Claims (29)
1. Способ извлечения стволовых/прародительских клеток из амниотической мембраны пуповины, при котором:
(а) отделяют амниотическую мембрану от других компонентов пуповины in vitro;
(б) культивируют ткань амниотической мембраны, полученной в этапе (а), в условиях, дающих возможность клеточной пролиферации; и
(в) извлекают стволовые/прародительские клетки.
2. Способ по п.1, при котором дополнительно
(а") отделяют клетки ткани амниотической мембраны перед культивированием способом, выбранным из группы, состоящей из ферментативного расщепления и прямого культивирования ткани эксплантата.
3. Способ по п.1, отличающийся тем, что стволовые/прародительские клетки имеют свойства, подобные свойствам эмбрионных стволовых клеток.
4. Способ по п.1, отличающийся тем, что стволовые/прародительские клетки являются эпителиальными и/или мезенхимными стволовыми/прародительскими клетками.
5. Способ по п.1, при котором дополнительно
(г) культивируют стволовые/прародительские клетки в условиях, дающих возможность клеткам претерпевать клональную экспансию.
6. Способ по п.5, при котором дополнительно
(д) культивируют стволовые/прародительские клетки в условиях, дающих возможность дифференцировки указанных клеток в эпителиальные клетки и/или мезенхимные клетки; и
(е) извлекают дифференцированные клетки.
7. Способ по п.6, отличающийся тем, что эпителиальные клетки выбраны из группы, состоящей из клеток эпителия кожи, клеток волосяных фолликул, клеток эпителия роговицы, клеток эпителия конъюнктивы, клеток эпителия сетчатки, клеток эпителия печени, клеток эпителия почек, клеток эпителия поджелудочной железы, клеток эпителия пищевода, клеток эпителия тонкой кишки, клеток эпителия толстой кишки, клеток эпителия легких, клеток эпителия дыхательных путей, клеток эпителия мочевого пузыря и клеток эпителия матки.
8. Способ по п.6, отличающийся тем, что мезенхимные клетки выбраны из группы, состоящей из: фибробластов кожи, хондроцитов, остеобластов, теноцитов, фибробластов связок, кардиомиоцитов, клеток гладких мышц, клеток скелетных мышц, адипоцитов, клеток, полученных из эндокринных желез, нейректодермальных клеток и всех разновидностей и производных нейректодермальных клеток.
9. Способ по любому из пп.1-8, при котором дополнительно
(ж) консервируют извлеченные стволовые/прародительские клетки для дальнейшего использования.
10. Способ по п.9, отличающийся тем, что консервацию осуществляют с использованием криоконсервации.
11. Стволовая/прародительская клетка, извлеченная из амниотической мембраны пуповины способом по любому из пп.1-10.
12. Клеточный банк, содержащий стволовую/прародительскую клетку, извлеченную способом по любому из пп.1-10.
13. Фармацевтическая композиция, содержащая стволовую/ прародительскую клетку, извлеченную из амниотической мембраны пуповины способом по любому из пп.1-10, или клеточный экстракт из нее.
14. Фармацевтическая композиция по п.13, отличающаяся тем, что фармацевтическая композиция приспособлена для системного или местного применения.
15. Фармацевтическая композиция по п.13 или 14, отличающаяся тем, что фармацевтическая композиция, приспособленная для местного применения, выбрана из группы, состоящей из мази, крема и лосьона.
16. Способ лечения имеющего расстройство индивидуума, включающий введение индивидууму эффективного количества стволовых/прародительских клеток, извлеченных способом по любому из пп.1-10.
17. Способ лечения по п.16, отличающийся тем, что расстройство выбрано из группы, состоящей из опухолевого заболевания, заболевания кожи, тканевых расстройств, висцеральной эндокринной недостаточности и нервного заболевания.
18. Способ по п.17, отличающийся тем, что тканевое расстройство представляет собой врожденную или приобретенную тканевую недостаточность.
19. Способ по п.17, отличающийся тем, что висцеральная эндокринная недостаточность выбрана из группы, состоящей из инсулиновой недостаточности, сахарного диабета, связанного с инсулиновой недостаточностью, недостаточности тестостерона, анемии, гипогликемии, гипергликемии, недостаточности поджелудочной железы, недостаточности надпочечников и ненормального функционирования щитовидной железы.
20. Способ по п.17, отличающийся тем, что нервное заболевание представляет собой болезнь Альцгеймера, болезнь Паркинсона, болезнь Крейтцфельда-Якоба, болезнь Лю-Герига и болезнь Хантингтона.
21. Способ по п.17, отличающийся тем, что заболевание кожи представляет собой ускоренное старение или рану.
22. Способ по п.17, отличающийся тем, что опухолевое заболевание представляет собой рак.
23. Способ по п.22, отличающийся тем, что рак выбран из группы, состоящей из карциномы сквамозных клеток, карциномы протоков и долек молочной железы, карциномы клеток печени, карциномы носоглотки, рака легких, рака костей, рака поджелудочной железы, рака кожи, рака головы или шеи, кожной или внутриглазной злокачественной меланомы, рака матки, рака яичников, рака прямой кишки, рака анальной области, рака желудка, рака толстой кишки, рака груди, рака яичек, рака матки, карциномы фаллопиевых труб, карциномы эндометрия, карциномы шейки матки, карциномы влагалища, карциномы вульвы, болезни Ходжкина, лимфомы не Ходжкина, рака пищевода, рака тонкой кишки, рака эндокринной системы, рака щитовидной железы, рака околощитовидной железы, рака надпочечника, саркомы мягких тканей, рака мочеиспускательного канала, рака пениса, рака простаты, хронических или острых лейкозов, детских твердых опухолей, лимфоцитарной лимфомы, рака мочевого пузыря, рака почки или мочеточника, карциномы клеток почки, карциномы почечных лоханок, новообразований центральной нервной системы (ЦНС), первичной лимфомы ЦНС, опухолевого ангиогенеза, опухоли позвоночного столба, глиомы ствола головного мозга, аденомы гипофиза, саркомы Калоши, эпидермоидного рака или любой комбинации таких видов рака, в том числе их диссеминированных (метастазирующих) форм.
24. Применение стволовой/прародительской клетки, как она определена в п.11, для продуцирования биологической молекулы.
25. Применение по п.24, отличающееся тем, что биологическая молекула выбрана из группы, состоящей из белка, пептида, низкомолекулярного органического соединения, олигосахарида, полисахарида, протеогликана и липида.
26. Применение стволовой/прародительской клетки, как она определена в п.11, в качестве питающего слоя в культивировании клеток млекопитающих.
27. Способ культивирования стволовых/прародительских клеток, как они определены в п.11, при котором
получают эксплантата ткани из амниотической мембраны пуповины;
культивируют эксплантата ткани в подходящих среде для культивирования и условиях культивирования в течение подходящего периода времени.
28. Способ по п.27, при котором дополнительно выдерживают эксплантат ткани в свежей среде для культивирования и продолжают культивирование в подходящих условиях в течение подходящего периода времени.
29. Применение бессывороточной среды для культивирования клеток для культивирования мезенхимных стволовых клеток, полученных способом по любому из пп.1-10.
