Claims (22)
1. Обработанный гомогенат, приготовленный из образца гетерогенной ткани, содержащий по существу гомогенно распределенные клеточные структуры, где соотношение клеточных структур в каждой подгруппе гомогената по существу аналогично соотношению клеточных структур в образце ткани.
2. Гомогенат по п. 1, где образец ткани выбран из группы, состоящей из: опухоли, лимфатического узла, метастаза, полипа, кисты, извлеченного резекцией материала, органа или их доли.
3. Гомогенат по п. 1 или 2, где образец ткани содержит пространственно обособленные клеточные структуры.
4. Гомогенат по любому из пп. 1-3, где клеточные структуры содержат кластер клеток, отдельную клетку, фрагмент клетки, органеллу, пептид, нуклеиновую кислоту, липид, метаболит или их комбинацию.
5. Гомогенат по любому из пп. 1-4, где гомогенат содержит до 25% включительно клеточных структур из образца ткани.
6. Гомогенат по любому из пп. 1-4, где гомогенат содержит 100% клеточных структур из образца ткани.
7. Гомогенат по любому из пп. 1-6, где гомогенат дополнительно содержит один или более чем один материал, выбранный из клетки, не являющейся клеткой человека, человеческой клетки, ненативного белка, нуклеиновых кислот или малой молекулы, красителя, вируса, бактерии, паразита, представителя простейших или химического вещества.
8. Композиция по п. 7, где малая молекула содержит гаптен, пептидную метку, белковую метку, флуоресцентную метку, нуклеиновокислотную метку и их комбинацию.
9. Способ получения репрезентативных данных, включающий анализ гомогената по любому из пп. 1-8.
10. Способ по п. 9, где анализ включает получение количественных и/или качественных данных для маркера в гомогенате.
11. Способ по п. 10, где маркер содержит ДНК, белок, РНК, липид, клеточную органеллу, метаболит или клетку.
12. Способ по п. 9, дополнительно включающий сравнение репрезентативных данных с предварительно заданным значением для этих данных.
13. Способ по п. 12, где предварительно заданное значение выбрано из данных клинических испытаний, данных для субъекта, данных из научной литературы, данных для находящейся в клинической разработке биологической или малой молекулы.
14. Способ по п. 9, где репрезентативные данные содержат репрезентативные онкологические данные, при этом репрезентативные онкологические данные содержат количественные и/или качественные данные по меньшей мере для одного опухолевого маркера из первого биологического образца, при этом указанный опухолевый маркер ассоциирован с наличием опухоли, включающий:
(a) измерение опухолевого маркера в первом биологическом образце, при этом измеренное значение для опухолевого маркера является нормированным;
(b) получение суммарной оценки в баллах на основании этого нормированного измеренного значения; и
(c) сравнение суммарной оценки в баллах с предварительно заданной оценкой в баллах с целью определения показателя риска рака у субъекта.
15. Способ определения фенотипического профиля образца ткани, который включает анализ клеточных структур гомогената по п. 1.
16. Способ приготовления гомогената по п. 1, включающий:
a) гомогенизацию образца;
b) реконструирование гомогенизированного образца с получением гомогената, причем указанный гомогенат содержит по существу гомогенные клеточные структуры,
при этом соотношение клеточных структур в подгруппе гомогената по существу аналогично или равно соотношению клеточных структур в образце ткани.
