Oppfinnelsens område
Foreliggende oppfinnelse omfatter monoklonale antistoffer mot IL-15 og farmasøytiske sammensetninger omfattende slike. Oppfinnelsen angår videre anvendelse av antistoffene for fremstilling av medikamenter for behandling av ulike sykdomstilstander hos pattedyr der en reduksjon i IL-15-aktivitet er ønsket.
Bakgrunn for oppfinnelsen
Interleukin-15 er en kjent T-cellevekstfaktor som underbygger proliferasjon av en IL-2-avhengig cellelinje, CTLL-2. IL-15 som først ble rapportert av Grabstein et al., i Science, 264:965 (1994), et sekretorisk cytokin omfattende et aminosyreforløperpolypeptid på 162 aminosyrer inneholdende en aminosyreledersekvens på 48 aminosyrer som leder til et modent protein på 114 aminosyrer, Grabstein et al. beskrev også kloningen av det humane fullengde cDNA som koder for aminosyreforløperen på 162 aminosyrer, som inneholder et 5' ikke kodende område på 316 bp og en åpen leseramme på 486 bp (eller en åpen leseramme på 489 bp inkludert stopp kodonet på 3 bp) og et 3' ikke-kodende område på 400 bp. IL-15 har mange egenskaper til felles med IL-2. Disse egenskapene omfatter proliferering og aktivering av humane og murine T-celler, induksjon av lymfokinaktivert drepercelle (LAK)-aktivitet, naturlig drepercelle (NK)-aktivitet og cytotoksisk T-lymfocytt (CTL)-aktivitet og kostimulering av B-celleproliferering og differensiering. IL-15 og IL-2 er dessuten strukturelt sett homologe molekyler som har evnen til å bindes til minst tre ulike reseptorsubenheter på T-cellemembranoverflaten. IL-2-reseptorer inneholder minst tre subenheter, a, p og y (Toshikazu et al., Science, 257:379 (1992). Både IL-15 og IL-2 deler binding med en felles P-y subenhetskompleks, mens IL-15 og IL-2 binder hver for seg spesifikt til en a-reseptorsubenhet (hhv. IL-15Ra og IL-2Ra). Nylig ble IL-15Ra oppdaget, denne er gjenstand for en annen pågående søknad med serie nr. 08/300,903. Antistoffer rettet mot y-kjeden til IL-2-reseptoren (anti-IL-2Ra) har ingen virkning på IL-15-binding (Grabstein et al.,Id). Antistoffer rettet mot |3-subenheten til IL-2-reseptoren, dvs. TU27, TUl 1 eller Mikpi har imidlertid evnen til blokkere aktiviteten til IL-15, noe som tyder på at IL-15 anvender p-subenheten for signalisering. Likeledes er y-kjeden av IL-2-reseptoren krevet for signaloverføring (Giri et al., EMBO J., 75:2822 (1994). Kombinasjonen av p- og y-subenhetene i IL-15-reseptorkomplekset bandt IL-15 til transfekterte COS-celler, men ikke subenhetene alene.

Publikasjonene beskriver ikke de monoklonale antistoffene ifølge foreliggende oppfinnelse.
Visse sykdomstilstander og fysiologiske tilstander er mediert av T-celler. Slike sykdommer omfatter organtransplantatavstøtnig, "graft-versus-host"-sykdom, autoimmun sykdom, reumatoid artritt, inflammatorisk tarmsykdom, dermatologiske forstyrrelser, insulinavhengig diabetes mellitus, synsforstyrrelser og idiopatisk nefrotisk syndrom/idiopatisk membranøs nefropati. Allografl avstøtning og "graft-versus-host"-sykdom (GVHD) er forbundet med økt ekspresjon av IL-2-reseptor. T-celler aktivert som respons på fremmede vevsforlikelighets-antigener ser ut til å uttrykke IL-2-reseptorkomplekset. Ulike behandlinger er foreslått og undersøkt. F.eks. rapporterte Tinubu et al. (J. Immunol., 755:4330 (1994) at anti-IL-2RP monoklonalt antistoff, Mikpi, forlenget allograft overlevelse av hjerte hos primater. Det er en økning i ekspresjon av IL-"RP-subenheten på CD4- og DC8-uttrykkende celler i forbindelse med akutt allograft avstøtning, noe som tyder på at uttrykket av IL-2Rp-subenheten ser ut til å øke på alloreaktive T-celler. Se, for eksempel, Niguma et al., Transplation, 52:296 (1991.
Forut for foreliggende oppfinnelse har det imidlertid ikke vært terapier som fokuserte på IL-15-ligand-reseptorinteraksjonen som en metode for behandling av GVHD eller for å fremme allograft overlevelse.
O ppsummering av oppfinnelsen.
Foreliggende oppfinnelse angår monoklonalt antistoff dannet mot interleukin-15 kjennetegnet ved SEQ JJD NO: 1 eller SEQ ID NO: 2 som forhindrer at IL-15 overfører et signal gjennom en av P- eller y-subenhetene til IL-15-reseptorkomplekset.
Monoklonale antistoffer ifølge oppfinnelsen omfatter antistoffer kjennetegnet ved SEQ UD NO: 1 eller 2 som forhindrer at IL-15 overfører et signal gjennom en av P- eller y-subenhetene til IL-15-reseptorkomplekset, der det monoklonale antistoffet interfererer med binding av aminosyren Asp<56> i IL-15-molekylet til P-subenheten av IL-15-reseptorkomplekset og/eller aminosyren Gin<156> i IL-15-molekylet til y-subenheten til H-15-reseptorkomplekset.
Foreliggende oppfinnelse angår videre en farmasøytisk sammensetning kjennetegnet ved at den omfatter et monoklonalt antistoff ifølge oppfinnelsen, og en farmasøytisk akseptable bærer eller fortynningsmiddel.
Oppfinnelsen er også rettet på anvendelse av de monoklonale antistoffene for fremstilling av et medikament for behandling av en sykdom eller tilstand der en reduksjon i IL-15-aktivitet på T-celler er ønskelig. Slike sykdommer omfatter organtransplantatavstøtning, "graft-versus-host"-sykdom, autoimmun sykdom, reumatoid artritt, inflammatorisk tarmsykdom, dermatologiske forstyrrelser, insulinavhengig diabetes mellitus, øyesykdommer og idiopatiske nefrotisk syndrom/idiopatisk membranøs nefropati. Særlig ved allograft avstøtning kan IL-15-aktiviteten føre til en vertsimmunrespons mot transplantatet og eventuelt en avstøtning. Likeledes ved GVAH viderefører transplantatet, vanligvis et benmargstransplantat, en immunrespons mot verten. Undertrykkelse av slike aktiviteter ved hjelp av IL-15-antistoffer ifølge oppfinnelsen kan være fordelaktig ved fremmelse av forlenget transplantatoverlevelse og ved behandling av
GVHD.
Ulike forskere har rapportert om forlengelse av transplantatoverlevelse ved anvendelse av antistoffer, slik som anti-TAC, et anti-human IL-2-a-reseptormonoklonalt antistoff, se (Reed et al., Transplantation, 47:55- 59 (1989), hvori anti-TAC er vist å bedre allograft transplantasjon av primatnyre. Også, Brown et al., Proe. Nati. Acad. Sei., 55:2663 (1991) beskriver anvendelse av humanisert anti-TAC forlenget primat hjerteallograft overlevelse. Kirkman et al., Transplantation, 57:107 (1991) beskriver også et klinisk forsøk der anti-TAC er involvert i forebyggelse av tidlig allograft avstøtning. Etter som JJL-15 har mange biologiske aktiviteter som ligner på IL-2 og særlig deler visse reseptorsubenheter med IL-2, som interfererer med en skadelig aktivitet av IL-15 ved sykdomstilstander har et særlig terapeutisk potensiale.
Detaljert beskrivelse av oppfinnelsen
Med "rekombinant DNA-teknologi" eller "rekombinant" som anvendt heri, menes teknikker og fremgangsmåte for fremstilling av spesifikke polypeptider fra mikrobielle- (f.eks. bakterie, insekt eller gjær) eller pattedyrceller eller organismer (f.eks. transgener) som er transformert eller transfektert med klonede eller syntetiske DNA-sekvenser for å muliggjøre biosyntese av heterologe peptider. Native glykosyleirngsmønstre vil kun oppnås med pattedyrcelleekspresjonssystemer. Gjær tilveiebringer et forskjellig glykosyleringsrnønster. Prokaryot celleeksplosjon (f.eks. E.coli) produserer generelt polypeptider uten glykosylering.
En "nukleotidsekvens" refererer seg til et polynukleotid i form av et separat fragment eller som en bestanddel av en større DNA-konstruksjon, som er avledet fra DNA eller RNA isolert minst en gang i vesentlig ren form (dvs. fri for forurensende endogene bestanddeler) og i en mengde eller konsentrasjon som muliggjør identifisering, manipulering og gjenvinning av polynukleotid- komponentnukleotidsekvensene ved hjelp av vanlige biokjemiske fremgangsmåte, slik som de beskrevet i Sambrook et al, Molecular Cloning: A Laboratory Manual 2. red., Cold Spring Harbor Laboratory, Cold Spring Harbor, NY (1989). Slike sekvenser tilveiebringes fortrinnsvis i form av en åpen leseramme uten interne ikke transplanterte sekvenser, eller introner som vanligvis er til stede i eukaryote gener. Sekvenser av ikke-translatert DNA kan være til stede i 5' eller 3' i en åpen leseramme der sekvensene ikke interfererer med manipulering eller ekspresjon av de kodede områder.
"Rekombinant ekspresjonsvektor" refererer seg til et plasmid omfattende en transkripsjonen enhet kjennetegnet ved en sammensetning av (1) et genetisk element eller elementer som har en regulatorisk rolle ved genekspresjon, f.eks. promotorer eller enhancere, (2) en strukturell er kodende sekvens som koder for IL-15 eller et IL-15-antistoff, og (3) egnede transkripsjons- og translasjonsinitieringssekvenser og, dersom dette er ønskelig termineringssekvenser. De foreliggende eksempler på ulike regulatoriske elementer som kan anvendes er diskutert under (se rekombinante DNA-teknikker). Strukturelle elementer ment for anvendelse i gjærekspresjonssystemer omfatter fortrinnsvis en leder sekvens som muliggjør ekstracellulærsekresjon av translaterte polypeptider i en gjærvertscelle. Alternativt, i et bakterielt ekspresjonssystem kan det rekombinante polypeptid omfatte en N-terminalmetoinrest. Den N-
terminale metioninrest kan deretter kløyves fra det uttrykte rekombinante polypeptid for å tilveiebringe et produkt som er egnet for ytterligere rensing.
"Rekombinant mikrobielt ekspresjonssystem" refererer seg til én i det vesentlige homogen monokultur av egnede vertsmikroorganismer, f.eks. bakterier slik som E. coli eller gjær, slik som S. cerviasiae, som har stabilt integrert en rekombinant transkripsjonen enhet in dets kromosomale DNA eller som bærer den rekombinante transskripsjonsenheten som en bestanddel av et iboende plasmid. Vertsceller bestående av et rekombinant mikrobielt ekspresjonssystem er generelt avkommet av en enkelt transformert stamcelle. Rekombinante mikrobielle ekspresjonssystemer uttrykker heterologe polypeptider etter induksjon av de regulatoriske elementer bundet til en strukturell nukleotidsekvens som uttrykkes.
Fremstilling av IL- 15
Human eller ape-IL-15 kan fremstilles ifølge fremgangsmåtenene beskrevet av Grabstein et al., Science, 264965 (1994), eller ved hjelp av vanlige fremgangsmåter, slik som polymerasekjedereaksjon (PCR). En avsetning av human IL-15 cDNA ble gjort i American Type Culture Collection, Rockville, MD, USA (ATCC) 19. februar 1993 med aksesjonsnr. 69245. Deponeringen ble kalt "141-hETF". Deponeringen ble gjort ifølge Budapesttraktaten.
Monoklonale antistoffer mot IL- 15
Oppfinnelsen er rettet mot et monoklonalt antistoff med IL-15-aktivitet som interfererer med signaloverføringen av IL-15 ved et reseptorkompleks. De monoklonale antistoffene ifølge
oppfinnelsen er monoklonale antistoff mot interleukin-15 (IL-15) kjennetegnet ved SEQ ID NO: 1 eller 2 som forhindrer at IL-15 overfører et signal gjennom en av P- eller y-subenhetene til IL-15-reseptorkomplekset, der det monoklonale antistoffet interfererer med binding av aminosyren Asp<56> i IL-15-molekylet til P-subenheten av IL-15-reseptorkomplekset og/eller aminosyren Gin<156> i IL-15-molekylet til y-subenheten til IL-15-reseptorkomplekset. I en utførelsesform er antistoffet humanisert. I en annen utførelsesform er antistoffet transgent. IL-15, deriblant derivater derav i tillegg til deler eller fragmenter av disse proteiner, slik som IL-15-peptider kan anvendes for fremstilling av antistoffer som spesifikt binder til IL-15. Uttrykket "antistoffer" forstås å omfatte polyklonale antistoffer, monoklonale antistoffer, fragmenter derav, slik som F(ab')2 og Fab-fragmenter, i tillegg til rekombinant fremstilte bindingspartnere. Affiniteten til et monoklonalt antistoff eller bindingspartner kan letthet bestemmes av en vanlig fagperson på området, (se Scatchard, Ann. N.Y. Acad. Sei., 51: 660-672 (1949). Spesifikke eksempler på slike antistoffer er tilveiebrakt i eksempel 1 heri.
Generelt kan monoklonale antistoffer mot IL-15 fremstilles ved anvendelse av følgende fremgangsmåte. Renset IL-15, et fragment derav, syntetiske peptider eller celler som uttrykker IL-15 kan anvendes for å generere monoklonale antistoffer mot IL-15 ved anvendelse av fremgangsmåter kjent perse, f.eks. teknikkene beskrevet i US patent 4,411,993.1 korte trekk immuniseres mus med IL-15 som immunogen emulgert i fullstendig Freud's adjuvans eller RJJ3I-adjuvans (RBBI Corp., Hamilton, Montana) og injiseres i mengder varierende fra 10-100 ig subkutant eller intrapeirtonealt. Ti til tolv dager senere, innsprøytes de immuniserte dyrene med ytterligere IL-15 emulgert i ufullstendig Freud's adjuvans. Mus innsprøytes periodevis deretter ukentlig eller annenhver uke. Serumprøver tas periodevis ved hjelp av retro-orbital blødning eller haletuppeksekusjon for testing av IL-15-antistoffer ved hjelp av dot blot analyse, ELISA (Enzyme-Linked Immunosorbent Assay) eller inhibering av IL-15-aktivitet på CTLL-2-celler.
Etter påvisning av egnet antistofftiter tilveiebringes positive dyr med en ytterligere intravenøs injeksjon av IL-15 i saltoppløsning. Tre til fire dager senere avlives dyrene, miltceller innhøstes og miltceller fusjoneres med en murinmyelomcellelinje, f.eks. NS1 eller fortrinnsvis P3x63Ag8.653 (ATCC CRL1580). Fusjoner danner hybridomceller, som utplates på flere mikrotiterplater i et HAT (hypoksantin, aminopterin, og tymidin) selektivt medium for å hemme proliferering av ikke-fusjonerte myelomceller og myelom hybrider.
Hybridomcellene screenes ved hjelp av ELISA med hensyn til reaktivitet mot renset IL-15 ved hjelp av tilpasninger av teknikker beskrevet i Engvall et al., Immunochem. 5:871,1971 og i US patent 4,703,004. En foretrukket screeningsteknikk er antistoffinnfangingsteknikken beskrevet i Beckmann et al., (J. Immunol. 744:4212,1990). Positive hybridomceller kan injiseres intraperitonealt inn i syngeneiske Balb/c-mus for å fremstille ascites inneholdende høye konsentrasjoner av monoklonale anti-IL-15 monoklonale antistoffer. Alternativt kan hybridomceller dyrkes in vitro i kolber eller rulle flasker ved hjelp av ulike fremgangsmåter. Monoklonale antistoffer fremstilles i museascites kan renses ved hjelp av ammoniumsulfatutfelling, etterfulgt av geleksklusjonskromatografi. Alternativt kan affinitetskromatografi basert på binding av antistoff til protein A eller protein G også anvendes, affinitetskromatografi basert på binding til IL-15 kan også anvendes.
Andre "antistoffer" kan fremstilles ved anvendelse av beskrivelsen tilveiebrakt heri og faller således innen rammen av oppfinnelsen. Fremgangsmåter anvendt for å generere humaniserte antistoffer kan finnes i US patent nr. 4,816,567 og WO 94/10332; fremgangsmåter for generering av mikro legemer kan finnes i WO 94/09817; og fremgangsmåter for fremstilling av transgene antistoffer kan finnes i GB 2 272 440.
For å bestemme hvilke monoklonale antistoffer som er antagonister er anvendelse av en screeningsanalyse foretrukket.
En CTLL-2-prrolifereringsanalyse er foretrukket for dette formål. Se Gillis og Smith, Nature 255:154 (1977).
Antistoffene ifølge oppfinnelsen finner, som beskrevet ovenfor og mer detaljert
nedenfor, anvendelse ved fremming av allograft overlevelse og ved behandling av pasienter med "graft-versus-host"-sykdom. En annen anvendelse for antistoffene omfatter behandling av senfase HTLV (human T-cellelymfosofisk virus) i-indusert voksen T-celleleukemilymfom, se Burton et al., Proe. Nati. Acad. Sei., 97:4935 (1994). Andre anvendelser omfatter evnen til å forebygge B-celle eller T-Celle stimulering in vitro. Undersøke reseptor-ligandinteraksjon, i diagnostiske sett mot infeksiøs sykdom og forstyrrelser i fordøyelseskanalen. Som følge av aktiviteten til
antistoffene ifølge oppfinnelsen kan nye fremgangsmåter for behandling av visse sykdommer etableres. Det er f.eks. beskrevet forebyggelse av allograft avstøtning hos en pasient med dette behov og behandling av GVHD hos en pasient med dette behov, hvor behandling omfatter administrering av et farmasøytisk preparat omfattende en mengde av en IL-15-antagonist som er effektiv til å hemme IL-15-aktivitet og en farmasøytisk akseptabel bærer eller fortynner. Andre sykdommer der målcellene (cellene som er antatt å være primærresponsive for sykdomstilstanden, eller et symptom på sykdomstilstanden) uttrykker EL-15-reseptorkomplekset og hvor en blokkering eller hemming av signaloverføringen gjennom P- eller y-subenhetene til IL-15-reseptoren er ønskelig, kan også behandles med medikamentet ifølge oppfinnelsen. Slike sykdomstilstander kan behandles etter undersøkelse av at målcellene uttrykker IL-15-reseptorkomplet. Tillegg til GVHD og allograft avstøtning kan slike sykdomstilstander f.eks. omfatte lymfomer, carcinomer, leukemier, rabdosarkomer og visse autoimmune sykdommer, slik som reumatoid artritt. Ovennevnte liste er ikke fullstendig for alle sykdomstilstander hvori målcellene uttrykker IL-15-reseptorkomplekset og må ikke anses å være begrensende for oppfinnelsen.
Farmasøytiske preparater omfattende de ovenfor beskrevne IL-15-antistoffene er også omfattet av oppfinnelsen.
Administrering av antistoffer mot IL- 15
Foreliggende oppfinnelse omfatter også anvendelse av antistoffer for fremstilling av farmasøytiske preparater omfattende en effektiv mengde av IL-15-antistoff i en egnet fortynner eller bærer. En IL-15-antistoff ifølge oppfinnelsen kan formuleres ifølge kjente fremgangsmåter anvendt for fremstilling av farmasøytiske anvendbare preparater. Et IL-15-antistoff kan kombineres i blanding, enten som eneste aktive bestanddel eller sammen med andre kjente aktive bestanddeler, med farmasøytisk egnede fortynnere, (f.eks. Tris-HCL, acetat, fosfat), konserveirngsmidler (f.eks. timerosal, benzylalkohol, parabener), emulgatorer, oppløsningsmidler, hjelpemidler og/eller bærere. Egnede bærere og deres formuleringer er beskrevet i Remington's Pharmaceutical Sciences, 16th ed. 1980, Mack Publishing Co. Slike preparater kan dessuten inneholde et IL-15-antistoff sammensatt av polyetylenglykol (PEG) metallioner eller inkorporert i polymere forbindelser, slik som polyeddiksyre, eller inkorporert i polymere forbindelser, slik som polyeddiksyre, hydrogeler, etc. eller inkorporert i liposomer, mikroemulsjoner, miceller, unilamellære eller multilamellære vesikler, erytrocytt spøkelser (ghosts) eller sfæroplaster. Slike preparater influerer på den fysiske tilstand, oppløselighet, stabilitet, in v/vø-frigjøringsgrad og fjerningsgrad av et IL-15-antistoff in vivo. Et IL-15-antistoff kan også konjugeres til antistoffer mot vevsspesifikke reseptorer, ligander eller antigener, eller kobles til vevsspesifikke reseptorligander. IL-15-antistoffene ifølge oppfinnelsen kan administreres topisk, parenteralt, rektalt eller ved inhalering. Med uttrykket "parenteralt" omfattes subkutane injeksjoner, intravenøs, intramuskulær, intrasisterneinjeksjon eller infusjonsteknikker. Disse preparatene inneholder vanligvis en effektiv mengde av en IL-15-antistoff, alene eller sammen med en effektiv mengde av enhver annen aktiv bestanddel. Slike doser og ønskede medikamentkonsentrasjoner i preparatene kan variere avhengig av mange faktorer, deriblant ønsket anvendelse, pasientens kroppsvekt og alder og administreringsvei. Forutbestemte doser kan bestemmes ut fra dyretester og doseskaleringen for human administrering kan utføres ifølge artsakseptable praktiseringer.
I tillegg til det ovenfor beskrevne er de følgende eksempler ment å illustrere særlige utførelsesformer.
Eksempel 2
Monoklonale antistoffer mot IL- 15
Foreliggende eksempel beskriver fremgangsmåten anvendt for å fremstille tre monoklonale anti-IL-15-antistoffer som virker som antagonister mot IL-15. Alle fremgangsmåter anvendt er vanlige teknikker bortsett fra når dette er bemerket.
Balb/c-mus ble injisert intraperitonealt ved to anledninger med tre ukers intervaller med 10 ig gjæravledet human-IL-15 i nærvær av RIBI-adjuvans (PJBI Corp., Hamilton, Montana). Museserum ble så analysert ved hjelp av vanlig dot blot teknikk, antistoiffnnfanging (ABC) og nøytraliseirngsanalyse (CTLL-2-analyse) for å bestemme hvilket dyr som var best til å fusere. Tre uker senere ble mus gitt en intravenøs innsprøyting av 3 ig av human-IL-15 suspendert i steril PBS. Tre dager senere ble mus avlivet og miltceller ble fusjonert Ag8,653 myelomceller (ATCC) etterfulgt av etablering av protokoller. I korte trekk ble Ag8.653-celler vasket flere ganger i serumfritt medium og fusjonert til musemiltceller i en grad på tre miltceller per myelomcelle. Fusjonsmidlet var 50% PEG: 10% DMSO (Sigma) Fusjonen ble platet ut i tyve 96-brønners flatbunnede plater (Corning) inneholdende HAT-supplementert DMEM-medium og tillatt å gro i åtte dager. Supernatanter fra resulterende hybridomer ble innsamlet og tilsatt til en 96-brønners plate i 60 minutter som først var dekket med geit-anti-mus Ig. Etter vask ble <125>I-IL-15 tilsatt til hver brønn, inkubert i 60 minutter ved romtemperatur og vasket fire ganger. Positive celler ble deretter detektert ved hjelp av autoradiografi ved -70<c> ved anvendelse av Kadok X-Omat S film. Positive kloner ble dyrket i svellingskultur og supernatanter ble deretter renset over en protein A-kolonne (Pharmacia). Klonene kalt Ml 10, Ml 11 og Ml 12 ble isomerket deretter som IgGl-monoklonale antistoffer. Hybridomer som produserte monoklonale antistomer Ml 10, Ml 11 og Ml 12 ble deponert i American Type Culture Collection, Rockville, MD, USA (ATCC) 13. mars, 1996 og merket med hhv. aksessjonnr. HB-12061, HB-12062 og HB-12063. Alle deponeringer ble gjort ifølge bestemmelsene i Budapesttraktaten.
Genererte monoklonale antistoffer kanaliseres med hensyn til IL-15-antagonistaktivitet ved anvendelse av CTLL-2-analyse som i det vesentlige er beskrevet av Gillis, et al., Id.
Eksempel 4
Inhibering av IL- 15- aktivitet ved CTLL- 2- analvse
Foreliggende eksempel illustrerer en fremgangsmåte for påvisning av hindringen av signaloverføring av IL-15 gjennom 0- og y-subenhetene av IL-15-reseptorkomplekset frembrakt av antistoffene ifølge oppfinnelsen.
Antagonistaktivitet til monoklonale antistoffer, PEGylert IL-15 og IL-15-muteiner kanaliseres ved anvendelse av en modifisert CTLL-2-celle <3>H-tymidininkorporeringsanalyse (Gillis, et al. Id.). Serielle fortynninger av antagonister kan utføres i 96-brønners flatbunnede veveskulturplater (Costar, Cambridge, MA) i DMEM-medium (supplert med 5% FCS, NEAA, NaPyruvat, HEPES pH 7,4,2-PS) i et endelig volum på 50 il. En ikke-optimal mengde av IL-15 (endelig konsentrasjon på 20-40 pg/ml) ble så tilsatt til alle analysebrønner (5 il/brønn) etter seriefortynning av prøver og før tilsetning av celler. Vaskede CTLL-2-celler ble tilsatt (ca. 2000 per brønn i 50 il) og platene ble inkubert i 24 timer ved 37°C i en fuktig atmosfære på 10% CO2 i luft. Dette ble etterfulgt av fem timers inkubering med 0,5 iCi av 3H-tymidin (25 Ci/mMol, Amersham, Arlington Heights, IL). Kulturene ble så innhøstet på glassfiberfiltere og tellet ved hjelp av overstrømsgassionisering, enten på en multidetektor direkte P-teller (Matrix 96, Packard Instrument Company, Meridien, CT) på en P-scintileirngsteller. Antallet per minutt (CPM) generert ved hjelp av analysen ble forandret til prosent inhibering og de prosentvise inhiberingsverdiene ved hver titrert antagonistprøve ble anvendt for å kalkulere antagonistaktivitet i enheter/ml.
Det var nødvendig å nøytralisere 40 pg/ml av IL-15 i en CTLL inhiberingsanalyse for å vise datene som viser konsentrasjonen i tabell I under. Tabell II under viser aktiviteten til IL-15 (agonistaktivitet) og IL-15-antagonister i CTLL og CTLL-inhibeirngsanalyser.
Claims (13)
1.
Monoklonalt antistoff dannet mot interleukin-15 (IL-15) karakterisert ved SEQ ID NO:
1 eller SEQ ID NO: 2 som forhindrer at IL-15 overfører et signal gjennom en av J3- eller y-subenhetene til IL-15-reseptorkomplekset.
2.
Monoklonalt antistoff mot interleukin-15 (IL-15) karakterisert ved SEQ ID NO: 1 eller 2 som forhindrer at IL-15 overfører et signal gjennom en av P- eller y-subenhetene til IL-15-reseptorkomplekset, der det monoklonale antistoffet interfererer med binding av aminosyren Asp<56> i IL-15-molekylet til P-subenheten av IL-15-reseptorkomplekset og/eller aminosyren Gin<156> i IL-15-molekylet til y-subenheten til IL-15-reseptorkomplekset.
3.
Monoklonalt antistoff ifølge krav 1 eller 2, karakterisert ved at antistoffet er humanisert.
4.
Monoklonalt antistoff ifølge et hvilket som helst av kravene 1 til 3,karakterisert ved at antistoffet er transgent.
5.
Farmasøytisk sammensetning karakterisert ved at den omfatter et monoklonalt antistoff ifølge et hvilket som helst av kravene 1 til 4, og en farmasøytisk akseptabel bærer eller fortynningsmiddel.

Monoklonalt antistoff ifølge et hvilket som helst av kravene 1 til 4 for anvendelse ved behandling av organtransplantatavstøtning, graft versus host disease, autoimmun sykdom, reumatoid artritt, inflammatorisk tarmsykdom, lymfom, karsinom, leukemi, rabdosarkom, dermatologisk forstyrrelse, forstyrrelse i gastrointestinalkanalen, insulinavhengig diabetes mellitus, en okular forstyrrelse, idiopatisk nefrotisk syndorm/idiopatisk membranøs nefropati og HTLV-1-indusert leukemi-lymfom.
7.
Monoklonalt antistoff ifølge et hvilket som helst av kravene 1 til 4 for anvendelse ved behandling av reumatoid artritt.
8.
Monoklonalt antistoff ifølge et hvilket som helst av kravene 1 til 4 for anvendelse ved behandling av en dermatologisk forstyrrelse.
9.
Anvendelse av et monoklonalt antistoff ifølge et hvilket som helst av kravene 1 til 4 for fremstilling av et medikament for behandling av organtransplantatavstøtning, graft versus host disease, autoimmun sykdom, reumatoid artritt, inflammatorisk tarmsykdom, lymfom, karsinom, leukemi, rabdosarkom, dermatologisk forstyrrelse, forstyrrelse i gastrointestinalkanalen, insulinavhengig diabetes mellitus, en okular forstyrrelse, idiopatisk nefrotisk syndorm/idiopatisk membranøs nefropati og HTLV-1 -indusert leukemi-lymfom.

Anvendelse av et monoklonalt antistoff ifølge et hvilket som helst av kravene 1 til 4 for fremstilling av et medikament for behandling av reumatoid artritt.

Anvendelse av et monoklonalt antistoff ifølge et hvilket som helst av kravene 1 til 4 for fremstilling av et medikament for behandling av en dermatologisk forstyrrelse.

Anvendelse av et monoklonalt antistoff ifølge et hvilket som helst av kravene 1 til 4 for fremstilling av et medikament for behandling av graft versus host disease.
13.
Anvendelse av et monoklonalt antistoff ifølge et hvilket som helst av kravene 1 til 4 for fremstilling av et medikament for forlengelse av allograft-overlevelse hos en pasient.
