放射性ハロゲン標識化合物の前駆体として、下記の一般式(I)〔式中、R1はハロゲン化物イオンにより求核置換可能な脱離基を表し、R2はアミノ基により求核アシル置換反応が進行する脱離基を表し、R3は水素原子又はハロゲン原子を表す。〕 で表される化合物を提供する。
Description
本発明は、放射性ハロゲン標識化合物、及びその前駆体、並びに放射性ハロゲン標識化合物の製造方法に関する。
放射性ハロゲン標識化合物は、PETやSPECTのような診断法、がんの治療など医療分野において幅広く用いられている。放射性ハロゲン標識化合物が診断、治療効果を発揮するためには、この化合物が生体内で安定に存在できることが必要である。本発明者らは、ネオペンチル構造が2つのヒドロキシ基と立体障害の大きい4級炭素を有することから、この構造を持つ放射性ハロゲン標識化合物が、生体内で安定に存在できることを見出した(特許文献1及び2)。
医療に用いられる放射性ハロゲンは半減期が短いことから、前駆体から短時間で得られる必要があり、このため、前駆体から放射性ハロゲン標識化合物への転化率が重要である。本発明は、このような背景の下、前駆体から高い転化率で得られる放射性ハロゲン標識化合物を提供することを目的とする。
本発明者は、上記課題を解決するため鋭意検討を重ねた結果、ネオペンチル構造を有する放射性ハロゲン標識化合物の前駆体にトリアゾール基を導入することにより、転化率が著しく向上することを見出した。また、このトリアゾール基はアジドとアルキンのクリック反応により容易に導入できることも見出した。
本発明は以上の知見に基づき完成されたものである。
即ち、本発明は、以下の(1)~(12)を提供するものである。
(1)下記の一般式(I)
〔式中、R1はハロゲン化物イオンにより求核置換可能な脱離基を表し、R2はアミノ基により求核アシル置換反応が進行する脱離基を表し、R3は水素原子又はハロゲン原子を表す。〕

本明細書は、本願の優先権の基礎である日本国特許出願、特願2022-114844の明細書及び/又は図面に記載される内容を包含する。
本発明は、新規な放射性ハロゲン標識化合物を提供する。この化合物は、前駆体から高い転化率で得ることができ、また、前駆体はクリック反応を利用し、容易に合成することができる。
以下、本発明を詳細に説明する。
本発明において「ハロゲン原子」とは、例えば、フッ素原子、塩素原子、臭素原子、ヨウ素原子、アスタチン原子などである。
本発明において「ハロゲン原子」とは、例えば、フッ素原子、塩素原子、臭素原子、ヨウ素原子、アスタチン原子などである。
本発明において「放射性ハロゲン原子」とは、例えば、18F、75Br、76Br、77Br、82Br、123I、124I、125I、131I、133I、209At、210At、211Atなどである。
本発明において「放射性ハロゲン化物イオン」とは、例えば、18F-、75Br-、76Br-、77Br-、82Br-、123I-、124I-、125I-、131I-、133I-、209At-、210At-、211At-などである。
本発明において「炭素数5~20のアルキル基」とは、炭素数が5以上20以下の直鎖又は分枝鎖アルキル基であり、例えば、ペンチル基、ヘキシル基、ヘプチル基、オクチル基、ノニル基、デシル基、ウンデシル基、ドデシル基、トリデシル基、テトラデシル基、ペンタデシル基、ヘキサデシル基、ヘプタデシル基、オクタデシル基、ノナデシル基、イコシル基などである。
本発明において「炭素数7~11のアルキル基」とは、炭素数が7以上11以下の直鎖又は分枝鎖アルキル基であり、例えば、ヘプチル基、オクチル基、ノニル基、デシル基、ウンデシル基などである。
本発明において「炭素数4~24のアルキル基」とは、炭素数が4以上24以下の直鎖又は分枝鎖アルキル基であり、例えば、ブチル基、ペンチル基、ヘキシル基、ヘプチル基、オクチル基、ノニル基、デシル基、ウンデシル基、ドデシル基、トリデシル基、テトラデシル基、ペンタデシル基、ヘキサデシル基、ヘプタデシル基、オクタデシル基、ノナデシル基、イコシル基、ヘンエイコシル基、ドコシル基、トリコシル基、テトラコシル基などである。
本発明において「炭素数1~24のアルキル基」とは、炭素数が1以上24以下の直鎖又は分枝鎖アルキル基であり、例えば、メチル基、エチル基、プロピル基、ブチル基、ペンチル基、ヘキシル基、ヘプチル基、オクチル基、ノニル基、デシル基、ウンデシル基、ドデシル基、トリデシル基、テトラデシル基、ペンタデシル基、ヘキサデシル基、ヘプタデシル基、オクタデシル基、ノナデシル基、イコシル基、ヘンエイコシル基、ドコシル基、トリコシル基、テトラコシル基などである。
本発明において「炭素数1~4のアルキル基」とは、炭素数が1以上4以下の直鎖又は分枝鎖アルキル基であり、例えば、メチル基、エチル基、プロピル基、iso-プロピル基、ブチル基、iso-ブチル基、sec-ブチル基、tert-ブチル基などである。
本発明において「炭素数1~4のアルコキシ基」とは、炭素数が1以上4以下の直鎖又は分枝鎖アルコキシ基であり、例えば、メトキシ基、エトキシ基、プロポキシ基、iso-プロポキシ基、ブトキシ基、iso-ブトキシ基、sec-ブトキシ基、tert-ブトキシ基などである。
本発明において「アリール基」とは、例えば、フェニル基、ナフタレン-1-イル基、ナフタレン-2-イル基などである。
本発明において「アリールオキシ基」とは、例えば、フェノキシ基、ナフタレン-1-イルオキシ基、ナフタレン-2-イルオキシ基などである。
一般式(I)においてR1は、ハロゲン化物イオンにより求核置換可能な脱離基を表す。このような脱離基は、放射性ハロゲン標識化合物の製造においてよく使用されるものであり(例えば、特開2017-52713、国際公開第2018/164043、国際公開第2019/151384に記載されている脱離基)、当業者に周知である。ハロゲン化物イオンにより求核置換可能な脱離基の具体例としては、下記の一般式(A)又は(B)で表される基を挙げることができる。
一般式(A)においてR11及びR12はそれぞれ独立して、炭素数5~20のアルキル基を表す。R11及びR12は異なる基であってもよいが、同一の基であることが好ましい。R11及びR12は前記した基であればよいが、好ましくは、炭素数7~11のアルキル基であり、より好ましくは、炭素数7又は11のアルキル基である。
一般式(A)においてX1及びX2はそれぞれ独立して、ハロゲン原子を表す。X1及びX2は異なる原子であってもよいが、同一の原子であることが好ましい。X1及びX2はハロゲン原子であればよいが、好ましくは、フッ素原子又は塩素原子であり、より好ましくは、フッ素原子である。
一般式(B)においてR15は、炭素数4~24のアルキル基、又は、-CONR18R19を表す。「炭素数4~24のアルキル基」は、好ましくは、直鎖のアルキル基であり、より好ましくは、炭素数8~16の直鎖アルキル基である。「-CONR18R19」において、R18及びR19は、それぞれ独立して、炭素数1~24のアルキル基、又は、置換基で置換されていてもよいアリール基を表す。「炭素数1~24のアルキル基」は、好ましくは、直鎖のアルキル基であり、より好ましくは、炭素数8~16の直鎖アルキル基である。「置換基で置換されていてもよいアリール基」における「アリール基」は、フェニル基のような単環アリール基でもよいが、ナフチル基のような縮合多環アリール基が好ましい。「置換基で置換されていてもよいアリール基」における「置換基」としては、炭素数1~4のアルキル基、炭素数1~4のアルコキシ基、ハロゲン原子などを挙げることができる。R18及びR19は異なる基であってもよいが、同一の基であることが好ましい。
一般式(B)においてR13、R14、R16、R17、及びR18は、それぞれ独立して、水素原子、炭素数1~4のアルキル基、又は炭素数1~4のアルコキシ基を表す。R13、R14、R16、R17、及びR18は、すべてが同一の基であってもよく、一部だけが同一の基であってもよく、全てが異なる基であってもよい。R13、R14、R16、R17、及びR18は前記した基であればよいが、好ましくは、水素原子である。
一般式(I)、(IIa)、及び(IIb)においてR2はアミノ基により求核アシル置換反応が進行する脱離基を表す。このような脱離基の具体例として、下記の一般式(C)、(D)、(E)、又は(F)で表される基を挙げることができる。
一般式(C)においてR21、R22、R23、R24、及びR25は、それぞれ独立して水素原子又はハロゲン原子を表す。R21、R22、R23、R24、及びR25は異なる原子であってもよく、同一の原子であってもよい。R23は前記した原子であればよいが、好ましくは、水素原子又はフッ素原子である。R21、R22、R24、及びR25は前記した原子であればよいが、好ましくは、フッ素原子である。
一般式(D)においてR26及びR27は、それぞれ独立して水素原子、炭素数1~4のアルキル基、又は置換基で置換されていてもよいアリール基を表す。「置換基で置換されていてもよいアリール基」における「置換基」としては、炭素数1~4のアルキル基、炭素数1~4のアルコキシ基、ハロゲン原子などを挙げることができる。R26及びR27は異なる基であってもよく、同一の基であってもよい。また、R26及びR27は互いに結合して環を形成してもよい。環は5員環であっても、6員環であっても、7員環であっても、8員環であってもよい。
一般式(E)においてR28及びR29は、それぞれ独立して水素原子、炭素数1~4のアルキル基、炭素数1~4のアルコキシ基、アミノ基[但し、アミノ基の1又は2個の水素原子は炭素数1~4のアルキル基に置換されていてもよい。]、又は置換基で置換されていてもよいアリール基を表す。「置換基で置換されていてもよいアリール基」における「置換基」としては、炭素数1~4のアルキル基、炭素数1~4のアルコキシ基、ハロゲン原子などを挙げることができる。R28及びR29は異なる基であってもよく、同一の基であってもよい。また、R28及びR29は互いに結合して環を形成してもよい。環は5員環であっても、6員環であっても、7員環であっても、8員環であってもよい。
一般式(F)においてR210及びR211は、それぞれ独立して水素原子、炭素数1~4のアルキル基、又は置換基で置換されていてもよいアリール基を表す。「置換基で置換されていてもよいアリール基」における「置換基」としては、炭素数1~4のアルキル基、炭素数1~4のアルコキシ基、ハロゲン原子などを挙げることができる。R210及びR211は異なる基であってもよく、同一の基であってもよい。また、R210及びR211は互いに結合して環を形成してもよい。環は4員環であっても、5員環であっても、6員環であっても、7員環であっても、8員環であってもよい。
一般式(I)、(IIa)、(IIb)、(IIIa)、及び(IIIb)においてR3は水素原子又はハロゲン原子を表す。R3は水素原子又はハロゲン原子であればよいが、好ましくは水素原子である。
一般式 (IIa)、(IIb)、(IIIa)、及び(IIIb) においてXは放射性ハロゲン原子を表す。ハロゲン原子の種類は特に限定されないが、臭素原子、ヨウ素原子、アスタチン原子であることが好ましい。Xは放射性ハロゲン原子であればよいが、好ましくは、18F、123I、131I、又は211Atであり、より好ましくは、123I、131I、又は211Atである。
一般式(IIIa)及び(IIIb)においてR4は生理活性物質から誘導される一価の基を表す。ここで、「生理活性物質」とは、生理機能の維持や調節に関わる物質を意味し、例えば、アミノ酸、ペプチド、タンパク質(抗体を含む)、糖などが含まれる。また、「生理活性物質から誘導される一価の基」とは、例えば、生理活性物質の分子中の一つの水素原子を除去することによって得られる一価の基を意味する。除去される水素原子は、例えば、生理活性物質がヒドロキシ基やアミノ基を含む場合、これらの基に含まれる水素原子である。一般式(IIIa)又は(IIIb)で表される化合物は疾患の診断や治療に用いられるので、この化合物を疾患部位に送り届けるため、生理活性物質は疾患部位に集積する性質を持つものであることが好ましい。疾患としては、がん、アルツハイマー病などを挙げることができる。疾患ががんである場合、生理活性物質はがん細胞と特異的に結合する物質とすることができ、疾患がアルツハイマー病である場合、生理活性物質はアミロイドβやタウタンパク質と親和性を示す物質とすることができる。
一般式(I)で表される化合物は、放射性ハロゲン標識化合物の標識前駆体試薬として使用することができる。放射性ハロゲン標識化合物の標識前駆体試薬は、通常、一般式(I)で表される化合物のみからなるが、他の物質を含んでいてもよい。一般式(I)で表される化合物は、分子内にアルキンを含む化合物とアジド化合物を反応させることにより製造することができる。より具体的には、実施例1に記載した製造方法に、必要に応じて、改変や修正を加えた方法に従って製造できる。
一般式(I)で表される化合物は、放射性ハロゲン標識化合物の前駆体化合物として以下のような利点を有する。
1)従来の前駆体化合物よりもハロゲン化転化率が高いので、短時間で放射性ハロゲン標識化合物を製造することができる。
2)分子内にアルキンを含む化合物とアジド化合物とのクリック反応により合成できるので、両化合物の構造を壊すことなく合成できる。

一般式(IIa)又は(IIb)で表される化合物は、放射性ハロゲン標識試薬として使用することができる。放射性ハロゲン標識試薬は、通常、一般式(IIa)又は(IIb)で表される化合物のみからなるが、他の物質を含んでいてもよい。一般式(IIa)又は(IIb)で表される化合物は、一般式(I)で表される化合物を放射性ハロゲン化物イオンと反応させることにより製造できる。使用する放射性ハロゲン化物イオンは特に限定されないが、好ましくは、18F、123I、131I、又は211Atであり、より好ましくは、123I、131I、又は211Atである。反応条件は、一般式(I)で表される化合物や放射性ハロゲン化物イオンの種類によって適宜設定することができる。当業者であれば、適切な反応温度、反応時間、各物質の濃度などを容易に決定することができる。
一般式(IIIa)又は(IIIb)で表される化合物は、疾患の治療薬又は診断薬として使用することができる。疾患の治療薬又は診断薬は、一般式(IIIa)又は(IIIb)で表される化合物以外の物質を含んでいてもよい。
一般式(IIIa)又は(IIIb)で表される化合物は、一般式(IIa)又は(IIb)で表される化合物を生理活性物質と反応させることにより製造できる。生理活性物質は特に限定されないが、好ましくは、疾患部位に集積する性質を持つ生理活性物質である。反応条件は、一般式(IIa)又は(IIb)で表される化合物や生理活性物質の種類によって適宜設定することができる。当業者であれば、適切な反応温度、反応時間、各物質の濃度などを容易に決定することができる。
以下に、実施例により本発明を更に詳細に説明するが、本発明はこれらの実施例に限定されるものではない。

NMRスペクトルは、JEOL Model ECP-400 (400 MHz for 1H)と Bruker AVANCE III HD 400 (400 MHz for 1H) を用いて、指定された溶媒で記録された。化学シフトは、CDCl3中の溶液の内部テトラメチルシラン(1Hは0ppm)の信号に対する百万分の一(ppm)の単位で報告される。1H NMRスペクトルのデータは以下のように報告される。CDCl3 (7.26 ppm), CD3OD-d4 (3.31 ppm) を外部標準とし、多重度は、次の略語を使用して報告される:s, singlet; br-s, broaded-singlet; d, doublet; br-d, broaded-doublet; dd, doublet of doublets; br-dd, broaded-doublet of doublets; t, triplet; dq, doublet of quartets; q, quartet, m, multiplet; J, coupling constants in Hertz.

すべての反応は、0.2 mm E. Merck シリカゲルプレート (60F-254) を用いて、p-アニスアルデヒド溶液、硫酸セリック又はエタノール型リンモリブデン酸で可視化した薄層クロマトグラフィーにより、UV光を照射してモニターした。カラムクロマトグラフィーによる分離は、シリカゲル(Merck silica gel 60, 0.063 - 0.200 mm)を用いて行った。
(5-(アジドメチル)-2,2-ジメチル-1,3-ジオキサン-5-イル)メタノール (6)の合成
9-(4-メトキシフェニル)-3,3-ジメチル-2,4,8,10-テトラオキサスピロ[5.5]ウンデカン(2)
乾燥 DMF (8 mL)中の(2-(4-メトキシフェニル)-1,3-ジオキサン-5,5-ジイル)ジメタノール (1) (1.00 g, 3.93 mmol, 1.0 eq.)及び(+)-10-カンファースルホン酸(45.7 mg, 197 μmol, 0.05 eq.)の撹拌溶液に、室温、Ar 雰囲気で 2,2-ジメトキシプロパン (1.4 mL, 11.8 mmol, 3.0 eq.)を添加した。室温で2時間撹拌した後、反応混合物をNaHCO3 aq.及びEtOAcに注いだ。水層をEtOAcで2回抽出した。合わせた抽出液をブラインで洗浄し、MgSO4上で乾燥させ、濾過し、減圧下で濃縮した。残渣をヘキサン/EtOAcから再結晶して精製し、9-(4-メトキシフェニル)-3,3-ジメチル-2,4,8,10-テトラオキサスピロ[5.5]ウンデカン(2)(659 mg, 2.24 mmol, 57%)を白色の結晶として得た。

2,3,5,6-テトラフルオロフェニルプロピオナート(9)

MeCN(1 mL)中の2,3,5,6-テトラフルオロフェニル 1-((5-((((2-(ジオクチルアミノ)-1,1-ジフルオロ-2-オキソエチル)スルホニル)オキシ)メチル)-2,2-ジメチル-1,3-ジオキサン-5-イル)メチル)-1H-1,2,3-トリアゾール-4-カルボキシレート (10)の撹拌溶液に、室温で、ヨウ化テトラブチルアンモニウム(120 mg, 325 μmol, 2.0 eq.)を添加した。室温で1時間撹拌した後、反応混合物をNH4Cl aq.及びEtOAcに注いだ。 水層をEtOAcで2回抽出した。合わせた抽出物をブラインで洗浄し、MgSO4上で乾燥させ、濾過し、減圧下で濃縮した。残渣をシリカゲル上のヘキサン/EtOAc(9:1)カラムクロマトグラフィーで精製し、2,3,5,6-テトラフルオロフェニル 1-((5-(ヨードメチル)-2,2-ジメチル-1,3-ジオキサン-5-イル)メチル)-1H-1,2,3-トリアゾール-4-カルボキシレート(11)(60.0 g, 113 μmol, 70%)を得た。
tert-ブチル(6-(1-((5-(ヨードメチル)-2,2-ジメチル-1,3-ジオキサン-5-イル)メチル)-1H-1,2,3-トリアゾール-4-カルボキサミド)-1-((4-メトキシフェニル)アミノ)-1-オキソヘキサン-2-イル)カルバメート (13)
EtOH (100 μL)中の2,3,5,6-テトラフルオロフェニル 1-((5-(ヨードメチル)-2,2-ジメチル-1,3-ジオキサン-5-イル)メチル)-1H-1,2,3-トリアゾール-4-カルボキシレート(11)(30.0 mg、56.7 μmol, 1.0 eq. ) の撹拌溶液に、室温で、トリエチルアミン (31.6 μL, 0.726 g/mL, 227 μmol, 4 eq.) 及びtert-ブチル(6-アミノ-1-((4-メトキシフェニル)アミノ)-1-オキソヘキサン-2-イル)カルバメート(12)(39.8 mg, 113 μmol, 2.0 eq.)を添加した。室温で10分間撹拌した後、反応混合物をNH4Cl aq.及びEtOAcに注いだ。 水層をEtOAcで2回抽出した。合わせた抽出液をブラインで洗浄し、MgSO4上で乾燥させ、濾過し、減圧下で濃縮した。残渣をCH3Cl/MeOH(9:1)を用いたシリカゲル上のカラムクロマトグラフィーにより精製し、tert-ブチル(6-(1-((5-(ヨードメチル)-2,2-ジメチル-1,3-ジオキサン-5-イル)メチル)-1H-1,2,3-トリアゾール-4-カルボキサミド)-1-((4-メトキシフェニル)アミノ)-1-オキソヘキサン-2-イル)カルバメート (13)(20. 3 mg, 28.4 μmol, 50%)を得た。

2,3,4,5,6-ペンタフルオロフェニル 1-((5-((((2-(ジオクチルアミノ)-1,1-ジフルオロ-2-オキソエチル)スルホニル)オキシ)メチル)-2,2-ジメチル-1,3-ジオキサン-5-イル)メチル)-1H-1,2,3-トリアゾール-4-カルボキシレート(14)
THF(400μL)中の(5-(アジドメチル)-2,2-ジメチル-1,3-ジオキサン-5-イル)メチル 2-(ジオクチルアミノ)-1,1-ジフルオロ-2-オキソエタン-1-スルホン酸 (27.5 mg, 47.2 μmol, 1.0 eq.)及びパーフルオロフェニルプロピオレート (16. 7 mg, 70.8 μmol, 1.5 eq.)の溶液に、水(100 μL)中の硫酸銅(II)五水和物(5.89 mg, 23.6 μmol, 0.5 eq.)及びL-アスコルビン酸ナトリウム(4.76 mg, 17.2 μmol, 0.5 eq.)を添加した。室温で24時間撹拌した後、反応混合物をNaHCO3 aq.及びEtOAcに注いだ。水層をEtOAcで2回抽出した。合わせた抽出物をブラインで洗浄し、MgSO4上で乾燥させ、濾過し、そして真空中で濃縮した。残渣をシリカゲル上のヘキサン/EtOAc(9:1)カラムクロマトグラフィーにより精製し、2,3,4,5,6-ペンタフルオロフェニル 1-((5-((((2-(ジオクチルアミノ)-1,1-ジフルオロ-2-オキソエチル)スルホニル)オキシ)メチル)-2,2-ジメチル-1,3-ジオキサン-5-イル)メチル)-1H-1,2,3-トリアゾール-4-カルボキシレート(14)(10. 4 mg, 12.7 μmol, 27%)を無色オイルとして得た。
THF(400μL)中の(5-(アジドメチル)-2,2-ジメチル-1,3-ジオキサン-5-イル)メチル 2-(ジオクチルアミノ)-1,1-ジフルオロ-2-オキソエタン-1-スルホン酸 (27.5 mg, 47.2 μmol, 1.0 eq.)及びパーフルオロフェニルプロピオレート (16. 7 mg, 70.8 μmol, 1.5 eq.)の溶液に、水(100 μL)中の硫酸銅(II)五水和物(5.89 mg, 23.6 μmol, 0.5 eq.)及びL-アスコルビン酸ナトリウム(4.76 mg, 17.2 μmol, 0.5 eq.)を添加した。室温で24時間撹拌した後、反応混合物をNaHCO3 aq.及びEtOAcに注いだ。水層をEtOAcで2回抽出した。合わせた抽出物をブラインで洗浄し、MgSO4上で乾燥させ、濾過し、そして真空中で濃縮した。残渣をシリカゲル上のヘキサン/EtOAc(9:1)カラムクロマトグラフィーにより精製し、2,3,4,5,6-ペンタフルオロフェニル 1-((5-((((2-(ジオクチルアミノ)-1,1-ジフルオロ-2-オキソエチル)スルホニル)オキシ)メチル)-2,2-ジメチル-1,3-ジオキサン-5-イル)メチル)-1H-1,2,3-トリアゾール-4-カルボキシレート(14)(10. 4 mg, 12.7 μmol, 27%)を無色オイルとして得た。
〔実施例2〕 放射性ハロゲン標識化合物の合成
化合物(3)[ 125 I] 2,3,5,6-テトラフルオロフェニル 1-(3-ヒドロキシ-2-(ヒドロキシメチル)-2-(ヨードメチル)プロピル)-1H-1,2,3-トリアゾール-4-カルボキシレートの作製
化合物(1) (0.4 mg, 5.0×10-7mol)を1%DIEA/MeCN溶液(50 μl)に溶解させた。溶液に[125I]NaI水溶液(0.5 μl, 61.6 μCi )を加え、50℃で1時間反応させ化合物(2)を作製した。続けて化合物(2)を含む溶液をWatars社製C18-ODS SEP-PAK(50 mg)に通し、水(1000 μl)で洗浄した。次いで、カラムに6M塩酸(2000 μl)を流し、カラム内に6M塩酸を満たしたまま、室温で10分間静置した。水(1000μl)で洗浄した後、MeCN(150 μl)により抽出し放射化学的収率76.5%で化合物(3)を得た(2 step)。HPLCにてImtakt Unison US-C18 150×4.6 mmを用い、移動相にはA相にMilliQ、B相にMeCNを使用し、0-30分でA相95%、B相5%からA相0%、B相100%まで変化する分析系により、放射化学的純度>99 %であることを確認した(図1)。
化合物(3)[ 125 I] 2,3,5,6-テトラフルオロフェニル 1-(3-ヒドロキシ-2-(ヒドロキシメチル)-2-(ヨードメチル)プロピル)-1H-1,2,3-トリアゾール-4-カルボキシレートの作製
化合物(4) [ 125 I] tert-ブチル(6-(1-((5-(ヨードメチル)-2,2-ジメチル-1,3-ジオキサン-5-イル)メチル)-1H-1,2,3-トリアゾール-4-カルボキサミド)-1-((4-メトキシフェニル)アミノ)-1-オキソヘキサン-2-イル)カルバメートの作製
化合物(1)(0.4 mg, 5.0×10-7mol)を1%DIEA/MeCN溶液(50 μl)に溶解させた。溶液に[125I]NaI水溶液(0.5 μl, 59 μCi )を加え、50℃で1.5時間反応した。溶液をWaters社製 Oasis HLB 1 cc Vac Cartridge(10 mg)に通し、水(500 μl)で洗浄した。続いてMeCNを流し、30~60 μlを流した画分に化合物(2)を回収した。この溶液にtert-ブチル(6-アミノ-1-((4-メトキシフェニル)アミノ)-1-オキソヘキサン-2-イル)カルバメートのエタノール溶液(2.0 mg/ml, 30 μl)を加え、37℃で1時間反応し放射化学的収率64.8%で化合物(4)を得た(2 step)。HPLCにてImtakt Unison US-C18 150×4.6 mmを用い、移動相にはA相にMilliQ、B相にMeCNを使用し、0-30分でA相95%、B相5%からA相0%、B相100%まで変化する分析系により、放射化学的純度>99 %であることを確認した(図2)。
〔実施例3〕 トリアゾール基を有する放射性ヨウ素化合物の収率
トリアゾール基を有する放射性ヨウ素化合物(化合物A)と他の化合物(化合物B及びC)の収率を比較した。
トリアゾール基を有する放射性ヨウ素化合物(化合物A)と他の化合物(化合物B及びC)の収率を比較した。
各化合物の前駆体(0.3 mg, 5.0×10-7mol)を1%DIEA/MeCN溶液(50 μl)に溶解させた。溶液に[125I]NaI水溶液(0.5 μl, 3 μCi )を加え、50℃で40分反応させた。反応溶液を順相TLC(トルエン:酢酸エチル=4:1(化合物B、C)あるいはトルエン:酢酸エチル-7:3(化合物A)にて分析した。
化合物B及びCの収率はそれぞれ49.2%及び23.8%であった。これに対し、トリアゾール基を有する化合物Aの収率は84.4%であり、化合物B及びCに比べ著しく高かった。
〔実施例4〕 ヨウ素標識抗体の作製
125Iヨウ素標識化合物の合成
化合物(1)(0.4 mg)を1%DIEA/MeCN(50 μl)に溶解させた。さらに、[125I]NaI aq.(0.5μl, 36.8 μCi)を加え、50度、3時間反応させた。メタノールで活性化させたWatars社製C18-ODS SEP-PAK(50mg)に反応溶液を全量加えた。続いて、SEP-PAKを水(1000μl)で洗浄した。さらに、6M塩酸(2000 μl)をSEP-PAKに加え、カラム内に6M塩酸を満たしたまま、室温で10分間静置した。MeCN:H2O, 1:1(500 μl)を用いて、カラム内より溶出させ、化合物(3)(26.0μCi)を得た(放射化学的収率70%)。
125Iヨウ素標識化合物の合成
標識抗体の作製
化合物(3)の溶液を窒素ガスにて留去した。本容器にEGFR抗体溶液( 2 mg/mL, 200μL, 0.16 M ホウ酸緩衝液pH 8-9 )を加えた。TLCにより反応の進行を確認した。60分後、スピンカラム(Sephadex G50, 0.1 M リン酸 pH 7.4)を用いて精製(1500 rpm, 2 min)することで、放射性標識抗体(8.85μCi)を得た(放射化学的収率 33%)。
〔実施例5〕 211Atアスタチン標識化合物の合成
化合物(1)(0.4 mg)を1%DIEA/MeCN(50 μl) + 10%アスコルビン酸Na aq.(0.5 μL)に溶解させた。さらに、[211At]At- in MeCN.(5 μl, 0.455 MBq)を加え、50度、1時間反応させた。メタノールで活性化させたWatars社製C18-ODS SEP-PAK(50mg)に反応溶液を全量加えた。続いて、SEP-PAKを水(1000μl)で洗浄した。さらに、6M塩酸(2000 μl)をSEP-PAKに加え、カラム内に6M塩酸を満たしたまま、室温で10分間静置した。MeCN:H2O, 1:1(500 μl)を用いて、カラム内より溶出させ、化合物(3)を0.244 MBqで得た(放射化学的収率53%)。

放射性ハロゲン標識化合物は、医薬として利用できるので、本発明は、医薬に関連する産業分野において利用可能である。

で表される基である請求項1に記載の化合物。 - 一般式(I)におけるR2が、下記の一般式(C)、(D)、(E)、又は(F)
で表される基である請求項1に記載の化合物。 - 下記の一般式(IIa)又は(IIb)
で表される化合物。 - 一般式(IIa)又は(IIb)におけるXが、放射性臭素原子、放射性ヨウ素原子、又は放射性アスタチン原子である請求項4に記載の化合物。
- 一般式(IIa)又は(IIb)におけるR2が、下記の一般式(C)、(D)、(E)、又は(F)
で表される基である請求項4に記載の化合物。 - 下記の一般式(IIIa)又は(IIIb)
で表される化合物。 - 一般式(IIIa)又は(IIIb)におけるXが、放射性臭素原子、放射性ヨウ素原子、又は放射性アスタチン原子である請求項7に記載の化合物。
- 一般式(IIIa)又は(IIIb)におけるR4が、疾患部位に集積する生理活性物質から誘導される一価の基である請求項7に記載の化合物。
- 一般式(IIIa)又は(IIIb)におけるR4が、抗体から誘導される一価の基である請求項7に記載の化合物。
- 請求項4乃至6のいずれか一項に記載の化合物の製造方法であって、請求項1乃至3のいずれか一項に記載の化合物を放射性ハロゲン化物イオンと反応させる工程を含む方法。
- 請求項7乃至10のいずれか一項に記載の化合物の製造法であって、請求項4乃至6のいずれか一項に記載の化合物を生理活性物質と反応させる工程を含む方法。
