Abstract
Description
すなわち、本発明は、以下の通りのものである。
(a)配列番号2に示すアミノ酸配列からなる蛋白質;
(b)配列番号2に示すアミノ酸配列と少なくとも90%の配列同一性を有するアミノ酸配列からなり、かつ、4-アミノ桂皮酸合成活性を有する蛋白質;
(c)配列番号2に示すアミノ酸配列において、1又は数個のアミノ酸が欠失、置換、挿入又は付加されたアミノ酸配列からなり、かつ、4-アミノ桂皮酸合成活性を有する蛋白質;及び
(d)配列番号2に示すアミノ酸配列をコードする塩基配列と相補的な塩基配列からなる核酸と高ストリンジェント条件下でハイブリダイズし、かつ、4-アミノ桂皮酸合成活性を有する蛋白質をコードするヌクレオチド配列からなる核酸によりコードされる蛋白質;
からなる群から選ばれる酵素を用いて、4-アミノフェニルアラニンを4-アミノ桂皮酸に変換することを特徴とする、4-アミノ桂皮酸の製造方法。

本発明において、注目した酵素はPhenyl alanine Ammonia Lyase(PAL)である。PALはフェニルアラニンを基質とし、桂皮酸を生産する酵素である。この反応と、本発明で目的としている脱アンモニア反応との違いはベンゼン環4位のアミノ基の有無である。前記したように、非特許文献3には、4-アミノ桂皮酸では、ベンゼン環上に電子吸引基を有すること、及び正のメソメリー効果の存在により変換が生じなかったことが記載されている。他方、非特許文献2には、酵素活性は極めて低いものの、シロイユナズナ由来のPALが4PAL活性を示したことが記載されている。かかる状況下、本発明者らは、PALの基質認識の非特異性を期待して酵素探索を実施した。これまでに知られているPALは植物と担子菌酵母に分布しているものが多いため、植物と担子菌酵母由来のPALを探索し、4APAL活性が高い酵素を選抜することとした。さらに、これらの酵素学的解析と大腸菌を用いた4-アミノ桂皮酸の休止菌体反応による生産を行った。
例えば、培養工程で得られる培養液を遠心分離によって培養上澄液と菌体とに分け、該菌体を生理食塩水で洗浄し、菌体濁度A600nm=40となるように滅菌精製水に懸濁し、これを休止菌体懸濁液として反応に用いることができる。
前記したように、RgPAL遺伝子の全配列は、特許文献2により公知であり、本明細書における配列番号2はかかるRgPAL遺伝子によりコードされるアミノ酸配列と同一である。当業者は、かかる配列情報に基づき、化学合成などの当業者に既知の任意の方法により配列番号2に示すアミノ酸配列からなる蛋白質を調製することができるため、当業者は該蛋白質を入手可能であり、入手方法を問わずに本発明を実施することができる。
ここで、「配列同一性」とは、ポリペプチド配列(若しくはアミノ酸配列)又はポリヌクレオチド配列(若しくは塩基配列)における2本の鎖の間で該鎖を構成している各アミノ酸残基同士又は各塩基同士の互いの適合関係において同一であると決定できるようなものの量(数)を意味し、二つのポリペプチド配列又は二つのポリヌクレオチド配列の間の配列相関性の程度を意味するものである。同一性は容易に算出できる。二つのポリヌクレオチド配列又はポリペプチド配列間の同一性を測定する方法は数多く知られており、「配列同一性」なる用語は、当業者には周知である。
変異DNAは、例えば、化学合成、遺伝子工学的手法、突然変異誘発などの当業者に既知の任意の方法により調製することができる。具体的には、配列番号2に示すアミノ酸配列をコードする配列番号1に示す塩基配列からなるDNAに対し、変異原となる薬剤と接触作用させる方法、紫外線を照射する方法、遺伝子工学的な手法等を用いて、これらDNAに変異を導入することにより、変異DNAを取得することができる。ここで、遺伝子工学的手法の一つである部位特異的変異誘発法は特定の位置に特定の変異を導入できる手法であることから有用であり、Sambrook,J. et al., Molecular Cloning: A Laboratory Manual, 2nd Ed., Cold Spring Harbor Laboratory Press, Cold Spring Harbor, N.Y., 1989等に記載の方法に準じて行うことができる。この変異DNAを適切な発現系を用いて発現させることにより、1又は数個のアミノ酸が欠失、置換、挿入又は付加されたアミノ酸配列からなる蛋白質を得ることができる。
本明細書において、「ストリンジェント(stringent)条件」とは、ポリヌクレオチドと、ゲノムDNAとの選択的かつ検出可能な特異的結合を可能とする条件である。ストリンジェント条件は、塩濃度、有機溶媒(例えば、ホルムアミド)、温度、その他公知の条件の適当な組み合わせによって定義される。すなわち、塩濃度を減じるか、有機溶媒濃度を増加させるか、又はハイブリダイゼーション温度を上昇させるかによってストリンジェンシーは増加する。更に、ハイブリダイゼーション後の洗浄の条件もストリンジェンシーに影響する。この洗浄条件もまた、塩濃度と温度によって定義され、塩濃度の減少と温度の上昇によって洗浄のストリンジェンシーは増加する。従って、「ストリンジェント条件」とは、各塩基配列間の同一性の程度が、例えば、全体の平均で約90%以上であるような、高い同一性を有する塩基配列間のみで、特異的にハイブリッドが形成されるような条件を意味する。具体的には、「ストリンジェント条件」とは、約45℃にて6.0×SSCでハイブリダイゼーションを行った後に、50℃で2.0×SSCで洗浄することを指す。ストリンジェンシーの選択のため、洗浄工程における塩濃度を、例えば、低ストリンジェントとしての約2.0×SSC、50℃から、高ストリンジェンシトとしての約0.1×SSC、50℃まで選択することができる。さらに、洗浄工程の温度を低ストリンジェント条件の室温、約22℃から、高ストリンジェント条件の約65℃まで増大させることができる。ハイブリダイゼーションは、当業界で公知の方法やそれに準じる方法に従って行なうことができる。また、市販のライブラリーを使用する場合、添付の使用説明書に記載の方法に従って行なうことができる。
実施例、比較例においては、以下の材料及び方法を用いた。
大腸菌(Escherichia coli)を、100mg/Lアンピシリンナトリウム又は100mg/Lカナマイシン硫酸塩を含むLB培地5mlに植菌し、37℃で一晩培養した。その後、これを同培地200mlに植菌し、O.D.が0.6になるまで培養後、イソプピル‐β‐チオガラクトピラノシド(IPTG)を終濃度0.5mMとなるように添加し、そのまま12時間28℃において培養した。
培養した菌体を集菌し、20mMリン酸カリウム緩衝液(pH7.5)に懸濁し、超音波破砕を行い、その上清を粗抽出液として、活性測定に用いた。酵素活性は、吸光光度計を用いて反応生成物の吸収波長の吸光度を5分間測定することによって測定し定量した。PAL活性は100mM Tris Buffer(pH8.0)、20mMフェニルアラニンに粗抽出液を加えて反応させ桂皮酸の生成を290nmの波長の吸光度の変化を測定し、4APAL活性については基質に20mM 4-アミノフェニルアラニンを用いて4-アミノ桂皮酸に由来する315nmの波長の吸光度を測定して定量した。
大腸菌(Escherichia coli)を100mg/Lアンピシリンナトリウム又は100mg/Lカナマイシン硫酸塩含むLB培地5mlに植菌し、37℃で一晩培養した。その後、これを同培地200mlに植菌し、O.Dが0.6になるまで培養後、IPTGを終濃度0.5mMとなるように添加し、さらに12時間28℃において培養した。これらの培養の際の回転数は120rpmとした。培養した菌体を集菌し、20mMリン酸カリウム緩衝液(pH7.5)に懸濁し、超音波破砕を行い、遠心分離の後に、その上清をHis-Trapカラムを用いて精製し、活性測定に用いた。酵素活性の測定方法は、前述の粗抽出液の測定方法に準じた。
(1)前培養
LB培地に100mg/Lアンピシリンナトリウム又は100mg/Lカナマイシン硫酸塩を含んだ培地を用い、37℃で大腸菌を培養した。培地5mlが入った試験管に予め生育させた寒天培地から、一白金耳植菌し、一晩120rpmで振盪培養した。
(2)本培養
前培養した菌体培養液をバッフル付きフラスコに入った同培地200mlに1%植菌した。37℃にて振とう培養を行い、培養液のO.D.が0.3になった時点で所定量のIPTGを加え、28℃で12時間培養し目的の遺伝子の発現誘導を行なった。
(3)反応方法
本培養後の菌体を回収し、50mMのKPi Buffer(pH7)で1回洗浄した後、20mMの基質(4-アミノフェニルアラニン、フェニルアラニン)が入った反応液5mlに懸濁させ、休止菌体反応を行なった。37℃で振盪しながら、0~24時間反応させた。反応後、上清を遠心管に写し、遠心分離によって回収し、以下のように高速液体クロマトグラフィー(HPLC)を用いて反応生成物を定量した。
(4)定量方法
HPLC(1200 infinity series: Hewlett Packerd)を用いて定量を行った。Purospher STAR RP-18 endcappedカラムを使用し、溶離液として20mMリン酸カリウム(pH7.0)と100%メタノールの混合液を用いた。210nm、254nm及び280nmの波長の吸収を用いて測定した。
大腸菌株として、NST37[ATCC 31882、米国特許第4,681,852号、遺伝子型aroG,aroF,pheA,tyrR,ryrA,trpE]と、BL21(DE3)[Novagen、遺伝子型F-,ompT,hsdSB(rB -mB -),gal(λcI857,ind1,Sam7,nin5,lacUV5-T7gene1),dcm(DE3)]を使用した。
大腸菌はLB培地中37℃で培養した。前培養は5mlの培地を入れた試験管を用いて行い、本培養は培地200mlをバッフル付きフラスコに入れ、前培養液を1%添加し、全培養と同じ条件下で、120rpmで震盪し培養を行った。
(1)pHSG298-AtPAL4の作製
植物シロイヌナズナ(Arabidopsis thaliana)由来のPAL(GenBank GI:30681254、gene locus:AT3G10340、塩基配列を配列番号3、アミノ酸配列を配列番号4に示す)を2つのプライマー(5’-CCGGATCCATGGAGCTATGCAATCAAAACAATC-3’及び5’-CCGCATGCTCAACAGATTGAAACCGGAGCTCCG-3’)を用いてPCRにより増幅後、BamHIとSphIにより処理し、予め同じ酵素で処理したpHSG298gxrA(Fujita, T. et al., Appl. Microbiol. Biothechnol. 97, 8887-8894 (2013))と連結してpHSG298-AtPAL4を作製した。
(2)pET28a-RgPALの作製
酵母ロドトルラ・グルチニス(Rhodotorula glutinis)由来のPAL(塩基配列を配列番号1、アミノ酸配列を配列番号2に示す)をNdeIとEcoRIを用いて制限酵素処理し、同じくNdeIとEcoRIを用いて制限酵素処理したプラスミドpET28a(Novagen)と連結して、pET28a-RgPALを作製した。
(3)pET28a-AtPAL4の作製
植物シロイヌナズナ(Arabidopsis thaliana)由来のPALを2つのプライマー(5’-CCCATATGGAGCTATGCAATCAAAACAATC-3’及び5’-CCGAATTCTCAACAGATTGAAACCGGAGCTCCG-3’)を用いてPCRにより増幅後、NdeIとEcoRIを用いて制限酵素処理し、同じくNdeIとEcoRIを用いて制限酵素処理したプラスミドpET28a(Novagen)と連結して、pET28a-RgPALを作製した。
これまでの研究からシロイヌナズナ由来のAtPAL4は高いPAL活性を示すことが示されていた。そこで、本発明者らは、AtPAL4はPAL活性だけでなく4APAL活性も高いのではないかと推定し、AtPAL4の組み換え酵素の4APAL活性を測定した。AtPAL4をpHSG298ベクターに接続し、大腸菌NST37に導入した株(NST37‐pHSG298‐AtPAL4)の粗抽出液は、以下の表2に示すように0.012 μmol/min/mgの4APAL活性を示した。
AtPAL4を導入した大腸菌NST37‐pHSG298‐AtPAL4は休止菌体反応において、4-アミノフェニルアラニンが4-アミノ桂皮酸に変換されたとはいえ、その生産量は最大67mg/Lであり、満足できるもではなかった。かかる4APAL活性は、更なる変換率の向上が望まれるグルコースを原料とした生産を行う際には不十分であると予想された。そこで、AtPAL4のアミノ酸配列の改変によって、AtPAL4の4APAL活性を更に上昇させることを試みた。Watts KT. et al. Discovery of a Substrate Selectivity Switch in Tyrosine Ammonia-Lyase, a Member of the Aromatic Amino Acid Lyase Family (2006) Chemistory & Biology, 13, 1317-1326には、芳香族アミノ酸のリアーゼファミリーに属する酵素(紅色光合成細菌由来のTyrosine Ammonia Lyaseとシアノバクテリア・トウモロコシ・パセリ・油脂蓄積酵母由来のPAL)に保存されている基質選択に関わる1アミノ酸をヒスチジンからフェニルアラニンに変えることにより基質特異性がチロシンからフェニルアラニンに変化することが報告されている。そこで、芳香族リアーゼファミリーに属する酵素とAtPAL4のアミノ酸配列を比較し、該当するアミノ酸を特定したところ、このアミノ酸はAtPAL4では123番目のヒスチジン(H123)に相当していることが分かった。本発明において基質としている4-アミノフェニルアラニンとPALの本来の基質であるフェニルアラニンとの違いは4位のアミノ基の有無である。そこで、H123をアミノ基とイオン結合しやすいと予想される酸性アミノ酸であるグルタミン酸又はアスパラギン酸に置換させることとした。これらの改変型AtPAL4を発現させた大腸菌(NST37-pHSG298-AtPAL4-H123D及びNST37-pHSG298-AtPAL4-H123E)を培養し、同様の方法で休止菌体反応とPAL活性と4APAL活性の測定を行なった。その結果、図1に示すように、休止菌体においては、H123E変異の導入により桂皮酸の生産量は0.06倍に減少したが、4-アミノフェニルアラニンの生産は1.25倍に増加した(図1(A)参照)。このことから、H123をグルタミン酸に変異させることよりAtPAL4の4-アミノフェニルアラニンへの選択性が上昇することが示された。また、活性測定の結果、AtPAL4-H123Eを導入した大腸菌の4APAL活性は、野生型活性の1.2倍であった(図1(B)参照)。この酵素活性の上昇が休止菌体反応における生産性の向上に貢献しているものと予想される。
NST37‐pHSG298‐AtPAL4の休止菌体反応をpH7、pH8、pH9の反応液を用いて行なった。コントロールとしてプラスミドを導入していない大腸菌NST37を用いた。まず、コントロールとの比較より、ベクターがある場合にのみ、4-アミノ桂皮酸のピークが顕著に検出されることからAtPAL4によって4-アミノフェニルアラニンから4-アミノ桂皮酸への反応が進むことが確認できた。図2に示すように、pH7では4-アミノ桂皮酸が51.3mg/Lしか生産されなかったが、pH8、pH9で休止菌体反応を行った場合、反応液中の4-アミノ桂皮酸がそれぞれ170mg/L、178mg/L生産されたことから、pH8とpH9の休止菌体反応によって4-アミノ桂皮酸が生成されることが分かった。
PAL活性を有することが知られている酵母ロドトルラ・グルチニス(Rhodotorula glutinis)由来のPAL(以下、RgPALともいう。)をpET28aにクローニングしたプラスミドを、大腸菌BL21(DE3)株に導入することによって大腸菌BL21‐pET28a‐RgPAL を作製し、これを用いて4-アミノ桂皮酸の生産を試みた。
大腸菌BL21‐pET28a‐RgPALの菌体を以下の方法で調製した。大腸菌BL21‐pET28a‐RgPALは、5 mlのLB培地(1%トリプトン、0.5%酵母抽出液、0.5%塩化ナトリウム)に100 mg/Lカナマイシン硫酸塩を含んだ培地を用いて37℃で一晩120 rpmで振盪培養した。得られた菌体培養液のうち2mLを500mL容の三角フラスコに入った同培地200 mlに移した後、37℃、120 rpmで振盪培養した。培養液の濁度が0.3になった時点でIPTGを加え、28℃、120 rpmで12時間、振盪培養した。得られた大腸菌BL21‐pET28a‐RgPALの菌体を遠心分離によって回収し、50 mMリン酸カリウム(pH 7)で1回洗浄したのち、20mMの基質(4-アミノフェニルアラニンまたはフェニルアラニン)が入った同水溶液に懸濁させ、37℃で振盪しながら、24時間保温した。反応後、生成した4-アミノ桂皮酸または桂皮酸をHPLCを用いて定量した。また、これと同じ方法を用い、pHを8または9とした50 mMリン酸カリウム水溶液中での反応もおこなった。その結果、図3に示すように、pH7では4-アミノ桂皮酸の生産量は610mg/Lであったが、pH8、pH9では4-アミノ桂皮酸がそれぞれ830mg/L、820mg/L生産された。いずれのpHにおいてもAtPAL4を発現させた場合よりも高い変換活性を示したことから、RgPALは4-アミノ桂皮酸の生産のために優れた酵素であると判断された。特に、pH7のときは変換効率が12倍に改善された。
グルコースから4-アミノ桂皮酸を生産する際には、フェニルアラニン生産性の大腸菌を用いるため、副産物としてフェニルアラニンが生産される。従って、目的とするPALは、フェニルアラニンが存在する条件下で4-アミノフェニルアラニンを生産できなければならない。そこで、フェニルアラニンと4-アミノフェニルアラニンの混合基質を加えた条件下での菌体によるPAL活性を測定した。具体的には、図4に示すように、4-アミノフェニルアラニンの濃度は20mMと固定し、それにフェニルアラニンを20mM、10mM、5mM、0mM加えた条件下でRgPALを発現させた大腸菌BL21(DE3)株又はAtPAL4を発現させた大腸菌NST37株の休止菌体反応を行い、フェニルアラニンが存在する条件下であってもPALは4-アミノフェニルアラニンを基質とするのか、その場合、どのくらいの割合で変換するのか調べた。その結果、4-アミノフェニルアラニンのみを基質とした場合、RgPALはAtPAL4の12倍の4-アミノ桂皮酸を生産することが示された。また、4-アミノフェニルアラニンのみを基質とした場合、RgPALはAtPAL4の36%の桂皮酸しか生産しなかった。Pheと4APheが共在する場合においても、RgPALはAtPAL4よりも多くの4-アミノ桂皮酸を生産し、少量の桂皮酸を生産することが示された。以上のことから、RgPALは、AtPAL4よりも4-アミノフェニルアラニンへの特異性が高いことが分かった。
pET28a-AtPAL4及びその誘導体並びにpET28a-RgPALをBL21(DE3)に導入して得られたAtPAL4及びその変異体並びにRgPALを発現する大腸菌の菌体が示す4APAL活性の違いは、各大腸菌におけるPALの発現量の違いによる可能性を考え、これらの大腸菌によるRgPALとAtPAL4の発現量を調べた。まず、SDS-PAGEによる発現の確認を行った。AtPAL4とその変異体の分子量は75.5kDa、RgPALの分子量は75.6kDaと予想されるところ、それぞれのPALを導入した大腸菌のいずれにおいても、SDS-PAGE上でこれらに該当するバンドが確認された。
さらに、図5に示すように、細胞粗抽出液を用いた活性測定を行い、これらの酵素の発現量を定量した。このとき、基質として用いる4-アミノフェニルアラニンおよび4-アミノ桂皮酸の濃度は20mMとした。その結果、RgPALを発現させた大腸菌の細胞粗抽出液が最も高いPAL活性と4APAL活性を示すことが分かった。RgPALのPAL活性と4APAL活性は、AtPAL4のものに比較して、ぞれぞれ、8.5倍と8倍であった。以上の結果より、休止菌体反応におけるRgPALを発現させたときの4-アミノ桂皮酸の生産量の多さの原因の一つはRgPALの発現量の多さによるものであると考えられた。
実施例3の大腸菌では、pET28AtPAL4及びその変異体並びにRgPALをHisタグとの融合タンパク質として発現させているので、これらの酵素をアフィニティーカラムを用いて簡易に精製することが可能である。これらを発現させた菌体を培養後、菌体を回収し、破砕後、精製を行った。精製された酵素を用いて、K m値とk cat値を算出した。K m値とk cat値の結果を、それぞれ、以下の表3と4に示す。
休止菌体反応後の反応液から、4-アミノ桂皮酸の抽出を試みた。まず、反応液から菌体を取り除いて得られる上清を20ml集め、エバポレーターで、水を蒸発させた。次に、4-アミノ桂皮酸がアセトンに可溶であることから、得られた固体に5mlのアセトンを加えて4-アミノ桂皮酸を抽出した後、アセトンを揮発させて4-アミノ桂皮酸を取り出すことを試みた。得られた抽出物をHPLC解析した結果を図6に示す。4-アミノ桂皮酸以外の主な3つのピークA、B、Cの高さをそれぞれ93%、89%、61%減少させることに成功した。これにより、純度は90%以上となった。4-アミノ桂皮酸の収量は消費された4-APheを100としたときに、純度換算して58%であった。

- 酵母ロドトルラ・グルチニス由来のアンモニアリアーゼ又はその改変体を用いて、4-アミノフェニルアラニンから4-アミノ桂皮酸を製造する方法。
- 前記アンモニアリアーゼが、フェニルアラニンアンモニアリアーゼであることを特徴とする請求項1に記載の方法。
- 前記改変体が、酵母ロドトルラ・グルチニス由来のアンモニアリアーゼのアミノ酸配列において、1又は数個のアミノ酸が欠失、置換、挿入又は付加されたアミノ酸配列であることを特徴とする、請求項1又は2に記載の方法。

からなる群から選ばれる酵素を用いて、4-アミノフェニルアラニンを4-アミノ桂皮酸に変換することを特徴とする、4-アミノ桂皮酸の製造方法。 - 前記酵素を発現する菌の休止菌体を、4-アミノフェニルアラニンを含む溶液中で培養又は反応させることを含む、請求項4に記載の方法。
- 前記菌が、大腸菌(Escherichia coli)である、請求項5に記載の方法。
- 前記休止菌体反応が、pH8~pH9で行われる、請求項5又は6に記載の方法。
- 前記休止菌体が、培養菌体、粉末菌体、及び固定化菌体からなる群から選択される、請求項5~7のいずれか1項に記載の方法。
- 請求項4~8のいずれか1項に記載の方法により製造された4-アミノ桂皮酸。
