本発明に係る放射性フッ素標識化合物は、上記のとおり、式(1)で表される化合物又はその塩であるが、特に好ましい実施形態によれば、例えば、下記式(4)で表される1-(2,2-ジヒドロキシメチル-3-[18F]フルオロプロピル)-2-ニトロイミダゾール、下記式(5)で表される1-(2,2-ジヒドロキシメチル-3-[18F]フルオロプロピル)-4-ヒドロキシメチル-2-ニトロイミダゾール、又は、下記(6)に表される1-(3,3-ジヒドロキシメチル-4-[18F]フルオロブチル)-2-ニトロイミダゾールとすることができる。
1.NMRスペクトルによる、非放射性化合物の分子構造の決定
実施例中、非放射性化合物の構造は、NMRスペクトルで同定した。NMRスペクトルは、NMR装置として、JNM-ECP-500(日本電子株式会社製)を使用して得た。共鳴周波数は、1H-NMRでは500MHz、19F-NMRでは470MHzとした。1H-NMRについては、重溶媒中の残留溶媒シグナルを参照として使用した(DMSO-d:δ2.5;CD3ODδ3.3;CDCl3δ7.26)。全ての化学シフトはデルタスケール(δ)上のppmであり、そしてシグナルの微細分裂については、略号(s:シングレット、d:ダブレット、t:トリプレット、dt:ダブルトリプレット、m:マルチプレット、brs:ブロードシングレット)を用いて示した。

2,2-ジメチル-5-[(2-ニトロ-1H-イミダゾール-1-イル)メチル]-5-(p-トルエンスルホニルオキシメチル)-1,3-ジオキサン)は、化合物1の標識前駆体である。図1は、この合成スキームを示す。
2-ブロモメチル-2-ヒドロキシメチル-1,3-プロパンジオール423mg(2.13mmol相当)をアセトン1.0mLに溶解し、10-カンファースルホン酸247mg(1.07mmol相当)を加え、室温(25℃)で2日間攪拌した。反応終了後、トリエチルアミンを加え、溶媒を留去した後、得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=3/1)にて精製を行い、5-ブロモメチル-2,2-ジメチル-5-ヒドロキシメチル-1,3-ジオキサン409mg(1.71mmol相当)を得た。
5-ブロモメチル-2,2-ジメチル-5-ヒドロキシメチル-1,3-ジオキサン409mg(1.71mmol相当)をジメチルホルムアミド5mLに溶解し、イミダゾール233mg(3.42mmol相当)とt-ブチルジメチルクロロシラン309mg(2.05mmol相当)を加え、室温(25℃)で18時間攪拌した。反応終了後、飽和塩化アンモニウム水溶液と水を加え、酢酸エチルで3回抽出を行なった。合わせた酢酸エチル層を、水及び飽和食塩水で洗浄した後、無水硫酸マグネシウムで乾燥後減圧濃縮し、得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=20/1)にて精製を行い、5-ブロモメチル-5-(t-ブチルジメチルシロキシメチル)-2,2-ジメチル-1,3-ジオキサン578mg(1.64mmol相当)を得た。
5-ブロモメチル-5-(t-ブチルジメチルシロキシメチル)-2,2-ジメチル-1,3-ジオキサン578mg(1.64mmol相当)をジメチルホルムアミド10mLに溶解した後、2-ニトロイミダゾール186mg(1.64mmol相当)と炭酸カリウム680mg(4.92mmol相当)とを加え、100℃で18時間加熱した。反応終了後、反応液を室温(25℃)まで冷却し、飽和塩化アンモニウム水溶液と水を加え、酢酸エチルで3回抽出を行った。合わせた酢酸エチル層を、水及び飽和食塩水で洗浄した後、無水硫酸マグネシウムで乾燥後減圧濃縮し、得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=3/1)にて精製を行い5-(t-ブチルジメチルシロキシメチル)-2,2-ジメチル-5-[(2-ニトロ-1H-イミダゾール-1-イル)メチル]-1,3-ジオキサン363mg(0.942mmol相当)を得た。
5-(t-ブチルジメチルシロキシメチル)-2,2-ジメチル-5-[(2-ニトロ-1H-イミダゾール-1-イル)メチル]-1,3-ジオキサン363mg(0.942mmol相当)をテトラヒドロフラン10.0mLに溶解し、テトラブチルアンモニウムフルオリド・テトラヒドロフラン溶液0.94mL(1.0mol/L溶液,0.94mmol相当)を加え、室温(25℃)で10分攪拌した。反応終了後、塩飽和塩化アンモニウム水溶液と水を加え、酢酸エチルで3回抽出を行った。合わせた酢酸エチル層を、水及び飽和食塩水で洗浄した後、無水硫酸マグネシウムで乾燥後減圧濃縮し、得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=1/3)にて精製を行い2,2-ジメチル-5-ヒドロキシメチル-5-[(2-ニトロ-1H-イミダゾール-1-イル)メチル]-1,3-ジオキサン221mg(0.815mmol相当)を得た。
2,2-ジメチル-5-ヒドロキシメチル-5-[(2-ニトロ-1H-イミダゾール-1-イル)メチル]-1,3-ジオキサン100mg(0.369mmol相当)をピリジン4.0mLに溶解し、0℃に冷却した後、p-トルエンスルホニルクロリド77.3mg(0.406mmol相当)を加え、室温(25℃)で1時間攪拌した。反応終了後、塩飽和塩化アンモニウム水溶液と水を加え、酢酸エチルで3回抽出を行った。合わせた酢酸エチル層を、水及び飽和食塩水で洗浄した後、無水硫酸マグネシウムで乾燥後減圧濃縮し、得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=1/1)にて精製を行い2,2-ジメチル-5-[(2-ニトロ-1H-イミダゾール-1-イル)メチル]-5-(p-トルエンスルホニルオキシメチル)-1,3-ジオキサン115mg(0.270mmol相当)を得た。
1-(2,2-ジヒドロキシメチル-3-フルオロプロピル)-2-ニトロイミダゾールは、化合物1中のフッ素原子をフッ素18からフッ素19に換えた以外は化合物1と同一の構造を有する化合物(化合物1のコールド体)である。図2は、この合成スキームを示す。
2,2-ジメチル-5-[(2-ニトロ-1H-イミダゾール-1-イル)メチル]-5-(p-トルエンスルホニルオキシメチル)-1,3-ジオキサン30mg(0.0705mmol相当)をアセトニトリル1.0mLに溶解し、クリプトフィックス222(商品名、メルク社製)39.8mg(0.106mmol相当)と、フッ化カリウム5.1mg(0.088mmol相当)と、炭酸カリウム2.0mg(0.014mmol相当)とを加え、加熱還流下で3時間攪拌した。反応終了後、反応液を室温(25℃)まで冷却した後、溶媒を留去し、得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=2/1)にて精製を行い2,2-ジメチル-5-フルオロメチル-5-[(2-ニトロ-1H-イミダゾール-1-イル)メチル]-1,3-ジオキサン9.2mg(0.034mmol相当)を得た。
2,2-ジメチル-5-フルオロメチル-5-[(2-ニトロ-1H-イミダゾール-1-イル)メチル]-1,3-ジオキサン56.3mg(0.206mmol相当)をメタノール2mLに溶解し、1mol/L塩酸2mLを加え、80℃で2時間加熱した。反応終了後、反応液を室温(25℃)まで冷却した後、溶媒を留去し、得られた粗生成物を酢酸エチルで洗浄することで1-(2,2-ジヒドロキシメチル-3-フルオロプロピル)-2-ニトロイミダゾール47.3mg(0.203mmol相当)を得た(図2、Step2)。

これをアルゴンガスの通気下110℃に加熱して水を蒸発させた後、アセトニトリル(0.3mL×2)を加えて共沸させ乾固させた。ここに実施例1にて合成した2,2-ジメチル-5-[(2-ニトロ-1H-イミダゾール-1-イル)メチル]-5-(p-トルエンスルホニルオキシメチル)-1,3-ジオキサン5mg(11.4μmol相当)を溶解したアセトニトリル溶液0.3mLを加え、110℃で10分加熱した。続けて1mol/L塩酸0.3mL加え、110℃で3分間加熱した。反応終了後、水1.0mLを加え、HPLC(移動相:0.1(v/v)%トリフルオロ酢酸水溶液/アセトニトリル(0.1(v/v)%トリフルオロ酢酸含有)(v/v)=85/15、流速:4.0mL/分)により、実施例2で得られた化合物1のコールド体を用いて保持時間10分のピークを化合物1の画分として同定し、同定した化合物1の画分を分取した。
当該画分に水10mLを添加した液をSep-Pak(登録商標) HLB Plas(商品名、日本ウォーターズ株式会社製)に通液し、化合物1を当該カラムに吸着捕集した。このカラムを水3mLで洗浄した後、エタノール2mLを通液して、化合物1を溶出させた。得られた放射能量は388MBq(合成開始後69分)であった。また、TLC分析を行ったところ、その放射化学的純度は98%であった。
2,2-ジメチル-5-[(4-メトキシメトキシメチル-2-ニトロ-1H-イミダゾール-1-イル)メチル]-5-(p-トルエンスルホニルオキシメチル)-1,3-ジオキサンは、化合物2の標識前駆体である。図4は、この合成スキームを示す。
5-ブロモメチル-2,2-ジメチル-5-ヒドロキシメチル-1,3-ジオキサン1.1g(4.6mmol相当)をジメチルホルムアミド23mLに溶解し、イミダゾール626mg(9.2mmol相当)と塩化t-ブチルジフェニルシラン1.43mL(5.5mmol相当)を加え、室温(25℃)で5時間攪拌した。反応終了後、飽和塩化アンモニウム水溶液を滴下し、酢酸エチルで3回抽出した。合わせた酢酸エチル層を水及び飽和塩化ナトリウム水溶液で洗浄し、無水硫酸マグネシウムで乾燥後減圧濃縮した。得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=20/1)で精製し、5-ブロモメチル-5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-1,3-ジオキサン1.70g(3.56mmol相当)を得た。
5-ブロモメチル-5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-1,3-ジオキサン1.70g(3.56mmol相当)をジメチルホルムアミド36mLに溶解し、2-ニトロイミダゾール402mg(3.56mmol相当)と炭酸カリウム1.48g(10.7mmol相当)を加え、油浴で80℃に加熱した後18時間攪拌した。反応終了後、水を滴下し、酢酸エチルで3回抽出した。合わせた酢酸エチル層を水及び飽和塩化ナトリウム水溶液で洗浄し、無水硫酸マグネシウムで乾燥後減圧濃縮した。得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=3/1)で精製し、5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-5-[(2-ニトロ-1H-イミダゾール-1-イル)メチル]-1,3-ジオキサン362mg(0.71mmol相当)を得た。
5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-5-(2-ニトロ-1H-イミダゾール-1-イル)-1,3-ジオキサン315mg(0.62mmol相当)をジメチルホルムアミド6mLに溶解し、N-ブロモスクシンイミド110mg(0.62mmol相当)を加え、室温(25℃)で17時間攪拌した。反応終了後、飽和炭酸水素ナトリウム水溶液を滴下し、続いて飽和チオ硫酸ナトリウム水溶液を滴下した後、酢酸エチルで3回抽出を行った。合わせた酢酸エチル層を無水硫酸マグネシウムで乾燥後減圧濃縮し、得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=4/1)で精製し、5-{(4-ブロモ-2-ニトロ-1H-イミダゾール-1-イル)メチル}-5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-1,3-ジオキサン131mg(0.22mmol相当)を得た。
5-{(4-ブロモ-2-ニトロ-1H-イミダゾール-1-イル)メチル}-5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-1,3-ジオキサン280mg(0.48mmol相当)をジメチルホルムアミド4.7mLに溶解し、トリブチルビニルスズ278μL(0.95mmol相当)とテトラキストリフェニルホスフィンパラジウム55mg(0.05mmol相当)を加え、油浴で80℃に加温した後、5時間攪拌した。反応終了後、反応液を減圧濃縮し、得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=4/1)で精製し、5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-5-[(4-ビニル-2-ニトロ-1H-イミダゾール-1-イル)メチル]-1,3-ジオキサン163mg(0.30mmol相当)を得た。
5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-5-[(4-ビニル-2-ニトロ-1H-イミダゾール-1-イル)メチル]-1,3-ジオキサン163mg(0.30mmol相当)に水/1,4-ジオキサン=3/1の混合溶液2.0mLを加え、酸化オスミウム38mg(マイクロカプセル、和光純薬社製、0.015mmol相当)とヨウ素酸ナトリウム130mg(0.60mmol相当)を加え、室温(25℃)で5日間攪拌した。反応終了後、酸化オスミウムを取り出し、反応液を酢酸エチルで3回抽出を行った。合わせた酢酸エチル層を水及び飽和塩化ナトリウム水溶液で洗浄し、無水硫酸マグネシウムで乾燥後減圧濃縮した。得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=5/1)で精製し、5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-5-[(4-ホルミル-2-ニトロ-1H-イミダゾール-1-イル)メチル]-1,3-ジオキサン107mg(0.20mmol相当)を得た。
5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-5-[(4-ホルミル-2-ニトロ-1H-イミダゾール-1-イル)メチル]-1,3-ジオキサン107mg(0.20mmol相当)をエタノール3.0mLに溶解し、水素化ホウ素ナトリウム10mg(0.24mmol相当)を加え、室温(25℃)で10分間攪拌した。反応終了後、アセトンを滴下し減圧濃縮した。得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=1/1)で精製し、5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-5-{(4-ヒドロキシメチル-2-ニトロ-1H-イミダゾール-1-イル)メチル}-1,3-ジオキサン101mg(0.19mmol相当)を得た。
5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-5-[(4-ヒドロキシメチル-2-ニトロ-1H-イミダゾール-1-イル)メチル]-1,3-ジオキサン101mg(0.19mmol相当)をジクロロメタン2.0mLに溶解し、氷浴にて約0℃に冷却した。N,N-ジイソプロピルエチルアミン125μL(0.72mmol相当)と塩化メトキシメチル41μL(0.51mmol相当)を加え、室温(25℃)へ昇温しながら26時間攪拌した。反応終了後、飽和炭酸水素ナトリウム水溶液を滴下し、酢酸エチルで3回抽出を行った。合わせた酢酸エチル層を水及び飽和塩化ナトリウム水溶液で洗浄し、無水硫酸マグネシウムで乾燥後減圧濃縮した。得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=2/1)で精製し、5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-5-[(4-メトキシメトキシメチル-2-ニトロ-1H-イミダゾール-1-イル)メチル]-1,3-ジオキサン86mg(0.15mmol相当)を得た。
5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-5-[(4-メトキシメトキシメチル-2-ニトロ-1H-イミダゾール-1-イル)メチル]-1,3-ジオキサン86mg(0.15mmol相当)をテトラヒドロフラン1.0mLに溶解し、テトラブチルアンモニウムフルオリド・テトラヒドロフラン溶液0.17mL(1mol/L溶液,0.17mmol相当)を加え、室温(25℃)で10分間攪拌した。反応終了後、溶媒を留去し、得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=1/1)にて精製を行い、2,2-ジメチル-5-ヒドロキシメチル-5-{(4-メトキシメトキシメチル-2-ニトロ-1H-イミダゾール-1-イル)メチル}-1,3-ジオキサン47mg(0.14mmol相当)を得た。
2,2-ジメチル-5-ヒドロキシメチル-5-[(4-メトキシメトキシメチル-2-ニトロ-1H-イミダゾール-1-イル)メチル]-1,3-ジオキサン23mg(0.11mmol相当)をトリエチルアミン1.0mLに溶解し、N,N-ジメチルアミノピリジン1mg(0.01mmol相当)とp-トルエンスルホニルクロリド23mg(0.12mmol相当)を加え、室温(25℃)で16時間攪拌した。反応終了後、飽和塩化アンモニウム水溶液を加え、酢酸エチルで3回抽出を行った。合わせた酢酸エチル層を、無水硫酸マグネシウムで乾燥後減圧濃縮した。得られた粗生成物をフラッシュシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=1/3)にて精製を行い、2,2-ジメチル-5-[(4-メトキシメトキシメチル-2-ニトロ-1H-イミダゾール-1-イル)メチル]-5-(p-トルエンスルホニルオキシメチル)-1,3-ジオキサン16mg(0.03mmol相当)を得た。
1-(2,2-ジヒドロキシメチル-3-フルオロプロピル)-4-ヒドロキシメチル-2-ニトロイミダゾールは、化合物2中のフッ素原子をフッ素18からフッ素19に換えた以外は化合物2と同一の構造を有する化合物(化合物2のコールド体)である。図5は、この合成スキームを示す。

これをアルゴンガスの通気下110℃に加熱して水を蒸発させた後、アセトニトリル(0.3mL×2)を加えて共沸させ乾固させた。ここに上記実施例4にて合成した2,2-ジメチル-5-[(4-メトキシメトキシメチル-2-ニトロ-1H-イミダゾール-1-イル)メチル]-5-(p-トルエンスルホニルオキシメチル)-1,3-ジオキサン5mg(10μmol相当)を溶解したアセトニトリル溶液0.3mLを加え、110℃で10分加熱した。続けて1mol/L塩酸0.3mL加え110℃で3分間加熱した。反応終了後、室温に冷却し注射用水1.0mLを加え、HPLC(移動相:0.1%(v/v)トリフルオロ酢酸水溶液/アセトニトリル(0.1%(v/v)トリフルオロ酢酸含有)(v/v)=90/10、流速:3.0mL/分)に付して、実施例5で得られた化合物2のコールド体を用いて保持時間13分のピークを化合物2の画分として同定し、同定した化合物2の画分を分取した。

2,2-ジメチル-5-[2-(2-ニトロ-1H-イミダゾール-1-イル)エチル]-5-(p-トルエンスルホニルオキシメチル)-1,3-ジオキサンは、化合物3の標識前駆体である。図7は、この合成スキームを示す。
水素化ナトリウム506mg(60%油性,13mmol相当)をテトラヒドロフラン5.0mLに添加し、氷浴で約0℃に冷却した。そこへマロン酸ジエチル3.2mL(20.7mmol相当)を加えた(溶液A)。2-ベンジルオキシ-1-ブロモエタン2.3g(10.7mmol相当)をテトラヒドロフラン3.0mLに溶解し、これを溶液Aに10分間かけて滴下し、一晩加熱還流を行った。反応終了後、反応液に0.5mol/L塩酸水溶液を滴下し、ジエチルエーテルで3回抽出を行った。合わせたジエチルエーテル層を飽和食塩水で洗浄した後、無水硫酸マグネシウムで乾燥後減圧濃縮し、得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=20/1)にて精製を行い2-(2-ベンジルオキシエチル)マロン酸ジエチル2.85g(9.67mmol相当)を得た。
2-(2-ベンジルオキシエチル)マロン酸ジエチル2.85g(9.67mmol相当)をアセトニトリル20mLに溶解し、炭酸水素カリウム1.60g(11.7mmol相当)とパラホルムアルデヒド465mg(ホルムアルデヒドとして11.7mmol相当)を添加し、室温(25℃)で一晩攪拌した。反応終了後、反応液に0.5mol/L塩酸水溶液を滴下し、クロロホルムで3回抽出を行った。合わせたクロロホルム層を、無水硫酸マグネシウムで乾燥後減圧濃縮し、得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=4/1)にて精製を行い、2-(2-ベンジルオキシエチル)-2-ヒドロキシメチルマロン酸ジエチル2.78g(8.76mmol相当)を得た。
2-(2-ベンジルオキシエチル)-2-ヒドロキシメチル-マロン酸ジエチル1.03g(3.24mmol相当)をメタノール10mLに溶解し、これを水素化ホウ素ナトリウム1.69g(44.6mmol相当)に20分間かけて滴下した。これを一晩加熱還流した後、室温(25℃)まで冷却した。反応液に水を添加し、30分間反応させた。反応液をクロロホルムで洗浄し、水相を減圧濃縮した。得られた残渣にエタノール100mLを添加し、2時間加熱還流した。反応終了後、ただちにシリカゲルカラムクロマトグラフィー(溶離液:エタノール)で処理し、4-ベンジルオキシ-2,2-ジヒドロキシメチルブタノール粗生成物543mgを得た。
4-ベンジルオキシ-2,2-ジヒドロキシメチルブタノール粗生成物543mgをアセトン3mLに溶解し、10-カンファースルホン酸232mg(1.0mmol相当)を添加し、室温(25℃)で一晩反応させた。反応終了後、トリエチルアミンを滴下し減圧濃縮した。得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=4/1)にて精製を行い、5-ベンジルオキシシエチル-2,2-ジメチル-5-ヒドロキシメチル-1,3-ジオキサン459mg(1.64mmol相当)を得た。
5-ベンジルオキシシエチル-2,2-ジメチル-5-ヒドロキシメチル-1,3-ジオキサン459mg(1.64mmol相当)をジメチルホルムアミド8mLに溶解し、イミダゾール197mg(3.20mmol相当)とt-ブチルジフェニルクロロシラン0.51mL(1.92mmol相当)を加え、室温(25℃)で22時間攪拌した。反応終了後、飽和塩化アンモニウム水溶液を加え、酢酸エチルで3回抽出を行った。合わせた酢酸エチル層を、水及び飽和食塩水で洗浄した後、無水硫酸マグネシウムで乾燥後減圧濃縮した。得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=20/1)にて精製を行い、5-ベンジルオキシエチル-2,2-ジメチル-5-(t-ブチルジフェニルシロキシメチル)-1,3-ジオキサン570mg(1.56mmol相当)を得た。
5-ベンジルオキシエチル-2,2-ジメチル-5-(t-ブチルジフェニルシロキシメチル)-1,3-ジオキサン570mg(1.56mmol相当)を酢酸エチル30mLに溶解し、アルゴン雰囲気下でパラジウム炭素100mgを加え、水素雰囲気下室温(25℃)で22時間攪拌した。反応終了後、沈殿物をろ過し、ろ液を減圧濃縮した。得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=4/1)にて精製を行い、5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-5-ヒドロキシエチル-1,3-ジオキサン174mg(0.41mmol相当)を得た。
5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-5-ヒドロキシエチル-1,3-ジオキサン48mg(0.112mmol相当)をテトラヒドロフラン1mLに溶解し、トリフェニエルホスフィン32mg(0.123mmol相当)とアザジカルボン酸ジイソプロピル24μL(0.123mmol相当)と2-ニトロイミダゾール38mg(0.336mmol相当)を加え、室温(25℃)で4時間攪拌した。反応終了後、減圧濃縮し得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=4/1)にて精製を行い、5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-5-[2-(2-ニトロ-1H-イミダゾール-1-イル)エチル]-1,3-ジオキサン46mg(0.088mmol相当)を得た。
5-(t-ブチルジフェニルシロキシメチル)-2,2-ジメチル-5-[2-(2-ニトロ-1H-イミダゾール-1-イル)エチル]-1,3-ジオキサン46mg(0.088mmol相当)をテトラヒドロフラン1mLに溶解し、テトラブチルアンモニウムフルオリド・テトラヒドロフラン溶液0.11mL(1mol/L溶液,0.11mmol相当)を加え、室温(25℃)で1時間攪拌した。反応終了後、溶媒を留去し、得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:クロロホルム/メタノール(v/v)=10/1)にて精製を行い、2,2-ジメチル-5-ヒドロキシメチル-5-[2-(2-ニトロ-1H-イミダゾール-1-イル)エチル]-1,3-ジオキサン25mg(0.086mmol相当)を得た。
2,2-ジメチル-5-ヒドロキシメチル-5-[2-(2-ニトロ-1H-イミダゾール-1-イル)エチル]-1,3-ジオキサン25mg(0.086mmol相当)をジクロロメタン1mLに溶解し、1,4-ジアザビシクロ[2,2,2]オクタン19mg(0.172mmol相当)とp-トルエンスルホニルクロリド19mg(0.10mmol相当)を加え、室温(25℃)で4時間攪拌した。反応終了後、飽和塩化アンモニウム水溶液を加え、酢酸エチルで3回抽出を行った。合わせた酢酸エチル層を、無水硫酸マグネシウムで乾燥後減圧濃縮した。得られた粗生成物をシリカゲルカラムクロマトグラフィー(溶離液:ヘキサン/酢酸エチル(v/v)=1/3)にて精製を行い2,2-ジメチル-5-[2-(2-ニトロ-1H-イミダゾール-1-イル)エチル]-5-(p-トルエンスルホニルオキシメチル)-1,3-ジオキサン18mg(0.041mmol相当)を得た。
1-(3,3-ジヒドロキシメチル-4-フルオロブチル)-2-ニトロイミダゾールは、化合物3中のフッ素18をフッ素19に換えた以外は化合物3と同一の構造を有する化合物(化合物3のコールド体)である。図8は、この合成スキームを示す。
2,2-ジメチル-5-[2-(2-ニトロ-1H-イミダゾール-1-イル)エチル]-5-(p-トルエンスルホニルオキシメチル)-1,3-ジオキサン5mg(11.4μmol相当)をアセトニトリル1mLに溶解し、クリプトフィックス22214mg(商品名,メルク社,34.5μmol相当)とフッ化カリウム0.82g(14.1μmol相当)と炭酸カリウム0.4mg(2.9μmol相当)を加え、5時間加熱還流した。反応終了後、水を加え、クロロホルムで3回抽出を行った。合わせたクロロホルム層を無水硫酸マグネシウムで乾燥後減圧濃縮した。得られた粗生成物を、シリカゲルカラムクロマトグラフィーにて精製し、2,2-ジメチル-5-フルオロメチル-5-[2-(2-ニトロ-1H-イミダゾール-1-イル)エチル]-1,3-ジオキサンを痕跡量得た。
2,2-ジメチル-5-フルオロメチル-5-[2-(2-ニトロ-1H-イミダゾール-1-イル)エチル]-1,3-ジオキサンをアセトニトリル0.5mLに溶解し、1mol/L塩酸0.5mLを加え、80℃にて30分攪拌した。反応終了後、減圧濃縮し1-(3,3-ジヒドロキシメチル-4-フルオロブチル)-2-ニトロイミダゾールを痕跡量得た。
図9は、この合成スキームを示す。
これをアルゴンガスの通気下110℃に加熱して水を蒸発させた後、アセトニトリル(0.3mL×2)を加えて共沸させ乾固させた。ここに上記実施例7にて合成した2,2-ジメチル-5-[2-(2-ニトロ-1H-イミダゾール-1-イル)エチル]-5-(p-トルエンスルホニルオキシメチル)-1,3-ジオキサン5mg(11.4μmol相当)を溶解したアセトニトリル溶液0.3mLを加え、110℃で10分加熱した。続けて1mol/L塩酸0.3mL加え、110℃で3分間加熱した。反応終了後、水1.0mLを加え、HPLC(移動相:0.1%(v/v)トリフルオロ酢酸水溶液/アセトニトリル(0.1%(v/v)トリフルオロ酢酸含有)(v/v)=85/15、流速:2.5mL/分)に付して、実施例8で得られた化合物3のコールド体を用いて保持時間16分のピークを化合物3の画分として同定し、同定した化合物3の画分を分取した。

化合物1、化合物2及び化合物3のインビボにおける腫瘍細胞への集積性を、マウス由来乳がん細胞株(EMT6)を用いて以下検討した。
担がんモデルマウスの作成:使用した動物は、6週齢の雌性Balb/c nu/nuマウス(入手先:日本SLC)である。この動物を用いて、マウス由来乳がん細胞株(入手先:ATCC)であるEMT6のマトリゲル縣濁液(細胞数2.5×106cells/0.05mL)を右脇腹から右肩にかけての部位に皮下移植した。腫瘍細胞は、移植3日後には定着していることを確認され、腫瘍体積は、移植9~11日後において200~400mm3に達していた。
実験方法:化合物1、化合物2、化合物3、及びRasey JSらの方法(Int J Radiat Oncol Biol Phys,Nov;17(5):985-991,1989)に従って合成した[18F]フルオロミソニダゾール([18F]FMISO)をそれぞれ10mg/mLアスコルビン酸含有生理食塩液にて希釈し、放射能濃度1000MBq/mLに調整し試料溶液とした。作製した担がんモデルマウスをイソフルラン麻酔下に置き、これらの試料溶液を、尾静脈より約20MBq投与した。投与60分後に動物用PET装置(形式:eXplore Vista,GE社製)を用い、static撮像を実施した。収集時間は、化合物1を投与した担がんモデルマウスでは20分、化合物2、化合物3及び[18F]FMISOを投与した担がんモデルマウスでは10分とした。収集データは、OSEM法により再構成して画像化し、画像解析ソフトウェアを用いて冠状断層像及び軸上断層像を作成した。
結果を図10~13に示す。図10は、化合物1を投与した担がんモデルマウスの冠状断層像である。図11は、化合物1を投与した担がんモデルマウスの軸上断層像である。図12(a)は化合物2、図12(b)は化合物3、図12(c)は[18F]FMISOを、それぞれ投与した担がんモデルマウスの冠状断層像である。図13(a)は化合物2、図13(b)は化合物3、図13(c)は[18F]FMISO、をそれぞれ投与した担がんモデルマウスの軸上断層像である。図11~13中、矢じりの尖端方向に腫瘍部位があることを示す。
実施例10で得られた軸上断層像を用いて、腫瘍内のSUVの最高値を測定した。腫瘍が描出された軸上断層像の全スライス面において、腫瘍全体に関心領域(以下ROI(region of interest))を設定し、測定を行った。なお、軸上断層像は61スライス面から構成されており、測定を行なったスライス面の中で最も高かったSUVを投与60分後におけるSUV最高値とした(腫瘍におけるSUV最高値)。一方、正常組織のSUVは、右体側部に腫瘍を移植した担がんモデルマウスにおける反対側である,左体側部の肺及び筋肉組織を対象部位とし、投与60分後における肺及び筋肉を含む左体側部全体にROIを設定し、測定を行った。ROI内の信号強度の平均値を測定し、各スライス面で測定した値の平均値を正常組織におけるSUVの平均値とした。下記数式(1)を用いて腫瘍正常組織比を算出し、腫瘍と正常組織とのコントラストに関する客観的指標として用いた。
表2に化合物1~3及び[18F]FMISOのSUV最高値及び腫瘍正常組織比を示す。投与60分後において化合物1~3が、いずれも、[18F]FMISOと比較して高い腫瘍正常組織比を示した。この結果から,化合物1、化合物2及び化合物3により、投与後早い時間点で腫瘍内の低酸素領域を高いコントラストをもって描出するする画像が得られることが示唆された。
化合物1~3及び[18F]FMISOを用い、本発明に係る化合物が腫瘍の低酸素領域を描出し得るか評価するため、下記の実験を行った。
各標識化合物における腫瘍内集積の局在を、オートラジオグラフィーを施行して確認した。実施例11の方法で作製した担がんモデルマウスのPET撮像終了後、心臓穿刺による放血により屠殺した直後に腫瘍を採取し、クリオスタット(形式:CM3050、Leica社製)を用いて組織切片(厚さ:10μm)の作製を行った。オートラジオグラフィーの測定には、18Fの半減期が短いことを考慮して、腫瘍組織切片の作製に時間を要しない未固定の新鮮凍結切片を用いた。当該腫瘍組織切片をイメージングプレートで8~10時間露光させた後、バイオイメージングアナライザー(形式:BAS-2500、FUJIFILM社製)を用いて画像を取り込んだ。取り込んだ画像につき、画像解析ソフトウェアを用いて画像解析を行った。
別に、腫瘍内の低酸素領域を確認する方法として、低酸素マーカーであるピモニダゾールを用いた免疫組織化学染色を、放射能減衰後の同切片を用いて、次の手順で実施した。腫瘍組織切片の固定と賦活化処理後、1次抗体にウサギポリクローナル抗ピモニダゾール抗体(入手先:Hypoxyprobe,Inc.、1:200)を、1次抗体に反応する2次抗体にビオチン標識抗ウサギ抗血清をそれぞれ使用して腫瘍組織切片と反応させ、ついで2次抗体に反応するHRP(horseradish peroxidase)標識ストレプトアビジンを用い、HRP活性をDAB(3,3'-Diaminobenzidine)を基質とした呈色反応で検出することで、腫瘍組織切片の低酸素領域を同定した。また細胞核を染色する核対比染色はマイヤーヘマトキシリンを用いた。近接切片として連続で薄切した1枚をネガティブコントロールとして用い、1次抗体だけを反応させない手順で上記同様の実験を行い、1次抗体以外の成分による腫瘍組織切片への非特異的な反応が認められないことを確認した。顕微鏡システム(形式:BZ‐9000、キーエンス社製)を用い、免疫組織化学染色により得られた標本画像の全体画像を取得した。取得した全体画像に対して画像処理を行い、低酸素領域を示すピモニダゾール陽性部位を抽出した。
結果を図14~17に示す。図14(b)は、化合物1の腫瘍内集積オートラジオグラフィーを示し、図14(a)は、図14(b)と同切片の免疫組織学的染色画像を示す。図15(b)は、化合物2の腫瘍内集積オートラジオグラフィーを示し、図15(a)は、図15(b)と同切片の免疫組織学的染色画像を示す。図16(b)は、化合物3の腫瘍内集積オートラジオグラフィーを示し、図16(a)は、図16(b)と同切片の免疫組織学的染色画像を示す。図17(b)は、[18F]FMISOの腫瘍内集積オートラジオグラフィーを示し、図17(a)は、図17(b)と同切片の免疫組織学的染色画像を示す。図14~16で示すように、化合物1~3の集積部位と低酸素マーカーであるピモニダゾールの局在は視覚的に一致していた。一方、図17に示すように、[18F]FMISOの集積部位と低酸素マーカーであるピモニダゾールの局在が視覚的に一致していない場所(図17中破線で囲んで示す)があった。
化合物1~3及び[18F]FMISOを用い、本発明に係る化合物が腫瘍の低酸素領域を定量的に描出し得るか評価するため、下記の実験を行った。
顕微鏡システム(形式:BZ-9000、キーエンス社製)を用い、実施例12で免疫組織化学染色により得られた病理像の全体画像及び10倍の対物レンズを用いてピモニダゾール陽性部位を強拡大した画像を取得した。 強拡大した画像は、無作為に選択した撮像領域について作成した。併せて顕微鏡システムにおけるナビゲーション機能により画像取得部位を記録し、ナビゲーション画像とした。取得した強拡大画像に対して画像処理を行い、強拡大画像に占めるピモニダゾール陽性面積を測定した。
各標識化合物のオートラジオグラフィー画像及び染色像は実施例12により取得したものを用いた。ナビゲーション画像の解像度をオートラジオグラフィー画像の解像度に合わせ、画像解析ソフトウェアを用いて両画像の幾何学的位置を合わせた。
オートラジオグラフィー画像において、強拡大画像取得部位に一致した部位にROIを設定し、ROIのPSL値(Photo‐Stimulated Luminescence value)を測定した。また、同一画像上において検体を含まない部位にROIを設定し、このPSL値をバックグランドとした。強拡大画像取得部位のPSL値をバックグラウンドのPSL値で減算し、正味のPSL値を求めた。低酸素マーカーであるピモニダゾール陽性面積を横軸、オートラジオグラフィー画像におけるPSL値を縦軸としてプロットし、相関係数を算出した。各標識化合物においてこの一連の操作を実施した。
結果を図18及び表3に示す。図18は、化合物1~3、及び、[18F]FMISOの、腫瘍内集積オートラジオグラフィーにおける低酸素領域の面積と信号強度との相関を示す図である。表3には、免疫組織学画像とオートラジオグラフィーとの相関係数を示す。図18においても明らかなように、[18F]FMISOでは、低酸素マーカーであるピモニダゾールの局在が認められる箇所において、低いPSL値を示す場合があり、PSL値と低酸素領域(ピモニダゾールの局在位置)とが視覚的に一致していない箇所が散見される。また、図18及び表3からも明らかなように、[18F]FMISOでは低酸素環境の占める割合と集積の強度を対応させた散布図において、そのプロットにばらつきが大きかった。一方、各標識化合物の集積は、実施例12における結果と同様、低酸素マーカーの局在が認められる箇所では、高いPSL値を示し、視覚的に一致している。図18からも明らかなように、各標識化合物の集積の強度は、同じ位置における低酸素環境の占める割合と相関していた。また、本発明に係る全ての標識化合物において、[18F]FMISOより高い相関係数が認められた。この結果から、各標識化合物の腫瘍への集積の強度は、[18F]FMISOと比較して、腫瘍内の低酸素領域の程度をより定量的に反映していることが示唆された。
化合物1~3及び[18F]FMISOを用い、担がんマウスにおける腫瘍及び各臓器への体内分布を測定した。
実験に使用した担がんモデルマウスは、実施例10で作製したものと同一であり、腫瘍体積が200~400mm3に達したものを1群につき3ないし4例選出した。担がんモデルマウスはイソフルラン麻酔下に置き、低酸素マーカーであるピモニダゾールを尾静脈より投与し、10分後に各標識化合物を投与した。投与してから60分及び120分後に担がんモデルマウスを屠殺した。担がんモデルマウスは全て心臓穿刺または腹部大動脈からの放血により屠殺し、その直後に解剖を行った。腫瘍の他に、血液、心臓、肺、胃、肝臓、胆嚢、腎臓、小腸及び大腸、筋肉、尿を回収し、それ以外の組織は残全身とした。採取した各組織の重量を測定した後、ガンマカウンターにより組織放射能量を測定し、各組織の%ID/g(%injection dose/g organ)に換算することで各標識化合物における体内分布の比較を行った。
結果を表4及び表5に示す。表4は、投与60分後の各組織への%ID/gを示す。表5は、投与120分後の各組織への%ID/gを示す。ただし、表4、5中、尿は%IDである。また、表4、5中、T/Bは、腫瘍/血液比を示し、T/Mは、腫瘍/筋肉比を示す。担がんモデルマウスにおける各標識化合物の腫瘍への放射能集積は、[18F]FMISOと比較して低い値を示していたものの、化合物1及び化合物3の血液への分布濃度は、各標識化合物投与60分後において、それぞれ1.70%ID/g及び2.81%ID/gであり、各標識化合物投与120分後では0.75%ID/g及び1.14%ID/gに低下した。また、尿への分布は各標識化合物投与60分後で62.13%ID及び45.80%IDであり、投与120分後で77.76%ID及び68.78%IDに増加した。この結果から、化合物1及び化合物3は、[18F]FMISOと比較して血液クリアランス及び尿中排泄が迅速であることが確認された。また、化合物2においては、各標識化合物投与60分後の血液への分布濃度及び尿への分布が0.60%ID/g及び80.42%IDを示したことにより、血液クリアランス及び尿中排泄がさらに迅速であることが確認された。さらに、各標識化合物投与60分後及び120分後における腫瘍血液比は[18F]FMISOと比較して高い値を示した。なお、免疫組織化学染色の結果から、腫瘍内が低酸素であることが確認された。
Claims (10)
- R2及びR3が、一緒になってメチレン基、1-メチルエタン-1,1-ジイル基、エタン-1,1-ジイル基、又は1-フェニルメタン-1,1-ジイル基を表し、その結果、1,3-ジオキサン環を形成したものである、請求項2記載の化合物。
- R4が非放射性の臭素原子、ヨウ素原子、又はp-トルエンスルホニルオキシ基である、請求項2又は3に記載の化合物。
- 請求項1の記載の化合物又はその塩を含む、放射性医薬組成物。
- 低酸素領域を画像化するために用いられる、請求項5記載の放射性医薬組成物。
- 腫瘍を画像化するために用いられる、請求項5又は6記載の放射性医薬組成物。
- 請求項2記載の式(2)で表される化合物又はその塩から、請求項1記載の式(1)で表される化合物又はその塩を製造する方法。
- 請求項2記載の式(2)で表される化合物又はその塩から、請求項1記載の式(1)で表される化合物又はその塩を製造するための装置。
