Abstract
Description
アンチジーンは、標的遺伝子の二重鎖DNA部位に対応するオリゴヌクレオチドである。該DNA部位とオリゴヌクレオチドで三重鎖を形成させることにより、DNAからmRNAへの転写を抑制する。
アプタマーは、特定の分子と特異的に結合するDNA、RNA及び/又はそれらの構造類似体である。標的タンパク質と結合することにより該タンパク質の機能を阻害する。
デコイ核酸は、特定の転写調節因子の結合部位と同じ配列を含む短いDNAである。該転写調節因子と遺伝子の結合を阻害し、該転写調節因子によって活性化される遺伝子群の発現を抑制する。

以下に本明細書中で使用する各用語を説明する。なお、本明細書中、各用語は単独で使用されている場合も、又は他の用語と一緒になって使用されている場合も、特に記載の無い限り、同一の意義を有する。
つまり、本発明における「核酸塩基部分」とは、核酸(DNA、RNA)の塩基部分を構成する置換若しくは非置換の複素環式基又は置換若しくは非置換の炭素環式基である。

本発明のオリゴヌクレオチドのリン酸部分としては、天然の核酸が有するリン酸ジエステル結合、S-オリゴ(ホスホロチオエート)、M-オリゴ(メチルフォスフォネート)、ボラノホスフェート等が挙げられる。
本発明のオリゴヌクレオチド中の式(II)で示されるヌクレオシド構造以外の塩基部分は、上記「Bx」について定義した任意の核酸塩基であってよい。
本発明のオリゴヌクレオチド中の式(II)で示されるヌクレオシド構造以外の糖部分としては、天然のリボース若しくはデオキシリボース、公知の修飾を有するリボース若しくはデオキシリボース等が挙げられる。公知の修飾としては、例えば、2’-O-CH2-CH2-O-CH3(2’MOE)、4’-CH2-O-2’(LNA、Locked nucleic acid)、AmNA(アミドBNA)(Bridged nucleic acid、WO2011/052436参照)等による修飾が挙げられる。
よって、式(I)で示される化合物中のBxが、保護基で保護された置換基を有する場合、オリゴヌクレオチド合成の際に、脱保護を行う。
経口投与用組成物としては、散剤、顆粒剤、水若しくは非水性媒体に溶解させた懸濁液又は溶液、カプセル、粉末剤、錠剤等が挙げられる。

窒素気流下、化合物1(合成法はJ.Med.Chem.2000,43,4516に記載の方法に準じる)(435mg,0.941mmol)のテトラヒドロフラン溶液(8.7mL)に、室温下、化合物2(合成法はChemMedChem 2011,6,840に記載の方法に準じる)(273mg,1.411mmol)、ジイソプロピルエチルアミン(164μL,0.941mmol)とヨウ化銅(17.9mg,0.094mmol)を加えて70℃下、14時間撹拌した。反応液をセライトでろ過した。溶媒を減圧留去して、得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン: 酢酸エチル=50:50→15:80)で精製し、化合物3(613mg,99%)を白色固体物質として得た。

窒素気流下、化合物3(589mg,0.899mmol)のジクロロメタン溶液(5.9mL)に、氷冷下、ピリジン(239μL,2.97mmol)とトリフルオロメタンスルホン酸無水物(228μL,1.35mmol)を加えて40分間撹拌した。反応液へ飽和重曹水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。溶媒を減圧留去して、化合物4の粗製物(714mg)を得た。

窒素気流下、化合物1(5.0g,9.92mmol)のジクロロメタン溶液(25mL)に-78℃下、三塩化ホウ素(1mol/Lジクロロメタン溶液、49.6mL,49.6mmol)を滴下し、30分間撹拌した後、氷冷下、2時間撹拌した。-78℃下、反応液にメタノール(50mL)とピリジン(30mL)を滴下して10分撹拌した後、室温まで昇温した。溶媒を減圧留去し、得られた残渣にピリジン(20mL)を加え、析出した固体をろ過により除去した。ろ液を濃縮して得られた残渣をピリジン(25mL)に溶かし、室温下、1,3-ジクロロ-1,1,3,3-テトライソプロピルジシロキサン(4.76mL,14.89mmol)を加え、一晩撹拌した。反応液へメタノール(10mL)を加えた後、溶媒を減圧留去した。得られた残渣を酢酸エチルに溶かし、5%クエン酸水溶液、飽和食塩水で洗浄後、硫酸マグネシウムで乾燥した。溶媒を減圧留去して、得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=80:20→50:50)で精製し、化合物9(4.66g,90%)を白色固体物質として得た。

窒素気流下、化合物13(589mg,1.036mmol)のピリジン溶液(10mL)に、室温下、4,4’-ジメトキシトリチルクロリド(702mg,2.073mmol)を加えて45時間撹拌した。溶媒を減圧留去した後、残渣へ飽和重曹水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。溶媒を減圧留去して、化合物14の粗製物(1.37g)を得た。
室温下、化合物14の粗製物(1.37g)のテトラヒドロフラン溶液(14mL)にトリエチルアミン(431μL,3.11mmol)とトリエチルアミン三フッ化水素酸塩(1.01mL,6.22mmol)を加えて30分間撹拌した。反応液へ飽和重曹水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄した後に、硫酸ナトリウムで乾燥させた。溶媒を留去して得られた粗製物をシリカゲルカラムクロマトグラフィー(クロロホルム:メタノール=100:0→94:6)で精製し、化合物8(626mg,99%)を白色固体物質として得た。

化合物23(合成法はJ.Med.Chem.2000,43.4516に記載の方法に準じる)(39.8g,100mmol)のテトラヒドロフラン溶液(400mL)に-78℃下、0.97Mメチルマグネシウムブロミド テトラヒドロフラン溶液(155mL,150mmol)をゆっくりと滴下した後、1時間撹拌した。反応液に飽和塩化アンモニウム水溶液を加え15分撹拌した後、酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄した後、硫酸ナトリウムで乾燥した。溶媒を減圧留去し、化合物24の粗製物(43g)を得た。
窒素気流下、ジクロロメタン(100mL)にオキサリルクロリド(13.1mL,150mmol)加えた後、-78℃下、ジメチルスルホキシド(21.3mL、300mmol)を滴下し、5分間撹拌した。続いて化合物24の粗製物(41.4g)のジクロロメタン溶液(20mL)を滴下し、トリエチルアミンを-78℃下滴下した。緩やかに室温まで昇温させ、室温で2時間撹拌した。反応液に水を加えてクエンチし、ジクロロメタンで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=95:5→65:35)で精製し、化合物25(28.5g,69.1%(工程1~工程2の通算収率))を淡黄色泡状物質として得た。

窒素気流下、化合物26の粗製物(29.1g)のアセトニトリル溶液(146mL)に室温下、チミン(10.5g,83mmol)、N,O-ビス(トリメチルシリル)アセトアミド(55.2mL,223mmol)を加え、50℃下5分間撹拌した。室温下、トリメチルシリルトリフルオロメタンスルホン酸(11.54mL,63.7mmol)を加え、1.5時間加熱還流した。反応液へ飽和重曹水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥して溶媒を減圧留去し化合物27の粗製物(33g)を白色固体物質として得た。

化合物27の粗製物(33g)のテトラヒドロフラン溶液(330mL)に40%メチルアミン水溶液(27mL)を加えて、氷冷下1.5時間撹拌した。テトラヒドロフランを減圧留去した後、残渣へ水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。溶媒を減圧留去して得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=80:20→50:50)で精製し、化合物28(14.1g、47%(工程3~工程5の通算収率))を白色固体物質として得た。

窒素気流下、化合物28(14g)のジクロロメタン溶液(70mL)に室温下、トリエチルアミン(10.1mL,72.8mmol)、メタンスルホニルクロライド(2.72mL,35.0mmol)を加えて45分間撹拌した。溶媒を減圧留去した後、残渣へ水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。溶媒を減圧留去し、化合物29の粗製物(16.3g)を得た。

化合物29の粗製物(16.3g)のテトラヒドロフラン溶液(81mL)に1mol/L水酸化ナトリウム水溶液(81mL)を加えて、室温下2時間撹拌した。N,N-ジメチルホルムアミド(35mL)を加えた後、70℃下18時間撹拌した。飽和塩化アンモニウム水溶液でクエンチし、テトラヒドロフランを減圧留去した後、残渣へ水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。溶媒を減圧留去して得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=60:40→10:90)で精製し、化合物30(14.1g)を白色固体物質として得た。

窒素気流下、化合物30(2.68g,5.58mmol)のN.N-ジメチルホルムアミド溶液(25mL)に、氷冷下、炭酸ナトリウム(3.08g,22.3mmol)、クロロメチルピバレート(1.83mL,12.3mmol)を加えて50℃下10時間撹拌した。飽和塩化アンモニウム水溶液を加えて撹拌した後、酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。得られた粗製物のN,N-ジメチルホルムアミド溶液(20mL)に、炭酸ナトリウム(1.86g,13.5mmol)、クロロメチルピバレート(1.11mL,7.41mmol)を加えて50℃下16時間撹拌した。反応液へ飽和塩化アンモニウム水溶液を加えて撹拌した後、酢酸エチルで抽出し、有機層を水、飽和食塩水で洗浄した。硫酸ナトリウムで乾燥した後、溶媒を減圧留去し、得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=65:35→50:50)で精製し、化合物31(2.8g,73%)を白色固体物質として得た。

窒素気流下、化合物32の粗製物(865mg)のエタノール溶液(8mL)に室温下、ヒドラジン一水和物(68.8μL,1.42mmol)を加えて、80℃下1時間撹拌した。反応液へ飽和塩化アンモニウム水溶液を加えて撹拌した後、酢酸エチルで抽出し、有機層を水、飽和食塩水で洗浄した。硫酸ナトリウムで乾燥した後、溶媒を減圧留去し、得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=90:10→65:35)で精製し、化合物33(815mg,98%(工程9~工程10の通算収率))を白色固体物質として得た。

窒素気流下、化合物35の組製物(1.4g)のアセトニトリル溶液(14mL)に室温下、1mol/L水酸化ナトリウム水溶液(7.0ml,7.0mmol)を加えて、2時間撹拌した。反応液へ飽和塩化アンモニウム水溶液を加えて撹拌した後、アセトニトリルを減圧留去した。残渣を酢酸エチルで抽出し、有機層を水、飽和食塩水で洗浄した。硫酸ナトリウムで乾燥した後、溶媒を減圧留去し、得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=65:35→40:60)で精製し、化合物36(403mg,42%(工程12~工程13の通算収率))を白色固体物質として得た。

化合物36(445mg,0.914mmol)のテトラヒドロフラン溶液(5mL)に室温下、20%水酸化パラジウム-炭素粉末(250mg)を添加し、水素気流下、3時間撹拌した。セライト濾過後、温メタノールで洗浄した。溶媒を減圧留去し、化合物37の粗製物(291mg)を得た。

窒素気流下、化合物37の粗製物(291mg)のピリジン溶液(3.0mL)に、室温下、4,4’‐ジメトキシトリチルクロリド(465mg,1.37mmol)を加えて2時間撹拌した。反応液に飽和重曹水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。溶媒を減圧留去して、得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=60:40→35:65)で精製し、化合物38(242mg,44%(工程14~工程15の通算収率))を白色固体物質として得た。

窒素気流下、化合物38(230mg,0.378mmol)のジクロロメタン溶液(2.8mL)に室温下、ジイソプロピルエチルアミン(198μL,1.13mmol)、2-シアノエチル-N,N,-ジイソプロピルクロロホスホロアミダイト(101μL,0.45mmol)を加え、1時間撹拌した。反応液へ飽和重曹水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄した後に、無水硫酸ナトリウムで乾燥させた。溶媒を留去して得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=50:50→25:75)で精製し、化合物IV-1(214mg,71%)を白色固体物質として得た。

(1)化合物I-1のヌクレオシド構造を含有するオリゴヌクレオチド
実施例1の(1-A)又は(1-B)で得られた化合物I-1から調製されるオリゴヌクレオチド(1)~(5)(表6)を、nS-8(ジーンデザイン社製)により0.2μmolスケールで合成した。化合物I-1 (アミダイトユニット) は、アセトニトリルに溶解して用いた。なお、表6において、Xは化合物I-1のヌクレオシド構造(下記式II-1)を意味する。アミダイトユニット (化合物I-1)と5’-末端の水酸基とのカップリング時間は32秒 (標準) から16分に延長した。5’-末端がDMTr基により保護されかつ固相に指示されたオリゴヌクレオチドを、オリゴヌクレオチド(1)~(4)については、室温下、28%アンモニア水:40%メチルアミン水溶液 (1:1)で、オリゴヌクレオチド(5)については、55℃下、28%アンモニア水 で処理し、溶媒を留去した。得られた粗製物をSep-Pak C18 Plus Short Cartridge(Waters社製)により粗精製した後、逆相HPLC(Gilson PLC2020、分取カラムとしてWatersXBridgeTM Shield RP18 Column 5.0 μm (10mm x 50mm)及びYMC Hydrosphere C18 Column 5.0 μm (10mm x 150mm)) により精製した。
合成したオリゴヌクレオチドの純度は、逆相HPLC(WatersXBridgeTM C18 Column 5.0μm(4.6mm x 50mm)及びYMC Hydrosphere C18 Column 5.0μm (4.6mm x 100mm)) により確認した (WatersXBridgeTM C18 Column条件:0.1Mトリエチルアンモニウム酢酸バッファー (pH 7.0)中、1mL/分で30分の5 → 8% (v/v) アセトニトリルのグラジエント、YMC Hydrosphere C18 Column条件:0.01 Mトリエチルアンモニウム酢酸バッファー (pH 7.0) 中、1mL/分で30分の6 → 10%(v/v)アセトニトリルのグラジエント及び1mL/分で30分の7.5 → 11.5% (v/v) アセトニトリルのグラジエント) 。また、分子量は、ESI-TOF-MASS測定により決定した。結果を表6に示す。
実施例1の(4)で得られた化合物IV-1を用いて、(1)と同様の方法にて、オリゴヌクレオチド(8)~(12)を合成した。ESI-TOF-MASS測定により決定した分子量を表7に示す。なお、表7において、Xpは化合物IV-1のヌクレオシド構造(下記式II-2)を意味する。
実施例1の(3)で得られた化合物VII-1を用いて、(1)と同様の方法にて、オリゴヌクレオチド(13)~(17)を合成した。ESI-TOF-MASS測定により決定した分子量を表8に示す。なお、表8において、Xqは化合物VII-1のヌクレオシド構造(下記式II-3)を意味する。
WO2011/052436を参考にアミド架橋型アミダイト体を合成した。実施例2(1)のオリゴヌクレオチド(1)と同様にアミド架橋型オリゴヌクレオチド(6)を合成した。分子量はMALDI-TOF-MASS測定により決定した。結果を表9に示す。
(1)化合物I-1のヌクレオシド構造を含有するオリゴヌクレオチド
実施例2の(1)で合成したオリゴヌクレオチド(1)~(4)(アンチセンス鎖) とセンス鎖 (3’-CGC AAA AAA CGA-5’)とをアニーリング処理した後、Tm値を測定することにより、オリゴヌクレオチド(1)~(4)のハイブリッド形成能を調べた。オリゴヌクレオチドのヌクレオシド部分が非修飾であるヌクレオチド(0)をコントロールとして利用した。
塩化ナトリウム100mM、リン酸ナトリウム緩衝液(pH7.2)10mM、オリゴヌクレオチド(アンチセンス鎖)4.0μM、及びセンス鎖4.0μMを含むサンプル溶液(150μM)をヒートブロック(95℃)で5分間、加熱した後、12時間かけて室温まで冷却した。分光高度計(SHIMADZU UV-1800)のセル室内に結露防止のために窒素気流を通し、サンプル溶液を5℃まで徐々に冷却し、さらに15分間5℃に保った後、測定を開始した。温度は90℃まで毎分0.5℃ずつ上昇させ、0.5℃間隔で260nmにおける紫外部吸収を測定した。なお、温度上昇による濃度変化を防止するため、セルは蓋付きのものを用いた。結果を表10に示す。
実施例2の(2)で合成したオリゴヌクレオチド(8)~(11)について、センス鎖(3’-CGC AAA AAA CGA-5’)とアニーリング処理した後、Tm値を測定した。Tm値の測定方法は、(1)と同様である。結果を表11に示す。
(3)化合物VII-1のヌクレオシド構造を含有するオリゴヌクレオチド
実施例2の(3)で合成したオリゴヌクレオチド(13)~(16)について、センス鎖(3’-CGC AAA AAA CGA-5’)と混合した後、Tm値を測定した。Tm値の測定方法は、(1)と同様である。結果を表12に示す。
実施例2で合成したオリゴヌクレオチド(5)(Triazole BNA)、オリゴヌクレオチド(12)(Pyrazole BNA)及びオリゴヌクレオチド(17)(Tetrazole BNA)について、オリゴヌクレオチドを3’側から分解するエキソヌクレアーゼに対する耐性を検討した。オリゴヌクレオチド(5)のX部分が非修飾であるヌクレオチド(nature)、オリゴヌクレオチド(5)のX部分が下記2’,4’‐BNA/LNAであるヌクレオチド(LNA)をコントロールとして利用した。

この結果、ヌクレオチド(nature)及びヌクレオチド(LNA)は40分後にはすべて分解している。ヌクレオチド(12)(Pyrazole BNA)は40分後の残存率は55%であり、ヌクレオチド(5)(Triazole BNA)は40分後の残存率は80%であった。オリゴヌクレオチド(17)(Tetrazole BNA)は40分後において、残存率がほぼ100%であった。よって、本発明のオリゴヌクレオチドは、非修飾のオリゴヌクレオチドや公知の人工ヌクレオチドから調整されるオリゴヌクレオチド(LNA)を大きく上回る酵素耐性を有することが分かった。従って、本発明のオリゴヌクレオチドは体内での持続性が非常によいと考えられることから、核酸医薬品を合成するための材料として有用であると考えられる。
