実施例4で得られた化合物28を用いて、10merのオリゴヌクレオチド類縁体(化合物29から32:以下の表4に示す)を、DNA/RNAオリゴヌクレオチド自動合成機nS-8(株式会社ジーンデザイン製)により0.2μmolスケールのCPG担体を用いて合成した。化合物28のアセトニトリル溶液(0.1M)のカップリング時間は8分間で行い、それ以外は天然のDNA合成と同条件にて行った。活性化剤は5-[3,5-ビス(トリフルオロメチル)フェニル]-1H-テトラゾール(0.25M)を用いた。合成したオリゴヌクレオチドは28%アンモニア水溶液を用いてCPG担体から切り出した。得られた化合物29から31の粗成績体については、逆相簡易カラム(Sep-Pak@Plus C18 Environmental Cartridges, Waters社)により精製し、続いてトリフルオロ酢酸(TFA)50%で24時間処理した後に逆相HPLC精製を行った。得られた化合物32の粗成績体については、逆相簡易カラム(Sep-Pak@Plus C18 Environmental Cartridges, Waters社)により精製し、さらに逆相HPLC精製を行った。
実施例5で得られた各種オリゴヌクレオチド(化合物28を用いて製造したオリゴヌクレオチド類縁体の化合物29から31、および天然型ヌクレオシドを含有するオリゴヌクレオチドの化合物33)と以下の表5および6に記載の標的鎖(10merのポリAおよび配列番号3~5)とをアニーリング処理して複合体を形成させた後、複合体の50%が解離する温度であるTm値を測定することにより、オリゴヌクレオチドのハイブリッド形成能を調べた。
実施例4で得られた化合物28を用いて、種々の塩基からなる9merのオリゴヌクレオチド類縁体(化合物34:以下の表7に示す)を、合成したオリゴヌクレオチドを28%アンモニア水溶液を用いてCPG担体から切り出した後に、55℃にて12時間かけて塩基部の保護基を除去した以外は、実施例5と同様にして合成および精製した。比較のために、天然型のヌクレオシドを含有するオリゴヌクレオチド(化合物35:以下の表7に示す)も、標準的なホスホロアミダイトプロトコルに従って同様に合成し精製した。
5’-d(TTTTTTTTXT)-3’の配列のXがそれぞれ以下に示す通りである、10merの各種オリゴヌクレオチドを調製した。すなわち、以下の各種オリゴヌクレオチドを調製した:実施例1のヌクレオシド類縁体(化合物8)を用いて製造した、Xがグアニジン架橋型人工核酸であるオリゴヌクレオチド類縁体(すなわち「化合物13」);実施例4のヌクレオシド類縁体(化合物28)を用いて製造した、Xがグアニジン架橋型人工核酸であるオリゴヌクレオチド類縁体(すなわち「化合物32」);XがLNA-T(チミンLNA)であるオリゴヌクレオチド(株式会社ジーンデザイン製:化合物38);XがDNA-T(チミンDNA)であるオリゴヌクレオチド(10merのオリゴdT、すなわち「化合物33」);およびXにS-オリゴ(酸化剤の代わりに硫化剤としてD-1,4-ジチオトレイトール(DDTT、ChemGene社製)を用いたこと以外は標準的なホスホロチオアート合成のプロトコルに従い、合成および精製した)を用いて標準的なホスホロアミダイトプロトコルに従って合成し精製したオリゴヌクレオチド(化合物39:陽性コントロールとして使用)。
5’-d(TTTTTTTTX)-3’の配列のXがグアニジン架橋型人工核酸であるオリゴヌクレオチド類縁体である、9merのオリゴヌクレオチド(化合物40)を、以下のように調製した。
以下の表9に記載する化合物33(10merのオリゴdTからなる天然型オリゴヌクレオチド);化合物29~31、42および43(化合物28のグアニジン架橋型核酸を含有するオリゴヌクレオチド類縁体);ならびに化合物44~48(LNA-Tを含有するオリゴヌクレオチド)について、10merのポリAにアニーリング処理して複合体を形成させた後、複合体の50%が解離する温度であるTm値を測定することにより、オリゴヌクレオチドのハイブリッド形成能を調べた。
表9のグアニジン架橋型人工核酸が5残基導入されているオリゴヌクレオチド類縁体(化合物43)およびLNAが5残基導入されているオリゴヌクレオチド(化合物48)について、完全相補配列を有するDNA標的鎖(フルマッチ型)および1塩基ミスマッチを有するDNA標的鎖(ミスマッチ型)のそれぞれに対する結合親和性を評価した。標的鎖の配列は、フルマッチ型:5’-(AAAAAAAAAA)-3’、ミスマッチ型:5’-(AAAAATAAAA)-3’である。結合親和性は、実施例11と同様に、アニーリング処理して複合体を形成させた後、複合体の50%が解離する温度であるTm値を測定することにより評価した。
(1)蛍光標識GuNA修飾オリゴヌクレオチド(F-GuNA-ODN)の合成と同定
まず、実施例4で得られた化合物28、天然型ヌクレオシド、および後述の蛍光修飾のためのアミダイト体を用いて、DNA/RNAオリゴヌクレオチド自動合成機nS-8(株式会社ジーンデザイン製)により、オリゴヌクレオチド類縁体を合成した。合成したオリゴヌクレオチド類縁体は表10に示す化合物53から57の前駆体(化合物49)になる化合物である。この前駆体(化合物49)の構造を以下に示す。

まず、前準備として、細胞観察用のガラスボトムディッシュ(品種コード:3970-035, Iwaki)のガラス部分に100μg/mLコラーゲン/塩酸(pH 3.0)(Cellmatrix Type I-C, 新田ゼラチン株式会社製)を1mL添加することにより、コラーゲンコーティングを行った。

Hoechst 33342: Ex 360/40nm, DM 400nm, BA 460/50nm (DAPI-B、株式会社キーエンス製), 2秒
LysoTracker(登録商標) Red DND-99: Ex 540/25 nm, DM 565 nm, BA 605/55 nm (TRITC, 株式会社キーエンス製), 1.2秒
Claims (6)
- 以下の式IまたはIIで表される化合物またはその塩:
B1は、α群から選択される任意の置換基を1以上有していてもよいプリン-9-イル基または2-オキソ-1,2-ジヒドロピリミジン-1-イル基を表し、ここで、該α群は、水酸基、核酸合成の保護基で保護された水酸基、炭素数1から6の直鎖アルキル基、炭素数1から6の直鎖アルコキシ基、メルカプト基、核酸合成の保護基で保護されたメルカプト基、炭素数1から6の直鎖アルキルチオ基、アミノ基、炭素数1から6の直鎖アルキルアミノ基、核酸合成の保護基で保護されたアミノ基、およびハロゲン原子からなり;
R1、R12およびR13は、それぞれ独立して、水素原子、分岐または環を形成していてもよい炭素数1から7のアルキル基、アミノ基の保護基、または
R2およびR3は、それぞれ独立して、水素原子、核酸合成の水酸基の保護基、分岐または環を形成していてもよい炭素数1から7のアルキル基、分岐または環を形成していてもよい炭素数2から7のアルケニル基、該α群から選択される任意の置換基を1以上有していてもよくそしてヘテロ原子を含んでいてもよい炭素数3から12のアリール基、該α群から選択される任意の置換基を1以上有していてもよくそしてヘテロ原子を含んでいてもよい炭素数3から12のアリール部分を有するアラルキル基、該α群から選択される任意の置換基を1以上有していてもよいアシル基、該α群から選択される任意の置換基を1以上有していてもよいシリル基、該α群から選択される任意の置換基を1以上有していてもよいリン酸基、核酸合成の保護基で保護されたリン酸基、-P(R4)R5[式中、R4およびR5は、それぞれ独立して、水酸基、核酸合成の保護基で保護された水酸基、メルカプト基、核酸合成の保護基で保護されたメルカプト基、アミノ基、炭素数1から5のアルコキシ基、炭素数1から5のアルキルチオ基、炭素数1から6のシアノアルコキシ基、および/または炭素数1から6のアルキル基で置換されたアミノ基を表す]を表す)。 - 前記式IまたはIIにおいて、前記B1が、6-アミノプリン-9-イル基、2,6-ジアミノプリン-9-イル基、2-アミノ-6-クロロプリン-9-イル基、2-アミノ-6-フルオロプリン-9-イル基、2-アミノ-6-ブロモプリン-9-イル基、2-アミノ-6-ヒドロキシプリン-9-イル基、6-アミノ-2-メトキシプリン-9-イル基、6-アミノ-2-クロロプリン-9-イル基、6-アミノ-2-フルオロプリン-9-イル基、2,6-ジメトキシプリン-9-イル基、2,6-ジクロロプリン-9-イル基、6-メルカプトプリン-9-イル基、2-オキソ-4-アミノ-1,2-ジヒドロピリミジン-1-イル基、4-アミノ-2-オキソ-5-フルオロ-1,2-ジヒドロピリミジン-1-イル基、4-アミノ-2-オキソ-5-クロロ-1,2-ジヒドロピリミジン-1-イル基、2-オキソ-4-メトキシ-1,2-ジヒドロピリミジン-1-イル基、2-オキソ-4-メルカプト-1,2-ジヒドロピリミジン-1-イル基、2-オキソ-4-ヒドロキシ-1,2-ジヒドロピリミジン-1-イル基、2-オキソ-4-ヒドロキシ-5-メチル-1,2-ジヒドロピリミジン-1-イル基、または4-アミノ-5-メチル-2-オキソ-1,2-ジヒドロピリミジン-1-イル基である、請求項1に記載の化合物またはその塩。
- 前記式IまたはIIにおいて、前記B1が、2-オキソ-4-ヒドロキシ-5-メチル-1,2-ジヒドロピリミジン-1-イル基である、請求項1に記載の化合物またはその塩。
- 以下の式I’またはII’で表されるヌクレオシド構造を少なくとも1つ含有するオリゴヌクレオチドまたはその薬理学上許容される塩:
B1は、α群から選択される任意の置換基を1以上有していてもよいプリン-9-イル基または2-オキソ-1,2-ジヒドロピリミジン-1-イル基を表し、ここで、該α群は、水酸基、核酸合成の保護基で保護された水酸基、炭素数1から6の直鎖アルキル基、炭素数1から6の直鎖アルコキシ基、メルカプト基、核酸合成の保護基で保護されたメルカプト基、炭素数1から6の直鎖アルキルチオ基、アミノ基、炭素数1から6の直鎖アルキルアミノ基、核酸合成の保護基で保護されたアミノ基、およびハロゲン原子からなり;
R1、R12、およびR13は、それぞれ独立して、水素原子、分岐または環を形成していてもよい炭素数1から7のアルキル基、アミノ基の保護基、または
R2およびR3は、それぞれ独立して、水素原子、核酸合成の水酸基の保護基、分岐または環を形成していてもよい炭素数1から7のアルキル基、分岐または環を形成していてもよい炭素数2から7のアルケニル基、該α群から選択される任意の置換基を1以上有していてもよくそしてヘテロ原子を含んでいてもよい炭素数3から12のアリール基、該α群から選択される任意の置換基を1以上有していてもよくそしてヘテロ原子を含んでいてもよい炭素数3から12のアリール部分を有するアラルキル基、該α群から選択される任意の置換基を1以上有していてもよいアシル基、該α群から選択される任意の置換基を1以上有していてもよいシリル基、該α群から選択される任意の置換基を1以上有していてもよいリン酸基、核酸合成の保護基で保護されたリン酸基、-P(R4)R5[式中、R4およびR5は、それぞれ独立して、水酸基、核酸合成の保護基で保護された水酸基、メルカプト基、核酸合成の保護基で保護されたメルカプト基、アミノ基、炭素数1から5のアルコキシ基、炭素数1から5のアルキルチオ基、炭素数1から6のシアノアルコキシ基、および/または炭素数1から6のアルキル基で置換されたアミノ基を表す]を表す)。 - 前記式I’またはII’において、前記B1が、6-アミノプリン-9-イル基、2,6-ジアミノプリン-9-イル基、2-アミノ-6-クロロプリン-9-イル基、2-アミノ-6-フルオロプリン-9-イル基、2-アミノ-6-ブロモプリン-9-イル基、2-アミノ-6-ヒドロキシプリン-9-イル基、6-アミノ-2-メトキシプリン-9-イル基、6-アミノ-2-クロロプリン-9-イル基、6-アミノ-2-フルオロプリン-9-イル基、2,6-ジメトキシプリン-9-イル基、2,6-ジクロロプリン-9-イル基、6-メルカプトプリン-9-イル基、2-オキソ-4-アミノ-1,2-ジヒドロピリミジン-1-イル基、4-アミノ-2-オキソ-5-フルオロ-1,2-ジヒドロピリミジン-1-イル基、4-アミノ-2-オキソ-5-クロロ-1,2-ジヒドロピリミジン-1-イル基、2-オキソ-4-メトキシ-1,2-ジヒドロピリミジン-1-イル基、2-オキソ-4-メルカプト-1,2-ジヒドロピリミジン-1-イル基、2-オキソ-4-ヒドロキシ-1,2-ジヒドロピリミジン-1-イル基、2-オキソ-4-ヒドロキシ-5-メチル-1,2-ジヒドロピリミジン-1-イル基、または4-アミノ-5-メチル-2-オキソ-1,2-ジヒドロピリミジン-1-イル基である、請求項4に記載のオリゴヌクレオチドまたはその薬理学上許容される塩。
- 前記式I’またはII’において、前記B1が、2-オキソ-4-ヒドロキシ-5-メチル-1,2-ジヒドロピリミジン-1-イル基である、請求項4に記載のオリゴヌクレオチドまたはその薬理学上許容される塩。
