Forbindelsen med formel (I) og syreaddisjonssaltene
derav utviser verdifull farmakodynamisk virkning, de har CNS-stimulerende aktivitet.
Fra litteraturen er det kjent noen 3,4-dihydro-5H-2,3-benzodiazepiner med strukturformler som er forskjellige fra forbindelsen fremstilt ifølge oppfinnelsen og som innehol-
der én eller flere alkoxygrupper i stillingene 7 og 8, og ingen substituent i stilling 1, i benzodiazepinkjernen [se f.eks. Chem. Ber. 95, 2012 (1962); Synthesis 1973, 159 og 1977, 1; Heiv. Chim. Acta 59, 2786 (1976)]. Det er imidlertid ikke publisert noen data vedrørende den biologiske aktivitet til disse forbindelsene. Andre 3,4-dihydro-5H-2,3-benzodiazepiner med en alkyl-, cykloalkyl-, fenyl- eller substituert fenyl-
gruppe i stilling 1 og én eller flere alkoxygrupper i stillin-
gene 7 og 8 i benzodiazepinkjernen er også kjent fra litteraturen. Disse forbindelsene har spontane bevegelsesaktivitet-nedsettende, narkoseforsterkende og milde CNS-nedsettende virkninger (BRD off.skrift nr. 3 209 100).
Foreliggende oppfinnelse er basert på det uventede
funn at, i motsetning til de kjente 3,4-dihydro-5H-2,3-benzo-diazepinderivater og til og med alle 5H-2,3-benzodiazepiner som hittil er kjent, forbindelsen med formel (I) fremstilt ifølge oppfinnelsen og syreaddisjonssaltene derav, har CNS-
stimulerende egenskaper, idet de utviser en virkning som er helt ny blant benzodiazepiner.
I en viss utstrekning er det farmakologiske virkningsområde for forbindelsen lik med området til dopamin-agonister som øker spontan bevegelsesaktivitet, forårsaker stereotypi og antagoniserer virkningen av dopaminreseptor-blokkerende neuroleptica (klorpromazin, haloperidol, etc.). I motsetning til kjente dopaminagonister (apomorfin, bromcriptin) utviser de imidlertid ikke emetisk virkning. Dessuten har forbindelsene også en sterk inhiberende virkning på skjelvingen som utløses av oxotremorin.
Når man tar i betraktning det faktum at påvirkning av dopaminergiske mekanismer resulterer i modifikasjon av flere funksjoner i nervesystemet (ko-ordinert muskelkontrak-sjon, læreprosesser, regulering av blodtrykket og det endocrine system), forventes den terapeutiske bruk av forbindelsene fremstilt ifølge oppfinnelsen å være for én av de sykdommer som står i forbindelse med. disse funksjonenene (f.eks. Parkinson's sykdom).
Forbindelsen med formel (I) og syreaddisjonssaltene derav fremstilles på kjent måte ved å redusere l-(4-amino-fenyl)-4-methyl-7,8-methylendioxy-5H-2,3-benzodiazepin med formel (II) i et egnet oppløsningsmiddel med et uorganisk eller uorganisk-organisk hydrid og/eller kompleks-metallhydrid og, om ønsket, omdanne basen eller basene til syre-addis jonssalter , eller omdanne saltet til en fri base.
Den selektive reduksjon av forbindelsen med formel (II) kan fortrinnsvis utføres med følgende kjente hydrider og kompleksmetallhydrider: natriumhydrid, lithiumhydrid, calsium-hydrid, diboran, silan, diethylsilan, lithiumaluminiumhydrid, natriumborhydrid, natriumborhydrid, natriumborhydrid aluminium-klorid, natriumdihydro-bis-(2-methoxy-ethoxy)-aluminat, natriumcyanborhydrid, lithiumtrimethoxy-aluminiumhydrid eller natriumborhydrid-triethyloxoniumfluorborat.
Reduksjonen utføres fortrinnsvis i de følgende opp-løsningsmidler eller oppløsningsmiddelblandinger: vann,
ethere, alkoholer, primære aminer, aromatiske hydrocarboner, klorerte alifatiske hydrocarboner, alifatiske carboxylsyrer og pyridin. Det reaksjonsmedium som brukes avhenger av det anvendte reduksjonsmiddel. Det er foretrukket å utføre reduksjonen i en oppløsningsmiddelblanding som ikke reagerer (eller bare svært sakte) med reduksjonsmidlet. Ved utførelsen av fremgangsmåten er pyridin det foretrukne oppløsningsmiddel.
Reduksjonen utføres fortrinnsvis ved en temperatur mellom 50 og 110° C.
Våre forsøk viser at reduksjonsmidlet må anvendes i et overskudd på 300 - 500 % for fullstendig reduksjon av forbindelsen med formel (II) , ettersom forbindelsen som skal reduseres også har en aminogruppe som binder noe av reduksjonsmidlet.
Ved utførelsen av fremgangsmåten fåes den påtenkte forbindelse som en fri base. De farmasøytisk akseptable syre-addis jonssalter av basen med formel (I) dannes på kjent måte: basen oppløses eller oppslemmes i et passende oppløsningsmid-del, f.eks. i vann, methanol, ethanol, isopropanol eller ethyl-acetat, og deretter tilsettes en passende syre eller en egnet oppløsning av syren. Det er foretrukket å bruke en ikke-toksisk, uorganisk syre, slik som saltsyre, hydrobromsyre, svovelsyre, fosforsyre eller en ikke-toksisk organisk syre, slik som eddiksyre, vinsyre, methansulfonsyre, maleinsyre, fumarsyre etc. Saltene isoleres enten ved filtrering eller ved avdamping av oppløsningsmidlet. Om ønsket oppslemmes det derved erholdte produkt, rekrystalliseres og/eller tørkes under redusert trykk.
Fremstillingen av utgangsstoffet til fremgangsmåten som brukes ved fremstillingen ifølge oppfinnelsen, [1-(4-aminofenyl)-4-methyl-7,8-methylendioxy-5H-2,3-benzodiazepin med formel (II)], er beskrevet i litteraturen (BRD off. skrift nr. 3 527 117) .
Som nevnt ovenfor utviser forbindelsen med formel (I) og syreaddisjonssaltene derav verdifulle CNS-stimulerende aktivitet. De farmakologiske virkningene av forbindelsene er fremlagt for 1-(4-aminofenyl)-4-methyl-7,8-methylendioxy-3,4-dihydro-5H-2,3-benzodiazepinbasen som et eksempel nedenunder.
l.a. Motilitetsundersøkelser på mus
For å undersøke hvordan forbindelsen fremstilt ifølge eksempel 1 øker motiliteten i mus av hannkjønn, ble det utført 2-timers forsøk. For å bestemme den spontane bevegelsesaktivitet, ble sultaktivitet-komponenten utelatt (musene ble ikke fastet), og følgelig ble bare utforskningsaktiviteten målt. Forsøksdyrene ble plassert i observasjonsboksen umiddel-bart etter den intraperitoneale behandling. De erholdte data er vist i Tabell 1
l.b. Motilitetsundersøkelser på rotter
CFY-rotter av hannkjønn som veide 120 -;140 g, ble behandlet intraperitonealt i grupper på tre med en dose på 10, 20, 25 eller 30 mg/kg av forbindelsen fremstilt ifølge eksempel 1, og ble øyeblikkelig plassert i observasjonsboksen. Forsøket varte i 2 timer. De erholdte resultater er vist i Tabell 2.
Dataene i Tabell 1 og 2 viser at forbindelsen fremstilt ifølge oppfinnelsen øker motiliteten eller den spontane bevegelsesaktivitet hos mus og rotter på en doseavhengig måte.
2. Måling av den haloperidol- antagonistiske virkning i mus
CFLP-mus av hannkjønn som veide 20 - 22 g, ble behandlet intraperitonealt med en dose av 0,5 mg/kg 4-[4-(4-klor-fenyl)-4-hydroxy-l-piperidinyl]-1-(4-fluorfenyl)-1-butanon (haloperidol) og den spontane bevegelsesaktivitet (SMA) ble målt i forhold til de erholdte data i kontrollene, ved hjelp av et kapasitans-følsomt "motimeter". En annen gruppe av for-søksdyrene ble også behandlet med en dose på 30 mg/kg av forbindelsen fremstilt ifølge eksempel 1, 15 minutter etter admi-nistreringen av haloperidol. Den haloperidol-induserte ned-settelse i SMA ble signifikant antagonisert ved hjelp av forbindelsen fremstilt ifølge eksempel 1. Resultatene er oppsum-mert i Tabell 3.
3. Undersøkelse av stereotypi hos rotter
For målingen av stereotypi ble fremgangsmåten ifølge B. Costall og R.I. Naylor [Eur. J. Pharmacol. 18. 95 (1972)] brukt. Poengverdiene i Tabell 4 er gjennomsnittlig poeng-verdier oppnådd på tidspunktet for avlesning. Resultatene er vist i Tabell 4.
Bromcriptin
På samme måte som bromcriptin, forårsaker forbindelsen fremstilt ifølge eksempel 1 stereotypi. Denne virkningen utvikles tidligere, men den er noe svakere enn virkningen av bromcriptin.
4. Inhibering av oxotremorin i mus
Undersøkelsene ble utført i overensstemmelse med metoden til R.C. Rathbun og I.H. Slater [Psychopharm. 4, 114
(1963)]. 1 time etter at sultende CFLP-mus av hannkjønn ble oralt forbehandlet med en dose på 6,25, 12,5 eller 25,0 mg/kg av forbindelsen fremstilt ifølge eksempel 1, ble de behandlet intraperitonealt med en dose på 1 mg/kg 1-[4-(1-pyrrolidinyl)-2-butinyl]-2-pyrrolidinon (oxotremorin).
Inhiberingen av oxotremorin-indusert skjelving og spyttutsondring utøvd av forbindelsen fremstilt ifølge eksempel 1 ble målt i forhold til kontrollgruppen behandlet med oxotremorin alene. Inhiberingsstyrken ble uttrykt som poenger (0, 0,5, 1, 2 eller 3). Resultatene er vist i Tabell 5.
Forbindelsen fremstilt ifølge eksempel 1 inhiberer ikke den oxotremorin-induserte spytt-utsondring, den nedsetter imidlertid skjelvingen signifikant.
5. AvhengighetsundersøkeIser
Undersøkelsene ble utført ved å bruke DAF-metoden (legemiddel-iblandet mat) [E. Tagashira et al. Psychopharma-cologia 57, 137 - 144 (1978)]. CFY-rotter av hannkjønn (antall dyr 24) ble foret i 6 uker med et rottefår inneholdende 0,05 % (vekt/vekt) av forbindelsen fremstilt ifølge eksempel 1. Den gjennomsnittlige daglige dose av det aktive middel var 40 mg/kg, 5-10 ganger mer enn den terapeutiske dose. Kropps-vekten, matforbruket, motiliteten, kroppstemperaturen og opp-førselen til dyrene ble undersøkt. Disse undersøkelsene ble fortsatt i 2 uker etter fjerningen av det aktive middel. Fjerningssymptomer eller tegn på avhengighet ble ikke iakttatt i løpet av undersøkelsene, i motsetning til psykostimulerende midler av amfetamintypen.
6. Toksisitetsdata

De erholdte farmakologiske data viser klart at forbindelsen fremstilt ifølge oppfinnelsen har egenskapene til et psykostimulerende middel og dopaminagonist, i motsetning til 5H-2,3-benzodiazepiner og 3,4-dihydro-derivater og derav beskrevet i litteraturen hittil, som er hovedsakelig CNS-nedsettende. Som et ytterligere trekk, har forbindelsen ikke de ugunstige bivirkninger til de kjente psykostimulerende midler (emetisk virkning, tilbøyelighet til avhengighet).
Basert på resultatene av de farmakologiske undersøkel-ser har forbindelsen med formel (I) fremstilt ifølge oppfinnelsen og syreaddisjonssaltene derav terapeutisk betydning ved behandlingen av sykdommer som står i forbindelse med de funk-sjonelle forstyrrelser i de dopaminergiske systemer (f.eks. Parkinson's sykdom, galaktorré, etc). De verdifulle farmakologiske egenskaper og den lave toksisistet gir tilsammen en fordelaktig terapeutisk sikkerhet og virkningsområde.
Fremgangsmåten som benyttes ved fremstillingen ifølge oppfinnelsen er illustrert nærmere ved hjelp av de følgende eksempler. De fremstilte forbindelser ble identifisert ved hjelp av elementæranalyser, IR, ^"H-NMR og/eller massespektro-metri.
Eksempel 1
Fremstilling av 1-( 4- aminofenyl)- 4- methyl- 7, 8- methylendioxy-3, 4- dihydro- 5H- 2, 3- benzodiazepin
En blanding av 8,80 g (0,03 mol)
1-(4-aminofenyl)-4-methyl-7,8-methylendioxy-5H-2,3-benzodiazepin, 5,675 g (0,15 mol) natriumborhydrid og 80 ml pyridin ble omrørt på et vannbad oppvarmet til koking i 5 timer.
Etter at reaksjonsblandingen var avkjølt, ble 90 ml vann tilsatt, og deretter ble 260 ml av en vandig saltsyreoppløsning inneholdende 110 ml konsentrert saltsyre tilsatt under isav-kjøling, og så ble 115 ml 40 % vandig natriumhydroxydoppløs-ning dryppet til reaksjonsblandingen. Ved tilsetning av vannet oppstår hydrogenutviklingen, ved tilsetning av basen fraskilles den påtenkte forbindelse i form av en olje som inneholder pyridin. Blandingen ekstraheres så med benzen, benzenfasen rystes med vann, tørkes, klares med aktivt kull og inndampeS under redusert trykk. Resten taes opp i 50 ml vann, råproduk-tet frafiltreres, vaskes fem ganger med 10 ml vann, tørkes ved 50 - 60° C, rekrystalliseres fra 16 ml 50 % vandig ethanol og krystallvannet fjernes til slutt ved 100 - 105° C under redusert trykk.
Utbytte: 6,3 g (71,1 %), sm.p.: 118 - 120° C.

Eksempel 2
Fremstilling av 1-( 4- aminofenyl)- 4- methyl- 7, 8- mcthylendioxy-3, 4- dihydro- 5H- 2, 3- benzodiazepin- dihydroklorid
2,95 g (0,01 mol) 1-(4-aminofenyl)-4-methyl-7,8-methylendioxy-3,4-dihydro-5H-2,3-benzodiazepin ble oppløst i 50 ml 99,5 % ethanol oppvarmet til koking, deretter ble 1,77 ml (0,022 mol) konsentrert saltsyre og 50 ml benzen tilsatt, og oppløsningsmidlet i den gule reaksjonsblanding ble fra-dampet under redusert trykk. Resten ble tatt opp i 20 ml isopropanol, frafiltrert og tørket ved 60 - 80° C. Råproduk-
tet (3,7 g, dekomponerte og ble forkullet over 223° C) ble oppslemmet i 30 ml isopropanol oppvarmet til koking (en del av produktet ble oppløst), etter avkjøling ble de fine kry-stallene frafiltrert, vasket og tørket ved 100 - 120° C. Utbytte: 3,4 g (92,3 %), sm.p.: dekomponerer og blir forkullet
over 227° C.

Denne forbindelsen ble fremstilt i henhold til fremgangsmåten beskrevet i eksempel 2, med methansulfonsyre. Sm.p.: 215 - ;217° C (dekomponerer).
Ifølge IR- og, NMR-spektraene er protonet bundet til N-atomet

- Analogifremgangsmåte for fremstilling av terapeutisk aktivt 1-(4-aminofenyl)-4-methyl-7,8-methylendioxy-3,4-dihydro-5H-2,3-benzodiazepin med formel (I)samt syreaddisjonssalter av denne forbindelsen, karakterisert ved at 1-(4-aminofenyl)-4-methyl-7,8-methylendioxy-5H-2,3-benzodiazepin med formel (II)reduseres i et egnet oppløsningsmiddel med et uorganisk eller organisk-uorganisk hydrid og/eller komplekst metallhydrid og, om ønsket, basen eller basene omdannes til syreaddisjonssalter, eller saltene omdannes til den eller de frie baser.
