Description
離水抑制のために、様々な技術が開発されており、例えば、タンパク質を使用する技術として改質卵白を用いる方法が知られている(特開2008−086306号公報、特開2008−035811号公報)。あるいは塩を活用する方法として炭酸ナトリウム及び/または炭酸水素ナトリウムを8~43%、塩化カリウムを3~20%およびアミノ酸、更に塩化ナトリウムを使用する方法が報告されている(特開2007−312771号公報)。その他素材を用いた技術として、カシアガムと他の食品素材を利用する方法(特開2006−223189号公報)、膨化穀類を添加する方法(特開2006−006236号公報)、ガラクトキシログルカンの側鎖ガラクトースを酵素的に部分除去した可逆的熱応答性ガラクトキシログルカンを配合したピックル液を使用する方法(特開2004−180550号公報)、微細セルロースと半精製カラギーナンからなる複合体を用いる方法(特開平11−046723号公報)、トレハロースを用いる方法(特開平09−056342号公報)、乾燥野菜及び増粘多糖類との乳化物を用いる方法(特開平08−084573号公報)、ゲル生成性食用ヒドロコロイド(EH)と食用糸状菌(EFF)を混合したゲル化物質を用いる方法(特表平10−511857号公報)、微細セルロースとゲル化剤からなる複合体を用いる方法(再表98/017126号公報)、ホエー蛋白質含有溶液、それを用いたホエー蛋白質ゲル化物を用いる方法(特許2529052号)、アルギン酸エステルを用いる方法(特開2002−281942号公報)が公知である。特に、特開2002−281942号にはアルギン酸エステルと酵素との併用が記載されており、酵素としてアミラーゼ、インベルターゼ、カタラーゼ、セルラーゼ、パパイン、プロテアーゼ、ペクチナーゼ、リゾチーム、リパーゼ、トリプシン、パンクレアチン、ブロメライン、ペプシン、ペプチダーゼ、アクチジニンが挙げられている。しかしアルギン酸エステルと酵素との併用効果はなんら明示されておらず、一般的な技術可能性を記載したのみである。更に製造工程での改良による離水抑制付与の技術も存在し、例えば超高速遠心ミキサーで混合し配合原料を高度に均質化する方法(特開2005−261351号公報)が公知である。
一方、リパーゼを用いた食品改質技術に関しても既に幾つかの報告がある。例えばホスホリパーゼA2による卵の品質改良(米国特許4,034,124号、Dutihl & Groger 1981 J.Sci.Food Agric.32,451−458)、酸性水中油型乳化組成物の安定性を高めるためエステラーゼ、リパーゼ及びホスホリパーゼが活用される方法(特開2004−201672号公報)、耐冷凍性卵白組成物製造のため、リパーゼを使用する方法(特開2000−014358号公報)、冷凍食品の解凍時における離水を防止し、冷凍損傷を防ぐために用いるためのゼラチン製造工程でリパーゼ処理を加える方法(特開平11−164655号公報)、冷凍食品の離水防止に用いる魚類ゼラチン粉末製造工程でリパーゼ処理する方法(特許2985953号)、リパーゼを生地に添加し、老化作用を改善する方法(EP0585988B)、リパーゼを生地に添加することによって、追加の脂肪/油を生地に添加せずに、柔らかさを改善する方法(WO94/04035)、外来性リパーゼがパンの体積を改善できる方法(Castello,P.ESEGP 89−10 Dec.1999 Helsinki)、ホスホリパーゼやガラクトリパーゼによる揚げ製品の品質改良(特開2009−65984号公報)などである。他にチーズなど乳製品のフレーバー増強効果は多数の報告が存在する。
またリパーゼ以外の脂質改質酵素による物性機能に関して、脂質アシルトランスフェラーゼの報告がある(WO2004/064537号、WO2005/066347号)。本特許記載内容によると、転移活性の高い脂質アシルトランスフェラーゼが乳化剤との併用条件化でリパーゼと比較して食品物性改質効果が高く、乳製品やパン、冷凍ドウ、めん類やホイップクリーム、チョコレートと共に畜肉加工製品での改質効果が報告されている。しかしリパーゼを利用すると遊離脂肪酸の増加のため、食品物性改質に適していないと記載されている。もっとも本特許記載酵素には弱い分解活性が確認されているが、ホスホリパーゼA2活性(又はホスホリパーゼA1活性であり、ホスホリパーゼC活性やホスホリパーゼD活性でなく、ホスホリパーゼC、Dの機能を類推することは不可能である。
特開昭58−152481号公報、特開2006−223119号公報、特開2006−180738号公報には、ホスホリパーゼCあるいはホスホリパーゼDの基本特性について記載されているが、畜肉加工製品への使用に関する記載は一切無く、ホスホリパーゼCやホスホリパーゼDによる畜肉加工製品の離水抑制効果や、硬さやしなやかさなどの物性保持、改質効果の報告はされていなかった。
本発明者は鋭意検討を重ねた結果、ホスホリパーゼCやホスホリパーゼDを活用することにより、畜肉加工製品の硬さ、しなやかさ等の食感を劣化させることなく、保存時の離水を抑制できることを見出し、本発明を完成させるに至った。即ち、本発明は以下の発明を包含する。
(1)ホスホリパーゼC及び/又はホスホリパーゼDを含有する畜肉加工製品改質用の酵素製剤。
(2)ホスホリパーゼC及び/又はホスホリパーゼDを畜肉原料に作用させることを特徴とする畜肉加工製品の製造方法。
(3)畜肉原料に作用させるホスホリパーゼC及び/又はホスホリパーゼDの量が、畜肉原料100g当り0.0005~0.05gである(2)記載の方法。
(4)畜肉原料に作用させるホスホリパーゼC及び/又はホスホリパーゼDの量が、畜肉原料100g当り15~1500ユニットである(2)記載の方法。
(5)畜肉加工製品が、ハム、ソーセージ、ハンバーグ、ミートローフ又はミートボールである(2)乃至(4)記載の方法。
以下に本発明について詳細に説明する。
本発明の、畜肉加工製品改質用の酵素製剤は、ホスホリパーゼC及び/又はホスホリパーゼDを含有するものである。ホスホリパーゼはリン脂質を分解する酵素であり、A1、A2、C、Dに大別されるが、本発明においては、ホスホリパーゼC及び/又はホスホリパーゼDが用いられる。本発明の酵素製剤には、ホスホリパーゼC、ホスホリパーゼDの他に、食品用賦形剤を配合してもよい。食品用賦形剤としては一般的な食品用賦形剤が使用できる。例えば、乳糖、マルチトール、ソルビトール、デキストリン、グルコース、澱粉類、多糖類、ガム類、ペクチン等があげられる。また酵素の働きを高める目的でホスホリパーゼの基質成分や酵素活性促進剤を配合しても良い。更に、本発明の該酵素製剤には、上記賦形剤のほかに、必要により卵白、大豆タンパク質などの食品タンパク質素材、調味料、食塩、砂糖、着色料、発色剤、エリソルビン酸及びその塩類、乳化剤等を適宜に含有せしめても良い。またこのような副剤の併用により、該酵素製剤の安定性向上、分散性向上、粉立ち防止機能を付与することが可能である。
本発明の畜肉加工製品を製造する方法は、ホスホリパーゼC及び/又はホスホリパーゼDを畜肉原料に作用させることを特徴とする。
本発明に使用される畜肉原料は、いわゆる食肉と総称される牛肉、豚肉、馬肉、めん羊肉、山羊肉、家兎肉、鶏肉、七面鳥肉、カモ肉、ガチョウ肉、鶉肉が挙げられるが、これらに限定されるものではない。
本発明の畜肉加工製品は、ソーセージ、ハム、ハンバーグ、ミートローフ、ミートボールなど、挽肉を主原料にしてつくられる加工品の総称で、これらの製品のそれぞれの製造方法自体はいずれも当業者に周知である。その製造の原理は、挽肉に食塩を加えて練り、塩溶性のタンパク質を溶かし出し、さらに混練り後、これをいろいろの形に成形してから加熱してゲル化させることにある。この際適宜、澱粉その他各種の副原料、調味料、香辛料、発色剤等を添加する。
ホスホリパーゼC及び/又はホスホリパーゼDは、練り工程のどの時点で添加してもよいが、好ましくは前記食塩添加後の混練り工程中に添加することにより、より弾力のある製品となる。
畜肉原料に作用させるホスホリパーゼC及び/又はホスホリパーゼDの量は、畜肉原料100g当り0.0005g~0.05gが好ましく、0.001~0.01gがより好ましい。あるいは、畜肉原料100g当り15~1500ユニットが好ましく、30~300ユニットがより好ましい。
ホスホリパーゼCの活性測定方法を以下に記す。まず、小試験管に第一反応試薬混合液0.9mlを正確に分注し、37℃で予備加温する。5分経過後、酵素試料液100μlを正確に加えて混和し、37℃で第一反応を開始する。10分経過後、反応停止液1mlを加えて混和後、直ちに第二反応試薬混合液1mlを加えて混和し、37℃で第二反応を開始する。盲検はこの時点で酵素試料液100μlを加える。20分経過後、500nmにおける吸高度を測定する。求めた吸光度を試料液はAs、肓検液はAbとする。ΔA=(As−Ab)≦0.30Absとした時、活性計算式は以下の通りとなる。

なお、第一反応試薬混合液の組成は0.2M DMG−NaOH緩衝液(pH7.5)0.2mlと10mM 卵黄レシチン溶液0.2ml、精製水0.5mlの混合液。10mM卵黄レシチン用液とは卵黄レシチン100mgをエーテル留去した後、ロータリーエバポレーターで減圧乾固し、乾固物に0.5%DOC溶液を13.5ml加えてホモゲナイズした溶液。0.5%DOC溶液とはデオキシコール酸を精製水40mlに懸濁して、4M NaOHを加えpH8.0くらいにして溶解した後、精製水で全量を50mlとした溶液である。第二反応試薬混合液は15mM 4−AA溶液0.1ml、0.2%(W/V)フェノール液0.1ml、30U/ml COD溶液0.1ml、90U/ml POD溶液0.1ml、80U/ml ALP溶液、50mM トリス−HCl緩衝液(pH8.0)0.5mlの混合物である。反応停止液は0.2%(W/V)ドデシル硫酸ナトリウムを含む1M トリス−HCl緩衝液(pH8.0)を指す。酵素溶解希釈用液として0.1%(W/V)BSAを含む10mM DMG−NaOH緩衝液(pH7.5)を用いた。
ホスホリパーゼDの活性測定方法を以下に記す。まず、小試験管に第一反応試薬混合液0.5mlを正確に分注し、37℃で予備加温する。5分経過後、酵素試料液50μlを正確に加えて混和し、37℃で第一反応を開始する。10分経過後、第二反応試薬混合液2.5mlを加えて混合し、37℃で第二反応を開始する。盲検はこの時点で酵素試料液50μlを加える。20分経過後、500nmにおける吸高度を測定する。求めた吸光度を試料液はAs、盲検液はAbとする。ΔA=(As−Ab)≦0.40Absとした時、活性計算式は以下の通りとなる。
活性(U)=(ΔA/10)/12.2×3.05/0.05

なお、第一反応試薬混合液の組成は0.1M トリス−HCl緩衝液(pH8.0)20mlと0.1M 塩化カルシウム溶液0.05ml、25mM基質溶液0.1ml精製水0.15mlの混合液で、基質液はジオレオイルフォスファチジルコリン88.5mgを5%(W/V)トリトンX−100溶液4.5mlで溶解したもの。第二反応試薬混合液は15mM 4−AA溶液0.1ml、0.2%(W/V)フェノール液0.1ml、60mM EDTA溶液(pH8.0)0.1ml、50mM トリス−HCl緩衝液(pH8.0)2ml、90U/ml POD液0.1ml、30U/ml COD溶液0.1mlの混合物である。酵素溶解希釈用液は0.05%(W/V)BSAと0.1%(W/V)トリトンX−100を含む10mM トリス−HCl緩衝液(pH8.0)を用いた。
酵素反応は短時間でも長時間でも効果発現が期待できる。例えば0~60℃で、0.05~48時間を畜肉加工製品の製造過程で確保すれば十分である。また酵素は畜肉加工製品製造過程で加熱失活していると考えられ、最終製品中での残存活性は無い。
図2は、本発明の実施例1に係るソーセージの硬さの比較を示す図である。
図3は、本発明の実施例1に係るソーセージのしなやかさの比較を示す図である。
図4は、本発明の実施例2に係るハムの離水率の比較を示す図である。
図5は、本発明の実施例2に係るハムのしなやかさの比較を示す図である。
図6は、本発明の実施例2に係るハムの破断応力の比較を示す図である。
また、官能評価により硬さとしなやかさを測定した。評価は畜肉製品の官能評価訓練を受けている5名で実施した。コントロールのスコアを3とし、1点から5点までの評点法とした。結果を硬さの結果を図2、しなやかさの結果を図3に記した。
離水抑制効果が認められたのはホスホリパーゼA1、ホスホリパーゼC、ホスホリパーゼDであった。しかしホスホリパーゼA1は官能評価で硬さの低下やしなやかさの低下が認められ、物性面の品質低下が示唆された。一方、ホスホリパーゼCやホスホリパーゼDは物性低下を起こさず、離水抑制効果を有することが明らかとなった。
Claims (5)
- ホスホリパーゼC及び/又はホスホリパーゼDを含有する畜肉加工製品改質用の酵素製剤。
- ホスホリパーゼC及び/又はホスホリパーゼDを畜肉原料に作用させることを特徴とする畜肉加工製品の製造方法。
- 畜肉原料に作用させるホスホリパーゼC及び/又はホスホリパーゼDの量が、畜肉原料100g当り0.0005~0.05gである請求の範囲第2項記載の方法。
- 畜肉原料に作用させるホスホリパーゼC及び/又はホスホリパーゼDの量が、畜肉原料100g当り15~1500ユニットである請求の範囲第2項記載の方法。
- 畜肉加工製品が、ハム、ソーセージ、ハンバーグ、ミートローフ又はミートボールである請求の範囲第2項記載の方法。
