200803752 (1) 九、發明說明 【發明所屬之技術領域】 本發明係關於含吸濕性粉末原料及芝麻素類之飲食品 組成物,更詳細而言,係關於藉由混合含有吸濕性粉末原 料及芝麻素類,上述吸濕性粉末原料之吸濕性被改善之飮 食品用粉末組成物及含有該組成物之飮食品。 【先前技術】 關於粉末原料’於製造中及/或貯藏中之吸濕經常成 爲問題。吸濕性程度係依原料而有差異,最嚴重時,粉末 吸收水至液狀化(潮解)。於製造中發生吸濕時,發生流 動特性改變、黏著性增加、凝聚發生、離型性降低之問題 ’不僅操作變得困難,亦有化學安定性降低,原料或製品 之組成改變。另外,於貯藏中發生吸濕時,不僅外觀劣化 ’亦有含有活性成份之化學安定性降低或因結晶形態變化 而溶解特性變化等之問題發生。如此地製造時或使用時之 操作困難之吸濕性粉末原料,例如有機酸或無機酸之鹽類 、無機鹽類及藥劑等所使用之化合物原料等,提出各種用 以解決該吸濕性或潮解性之方法。 揭示例如添加粉末狀的油脂及碳酸鈣於粉狀物中以防 止潮解及結塊的方法(專利文獻1 ),添加蔗糖脂肪酸酯 以防止結塊的方法(專利文獻2 ),使吸附性物質吸附潮 解性物質之防止潮解的方法(專利文獻3 ),以腸溶性皮 膜劑被覆具有吸濕性之粉末原料之粉末改良品(專利文獻 -5- 200803752 4 ),由改變結晶形態以延緩潮解性之方法(專利文獻5 )等。 然而,天然存在之左旋肉鹼係特殊胺基酸中之一種, 已知爲用以燃燒體內脂肪,改變成能量之必要而不可或缺 的營養素,由消耗多餘的脂肪,可期待預防生活習慣、消 除疲勞或提昇體力、提昇運動或日常生活中表現、提昇基 礎代謝等之效果。體內存在之左旋肉鹼量,因爲於二十多 歲爲高峰,隨著年齢增加,逐漸減少而不足,所以以適當 地攝取左旋肉鹼爲目的,近年來使用於營養補助食品、飮 食療法用製品、健康食品等之飲食品或飲食品組成物。然 而,已知左旋肉鹼及該衍生物具有極高的吸濕性,形成下 式:

如上所述,以左旋肉鹼爲首之各種吸濕性粉末原料, 提出改善該吸濕性及潮解性之方法。然而,並未存在可容 易適用,而且滿足防止或抑制吸濕性之方法。另外,由添 加劑改善吸濕性時,亦有作爲該添加劑之安全性及副作用 ,以及飲食物使用時之香味等之問題。進一步’改變結晶 形態或鹽之形態以改善吸濕性時,亦包含許多必須有纟頁_ 的製造方法等之製造上的問題者。例如,上述專利文獻9 中記載之組成物時,以90%的乙醇水煮沸L ( + )酒石酸 ,爲添加左旋肉鹼分子內鹽於其中,必須於高溫、高壓T (4) (4)200803752 濃縮大量的含肉鹼鹽溶液,不僅伴隨著明顯的能量浪費, 使用有機溶劑時亦伴隨著成本增加、溶劑的回收使用、環 境污染及有毒廢棄物之廢棄處理等之嚴重問題。 因此,本發明之課題係提供藉由工業上容易使用,且 生產成本亦低之方法,吸濕性粉末原料,尤其肉鹼或該鹽 或該衍生物之吸濕性被改善之含有吸濕性粉末原料之飲食 品組成物。 課題之解決手段 本發明者爲解決上述課題,努力硏究的結果,混合粉 末之芝麻素類於吸濕性粉末原料,上述吸濕性粉末原料之 吸濕性被改善,尤其吸濕性粉末原料爲肉鹼或該鹽或該衍 生物時,發現該吸濕性明顯地被改善。接著,因爲粉末狀 之芝麻素類無臭無味,所以確認容易適用飲食品,完成本 發明。 亦即’本發明係提供含吸濕性粉末原料及芝麻素類, 以粉末芝麻素類爲宜之飮食品組成物,飲食品粉末組成物 爲宜。 本發明係提供混合、含有吸濕性粉末原料及芝麻素類 之飲食品粉末組成物,上述吸濕性粉末原料之吸濕性被改 善之上述組成物。 本發明又提供上述吸濕性粉末原料係肉鹼或該鹽或該 衍生物之上述組成物。本發明又提供上述芝麻素類係芝麻 素及/或表芝麻素,更特定時,芝麻素及表芝麻素之上述 -8- (5) (5)200803752 組成物。 進一步,本發明係含有上述組成物之飲食品。 用以實施發明之最佳型態 本說明書中,所謂「吸濕性」係指對象物暴露於空氣 中時,捕捉大氣中水蒸氣之性質,特別稱爲潮解性,捕捉 大氣中水蒸氣而溶解於該水之性質亦包含於本說明書中之 「吸濕性」。另外,吸濕性粉末原料之「吸濕性改善」, 係指與未處理之吸濕性粉末原料相比較,吸濕性降低,防 止潮解及/或結塊,或抑制潮解及/或結塊。 吸濕性係例如後述之實施例記載,使用該業者已知手 法,可測定及/或評估。具體上,於室溫或高溫高濕度下 等之一定條件下’將測量的原料,放置適當的時間後,可 藉由目測觀察變化(確認原料表面的水份、確認固體的消 失、確認粉末的流動性或結塊化程度、確認變色等)、或 測量重量增加分(吸濕水量)檢討等之手法。 本說明書中,所謂「吸濕性粉末原料」係指所需製品 之製造中及/或保存中等,只要上述之吸濕性成爲問題之 飮食品用或醫藥用所容許之物質皆可,並無限制。本說明 書中,「吸濕性成爲問題」原料係例如本說明書中之有關 吸濕性之記載爲參考,若相關業者應可容易判斷,具體上 係有關於製造上或製品之貯藏中發生之吸濕。尤其測定製 品之貯藏安定性用之加速試驗所使用之溫度爲4 0 °C,相 對濕度爲7 5 %之條件下顯示吸濕(或潮解)之原料。作爲 (6) 200803752 吸濕性粉末原料之具體例,可舉例如偏磷酸鈉、焦磷酸鉀 、醋酸鈉、明礬胺(ammonium alum )等之有機酸或無機 酸之鹽類、氯化鎂、氯化鈣等之無機鹽類、木寡醣等之糖 類,進一步如帝拔癲(sodium valproate)、甲磺司特( Suplatast Tosilate )、膽驗之鹽、卡巴拉汀 ( Rivastigmine )、溴斯的明(Pyridostigmine Bromide)、 巴氨西林(bacampicillin )、左旋肉鹼及其衍生物、 Moricizine Hydrochloride、及白背櫟萃取物等之生藥萃取 物等之藥劑所使用之活性成份。 本發明之吸濕性粉末原料係包含於飮食品組成物,所 以若爲具有相同的生理活性之原料時,以使用香味問題少 之吸濕性粉末原料爲宜。例如吸濕性原料爲左旋肉鹼或該 鹽或該衍生物時,使用無臭無味之左旋肉鹼比使用酸味強 的左旋肉鹼酒石酸鹽爲宜。 本說明書中,所謂肉鹼或該鹽或該衍生物亦包含肉鹼 [(CH3)3N+CH2CH(OH)CH2CO〇-、7 — 三甲基銨一沒—羥 基丁酸]、該異構物、該鹽、及肉鹼、該異構物及該鹽之 衍生物(例如生物體內轉換成肉鹼之衍生物)。包含如肉 鹼、左旋肉鹼、氯化肉鹼(右旋左旋體、左旋體)、左旋 肉鹼酒石酸鹽、左旋肉鹼馬來酸鹽、左旋肉鹼富馬酸鹽、 左旋肉驗硫酸鹽、左旋肉驗草酸鹽、左旋肉驗鎂鹽、醯基 左旋肉鹼、乙醯基左旋肉鹼、丙醯基左旋肉鹼、乳清酸肉 鹼等。此等係對該製作方法不做任何限定,天然萃取物、 微生物發酵品、合成品中之任一種皆可,以天然存在之左 -10- (7) (7)200803752 旋肉鹼或該鹽或該衍生物爲宜,例如下式:
OR (式中,R係Η或Q至C5之低級鏈烷醯基(alkanoyi ) ° ) 所表示。另外,關於本發明書中之肉鹼之說明係合乎肉鹼 的鹽、肉鹼或該鹽之衍生物。 本發明書中’ 「芝麻素類」係包含芝麻素類及該類似 體’作爲具體例,可舉例如芝麻素、芝麻醇(sesaminol )、表芝麻醇、芝麻素酚(sesamolin )、表芝麻素( episesamin)、二氧雙環[3.3.0]辛烷衍生物(例如特開平 4 一 93 3 1號記載)等。其中,本發明中係以芝麻素、表芝 麻素爲宜。進一步,「芝麻素類」係包含芝麻素及該類似 體之糖苷以及此等之代謝物,只要適合提供本發明之飲食 品組成物’該任一種皆可含於本發明之飲食品組成物。 本發明所使用之芝麻素類係只要爲固體狀,對於製造 方法等無任何限制。例如使用芝麻素及/或表芝麻素作爲 芝麻素類時,通常係可使用由已知方法(例如加入熱甲醇 於芝麻油萃取,除去甲醇後,加入丙酮於殘渣中萃取之方 法(記載於特開平4 一 9 3 3 1號公報))自芝麻油萃取之芝 麻素(亦稱爲芝麻素萃取物或芝麻素精製物)。成爲原料 之芝麻油並無特別的限制,但使含於本發明之飮食品組成 物時,因爲有時芝麻油特有的風味,於官能評價上並不喜 好,所以以使用無臭無味之芝麻油所萃取之芝麻素萃取物 -11 - (8) (8)200803752 或芝麻素精製物爲宜。另外,使用精製度低的芝麻素類時 ’若配合後述之適合量之芝麻素時,所得之飲食品組成物 或飲食品之體積過大,發生不適合攝取之情況。尤其,製 劑化成經口投予用時,製劑(錠劑、膠囊等)變大,發生 攝取障礙。因此,就攝取量少爲宜之觀點,以使用芝麻素 精製物爲宜。 本發明之芝麻素類,就關於改善吸濕性粉末原料之吸 濕性之觀點,以粉末芝麻素類爲宜。該粉末芝麻素類之粒 徑雖依與吸濕性粉末原料之混合方法(均質性)而定,但 通常爲JIS標準篩10至250網目,以60至200網目之範 圍爲宜。 本發明之飲食品組成物中與芝麻素類一同含有之吸濕 性粉末原料係具有增進健康等之作用時,上述芝麻素類, 尤其以芝麻素及/或表芝麻素爲宜。因爲認爲此等芝麻素 類之各種生理作用係可與上述吸濕性粉末原料之作用相加 或加乘地發揮。例如吸濕性粉末原料係肉鹼時,提昇肉鹼 能量代謝之作用、提昇運動性能之作用係芝麻素及表芝麻 素具有之生理作用之一 ’認爲可相加或加乘地發揮提昇能 量代謝(Biosci. Biotechnol. Biochem·,69,179-188,2005 )、或提昇運動能作用(Int· J. Sports Med.,24,530-534, 2003,Biofactors,21,191-196,2004 ),將可提供優異的 健康食品。 本發明之飲食品組成物係含吸濕性粉末原料及芝麻素 類,可由混合吸濕性粉末原料及芝麻素類以及因應所需之 -12- 200803752 (9) 其他添加劑而得。混合手法並無任何限制,只要可均質地 混合,任何手法皆可。使用一般已知之混合機,例如水平 圓筒型混合機、V型混合機、雙重圓錐型混合機、立方體 型混合機、螺旋型混合機、螺帶型混合機、介質攪拌型硏 磨機、高速旋轉磨碎·衝擊硏磨機、噴射式硏磨機等之空 氣力學的粉碎器等混合、可使用以硏缽或球磨機等粉碎混 合、利用聚乙嫌(P E )製、尼龍製或依該性質而成的袋攪 拌混合等。就操作容易,無須特別機械之觀點,可使用利 用袋之攪拌混合爲宜。吸濕性粉末原料及芝麻素類之混合 程度高時,可得到更高的吸濕性改善效果。 本發明之飮食品組成物,芝麻素類之配合量係依一同 所含有之吸濕性原料之種類或其他添加劑之有無等而異, 相對於100份之吸濕性原料,作爲重量比係以0.00丨至 1〇〇之芝麻素類,0.01至10程度爲適當的。 本發明之飮食品組成物係可直接作爲後述之飮食品使 用’亦可使用該業者已知的手法,使適當含於該飲食品而 使用。本發明之飲食品組成物因爲含有該吸濕性被改善狀 態之吸濕性粉末原料,所以化學上安定,適合於貯藏或製 造飮食品等。 本說明書中,所謂飮食品係包含調味料、營養補助食 品、健康食品、飲食療法食品、綜合健康食品、營養補給 品及飲料以及經口投予醫藥品。本發明之飲食品係可爲固 體狀(例如結晶、膠囊、锭劑、粉末)、半固體狀(例如 凝膠、糊狀物)、液狀(例如礦泉水、清涼飲料水、果實 -13- (10) (10)200803752 飲料、運動飲料、酒類)。本發明之飲食品係以錠劑、顆 粒品、飲食前溶解於液體之錠劑或顆粒品爲宜。 本發明之飲食品係可使用該業者已知之手法,可使含 有本發明之飲食品組成物而製造。本發明之飮食品係可期 待安定的吸濕性粉末原料及芝麻素的所需效果。 本發明之飲食品中吸濕性粉末原料及芝麻素之配合量 並無特別的限制,以此等適合之1日攝取量作爲參考,若 爲該業者’因應該飮食品之攝取形態,可適當地設定配合 本發明之飲食品組成物係可因應需要,含有上述吸濕 性粉末原料及芝麻素類以外之任意添加劑。另外,本發明 之飲食品係可含有上述本發明之飲食品組成物以外之通常 飲食品所使用之任意成份。作爲此等添加劑及/或成份, 可舉例如維生素E、維生素C等之維生素類、糖類、賦形 劑、崩壞劑、結合劑、潤澤劑、乳化劑、緊張化劑(等張 化劑)、緩衝劑、助溶劑、防腐劑、安定化劑、抗氧化劑 、著色劑、矯味劑、香料、凝固劑、pH調整劑、增黏劑 、萃取物粉末、生藥、無機鹽等。 另外,本發明之飲食品組成物或飲食品可表示該具體 之用途(例如補給營養用、維持健康用、燃燒脂肪用、預 防肥胖用、維持瘦身後體型用等)及/或此等之具體用法 (例如攝取量、攝取次數、攝取方法等)。 【實施方式】 -14- (11) (11)200803752 以下係基於實施例說明本發明,但本發明並不侷限於 此等實施例者。 實施例1 肉鹼之吸濕性改善 使用肉鹼作爲吸濕性原料,對於藉由混合芝麻素類, 改善肉鹼之吸濕性,進行檢討。 實驗材料 使用左旋肉鹼結晶性粉末(三榮源FFI股份有限公司 ’左旋肉鹼結晶性粉末)作爲吸濕性粉末原料。使用經精 製之粉末狀之芝麻素及表芝麻素之混合物(芝麻素:表芝 麻素=51·1 : 48.2 (重量比))作爲芝麻素類。 另外,作爲比較例係取代芝麻素類,使用如下述之製 劑化中所經常使用之添加劑,對於改善肉鹼之吸濕性,進 行檢討。 比較例1 :環狀糊精類(α — CD/冷—CD/γ — CD)(鹽 水港精製股份有限公司,Dexypala — 100 /石一 ΐ〇〇/γ — 100 ) 比較例2 : Si〇2 (富士 Sili sea化學股份有限公司, sylopage) 比較例3 :殿粉(日澱化學股份有限公司,perfiler— ι〇2 ) 比較例4 :水溶性食物纖維(太陽化學股份有限公司, Sunfiber ) -15- 200803752 (12) 比較例5 ··蔗糖脂肪酸酯(第一工業製藥股份有限公司, DK 酯 F - 1 〇) 實驗方法 1 *於培養皿(40必)中,秤量各200mg之肉鹼或以 1 : 1混合肉鹼與芝麻素類(使用PE製塑膠袋混合粉體) 之粉末。 2 ·培養皿加蓋之狀態下,靜置於(A )室溫(2 0 °C )、(B)培養箱(40°C,75%RH),目測觀察12小時後 之性狀變化。 3 .分別使用比較例1至5之粉末以取代芝麻素類, 同樣地進行檢討。 結果 圖1係表示混合芝麻素類於肉鹼時之改善肉鹼吸濕性 之結果。單獨肉鹼時吸濕性高,不僅於高溫高濕度下(40 °C,7 5 %RH ),即使於室溫下亦發生潮解(吸濕)(圖1 中(b ) ( c ))。相對於此,等量混合芝麻素類及肉鹼時 ’於室溫下,高溫高濕度下’混合物仍爲粉末狀態(圖1 中(e ) ( f ))。由此顯示芝麻素類具有吸濕性原料之吸 濕性(潮解性)改善作用。另外,改變肉鹼及芝麻素之混 合條件時’顯不混合比率高者吸濕性改善效果高(未表示 實驗結果)。 圖2係表示混合比較例1至5之各添加劑於肉鹼時之 -16- (13) (13)200803752 改善肉鹼吸濕性之結果。認爲環狀糊精類(比較例1、圖 2 中(Ια) 、 (1/3) 、 (lr) 、Si02 (比較例 2、圖 2 中(2 ))無吸濕性改善作用。認爲澱粉(比較例3、圖2 中(3 ))、水溶性食物纖維(比較例4、圖2中(4 )) 部份潮解。認爲蔗糖脂肪酸酯(比較例5、圖2中(5 ) )改善吸濕性。 由以上結果顯示芝麻素類或蔗糖脂肪酸酯係對於改善 肉鹼之吸濕性有效。芝麻素類係無臭無味,認爲特別適合 使用。 實施例2 各種吸濕性粉末原料之吸濕性改善 使用作爲吸濕性粉末原料之左旋肉鹼結晶性粉末(三 榮源F F I股份有限公司,左旋肉鹼左旋酒石酸鹽;5、雖左 旋肉鹼吸濕性被改善,但仍具有吸濕性)、MgCl2 (富田 製藥股份有限公司,氯化鎂S)、木寡糖(Suntory股份 有限公司,木寡糖95P)。以與實施例1相同步驟,檢討 芝麻素類對此等濕性粉末原料之吸濕性改善作用。該結果 係可確認對於任一種吸濕性粉末原料,吸濕性被改善。 實施例3 製劑例 於下述之製造例中’芝麻素類及肉鹼係使用實施例1 記載者。 (製劑例1 )顆粒劑 -17- (14) (14)200803752 芝麻素類 0.0 1 g 肉驗 1 〇〇g 生育酚乙酸酯 〇.25g 矽酸酐 2 0.5 g 玉米澱粉 79.0g 將上述粉體均勻混合後,加入100ml之10%之羥丙基 纖維素·乙醇溶液,如常法混練、押出、乾燥而得顆粒劑 (製劑例2 )錠劑 芝麻素類 O.Olg 肉驗 l〇〇g 澱粉 172g 蔗糖脂肪酸酯 9.0g 氧化石夕 9.0g 將此等混合,以單發式打錠機打錠,製造直徑爲 9mm,重量爲300mg之錠劑。 (製劑例3 )膠囊劑 果膠 6 0.0% 甘油 3 0.0% 對羥基苯甲酸甲酯 0.15% 對羥基苯甲酸丙酯 0.51% 水 適量 -18- (15) (15)200803752 由上述成份所形成之軟膠囊劑皮之中,由常法充塡如 下所示之組成物,得到1粒20〇g之軟膠囊。 芝麻素類 1 O.Omg 肉鹼 1 OOmg 甘油脂肪酸酯 1 5 · Omg 蜜蠟 1 5 . Omg 小麥胚芽油 1 60mg (製劑例4 ) 口服劑 呈味·· 右旋左旋酒石酸鈉 〇.lg 琥珀酸 0.009g 甘味: 液糖 800g 酸味: 檸檬酸 12g 維生素:維生素c 10g 芝麻素類 1 g 肉鹼 10g 維生素E 3 〇g 環狀糊精 5g 香料 15 ml 氯化鉀 1 g 硫酸鎂 〇.5g 配合上述成份,加水成1 0 L。 1 0 0 m 1 〇 此口服劑係每次飲用約 -19- (16) (16)200803752 (製劑例5 )錠劑 調製配合左旋肉鹼衍生物之以維持健康及/或維持痩 身後體型爲目的之健康食品(錠劑)。使用左旋肉鹼左旋 酒石酸鹽(Lonza Japan股份有限公司)作爲左旋肉鹼衍 生物,配合如下述所示含量(每一粒)之作爲其他功能性 成份之芝麻素類(竹本油脂股份有限公司)、荷葉萃取粉 末(松浦樂業股份有限公司)、辣椒素(capsaicin)(日 本新藥股份有限公司)、咖啡因(白鳥製藥股份有限公司 )、大花紫薇(Ban aba )葉萃取粉末(常盤植物化學硏究 所股份有限公司)及維生素B 1 (田邊製藥股份有限公司 )。另外,混合作爲賦型劑之蔗糖脂肪酸酯、纖維素、二 氧化矽、澱粉、乳糖,以打錠劑製造每一粒爲直徑8mm ,重量25 Omg之錠劑。確認製造中無吸濕或潮解之問題 ,以及所調製之錠劑於貯藏中未發生吸濕或潮解之問題。 左旋肉鹼左旋酒石酸鹽 7 4.3 m g 芝麻素類 1 · 7mg 荷葉萃取粉末 1 6.7 m g 辣椒素 4.2 m g 咖啡因 1 6.7 m g 大花紫薇葉萃取粉末 1 6.7 m g 維生素B1 0.1 8 m g 實施例4 香味試驗 於下述製造例中’芝麻素類及肉驗係使用實施例1記 -20- 200803752 (17) 載者,左旋肉鹼左旋酒石酸鹽係使用實施例2記載者。1 3 名品評員舔嚐粉末狀態之等量混合芝麻素類及肉鹼者(P )、及左旋肉鹼左旋酒石酸鹽(Q ),進行(1 )甜味、 (2 )刺激、(3 ) 口中殘留味道、(4 )難以食用、(5 ) 喜歡何者之5個詢問。然而,對於(1 )至(4 )之詢問, 選擇1.感覺強烈、2.稍微感覺到、3·無特別感覺之3種回 答,對於(5 )之詢問,p、Q二者中擇一。 該結果如表1所示,可知喜歡P比Q多(表i中的 數係橫軸記載分別對於P、Q,選擇各回答之人數)。亦 即’與由傳統的步驟使改善肉鹼吸濕性之肉鹼衍生物之左 旋肉鹼左旋酒石酸鹽(Q )相比較,可確認等量混合芝麻 素類及肉鹼之本發明組成物(p ) ( 1 )稍有甜味、(2 ) 刺激弱、(3 ) 口中未感到殘留味道、(4 )容易食用、( 5 )受喜愛,判定本發明之組成物係作爲飮食品組成物, 就香味方面,亦爲優異者。 表1⑴ ⑵
