Description
本發明係關於含吸濕性粉末原料及芝麻素類之飲食品組成物,更詳細而言,係關於藉由混合含有吸濕性粉末原料及芝麻素類,上述吸濕性粉末原料之吸濕性被改善之飲食品用粉末組成物及含有該組成物之飲食品。
關於粉末原料,於製造中及/或貯藏中之吸濕經常成為問題。吸濕性程度係依原料而有差異,最嚴重時,粉末吸收水至液狀化(潮解)。於製造中發生吸濕時,發生流動特性改變、黏著性增加、凝聚發生、離型性降低之問題,不僅操作變得困難,亦有化學安定性降低,原料或製品之組成改變。另外,於貯藏中發生吸濕時,不僅外觀劣化,亦有含有活性成份之化學安定性降低或因結晶形態變化而溶解特性變化等之問題發生。如此地製造時或使用時之操作困難之吸濕性粉末原料,例如有機酸或無機酸之鹽類、無機鹽類及藥劑等所使用之化合物原料等,提出各種用以解決該吸濕性或潮解性之方法。
揭示例如添加粉末狀的油脂及碳酸鈣於粉狀物中以防止潮解及結塊的方法(專利文獻1),添加蔗糖脂肪酸酯以防止結塊的方法(專利文獻2),使吸附性物質吸附潮解性物質之防止潮解的方法(專利文獻3),以腸溶性皮膜劑被覆具有吸濕性之粉末原料之粉末改良品(專利文獻4),由改變結晶形態以延緩潮解性之方法(專利文獻5)等。
然而,天然存在之左旋肉鹼係特殊胺基酸中之一種,已知為用以燃燒體內脂肪,改變成能量之必要而不可或缺的營養素,由消耗多餘的脂肪,可期待預防生活習慣、消除疲勞或提昇體力、提昇運動或日常生活中表現、提昇基礎代謝等之效果。體內存在之左旋肉鹼量,因為於二十多歲為高峰,隨著年齡增加,逐漸減少而不足,所以以適當地攝取左旋肉鹼為目的,近年來使用於營養補助食品、飲食療法用製品、健康食品等之飲食品或飲食品組成物。然而,已知左旋肉鹼及該衍生物具有極高的吸濕性,形成下式:

左旋肉鹼吸濕時,變化成泥狀或糊狀,以及因化學安定性降低,所以三甲基胺游離,成為不愉快的魚臭味。由此等理由,對於左旋肉鹼或該鹽或該衍生物,提出降低吸濕性之方法。例如吸濕低之乳清酸肉鹼之製造方法(專利文獻6)、使用L(-)肉鹼之馬來酸及富馬酸鹽(專利文獻7)、L(-)肉鹼之硫酸鹽及草酸鹽(專利文獻8)、使用L(-)肉鹼L(+)酒石酸(2:1)(專利文獻9)等。

如上所述,以左旋肉鹼為首之各種吸濕性粉末原料,提出改善該吸濕性及潮解性之方法。然而,並未存在可容易適用,而且滿足防止或抑制吸濕性之方法。另外,由添加劑改善吸濕性時,亦有作為該添加劑之安全性及副作用,以及飲食物使用時之香味等之問題。進一步,改變結晶形態或鹽之形態以改善吸濕性時,亦包含許多必須有煩雜的製造方法等之製造上的問題者。例如,上述專利文獻9中記載之組成物時,以90%的乙醇水煮沸L(+)酒石酸,為添加左旋肉鹼分子內鹽於其中,必須於高溫、高壓下濃縮大量的含肉鹼鹽溶液,不僅伴隨著明顯的能量浪費,使用有機溶劑時亦伴隨著成本增加、溶劑的回收使用、環境污染及有毒廢棄物之廢棄處理等之嚴重問題。
因此,本發明之課題係提供藉由工業上容易使用,且生產成本亦低之方法,吸濕性粉末原料,尤其肉鹼或該鹽或該衍生物之吸濕性被改善之含有吸濕性粉末原料之飲食品組成物。
本發明者為解決上述課題,努力研究的結果,混合粉末之芝麻素類於吸濕性粉末原料,上述吸濕性粉末原料之吸濕性被改善,尤其吸濕性粉末原料為肉鹼或該鹽或該衍生物時,發現該吸濕性明顯地被改善。接著,因為粉末狀之芝麻素類無臭無味,所以確認容易適用飲食品,完成本發明。
亦即,本發明係提供含吸濕性粉末原料及芝麻素類,以粉末芝麻素類為宜之飲食品組成物,飲食品粉末組成物為宜。
本發明係提供混合、含有吸濕性粉末原料及芝麻素類之飲食品粉末組成物,上述吸濕性粉末原料之吸濕性被改善之上述組成物。
本發明又提供上述吸濕性粉末原料係肉鹼或該鹽或該衍生物之上述組成物。本發明又提供上述芝麻素類係芝麻素及/或表芝麻素,更特定時,芝麻素及表芝麻素之上述組成物。
進一步,本發明係含有上述組成物之飲食品。
本說明書中,所謂「吸濕性」係指對象物暴露於空氣中時,捕捉大氣中水蒸氣之性質,特別稱為潮解性,捕捉大氣中水蒸氣而溶解於該水之性質亦包含於本說明書中之「吸濕性」。另外,吸濕性粉末原料之「吸濕性改善」,係指與未處理之吸濕性粉末原料相比較,吸濕性降低,防止潮解及/或結塊,或抑制潮解及/或結塊。
吸濕性係例如後述之實施例記載,使用該業者已知手法,可測定及/或評估。具體上,於室溫或高溫高濕度下等之一定條件下,將測量的原料,放置適當的時間後,可藉由目測觀察變化(確認原料表面的水份、確認固體的消失、確認粉末的流動性或結塊化程度、確認變色等)、或測量重量增加分(吸濕水量)檢討等之手法。
本說明書中,所謂「吸濕性粉末原料」係指所需製品之製造中及/或保存中等,只要上述之吸濕性成為問題之飲食品用或醫藥用所容許之物質皆可,並無限制。本說明書中,「吸濕性成為問題」原料係例如本說明書中之有關吸濕性之記載為參考,若相關業者應可容易判斷,具體上係有關於製造上或製品之貯藏中發生之吸濕。尤其測定製品之貯藏安定性用之加速試驗所使用之溫度為40℃,相對濕度為75%之條件下顯示吸濕(或潮解)之原料。作為吸濕性粉末原料之具體例,可舉例如偏磷酸鈉、焦磷酸鉀、醋酸鈉、明礬胺(ammonium alum)等之有機酸或無機酸之鹽類、氯化鎂、氯化鈣等之無機鹽類、木寡醣等之糖類,進一步如帝拔癲(sodium valproate)、甲磺司特(Suplatast Tosilate)、膽鹼之鹽、卡巴拉汀(Rivastigmine)、溴斯的明(Pyridostigmine Bromide)、巴氨西林(bacampicillin)、左旋肉鹼及其衍生物、Moricizine Hydrochloride、及白背櫟萃取物等之生藥萃取物等之藥劑所使用之活性成份。
本發明之吸濕性粉末原料係包含於飲食品組成物,所以若為具有相同的生理活性之原料時,以使用香味問題少之吸濕性粉末原料為宜。例如吸濕性原料為左旋肉鹼或該鹽或該衍生物時,使用無臭無味之左旋肉鹼比使用酸味強的左旋肉鹼酒石酸鹽為宜。
本說明書中,所謂肉鹼或該鹽或該衍生物亦包含肉鹼[(CH3
)3

COO-
、γ-三甲基銨-β-羥基丁酸]、該異構物、該鹽、及肉鹼、該異構物及該鹽之衍生物(例如生物體內轉換成肉鹼之衍生物)。包含如肉鹼、左旋肉鹼、氯化肉鹼(右旋左旋體、左旋體)、左旋肉鹼酒石酸鹽、左旋肉鹼馬來酸鹽、左旋肉鹼富馬酸鹽、左旋肉鹼硫酸鹽、左旋肉鹼草酸鹽、左旋肉鹼鎂鹽、醯基左旋肉鹼、乙醯基左旋肉鹼、丙醯基左旋肉鹼、乳清酸肉鹼等。此等係對該製作方法不做任何限定,天然萃取物、微生物發酵品、合成品中之任一種皆可,以天然存在之左旋肉鹼或該鹽或該衍生物為宜,例如下式:

之低級鏈烷醯基(alkanoyl)。)所表示。另外,關於本發明書中之肉鹼之說明係合乎肉鹼的鹽、肉鹼或該鹽之衍生物。
本發明書中,「芝麻素類」係包含芝麻素類及該類似體,作為具體例,可舉例如芝麻素、芝麻醇(sesaminol)、表芝麻醇、芝麻素酚(sesamolin)、表芝麻素(episesamin)、二氧雙環[3.3.0]辛烷衍生物(例如特開平4-9331號記載)等。其中,本發明中係以芝麻素、表芝麻素為宜。進一步,「芝麻素類」係包含芝麻素及該類似體之糖苷以及此等之代謝物,只要適合提供本發明之飲食品組成物,該任一種皆可含於本發明之飲食品組成物。
本發明所使用之芝麻素類係只要為固體狀,對於製造方法等無任何限制。例如使用芝麻素及/或表芝麻素作為芝麻素類時,通常係可使用由已知方法(例如加入熱甲醇於芝麻油萃取,除去甲醇後,加入丙酮於殘渣中萃取之方法(記載於特開平4-9331號公報))自芝麻油萃取之芝麻素(亦稱為芝麻素萃取物或芝麻素精製物)。成為原料之芝麻油並無特別的限制,但使含於本發明之飲食品組成物時,因為有時芝麻油特有的風味,於官能評價上並不喜好,所以以使用無臭無味之芝麻油所萃取之芝麻素萃取物或芝麻素精製物為宜。另外,使用精製度低的芝麻素類時,若配合後述之適合量之芝麻素時,所得之飲食品組成物或飲食品之體積過大,發生不適合攝取之情況。尤其,製劑化成經口投予用時,製劑(錠劑、膠囊等)變大,發生攝取障礙。因此,就攝取量少為宜之觀點,以使用芝麻素精製物為宜。
本發明之芝麻素類,就關於改善吸濕性粉末原料之吸濕性之觀點,以粉末芝麻素類為宜。該粉末芝麻素類之粒徑雖依與吸濕性粉末原料之混合方法(均質性)而定,但通常為JIS標準篩10至250網目,以60至200網目之範圍為宜。
本發明之飲食品組成物中與芝麻素類一同含有之吸濕性粉末原料係具有增進健康等之作用時,上述芝麻素類,尤其以芝麻素及/或表芝麻素為宜。因為認為此等芝麻素類之各種生理作用係可與上述吸濕性粉末原料之作用相加或加乘地發揮。例如吸濕性粉末原料係肉鹼時,提昇肉鹼能量代謝之作用、提昇運動性能之作用係芝麻素及表芝麻素具有之生理作用之一,認為可相加或加乘地發揮提昇能量代謝(Biosci.Biotechnol.Biochem.,69,179-188,2005)、或提昇運動能作用(Int.J.Sports Med.,24,530-534,2003,Biofactors,21,191-196,2004),將可提供優異的健康食品。
本發明之飲食品組成物係含吸濕性粉末原料及芝麻素類,可由混合吸濕性粉末原料及芝麻素類以及因應所需之其他添加劑而得。混合手法並無任何限制,只要可均質地混合,任何手法皆可。使用一般已知之混合機,例如水平圓筒型混合機、V型混合機、雙重圓錐型混合機、立方體型混合機、螺旋型混合機、螺帶型混合機、介質攪拌型研磨機、高速旋轉磨碎.衝擊研磨機、噴射式研磨機等之空氣力學的粉碎器等混合、可使用以研缽或球磨機等粉碎混合、利用聚乙烯(PE)製、尼龍製或依該性質而成的袋攪拌混合等。就操作容易,無須特別機械之觀點,可使用利用袋之攪拌混合為宜。吸濕性粉末原料及芝麻素類之混合程度高時,可得到更高的吸濕性改善效果。
本發明之飲食品組成物,芝麻素類之配合量係依一同所含有之吸濕性原料之種類或其他添加劑之有無等而異,相對於100份之吸濕性原料,作為重量比係以0.001至100之芝麻素類,0.01至10程度為適當的。
本發明之飲食品組成物係可直接作為後述之飲食品使用’亦可使用該業者已知的手法,使適當含於該飲食品而使用。本發明之飲食品組成物因為含有該吸濕性被改善狀態之吸濕性粉末原料,所以化學上安定,適合於貯藏或製造飲食品等。
本說明書中,所謂飲食品係包含調味料、營養補助食品、健康食品、飲食療法食品、綜合健康食品、營養補給品及飲料以及經口投予醫藥品。本發明之飲食品係可為固體狀(例如結晶、膠囊、錠劑、粉末)、半固體狀(例如凝膠、糊狀物)、液狀(例如礦泉水、清涼飲料水、果實飲料、運動飲料、酒類)。本發明之飲食品係以錠劑、顆粒品、飲食前溶解於液體之錠劑或顆粒品為宜。
本發明之飲食品係可使用該業者已知之手法,可使含有本發明之飲食品組成物而製造。本發明之飲食品係可期待安定的吸濕性粉末原料及芝麻素的所需效果。
本發明之飲食品中吸濕性粉末原料及芝麻素之配合量並無特別的限制,以此等適合之1日攝取量作為參考,若為該業者,因應該飲食品之攝取形態,可適當地設定配合量。
本發明之飲食品組成物係可因應需要,含有上述吸濕性粉末原料及芝麻素類以外之任意添加劑。另外,本發明之飲食品係可含有上述本發明之飲食品組成物以外之通常飲食品所使用之任意成份。作為此等添加劑及/或成份,可舉例如維生素E、維生素C等之維生素類、糖類、賦形劑、崩壞劑、結合劑、潤澤劑、乳化劑、緊張化劑(等張化劑)、緩衝劑、助溶劑、防腐劑、安定化劑、抗氧化劑、著色劑、矯味劑、香料、凝固劑、pH調整劑、增黏劑、萃取物粉末、生藥、無機鹽等。
另外,本發明之飲食品組成物或飲食品可表示該具體之用途(例如補給營養用、維持健康用、燃燒脂肪用、預防肥胖用、維持瘦身後體型用等)及/或此等之具體用法(例如攝取量、攝取次數、攝取方法等)。
以下係基於實施例說明本發明,但本發明並不侷限於此等實施例者。
使用肉鹼作為吸濕性原料,對於藉由混合芝麻素類,改善肉鹼之吸濕性,進行檢討。
實驗材料使用左旋肉鹼結晶性粉末(三榮源FFI股份有限公司,左旋肉鹼結晶性粉末)作為吸濕性粉末原料。使用經精製之粉末狀之芝麻素及表芝麻素之混合物(芝麻素:表芝麻素=51.1:48.2(重量比))作為芝麻素類。
另外,作為比較例係取代芝麻素類,使用如下述之製劑化中所經常使用之添加劑,對於改善肉鹼之吸濕性,進行檢討。
比較例1:環狀糊精類(α-CD/β-CD/γ-CD)(鹽水港精製股份有限公司,Dexypal α-100/β-100/γ-100)比較例2:SiO2
(富士Silisea化學股份有限公司,sylopage)比較例3:澱粉(日澱化學股份有限公司,Perfiler-102)比較例4:水溶性食物纖維(太陽化學股份有限公司,Sunfiber)比較例5:蔗糖脂肪酸酯(第一工業製藥股份有限公司,DK酯F-10)
實驗方法1.於培養皿(40)中,秤量各200mg之肉鹼或以1:1混合肉鹼與芝麻素類(使用PE製塑膠袋混合粉體)之粉末。
2.培養皿加蓋之狀態下,靜置於(A)室溫(20℃)、(B)培養箱(40℃,75%RH),目測觀察12小時後之性狀變化。
3.分別使用比較例1至5之粉末以取代芝麻素類,同樣地進行檢討。
結果圖1係表示混合芝麻素類於肉鹼時之改善肉鹼吸濕性之結果。單獨肉鹼時吸濕性高,不僅於高溫高濕度下(40℃,75%RH),即使於室溫下亦發生潮解(吸濕)(圖1中(b)(c))。相對於此,等量混合芝麻素類及肉鹼時,於室溫下,高溫高濕度下,混合物仍為粉末狀態(圖1中(e)(f))。由此顯示芝麻素類具有吸濕性原料之吸濕性(潮解性)改善作用。另外,改變肉鹼及芝麻素之混合條件時,顯示混合比率高者吸濕性改善效果高(未表示實驗結果)。
圖2係表示混合比較例1至5之各添加劑於肉鹼時之改善肉鹼吸濕性之結果。認為環狀糊精類(比較例1、圖2中(1 α)、(1 β)、(1 γ)、SiO2
(比較例2、圖2中(2))無吸濕性改善作用。認為澱粉(比較例3、圖2中(3))、水溶性食物纖維(比較例4、圖2中(4))部份潮解。認為蔗糖脂肪酸酯(比較例5、圖2中(5))改善吸濕性。
由以上結果顯示芝麻素類或蔗糖脂肪酸酯係對於改善肉鹼之吸濕性有效。芝麻素類係無臭無味,認為特別適合使用。
使用作為吸濕性粉末原料之左旋肉鹼結晶性粉末(三榮源FFI股份有限公司,左旋肉鹼左旋酒石酸鹽S、雖左旋肉鹼吸濕性被改善,但仍具有吸濕性)、MgCl2
(富田製藥股份有限公司,氯化鎂S)、木寡糖(Suntory股份有限公司,木寡糖95P)。以與實施例1相同步驟,檢討芝麻素類對此等濕性粉末原料之吸濕性改善作用。該結果係可確認對於任一種吸濕性粉末原料,吸濕性被改善。
於下述之製造例中,芝麻素類及肉鹼係使用實施例1記載者。
(製劑例1)顆粒劑芝麻素類 0.01g肉鹼 100g生育酚乙酸酯 0.25g矽酸酐 20.5g玉米澱粉 79.0g
將上述粉體均勻混合後,加入100ml之10%之羥丙基纖維素.乙醇溶液,如常法混練、押出、乾燥而得顆粒劑。
(製劑例2)錠劑芝麻素類 0.01g肉鹼 100g澱粉 172g蔗糖脂肪酸酯 9.0g氧化矽 9.0g
將此等混合,以單發式打錠機打錠,製造直徑為9mm,重量為300mg之錠劑。
(製劑例3)膠囊劑果膠 60.0%甘油 30.0%對羥基苯甲酸甲酯 0.15%對羥基苯甲酸丙酯 0.51%水 適量
由上述成份所形成之軟膠囊劑皮之中,由常法充填如下所示之組成物,得到1粒200g之軟膠囊。
芝麻素類 10.0mg肉鹼 100mg甘油脂肪酸酯 15.0mg蜜蠟 15.0mg小麥胚芽油 160mg
(製劑例4)口服劑呈味:右旋左旋酒石酸鈉 0.1g琥珀酸 0.009g甘味:液糖 800g酸味:檸檬酸 12g維生素:維生素C 10g芝麻素類 1g肉鹼 10g維生素E 30g環狀糊精 5g香料 15ml氯化鉀 1g硫酸鎂 0.5g
配合上述成份,加水成10L。此口服劑係每次飲用約100ml。
(製劑例5)錠劑調製配合左旋肉鹼衍生物之以維持健康及/或維持瘦身後體型為目的之健康食品(錠劑)。使用左旋肉鹼左旋酒石酸鹽(Lonza Japan股份有限公司)作為左旋肉鹼衍生物,配合如下述所示含量(每一粒)之作為其他功能性成份之芝麻素類(竹本油脂股份有限公司)、荷葉萃取粉末(松浦藥業股份有限公司)、辣椒素(capsaicin)(日本新藥股份有限公司)、咖啡因(白鳥製藥股份有限公司)、大花紫薇(Banaba)葉萃取粉末(常盤植物化學研究所股份有限公司)及維生素B1(田邊製藥股份有限公司)。另外,混合作為賦型劑之蔗糖脂肪酸酯、纖維素、二氧化矽、澱粉、乳糖,以打錠劑製造每一粒為直徑8mm,重量250mg之錠劑。確認製造中無吸濕或潮解之問題,以及所調製之錠劑於貯藏中未發生吸濕或潮解之問題。
左旋肉鹼左旋酒石酸鹽 74.3mg芝麻素類 1.7mg荷葉萃取粉末 16.7mg辣椒素 4.2mg咖啡因 16.7mg大花紫薇葉萃取粉末 16.7mg維生素B1 0.18mg
於下述製造例中,芝麻素類及肉鹼係使用實施例1記載者,左旋肉鹼左旋酒石酸鹽係使用實施例2記載者。13名品評員舔嚐粉末狀態之等量混合芝麻素類及肉鹼者(P)、及左旋肉鹼左旋酒石酸鹽(Q),進行(1)甜味、(2)刺激、(3)口中殘留味道、(4)難以食用、(5)喜歡何者之5個詢問。然而,對於(1)至(4)之詢問,選擇1.感覺強烈、2.稍微感覺到、3.無特別感覺之3種回答,對於(5)之詢問,P、Q二者中擇一。
該結果如表1所示,可知喜歡P比Q多(表1中的數係橫軸記載分別對於P、Q,選擇各回答之人數)。亦即,與由傳統的步驟使改善肉鹼吸濕性之肉鹼衍生物之左旋肉鹼左旋酒石酸鹽(Q)相比較,可確認等量混合芝麻素類及肉鹼之本發明組成物(P)(1)稍有甜味、(2)刺激弱、(3)口中未感到殘留味道、(4)容易食用、(5)受喜愛,判定本發明之組成物係作為飲食品組成物,就香味方面,亦為優異者。
圖1係表示於實施例1中,混合芝麻素類於肉鹼時之肉鹼吸濕性改善結果圖。
圖2係表示於實施例1中,混合各種添加劑於肉鹼時之肉鹼吸濕性改善結果圖。
Claims (5)
- 一種飲食品組成物,其特徵為,含吸濕性粉末原料及芝麻素(sesamin)類。
- 如申請專利範圍第1項之組成物,其中該吸濕性粉末原料係肉鹼或該鹽或該衍生物。
- 如申請專利範圍第1項或第2項之組成物,其中該芝麻素類係芝麻素及/或表芝麻素(episesamin)。
- 如申請專利範圍第1或2項之組成物,其為含有混合吸濕性粉末原料及芝麻素類之飲食品粉末組成物,該吸濕性粉末原料之吸濕性被改善。
- 一種飲食品,其特徵為含有如申請專利範圍第1項至第4項中任一項之組成物。
