Abstract
Description
α化した澱粉を常温や低温で放置すると、水分を分離し硬くなる。この現象を老化といい、澱粉の老化現象については数多く研究されている。一般に老化防止のためには温度を80℃以上に保つ、急速に乾燥させて水分を15%以下にする、pH13以上のアルカリ性に保つことが必要である。また、老化を防止する方法として澱粉含有食品に糖類(ブドウ糖、果糖、液糖等)や大豆タンパク、小麦グルテン、脂肪酸エステル、多糖類(山芋、こんにゃく等)を添加する方法が一般に知られており、特開昭59−2664号公報には増粘剤、界面活性剤等を添加する方法が記載されている。しかし、これらの方法では食味が大きく変化し、また効果も不安定で十分な解決法とはなっていない。
また、老化防止の手段として酵素を添加する方法も知られている。例えば、特開昭58−86050号公報には、精白米にアミラーゼ、プロテアーゼ、リパーゼ等の酵素と、食塩及びサイクロデキストリンを混合して炊飯する米飯の改良方法が記載されている。特開昭60−199355号公報には、炊飯後の米飯に糖化型アミラーゼ(β−アミラーゼ、グルコアミラーゼ)の水溶液を噴霧添加する米飯の老化防止方法が記載されている。
澱粉含有食品の一つである、麺類の食感改良方法に関しては多くの知見がある。すなわち、茹で麺の食感を改良するためにタンパク質素材(活性グルテン、大豆タンパク質、卵白、全卵、カゼイン等)や澱粉等(各種澱粉、多糖類、乳化剤等)を添加することが行われている(特開平2−117353号公報)。また、レトルト殺菌処理の場合に食感を維持させるために高温、短時間処理を行っている(特開平2−186954号公報)。また、トランスグルタミナーゼを使用し、食感を改善させる方法も知られている(特開平2−286054号公報、特開平6−14733号公報)。これらの方法によれば、トランスグルタミナーゼの作用によりタンパク質間及びタンパク質内のネットワーク構造を麺体の中に形成させて麺体内での水分の均一化を防止することにより、茹で後の弾力(こし)のある好ましい食感を維持することができる。しかしながら、全体が均一な食感となり、アルデンテと呼ばれる、中芯感のある食感(外側に比べ内側が硬い)を得るには改善の余地があった。
また、WO2005/096839によれば澱粉含有食品の物性改良剤として、α−グルコシダーゼを小麦混練時に添加することによって、硬さ、粘りが増し、かつ時間が経つと無添加に比べ中芯感もあるうどんを得ることができる。一定の効果が見られるものの、茹で直後での物性改良効果において改善の余地が残っていた。しかし最近、α−グルコシダーゼとトランスグルタミナーゼを適切な比率にて併用することで、茹で上げ直後の食感を向上させ、かつその優れた食感を長時間にわたって維持するという2つの両立に成功したとの報告がなされた(WO2008/001940)。かなりの効果が見られるが、好ましい食感バランスを保ちつつ強い弾力を持たせるには限界があった。
グルコースオキシダーゼの麺類への使用に関しては、グルコースオキシダーゼ及びアミラーゼ及びグルコアミラーゼを併用することによりコシが強くなるという報告があるが(特開平6−296467号公報)、上記α−グルコシダーゼやトランスグルタミナーゼとの併用に関する記載はない。また、グルコースオキシダーゼ及びグルコースの併用により斑点の発生が抑制されるとの報告があるが、食感においてはむしろ低下する傾向にある(特開平11−137197号公報)。更に、グルコースオキシダーゼにより日持ちが向上するという報告があるが(農林水産消費安全技術センター調査研究報告第19号,p95−101)、食感向上に関する記載はない。また、特開2000−60431号公報には、トランスグルタミナーゼとグルコーシオキシダーゼの併用により麺の喉ごし、歯ごたえ等の麺の食感を改良する方法が開示されている。この方法はかなりの効果があるが、「もちもち感」と強い「弾力」を同時に有する食感の麺を得るには限界があった。
本発明者等は、鋭意研究を行った結果、本発明は、α−グルコシダーゼ及びグルコースオキシダーゼ、もしくはα−グルコシダーゼ及びグルコースオキシダーゼ及びトランスグルタミナーゼを用いることにより、上記目的を達成しうることを見出し、本発明を完成するに至った。即ち、本発明は以下の通りである。
(1)α−グルコシダーゼの量が原料穀粉1g当たり1.5~300,000Uであり、グルコースオキシダーゼの量が原料穀粉1g当たり0.002~500Uかつα−グルコシダーゼ1U当たり0.00003~30Uであることを特徴とする、α−グルコシダーゼ及びグルコースオキシダーゼを用いる麺類の製造方法。
(2)α−グルコシダーゼの量が原料穀粉1g当たり3~15,000Uであり、グルコースオキシダーゼの量が原料穀粉1g当たり0.005~50Uである(1)記載の方法。
(3)グルコースオキシダーゼの量がα−グルコシダーゼ1U当たり0.00006~3Uである(1)記載の方法。
(4)さらにトランスグルタミナーゼを用いる(1)記載の方法。

(11)グルコースオキシダーゼの含有量がα−グルコシダーゼ1U当たり0.00006~3Uである(10)記載の酵素製剤。
(12)さらにトランスグルタミナーゼを有効成分として含有する(10)記載の酵素製剤。
(13)トランスグルタミナーゼの含有量がα−グルコシダーゼ1U当たり0.0000001~1Uである(12)記載の酵素製剤。
(14)トランスグルタミナーゼの含有量がα−グルコシダーゼ1U当たり0.000001~0.1Uである(12)記載の酵素製剤。
本発明により、麺類の品質を向上することができる。特に、「もちもち感」と強い「弾力」を同時に有する麺類を製造することができ、時間経過による麺類の品質劣化を抑制することができる。
本発明による麺類の製造方法及び麺類改質用の酵素製剤には、α−グルコシダーゼ及びグルコースオキシダーゼ、もしくはα−グルコシダーゼ及びグルコースオキシダーゼ及びトランスグルタミナーゼを用いる。
本発明のα−グルコシダーゼは、非還元末端α−1,4−グルコシド結合を加水分解し、α−グルコースを生成する酵素である。α−グルコシダーゼのうち、α−1,4結合をα−1,6結合へと変換する糖転移活性を有するトランスグルコシダーゼが好ましい。尚、トランスグルコシダーゼL「アマノ」という商品名で天野エンザイム(株)より市販されている酵素が、α−グルコシダーゼの一例である。
本発明のグルコースオキシダーゼは、グルコース、酸素、水を基質としてグルコン酸と過酸化水素を生成する反応を触媒する酸化酵素である。この反応により生成された過酸化水素は、タンパク中のSH基を酸化することでSS結合(ジスルフィド結合)生成を促進し、タンパク中に架橋構造を作るものと推定している。グルコースオキシダーゼは、微生物由来、植物由来のものなど種々の起源のものが知られているが、本発明で用いる酵素はこの活性を有している酵素であれば構わず、その起源としてはいずれのものでも構わない。また、組み換え酵素であっても構わない。「スミチームPGO」という商品名で新日本化学工業(株)より市販されている微生物由来のグルコースオキシダーゼが一例である。尚、グルコーシオキシダーゼにカタラーゼ製剤が混合されている市販品も多く見られるが、グルコースオキシダーゼ活性を有していれば、他の製剤との混合物であっても構わない。
本発明のトランスグルタミナーゼはタンパク質やペプチド中のグルタミン残基を供与体、リジン残基を受容体とするアシル転移反応を触媒する活性を有する酵素のことを指し、哺乳動物由来のもの、魚類由来のもの、微生物由来のものなど、種々の起源のものが知られている。本発明で用いる酵素はこの活性を有している酵素であれば構わず、その起源としてはいずれのものでも構わない。また、遺伝子組み換え酵素であっても構わない。味の素(株)より「アクティバ」TGという商品名で市販されている微生物由来のトランスグルタミナーゼが一例である。
麺類としては様々なものが考えられるが、市場の大きさや、ニーズ等と照らし合わせると、うどん、パスタ、日本そば、中華麺、焼きそば、フライ工程や乾燥工程を経る即席麺等の麺類、餃子、焼売の皮等が特に有効であると考えられる。
麺類(餃子の皮、焼売の皮等も含む)の製造において、小麦粉等の原料穀粉にα−グルコシダーゼ及びグルコースオキシダーゼ、もしくはα−グルコシダーゼ及びグルコースオキシダーゼ及びトランスグルタミナーゼを作用させる場合は、麺類製造工程のどの段階で作用させてもかまわない。すなわち原料混合時に酵素を添加してもよいし、混合後に酵素を振りかけて作用させてもよい。トランスグルタミナーゼ、α−グルコシダーゼ、グルコースオキシダーゼを麺類に作用させる順序は特に問わず、いずれかの1種もしくは2種の酵素を先に作用させた後、残りの酵素を作用させてもよいが、3種を同時に作用させるのが好ましい。さらに、これらの酵素以外の他の酵素や物質(デキストリン、澱粉、加工澱粉等の糖類、畜肉エキス等の調味料、植物蛋白、グルテン、卵白、ゼラチン、カゼイン等の蛋白質、蛋白加水分解物、蛋白部分分解物、乳化剤、クエン酸塩、重合リン酸塩等のキレート剤、グルタチオン、システイン等の還元剤、アルギン酸、かんすい、色素、酸味料、香料等その他の食品添加物等)と併用し使用してもかまわない。
原料穀粉としては小麦粉、米粉、大麦粉、ライ麦粉が挙げられる。小麦粉を用いる場合はどのような品種の小麦粉でもよく、強力粉、準強力粉、中力粉、薄力粉、デュラムセモリナ粉でもかまわない。また、米粉等の小麦粉以外の穀粉、澱粉(加工澱粉を含む)と混合して使用してもかまわない。
本発明において、α−グルコシダーゼの添加量は、原料穀粉1gに対して酵素活性が1.5U以上、好ましくは1.5~300,000U、より好ましくは3~15,000Uの範囲が適正である。尚、α−グルコシダーゼの酵素活性については1mMα−メチル−D−グルコシド1mlに0.02M酢酸バッファー(pH5.0)1mlを加え、酵素溶液0.5ml添加して、40℃、60分間を作用させた時に、反応液2.5ml中に1μgのブドウ糖を生成する酵素量を1U(ユニット)と定義した。
本発明において、グルコースオキシダーゼの添加量は、原料穀粉1gに対して酵素活性が0.001U以上、好ましくは0.002~500U、より好ましくは0.005~50Uの範囲が適正である。更に、グルコースオキシダーゼの添加量がα−グルコシダーゼ1U当たり0.00003~30U、好ましくは0.00006~3Uとなるように添加するのが望ましい。尚、グルコースオキシダーゼの酵素活性については、グルコースを基質として、酸素存在下でグルコースオキシダーゼを作用させることで過酸化水素を生成させ、生成した過酸化水素にアミノアンチピリン及びフェノール存在下でペルオキシダーゼを作用させることで生成したキノンイミン色素が呈する色調を、波長500nmで測定し定量する。1分間に1μmolのグルコースを酸化するのに必要な酵素量を1U(ユニット)と定義した。
本発明において、トランスグルタミナーゼの添加量は、穀粉1gに対して酵素活性が0.0001~100U、好ましくは0.0001~10Uの範囲が適正である。更に、トランスグルタミナーゼの添加量がα−グルコシダーゼ1U当たり0.0000001~1U、好ましくは0.000001~0.1Uとなるように添加するのが望ましい。尚、トランスグルタミナーゼの酵素活性についてはベンジルオキシカルボニル−L−グルタミニルグリシンとヒドロキシルアミンを基質として反応を行い、生成したヒドロキサム酸をトリクロロ酢酸存在下で鉄錯体を形成させた後525nmの吸光度を測定し、ヒドロキサム酸の量を検量線より求め活性を算出する。37℃、pH6.0で1分間に1μmolのヒドロキサム酸を生成する酵素量を1U(ユニット)と定義した。
繰り返しになるが、α−グルコシダーゼ及びグルコースオキシダーゼ、もしくはα−グルコシダーゼ及びグルコースオキシダーゼ及びトランスグルタミナーゼを作用させて麺類の製造をする場合の各酵素の添加量比は、酵素活性(ユニット数)において、グルコースオキシダーゼがα−グルコシダーゼ1U当たり0.00003~30U、好ましくは0.00006~3Uが適当であり、トランスグルタミナーゼがα−グルコシダーゼ1U当たり0.0000001~1U、好ましくは0.000001~0.1Uが適当である。パスタの場合、グルコースオキシダーゼの添加量は、α−グルコシダーゼ1U当たり0.0006~3Uが特に好ましく、トランスグルタミナーゼの添加量はα−グルコシダーゼ1U当たり0.000001~0.1Uが特に好ましい。うどんの場合、グルコースオキシダーゼの添加量は、α−グルコシダーゼ1U当たり0.00006~0.3Uが特に好ましく、トランスグルタミナーゼの添加量はα−グルコシダーゼ1U当たり0.000001~0.1Uが特に好ましい。各酵素の添加量比が上記の範囲にあるとき、もちもち感と強い弾力を同時に有する好ましい食感が得られ、時間経過による品質劣化の抑制された麺類を製造することができる。また、ゆでた際のゆで汁濁度も低減することができる。
各酵素の反応時間は、酵素が基質物質に作用することが可能な時間であれば特に構わず、非常に短い時間でも逆に長時間作用させても構わないが、現実的な作用時間としては5分~24時間が好ましい。また、反応温度に関しても酵素が活性を保つ範囲であればどの温度であっても構わないが、現実的な温度としては0~80℃で作用させることが好ましい。すなわち、通常の製麺工程を経ることで十分な反応時間が得られる。
α−グルコシダーゼ、グルコースオキシダーゼ、トランスグルタミナーゼにデキストリン、澱粉、加工澱粉等の賦形剤、畜肉エキス等の調味料、植物蛋白、グルテン、卵白、ゼラチン、カゼイン等の蛋白質、蛋白加水分解物、蛋白部分分解物、乳化剤、クエン酸塩、重合リン酸塩等のキレート剤、グルタチオン、システイン等の還元剤、アルギン酸、かんすい、色素、酸味料、香料等その他の食品添加物等を混合することにより、麺類改質用の酵素製剤を得ることができる。本発明の酵素製剤は液体状、ペースト状、顆粒状、粉末状のいずれの形態でも構わない。また、酵素製剤における各酵素の配合量は0%より多く、100%より少ないが、トランスグルタミナーゼの場合、配合量は0%でも構わない。
表3の試験区1~9に示す通り、AG1U当たりのGO添加量が0.00003~30Uにおいて、もちもち感と強い弾力を併せ持つ好ましい食感すなわち官能評価結果△以上が得られた。同じく試験区10~18に示す通り、AG1U当たりのTG添加量が1U以下において好ましい食感が得られた。また、少なくとも0.0000001U以上にて、好ましい食感が得られることが確認された。同じく試験区19~27に示す通り、原料穀粉1gに対するAG活性が1.5~300000U、原料穀粉1gに対するGO活性が0.002~500Uにおいて、好ましい食感が得られた。同じく試験区28~35に示す通り、原料穀粉1gに対するTG活性が100U以下において、好ましい食感が得られた。また、少なくとも0.0001U以上にて、好ましい食感が得られることが確認された。一方、上記範囲外の試験区においては、各酵素の効果が弱すぎるもしくは強すぎることにより全体としての食感バランスが崩れてしまい、好ましい効果は得られなかった。
以上より、麺類にAG及びGO、もしくはAG及びTG及びGOを併用する場合、原料穀粉1gに対するAG添加量は1.5~300000U、原料穀粉1gに対するGO添加量は0.002~500U、原料穀粉1gに対するTG添加量は0.0001~100U、AG1U当たりのGO添加量は0.00003~30U、AG1U当たりのTG添加量は0.0000001~1Uにて好ましい食感が得られることが確認された。
表5に示す通り、GOのみ添加した場合、GOにより強い弾力が付与されるものの、やきそばの好ましい食感において重要とされるもちもち感が全く付与されなかった。また、AGのみ添加した場合、AGによりもちもち感および弾力が付与されるものの、付与される弾力の強度が若干弱かった。一方、AG及びGO、もしくはAG及びTG及びGOを併用した試験区では、もちもち感と強い弾力が共に付与された。以上のように、AG及びGO、もしくはAG及びTG及びGOを併用することで、もちもち感と強い弾力を併せ持つ好ましい食感のやきそばを製造できることが示された。
比較例
特開平6−296467号公報には、グルコースオキシダーゼ及びアミラーゼ(AM)及びグルコアミラーゼ(GA)を併用することによりコシが強くなると記載されているため、本発明との比較を行なう目的で、実施例2と同様の原料、同様の方法にてうどんを作製し、官能評価を行った。官能評価は、もちもち感、弾力に関して、コントロール区分を0点とし、−2点から2点までの評点法にて評価人数4人で行った。結果を表7に示す。評点は、0.5点が「差あり」、1点が「顕著な差あり」、2点が「非常に顕著な差あり」とした。尚、うどんの好ましい食感として、もちもち感と強い弾力を共に有することが重要とされるため、もちもち感が付与された(評点が0点より大きい)試験区に「*」、弾力が顕著に付与された(評点が1点以上)試験区に「*」を記した。試験区分は、表7に示す通りである。尚、α−アミラーゼはアミラーゼAD「アマノ」1(天野エンザイム社製)、グルコアミラーゼはグルクザイムAF6(天野エンザイム社製)を用いた。表7の試験区1~3に設定したα−アミラーゼ、グルコアミラーゼおよびGOの添加量は、特開平6−296467号公報の実施例1、比較例2、比較例3に従った。
Claims (14)
- α−グルコシダーゼの量が原料穀粉1g当たり1.5~300,000Uであり、グルコースオキシダーゼの量が原料穀粉1g当たり0.002~500Uかつα−グルコシダーゼ1U当たり0.00003~30Uであることを特徴とする、α−グルコシダーゼ及びグルコースオキシダーゼを用いる麺類の製造方法。
- α−グルコシダーゼの量が原料穀粉1g当たり3~15,000Uであり、グルコースオキシダーゼの量が原料穀粉1g当たり0.005~50Uである請求の範囲第1項記載の方法。
- グルコースオキシダーゼの量がα−グルコシダーゼ1U当たり0.00006~3Uである請求の範囲第1項記載の方法。
- さらにトランスグルタミナーゼを用いる請求の範囲第1項記載の方法。
- トランスグルタミナーゼの量が原料穀粉1g当たり0.0001~100Uである請求の範囲第4項記載の方法。
- トランスグルタミナーゼの量が原料穀粉1g当たり0.0001~10Uである請求の範囲第4項記載の方法。
- トランスグルタミナーゼの量がα−グルコシダーゼ1U当たり0.0000001~1Uである請求の範囲第4項記載の方法。
- グルコースオキシダーゼの量がα−グルコシダーゼ1U当たり0.00006~3Uであり、トランスグルタミナーゼの添加量がα−グルコシダーゼ1U当たり0.000001~0.1Uである請求の範囲第4項記載の方法。
