Description
Foreliggende oppfinnelse omhandler nye metalloprotease inhibitorer, en fremgangsmåte for fremstilling av dem og farmasøytiske sammensetninger inneholdende dem.
I den fysiologiske tilstand er syntesen av bindevev i dynamisk likevekt med degraderingen av den ekstracellulære matriks. Den degraderingen skyldes at sinkproteaser (metalloproteaser) utsondret av cellene i den eksisterende matriks: de er, uten å antyde noen begrensning, kol-Tagenaser (MMP-1, MMP-8, MMP-13), gelatinaser eller kolla-genaser av type IV (MMP-2, MMP-9) og stromelysiner (MMP-3).
I normaltilstanden reguleres de katabolske enzymer hva angår deres syntese og deres sekresjon, og hva angår deres ekstracellulære enzymatiske aktivitet, av naturlige inhibitorer, slik som a2-makroglobulin eller TIMP'ene
(vevsinhibitorer av metalloproteinaser), som danner inak-tive komplekser med metalloproteasene.
En vanlig faktor i patologier i hvilke disse enzymene er implisert er en ubalanse mellom aktiviteten av de aktiverte enzymer og de av deres naturlige inhibitorer, konsekvensen av dette er usedvanlig stor vev degradering.
Ukontrollert og akselerert membrandegradering ved resorp-sjon av den ekstracellulære matriks katalysert av metalloproteasene er en parameter vanlig for mange patologiske tilstander, slik som revmatoid artritt, artrose, tumorinvasjon og vekst, inkludert malign spredning og dannelsen av metastaser, ulcerasjoner, aterosklerose, etc.
BB94, en metalloproteaseinhibitor, har nylig utvist anti-tumoraktivitet i klinisk anvendelse, hvor den har vist seg å være aktiv mot eggstokkreft (Becket et al., DDT 1996, 1 (1), 16).
Det kan derfor forventes at en metalloproteaseinhibitor vil gjenopprette likevekten mellom protease og inhibitor og derfor positivt modifisere utviklingen av slike patologier.
Et visst antall metalloproteaseinhibitorer har blitt beskrevet i litteraturen. Det skal mer spesielt nevnes forbindelsene beskrevet i patentbeskrivelsene WO 95/35275, WO 95/35276, EP 606 046, WO 96/00214, EP 803 505, WO 97/20824 og EP 780 386.
Forbindelsene ifølge foreliggende oppfinnelse er ikke bare nye, men har også vist seg å være kraftigere metalloproteaseinhibitorer enn de beskrevet i litteraturen, hvilket gjør dem potensielt nyttige i behandlingen av kreft, revmatiske sykdommer, slik som artrose og revmatoid artritt, aterosklerose, etc.

Fremstillingene gir syntesemellomprodukter for anvendelse i fremstillingen av forbindelsene ifølge oppfinnelsen.
Strukturene av forbindelsene beskrevet i eksemplene og fremstillingene ble bestemt ifølge de vanlige spektrofoto-metriske teknikker (infrarød, NMR, massespektrometri, etc.).
FREMSTILLINGA: Etvl 4-( sulfanvlmetvl) tetrahvdro- 2H- t>vran-4- karboksylat
TRINNA: Etyl 4-[ ( acetylsulfanyl) metyl] tetrahydro- 2H- pyran-4- karboksylat
Under argon løses 47 g trifenylfosfin i 350 ml tetrahydrofuran (THF). Etter kjøling til 0°C tilsettes 34,9 ml diisopropylazodikarboksylat (DIAD) til denne løsningen. Etter 30 minutters røring tilsettes en løsning inneholdende 89 mmol etyl 4-(hydroksymetyl)tetrahydro-2H-pyran-4-karboksylat og 12,8 ml tioeddiksyre i 300 ml THF.
Etter røring natten over ved romtemperatur og inndampning til tørrhet, tas residuet opp i eter. Etter filtrering, dampes filtratet inn og gir det forventede produkt i form av en olje, som renses ved kromatografi over en silikakolonne ved anvendelse av en blanding av diklormetan/etylacetat (90/10) som eluent.
TRINN B: Etyl 4-( sulfanylmetyl) tetrahydro- 2H- pyran- 4- kar-boksylat 92 ml av en 2,2 N saltsyreløsning i etanol settes til 67,4 mmol av forbindelsen oppnådd i det foregående trinn oppløst i 50 ml etanol.
Etter røring natten over dampes det hele inn til tørrhet for å gi det forventede produkt i form av en olje.
FREMSTILLING B: 5- brom- 2-( 4- pyridyl) benzofuran
TRINN A: ( 5- brom- 2- hydroksybenzyl) trifenylfosfoniumbromid
169 g trifenylfosfinbromid settes til 490 mmol 4-brom-2-(hydroksymetyl)fenol suspendert i 500 ml acetonitril. Det hele varmes ved 100°C i 2 timer. Etter kjøling filtreres
bunnfallet som har blitt dannet og tørkes for å gi det forventede produkt.
Smeltepunkt: 260°C
TRINN B: 5- brom- 2-( 4- pyridyl) benzofuran
256 ml trietylamin settes til 243 g av produktet oppnådd i det foregående trinn i 2 liter toluen i nærvær av 90,1 g isonikotinoylklorid. Det hele varmes ved 100°C i 24 timer. Etter kjøling filtreres bunnfallet som har blitt dannet fra og gir det forventede produkt etter omkrystallisering fra etylacetat.
Smeltepunkt: 160°C
FREMSTILLING C: 2-( 4- pyridyl)- l- benzofuran- 5- sulfonylklorid TRINN A: {[ 2-( 4- pyridyl)- l- benzofuran- 5- yl] sulfonyl} litium
n-Butyllitium settes, ved -72°C til 60,2 mmol av forbindelsen oppnådd i fremstilling B suspendert i tetrahydrofuran. Etter 90 minutter ved -72°C føres en strøm av S02 gjennom blandingen i 1 time. Etter 2 dager ved romtemperatur filtreres faststoffet som har blitt dannet fra og renses med eter for å gi det forventede produkt.
TRINN B: 2-( 4- pyridyl)- l- benzofuran- 5- sulfonylklorid
59 mmol av produktet oppnådd i det foregående trinn suspenderes i 80 ml diklormetan. Etter kjøling til 0°C tilsettes 5,7 ml sulfurylklorid dråpevis. Etter en natt ved romtemperatur filtreres bunnfallet som har blitt dannet fra og renses med eter for å gi det forventede produkt. Smeltepunkt: 210°C
EKSEMPEL 1: N- hvdroksv-( 5R)- 6-{\ 2 -( 4- pvridvl)- 1- benzofuran-5- yl] sulfonyl}- 4. 5. 6, 7- tetrahvdrofuro[ 2. 3- c] pyridin- 5-karboksamid hvdroklorid
TRINN A: ( 5R)- 6-{ T2-( 4- pvridyl)- l- benzofuran- 5- yll sulfonyl}- 4, 5, 6. 7- tetra- hydrofuro[ 2, 3- c] pyridin- 5- karboksylsyre
. tert- butylester
30 mmol (5R)-4,5,6,7-tetrahydrofuro[2,3-c]pyridin-S-karboksylsyre-tert-butylester hydroklorid plasseres i 150 ml pyridin. 30 mmol 2-(4-pyridyl)-1-benzofuran-5-sulfonylklorid, beskrevet i fremstilling C, tilsettes deretter ved romtemperatur og det hele varmes ved 60°C natten over.
Etter fjerning av pyridinet ved inndampning, opptak av residuet i diklormetan, vasking med vann, tørking og inndampning, oppnås det forventede produkt i form av en olje, som renses ved kromatografi over silika ved anvendelse av en blanding av diklormetan/etanol (98/2) som eluent.
TRINN B: ( 5R)- 6-{ \ 2 -( 4- pvridyl)- 1-( benzofuran- 5- vllsulfon-yl}- 4. 5. 6, 7- tetrahydrofuro[ 2. 3- c] pyridin- 5- karboksylsyre
2,4 ml anisol settes til 22 mmol av esteren oppnådd i det foregående trinn oppløst i 250 ml diklormetan. Det hele avkjøles til 0°C og 17 ml trifluoreddiksyre tilsettes. Det hele holdes ved romtemperatur over natten. Etter inndampning til tørrhet renses residuet ved kromatografi over silika ved anvendelse av en blanding av diklormetan/metanol (85/15) som eluent for å gi det forventede produkt.
TRINN C; N- allvloksy-( 5R)- 6-{\ 2 -( 4- pyridvl)- 1- benzofuran- 5-yl] sulfonyl}- 4, 5. 6. 7- tetrahydrofuro[ 2. 3- c] pyridin- 5- karboksamid
Til en løsning, kjølt til 0°C, inneholdende 12 mmol av sy-ren oppnådd i det foregående trinn i 150 ml diklormetan, tilsettes 10,3 ml diisopropyletylamin, 1,65 g 1-hydroksybenzotriazol, en løsning inneholdende 1,6 g O-allylhydroksylamin-hydroklorid i 50 ml dimetylformamid og 4,65 g O-benzotriazolyl-tetrametylisouronium tetrafluorbo-rat (TBTU). Det hele holdes ved romtemperatur over natten. Etter inndampning til tørrhet tas residuet opp i diklormetan. Etter vasking med vann, tørking og inndampning oppnås et residu som gir det forventede produkt etter rensing over en silikakolonne ved anvendelse av en blanding av diklormetan/etanol/ammoniakk (98/2/0,2) som eluent.
TRINN D: N- hydroksy-( 5R)- 6-{[ 2-( 4- pyridvl)- 1- benzofuran- 5-yl] sulfonyl}- 4. 5. 6, 7- tetrahydrofuro[ 2. 3- c] pyridin- 5- karboksamid hydroklorid
300 mg av Pd katalysatoren (PPh3)2Cl2 og 1,5 ml eddiksyre settes til 8,55 mmol av produktet oppnådd i det foregående trinn i 150 ml diklormetan. Etter 5 minutter tilsettes 4,9 ml tributyltinnhydrid. Det hele holdes ved romtemperatur i 30 minutter og dampes deretter inn. Etter at residuet har blitt tatt opp i acetonitril, tilsettes 20 ml IN saltsyre og det hele fortynnes med vann. Den vandige fasen vaskes med eter og lyofiliseres deretter for å gi det forventede produkt.

EKSEMPEL 2: N- hydroksv-( 3S)- 2. 2- dimetvl- 4- f[ 2 -( 4- pyridvl)-l- benzofuran- 5- yllsulfonyl}- 3- tiomorfolinkarboksamid TRINN A: 4-{[ 2-( 4- aminofenyl)- l- benzofuran- 5- yllsulfonyl}-2. 2- dimetyl- 3- tiomorfolinkarboksylsyre tert- butyl( dimetyl)-silylester 58,7 mmol 2,2-dimetyl-3-tiomorfolinkarboksylsyre tert-butyl (dimetyl) silylester løses i 500 ml vannfri diklormetan. Ved -20°C tilsettes 16 ml N-metylmorfolin, fulgt av 57,5 mmol 2-(4-pyridyl)-1-benzofuran-5-sulfonylklorid beskrevet i fremstilling C. Det hele blir rørt i 48 timer ved romtemperatur og tømmes deretter over i 300 ml vann. Etter dekantering, vasking med vann, tørking og inndampning, oppnås det forventede produkt i form av en olje. TRINN B: 4-{[ 2-( 4- aminofenyl)- l- benzofuran- 5- yl}- 2. 2- dimetyl- 3- 1 iomorfolinkarboksylsyre 33 g av forbindelsen oppnådd i det foregående trinn løses i 400 ml vannfri metanol. Det hele reflukseres i 2 timer og dampes deretter inn. Det forventede produkt oppnås ved krystallisasjon av residuet fra eter.
TRINN C: N-( allyloksy)- 4-{[ 2-( 4- aminofenyl)- 1- benzofuran- 5-yll sulfonyl}- 2. 2- dimetyl- 3- tiomorfolinkarboksamid
Det forventede produkt oppnås ifølge fremgangsmåten beskrevet i trinn C i eksempel 1 ved å starte fra produktet beskrevet i det foregående trinn.
TRINN D: N- hydroksy-( 3S)- 2. 2- dimetvl- 4-{\ 2 -( 4- pvridyl)- 1-benzofuran- 5- yll- sulfonyl}- 3- tiomorfolinkarboksamid
Til 10,2 mmol av forbindelsen beskrevet i det foregående trinn oppløst i 70 ml diklormetan, tilsettes 360 mg av Pd katalysatoren (PPh3)2Cl2 og 1,75 ml eddiksyre, fulgt 5 minutter senere av 5,8 ml tributyltinnhydrid. Etter 20 minutters røring, tilsettes 70 ml eter. Det uløselige stoffet filtreres fra og vaskes med eter, tas opp i en blanding av acetonitril/vann (50/50) og gir, etter lyofi-lisering, det forventede produkt.

23,9 mmol 5-brom-2-(4-pyridyl)benzofuran beskrevet i fremstilling B, 7,32 g av forbindelsen beskrevet i fremstilling A, 431 mg tris-(dibenzylidenaceton)dipalladium og 1,05 g 1,1'-bis(difenyl)fosfinoferrocen plasseres i 75 ml N-metyl-pyrrolidon. Det hele varmes ved 100°C i 48 timer. Etter inndampning, tas residue opp i etylacetat og en mettet natriumkloridløsning. Etter filtrering, ekstrahering med etylacetat og inndampning, renses residuet ved kromatografi over en silikakolonne ved anvendelse av en blanding av diklormetan/etylacetat (9/1) som eluent. Det forventede produkt krystalliserer deretter ut.
Smeltepunkt; 92°C
TRINN B: 4-{(\ 2 -( 4- pyridvl)- 1- benzofuran- 5-vl] sulfanyl} metyl} tetrahydro- 2H- pyran- 4- karboksylsvre
7,5 g av esteren oppnådd i det foregående trinn plasseres i 200 ml 6N saltsyre. Det hele reflukseres over natten. Etter kjøling justeres løsningens pH til 7 ved tilsetningen av natriumhydroksid. Bunnfallet som dannes filtreres fra og
vaskes med vann for å gi det forventede produkt.
Smeltepunkt: 227°C
TRINN C: 4-{{[ 2-( 4- pvridyl)- 1- benzofuran- 5-yllsulfonyl} metyl} tetrahydro- 2H- pyran- 4- karboksylsyre 18 mmol av forbindelsen oppnådd i det foregående trinn plasseres i 116 ml vann og 140 ml acetonitril. Det hele avkjøles i et isbad og 17,65 g okson tilsettes i porsjoner. Blandingen etterlates ved romtemperatur i 48 timer. Etter inndampning, justeres pH til 7 og det forventede produkt feller ut.
Smeltepunkt: 217°C
TRINN D: 4-{{[ 2-( 4- pvridyl)- 1- benzofuran- 5-yl] sulfonyl} metyl}- N-( allyloksy)- tetrahydro- 2H- pyran- 4-karboksamid
Det forventede produkt oppnås ifølge fremgangsmåten beskrevet i trinn C i eksempel 1 ved å starte fra forbindelsen beskrevet i det foregående trinn.

FARMAKOLOGISK STUDIE AV FORBINDELSENE IFØLGE OPPFINNELSEN
EKSEMPEL A: Enzymatisk inhibisjon av metalloproteaser
Seks rekombinante humane enzymer, MMP-1 (interstitial kollagenase), MMP-2 (gelatinase A), MMP-3 (stromelysin 1), MMP-8 (neøtrofil kollagenase), MMP-9 (gelatinase B) og MMP-13 (kollagenase 3) blir aktivert med APMA (4-aminofe-nylkvikksølvacetat).
De enzymatiske testene på MMP-1, -2, -8, -9 og -13 utføres ved anvendelse av det følgende peptidomimetiske substrat:
DnpProGhaGlyCys(Me)HisAlaLys(Nma)NH2,
som spaltes mellom glysinet og cysteinet for å gi et fluorescerende produkt beskrevet av D.M. BICKETT et al.
(Anal. Biochem., 212, 58-64, 1993).
Den enzymatiske testen på MMP-3 utføres ved anvendelse av det følgende peptidomimetiske substrat:
McaArgProLysProTyrAlaNvaTrpMetLys(Dnp)NH2,
som spaltes mellom alanin og norvalin for å gi et fluorescerende produkt beskrevet av H. NAGASE et al. (J. Biol. Chem., 269, 20952-20957, 1994).
Reaksjonene, utført i en buffer av 50 mM Tris, 200 mM NaCl, 5 mM CaCl2, 0,1% Brij 35 ved pH 7,7, blir initiert ved anvendelse av 20 /xM substrat i et totalvolum på 100 fil ved 37°C.
Fluorescensen oppnådd etter seks timer avleses i en 96-brønns plate i et fluorimeter utstyrt med en kombinasjon av 360 nm og 460 nm filtre for eksitering og emisjon. Forbindelsene ifølge oppfinnelsen har IC50 verdier fra 10"<10 >til 10"<8>M for alle MMP'ene med unntak av MMP-1. Kollagena-sene MMP-13 og MMP-8 utviser en spesifisitet på en faktor 1000 sammenlignet med kollagenase MMP-1.
EKSEMPEL B: In vitro- deqraderinq av bruskmatriksen
Forbindelsene ifølge oppfinnelsen ble undersøkt i en modell for skade på bruskmatriksen indusert av IL-lp. Under-søkelsene, utført på kaninbrusk, omhandler: - på en side, degraderingen av kollagen: kolorimetrisk analyse, ifølge teknikken til Grant (GRANT R.A. Estima-tion of OH-proline by the autoanalyser, J. Clin. Path., 17, 685, 1964), av OH-prolin andelen frigitt av vevet i kontakt med IL-ip (10 ng/ml) og plasmin (0,1 U/ml) i 2 dager; - på den andre side, degraderingen av proteoglykaner: radio-isotopisk måling av andelen glykosaminoglykaner frigitt etter 24 timer's stimulering med IL-ip (10 ng/ml) av vevet formerket med <35>S04, i løpet av 24 timer i kontakt med APMA (5xlO"4M) .
Forbindelsene ifølge oppfinnelsen ble studert ved tilset-ning til kultiveringsmediet i testens 3 dager. For konsentrasjoner fra IO"<9> til 10"<6>M, inhiberte de sterkt degraderingen av kollagen og av proteoglykaner.
EKSEMPEL C: In vitro angiogenese
Porsjoner av torakal aorta fra hann Fischer 344-rotter som var fra 8 til 12 uker gamle ble dyppet ned i en type I kollagengel ifølge fremgangsmåten til Nicosia og Ottinetti (Lab. Invest., 63, 115, 1990). Etter fem dager med dyrking i et medium uten serum, blir preparatene undersøkt under et mikroskop og dannelsen av pseudo-blodkar kvantifiseres uttrykt som vaskulær tetthet etter digitalisering og bildeanalyse.
Ved konsentrasjoner fra 10"<9> til 10"<6>M blokkerer forbindelsene ifølge oppfinnelsen selektivt dannelsen av pseudo-blodkar fra endoteliale celler, uten å påvirke fibroblas-tiske celler.
