EMI2.1
mit einer Verbindung der Formel
EMI2.2
in denen m = 0 und n = 1, oder m = I und n = 0 sind, eines der beiden Symbole A und B die Formylgruppe, und das andere Symbol eine Triarylphosphoniummethylgruppe der Formel -CH2-P[X]3#Y#, worin X einen Arylrest und Y das Anion einer organischen oder anorganischen Säure darstellt, die Reste R1, R2, R3, R4 und R die oben gegebenen Bedeutung haben, und R7 Carboxy, Alkoxycarbonyl, Di-nieder-Alkylcarbamoyl oder N-Heterocyclylcarbonyl bedeutet und R7, falls B die Formylgruppe darstellt, zusätzlich Alkoxymethyl und Alkanoyloxymethyl;
Die in der Triarylphosphoniummethylgruppe der Formel -CH2-P[X]3#Y# mit X bezeichneten Arylgruppen umfassen gemeinhin alle bekannten Arylreste, insbesondere aber einkernige Reste wie Phenyl oder nieder-Alkyl- bzw. nieder Alkoxy-substituiertes Phenyl, wie Tolyl, Xylyl, Mesityl und p-Methoxyphenyl. Von den anorganischen Säureanionen Y ist das Chlor-, Brom- und Jodion oder das Hydrosulfat-ion, von den organischen Säureanionen ist das Tosyloxy-ion bevorzugt;

EMI2.1
with a compound of the formula
EMI2.2
in which m = 0 and n = 1, or m = I and n = 0, one of the two symbols A and B is the formyl group, and the other symbol is a triarylphosphonium methyl group of the formula -CH2-P [X] 3 # Y #, wherein X is an aryl radical and Y is the anion of an organic or inorganic acid, the radicals R1, R2, R3, R4 and R have the meanings given above, and R7 is carboxy, alkoxycarbonyl, di-lower-alkylcarbamoyl or N-heterocyclylcarbonyl and R7 if B represents the formyl group, additionally alkoxymethyl and alkanoyloxymethyl;
and R7, if B is a triarylphosphonium methyl group, also denotes formyl.
The aryl groups denoted by X in the triarylphosphonium methyl group of the formula -CH2-P [X] 3 # Y # generally include all known aryl radicals, but in particular mononuclear radicals such as phenyl or lower-alkyl- or lower-alkoxy-substituted phenyl, such as tolyl, xylyl , Mesityl and p-methoxyphenyl. Of the inorganic acid anions Y, the chlorine, bromine and iodine ion or the hydrosulfate ion is preferred; of the organic acid anions, the tosyloxy ion is preferred;
The starting substances of formulas II and III are partly new compounds.
You are e.g. available in the following ways:
Compounds of formula 11 in which m = 0 and A is a triarylphosphonium methyl group. [IIa] can e.g. be obtained by adding a corresponding benzene substituted by the radicals R1-R ,, in the presence of a hydrohalic acid, e.g. in the presence of conc.
Hydrochloric acid optionally treated with formaldehyde in a solvent, especially in glacial acetic acid, and the resulting benzyl halide, substituted by the radicals R, -R, is a halide of the formula Iii, in which m = 0], in a manner known per se with a triarylphosphine in one Solvent, primarily with triphenylphosphine in toluene or benzene, is reacted.
An alkoxy group present in the above-mentioned benzene substituted by the radicals Rl-R, can e.g. are introduced by alkylating an existing hydroxy group. For example, the corresponding phenol is used, preferably in a solvent, e.g. in an alkanol and in the presence of a base such as potassium carbonate with an alkyl halide, e.g. with methyl iodide or dimethyl sulfate.
Compounds of formula II in which m = 1 and A is a triarylphosphonium methyl group [1 Ib] are e.g. obtainable in the following way: The corresponding benzene substituted by the radicals R, -R, is first subjected to a formylation reaction by, for example, letting a formylating agent act on the starting compound. This can e.g. done in such a way that the starting compound is formylated in the presence of a Lewis acid. The following substances are particularly suitable as formylating reagents: orthoformic acid ester, formyl chloride and dimethylformamide.
Of the Lewis acids, the halides of zinc, aluminum, titanium, tin and iron, such as zinc chloride, aluminum trichloride, titanium tetrachloride, tin tetrachloride and iron trichloride, and also the halides of inorganic and organic acids such as phosphorus oxychloride and methanesulfonyl chloride are particularly suitable.
If the formylating agent is present in excess, the formylation can optionally be carried out without the addition of a further solvent. In general, however, it is advisable to carry out the reaction in an inert solvent, e.g. in nitrobenzene, or in a chlorinated hydrocarbon such as methylene chloride, by zuffibren. The reaction temperature can be between 0 and the boiling point of the reaction mixture.
The benzaldehyde obtained, substituted by the radicals R1-R-, can then in a manner known per se by condensation with acetone in the cold, i.e. in a temperature range of about 0-300C in the presence of alkali e.g. in the presence of dilute aqueous sodium hydroxide solution to the phenyl-but-3-en-2-one substituted by the radicals R1-R-, which is produced in a manner known per se with the aid of an organometallic reaction e.g. can be converted with a Grignard reaction by addition of acetylene into the corresponding phenyl-3-methyl -3-hydroxy-penta-4-en-1-yne substituted by the radical R, -R,.
The tertiary acetylene carbinol obtained is then partially hydrogenated in a manner known per se with the aid of a partially poisoned noble metal catalyst (Lindlar catalyst).
The resulting tertiary ethylene carbinol can then be treated with a triarylphosphine, in particular with triphenylphosphine, in the presence of a mineral acid, e.g. in the presence of a hydrogen halide such as hydrogen chloride or hydrogen bromide or in the presence of sulfuric acid in a solvent e.g. in benzene into the desired phosphonium salt of the formula IIb, in which m = list, are converted.
Compounds of the formula IIg in which m = 0 and A is the formyl group can e.g. can be prepared by formylating a benzene substituted by the radicals R, -R3, as described above. In this way, starting from the benzene substituted by the radicals R1-R5, the benzaldehyde substituted by the radicals R1-R is obtained directly.
Compounds of the formula IIh in which m = list and A denotes the formyl group can be found e.g. produce in such a way that the phenyl-hut-3-en-2-one described above in the preparation of compounds of the formula IIb, substituted by the radicals R, -R, according to Wittig with ethoxycarbonyl-methylene-triphenylphosphorane or with diethyl phosphonoacetic acid ethyl ester.
The phenyl-3-methyl-penta -2,4-diene-1-silurellethyl ester obtained, which is substituted by the radicals R1-R5, is then in the kiltie with the help of a mixed metal hydride, in particular with lithium aluminum hydride, in an organic solvent, e.g. reduced in anther or in tetrahydrofuran to the phenyl-3-methyl-penta-2,4-dien-1 -ol substituted by the radicals R1-R5.
The alcohol obtained is then treated with an oxidizing agent, e.g. with manganese dioxide in an organic solvent, such as acetone or methylene chloride, in a temperature range between 0 and the boiling point of the reaction mixture to the desired phenyl-3-methyl-penta-2,4-diene- 1- al of the formula IIh oxidized.
Some of the compounds of the formula III are also new:
Compounds of the formula III in which n = 0 and B is a triarylphosphonium methyl group [IIla] can be prepared in a simple manner by reacting an optionally esterified 3-halomethylcrotonic acid or an etherified 3-halomethylcrotyl alcohol with a Triarylphosphine in a solvent, primarily with triphenyiphosphine in toluene or benzene, is reacted.
Compounds of formula III in which n = 1 and B denotes a triarylphosphonium methyl group [IIIb] can be e.g. win in such a way that the formyl group of an aldehyde of formula IIIh, in which n = 1, with the aid of a metal hydride e.g. with the aid of sodium borohydride in an alkanol e.g. reduced to the hydroxymethyl group in ethanol or isopropanol. The alcohol obtained can be purified by means of one of the usual halogenating agents, e.g.
with phosphorus oxychloride, halogenated and the 8-halo-3,7-dimethyl-octa-2,4,6-triene-1-carboxylic acid obtained, a halide of the formula IIIk, in which n = 1, or a derivative of this azure with a triarylphosphine in a solvent, primarily with triphenylphosphine in toluene or benzene, to form the desired phosphonium salt of the formula IIIb.
The compounds of the formula IIIg, in which n = 0 and B denotes the formyl group, can be obtained, for example, by oxidative cleavage of an optionally esterified tartaric acid, e.g. by exposure to lead tetraacetate at room temperature in an organic solvent such as benzene. The glyoxalic acid derivative obtained is then treated in a manner known per se, possibly in the presence of an amine, with propionaldehyde at elevated temperature, e.g. condensed in a temperature range between 60 and 110 ° C., with elimination of water, to give the desired 3-formylcrotonic acid derivative.
Compounds of the formula IIIh, in which n = list and B denotes the formyl group, can be prepared, for example, by adding 4,4-dimethoxy-3-methyl-but-1-en-3-ol in the kite , preferably at -10 to -20 C, in the presence of a tertiary amine, such as pyridine, allows phosgene to act and the 2-formyl-4-chloro-but-2-ene obtained with the aid of a Wittig reaction with an optionally esterified 3- Formylcrotonic acid or linked to an esterified or etherified 3-formylcrotyl alcohol to form the desired aldehyde of the formula Ilib.
According to the invention: - Phosphonium salts of the formula IIa or IIb are reacted with aloes of the formula IIIh or IIIg or - Phosphonium salts of the formula IIIa or IIIb with aldehydes of the formula IIh or Hg.
According to the procedure given by Wittig, the components are in the presence of an acid-binding agent, e.g. in the presence of an alkali metal alcoholate, such as sodium methylate, or in the presence of an optionally alkyl-substituted ethylene oxide, in particular in the presence of ethylene oxide or 1,2-butylene oxide, optionally in a solvent, e.g. reacted with one another in a chlorinated hydrocarbon, such as methylene chloride, or else in dimethylformamide, in a temperature range between room temperature and the boiling point of the reaction mixture.
In certain cases it has proven to be unsuitable to carry out the aforementioned reactions in situ, i.e. to combine the condensation components without isolating the phosphonium salt concerned.
A carboxylic acid of the formula I can be used in a manner known per se, e.g. by treatment with thionyl chloride, preferably in pyridine, can be converted into the acid chloride, which can be converted into an ester by reaction with an alkanol, and into the amide with ammonia.
A carboxylic acid ester of formula I can be used in a manner known per se, e.g. hydrolyzed by treatment with alkalis, in particular by treatment with aqueous alcoholic sodium or potassium hydroxide solution in a temperature range between room temperature and the boiling point of the reaction mixture, and either via an acid halide or, as described below, directly amidated.
A carboxylic acid ester of formula I can e.g. being converted directly into the corresponding amide by treatment with lithium amide. The lithium amide is advantageously reacted with the ester in question at room temperature.
A carboxylic acid or a carboxylic acid ester of the formula I can be reduced to the corresponding alcohol of the formula I in a manner known per se. The reduction is advantageously carried out with the aid of a metal hydride or alkyl metal hydride in an inert solvent.
Mixed metal hydrides, such as lithium aluminum hydride or bis [methoxy-ethyleneoxy] - -sodium aluminum hydride, have proven particularly suitable as hydrides. The solvents that can be used include Ether, tetrahydrofuran or dioxane if lithium aluminum hydride is used, and ether, hexane, benzene or toluene if diisobutylaluminum hydride or bis [methoxy-ethyleneoxyl-sodium aluminum hydride are used.
An alcohol of formula I can e.g. in the presence of a base, preferably in the presence of sodium hydride, in an organic solvent such as dioxane, tetrahydrofuran, 1,2-dimethoxyethane, dimethylformamide, or in the presence of an alkali metal alcoholate in an alkanol, in a temperature range between 0 and room temperature with an alkyl halide , e.g. with ethyl iodide, are etherified.
An alcohol of the formula I can also be esterified by treatment with an alkanoyl halide or anhydride, conveniently in the presence of a base, for example in the presence of pyridine or triethylamine in a temperature range between room temperature and the boiling point of the reaction mixture.
An alcohol ester can be used in a manner known per se, e.g.
as described above for the saponification of the carboxylic acid ester be saponified.
An alcohol of the formula I or an ester of this alcohol can be oxidized to the corresponding satire of the formula I in a manner known per se. The oxidation is advantageously carried out with silver (I) oxide and alkali in water or in a water-miscible organic solvent in a temperature range between room temperature and the boiling point of the reaction mixture.
An amine of the formula I forms addition salts with inorganic or organic acids. Examples include: salts with hydrohalic acids, in particular with hydrochloric or hydrobromic acid, salts with mineral acids, e.g. with sulfuric acid, or also salts with organic acids? e.g. with benzoic acid, acetic acid, citric acid or lactic acid,
A carboxylic acid of the formula I forms with bases, in particular special with the alkali metal hydroxides, preferably with
Sodium or potassium hydroxide salts.
The compounds of the formula I can be used as cis / trans
Mixtures are obtained which, if necessary, can be separated into the cis and trans components or isomerized to the all-trans compounds in a manner known per se.
