Description
ヒドロキシイミノアルケニルシクロプロパンカルボン酸エステルの製造方法としては、例えば、エタノールおよびピリジン中、2,2−ジメチル−3−(2−ホルミル−1−プロペニル)シクロプロパンカルボン酸メチルとヒドロキシルアミン塩酸塩とを加熱還流することにより反応させる方法が知られている(例えば、特公昭38−2272号公報実施例参照。)。
特公昭38−2272号公報には、シアノアルケニルシクロプロパン化合物の製造方法として、2,2−ジメチル−3−[3−(ヒドロキシイミノ)−2−メチル−1−プロペニル]シクロプロパンカルボン酸メチルと無水酢酸とを混合して、得られる混合物を加熱煮沸する方法も記載されている。
すなわち本発明は、以下の通りである。

二重結合に基づく幾何異性体について、ホルミル化合物(1)は、E体であってもよいし、Z体であってもよいし、E体とZ体とのあらゆる比率の混合物であってもよい。
また、シクロプロパン環に基づく幾何異性体について、ホルミル化合物(1)は、シス体であってもよいし、トランス体であってもよいし、シス体とトランス体とのあらゆる比率の混合物であってもよい。
ホルミル化合物(1)において、シクロプロパン環を形成する炭素原子のうち、2個の炭素原子は不斉炭素原子であり、ホルミル化合物(1)は、光学活性を示す化合物であってもよいし、光学活性を示さない化合物であってもよい。
ホルミル化合物(1)において、R1は、置換基を有していてもよいアルキル基またはハロゲン原子を表す。
ハロゲン原子としては、フッ素原子、塩素原子、臭素原子およびヨウ素原子等が挙げられ、ハロゲン原子は塩素原子または臭素原子であることが好ましい。
R1で表わされるアルキル基としては、炭素数1~10程度の直鎖状アルキル基、炭素数3~10程度の分枝状アルキル基、炭素数3~10程度のシクロアルキル基等が挙げられる。
炭素数1~10程度の直鎖状アルキル基としては、メチル基、エチル基、n−プロピル基等が挙げられ、炭素数3~10程度の分枝状アルキル基としては、イソプロピル基、イソブチル基、tert−ブチル基、イソアミル基、2−エチルヘキシル基等が挙げられ、炭素数3~10程度のシクロアルキル基としては、シクロプロピル基、シクロペンチル基、シクロヘキシル基等が挙げられる。
R1で表わされるアルキル基における置換基としては、フッ素原子、塩素原子、臭素原子等のハロゲン原子;メトキシ基、エトキシ基等の炭素数1~4程度のアルコキシ基;等が挙げられる。置換基を有するアルキル基としては、フルオロメチル基、クロロメチル基、トリフルオロメチル基、トリクロロメチル基等が挙げられる。
R1としては、好ましくは、炭素数1~4の直鎖状アルキル基、塩素原子、臭素原子等が挙げられ、より好ましくは、メチル基、塩素原子等が挙げられる。
ホルミル化合物(1)において、R2は、置換基を有していてもよいアルキル基または置換基を有していてもよいベンジル基を表す。
R2で表わされるアルキル基としては、上述したR1におけるアルキル基と同様のものが挙げられる。
R2で表わされるアルキル基における置換基としては、フッ素原子、塩素原子、臭素原子等のハロゲン原子;アリル基等の炭素数3~4程度のアルケニル基;プロパルギル基等の炭素数3~4程度のアルキニル基;メトキシ基、エトキシ基等の炭素数1~4程度のアルコキシ基;メチルチオ基等の炭素数1~4程度のアルキルチオ基;等が挙げられる。
上記ベンジル基における置換基としては、例えば、フッ素原子、塩素原子、臭素原子等のハロゲン原子;アリル基等の炭素数3~4程度のアルケニル基;プロパルギル基等の炭素数3~4程度のアルキニル基;メトキシ基、エトキシ基等の炭素数1~4程度のアルコキシ基;メチルチオ基等の炭素数1~4程度のアルキルチオ基等;メチル基、エチル基、n−プロピル基等の炭素数1~10程度の直鎖状アルキル基;イソプロピル基、イソブチル基、tert−ブチル基、イソアミル基、2−エチルヘキシル基等の炭素数3~10程度の分枝状アルキル基;シクロプロピル基、シクロペンチル基、シクロヘキシル基等の炭素数3~10程度のシクロアルキル基;メトキシメチル基、エトキシメチル基などの炭素数2~4程度のアルコキシメチル基;メチルチオメチル基等の炭素数1~4程度のアルキルチオメチル基等が挙げられる。
R2としては、好ましくは、炭素数1~4の直鎖状アルキル基、置換基を有していてもよいベンジル基等が挙げられ、より好ましくは、炭素数1~4の直鎖状アルキル基;ベンジル基;および、ハロゲン原子、炭素数3~4のアルケニル基、炭素数1~4の直鎖状アルキル基、炭素数2~4のアルコキシメチル基および炭素数1~4のアルキルチオメチルからなる群より選択される基で置換されたベンジル基が挙げられ、更に好ましくはメチル基、エチル基、ベンジル基、4−メチル−2,3,5,6−テトラフルオロベンジル基、4−メトキシメチル−2,3,5,6−テトラフルオロベンジル基、4−アリル−2,3,5,6−テトラフルオロベンジル基、4−プロパルギル−2,3,5,6−テトラフルオロベンジル基、4−メチルチオメチル−2,3,5,6−テトラフルオロベンジル基等が挙げられる。
ホルミル化合物(1)としては、具体的には、表1、2および3に記載される化合物が例示される。
ホルミル化合物(1)は、例えば日本国特許公開2008−44900号公報に記載される方法により製造することができる。具体的には、式(4)

で示される化合物とを塩基の存在下で縮合させる方法等が挙げられる。
次いで、オキシム化工程を説明する。
オキシム化工程において、カルボン酸およびカルボン酸の金属塩からなる群より選択される少なくとも1種の化合物の存在下で、例えばカルボン酸またはカルボン酸の金属塩の存在下で、ホルミル化合物(1)とヒドロキシルアミンとを反応させることにより、ホルミル化合物(1)は式(3)で示されるヒドロキシイミノアルケニルシクロプロパンカルボン酸エステル(以下、ヒドロキシイミノ化合物(3)と称することがある。)に変換される。
オキシム化工程に用いられるカルボン酸としては、例えばプロパン酸、ブタン酸、ペンタン酸、ヘキサン酸、ヘプタン酸、オクタン酸、2−エチルヘキサン酸、ノナン酸、デカン酸、ミスチリン酸等の炭素数3以上のカルボン酸が挙げられる。カルボン酸は、ヒドロキシイミノ化合物(3)を高収率で得る点で、ヘキサン酸、ヘプタン酸、オクタン酸、2−エチルヘキサン酸、ノナン酸、デカン酸、ミスチリン酸等の炭素数6以上のカルボン酸が好ましく、炭素数6以上、炭素数14以下のカルボン酸がより好ましい。ホルミル化合物(1)とヒドロキシルアミンとを反応させた後にカルボン酸を除去する場合、または後述するニトリル化工程においてヒドロキシイミノ化合物(3)と脱水剤とを反応させた後にカルボン酸を除去する場合、さらにカルボン酸を容易に除去できる点で、ヘキサン酸、ヘプタン酸、オクタン酸、2−エチルヘキサン酸等の炭素数6~8のカルボン酸がより好ましく、ヘキサン酸、ヘプタン酸、オクタン酸等の炭素数6~8の直鎖状カルボン酸がさらに好ましく、オクタン酸がより一層好ましい。
カルボン酸の金属塩としては、上述した炭素数3以上のカルボン酸のカリウム塩、ナトリウム塩、リチウム塩等のアルカリ金属塩が挙げられる。かかるカルボン酸の金属塩は、上述したカルボン酸と、水酸化カリウム、水酸化ナトリウム、水酸化リチウム等のアルカリ金属水酸化物、または炭酸カリウム、炭酸ナトリウム、炭酸リチウム等のアルカリ金属炭酸化物とを反応させることにより、オキシム化工程の反応系中で発生させてもよい。
カルボン酸またはカルボン酸の金属塩の使用量としては、合計で、ホルミル化合物(1)1モルに対し、好ましくは0.01~0.5モルの範囲内であり、より好ましくは0.05~0.2モルの範囲内である。
オキシム化工程に用いられるヒドロキシルアミンの使用量は、ホルミル化合物(1)1モルに対して、好ましくは0.8~2モルの範囲内であり、より好ましくは1.0~1.3モルの範囲内である。
ヒドロキシルアミンは、水溶液として用いることが好ましい。ヒドロキシルアミンは、塩酸塩、硫酸塩等の塩を形成していてもよいが、ヒドロキシルアミンが塩を形成している場合には、無機塩基と接触させる等によりヒドロキシルアミンを遊離させて用いることが好ましい。無機塩基としては、例えば、水酸化カリウム、水酸化ナトリウム、水酸化リチウム等のアルカリ金属水酸化物、炭酸カリウム、炭酸ナトリウム、炭酸リチウム等のアルカリ金属炭酸化物が挙げられる。無機塩基は、水溶液として用いることが好ましい。
無機塩基の使用量は、ホルミル化合物(1)1モルに対して、好ましくは0.9~2.5モルの範囲内であり、より好ましくは1.0~1.4モルの範囲内である。
オキシム化工程において、ホルミル化合物(1)とヒドロキシルアミンとの反応は、溶媒中で行われる。かかる溶媒としては、例えば、トルエン、キシレン、エチルベンゼン等の芳香族炭化水素溶媒;n−ペンタン、n−ヘキサン等の鎖状炭化水素溶媒;シクロペンタン、シクロヘキサン等の環状炭化水素溶媒;モノクロロベンゼン等のハロゲン化芳香族炭化水素溶媒;ジエチルエーテル、tert−ブチルメチルエーテル、1,2−ジメトキシエタン、1,4−ジオキサン、テトラヒドロフラン、アニソール等のエーテル溶媒;メタノール、エタノール、イソプロピルアルコール等のアルコール溶媒;水が挙げられ、水と、水と非混和性の有機溶媒(以下、「水と非混和性の有機溶媒」を「非混和性有機溶媒」と称することがある。)との混合溶媒であることが好ましい。
非混和性有機溶媒としては、例えば、トルエン、キシレン、エチルベンゼン等の芳香族炭化水素溶媒;n−ペンタン、n−ヘキサン等の鎖状炭化水素溶媒;シクロペンタン、シクロヘキサン等の環状炭化水素溶媒;モノクロロベンゼン等のハロゲン化芳香族炭化水素溶媒;ジエチルエーテル、tert−ブチルメチルエーテル、1,2−ジメトキシエタン、1,4−ジオキサン、テトラヒドロフラン、アニソール等のエーテル溶媒が挙げられる。
溶媒の使用量は、ホルミル化合物(1)1重量部に対して、好ましくは1~40重量部の範囲内である。溶媒として、水と、非混和性有機溶媒との混合物を用いる場合、ホルミル化合物(1)1重量部に対し、水の使用量は0.5~20重量部の範囲内であることが好ましく、非混和性有機溶媒の使用量は0.5~20重量部の範囲内であることが好ましい。
オキシム化工程における混合順序は制限されず、例えば、溶媒中、ホルミル化合物(1)とカルボン酸またはカルボン酸の金属塩とを混合し、得られる混合物にヒドロキシルアミンを添加する方法、溶媒中、ホルミル化合物(1)とヒドロキシルアミンとを混合し、得られる混合物にカルボン酸またはカルボン酸の金属塩を添加する方法が挙げられる。
ヒドロキシルアミンが塩を形成している場合には、溶媒中、ホルミル化合物(1)とヒドロキシルアミンの塩とカルボン酸またはカルボン酸の金属塩とを混合し、得られる混合物に無機塩基を添加する方法、溶媒中、ホルミル化合物(1)とカルボン酸またはカルボン酸の金属塩とを混合し、得られる混合物にヒドロキシルアミンの塩を添加しながら、無機塩基を添加する方法が好ましい。
ヒドロキシルアミンの塩を添加しながら無機塩基を添加する方法において、ヒドロキシルアミンの塩の添加時間と無機塩基の添加時間とを略同一にすることがより好ましい。ここで、ヒドロキシルアミンの塩の添加時間と無機塩基の添加時間とが略同一であるとは、ヒドロキシルアミンの塩の添加時間に対して、無機塩基の添加時間が、例えば0.8~1.2倍の範囲内であることを意味する。ヒドロキシルアミンの塩の添加時間と無機塩基の添加時間とは、例えば1~30時間の範囲内であり、好ましくは3~10時間の範囲内である。
オキシム化工程において、得られる混合物をpH7.5以下に保持することが好ましい。pH7.5以下に保持する方法としては、例えば添加するヒドロキシルアミンの塩の10~20%を予めホルミル化合物(1)およびカルボン酸と混合し、そこへヒドロキシルアミンの塩を添加しながら、無機塩基を添加する方法が挙げられる。
オキシム化工程は、加熱を必要とせず、常温(20℃程度)で実施しても実用的な収率でヒドロキシイミノ化合物(3)を得ることができる。所望する反応時間に応じて、エネルギー消費量を低く抑制できる範囲内で反応温度を調整することもできる。反応温度は、例えば0~60℃の範囲内であり、好ましくは5~50℃の範囲内であり、より好ましくは10~35℃の範囲内である。
オキシム化工程において、上記添加時間を除く反応時間は、例えば1分~40時間の範囲内であり、好ましくは10分~24時間の範囲内であり、さらに好ましくは30分~10時間の範囲内であり、より一層好ましくは1時間~6時間の範囲内である。
オキシム化工程における反応の進行度合いは、ガスクロマトグラフィー、高速液体クロマトグラフィー等の分析手段により確認することができる。
反応後に得られる反応混合物には、カルボン酸またはカルボン酸の金属塩が含まれている。反応混合物に含まれるカルボン酸またはカルボン酸の金属塩は、反応混合物をアルカリ水溶液等で洗浄することにより、pH10以上の水溶液中に除去することができるが、ヒドロキシイミノ化合物(3)を式(2)で示されるシアノアルケニルシクロプロパンカルボン酸エステル(以下、シアノ化合物(2)と称することがある。)に変換する場合、カルボン酸またはカルボン酸の金属塩は、後述するニトリル化工程においてヒドロキシイミノ化合物(3)と脱水剤とを反応させた後に除去されることが、シアノ化合物(2)の収率を向上させる点で特に好ましい。
反応混合物を、水洗等の後処理に付した後、必要に応じて脱水を行うこともできるし、反応混合物を、中和、抽出、水洗等の後処理に付した後、結晶化、抽出、蒸留、活性炭、シリカ、アルミナ等の吸着、シリカゲルカラムクロマトグラフィー等のクロマト精製等の精製を行うこともできる。
かくして得られるヒドロキシイミノ化合物(3)としては、具体的には、表4、5および6に記載された化合物が例示される。
次いで、ニトリル化工程を説明する。
ニトリル化工程において、ヒドロキシイミノ化合物(3)と脱水剤とを反応させることにより、ヒドロキシイミノ化合物(3)はシアノ化合物(2)に変換される。
本明細書において、脱水剤とは、ヒドロキシイミノ化合物(3)における−CH=NOHの水酸基と水素とを脱離させ、−CH=NOHを−CNに変換させる作用を示す化合物を意味する。
上記脱水剤としては、無水酢酸等のカルボン酸無水物;カルボン酸ハロゲン化物;スルホン酸ハロゲン化物;ハロゲン化チオニル等が挙げられる。
カルボン酸ハロゲン化物(「ハロゲン化アシル」ともいう)としては、塩化アセチル、臭化アセチル等のハロゲン化アセチル; 塩化プロピオニル、臭化プロピオニル等のハロゲン化プロピオニル;および、塩化ブチロイル、臭化ブチロイル等のハロゲン化ブチロイル;等が挙げられる。
スルホン酸ハロゲン化物としては、塩化メタンスルホニル、臭化メタンスルホニル等のハロゲン化メタンスルホニル等が挙げられる。
ハロゲン化チオニルとしては、塩化チオニル、臭化チオニル等が挙げられる。
カルボン酸無水物としては、炭素数2~8のカルボン酸無水物等が挙げられ、具体的には無水酢酸、プロピオン酸無水物およびブタン酸無水物等が挙げられる。カルボン酸無水物は、無水酢酸であることが好ましい。
これらの脱水剤のうち、カルボン酸ハロゲン化物、スルホン酸ハロゲン化物およびハロゲン化チオニルは、ヒドロキシイミノ化合物(3)との反応する際、酸を発生する。
ニトリル化工程において、脱水剤は1種類用いてよいし、2種以上用いてよい。
かかる脱水剤は、好ましくはカルボン酸無水物およびカルボン酸ハロゲン化物からなる群より選ばれる少なくとも1種の化合物である。
かかる脱水剤を2種類以上用いる場合、脱水剤の1種としてカルボン酸無水物を用いることが好ましく、カルボン酸無水物と、カルボン酸ハロゲン化物、スルホン酸ハロゲン化物およびハロゲン化チオニルからなる群より選択される少なくとも1種の化合物とを用いることがより好ましく、カルボン酸無水物と、塩化アセチル、臭化アセチル、塩化チオニルおよび塩化メタンスルホニルからなる群より選択される少なくとも1種の化合物とを用いることが更に好ましい。
脱水剤の使用量は、合計で、ヒドロキシイミノ化合物(3)1モルに対して、好ましくは0.8~3.0モルの範囲内、より好ましくは1.0~2.0モルの範囲内である。
なお、脱水剤としてカルボン酸無水物を用いる場合、カルボン酸無水物の使用量は、ヒドロキシイミノ化合物(3)1モルに対して、好ましくは0.8~2モルの範囲内であり、より好ましくは1.0~1.5モルの範囲内である。
ニトリル化工程は、ハロゲン化水素および有機スルホン酸からなる群より選ばれる少なくとも1種の酸の存在下で行うことが、反応を促進するので好ましい。該酸の詳細については、後述する。
なお、上述のように、ニトリル化工程において、カルボン酸ハロゲン化物、スルホン酸ハロゲン化物およびハロゲン化チオニルからなる群より選択される化合物を脱水剤として用いる場合、ニトリル化工程を上記酸の存在下で行うことができる。ゆえに、脱水剤を2種以上用いる場合、カルボン酸ハロゲン化物、スルホン酸ハロゲン化物およびハロゲン化チオニルの何れかを脱水剤の1種として用いることが好ましい。
ニトリル化工程において、ヒドロキシイミノ化合物(3)と脱水剤との反応は、有機塩基の存在下で行うことが好ましい。
有機塩基としては、例えば、ピリジン、メチルエチルピリジン、ジメチルアミノピリジン等の芳香族アミン、トリエチルアミン等の脂肪族第3級アミンが挙げられる。有機塩基は、ピリジンであることが好ましい。
有機塩基は、ハロゲン化水素および有機スルホン酸からなる群より選ばれる少なくとも1種の酸と塩を形成しているものであってもよい。
ハロゲン化水素としては、塩酸、臭素酸、ヨウ素酸等が挙げられる。有機スルホン酸としては、炭素数1~4のアルキルスルホン酸等の脂肪族スルホン酸;および、炭素数6~10の芳香族炭化水素スルホン酸等の等の芳香族スルホン酸が挙げられる。
脂肪族スルホン酸としては、メタンスルホン酸、エタンスルホン酸、プロパンスルホン酸が挙げられる。芳香族スルホン酸としては、ベンゼンスルホン酸、トルエンスルホン酸等の有機スルホン酸等が挙げられる。
有機塩基と上記酸との塩としては、芳香族アミンの塩酸塩、脂肪族第3級アミンの塩酸塩、芳香族アミンの臭素酸塩、芳香族アミンのヨウ素酸塩、脂肪族第3級アミンの塩酸塩、芳香族アミンのアルキルスルホン酸塩等が挙げられる。かかる塩としては、具体的には、ピリジン塩酸塩、メチルエチルピリジン塩酸塩、ジメチルアミノピリジン塩酸塩、トリエチルアミン塩酸塩、ピリジン臭素酸塩、ピリジンヨウ素酸塩、ピリジンメタンスルホン酸塩、ピリジンエタンスルホン酸塩、ピリジンプロパンスルホン酸塩、ピリジンベンゼンスルホン酸塩およびピリジントルエンスルホン酸塩が挙げられ、ピリジン塩酸塩またはピリジン臭素酸塩が好ましく、ピリジン塩酸塩がより好ましい。
ニトリル化工程は、有機塩基、および、有機塩基と上記酸との塩の存在下で行うことが好ましい。
有機塩基の使用量は、ヒドロキシイミノ化合物(3)1モルに対して、好ましくは0.8~2モルの範囲内、より好ましくは1.0~1.5モルの範囲内である。なお、有機塩基の使用量は、有機塩基と上記酸との塩を用いた場合、該塩に由来する塩基の量も含めた合計量である。
ニトリル化工程において、ヒドロキシイミノ化合物(3)と脱水剤との反応は、溶媒の存在下で行うことが好ましい。かかる溶媒としては、例えば、オキシム化工程において例示した非混和性有機溶媒が挙げられる。オキシム化工程において非混和性有機溶媒を用いた場合には、当該非混和性有機溶媒をヒドロキシイミノ化合物(3)との混合物としてそのまま用いることもできるし、必要に応じて、溶媒により希釈して用いることもできる。溶媒の使用量は、ヒドロキシイミノ化合物(3)1重量部に対して、通常0~20重量部、好ましくは0.5~20重量部、より好ましくは0.5~5重量部の範囲内である。
ニトリル化工程における混合順序は制限されず、例えば、溶媒と有機塩基とを混合し、そこへヒドロキシイミノ化合物(3)と脱水剤とを添加する方法、溶媒と有機塩基とヒドロキシイミノ化合物(3)とを混合し、そこへ脱水剤を添加する方法が挙げられ、溶媒と有機塩基とを混合し、そこへヒドロキシイミノ化合物(3)と脱水剤とを添加する方法が好ましい。
かかる好ましい方法において、溶媒と有機塩基との混合物に、ヒドロキシイミノ化合物(3)を添加しながら、脱水剤を添加することが好ましい。溶媒と有機塩基との混合物に、ヒドロキシイミノ化合物(3)を添加しながら脱水剤を添加する場合、ヒドロキシイミノ化合物(3)の添加時間と脱水剤の添加時間とを略同一にすることがより好ましい。
溶媒と有機塩基との混合物における溶媒の量は、ヒドロキシイミノ化合物(3)1重量部に対して、好ましくは0~10重量部の範囲内、より好ましくは0.5~2重量部の範囲内である。ここで、ヒドロキシイミノ化合物(3)の添加時間と脱水剤の添加時間とが略同一であるとは、ヒドロキシイミノ化合物(3)の添加時間に対して、脱水剤の添加時間が、0.8~1.2倍の範囲内であることを意味する。
かかる好ましい方法において、ヒドロキシイミノ化合物(3)の添加時間は、通常1~30時間の範囲内であり、好ましくは3~10時間の範囲内である。
ニトリル化工程における反応温度は、好ましくは80~150℃の範囲内であり、より好ましくは90~110℃の範囲内である。
ニトリル化工程において、上記添加時間を除く反応時間は、例えば1分~24時間の範囲内であり、好ましくは1時間~10時間の範囲内であり、さらに好ましくは1~5時間の範囲内である。
ニトリル化工程の好ましい態様として、ハロゲン化水素およびスルホン酸からなる群より選ばれる少なくとも1種の酸と有機塩基との存在下、ヒドロキシイミノ化合物(3)とカルボン酸無水物とを反応させる工程が挙げられる。
本態様において、カルボン酸無水物および有機塩基としては、それぞれ上述したものが好ましく挙げられる。カルボン酸無水物は、脱水剤として用いられる。
本態様において、酸は、ハロゲン化水素および有機スルホン酸からなる群より選ばれる。
上記ハロゲン化水素としては、塩化水素、臭化水素、ヨウ化水素等が挙げられる。
上記有機スルホン酸としては、メタンスルホン酸、エタンスルホン酸、プロパンスルホン酸等の脂肪族スルホン酸;および、ベンゼンスルホン酸、トルエンスルホン酸等の芳香族スルホン酸が挙げられる。
上記酸は、ハロゲン化水素であることがより好ましい。ハロゲン化水素は、塩化水素または臭化水素であることが好ましい。
上記酸は、市販品であってもよいし、任意の公知の方法により生成させたものであってもよいし、ニトリル化工程の反応系中で生成させたものであってもよい。
ニトリル化工程の反応系中で上記酸を生成させる方法として、例えば、ヒドロキシイミノ化合物(3)と接触することにより上記酸が生成する化合物(この化合物を、「酸発生剤」と称することがある。)を、上記酸に代え、または上記酸と共に、ヒドロキシイミノ化合物(3)とカルボン酸無水物との反応に用いる方法が挙げられる。
酸発生剤として、以下の化合物が挙げられる。
・カルボン酸ハロゲン化物、スルホン酸ハロゲン化物およびハロゲン化チオニルからなる群より選択される化合物
・金属ハロゲン化物
・ハロゲン化ホウ素
カルボン酸ハロゲン化物、スルホン酸ハロゲン化物およびハロゲン化チオニルからなる群より選択される化合物は、上記酸を発生させる作用とともに、上述のように、脱水剤として、ヒドロキシイミノ化合物(3)の−CH=NOHを−CNに変換する作用を示す。ゆえに、脱水剤として、カルボン酸無水物と、カルボン酸ハロゲン化物およびスルホン酸ハロゲン化物からなる群より選択される化合物とを併用すると、ヒドロキシイミノ化合物(3)のニトリル化を効率よく行うことができる。
金属ハロゲン化物としては、塩化アルミニウム、臭化アルミニウム等のハロゲン化アルミニウム;塩化ジルコニウム、臭化ジルコニウム等のハロゲン化ジルコニウムが挙げられる。
ハロゲン化ホウ素としては、塩化ホウ素、臭化ホウ素等のハロゲン化ホウ素が挙げられる。
ニトリル化工程の反応系中で生成させた酸は、好ましくはカルボン酸ハロゲン化物、有機スルホン酸ハロゲン化物、金属ハロゲン化物およびハロゲン化チオニルからなる群より選ばれる少なくとも1種の化合物から生成した酸であり、より好ましくはハロゲン化アセチルおよびハロゲン化アルミニウムからなる群より選ばれる少なくとも1種の化合物から生成した酸である。
上記酸および酸発生剤は、単独で用いることもできるし、上述の溶媒や酢酸等と混合して用いることもできる。
上記酸の使用量は、ヒドロキシイミノ化合物(3)1モルに対して、好ましくは0.05~1モルの範囲内であり、より好ましくは0.1~0.5モルの範囲内である。なお、該酸の使用量は、酸発生剤や有機塩基と上記酸との塩を用いた場合、酸発生剤や該塩に由来する酸の量も含めた合計量である。
上記酸をニトリル化工程の反応系中で酸発生剤から生成させる場合、酸発生剤の使用量は、酸発生剤1モルから発生しうる酸のモル数により異なり、例えば酸発生剤1モルから酸が1モル発生しうるのであれば、ヒドロキシイミノ化合物(3)1モルに対して、好ましくは0.05~1モルの範囲内であり、より好ましくは0.1~0.5モルの範囲内である。
ニトリル化工程において、反応試剤の混合方法は限定されないが、
(a)カルボン酸無水物と有機塩基と酸もしくは酸発生剤とを混合し、得られる混合物へヒドロキシイミノ化合物(3)を添加する方法、
(b)ヒドロキシイミノ化合物(3)と有機塩基と酸もしくは酸発生剤とを混合し、得られる混合物へカルボン酸無水物を添加する方法、または
(c)有機塩基と酸もしくは酸発生剤とを混合し、得られる混合物へヒドロキシイミノ化合物(3)とカルボン酸無水物とを添加する方法
が好ましい。
(a)~(c)に記載される混合物を調製する際に、さらに溶媒を混合することが好ましい。また、(a)に記載される方法において、ヒドロキシイミノ化合物(3)を溶媒と混合して添加してもよいし、(b)に記載される方法において、カルボン酸無水物を溶媒と混合して添加してもよいし、(c)に記載される方法において、ヒドロキシイミノ化合物(3)および/またはカルボン酸無水物を溶媒と混合して添加してもよい。
(c)に記載される方法がより好ましく、(c)に記載される方法において、ヒドロキシイミノ化合物(3)を溶媒と混合して添加することがさらに好ましい。
(a)に記載される混合物へのヒドロキシイミノ化合物(3)の添加時間、(b)に記載される混合物へのカルボン酸無水物の添加時間、(c)に記載される混合物へのヒドロキシイミノ化合物(3)とカルボン酸無水物との添加時間は、ヒドロキシイミノ化合物(3)から生じる式(3−1)
(式中、R1およびR2は、上記で定義したとおり。R3はカルボン酸無水物由来の基を表す。)
で示される反応中間体をシアノ化合物(2)に逐次変換させる点で、好ましくは1時間以上であり、より好ましくは3時間以上である。また、生産性の点で、好ましくは30時間以内であり、より好ましくは10時間以内である。
ここで、添加時間とは、(a)に記載される混合物へのヒドロキシイミノ化合物(3)の添加開始から終了までに要する時間、(b)に記載される混合物へのカルボン酸無水物の添加開始から終了までに要する時間、あるいは(c)に記載される混合物へのヒドロキシイミノ化合物(3)の添加開始から終了までに要する時間および(c)に記載される混合物へのカルボン酸無水物の添加開始から終了までに要する時間を意味する。
(c)に記載される方法を行う場合、ヒドロキシイミノ化合物(3)の添加時間とカルボン酸無水物の添加時間とは略同一であることが好ましい。
ここで、ヒドロキシイミノ化合物(3)の添加時間とカルボン酸無水物の添加時間とが略同一であるとは、ヒドロキシイミノ化合物(3)の添加時間に対して、カルボン酸無水物の添加時間が、例えば0.8~1.2倍の範囲内であることを意味する。ヒドロキシイミノ化合物(3)の添加時間に対して、カルボン酸無水物の添加時間が0.9~1.1倍の範囲内であることが好ましく、0.95~1.05の範囲内であることがより好ましい。ヒドロキシイミノ化合物(3)の添加とカルボン酸無水物の添加とを同時に開始し、同時に終了することがさらに好ましい。
ニトリル化工程における反応の進行度合いは、ガスクロマトグラフィー、高速液体クロマトグラフィー等の分析手段により確認することができる。
反応を水と非混和性溶媒の存在下で行った場合、例えば、反応後に得られる反応混合物と水とを混合し、抽出、水洗、濾過等の後処理を施し、次いで、蒸留や結晶化等の単離を施せば、シアノ化合物(2)を取り出すことができる。
非混和性溶媒は、例えば、上述した芳香族炭化水素溶媒、飽和鎖式炭化水素溶媒、脂環式炭化水素溶媒およびハロゲン化芳香族炭化水素溶媒が挙げられる。
反応を水と混和性の溶媒の存在下または溶媒の非存在下で行った場合、例えば、反応後に得られる反応混合物と上述した非混和性溶媒と水とを混合し、抽出、水洗、濾過等の後処理を施し、次いで、蒸留や結晶化等の単離を施せば、シアノ化合物(2)を取り出すことができる。
反応混合物に有機塩基が含まれる場合、後処理において、硫酸、塩酸、メタンスルホン酸等の強酸の水溶液で反応混合物を洗浄することにより、反応混合物から除去することができる。洗浄に用いる強酸の使用量は、有機塩基1モルに対して、1塩基酸であれば0.5モル以上が好ましく、2塩基酸であれば0.25モル以上が好ましい。
また、反応混合物にホルミル化合物(1)が含まれている場合には、後処理において、反応混合物を、水酸化カリウム水溶液、水酸化ナトリウム水溶液、水酸化リチウム水溶液、炭酸カリウム水溶液、炭酸ナトリウム水溶液等のアルカリ水溶液および/または水で洗浄することにより、反応混合物からホルミル化合物(1)を除去することができる。
反応後に得られる反応混合物には、オキシム化工程で用いたカルボン酸またはカルボン酸の金属塩が含まれている場合がある。
反応混合物にカルボン酸またはカルボン酸の金属塩が含まれている場合には、該反応混合物を硫酸、塩酸等の強酸水溶液により洗浄した後、または該反応混合物をそのまま、アルカリ水溶液で洗浄することにより、カルボン酸またはカルボン酸の金属塩をアルカリ水溶液中に除去することができる。洗浄後のアルカリ水溶液はpH10以上であることが好ましく、かかるアルカリ水溶液としては、水酸化カリウム水溶液、水酸化ナトリウム水溶液、水酸化リチウム水溶液等のアルカリ金属水酸化物水溶液、炭酸カリウム水溶液、炭酸ナトリウム水溶液等のアルカリ金属炭酸化物水溶液が挙げられる。
アルカリ水溶液の濃度としては、例えば0.1~30重量%の範囲内が挙げられ、好ましくは5~15重量%の範囲内である。アルカリ水溶液の使用量は、ヒドロキシイミノ化合物(3)1モルに対して、0.1~2モルの範囲内、好ましくは0.4~1.0モルの範囲内である。アルカリ水溶液で洗浄した後、さらに水洗することが好ましい。
得られたシアノ化合物(2)は、例えば再結晶;抽出;蒸留;活性炭、シリカ、アルミナ等への吸着処理;シリカゲルカラムクロマトグラフィー等のクロマトグラフィー法等により、精製することもできる。
かくして得られるシアノ化合物(2)としては、具体的には、表7、8および9に記載される化合物が例示される。
ホルミル化合物(1)、ヒドロキシイミノ化合物(3)およびシアノ化合物(2)の含有量は、蒸留、再結晶等により別途調製したホルミル化合物(1)、ヒドロキシイミノ化合物(3)およびシアノ化合物(2)それぞれの標準品を用いて、ガスクロマトグラフィー内部標準法によって求めた。
<実施例1>
トランス−2,2−ジメチル−3−[(1E)−2−ホルミル−1−プロペニル]シクロプロパンカルボン酸メチルの混合キシレン溶液185.68g(濃度47.54重量%)に、ヒドロキシルアミン硫酸塩水溶液(濃度24重量%)16.18gとオクタン酸6.63g(トランス−2,2−ジメチル−3−[(1E)−2−ホルミル−1−プロペニル]シクロプロパンカルボン酸メチル1モルに対して0.1モル)を加えた。得られた混合物はpH2を呈した。そこへ、20℃にてヒドロキシルアミン硫酸塩水溶液(濃度24重量%)160.89gを滴下しながら、23%水酸化ナトリウム溶液97.42gを滴下した。滴下時間はいずれも4.5時間であった。滴下終了時、得られた混合物はpH6.5を呈した。20℃で3時間保温して得られた反応混合物から水層を分液し、得られた油層を水で2回洗浄した後、圧力5kPaで減圧し、加熱還流下脱水することにより、トランス−2,2−ジメチル−3−[(1E)−3−(ヒドロキシイミノ)−2−メチル−1−プロペニル]シクロプロパンカルボン酸メチルの混合キシレン溶液211.82g(濃度42.73重量%)を得た。水分52ppm、収率95.32%。
<実施例2>
窒素雰囲気下、ピリジン10.35gと混合キシレン21.24gと塩化アセチル2.41gとを混合し、得られた混合物を100℃へ昇温した。そこへ、トランス−2,2−ジメチル−3−[(1E)−3−(ヒドロキシイミノ)−2−メチル−1−プロペニル]シクロプロパンカルボン酸メチルの混合キシレン溶液52.63g(濃度42.73重量%)を滴下しながら、無水酢酸10.40gを滴下した。滴下時間はいずれも6時間であった。滴下終了後、得られた混合物を2時間保温し、20℃へ冷却した。冷却した反応混合物へ水21.05gと97重量%硫酸5.60gとを加えて混合し、分液して油層1と水層1とを得た。水層1へ混合キシレン10.67gを加えて混合し、分液して得られた油層2と、先に得られた油層1とを合わせ、油層3を得た。そこへ、水21.12gと23重量%水酸化ナトリウム水溶液11.37gとを加えて混合し、油層3に含まれていたオクタン酸を水層中へと除去した後、水層から分液して油層4を得た。水層はpH12.3を呈した。得られた油層4に水21.15gを加えて混合し、分液して得られた油層を濃縮することにより、トランス−2,2−ジメチル−3−[(1E)−2−シアノ−1−プロペン−1−イル]シクロプロパンカルボン酸メチルを含む固形物24.45g(トランス−2,2−ジメチル−3−[(1E)−2−シアノ−1−プロペン−1−イル]シクロプロパンカルボン酸メチルの含量:82.34重量%)を得た。収率97.80%、
固形物に含まれるオクタン酸は、トランス−2,2ジメチル−3−[(1E)−2−シアノ−1−プロペン−1−イル]シクロプロパンカルボン酸メチルに対して、0.1重量%以下であった。
<実施例3>
窒素雰囲気下、ホルミル化合物(1)としてトランス−2,2−ジメチル−3−[(1E)−2−ホルミル−1−プロペニル]シクロプロパンカルボン酸メチルの混合キシレン溶液41.32g(濃度47.54重量%)に、ヒドロキシルアミン硫酸塩水溶液(濃度24重量%)37.68gと、カルボン酸としてオクタン酸1.48g(トランス−2,2−ジメチル−3−[(1E)−2−ホルミル−1−プロペニル]シクロプロパンカルボン酸メチルに対して10モル%)とを加え、そこへ、20℃にて23%水酸化ナトリウム溶液22.16gを5.5時間かけて滴下した。20℃にて1.5時間保温した後、水層を分液し、得られた油層を水で2回洗浄することにより、ヒドロキシイミノ化合物(3)としてトランス−2,2−ジメチル−3−[(1E)−3−(ヒドロキシイミノ)−2−メチル−1−プロペニル]シクロプロパンカルボン酸メチルの混合キシレン溶液41.98g(濃度49.60重量%)を得た。 収率98.54%
<実施例4~8および参考例1>
実施例3において、表10に示す条件を変更し、23%水酸化ナトリウムの滴下時間を5.5時間から4.5時間へ変更した以外は、実施例3と同様の条件により、トランス−2,2−ジメチル−3−[(1E)−3−(ヒドロキシイミノ)−2−メチル−1−プロペニル]シクロプロパンカルボン酸メチルの混合キシレン溶液を得た。結果を表11に示す。
窒素雰囲気下、トランス−2,2−ジメチル−3−[(1E)−3−(ヒドロキシイミノ)−2−メチル−1−プロペニル]シクロプロパンカルボン酸メチルを混合キシレンに溶解させた溶液3.07g(ホルミル化合物(1)の濃度:32.74重量%)にピリジン0.49gと塩化アセチル0.11gとを加えた。得られた混合物を100℃に昇温し、そこへ無水酢酸0.49gを加えて2時間保温した。反応混合物を冷却した後、混合キシレンで希釈して2,2ジメチル−3−[(1E)−2−シアノ−1−プロペン−1−イル]シクロプロパンカルボン酸メチルの混合キシレン溶液9.60g(シアノ化合物(2)の含量:9.11重量%)を得た。収率95.6%。
<実施例10>
窒素雰囲気下、混合キシレン42.35g、ピリジン20.65g、塩化アセチル3.19gおよび無水酢酸22.56gを混合した。得られた混合物を100℃に昇温し、そこへトランス−2,2−ジメチル−3−[(1E)−3−(ヒドロキシイミノ)−2−メチル−1−プロペニル]シクロプロパンカルボン酸メチルを混合キシレンに溶解させた溶液95.61g(ホルミル化合物(1)の濃度:44.20重量%)を6時間かけて滴下した後、2時間保温した。反応混合物を冷却した後、そこへ水42.35gと97重量%の硫酸12.15gとを加えて混合し、分液して油層1と水層1とを得た。水層1へ混合キシレン21.19gを加えて混合し、分液して得られた油層2と、先に得られた油層1とを合一した。そこへ、水42.50gと23重量%の水酸化ナトリウム21.98gとを加えて混合し、分液した。得られた油層3を、更に水42.31gと混合して分液し、得られた油層を濃縮することにより、2,2−ジメチル−3−[(1E)−2−シアノ−1−プロペン−1−イル]シクロプロパンカルボン酸メチルの混合キシレン溶液122.57g(シアノ化合物(2)の含量:29.79重量%)を得た。 収率95.3%
<実施例11~14>
実施例9において、表12に示す条件を変更した以外は、実施例9と同様の操作を行って、シアノ化合物(2)として2,2ジメチル−3−[(1E)−2−シアノ−1−プロペン−1−イル]シクロプロパンカルボン酸メチルの混合キシレン溶液を得た。結果を表13に示す。
窒素雰囲気下、ピリジン17.40gと混合キシレン42.25gと塩化ピリジニウム4.62gとを混合し、得られた混合物を100℃へ昇温した。そこへ、同温度にて、トランス−2,2−ジメチル−3−[(1E)−3−(ヒドロキシイミノ)−2−メチル−1−プロペニル]シクロプロパンカルボン酸メチルを混合キシレンに溶解させた溶液95.59g(ホルミル化合物(1)の濃度:44.20重量%)の滴下と無水酢酸26.54gの滴下とを同時に開始した。前記溶液の滴下時間と無水酢酸の滴下時間とは、いずれも6時間であった。滴下終了後、得られた混合物を2時間保温した。混合物を20℃へ冷却した後、そこへ水42.25gと97重量%の硫酸12.13gとを加えて混合し、分液して油層1と水層1とを得た。水層1へ混合キシレン21.13gを加えて混合し、分液して得られた油層2と、先に得られた油層1とを合一した。そこへ、水42.25gと23重量%の水酸化ナトリウム24.35gとを加えて混合し、分液した。得られた油層3を、更に水42.25gと混合して分液し、得られた油層を濃縮すると2,2−ジメチル−3−[(1E)−2−シアノ−1−プロペン−1−イル]シクロプロパンカルボン酸メチルを含む溶液82.26g(シアノ化合物(2)の含量:44.69重量%)が得られた。収率95.0%
<実施例16>
窒素雰囲気下、ピリジン17.40gと混合キシレン42.25gと塩化ピリジニウム4.62gとを混合し、得られた混合物を100℃へ昇温した後、無水酢酸1.06gを添加した。そこへ、同温度にてトランス−2,2−ジメチル−3−[(1E)−3−(ヒドロキシイミノ)−2−メチル−1−プロペニル]シクロプロパンカルボン酸メチルを混合キシレンに溶解させた溶液87.97g(ホルミル化合物(1)の濃度:47.80重量%)の滴下と無水酢酸25.52gの滴下とを同時に開始した。前記溶液の滴下時間と無水酢酸の滴下時間とは、いずれも6時間であった。滴下終了後、得られた混合物を2時間保温した。混合物を20℃へ冷却した後、そこへ水42.25gと97重量%の硫酸11.10gとを加えて混合し、分液して油層1を得た。そこへ、水42.25gと23重量%の水酸化ナトリウム24.35gとを加えて混合し、分液した。得られた油層2を、更に水42.25gと混合して分液し、得られた油層を濃縮すると2,2−ジメチル−3−[(1E)−2−シアノ−1−プロペン−1−イル]シクロプロパンカルボン酸メチルを含む溶液64.51g(シアノ化合物(2)の含量:56.09重量%)が得られた。収率94.1%
<実施例17>
窒素雰囲気下、ピリジン3.96gと混合キシレン52.82gと35%塩酸5.19gとを混合した後、圧力5kPaまで減圧し、加熱還流下脱水することにより塩化ピリジニウムを含むキシレン溶液を得た。得られたキシレン溶液にピリジン21.75gを加え、100℃へ昇温した。そこへ、同温度にて、トランス−2,2−ジメチル−3−[(1E)−3−(ヒドロキシイミノ)−2−メチル−1−プロペニル]シクロプロパンカルボン酸メチルを混合キシレンに溶解させた溶液109.62g(ホルミル化合物(1)の濃度:48.18重量%)の滴下と無水酢酸30.63gの滴下とを同時に開始した。前記溶液の滴下時間と無水酢酸の滴下時間とは、いずれも6時間であった。滴下終了後、得られた混合物を2時間保温し、2,2−ジメチル−3−[(1E)−2−シアノ−1−プロペン−1−イル]シクロプロパンカルボン酸メチルを含む溶液205.44g(化合物(2)の含量:22.15重量%)が得られた。収率94.2%
<参考例1>
窒素雰囲気下、トランス−2,2−ジメチル−3−[(1E)−3−(ヒドロキシイミノ)−2−メチル−1−プロペニル]シクロプロパンカルボン酸メチル(純度98.55重量%)1.00gを混合キシレン2.48gに溶解し、得られた溶液にピリジン0.49gを加えた。得られた混合物を100℃に昇温し、そこへ無水酢酸0.64gを加えて16時間保温した。反応中間体のトランス−2,2−ジメチル−3−[(1E)−3−(アセトキシイミノ)−2−メチル−1−プロペニル]シクロプロパンカルボン酸メチルが2,2−ジメチル−3−[(1E)−2−シアノ−1−プロペン−1−イル]シクロプロパンカルボン酸メチルとの合計に対する面積百分率として14%残存した。冷却後、混合キシレンで希釈しトランス−2,2−ジメチル−3−[(1E)−3−(アセトキシイミノ)−2−メチル−1−プロペニル]シクロプロパンカルボン酸メチルと2,2ジメチル−3−[(1E)−2−シアノ−1−プロペン−1−イル]シクロプロパンカルボン酸メチルの混合キシレン溶液8.83g(含量8.31重量%)を得た。収率80.3%。
本発明は、かかる化合物の製造方法として利用可能である。
Claims (22)
- カルボン酸およびカルボン酸の金属塩からなる群より選択される少なくとも1種の化合物が炭素数3以上のカルボン酸または炭素数3以上のカルボン酸の金属塩である請求項1記載の製造方法。
- 溶媒が、水と、水に非混和性の有機溶媒との混合溶媒である請求項1または2記載の製造方法。
- 溶媒中、カルボン酸およびカルボン酸の金属塩からなる群より選択される少なくとも1種の化合物の存在下で、式(1)
(式中、R1は置換基を有していてもよいアルキル基またはハロゲン原子を表し、R2は置換基を有していてもよいアルキル基または置換基を有していてもよいベンジル基を表わす。)
で示されるホルミルアルケニルシクロプロパンカルボン酸エステルと、ヒドロキシルアミンとを反応させるオキシム化工程と、
前記オキシム化工程で得られる式(3)
(式中、R1およびR2は前記で定義された通り。)
で示されるヒドロキシイミノアルケニルシクロプロパンカルボン酸エステルと、脱水剤とを反応させるニトリル化工程と
を有する式(2)
(式中、R1およびR2は前記で定義された通り。)
で示されるシアノアルケニルシクロプロパンカルボン酸エステルの製造方法。 - 脱水剤がカルボン酸無水物およびハロゲン化アシルからなる群より選ばれる少なくとも1種の化合物である請求項4記載の製造方法。
- 脱水剤がカルボン酸無水物である請求項4または5記載の製造方法。
- ニトリル化工程は、ハロゲン化水素および有機スルホン酸からなる群より選ばれる少なくとも1種の酸の存在下で行われる請求項4または5記載の製造方法。
- ニトリル化工程は、有機塩基の存在下で行われる請求項4または5記載の製造方法。
- 更に、ニトリル化工程における反応により得られる混合物からカルボン酸およびカルボン酸の金属塩からなる群より選択される少なくとも1種の化合物を除去する工程を含む請求項4記載の製造方法。
- カルボン酸およびカルボン酸の金属塩からなる群より選択される少なくとも1種の化合物は、アルカリ水溶液中に除去される請求項9記載の製造方法。
- アルカリ水溶液はpH10以上のアルカリ水溶液である請求項10記載の製造方法。
- アルカリ水溶液は、アルカリ金属水酸化物水溶液またはアルカリ金属炭酸化物水溶液である請求項10または11記載の製造方法。
- 前記工程が、溶媒の存在下で、式(1)で示されるヒドロキシイミノアルケニルシクロプロパン化合物とカルボン酸無水物とを反応させる工程である請求項13記載の製造方法。
- 有機塩基は、酸と有機塩基との塩を形成している請求項14記載の製造方法。
- カルボン酸無水物は、炭素数2~8のカルボン酸無水物である請求項13または14記載の製造方法。
- カルボン酸無水物は、無水酢酸である請求項13または14記載の製造方法。
- 有機塩基は、ピリジン、メチルエチルピリジン、ジメチルアミノピリジンおよびトリエチルアミンからなる群より選択される少なくとも1種である請求項13または14記載の製造方法。
- 有機塩基は、ピリジンである請求項13または14記載の製造方法。
- 前記酸は、ハロゲン化水素である請求項13または14記載の製造方法。
- 前記酸は、塩化水素である請求項13または14記載の製造方法。
- 前記酸は、カルボン酸ハロゲン化物、有機スルホン酸ハロゲン化物、金属ハロゲン化物およびハロゲン化チオニルからなる群より選ばれる少なくとも1種の化合物から生成した酸である請求項13または14記載の製造方法。
