Abstract
Винахід належить до способу виявлення та кількісного визначення молекули нуклеїнової кислоти, що становить інтерес, в реакції ампліфікації із внесенням одноланцюгових розривів і добудовою, причому спосіб включає приведення цільової молекули нуклеїнової кислоти в контакт у практично ізотермічних умовах з полімеразою, двома або більше олігонуклеотидними праймерами, де кожний з вказаних олігонуклеотидних праймерів містить у напрямку 5'-3' послідовність розпізнавання для ферменту, що вносить одноланцюговий розрив, послідовність, комплементарну цільовій молекулі нуклеїнової кислоти та один або більше нуклеотидів з 2'-О-метильною модифікацією розташованих на 3'-кінці послідовності комплементарної цільовій молекулі нуклеїнової кислоти; фермент, що вносить одноланцюговий розрив, який зв'язується з послідовністю розпізнавання для ферменту, що вносить одноланцюговий розрив, у дволанцюговій ДНК та розщеплює один ланцюг дволанцюгової ДНК в сайті одноланцюгового розриву; утворення ампліконів, які містять щонайменше частину зазначеної цільової молекули нуклеїнової кислоти; виявлення сигналу, специфічного для гібридизації олігонуклеотидного зонда з цільовою молекулою нуклеїнової кислоти або її ампліконом, де сигнал указує на кількість цільової молекули нуклеїнової кислоти присутньої у зразку, або її амплікону.
