Description
Oppfinnelsen vedrører cyaninfargestoffer av den generelle formel som angitt i krav 1. Slike cyaninfargestoffer kan brukes som kontrastmidler i fluorescens- og transilluminasjonsdiagnostikken i NIR-området.
For å kunne fastslå sykdommer er man i vesentlig grad av-hengig av om det lykkes å oppnå informasjoner om strukturer og deres forandringer i de primært ikke-tilgjengelige dypere vevsjikt. Dette kan i tillegg til føling, blottlegging, eller punktering, skje ved de moderne avbildningsteknikker, så som røntgen, magnetresonanstomografi eller ultralyddi-agnostikk.
Da biologisk vev har en relativt høy gjennomtrengelighet for langbølget lys i bølgelengdeområdet fra 650 - 100 nm, har dermed diagnostikeren en helt annen fremgangsmåte for fremstilling av vevsbilder til rådighet. Den kjensgjerning at nærinfrarødt lys kan trenge gjennom vev i flere centime-ters dybde utnyttes i transilluminasjonsavbildning. Denne teknikk har hittil gjort det mulig å diagnostisere beten-nelser i nesebihulene og kjevehulene eller å påvise væske-opphopninger eller blod i overflatenære vevsområder (Beuthan J., MUller G.) Infrarotdiaphanoskopie, Med. Tech. 1 (1992) 13-17).
Forsøk for å påvise brysttumorer har hittil forløpt util-fredsstillende {Navarro, G.A.; Profia, A. E.; Contrast on diaphanography of the breast; Med. Phys. 150 (1988) 181-187; Aspegren, K.; Light Scanning Versus Mammography for the detection of Breast Cancer in Screening and Clinical Practice, Cancer 65 (1990) 1671-77), men på grunn av de nyeste apparattekniske utviklinger ser det mer lovende ut (Klingenbeck J.; Laser-Mammography with NTR-Light; Gynåkol. Geburtsh.- Rundschau 33 Suppl. 1 (1993) 299-300); Benaron D.A.; Optical Imaging reborn with technical ad-vances, Diagnostic Imaging (1994) 69-76).
I tillegg til påvisningen av ikke-absorberende stråling kan også fluorescensstrålingen som emitteres etter bestråling med nærinfrarødt lys gi vevsspesifikke informasjoner. Den såkalte autofluorescens utnyttes for å skille mellom aterosklerotisk og normalt vev. {Henry, P.D., et al., Laser-Induced Autofluorescence of human Arteries, Circ. Res. 63
(1988) 1053-59).
Det vesentlige problem ved utnyttelse av nærinfrarød stråling er den overordentlig sterke brytning av lyset, slik at selv med forskjellige fotofysikalske egenskaper avtegner en skarp avgrenset gjenstand seg bare uskarpt fra sine omgi-velser. Problemet øker med voksende avstand fra overflaten og kan som hovedsakelig begrensende faktor bli anselig, både ved transilluminasjonen og ved påvisningen av fluorescens st rål ing.
Til forbedring av differensieringen mellom normalt og sykt vev kan egnede fluorescensfargestoffer bidra, da de anriker seg i sykt vev (spesielt tumorer) og har en spesifikk ad-sorpsjons- og emisjonsadferd. Endringen av det (brutte) innstrålte lys som bevirkes ved adsorpsjon av fargestoffet, eller fluorescensen som induseres ved aktiveringsstrålingen, påvises og gir de egentlige vevsspesifikke informasjoner .
Eksempler på anvendelse av fargestoffer for in vivo-diagnostikk på mennesker er overvåking i blodet med fotometriske metoder for å påvise fordelingsrom, blodstrøm eller metabo-liserings- og utskillelsesfunksjoner, samt synliggjøring av gjennomsiktige strukturer i øyet (oftalmologi). Foretrukne fargestoffer for disse anvendelser er indocyaningrønt og fluorescein (Googe, J. M. et al., Intraoperative Fluorescein Angiography; Ophthalmology, 100 (1993), 1167-70).
Indocyaningrønt (cardiogrønt) anvendes for måling av lever-funksjon, hjertetid- og slagvolumer, samt organblodomløpet og det perifere blodomløpet. ( I. Med. 24 (1993) 10-27) og utprøves som kontrastmiddel for påvisning av tumorer. Indo-cyaningrønt bindes 100% til albumin og frigis i leveren. Fluorescenskvanteutbyttet er lavt i vandig miljø. LDso-ver-dien (0,84 mmol/kg) er tilstrekkelig høy, men det kan frem-kalles sterke anafylaktiske reaksjoner. Indocyaningrønt er ustabilt i løsning, og det kan ikke anvendes i saltholdige medier, da det skjer en utfeining.
For å lokalisere og avbilde tumorer har det hittil blitt anvendt fotosensibilatorer (bl.a. hematoporfyrinderivater, fotofrin II, benzoporfyrin, tetrafenylporfyrin, klorin, ftalocyanin) som var ment for anvendelse i den fotodynami-ske terapi (PDT) (Bonnett R.; New photosensitizers for the photodynamic therapy of tumors, SPIE vol. 2078 (1994)). De angitte forbindelser har den felles ulempe at de oppviser dårlig absorpsjon i bølgelengdeområdet fra 650 til 1200 nm. Den fototoksisitet som er nødvendig for PDT, er forstyr-rende for rent diagnostiske formål. Ytterligere skrifter vedrørende dette tema: U. I U. S.- PS 4943239 beskrives tallrike apparatanordninger for påvisning av brystkreft, og som kontrastforhøyende absorp-sjonsfargestoff nevnes det kjente fluorescein, fluorescamin og riboflavin. Disse fargestoffer har den felles ulempe at de absorberes i det synlige bølgelengdeområde fra 400-600 nm, i hvilket lysgjennomsiktigheten av vev er meget liten.
I DE- QS 4136869 beskrives et apparat for fluorescenspåvis-ning av vevsomrader som er anriket med fluorescerende substanser. Substansene er bakterioklorofyll og derivater, samt naftalocyanin. Disse strukturer utmerker seg ved adsorpsjoner i området fra 700-899 nm med ekstinksjons-koeffisienter på inntil 70000 l.mol"<1>. De her angitte forbindelser har i tillegg til fluorescensegenskapen den evne at de ved bestråling kan generere singulettoksygen, og dermed virker de cytotoksisk (fotosensibilatorer for foto-dynamisk terapi). Denne fotosensibiliserende virkning er i høyeste grad uønsket for et rent, virkningsfritt diagnos-tikum.
Dessuten er syntesen av bakterioklorofyllforbindelsene både omstendelig og dyr, da det kreves naturprodukter som ut-gangssubstanser; naftalocyaninene utmerker seg ofte ved meget liten fotostabilitet. De kjente forbindelser i disse klasser har liten vannoppløselighet, og syntesen av enhet-lige, hydrofile derivater er omstendelig.
WO 84/ 04665 beskriver en in-vivo-fremgangsmåte for fluore-scenspåvisning av tumorer under anvendelse av fotosensi-bilisatorene hematoporfyrin og -derivat. (Hp og HpD), uro-og copro- og protoporfyrin og tallrike mesosubstituerte porfyriner, samt fargestoffene riboflavin, fluorescein, acridin-orange, berberinsulfat og tetracykliner. De oven-nevnte fotofysikalske og farmakokjemiske krav oppfylles ikke av de nevnte substanser.
Folli et al., Cancer Research 54, 2543-2649 (1994) beskriver et med et cyaninfargestoff forbundet monoklonalt antistoff som ble anvendt for påvisning av en subkutant implantert tumor. Påvisningen av dypereliggende patologiske prosesser gjør det riktignok nødvendig å anvende ytterligere forbedrede fargestoffer. Videre synes det som om høyere fargestoffdoseringer er uegnet for anvendelse av antistoffer som bærere på grunn av forventede bivirkninger.
Cyaninfargestoffer og beslektede polymetinfargestoffer finner anvendelse også som bestanddeler i fotografiske sjikt. Slike fargestoffer trenger ingen luminescensegen-skaper. Cyaninfargestoffer med luminescens-(fluorescens-) egenskaper er blitt syntetisert for anvendelse i fluore-scensmikroskopi og gjennomstrømningscytometri og er blitt koblet til biomolekyler, f.eks. forbindelser med jodacetyl-grupper som spesifikke markører for sulfhydrylgrupper i proteiner (Waggoner, A.S. et al.; Cyanine dye Labeling Reagents for Sulfhydryl Groups. Cytometry, 10, (1989) 3-10). På denne måte markeres proteiner og isoleres. Ytterligere litteratureksempler : Cytometry 12 (1990) 723-30; Anal. I DE- OS 39 12 046 tilhørende Waggoner, A.S., beskrives en fremgangsmåte ved merking av biomolekyler med cyanin- og beslektede fargestoffer, så som merocyanin og styrylfarge-stoffer som inneholder minst en sulfonat- eller sulfonsyre-gruppering. Det nevnte skrift vedrører en et-trinns samt to-trinns markeringsfremgangsmåte i et vandig medium, hvorved det finner sted en kovalent reaksjon mellom fargestoff og en amin-, hydroksy-, aldehyd- eller sulfhydrylgruppe på proteiner eller andre biomolekyler.
DE- OS 3828360 vedrører en fremgangsmåte ved merking av antitumorantistoffer, spesielt melanom- og kolonkarsinom-spesifikke antistoffer, med fluorescein og indocyaningrønt for oftalmologiske formål. Dannelsen av indocyaningrønt på biomolekyler er ikke kovalent (fargestoffantistoffkombina-sjon, blanding).
Fremgangsmåtene som er kjent fra teknikkens stand for in-vivo-diagnostikk ved hjelp av NIR-stråling oppviser føl-gelig en rekke ulemper som gjorde det umulig å anvende dem i utstrakt medisinsk diagnostikk.
Den direkte anvendelse av synlig lys eller NIR-stråling er begrenset til overflatenære kroppsområder, og dette henger sammen med den sterke brytning av det innstrålte lys.
Tilsetningen av fargestoffer for forbedring av kontrasten og oppløsningsevnen fremkaller dog en rekke ytterligere problemer. Man må nemlig anbringe målestokker for disse fargestoffene som gjelder generelt for diagnostikk. Således må slike stoffer bare oppvise liten toksisitet, da de for det meste iakttas i høyere doser og også over et lengre diagnosetidsrom. Videre må disse for diagnose egnede fargestoffer være lett oppløselige i vann og tilstrekkelig, dvs. i det minste så lenge diagnosen varer, kjemisk og fotofysi-kalsk stabile. Dessuten settes det krav til stabilitet ved-rørende metaboliseringen i organismen.
Hittil har verken fargestoffer med slike egenskaper eller en egnet fremgangsmåte ved in-vivo-diagnostikk ved hjelp av NIR-bestråling stått til rådighet.
Til grunn for oppfinnelsen ligger derfor den oppgave å til-veiebringe et fargestoff egnet for in-vivo-diagnostikk som overvinner ulempene i teknikkens stand.
Forbindelsene ifølge oppfinnelsen, utmerker seg ved at de absorberer i bølgelengdeområdet fra 650 til 1200 nm og fluoriserer, de har absorpsjonskoeffisienter fra ca. 100.000 1'mol-1 •cm-1 og høyere og, såfremt fluorescens er ønsket, har de fluorescenskvanteutbytter større enn 5%, de har en tilstrekkelig vannoppløselighet, fordragelighet og in vitro- og in vivo- samt fotostabilitet. De utskilles på kortest mulig tid nesten fullstendig. Forbindelsene ifølge oppfinnelsen er lette og økonomiske å syntetisere i færrest mulig reaksjonstrinn fra kommersielt tilgjengelige utgangs-materialer.
Cyaninfargestoffene ifølge oppfinnelsen tilføres vevene ved in vivo-diagnostikk for eksempel ved intravenøs injeksjon, og lys fra det synlige til nærinfrarøde område fra 650 til 1200 nm innstråles. Den ikke-absorberte stråling og fluorescensstrålingen registreres enkeltvis eller begge sammen eller begge tidsforskjøvet mot hverandre. Av de erholdte data oppnås et syntetisk bilde.
Opptaket av fluorescensbilder kan skje ifølge forskjellige metoder. Foretrukket er de metoder hvor vevet bestråles i en stor flate og fluorescensinformasjonen oppløses på stedet ved opptak med et CCD-kamera, eller vevsarealet som skal avbildes, avføles med en lysstråle som er innbundet i en optisk lysleder, og de erholdte signaler omsettes tall-messig til et bilde. Lyset innstråles i et smalt bånd i området for henholdsvis absorpsjonsmaksimum og fluorescens-aktiveringsbølgelengden for de forbindelsesmessige forbindelser. Den ikke-absorberte bestråling kan likeledes påvises på den beskrevne måte, og de erholdte signaler kan bearbeides.
Innstrålings- og iakttagelsesvinkelen kan velges ifølge de anatomiske forhold og den optimale kontrast fra tilfelle til tilfelle. Ved substraksjon av bildene før og etter fargestofftilsetningen kan metodens følsomhet ytterligere forbedres. Ved vurdering av tidsforløpet for de fargestoff-betingede forandringer kan det erholdes ytterligere informasjoner som er nyttige for diagnostikken.
De måletekniske fremgangsmåter er kjent for fagmannen. Fagmannen er videre kjent med hvilke apparatmessige parametere som må velges for å oppnå optimale nedtegnelses- og bear-beidelsesbetingelser ved på forhånd gitte absorpsjons, henholdsvis fluorescensbølgelengder for de oppfinnelsesmessig anvendte fargestoffer med den generelle formel I.
Ved hjelp av de ovenfor beskrevne teknikker kan man oppsøke selv små objekter med noen få mm<3> volum i dypere sjikt av vevet eller i ikke-transparente kroppsvæsker. På grunn av lysbrytninger og den dermed forbundne begrensede oppløsning forblir det vanskelig å bestemme den nøyaktige form og størrelse av objektet, hvilket for noen viktige diagnostiske spørsmålsstillinger heller ikke er nødvendig. Overraskende viste et etter applikasjon av et cyaninfargestoff utført fluorescenslysopptak av en naken mus med et CCD-kamera en fluorescensintensitet som var tusen ganger høyere enn med et tilsvarende dosert porfyrin.
Den beskrevne fremgangsmåte under anvendelse av de oppfinnelsesmessige forbindelser egner seg spesielt for fremstilling av bilder av vev som ikke er patologisk forandret, systemiske sykdommer, tumorer, blodkar, aterosklerotisk plaque og av perfusjon og diffusjon.
Cyaninfargestoffer ifølge oppfinnelsen anbringes i vevet på forskjellige måter. Spesielt foretrukket er intravenøs til-førsel av fargestoffene.
Doseringen kan være meget forskjellig, alt etter anvendel-sesformålet. Målet er en påviselig fargestoffkonsentrasjon i vevsområdet som skal diagnostiseres, hvorved en konsentrasjon på 1-100 ug/l i vevet eller i kroppsvæsken for det meste er tilstrekkelig. Dette mål oppnås ved direkte injeksjon i små kroppshulrom eller små blod- eller lymfekar generelt ved administrasjon av 0,1-100 mg av det aktuelle fargestoff i 0,1-10 ml bærervæske. Foretrukket er i dette tilfelle 1-10 mg fargestoff. For farging av blodkarene eller for påvisning av spesielle vev eller strukturer etter intravenøs injeksjon er det for det meste nødvendig med høyere doseringer (større eller lik 100 mg). Den øvre gren-se for doseringen fastsettes bare ved de enkelte substan-sers og tilberedningers fordragelighet.

Oppfinnelsen angår videre anvendelse av slike cyaninfargestoffer ved fremstilling av NIR-kontrastmidler som angitt i krav 3, samt slike midler for in vivo-diagnostikk som angitt i krav 4.
En vesentlig egenskap hos de oppfinnelsesmessige forbindelser og de oppfinnelsesmessig anvendte forbindelser består i at de har en hydrofilisitet som kjennetegnes ved en n-oktanol/vann-fordelingskoeffisient på mindre enn 2,0 (fordelingskoeffisient n-oktanol/0,01 M Tris-buffer med 0,9% kok-salt, innstilt på pH 7,4, begge faser mettet mot hverandre) . Forbindelsene har ingen utpregede, for et rent diag-nostikum uønskede, fotosensibiliserende eller fototoksiske egenskaper. De er videre lett fordragelige og de utskilles.
Ved disse egenskaper atskiller forbindelsene seg fra de tidligere kjente fargestoffer som er blitt foreslått eller anvendt i in vivo-diagnostikken. Fluorescenskvanteutbyt-tene, som spesielt hos cyaninfargestoffet synker drastisk i vandige medier ved aggregering, kan sammenlignes med dem som kan måles i apolare løsningsmidler, da en aggregat- og micelledannelse undertrykkes på grunn av den forhøyde vann-oppløselighet og de hydrofile gruppers plassbehov. Proteinaffiniteten av en gruppe av de foretrukne forbindelser er liten, den farraakokinetiske atferd er tilnærmelses-vis den samme som for f.eks. insulin eller sakkarose.
Overraskende viste det seg at også hos disse forbindelser, til tross for den enkle molekyloppbygning, skjer det en diagnostisk tilstrekkelig anrikning i bestemte strukturer i organismen, f.eks. også i tumorer, og etter jevn fordeling av fargestoffet i organismen finner eliminasjonen fra tu-morområdene forsinket sted sammenlignet med det omliggende vev.
Fordrageligheten av substansene er meget høy. Spesielt foretrukket er substanser med LD5o-verdier på mer enn 0,5 mmol/kg kroppsvekt beregnet på et enkelt fargestoffmolekyl.
De oppfinnelsesmessige og oppfinnelsesmessig anvendte forbindelser utmerker seg ved høy in vitro- og in vivo- samt fotostabilitet. Ved henstand av en vandig løsning i dags-belyste rom er spesielt de foretrukne forbindelser etter 2 dager til 98% og etter 12 til 70% uforandret.
De beskrevne fotofysikalske og farmakokinetiske egenskaper hos de oppfinnelsesmessige og oppfinnelsesmessig anvendte forbindelser atskiller seg fra det eneste cyaninfargestoff som er tillatt for anvendelse på pasienter, indocyaningrønt (cardiogreen).
Det er mulig å syntetisere cyaninfargestoffer som absorberer ved bølgelengder på 650 til 1200 nm med høye ekstink-sjonskoeffisienter og fluoriserer med høy virkningsgrad. Syntesen av de oppfinnelsesmessige og oppfinnelsesmessig anvendte cyaninfargestoffer skjer overveiende ifølge litte-raturkjente metoder, f.eks. F.M. Hamer i The Cyanine Dyes and Related Compounds, John Wiley and Sons, New York, 1964; Cytometry, 10, (1988) 3-10; 11 (1990) 418-430; 12 (1990) 723-30; Bioconjugate Chem. 4 (1993) 105-11, Anal. Biochem., 217 (1994) 197-204; Tetrahedron 45 (1989) 4845-66, Euro-peisk patentsøknad 0 591 820 Al.
En gjenstand for foreliggende oppfinnelse er videre også diagnostiske midler som inneholder forbindelser med den generelle formel V.
Disse midler fremstilles ifølge kjente metoder, eventuelt under anvendelse av vanlige hjelpe- og/eller bærerstoffer samt fortynningsmidler og lignende. Dertil hører fysiologisk akseptable elektrolytter, buffere, detergenser og substanser for justering av osmolaliteten samt for forbedring av stabiliteten og løseligheten, som f.eks. cyklo-dekstrin. Ved de forholdsregler som normalt anvendes i farmasien sørges det for sammensetningenes sterilitet ved fremstillingen og spesielt før applikasjonen.

Av 5-karboksymetyl-2-[7-[5-karboksymetyl-l,3-dihydro-3,3-dimetyl-1- (4-sulfobutyl) -2H-indol-2-yliden] -1,3, 5-heptatri-enyl]-3, 3-dimetyl-l-(4-sulf obutyl)-3ff-indolium, indre salt, monokaliumsalt fremstilles ifølge kjente fremgangsmåter di-N-hydroksysuccinimidesteren ( Cytometry 11 (1990) 418-430). Til en løsning av 0,5 g (0,51 mmol) av disuccinimididyl-esteren i 5 ml DMF tilsettes 0,16 g (1,22 mmol) asparagin-syre i 1 ml DMF. Reaksjonsblandingen røres i 48 t ved værelsestemperatur, og produktet utfelles ved tilsetning av eter. Rensing på RP-18 (LiChroprep, 15-25u, H20:MeOH 99:1 til 1:1) og påfølgende frysetørking, samt tørking i 24 t ved 50°C/0,01 mbar gir 0,27 g (51%) produkt.

Til en løsning av 0,5 g (0,73 mmol) 2-(7-[5-{karboksy-metyl) -1,3-dihydro-3,3-dimetyl-l-etyl-2H-indol-2-yliden]-1,3,5-heptatrienyl]-3,3-dimetyl-l-(4-sulfobutyl)-3ff-indo-lium-N-succinimidylester, indre salt ( Cytometry 11 (1990) 418-430) i 5 ml metanol tilsettes langsomt dråpevis 43 mg (0,65 mmol) 85% hydrazinhydrat i 1 ml metanol ved -10°C og omrøres i 2 timer ved denne temperatur. Reaksjonsløsningen inndampes under vakuum til ca. 3 ml, tilsettes 1 ml isopropanol og oppbevares over natten ved -20°C. De utfelte krystaller suges opp, og tørkes ved en oljepumpe. Man erholder 0,27 g (61%) trikarbocyaninkarboksylsyrehydrazid.
Til en løsning av 0,21 g (0,51 mmol) dietylentriaminpenta-eddiksyremonoetylestermonoanhydrid i 20 ml DMF og 0,2 ml trietylamin tilsettes under omrøring 0,27 g (0,45 mmol) av hydrazidet, blandingen omrøres i 48 timer ved værelsestemperatur. Etter filtrering inndampes løsningsmidlet ved 0,2 mbar, residuet røres med CH2CI2, filtreres og tørkes i høyvakuum. Det erholdte produkt røres i 5 ml 3M vandig NaOH ved værelsestemperatur, deretter justeres med halvkon-sentrert HC1 til pH 2,0, 1 ml isopropanol tilsettes og de krystaller som erholdes etter 18 t henstand ved 4°C, suges av og tørkes i høyvakuum i 25 t ved 60°C. Utbytte 0,23 g (52%) som et mørkt, rødglinsende granulat.

Til en løsning av 800 mg metoksypolyoksyetylenamin (ca. 0,16 mmol; midlere molmasse ca. 5000) i 10 ml CH2CI2 til-setter man en løsning av 0,08 mmol av N,N-disuccinimidyl-esteren fra eksempel 1 i 1 ml DMF, og det omrøres i 24 t ved værelsestemperatur. Det faste stoff som faller ut etter tilsetning av eter, avfiltreres og renses kromatografisk (Sephadex G50-medium, H2O som eluens), utbytte ca. 58% som grønnblått pulver etter frysetørking og tørking over P2Os.

2-[7-[1/3-dihydro-3,3-dimetyl-l-(4-sulfobutyl)-2H-indol-2-yliden]-1,3,5-heptatrienyl]-3,3-dimetyl-5-(metoksypolyoksy-etylen)aminokarbonyl-1-(4-sulfobutyl)-3H-indolium, natriumsalt .
0,41 g (0,5 mmol) 2-[7-[1,3-dihydro-3,3-dimetyl-l-(4-sulfo-butyl-1-(4-sulfobutyl)-2H-indol-2-yliden]-1,3,5-hepatri-enyl]-3,3-dimetyl-5-karboksy-l-(4-sulfobutyl)-3H-indolium-N-succinimidylester, natriumsalt røres sammen med 2,3 g metoksypolyoksyetylenamin (0,46 mmol; midlere molmasse 5000) i 18 t 70 ml CH2CI2 ved værelsestemperatur under argon, løsningsmidlet konsentreres i vakuum til halvparten, og produktet isoleres som beskrevet i eksempel 3. Det erholdes 2,1 g produkt som grønnblått pulver.

3-(3-karboksypropyl)-2-[7-[3-(3-karboksypropyl)-1,3-di-hydro-3-metyl-l-(4-sulfobutyl)-2ff-indol-2-yliden]-1,3,5-heptatrienyl]-3-metyl-l-(4-sulfobutyl)-3tf-indolium, natriumsalt
6,5 g (50 mmol) fenylhydrazinhydroklorid og 8,7 g (55 mmol) 5-metyl-6-oksoheptansyre røres i 50 ml konsentrert eddik-syre 1 t ved værelsestemperatur og 5 t ved 120°C. Etter konsentrering røres residuet med 20 ml vann, og de oppståtte krystaller filtreres vekk og tørkes ved hjelp av en oljepumpe.
Man erholder 6,9 g (83%) brunlige krystaller som suspenderes i 60 ml diklorbenzen og etter tilsetning av 11,6 g (85 mmol) 1,4-butansulfon oppvarmes i 8 t ved 150°C. Etter avkjøling til værelsestemperatur tilsettes 200 ml aceton, og den oppståtte feining filtreres vekk. Fremstillingen av indotrikarbocyaninfargestoffet skjer ved 30 min oppvarming av 5,0 g (13,6 mmol) indolenin og 1,9 g (6,8 mmol) glutaconaldehyddianilhydroklorid i 100 ml eddik-syreanhydrid under tilsetning av 25 ml konsentrert eddik-syre og 2,3 g (27,6 mmol) vannfritt natriumacetat ved 120°C. Felningen som oppnås etter tilsetning av 500 ml eter, renses kromatografisk (i 1,0 g porsjoner, RP-18, LiChroprep, 15-25 u, MeOH:H20 som eluens), og deretter frysetørkes den. Man erholder 2,5g (45%) produkt.

Fremstilling av
2-[[3-[[3-(karboksypropyl)-1,3-dihydro-3-metyl-l-(4-sulfo-butyl) -2H-indol-2-yliden] metyl] -2-hydroksy-4-okso-2-cyklo-buten-l-yliden]metyl]-1,l-dimetyl-3-etyl-lH-benz(e)indol-ium, indre salt
Til en løsning av 1,36 g (8,0 mmol) kvadratsyredietylester og 1,6 ml trietylamin i 12 ml etanol, oppvarmet til 70°C, tilsettes 3,65 g (10,0 mmol) 3-etyl-l,1,2-trimetyl-lH-benz(e)indoliumjodid. Etter 10 min omrøring ved 80°C avkjø-les til 0°C, og de utfelte, rødfarvede krystaller filtreres vekk, vaskes med eter og tørkes i vakuum. Rensing ved kro-matograf i på kiselgel (CH2C12 : AcOH 9:1 til 7:3) gir 1,33 g (46%) 2-etoksy-l-[(3-etyl-l,1-dimetyl-lH-bens(e)indol-2-yliden)-metyl]-cyklobuten-3,4-dion. Dette suspenderes i 15 ml kokende etanol, og det tilsettes under omrøring 0,5 ml 40-% NaOH. Den erholdte løsning røres 5 min ved 80°C, og etter avkjøling til værelsestemperatur tilsettes 5 ml 2N HC1. Det etter konsentrasjon utfelte 1-[(3-etyl-l,1-di-metyl-lH-benz(e)indol-2-yliden)-metyl]-2-hydroksycyklo-buten-3,4-dion (1,30 g) filtreres, tørkes og anvendes uten ytterligere rensing i neste syntesetrinn. Fremstillingen av squarainfargestoffet skjer ved omsetning av 1,30 g (3,9 mmol) av det erholdte kvadratsyrederivat med 1,43 g (3,9 mmol) 3-(3-karboksypropyl)-2,3-dimetyl-l-(4-sulfobutyl)-3H-indolenin. Komponentene oppvarmes i 80 ml toluen og 80 ml 1-butanol i 18 t med vannutskiller og deretter befries de for løsningsmidlene i vakuum. Residuet tilsettes eter, og de oppståtte krystaller avfiltreres eter 16 timers omrøring ved værelsestemperatur og renses kromatografisk (RP-18, LiChroprep, 15-25 u, MeOH:H20 som eluens). Utbytte 0,95 g (35%) .

Fremstilling av
2-[7-[1,3-dihydro-5-[2-[(2,3-dihydroksypropyl)amin]-2 oksoetyl]-3,3-dimetyl-l-(4-sulfobutyl)-2H-indol-2-yliden]-1,3,5-heptatrienyl]-5-[2-[(2,3-dihydroksypropyl)amino]-2-oksoetyl]-3,3-dimetyl-l-(4-sulfobutyl)-3H-indolium, natriumsalt
Til 2,0 g (6,4 mmol) 2,3,3-trimetyl-3H-indol-5-yleddiksyre-succinimidyl-ester i 50 ml CH2CI2 tilsettes 0,84 g (6,4 mmol) 4-aminometyl-2,2-dimetyl-l,3-dioksolan. Etter 5 t omrøring ved værelsestemperatur helles det på 100 ml vann, ekstraheres med CH2CI2, og de organiske faser konsentreres. Etter kromatografisk rensing (kiselgel CH2CI2 ; MeOH 98:2) erholder man 1,86 g (88%) av amidet, som røres i 20 ml diklorbenzen med 1,36 g (10,0 mmol) 1,4-butansulfon i 7 t ved 100°C og i 12 t ved værelsestemperatur. Granulatet som oppstår etter omrøring med 50 ml aceton, avfiltreres og renses kromatografisk (RP-18, LiChroprep, 15-25 u, MeOH:H20 som eluens). Man erholder 0,85 g (28% beregnet på utgangs-forbindelsen) 5-[2,2-dimetyl-l,3-dioksa-4-cyklopentyl)-metyl]amino-2-oksoetyl]-2,2,2-trimetyl-l-(4-sulfobutyl-3H-indolenin.
Omsetningen av fargestoffet skjer ifølge eksempel 4, idet man oppvarmer 10 min ved 120°C. Råproduktet røres i 5 ml MeOH under tilsetning av 100 mg p-toluensulfonsyre 16 t ved værelsestemperatur. Uløselige deler separeres, og filtra-tet oppbevares deretter etter tilsetning av 3 ml isopropanol ved -20°C. Det utfelte pulver renses kromatografisk (RP-18, LiChroprep, 15-25 u, MeOH:H20 som eluens), fryse-tørkes og tørkes 24 t ved 50°C/0,01 mbar. Utbytte 0,32 g (37%) .

Til en bedøvet, tumorbærende nakenmus (Swiss-Nude, tumor LS 174 T i den høyre bakpart) ble det administrert 2-[7-(1,3-dihydro-3,3-dimetyl-5-(metoksykarbonyl)-1-(4-sulfobutyl)-2H-indol-2-yliden]-1,3,5-heptatrienyl]-3,3-dimetyl-5-(metoksykarbonyl)-1-(4-sulfobutyl)-3H-indolium, natriumsalt i en dose på 3,8 umol/kg kroppsvekt i.v.
De laserinduserte fluorescensopptak ble utført før og til forskjellige tider etter substansapplikasjonen med en fluorescens-imaginator (Physikalisch-Technische Bundesan-stalt, Berlin Charlottenburg). Aktiveringen skjedde med monokromatisk laserlys ved 740 nm ved utkobling av strål-ingen over et lysledersystem. Aktiveringsstrålingen under 740 nm ble fjernet ved et kantfilter, og det laserinduserte fluorescenslys over 740 nm ble opptatt med et CCD-kamera (Charge Coupled Device), og dataene ble lagret som svart/- hvitt-bilder.
Opptakssekvensen på figur 1 viser tydelig den generelle økning av fluorescensintensiteten etter substansapplikasjonen (Bilde A, B). Etter 30 s iakttas en jevnt fordelt intensitet med forhøyede verdier i lever-lunge-området og tumoren (B). I det ytterligere tidsforløp inntil 1 t (C, D, E) fordeler substansen seg i dyret økende grad. Etter 18 t kan man iaktta i tumoren (den høyre bakpart) en fluorescensintensitet som i forhold til resten av kroppen er tydelig forhøyet.
Figur 1 viser fluorescenslysopptak (svart/hvitt-bilde) av en nakenmus (Swiss Nude) til forskjellige tidspunkter etter i.v.-applikasjon av 3,8 umol/kg kroppsvekt av 2-[7-[1,3-dihydro-3,3-dimetyl-5-metoksykarbonyl)-1-(4-sulfobutyl)-2H-indol-2-yliden]-1,3,5-heptatrienyl]-3,3-dimetyl-5-(metoksykarbonyl)-1-(4-sulfobutyl)-3H-indolium, natriumsalt.

R<20> til R<29>, R<32> og R3<3> betyr uavhengig av hverandre et hydrogenatom, en hydroksygruppe, en karboksy-, en sulfon-syrerest eller en karboksyalkyl-, alkoksykarbonyl- eller alkoksyoksoalkylrest med inntil 10 C-atomer eller en sulfoalkylrest med inntil 4 C-atomer
eller betyr et polylysin, polyetylenglykol, metoksypolyetylenglykol, polyvinylalkohol, dekstran, karboksydekstran eller en kaskadepolymerlignende struktur, eller representerer en rest med den generelle formel VI
