Description
ポリアミンは、第一級アミノ基を2つ以上もつ直鎖状の脂肪族炭化水素の総称であり、代表的なポリアミンとして、プトレスシン、スペルミジン、スペルミンを挙げることができる。ポリアミンが生理作用を示すのに必要な濃度はプトレスシン>スペルミジン>スペルミンの順であり、その生理作用に関しては各々に4~8倍の差があり、スペルミンが一番低濃度で効果を発揮する(神秘の生命物質−ポリアミン− 未来の生物科学シリーズ28、共立出版 1993 p46−48、78−80)。
ポリアミンの生理作用としては、(1)細胞増殖作用、(2)細胞分化促進作用、(3)免疫必須因子、(4)抗アレルギー作用、(5)タンパク質合成促進作用、(6)核酸との相互作用による構造の安定化、(7)酵素活性調節作用等が知られている。最近では、急性の炎症を抑制する作用(J Exp Med.May 19;185(10):1759−1768.1997、Lancet.Sep 20;350(9081):861−862 1997.)と共に、慢性炎症を抑制する作用(再表2004/073701号公報、J Immunol.Jul 1;175(1):237−245.2005.)を有している為に、慢性炎症で誘発される加齢に伴う疾患の進行を抑制する作用が期待されている。
このように、ポリアミンが持つ種々の効果が明らかになるにつれて、ポリアミンを日常的に摂取することは望ましいと考えられるようになってきた。食物中のポリアミンは腸管から速やかに吸収され、短時間で体の各組織の細胞内に移行することが明らかになっている。ポリアミンの1日の摂取量は、成人でほぼ350−550μmolと考えられているが、食物によりポリアミン含量の偏りが激しい。例えば豆類やきのこ類にスペルミジンやスペルミンが多いが、地域の食生活の違いにより、摂取するポリアミンの種類と量に差が大きい。従って、手軽にポリアミンを摂取できる製品があれば、様々な生理作用をもつポリアミンを量、質ともに補うことができる。
このようなポリアミンの重要性に鑑みて、従来から種々のポリアミンを抽出する方法が検討されている。
特開2001−95483号公報および特開2001−8663号公報は、乳、乳素材からのポリアミンの製造を開示している。この方法は、イオン交換樹脂、合成吸着剤の使用および、その脱着に塩酸、硫酸等の無機酸、カチオンを含有する無機酸を使用する方法、またはpH5.5以下で限外ろ過する方法といった、複雑な抽出方法である。よって酸を使用したりpHを調整することが必要となり、抽出後のポリアミンの精製も困難である。
特開2000−245493号公報および特開平10−52291号公報は、酵母菌体からのポリアミンの製造を開示している。この方法は、ヌクレアーゼ処理、アルカリ加水分解または酸性条件下での処理を行っており、酸やアルカリで抽出しているために、抽出後のポリアミンの精製が困難である。
特開平9−206025号公報は、動物臓器からのポリアミンの製造を開示している。この方法は、酸溶液を添加することによるポリアミンの抽出のため、抽出後のポリアミンの精製が困難である。
特開平8−238094号公報は、白子からのポリアミンの製造を開示している。この方法は、タンパク質分解酵素や核酸分解酵素を用いる方法、もしくは鉱酸を添加し酸性条件下でポリアミンを抽出する方法であり、酵素や酸を用いるために処理の手間、コストがかかり、さらに抽出後のポリアミンの精製が困難である。
特開平10−101624号公報は、茶類からのポリアミンの製造を開示している。この方法も酸性条件下で抽出しているため、抽出後のポリアミンの精製が困難である。
このように従来のポリアミンの抽出方法においては、複雑な抽出工程を採用しているため、製造コストがかかると共に、酸やアルカリを用いた処理を含むため、飲食品に用いるには必ず精製工程が必要となるという不都合が依然として存在していた。従って、ポリアミンを摂取できる飲食品を製造するためには、安全かつコストのかからない新たなポリアミンの抽出方法が必要である。
本発明者は上記の目的を達成する為に、各種ポリアミン抽出方法を検討した結果、酸、塩基の使用または酸、アルカリへのpH調整、イオン交換樹脂等を用いることなく、非常に簡便な塩のみを用いた抽出によりポリアミンが動植物素材より抽出されることを見出した。
すなわち、本発明は、動植物素材に塩を加えて抽出することを特徴とするポリアミン抽出物の製造方法である。さらに、本発明は、上記の製造方法によって製造されたことを特徴とするポリアミン抽出物を含有する飲食品である。また、本発明は、上記の製造方法によって製造されたことを特徴とするポリアミン抽出物を、さらに精製して得られるポリアミン製剤である。さらに、本発明は、上記のポリアミン製剤を含有する飲食品である。
本発明品は、チューインガム、キャンディ、錠菓、グミゼリー、チョコレート、ビスケット等の菓子、アイスクリーム、シャーベット、氷菓等の冷菓、飲料、スープ並びにジャム等の飲食品に配合、もしくは飲食品原材料から直接抽出することで配合でき、日常的に利用することが可能である。
図2は、ポリアミン茶の第二の製造方法を示すフローチャートである。
本発明の、動植物素材に塩を加えて抽出することを特徴とするポリアミン抽出物の製造方法において、塩の種類は特に限定されないが、風味の面から中性塩が好ましく、具体的には塩化ナトリウム、塩化カルシウム、塩化マグネシウムといった塩が挙げられる。特に抽出力が高く、また栄養面から考えても好ましい塩として、塩化カルシウム、塩化マグネシウムが挙げられる。
従来のポリアミンの抽出には酸を用いるのが一般的であったが、飲食品とするには味に影響が大きく必ず精製する必要があった。一方、塩により動植物素材からポリアミンが効率よく抽出されることは知られておらず、本発明のポリアミンの抽出方法は、塩のみの使用であるため、飲食品添加時に希釈され、製品として問題のない塩濃度に調整することができ、飲食品への使用が容易であり、さらに簡便な製法であるためコストも抑えられる画期的な抽出法である。
動植物素材からのポリアミンの抽出効果を、水、酸、および塩を使用して検討した例を以下の表1に示す。
上表1から明らかなように、ポリアミンは水抽出では抽出されず、一般に酸を用いて抽出されるが、塩を用いても抽出されることがわかる。
使用した動植物素材は、鮭白子、豚の肝臓、麦芽、大豆、納豆、烏龍茶葉、紅茶葉、碁石茶葉、黒豆、番茶、柿の葉、および緑茶葉であるが、特に、動植物素材の中でも、茶葉には高い濃度でポリアミンが存在し、かつ生理活性の高いスペルミンが豊富に含まれる、極めてよい素材である。ポリアミンの生理作用を効果的に発現させるために、緑茶は容易に摂取しやすい飲料形態であり、嗜好性、市場性から望ましい。
しかし、通常の緑茶飲料製法では緑茶中にポリアミンは抽出されないことが明らかになっている(J Biochem.Jan;139(1):81−90.2006、Biosci Biotechnol Biochem.Sep;61(9):1582−1584.1997.)。この問題の解決手段として、本法を茶葉の抽出に用いることにより、ポリアミンを高い濃度で含む新たな緑茶飲料を製造することができる。本法によると、茶葉1gよりプトレスシン1~1000nmol、スペルミジン1~1000nmol、スペルミン1~1000nmolを抽出することができ、茶葉使用量によって緑茶中に1nmol~1000μmolのポリアミンを含有するまったく新規な機能性飲料が得られる。
また、以下の実施例から明らかなように、動植物素材が、Camellia属の植物である茶樹の葉および/または茎からとれる緑茶葉であることが好ましい。
本発明の動植物素材に塩を加えて抽出するポリアミン抽出物の製法によって製造されたことを特徴とするポリアミン抽出物を含有する飲食品としては、特に限定されないが、チューインガム、キャンディ、錠菓、グミゼリー、チョコレート、ビスケット等の菓子、アイスクリーム、シャーベット、氷菓等の冷菓、飲料、スープ並びにジャム等の飲食品が挙げられる。このような飲食品におけるポリアミンの含有量は、飲食品の種類によって変わりうるが、ポリアミンを一食分で0.2~500μmol含有することが好ましい。さらに、飲食品として緑茶飲料も挙げられ、この緑茶飲料におけるポリアミンの含有量は、ポリアミンを0.2~500μmol/L含有することが好ましい。
本発明に係る茶の塩によるポリアミンの抽出例を以下に説明する。
試料としては上記の塩によるポリアミン抽出例で示した緑茶葉(煎茶)を使用した。茶はCamellia属の植物である茶樹の葉および/または茎からとれるものであって、生のものでも加工したものでも使用できる。また茶そのものでも熱水抽出後の茶殻でも使用することができ、粉末など形態は問わない。
なお、具体的なポリアミン茶の製造方法は図1、2に示した。
抽出溶液:5%トリクロロ酢酸溶液
抽出条件:1g茶葉に対し抽出溶液5mlを加え、80℃5分抽出
試験結果を以下の表9、10、11に示す:
産地別緑茶の検討結果から、使用する茶葉は深煎りしていない静岡茶や鹿児島茶がより好ましい。新芽新茶と通常新茶との検討結果から、より若い新芽の方が、抽出効率、ポリアミン量ともによい傾向があるが、一般的な緑茶であれば十分ポリアミン含量の高い緑茶が調製できる。生葉、荒茶、煎茶の検討結果から、生葉はポリアミンが減少しやすく採取後、早期に加工する必要があるが、荒茶、煎茶では大差なく、使用することができる。
使用茶葉は狭山茶Bを採用し、抽出溶液、抽出条件および試験結果を表12、13に示した。
通常の緑茶中のカフェイン量は約0.20mg/ml、緑茶中のカテキン量は約0.36mg/ml程度であり、ポリアミン茶もほぼ同様の含量を有している。
塩化カルシウム抽出液では、茶殻より緑茶葉を使用したほうがカフェイン、カテキン量が多い抽出物が得られる傾向があるが、茶殻からの抽出においても、特にカテキン量は条件次第で高含有させることができる。さらに、カフェイン、EGCg量は塩化カルシウム濃度が低いほど、抽出温度が高いほど高くなる傾向が示され、粉末化した場合、カフェイン、カテキン量が多いものから少ないものまで抽出条件により調製できる。
この出願は2009年5月13日に出願された日本国特許出願番号2009−116672からの優先権を主張するものであり、その内容を引用してこの出願の一部とするものである。
Claims (10)
- 動植物素材に塩溶液を加えて抽出することを特徴とする、ポリアミン抽出物の製造方法。
- 前記塩が、塩化ナトリウム、塩化マグネシウムおよび塩化カルシウムから成る群から選択される、請求項1に記載のポリアミン抽出物の製造方法。
- 前記塩が塩化カルシウムである、請求項2に記載のポリアミン抽出物の製造方法。
- 前記動植物素材が、鮭白子、豚の肝臓、麦芽、大豆、納豆、烏龍茶葉、紅茶葉、碁石茶葉、黒豆、柿の葉、および緑茶葉から成る群から選択される請求項1乃至3に記載のポリアミン抽出物の製造方法。
- 前記動植物素材が、Camellia属の植物である茶樹の葉および/または茎からとれる緑茶葉である請求項4に記載のポリアミン抽出物の製造方法。
- 請求項1乃至5のいずれかに記載の製造方法によって製造されたことを特徴とするポリアミン抽出物を含有する飲食品。
- 請求項1乃至5のいずれかに記載の製造方法によって製造されたことを特徴とするポリアミン抽出物を、さらに精製して得られるポリアミン製剤。
- 請求項7に記載のポリアミン製剤を含有する飲食品。
- 請求項6に記載の飲食品であって、ポリアミンを一食分で0.2~500μmol含有する飲食品。
- 請求項6に記載の飲食品であって、ポリアミンを0.2~500μmol/L含有する緑茶飲料。
