Description
Foreliggende oppfinnelse vedrører en analogifremgangsmåte ved fremstilling av nye terapeutisk aktive kynurensyrederivater, samt mellomprodukter for deres fremstilling.
Forbindelsene ifølge oppfinnelsen har den følgende generelle formel I
hvori:
R er en hydroksygruppe eller lavere alkoksy,
Ri er hydrogen
R2, R3 eller R4, som er like eller forskjellige, er hydrogen, halogen (klor, brom, jod eller fluor),
Cl-C6 alkoksy- eller trifluormetylgrupper.
Fysiologisk akseptable grupper for R er velkjente og
anvendt innen den medisinske kjemi; typiske eksempler for R grupper innbefatter estergrupper såsom metyl, etyl, t-butyl, pivaloyloksymetyl, t-butoksymetyl.
I forbindelsene med formel I er C^-C^ alkoksy fortrinnsvis metoksy.

Foretrukne forbindelser med formel I er de hvori R og R^ er hydrogen, minst én av R2, R3 og R4 er hydrogen og de andre to kan ha de ovenfor angitte betydninger; mere foretrukket er én av R2, R3, R4 hydrogen og de to andre er uavhengig
hydrogen, klor, trifluormetyl eller metoksy.
Når en eller flere av 83» R3 eller R4 gruppene er halogen, er de fortrinnsvis klor i posisjon 5 og/eller 6 og/eller 7.
Oppfinnelsen vedrører også farmasøytisk akseptable salter av forbindelser med formel I med ikke-toksiske syrer eller baser.

Forbindelsene ifølge oppfinnelsen er i stand til å motvirke effekten av glutamat ved å påvirke glycingjenkjenningsposi-sjonen tilstede i supermolekylkomplekset kjent som "NMDA reséptor-ion-kanalkompleks".
Forbindelsene I kan derfor anvendes ved humanterapi for behandlingen av mange neurologiske forstyrrelser som tilskrives svekket stimulering av glutamatergisk transmisjon. I realiteten, for høy tilstedeværelse av glutamat på det synaptiske nivå kan forårsake en abnorm stimulering og endelig neuronal død. Denne død er blitt definert som excito-toksisk type (Olney J. W., Advances Biochemical Psychopharmac. 27; 375-381, 1981).
Patologien hvor den abnormale funksjon av glutamatergisk transmisjon er antatt å spille en viktig rolle er vidtfav-nende og innbefatter ekstremt forskjellige kliniske tilstander, slik som "kinesisk restaurant-syndrom", visse hukommelse- eller læresvekkelser, hepatisk koma, visse nedbrytende sykdommer av CNS, slik som Huntington's chorea, olivopontocerebellar degenerering, amytrofisk lateral sklerose, visse retinale degenererende tilstander og lignende. En abnormal stimulering av glutamatreseptorer forårsaker også konvulsjoner og er involvert i hypoglycemisk neuronal ødeleggelse, latyrisme og neurologiske forstyrrelser forbundet med disse (amyotropisk lateral sklerose, Parkinson, dementia) som påvirker visse befolkningsgrupper nær sjø eller hav som spiser mat inneholdende store mengder av aminosyrer lignende glycin og glutamat (Science 237; 517--522; 1987).
Et annet felt av den neurologiske patologi, hvor glutamat nesten med sikkerhet er involvert er cerebrale ødeleggelser indusert av hypoxia eller ischemia.
Forbindelsene fremstilt i henhold til oppfinnelsen som kan anvendes innen det enorme patalogiske felt, kan betraktes som derivater av kynurensyre, hvis tilstedeværelse i lave konsentrasjoner i hjerner i forskjellige dyrearter er anerkjent såvel som dens antagonistiske aktivitet på glutamateffekten i visse klassiske farmakologiske forsøk, slik som marsvin myenterisk plexus (Moroni F. et al, Neuroscience Letters 68, 57-63; 1986; Moroni F. et al, European J. Pharmacol., 163, 123-126; 1989).
Det er vist at virkningen av kynurenat hovedsakelig kan tilskrives dets samvirke med glycin: relativt lave konsentrasjoner av glycin kan i realiteten antagonisere den inhiberende effekt av kynurenat. Glycin bør derfor være en agonist for en reseptor som modulerer glutamataktiviteten, mens kynurenat bør være en antagonist for denne reseptor.
Kynurensyrederivater er vist i EP-A-0303387, mens 2-klor-kynurensyre er tidligere studert (Kemp J. A. et al., Proe. Nati. Acad. Sei. Vol. 85 (17), 6547-50, 1988).
Forbindelsene ifølge foreligende oppfinnelse er vist å besitte mere gunstige farmakologiske aktiviteter enn stamforbindelsen. Mere spesielt er forbindelsene I og tiokynurensyre (R = OH, R2, R3 og R4 = H, R^ = H) konkurrerende antagonister for glycinreseptorer, hvilket er vist av de følgende resultater: 1) de fortrenger merket glycin fra kortikale membraner med en ECsg-verdi 5-10 ganger lavere enn den for kynurenat; 2) de antagoniserer på en ikke-konkurrerende måte glutamateffekten på NMDA-reseptorer tilstede i marsvin ileum: ved denne prøve er deres effekt konkurrerende antagonisert av glycin.
Innføringen av et halogenatom på benzenringen i tiokynure-nat har forsterket dets affinitet for glycinreseptorer.
Forbindelsene I var også aktive i musecerebralkorteks-snitt i henhold til Proe. Nati. Acad. Sei. USA 85; 6547-6550 - 1988, ved å forhindre glutamattoksisitet i kultiverte celler og for å forhindre NMDA induserte konvulsjoner hos mus.
I dette forsøk var forbindelsene I fra 3 til 50 ganger mere aktive enn de tilsvarende kynurensyrederivater. Mere spesielt vil 7-klor-4-tiokynurensyre ved en konsentrasjon på 0,08 uM antagonisere glutamatneurotoksisiteten i kultiverte celler; ved en konsentrasjon på 1 pM vil den fortrenge 50$ merket glycin fra bindingssetene i kortikale membraner og ved en konsentrasjon på 5 pM vil den antagonisere 50$ av glutamat eller NMDA virkninger på marsvin-terminal ileum eller på hjernesnitt, mens 5,7-diklortiokynurensyre antagoniserer NMDA induserte konvulsjoner hos mus i doser på
5-20 mg/kg.
Fra de ovenfor viste resultater er det åpenbart at 4-tiokynurensyre og dens derivater I er konkurrerende antagonister for en spesiell type glycinreseptorer. De er derfor nyttige for aminosyrestimulert neuro-transmisjon, spesielt for behandling av patologiske tilstander særpreget ved abnormal stimulering av NMDA reseptorer, slik som: 1) cerebral hypoxia eller ischemiske syndromer hvor tiokynureat også ville virke som en radikalfjerner.
2) Konvuls j oner
3) Hypoglycemia
4) Cerebral og spinal trauma
5) Muskelspasmer av sentral opprinnelse
6) Neurodegenererende sykdommer (amyotrofisk lateral sklerose, olivopontocerebellar atropi, ataxis etc.) 7) Cephalgia av forskjellig opprinnelse (utbredelse av depresjon, kombinert med visse former for cephalgia,
antagoniseres av NMDA antagonister)
8) Smertefulle syndromer som er vanskelige å behandle eller diagnostisere (trigeminus neuralgia, smerter på
grunn av "ghost limb", etc).
9) Encephalopati forbundet med AIDS
10) Hepatitisk encephalopati
11) Hydrocephalus
12) Lathyrisme og skalldyrforgiftning
13) Psycho-motorisk opphisselse
14) Insomnia
15) Psykiatriske sykdommer (schizofreni type 2; mani, etc).
For terapeutisk anvendelse vil forbindelsene I kunne formuleres til egnete farmasøytiske blandinger under anvendelse av konvensjonelle eksipienter og fremgangsmåter.
Forbindelsen kan administreres oralt eller parenteralt i doser i området 0,1 - 10 g/døgn.
De nye forbindelser kan fremstilles ved en fremgangsmåte som omfatter å omsette forbindelser med formel II
hvori R2» R3°S R4 eI* som tidligere definert, og R' er OH eller en gruppe som kan omdannes til en hydroksygruppe, med N,N-dimetyltiokarbamoylklorid.
De erholdte forbindelser med formel III
hvor R2, R3, R4 og R' er som ovenfor definert, omdannes til forbindelser IV ved behandling med syrer
hvor R2, R3, R4 og R' er som tidligere definert,
som omdannes til forbindelser I ved hydrolyse og avspaltning av den eventuelle beskyttende gruppe.
De således erholdte forbindelser I kan omdannes til "pro-medisiner" ved kjente fremgangsmåter, slik som alkylering eller acylering av tiogrupper og/eller forestring av karboksygruppen.
Forbindelsene III og IV er nye og faller inn under omfanget av foreliggende oppfinnelse som mellomprodukter som er nyttige ved fremstillingen av forbindelser I.
Forbindelsene II er kjente og de kan fremstilles i henhold til kjente fremgangsmåter.
Eksempelvis kan lett tilgjengelige aniliner V
hvor R2, R3 og R4 er som tidligere definert,
omsettes med acetylendikarboksylatdimetylester til å gi forbindelse II, hvor R' er -OCH3.
TTK-STflVTP EL 1
a) Kynurensyremetylester.
En eteroppløsning av diazometan(100 ml, 1,5 g) ble inndryp-pet i løpet av 15 min. til en suspensjon av kynurensyre (3,0 g, 0,016 mmol) i destillert tetrahydrofuran (200 ml) ved 0°C og under magnetisk omrøring. Deretter fikk reak-sjon sblandingen henstå til oppvarmning til romtemperatur og omrøringen fortsatte inntil fullstendig oppløsning av utgangsproduktet. Oppløsningsmidlet ble avdampet til å gi et råprodukt som ble underkastet flash-kromatografi på S102 (diameter 4 cm; høyde I6cm) eluert med kloroform (800 ml) og 95:5 kloroform-metanol (800 ml). 2,2 g av tittelforbindelsen ble erholdt, utbytte 73*, smp. 224-226'C.
%-NMR (CDC13 + CD3) S: 3,95 (s, 3H); 7,45 (m, 4H); 8.15 (d, 1H) ppm.
b) N,N-dimetyl-tiokarbamoyl-kynurensyremetylester (III).
En oppløsning av produktet fra trinn a) (700 mg, 13,4 mmol)
i destillert dimetylformamid (5 ml) ved 25°C og under magnetisk omrøring, ble tilsatt 1,4-diazabicyklooktan (1,35 g, 12,04 mmol) og N,N'-dimetyl-tiokarbamoylklorid (425 mg, 13,4 mmol). Reaksjonsblandingen ble omrørt inntil ut-gangsforbindelsen forsvant fullstendig, deretter ble blandingen fortynnet med vann (20 ml) og avkjølt til -5°C. Det utfelte faststoff ble filtrert fra og tørket under vakuum. 900 mg (Illb) ble erholdt i 92 £'s utbytte, smp. 131-134°C.
<1->H-NMR (CDC1<3>) S: 3,5 (d, 6H); 4,05 (s, 3H); 7,8 (m, 4H); 8,4 (d, 1H) ppm.
IR (CDCI3) : 1720, 1530 cm-<1>. c) N,N-dimetyl-tiolkarbamoyl-kynurensyremetylester (IV).
En oppløsning av (Illb) (200 mg, 0,70 mmol) i metanol (20
ml) under magnetisk omrøring, ble tilsatt 37 £'ig HC1 (0,5 ml). Reaksjonsblandingen ble omrørt i 10 min. og oppløs-
ningsmidlet avdampet under redusert trykk under oppvarmning til 40°C. 200 mg av (IVc) ble erholdt, smp. 121-124°C. %-NMR (CDC13) S : 3,1 (s bredt, 6H); 4,05 (s, 3H); 7,8 (m, 2H); 8,2 (m, 1H); 8,6 (s, 1H); 9,1 (d, 1H) ppm.
IR (CDCI3) :1720, 1680, 1625 cm-<1>.
d) tiokynurensyre (Id).
(IVc) (2,8 g, 9,0 mmol) ble suspendert i 10*'ig NaOH
oppløsning (30 ml) under magnetisk omrøring i 15 timer ved romtemperatur. Reaksjonsblandingen ble surgjort med 10*'ig HC1 til pH = 4, og deretter ble det utfelte faststoff filtrert og tørket under vakuum. 2,1 g av (Id) (100* utbytte) ble erholdt, smp. 230-233<*>C (spaltning).
^H-NMR (CD3OD) S : 7,5 (m, 1H); 7,5 (m, 1H); 7,90 (d, 1H); 8,05 (s, 1H); 8,75 (d, 1H) ppm.
<13>XC-NMR (CD3OD) 5 : 121,09, 125,96, 127,16, 129,81, 134,23, 165,51 ppm.
Masse (m/z): 205,6 (M<+>).
TTCSEMPEL 2
a) 7-klor-kynurensyremetylester.
En oppløsning av 3-kloranilin (16,6 ml, 0,15 mol) i metanol
(156 ml) under magnetisk omrøring, ble tilsatt en oppløsning av acetylendikarboksylatdimetylester (19,2 ml, 0,15 mol) i metanol (156 ml). Den resulterende oppløsning ble kokt under tilbakeløp i 30 min. og oppløsningsmidlet dampet av og resten tatt opp i 200 g difenyleter. Oppløsningen ble kokt under tilbakeløp i 15 min, deretter avkjølt og fortynnet med petroleumeter (400 ml). Den dannete utfelling ble filtrert og det ble erholdt 37 g av en blanding av 5-klor- og 7-klor-kynurenat. Denne utfellingen ble oppvarmet til 70° C i 160 g iseddik, og etter avkjøling ble den uoppløselige 7-klor-kynurensyremetylester filtrert fra. 26,5 g av produktet (72 % utbytte) ble erholdt ved avkjøling; smp. 291-292°C.
b) 7-klor-kynurensyre.
Metylesteren fra trinn a) ble tilsatt til en 10*'ig natriumhydroksydoppløsning (5 ml). Den resulterende suspensjon ble magnetisk omrørt ved romtemperatur i 30 min., deretter vasket med etyleter (2x 3 ml). De vandige faser ble surgjort med 10*'ig saltsyre til å gi en utfelling som ble filtrert fra og tørket til å gi 0,170 g av tittelproduktet (76 *'ig utbytte); smp. 285-287"C.
%-NMR (NaOD) S : 6,75 (s, 1H, H3); 6,85 (dd, 1H, J6_5 = 7m,5 Hz; Jfc_g <=> 1,5 Hz, H6); 7,22 (d, 1H, Jg-6 - <1,>5 Hz, H8); 7,58 (d, 1H, J5_6 = 7,5 - 7,5 Hz, H5) <pp>m. c) 7-klor-kynurensyremetylester N,N-dimetyl-tion-karbamat.
En suspensjon av esteren fra trinn a) (50 g, 0,21 mol i vandig dimetylformamid ble tilsatt DABC0 (82,5 g, 0,73 mol) og deretter N,N-dimetyl-tiokarbamoylklorid (25,9 g, 0,21 mol). Den resulterende suspensjon ble omrørt magnetisk ved romtemperatur i 3 timer, deretter fortynnet med vann (800 ml) og det utfelte faststoff ble filtrert fra og tørket til å gi 65 g av tittelforbindelsen (97 *'ig utbytte). l-H-NMR (CDC13) S : 3,52 (s, 6H, NMe2); 4,07 (s, 3H, MeO) 7,5 (dd, 1H, J6_5 = 8,5 Hz; <J>6_8 = 1 Hz, H6); 7,82 (d, 1H, <J>5_6 = 8,5 Hz, H5); 7,97 (s, 1H, H3); 8,27 (d, 1H, J8_6 = <1> Hz, H8) ppm. d) 7-klor-kynurensyremetylester N,N-dimetyl-tiol-karbarnat.
En suspensjon av tiokarbamatet (65 g, 0,2 mol) fra trinn c) i metanol (1200 ml) ble tilsatt 16,7 ml 37*'ig saltsyre. Suspensjonen hvis farge ble gul, ble magnetisk omrørt ved romtemperatur i 30 min.. Dereter ble faststoffet frafiltrert, oppløsningsmidlet ble delvis inndampet til å gi en andre utfelling av et stoff som også ble filtrert og tørket til å gi 64,5 g av tittelforbindelsen (99*'ig utbytte).
%-NMR (CDCI3) S : 3,11 (bs, 6H, NMe2);4,06 (s; 3H, MeO) 7,56 (dd, 1H, J6_5 - 7,5 Hz; J6_8 - 1,5 Hz, H6); 8,2 (d, 1H, <J>5-6 = 7>5 Hz» H5); 8»3 (d» 1H, J6_8 = 1,5 Hz, Hg) 8,38 (d, 1H, H3) ppm.
e) '_ 7-klor-tio-kynurensyre
Tiolkarbamatet fra trinn d) (64,5 g, 0,2 mol) ble tilsatt
til en 10*'ig natriumhydroksydoppløsning. Den resulterende suspensjon ble magnetisk omrørt ved romtemperatur i 16 timer og deretter vasket med etyleter (3 x 150 ml). De vandige faser ble surgjort med 10 *'ig saltsyre for å utfelle et rød-oransj faststoff som ble frafiltrert og tørket til å gi 47 g av tittelforbindelsen (98*'ig utbytte); smp. 200-203°C.

omdannes til en forbindelse med formel I ved hydrolyse, og avspaltning av en eventuell beskyttende gruppe.

karakterisert ved at man utgår fra den tilsvarende substituerte utgangsforbindelse.
