Description
Foreliggende oppfinnelse vedrører fremstilling av nye 4-pyridonderivater, som anvendelse for behandling av bakterielle infeksjoner.
4-pyridonderivater og spesielt oksoderivater av substituerte kinolin-, naftyridin-, benzoksazin- og benzokinolinkarboksy1 - syrer med antibakteriell virkning er kjente. Fra denne forbindelsesklassen er, f.eks. i US-PS 4 146 719, beskrevet, derivater av kinolin-3-karboksylsyren, som som spesielt strukturelement oppviser en cyklisk nitrogenbase i 7-stilling på kinolinringen og har antibakteriell aktivitet.
I EP 131 839 beskrives blant andre antibakterielt virksomme kinolinkarboksylsyrederivater som i I-stilling kan være substituert med p-fluorfenyl, og som, som karakteristisk substituent i 7-stilling, kan bære en alifatisk, ringformig heterocyklisk gruppe. Brodannede, bicykliske baser nevnes imidlertid ikke som substituenter.
Endelig er fra EP-A-159174 oksoderivater av substituerte kinolin-, naftyridin- og benzoksazinkarboksylsyrer kjente, som som karakteristiske substituenter i 7-stilling av kinolin- eller naftyridinringen, hhv. i 10-stilling på benzoksazinringen, bærer en brodannet, bicyklisk nitrogenbase og disse er også antibakterielt virksomme.
På tross av det store antallet av de i litteraturen omtalte, antibakterielt virksomme 4-pyridonderivatene består fremdeles et behov for nye forbindelser fra denne stoffklassen, i det kjente 4-pyridonderivatene kan forbedres når det gjelder aktivitet, virksomhet mot bestemte typer av bakterielle sykdomsfremkallere, stabilitet, farmakokinetiske egenskaper og/eller toksiske bivirkninger.
Det er nå funnet nye, med brodannede nitrogenbaser substituerte oksoderivater av spesielt kinolin- og benzoksasinkarbok-sylsyrer, som oppviser, interessante farmakologiske egenskaper .
Ved foreliggende oppfinnelse fremstilles nye 4-pyridonderivater med den generelle formelen
der
Rj er 2,4-difluorfenyl og
Y er en base med den generelle formel
der
R2 er hydrogen, metyl,, benzyl, formyl, acetonyl eller en rest med den generelle formel
samt addisjonssalter av forbindelser med den generelle formel I med farmasøytisk godtagbare uorganiske eller organiske syrer eller baser og/eller av deres hydrater.
Av forbindelsene av formel I er slike foretrukket hvori Y står for en base av den generelle formel Ila hvori R2 står for hydrogen, metyl eller formyl.
Spesielt foretrukket er forbindelsene 6-fluor-l-(2,4-difluorfenyl )-l,4-dihydro-7-(5-metyl-2,5-diaza-bicyklo-(2,2,1)hept-2-yl)-4-okso-kinolin-3-karboksylsyre og 7-(2,5-diazabicyklo(2,2,1 )hept-2-yl )-6-f luor-l-(2 ,4-dif luorferiy.1 )-1,4-dihydro-4-okso-;-kinolin-3-karboksylsyre.
Forbindelsene fremstilt ifølge foreliggende oppfinnelse har delvis optiske sentre og kan foreligge som racemater eller i form av optiske isomerer.
Forbindelsene med generell formel I fremstilles ifølge oppfinnelsen ved at man
a) omsetter forbindelser med den generelle formelen
der Ri har den ovenfor angitte betydning, L er en nukleofil avspaltbar gruppe klor eller fluor og R3 hydrogen eller et borkompleks -B(R4R5), der R4 og R5 er like og betyr fluor, med forbindelser med formelen
der R2 nar den ovenfor angitte betydning eller i tillegg betyr en aminobeskyttelsesgruppe, hvoretter man
b) 1 de således oppnådde forbindelser der R3 betyr gruppen
-B(R4R5), spalter av denne gruppe,
c) 1 de oppnådde forbindelser som 1 posisjon R2 bærer en aminobeskyttelsesgruppe, spalter av denne, d) eventuelt metylerer, benzylerer, formylerer eller omsetter med bromaceton eller en forbindelse R^, Hal der R5 er en rest
med formel III, de oppnådde forbindelser med den generelle formel I der R2 betyr hydrogen, og
eventuelt overfører de i trinnene a) til d) oppnådde forbindelser med den generelle formel I til et farmasøytisk akseptabelt addisjonssalt med syrer eller baser.
Ved omsetningen av forbindelsene av den generelle formel IV med forbindelser av formel V ifølge fremgangsmåtetrinn a) anvender man baser som HX-akseptorer. Som sådanne kan det f.eks. anvendes baser i overskudd av den generelle formelen Va eller Vb. For dette formålet anvendes det til 1 mol av forbindelen av den generelle formel IV 1-20 mol, fortrinnsvis 1-5 mol av forbindelsene av formelen Va eller Vb. Basen som anvendes i overskudd av.den generelle formel Va eller Vb kan deretter tilbakevinnes ved opparbeiding av moderluten. Ved anvendelse av ekvivalente mengder av forbindelsene av den generelle formel IV og basene av den generelle formel Va eller Vb tilsettes det som syrebindende middel eksempelvis 1,4 diazabicyklo(2,2,2)oktan, trietylamin eller 1,8-diaza-bicyklo(5,4,0)undec-7-en.
Omsetningen ifølge fremgangsmåtetrinn a) gjennomføres fordelaktig i nærvær av oppløsnings- eller fortynningsmidler som eksempelvis dimetylsulfoksyd, 1,3-dimetyl-tetrahydro-2(1H)-pyrlmidinon, heksametylfosforsyretriamid, sulfolan, pyridin, lutidin eller kollidin eller gjennomføres i blandinger av disse oppløsningsmidlene. Det er imidlertid også mulig som oppløsningsmiddel å anvende en av de ovenfor nevnte basene eller blandinger av disse basene med et av de ovenfor nevnte oppløsningsmidlene.
Reaksjonsvarigheten for reaksjonen I fremgangsmåtetrinn a) utgjør som regel 1 til 5 timer, reaksjonstemperaturen 100-200°C, hvorved 130-170°C er foretrukket.
Dersom produktet etter gjennomføring av reaksjonen ikke utkrystalliserer ved avkjøling og kan utvinnes ved en enkel filtrering, foregår isoleringen valgfritt ved avdestillering av den flyktige andelen eller ved utfelling av produktet eller et av dets salter, eller ved en kombinasjon av disse mulighetene.
Dersom det i fremgangsmåtetrinn a) anvendes forbindelser av formel Va eller Vb, hvori R£ står for hydrogen, er det ikke nødvendig, men i mange tilfeller fordelaktig, å gjøre forbindelsene av formel Va eller Vb inerte mot nukleofil substitusjon ved omsetning med aminobeskyttelsesgrupper. På denne måten kan f.eks. følgende beskyttelsesgrupper finne anvendelse: karboksyl syregrupper som f.eks. formyl, acetyl, trifluorace-tyl; alkoksykarbonylgrupper som f.eks. etoksykarbonyl , t-butyoksykarbonyl, 2,2,2-trikloretoksykarbonyl; aryloksykarbo-nylgrupper som f.eks. benzyloksykarbonyl, fenoksykarbonyl; og grupper som f.eks. trimetylsilyl, trityl, p-toluensulfonyl og benzyl.
Avspaltningen av R3 med betydningen -B(R4R5) ifølge f rem-gangsmåtetrinn b) foregår ved 1 times oppvarming under tilbakeløp av det under a) oppnådde produktet i et ca. 10 gangers overskudd av fortynnet alkalihydroksydoppløsning. Etter oppvarmingen nøytraliseres oppløsningen med HC1, det utfelte bunnfallet frasuges og opparbeides ved vanlige fremgangsmåter som vasking og omkrystallisering. Avspaltningen av aminobeskyttelsesgruppen Ifølge fremgangs-måtetrlnn c) kan foregå ved kjente fremgangsmåter før eller etter isolering av produktet av formel I. Følgelig avspaltes f.eks. etoksykarbonylgruppen ved hydrolyse i surt eller alkalisk miljø, tritylgruppen kan fjernes ved hydrogenolyse.
Metyleringen av forbindelser av formel I, hvori R2 nar betydningen hydrogen, ifølge fremgangsmåtetrinn d) kan foregå ved hjelp av vanlige metyleringsmidler som metyljodid eller dimetylsulfat eller ved oppvarming av det sekundære aminet under tilbakeløp i et 10-20 gangers overskudd i en blanding av maursyre og 40$ formaldehydoppløsningen. Reaksjonstiden utgjør 0,5-5 timer. Reaksjonsproduktet inndampes deretter, opptas i vann, bringes til pH 7 med fortynnet ammoniakk og ekstraheres flere ganger med en blanding av metylenklorid og kloroform. Inndampning av de samlede organiske fasene og omkrystallisasjon, f.eks. fra metanol/eter gir det ønskede alkylerte produktet.
Formyleringen av forbindelser av formel I hvori R2 har betydningen hydrogen, foregår ved hjelp av vanlige formyle-ringsmidler som eksempelvis de blandede anhydridene av eddiksyre og maursyre.. Omsetningen med bromaceton eller en forbindelse R^Hal kan eksempelvis foregå i et inert fortyn-ningsmiddel som dimetylformamid under tilsats av syrebindende midler som kaliumhydrogenkarbonat.
Om ønsket overfører man i et etterfølgende reaksjonstrinn de ifølge a) til d) oppnådde forbindelsene som er amfotære, med baser eller syrer til deres salter og/eller fysiologisk tålbare estere. Som salter som kan oppnås ved reaksjon av baser med karboksylgruppen av forbindelsene ifølge oppfinnelsen kommer eksempelvis alkali-, jordalkali-, ammoniakk- og sølvsalter, samt saltene med farmasøytisk tålbare baser som kollin, dietanolamin, etylendiamin, guanidin og andre på tale. Salter med farmasøytisk tålbare uorganiske eller organiske syrer dannes ved reaksjon ved basiske sentre. Eksempler på slike syrer er saltsyre, svovelsyre, fosforsyre, eddiksyre, sltronsyre, oksalsyre, malonsyre, sal icylsyre, metansulfonsyre osv. Dannelsen av den fysiologisk tålbare esteren kan foregå ved generelt kjente fremgangsmåter.
Stoffer som trenges som utgangsforbindelser av strukturforme-len IV er kjente eller kan fremstilles analogt kjente fremgangsmåter, som eksempelvis beskrevet i EP 47005, EP 159174, EP 203, EP 78362 og i J.Med.Chem. 1980, 23, 135S eller J.Med.Chem. 1985, 28, 1558. 6,1 difluor-l-(2,4-fluorfenyl )-l ,4-dihydro-4-okso-kinolin-3-karboksylsyren ble fremstilt ifølge formelskjema II. Syrekloridet VI ble omsatt med magnesiumsaltet av malonsyre-etyl-tert-butylesteren til acylmalonesteren VII, som ved behandling med trifluoreddik-syre ble overført til aroyleddiksyreetylesteren VIII. Ved kondensasjon av VIII med o-maursyre-trietylester/acetanhydrid ble 2-(2-klor-4,5-difluor-benzoyl)-3-etoksy-akrylsyreetyl-esteren VIIII oppnådd, som ble omsatt med 2,4-fluor-anilin til enamin X. Enaminet X ble ringslutet med natriumhyririd som base til kinolonester XI (R = etyl), som deretter ble forsåpet til den ønskede.syren XI (R = H).
Det som utgangsmaterial for denne syntesen anvendte 2-klor-4,5-difluorbenzoylklorid VI ble oppnådd fra den tilsvarende syren med tionylklorid.
2-klor-4,5-difluorbenzosyren ble oppnådd ved Sandmeyer-reaksjon fra ifølge EP-55 629 fremstilt 4,5-difluorantranil-syre.
Fremstillingen av borkompleksene av den generelle formel IV er også kjent fra litteraturen og kan f.eks. foregå ved fremgangsmåten beskrevet i de japanske søknadene 82/233684, 83/188138 og 85/126290.

Forbindelsene som fremstilles ifølge oppfinnelsen har en fremragende antibakteriell virkning mot forskjellige patogene bakteriestammer. Eksempler på slike stammer er Escherichia, Klebsiella, Enterobacter, Serrattia, Acinetobacter, Pseudomonas, Staphylococcus, Streptococcus, Bacteroider, Clostridium og Peptococcus.
For bestemmelse av den antibakterielle virksomheten ble MHK-verdiene (minimale hemningskonsentrasjonsverdier) målt in vitro, samt EDso-verdiene in vivo. Som sammenligningsstoffer tjente ciprofloksasin (det foretrukne stoffet ifølge EP 78 362), pefloksasin (det foretrukne stoffet ifølge DE-OS 2843066), samt enoksasin (det foretrukne stoffet fra EP 9 425). Overlegenheten av stoffene fremstilt ifølge oppfinnelsen viser seg spesielt overfor gramposltive bakterier og ved oral tilførsel, idet resorpsjonen av forbindelsene fremstilt ifølge foreliggende oppfinnelse er betydelig forbedret sammenlignet med ciprofloksasin, pefloksasin og enoksasin.
EKSEMPEL 1
15,0 g (37,36 mmol ) av det blandede anhydridet av 7-klor-6-fluor-l-(2,4-difluor-fenyl)-l,4-dihydro-4-okso-kinolin-3-karboksylsyre og difluorborsyre, fremstilt ved oppvarming av 7-klor-6-fluor-l-(2,4-di fluorfeny1 )-l,4-dihydro-4-okso-kinolin-3-karboksylsyre i 60$ vandig fluorborsyre, 15,36 g (56 mmol) av dihydrobromidet av 2-metyl-2,5-diaza-bicyk1o-(2,2,1 )heptan og 31 ml (0,224 mol) trietylamin tilsettes til 200 ml tørr dimetylsulfoksyd og omrøres i 6 dager ved romtemperatur. Deretter helles reaksjonsblandingen i 800 ml kaldt vann. Bunnfallet som dannes frafUtreres, vaskes med vann og oppvarmes deretter i en blanding av 100 ml dioksan og 150 ml 3N natronlut i 1 time til tilbakeløp. Etter avkjøling tilsettes under omrøring 500 ml konsentrert saltsyre. Det omrøres i 1 time, de uoppløslige bestanddelene frafiltreres og det klart gule filtratet ekstraheres flere ganger med metylenklorid. Den vandige fasen bringes deretter under isavkjøling, ved tilsats av fast natriumhydroksyd, til pH-verdien 7,4. Deretter ekstraheres med kloroform. Kloroform-fasen tørkes med natriumsulfat og kloroformen avdestilleres i vakuum. Den derved oppnådde mørkebrune oljen gir etter krystallisasjon fra 450 ml etanol 3,15 g 6-fluor-l-(2,4-difluorfenyl )-l,4-dihydro-7-(5-metyl-2,5-diaza-bicyk1o-(2,2,1)-hept-2-yl)-4-okso-kinolin-3-karboksylsyre som hydrat, smeltepunkt 215-220°C (dekomponering).
Utbytte: 3,15 g (18,8& av teoretisk)
EKSEMPEL 2
Analogt eksempel 6 oppnås ved anvendelse av 5,56 g (13,85 mmol) av det blandede anhydridet av 7-klor-6-fluor-l-(2,4-dif luorf enyl )-l,4-dihydro-4-okso-kinolin-3-karboksylsyre og difluorborsyre, 5,82 g (16,62 mmol) 2-fenylmetyl-2,5-diazabi-cyklo(2,2,1)heptan dihydrobromid og 11,58 ml (83 mmol) trietylamin i 50 ml tørr dimetylsulfoksyd, 2,73 g av 6-fluor-1-( 2 ,4-difluorfenyl)-l,4-dihydro-4-okso-7-(5-fenylmetyl-2,5-diaza-bicyklo(2,2,1)hept-2-yl)kinolin-3-karboksylsyre med et smeltepunkt på 233-238°C (dekomponering) etter omkrystallisasjon fra toluen.
Utbytte: 2,73 g (39# av teoretisk)
EKSEMPEL 3
2,57 g (5,08 mmol) av 6-fluor-1-(2,4-difluorfenyl)-l,4-dihydro-4-okso-7-(5-fenylmetyl-2,5-diazabicyklo(2,2,1)hept-2-yl )kinolin-3-karboksylsyre oppløses i 170 ml absolutt etanol som inneholder 18 g saltsyre og blandes med 2,1 g 10$ palladium-aktivt kull. Det hydreres i 2 timer ved 103,4 kPa og 60°C. Etter fjernelse av katalysatoren inndampes under redusert trykk. Det oppnådde faste stoffet opptas i 80 ml vann og innstilles på en pH-verdi på 7. Den oppnådde suspensjonen oppvarmes i 20 minutter til 60°C. Etter avkjøling frafiltreres bunnfallet. Etter omkrystallisasjon fra dimetylformamid oppnås 1,53 g 7-(2,5-diazabicyklo(2 ,2 ,1 )-hept-2-yl )-6-fluor-l-(2, 4-dif luorf enyl )-l, 4-dihydro-4-okso-kinolin-3-karboksylsyre som hydrat, smeltepunkt 198-204°C (dekomponering).
Utbytte: 1,53 g (69,5^ av teoretisk)
EKSEMPEL 4
0,5 g (1,2 mmol) 7-(2,5-diazabicyklo(2,2,1)-6-fluor-1-(2,4-difluorfenyl )-l , 4-dihydro-4-okso-kinolin-3-karboksy1 syre suspenderes i 20 ml dimetylformamid og blandes med 3 ml blandet anhydrid av eddiksyre og maursyre. Etter 90 minut-ters omrøring ved romtemperatur avdestilleres oppløsningsmid-let under redusert trykk. Den oljeformige resten blir fast etter utrøring med eter og kan frafiltreres. Omkrystallisasjon fra en blanding av 20 ml dimetylformamid og 5 ml ligroin gir 0,24 g 7-(5-formyl-2,5-diazabicyklo(2,2,l)hept-2-yl)-6-fluor-l-(2,4-difluorfenyl)-l, 4-dihydro-4-okso-kinolin-3-karboksylsyre. Smeltepunkt: 252-258°C (dekomponering).
Utbytte: 0,24 (45,3# av teoretisk)
EKSEMPEL 5
188 mg (0,434 mmol) 7-(2,5-diazabicyklo(2,2,1)-hept-2-yl)-6-fluor-l-(2,4-difluorfenyl)-l,4-dihydro-4-okso-kinolin-3-karboksylsyremonohydrat (eksempel 3) suspenderes i 20 ml tørr dimetylformamid, blandes med 96 mg (0,95 mmol) vannfritt kaliumhydrogenkarbonat og 92 mg (0,48 mmol) 4-brommetyl-5-metyl-1,3-dioksolen-2-on og omrøres i 10 timer ved romtemperatur. Deretter helles det på en blanding av 150 ml etylacetat og 50 ml vann, og fasene skilles. Den organiske fasen vaskes 5 ganger, hver gang med 30 ml vann, tørkes over natriumsulfat, filtreres og inndampes under redusert trykk ved romtemperatur. Omkrystallisasjon fra etylacetat gir 63 mg l-(2,4-difluorfenyl)-6-fluor-1,4-dihydro-7-(5((5-metyl-2-okso-1 ,3-dioksolen-4-yl)-metyl)-2,5-diazabicyklo(2,2,1 )hept-2-yl)-4-okso-kinolin-3-karboksylsyre.
Smeltepunkt: 133-138°C (dekomponering)
Utbytte: 63 mg (27 ,556 av teoretisk).
EKSEMPEL
Bestemmelse av de antibakterielle in vitro-egenskapene.
De antibakterielle egenskapene for forbindelsene fra eksempler 1 og 3 ble bestemt ved MHK-forsøk (agarfortynningsfor-søk, medium ifølge Mtlller-Hinton) overfor forskjellige grampositive og gramnegative bakteriestammer og sammenlignet med pefloksasin. Resultatene er sammenfattet i tabell III.
Farmakologiske forsøk
I et sammenligningsforsøk ble forbindelsen l-(2,4-difluorfenyl)-6-fluor-l,4-dihydro-7-(5-metyl-2,5-diazabicyklo-[2,2,l]hept-2-yl)-4-okso-3-kinolinkarboksylsyre (forbindelse I) og 6-fluor-l-(4-fluorfenyl)-l,4-dihydro-7-(5-metyl-2,5-diazabicyklo-[2,2,l]hept-2-yl)-4-okso-3-kinolinkarboksylsyre (forbindelse II) testet med henblikk på deres antibakterielle virkning.
Testen for bestemmelse av den antibakterielle aktiviteten var standardtesten ved agar-fortynningsmetoden i en nærings-oppløsning ifølge Muller-Hinton, hvorved prøvene ble inkubert i 18 timer ved 37°C.
I de følgende tabellene er resultatene fra denne testen angitt i MIC-verdier (minimal inhibitory concentration) i mikrogram/ml.
Claims (1)
- Analogifremgangsmåte for fremstilling av terapeutisk aktive antibakterielt virksomme 4-pyridonderivater med den generelle formel der Ri er 2,4-difluorfenyl og Y er en base med den generelle formel derR2 er hydrogen, metyl, benzyl, formyl, acetonyl eller en rest med den generelle formel samt addlsjonssalter av forbindelser med den generelle formel I med farmasøytisk godtagbare uorganiske eller organiske syrer eller baser og/eller av deres hydrater, karakterisert ved at man a) omsetter forbindelser med den generelle formelender Ri har den ovenfor angitte betydning, L er en nukleofil avspaltbar gruppe klor eller fluor og R3 hydrogen eller et borkompleks -B(R4R5), der R4 og R5 er like og betyr fluor, med forbindelser med formelender R2 har den ovenfor angitte betydning eller i tillegg betyr en aminobeskyttelsesgruppe, hvoretter man b) i de således oppnådde forbindelser der R3 betyr gruppen —B(R4R5), spalter av denne gruppe, c) i de oppnådde forbindelser som i posisjon R2 bærer en aminobeskyttelsesgruppe, spalter av denne, d) eventuelt metylerer, benzylerer, formylerer eller omsetter med bromaceton eller en forbindelse R5 Hal der R5 er en rest med formel III, de oppnådde forbindelser med den generelle formel I der R2 betyr hydrogen, og eventuelt overfører de 1 trinnene a) til d) oppnådde forbindelser med den generelle formel I til et farmasøytisk akseptabelt addisjonssalt med syrer eller baser.
