber auch höhere Alkoxy-reste mit 7 bis 20 Kohlenstoffatomen, von diesen insbesondere der Cetyloxyrest, in Frage.
Die Alkanoyloxymethylgruppen leiten sich vornehmlich von niederen Alkancarbonsäuren mit 1 bis 6 Kohlenstoffatomen, z. B. von der Essig-, Propion- oder Pivalinsäure, gegebenenfalls aber auch von höheren Alkancarbonsäuren mit 7 bis 20 Kohlenstoffatomen, z. B. von der Palmitin- oder Stearinsäure, ab.
Beispiele für mono- oder di-nieder-Alkylcarbamoyl sind Methylcarbamoyl, Dimethylcarbamoyl und Diäthylcarbamoyl.
Die N-Heterocyclylreste der N-Heterocyclylcarbonylgrup-pen sind vornehmlich 5- oder 6gliedrig heterocyclische Reste, die gegebenenfalls neben dem Stickstoffatom, Sauerstoff, Stickstoff oder Schwefel als weiteres Heteroatom enthalten. Beispiele hierfür sind der Piperidino-, Morpholino-, Thiomor-pholino- oder Pyrrolidinorest.
Vorzugsweise werden all-trans-Verbindungen hergestellt.
Wenn im folgenden spezifisch genannte Verbindungen der Formel I, II oder III angegeben sind, so sind darunter stets all-trans-Verbindungen zu verstehen, sofern nicht ein besonderer Hinweis auf eine cis-Verbindung oder ein cis/trans-Gemisch existiert.
Als repräsentative Vertreter der erfindungsgemäss herstellbaren Verbindungsklasse können genannt werden: 9-(3-Chlor-2,4,6-trimethyl-phenyl)-3,7-dimethyl-nona-
2,4,6,8-tetraen-l-säureäthylester, 9-(3-Chlor-2,4,6-trimethyl-phenyl)-3,7-dimethyl-nona-
2,4,6,8-tetraen-l-säure, 9-(3,5-Dichlor-2,4,6-trimethyl-phenyl)-3,7-dimethyl-nona-
2,4,6,8-tetraen-l-säure.

in denen m = 0 und n = 1 oder m = 1 und n = 0 sind, eines der beiden Symbole A und B die Formylgruppe und das andere Symbol eine Triarylphosphoniummethylgruppe der Formel -CH2-P[X]3® Y©, worin X einen Arylrest und Y das Anion einer organischen oder anorganischen Säure darstellt, die Reste Ri, R2, R3, R4 und Rs die oben gegebene Bedeutung haben und R7 Carboxyl, Alkoxycarbonyl, di-nieder-Alkylcarb-amoyl oder N-Heterocyclylcarbonyl bedeutet und R7, falls B die Formylgruppe darstellt, zusätzlich Alkoxymethyl und Al-kanoyloxymethyl und R7, falls B eine Triarylphosphoniummethylgruppe darstellt, zusätzlich Formyl bedeutet,
umsetzt.
Die in der Triarylphosphoniummethylgruppe der Formel -CH2-P[X]3® Y9 mit X bezeichneten Arylgruppen umfassen gemeinhin alle bekannten Arylreste, insbesondere aber einkernige Reste wie Phenyl oder nieder-Alkyl- bzw. nieder-Alkoxy-substituiertes Phenyl, wie Tolyl, Xylyl, Mesityl und p-Methoxy-phenyl. Von den anorganischen Säureanionen Y ist das Chlor-, Brom- und Jodion oder das Hydrosulfation, von den organischen Säureanionen ist das Tosyloxyion bevorzugt.
Die Ausgangssubstanzen der Formeln II und III sind zum Teil neue Verbindungen. Sie sind z. B. auf folgendem Wege erhältlich:
Verbindungen der allgemeinen Formel II, in der m = 0 ist und A eine Triarylphosphoniummethylgruppe [IIa] bedeutet, können z. B. dadurch erhalten werden, dass man ein entsprechendes durch die Reste Ri—Rs substituiertes Benzol in Gegenwart einer Halogenwasserstoffsäure, z. B. in Gegenwart von konz. Salzsäure, gegebenenfalls in einem Lösungsmittel, insbesondere in Eisessig mit Formaldehyd, behandelt und das entstehende, durch die Reste Ri-Rs substituierte Benzylhalo-genid [ein Halogenid der Formel Iii, in der m = 0 ist] in an sich bekannter Weise mit einem Triphenylphosphin in Toluol oder Benzol umsetzt.
Eine, in dem vorstehend genannten, durch die Reste Ri—R5 substituiertes Benzol vorhandene Alkoxygruppe kann z. B. durch Alkylieren einer vorhandenen Hydroxygruppe eingeführt werden. Man setzt beispielsweise das entsprechende Phenol, vorzugsweise in einem Lösungsmittel, z. B. in einem Alkanol und in Gegenwart einer Base wie Kaliumcarbonat, mit einem Alkylhalogenid, z. B. mit Methyljodid oder Dimethyl-sulfat, um.
Verbindungen der Formel II, in der m = 1 ist und A eine Triarylphosphoniummethylgruppe [IIb] bedeutet, sind z. B. auf folgendem Wege erhältlich: Man unterwirft das entsprechende, durch die Reste Ri-Rs substituierte Benzol zunächst einer Formylierungsreaktion, indem man beispielsweise auf die Ausgangsverbindung ein Formylierungsmittel einwirken lässt. Dies kann z. B. in der Weise geschehen, dass man die Ausgangsverbindung in Gegenwart einer Lewis-Säure formyliert. Als Formylierungsreagenzien kommen insbesondere folgende Substanzen in Frage: Orthoameisensäureester, Formylchlorid und Dimethylformamid. Von den Lewis-Säuren sind insbesondere geeignet die Halogenide von Zink, Aluminium, Titan, Zinn und Eisen, wie Zinkchlorid, Aluminiumtrichlorid, Titantetrachlorid, Zinntetrachlorid und Eisentrichlorid, sowie ferner auch die Halogenide von anorganischen und organischen Säuren, wie beispielsweise Phosphoroxychlorid und Methan-sulfochlorid.
Die Formylierung kann, wenn das Formylierungsmittel im Überschuss anwesend ist, gegebenenfalls ohne Zusatz eines weiteren Lösungsmittels durchgeführt werden. Im allgemeinen empfiehlt es sich jedoch, die Reaktion in einem inerten Lösungsmittel, z. B. in Nitrobenzol, oder in einem chlorierten Kohlenwasserstoff, wie Methylenchlorid, durchzuführen. Die Reaktionstemperatur kann zwischen 0 und dem Siedepunkt des Reaktionsgemisches liegen.
Der erhaltene durch die Reste R1-R5 substituierte Benzaldehyd kann anschliessend in an sich bekannter Weise durch Kondensation mit Aceton in der Kälte, d. h. in einem Temperaturbereich von etwa 0-30° C, in Gegenwart von Alkali z. B. in Gegenwart von verdünnter wässriger Natronlauge zu dem durch die Reste Ri—Rs substituierten Phenyl-but-3-en-2-on verlängert werden, das in an sich bekannter Weise mit Hilfe einer metallorganischen Reaktion z. B. mit einer Grignardreak-tion durch Addition von Acetylen in das entsprechende durch den Rest Rx-R5 substituierte Phenyl-3-methyl-3-hydroxy-penta-4-en-l-in übergeführt werden kann. Das erhaltene tertiäre Acetylencarbinol wird anschliessend in an sich bekannter Weise mit Hilfe eines teilweise vergifteten Ed.elmetallkataly-sators (Lindlar-Katalysator) partiell hydriert. Das entstehende tertiäre Äthylencarbinol kann anschliessend unter Allylumla-gerung durch Behandeln mit einem Triarylphosphin insbesondere mit Triphenylphosphin in Gegenwart einer Mineralsäure z. B. in Gegenwart eines Halogenwasserstoffs wie Chlor- oder Bromwasserstoff oder in Gegenwart von Schwefelsäure in einem Lösungsmittel z. B. in Benzol in das gewünschte Phos-phoniumsalz der Formel IIb, in der m = 1 ist, übergeführt werden.
Verbindungen der Formel Hg, in der m = 0 ist und A die Formylgruppe bedeutet, können z. B. dadurch hergestellt werden, dass man ein durch die Reste R1-R5 substituiertes Benzol, wie vorangehend beschrieben, formyliert. Man erhält auf diese Weise ausgehend von dem durch die Reste Ri—Rs substituierten Benzol unmittelbar den durch die Reste Rt—Rs substituierten Benzaldehyd.
Verbindungen der Formel Ilh, in der m = 1 ist und A die Formylgruppe bedeutet, lassen sich z. B. in der Weise herstellen, dass man das vorstehend bei der Herstellung von Verbindungen der Formel IIb näher beschrieben, durch die Reste R1-R5 substituierte, Phenyl-but-3-en-2-on nach Wittig mit Äthoxycarbonyl-methyl-en-triphenylphosphoran oder mit Diäthyl-phosphonoessigsäureäthylester umsetzt. Der erhaltene durch die Reste Ri—R5 substituierte Phenyl-3-methyl-penta-2,4-dien-l-säureäthylester wird anschliessend in der Kälte mit Hilfe eines gemischten Metallhydrids, insbesondere mit Lithiumaluminiumhydrid, in einem organischen Lösungsmittel z. B. in Äther oder in Tetrahydrofuran zu dem durch die Reste R1-R5 substituierten Phenyl-3-methyl-penta-2,4-dien-l-ol reduziert. Der erhaltene Alkohol wird danach durch Behandeln mit einem Oxydationsmittel z. B. mit Mangandioxyd in einem organischen Lösungsmittel, wie Aceton oder Methylenchlorid, in einem zwischen 0° und dem Siedepunkt des Reaktionsgemisches liegenden Temperaturbereich zu dem gewünschten durch Ri—R5 substituierten Phenyl-3-methyl-penta-2,4-dien-l-al der Formel Ilh oxydiert.
Auch die Verbindungen der Formel III sind zum Teil neu:
Verbindungen der Formel III, in der n = 0 ist und B eine Triarylphosphoniummethylgruppe [lila] bedeutet, lassen sich in einfacher Weise dadurch herstellen, dass man eine gegebenenfalls veresterte 3-Halogenmethylcrotonsäure oder einen verätherten 3-Halogenmethyl-crotyl-alkohol mit einem Triarylphosphin in einem Lösungsmittel, vornehmlich mit Triphenylphosphin in Toluol oder Benzol, umsetzt.
Verbindungen der Formel III, in der n = 1 ist und B eine Triarylphosphoniummethylgruppe [Illb] bedeutet, lassen sich z. B. in der Weise gewinnen, dass man die Formylgruppe eines Aldehyds der Formel Illh, in der n = 1 ist, mit Hilfe eines Metallhydrids z. B. mit Hilfe von Natriumborhydrid in einem Alkanol z. B. in Äthanol oder Isopropanol zur Hydroxy-methylgruppe reduziert. Der erhaltene Alkohol kann mit Hilfe eines der üblichen Halognierungsmittel, z. B. mit Phosphoroxychlorid, halogeniert und die erhaltene 8-Halogen-3,7-dimethylocta-2,4,6-trien-l-carbonsäure, ein Halogenid der Formel Ulk, in der n = 1 ist, oder ein Derivat dieser Säure mit einem Triarylphosphin in einem Lösungsmittel, vornehmlich

mit Triphenylphosphin in Toluol oder Benzol, zu dem gewünschten Phosphoniumsalz der Formel Illb umgesetzt werden.
Verbindungen der Formel Illg, in der n = 0 ist und B die Formylgruppe bedeutet, können beispielsweise dadurch erhalten werden, dass man eine gegebenenfalls veresterte Weinsäure oxydativ spaltet, z. B. durch Einwirkung von Bleitetra-acetat bei Raumtemperatur in einem organischen Lösungsmittel wie Benzol. Das erhaltene Glyoxalsäurederivat wird anschliessend in an sich bekannter Weise, tunlich in Gegenwart eines Amins, mit Propionaldehyd bei erhöhter Temperatur, z. B. in einem zwischen 60 und 110° C liegenden Temperaturbereich, unter Wasserabspaltung zu dem gewünschten 3-Formyl-croton-säurederivat kondensiert.
Verbindungen der Formel Illh, in der n = 1 ist und B die Formylgruppe bedeutet, lassen sich z. B. in der Weise herstellen, dass man auf 4,4-Dimethoxy-3-methyl-but-l-en-3-ol in der Kälte, vorzugsweise bei —10 bis -20° C, in Gegenwart eines tertiären Amins, wie Pyridin, Phosgen einwirken lässt und das erhaltene 2-Formyl-4-chlor-but-2-en mit Hilfe einer Wittig-Reaktion mit einer gegebenenfalls veresterten 3-Formyl-crotonsäure oder mit einem veresterten oder verätherten 3-Formyl-crotylalkohol zu dem gewünschten Aldehyd der Formel Illh verknüpft.
Gemäss der Erfindung werden:
- Phosphoniumsalze der Formel IIa oder IIb mit Aldehyden der Formel Illh oder Illg oder
- Phosphoniumsalze der Formel lila oder Illb mit Aldehyden der Formel Ilh oder Hg umgesetzt.
Nach der von Wittig angegebenen Arbeitsweise werden die Komponenten in Gegenwart eines säurebindenden Mittels, z. B. in Gegenwart eines Alkalimetallalkoholates, wie Natrium-methylat, oder in Gegenwart eines gegebenefalls alkylsubsti-tuierten Ätylenoxyds, insbesondere in Gegenwart von Äthylenoxyd oder 1,2-Butylenoxyd, gegebenenfalls in einem Lösungsmittel, z. B. in einem chlorierten Kohlenwasserstoff, wie Methylenchlorid, oder auch in Dimethylformamid, in einem zwischen der Raumtemperatur und dem Siedepunkt des Reaktionsgemisches liegenden Temperaturbereich miteinander umgesetzt.
Es hat sich in bestimmten Fällen als zweckmässig erwiesen, die vorstehend genannten Reaktionen in situ, d. h. die Kondensationskomponenten, ohne das betreffende Phosphoniumsalz zu isolieren, mit einander zu verknüpfen.
Eine Carbonsäure der Formel I kann in an sich bekannter Weise, z. B. durch Behandeln mit Thionylchlorid, vorzugsweise in Pyridin, in das Säurechlorid übergeführt werden, das durch Umsetzen mit einem Alkanol in einen Ester, mit Ammoniak in das Amid umgewandelt werden kann.
Ein Carbonsäureester der Formel I kann in an sich bekannter Weise, z. B. durch Behandeln mit Alkalien, insbesondere durch Behandeln mit wässriger alkoholischer Natron- oder Kalilauge, in einem zwischen der Raumtemperatur und dem Siedepunkt des Reaktionsgemisches liegenden Temperaturbereich hydrolysiert und entweder über ein Säurehalogenid oder, wie nachstehend beschrieben, unmittelbar amidiert werden.
Ein Carbonsäureester der Formel I kann z. B. durch Behandeln mit Lithiumamid direkt in das entsprechende Amid umgewandelt werden. Das Ltihiumamid wird vorteilhaft bei Raumtemperatur mit dem betreffenden Ester zur Reaktion gebracht.
Ein Amin der Formel I bildet mit anorganischen oder organischen Säuren Additionssalze. Als Beispiele können genannt werden: Salze mit Halogenwasserstoffsäuren, insbesondere mit der Chlor- oder Bromwasserstoffsäure, Salze mit Mineralsäuren, z. B. mit Schwefelsäure, oder auch Salze mit organischen Säuren, z. B. mit der Benzoesäure, Essigsäure, Zitronensäure oder Milchsäure.
Eine Carbonsäure der Formel I bildet mit Basen, insbesondere mit den Alkalimetallhydroxyden, vorzugsweise mit Natrium- oder Kaliumhydroxyd, Salze.
Die Verbindungen der Formel I können als cis/trans-Ge-mische anfallen, welche in an sich bekannter Weise erwünsch-tenfalls in die eis und trans-Komponenten aufgetrennt oder zu den all-trans-Verbindungen isomerisiert werden können.
Die Verfahrensprodukte der Formel I stellen pharmako-dynamisch wertvolle Verbindungen dar. Sie können zur topischen und systematischen Therapie von benignen und malignen Neoplasien, von prämalignen Läsionen sowie ferner auch zur systemischen und topischen Prophylaxe der genannten Affektion verwendet werden. Sie sind des weiteren für die topische und systemische Therapie von Akne, Psoriasis und anderen mit einer verstärkten oder pathologisch veränderten Verhornung einhergehenden Dermatosen, wie auch von entzündlichen und allergischen dermatologischen Affektionen geeignet. Die Verfahrensprodukte der Formel I können femer auch zur Bekämpfung von Schleimhauterkrankungen mit entzündlichen oder degenerativen bzw. metaplastischen Veränderungen eingesetzt werden.
Verbindungen der Formel I, in der einer oder beide Reste R3 und Rs Halogen und R4 Alkoxy darstellen, sind nur • schwach pharmakodynamisch wirksam. Die Herstellung dieser Verbindungen wird deshalb nicht beansprucht.
Die Toxizität der neuen Verbindungsklasse ist gering. Die akute Toxizität [DL501 der 9-(3-Chlor-2,4,6-trimethyl-phenyl)-3,7-dimethyl-nona-2,4,6,8-tetraen-l-l-säure [A] liegt-wie aus der in der nachstehenden Tabelle verzeichneten Spättoxi-zität nach 20 Tagen ersichtlich — bei der Maus nach intraperitonealer Verabreichung in Rüböl bei 2800 mg/kg.

Die tumorhemmende Wirkung der Verfahrensprodukte ist signifikant. Im Papillomtest regressieren mit Dimethylbenz-anthracen und Krotonöl induzierte Tumoren. Die Durchmesser der Papillome nehmen innerhalb von 2 Wochen bei intraperitonealer Applikation von Substanz A: bei 400 mg/kg/Woche um 65 %
bei 200 mg/kg/Woche um 46% bei 50 mg/kg/Woche um 10% ab.
Die Verbindungen der Formel I können deshalb als Heilmittel, z. B. in Form pharmazeutischer Präparate, Anwendung finden.
Die zur systemischen Anwendung dienenden Präparate können z. B. dadurch hergestellt werden, dass man eine Verbindung der Formel I als wirksamen Bestandteil nichttoxischen, inerten an sich in solchen Präparaten üblichen festen oder flüssigen Trägern zufügt.
Die Mittel können enterai oder parenteral verabreicht werden. Für die enterale Applikation eignen sich z. B. Mittel in Form von Tabletten, Kapseln, Dragées, Sirupen, Suspensionen, Lösungen und Suppositorien. Für die parenterale Applikation sind Mittel in Form von Infusions- oder Injektionslösungen geeignet.

Die Dosierungen, in denen die Verfahrensprodukte verabreicht werden, können je nach Anwendungsart und Anwendungsweg sowie nach den Bedürfnissen der Patienten variieren.
Die Verfahrensprodukte könnnen in Mengen von 5 bis 200 mg täglich in einer oder mehreren Dosierungen verabreicht werden. Eine bevorzugte Darreichungsform sind Kapseln mit einem Gehalt von ca. 10 bis ca. 100 mg Wirkstoff.
Die Präparate können inerte oder auch pharmakoxynamisch aktive Zusätze enthalten. Tabletten oder Granulate z. B. können eine Reihe von Bindemitteln, Füllstoffen, Trägersubstanzen oder Verdünnungsmitteln enthalten. Flüssige Präparate können beispielsweise in Form einer sterilen, mit Wasser mischbaren Lösung vorliegen. Kapseln können neben dem Wirkstoff zusätzlich ein Füllmaterial oder Verdickungsmittel enthalten. Des weiteren können geschmacksverbessernde Zusätze sowie die üblicherweise als Konservierungs-, Stabilisie-rungs-, Feuchthalte- oder Emulgiermittel verwendeten Stoffe, ferner auch Salze zur Veränderung des osmotischen Druckes, Puffer und andere Zusätze vorhanden sein.
Die vorstehend erwähnten Trägersubstanzen und Verdünnungsmittel können aus organischen oder anorganischen Stoffen, z. B. aus Wasser, Gelatine, Milchzucker, Stärke, Magne-siumstearat, Talkum, Gummi arabicum, Polyalkylenglykolen u. dgl. bestehen. Voraussetzung ist, dass alle bei der Herstellung der Präparate verwendeten Hilfsstoffe untoxisch sind.
Zur topischen Anwendung werden die Verfahrensprodukte zweckmässig in Form von Salben, Tinkturen, Crèmen, Lösungen, Lotionen, Sprays, Suspensionen u. dgl. verwendet. Bevorzugt sind Salben und Crèmen sowie Lösungen. Diese zur topischen Anwendung bestimmten Präparate können dadurch hergestellt werden, dass man die Verfahrensprodukte als wirksamen Bestandteil nichttoxischen, inerten, für topische Behandlung geeigneten, an sich in solchen Präparaten üblichen festen oder flüssigen Trägern zumischt.
Für die topische Anwendung sind zweckmässig ca. 0,01 bis ca. 0,3prozentige, vorzugsweise 0,02 bis 0,lprozentige, Lösungen sowie ca. 0,05 bis ca. 5prozentige, vorzugsweise ca. 0,1 bis ca. 2,0prozentige, Salben oder Crèmen geeignet.
Den Präparaten kann gegebenenfalls ein Antioxydationsmittel, z. B. Tocopherol, N-Methyl-y-tocopheramin sowie butyliertes Hydroxyanisol oder butyliertes Hydroxytoluol beigemischt sein.
Beispiel 1
16 g 3-Chlor-2,4,6-trimethyl-benzyl-triphenyl-phospho-niumchlorid und 10 g 7-Formyl-3-methyl-octa-2,4,6-trien-l-säure-butylester werden nach Zugabe von 40 g 1,2-Butylen-oxyd unter Rühren zum Sieden erhitzt. Das 1,2-Butylenoxyd wird langsam abdestilliert. Das Reaktionsgemisch wird 30 Minuten bei 80—82° C gerührt, danach gekühlt und erschöpfend mit Hexan extrahiert. Der Hexanextrakt wird 5mal mit je 50 ml Methanol/Wasser 70:30 ausgeschüttelt, danach über Natriumsulfat getrocknet und unter vermindertem Druck eingedampft. Der zurückbleibende 9-(3-Chlor-2,4,6-trimethyl-phenyl)-3,7-dimethyl-nona-2,4,6,8-tetraen-l-säure-butylester kann wie folgt in die freie Säure übergeführt werden:
5 g 9-(3-Chlor-2,4,6-trimethyl-phenyl)-3,7-dimethyl-nona-2,4,6,8-tetraen-l-säurebutylester werden in 50 ml einer 5pro-zentigen äthanolischen Kaliumhydroxydlösung unter Stickstoffbegasung zum Sieden erhitzt. Die in der Siedehitze klar werdende Lösung wird nach 30 Minuten abgekühlt, in Wasser eingetragen und durch Zugabe von Essigsäure sauer gestellt. Die ausfallende 9-(3-Chlor-2,4,6-trimethyl-phenyl)-3,7-dimethyl-nona-2,4,6,8-tetraen-l-säure schmilzt nach dem Umkristallisieren aus Benzol bei 208-209° C.
Das als Ausgangsverbindung eingesetzte 3-Chlor-2,4,6-tri-methyl-benzyl-triphenylphosphoniumchlorid kann z. B. wie folgt hergestellt werden:
119 g Chlormesitylen, 11,9 g Paraformaldehyd und 5,95 g Zinkchlorid (wasserfrei) werden auf 60° C erhitzt und unter Rühren zunächst 8 Stunden und nach Zugabe von weiteren 11,9 g Paraformaldehyd weitere 8 Stunden mit Chlorwasserstoff begast. Das Reaktionsgemisch wird danach auf Eis gegossen und erschöpfend mit Äther extrahiert. Der Ätherextrakt wird nacheinander mit Wasser, mit einer gesättigten wässrigen Natriumhydrogencarbonatlösung und mit Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft. Das zurückbleibende 3-Chlor-2,4,6-trimethyl-benzyIchlorid siedet nach der Rektifikation bei 138° C/17 Torr.
71,25 g 3-Chlor-2,4,6-trimethyl-benzylchlorid, 92 g Triphenylphosphin und 375 ml abs. Toluol werden 12 Stunden auf 100° C erhitzt. Das beim Abkühlen ausfallende 3-Chlor-2,4,6-trimethyl-benzyl-triphenylphosphoniumchlorid schmilzt bei 233-235° C.
Beispiel 2
In Analogie zu der in Beispiel 1 beschriebenen Arbeitsweise können hergestellt werden:
— aus 3,5-Dichlor-2,4,6-trimethyl-benzyl-triphenyl-
phosphoniumchlorid,
durch Umsetzen mit 7-Formyl-3-methyl-octa-2,4,6-trien-1 -säureäthylester,
— der 9-(3,5-Dichlor-2,4,6-trimethyl-phenyl)-3,7-dimethyl-
nona-2,4,6,8-tetraen-l-säureäthylester,
und durch Verseifen des erhaltenen Esters,
— die 9-(3,5-Dichlor-2,4,6-trimethyl-phenyl)-3,7-dimethyl-
nona-2,4,6,8-tetraen-l-säure; Fp.: 220-222°C.
— aus 5-Fluor-2,4,6-trimethyl-benzyl-triphenyl-
phosphoniumchlorid,
durch Umsetzen mit 7-Formyl-3-methyl-octa-2,4,6-trien-1-säure,
— die 9-(5-Fluor-2,4,6-trimethyl-phenyl)-3,7-dimethyl-
nona-2,4,6,8-tetraen-l-säure;
Fp.: 206—209°C.
— aus 5-Brom-2,4,6-trimethyl-benzyl-triphenyl-
phosphoniumchlorid,
durch Umsetzen mit 7-Formyl-3-methyl-octa-2,4,6-trien-1-säure,
— die 9-(5-Brom-2,4,6-trimethyl-phenyl)-3,7-dimethyl-
nona-2,4,6,8-tetraen-l-säure; Fp. 209-214°C.
Beispiel 3
In Analogie zu der in Beispiel 4 beschriebenen Arbeitsweise kann hergestellt werden:
— aus 3-ChIor-2,4,6-trimethyl-benzyl-triphenyl-
phosphoniumchlorid,
durch Umsetzen mit 7-Formyl-3-methyl-octa-2,4,6-trien-1-säureäthylester,
— der 9-(3-Chlor-2,4,6-trimethyl-phenyl)-3,7-dimethyl-
nona-2,4,6,8-tetraen-l-säureäthylester; Fp.: 84 bis 85°C.
Das als Ausgangsverbindung eingesetzte 3-Chlor-2,4,6-tri-methyl-benzyl-triphenylphosphoniumchlorid ist in analoger Weise, wie in den vorangehenden Beispielen beschrieben, z. B. ausgehend von 2,4,6-Trimethylanilin,
über 2,4,6-Trimethyl-chlorbenzol
— 3-Chlor-2,4,6-trimethyl-benzylchlorid zugänglich.

2. Verfahren nach Patentanspruch 1, dadurch gekennzeichnet, dass man all-trans-Verbindungen herstellt.
3. Verfahren nach Patentanspruch 1 oder 2, dadurch gekennzeichnet, dass man ein Phosphoniumsalz der Formel II oder III mit einem Aldehyd der Formel III oder II in Gegenso wart eines Epoxyds, insbesondere in Gegenwart eines gegebenenfalls durch niederes Alkyl substituierten Äthylenoxyds, wie 1,2-Butylenoxyd, gegebenenfalls unter Zusatz eines Lösungsmittels, umsetzt.
4. Verfahren nach Patentanspruch 1 oder 2, dadurch gekennzeichnet, dass man einen erhaltenen Carbonsäureester zur entsprechenden Carbonsäure verseift.
5. Verfahren nach Patentanspruch 1 oder 2, dadurch gekennzeichnet, dass man eine erhaltene Carbonsäure oder ein erhaltenes Amin in ein Salz überführt.
6. Verfahren nach Patentanspruch 1 oder 2, dadurch gekennzeichnet, dass man eine erhaltene Carbonsäure in einen Ester überführt.
7. Verwendung einer nach Patentanspruch 1 oder 2 erhaltenen Carbonsäure zur Herstellung eines Amides, eines mono-oder di-nieder- Alkyl-amides.
