Abstract
1. Способ выращивания плюрипотентных стволовых клеток, включающий следующие этапы:а. прикрепление популяции плюрипотентных стволовых клеток к первому объему микроносителей;b. культивирование плюрипотентных стволовых клеток на первом объеме микроносителей;c. отделение плюрипотентных стволовых клеток от первого объема микроносителей;d. прикрепление популяции плюрипотентных стволовых клеток ко второму объему микроносителей.2. Способ по п.1, в котором этапы культивирования, отделения и прикрепления плюрипотентных стволовых клеток на микроносителях повторяются с использованием следующих объемов микроносителей.3. Способ по п.1, в котором первый объем микроносителей выбирается из группы, состоящей из декстрановых микроносителей и полистироловых микроносителей.4. Способ по п.1, в котором второй объем микроносителей выбирается из группы, состоящей из декстрановых микроносителей и полистироловых микроносителей.5. Способ по п.1, в котором плюрипотентные стволовые клетки прикрепляются к первому объему микроносителей в среде, содержащей ингибитор Rho-киназы.6. Способ по п.1, в котором плюрипотентные стволовые клетки прикрепляются ко второму объему микроносителей в среде, содержащей ингибитор Rho-киназы.7. Способ по п.1, в котором плюрипотентные стволовые клетки отделяются от первого объема микроносителей путем ферментативной обработки.8. Способ по п.1, в котором плюрипотентные стволовые клетки отделяются от второго объема микроносителей путем ферментативной обработки.9. Способ по п.1, в котором первый объем микроносителей отделяется от плюрипотентных стволовых клеток перед прикреплением клеток ко второму объему микро
Claims (10)
1. Способ выращивания плюрипотентных стволовых клеток, включающий следующие этапы:
а. прикрепление популяции плюрипотентных стволовых клеток к первому объему микроносителей;
b. культивирование плюрипотентных стволовых клеток на первом объеме микроносителей;
c. отделение плюрипотентных стволовых клеток от первого объема микроносителей;
d. прикрепление популяции плюрипотентных стволовых клеток ко второму объему микроносителей.
2. Способ по п.1, в котором этапы культивирования, отделения и прикрепления плюрипотентных стволовых клеток на микроносителях повторяются с использованием следующих объемов микроносителей.
3. Способ по п.1, в котором первый объем микроносителей выбирается из группы, состоящей из декстрановых микроносителей и полистироловых микроносителей.
4. Способ по п.1, в котором второй объем микроносителей выбирается из группы, состоящей из декстрановых микроносителей и полистироловых микроносителей.
5. Способ по п.1, в котором плюрипотентные стволовые клетки прикрепляются к первому объему микроносителей в среде, содержащей ингибитор Rho-киназы.
6. Способ по п.1, в котором плюрипотентные стволовые клетки прикрепляются ко второму объему микроносителей в среде, содержащей ингибитор Rho-киназы.
7. Способ по п.1, в котором плюрипотентные стволовые клетки отделяются от первого объема микроносителей путем ферментативной обработки.
8. Способ по п.1, в котором плюрипотентные стволовые клетки отделяются от второго объема микроносителей путем ферментативной обработки.
9. Способ по п.1, в котором первый объем микроносителей отделяется от плюрипотентных стволовых клеток перед прикреплением клеток ко второму объему микроносителей.
10. Способ дифференцирования плюрипотентных стволовых клеток в инсулинэкспрессирующие клетки, состоящий из следующих этапов:
а. прикрепление плюрипотентных стволовых клеток к объему микроносителей;
b. дифференцирование плюрипотентных стволовых клеток в клетки, экспрессирующие маркеры, характерные для линии сформированной эндодермы;
c. дифференцирование клеток, экспрессирующих маркеры, характерные для линии сформированной эндодермы, в клетки, экспрессирующие маркеры, характерные для линии панкреатической эндодермы;
d. дифференцирование клеток, экспрессирующих маркеры, характерные для линии панкреатической эндодермы, в клетки, экспрессирующие маркеры, характерные для линии панкреатических эндокринных клеток;
e. дифференцирование клеток, экспрессирующих маркеры, характерные для линии панкреатических эндокринных клеток, в инсулинэкспрессирующие клетки.
