Abstract
Oppfinnelsen angår fremstilling av organiske amider med formel:. hvor R.-CO er en karboksylsyrerest som ikke utgjør en naturlig fettsyre normalt bundet til nitrogen i nitrogenholdige lipider, Rer et hydrogenatom, en mettet-alkylgruppe eller en mettet C_kloalkylgruppe og Rj-N er en nitrogenholdig lipid-rest. Forbindelsene kan benyttes til å øke eller stimulere den biologiske in vivo aktivitet av karboksylsyrer som er biologisk aktive in vitro.
Description
Foreliggende oppfinnelse angår en fremgangsmåte for fremstilling av en hittil ukjent antibiotisk stoffblanding eller dens bestanddeler eller salter derav.
Den her omhandlede blanding av hittil ukjente antibiotiske stoffer, kalles i det følgende generisk prasinomycin, og de enkelte bestanddeler betegnes prasinomycin A, prasinomycin B, prasinomycin C, prasinomycin D og prasinomycin E.
Foreliggende fremgangsmåte karakteriseres ved at en av mikroorganismene Streptomyces prasinus ATCC 15.825, Streptomyces hirsutus ATCC 19.091 eller Streptomyces prasino-pilosus ATCC 19.o92 dyrkes i et vandig næringsmedium inneholdende assimilerbare kilder for karbon og nitrogen under aerobe betingelser, hvoretter den antibiotiske blanding utvinnes fra dyrkningsmediet og om ønsket oppdeles i sine bestanddeler eller omdannes til et salt eller begge deler.
For å danne de antibiotiske stoffer kan den ønskede organisme dyrkes ved 2 5°C under submerse, aerobe betingelser i et vandig næringsmedium inneholdende eri assimilerbar karbonhydrat-og nitrogenkilde. Fermenteringen utføres i ca. 168 timer, og etter denne tid er de antibiotiske stoffer dannet.
Etter at fermenteringen er ferdig, ekstraheres prasinomycin fra myceliet med metanol, eller, fortrinnsvis fra hele fermenteringsbuljongen, ved at først justeres buljongen til en
pH på ca. 1,5 til 3,o med saltsyre, alle de faste stoffer fjernes ved filtrering og filterkaken ekstraheres med metanol.
De metanoliske ekstrakter nøytraliseres til pH 7 og konsentreres i vakuum og man får tilbake en vandig suspensjon.
Den vandige suspensjon behandles med en mineralsyre for å justere pH-verdien til 1,5 og ekstraheres deretter med kloroform eller butahol. Den organiske fase kan deretter tørkes med vannfri Na2SC>4 og konsentreres til et lite volum. Konsentratet fortynnes med tre til ti volumdeler aceton, hvorved det rå antibiotiske

stoff, utfelles,.
Det. rå prasinomycin kan renses ytterligere ved at det suspenderes i destillert vann og pH-verdien justeres til ca.
8,5 ved tilsetning av natriumhydroksyd. Den basiske vandige oppløsning ekstraheres med n-butanol for å fjerne nøytralt materiale. Den vandige fase ansyres til pH 1,5 med HCl og ekstraheres med n-butanol. Butanolekstrakten konsentreres under vakuum for å gi et mørkebrunt, amorft produkt.
Det rå prasinomycin renses fortrinnsvis ved å suspen-dere det i ti ganger sin egen vekt av destillert vann og justere- - pH-verdien til ca. 8,5 ved tilsetning av natriumhydroksyd. Den basiske, vandige oppløsning vaskes med n-butanol for å fjerne nøy-tralt materiale. Den vandige fase justeres til pH 5 og vaskes på nytt med n-butanol. Den vandige fase justeres til pH 1,5 - 2,o og ekstraheres med n-butanol. Butanolekstrakten nøytraliseres med natriumhydroksyd, <p>g blandingen inndampes til tørrhet. Man får den frie syre, og natriumionene fjernes ved å bringe en vandig oppløsning av natriumsaltet i kontakt med en sterkt sur ionebytterharpiks.
Adskillelse av de fem komponenter av den antibiotiske blanding utføres ved motstrømsfordeling i en atmosfære av. nitrogen ved papir-fordelingskromatografi.
Saltene av prasinomycinene fremstilles ved omsetning,
av det ønskede prasinomycin med den ønskede base. Enhver base kan anvendes. Slike baser omfatter metallbaser, så som alkalime-tallhydroksyder (f.eks. natriumhydroksyd og kaliumhydroksyd), og jordalkalimetallhydroksyder (f.eks. bariumhydroksyd); og organiske baser, så som primære, sekundære og tertiære aminer (f.eks. diety-lamin) og N-etylmorfolin-amin. Således fremstilles f.eks. natrium-saltene av prasinomycinene ved at det ønskede prasinomycin (i form av den frie syre) suspenderes i vann, og o,5N vandig oppløsning av natriumhydroksyd tilsettes inntil en pH-verdi på 8 er oppnådd. Blandingen omrøres deretter, og fem volumdeler etanol tilsettes
for å utfelle natriumsaltet. Bunnfallet filtreres fra og vaskes med etanol.
De ved fremgangsmåten ifølge oppfinnelsen anvendte mikroorganismer er kjente organismer, se L. Ettlinger, R. Corbaz og R. Hiitter "Zur Systematik der Aktinomyceten" , Arkiv Fiir Mikro-biologie, bind 31, side 326-358 (1958)... Til ytterligere karakteri-sering av de anvendte mikroorganismer gis nedenstående opplysninger.

Streptomyces prasinus som er isolert fra jordprøver fra.Colorado Springs, Colorado, kan isoleres og karakteriseres ved først å ryste en porsjon av jordprøven i sterilt, destillert vann og fordele den på et agarmedium inneholdende de følgende materialer:
Mediet justeres til pH 7,o og steriliseres i en^auto-klav ved 121°C i tretti.minutter. Etter syv til ti dagers inku-bering ved 25°C isoleres kolonier av Streptomyces prasinus fra den fordelte jord. Disse isolerte kolonier dyrkes deretter i et medium inneholdende:
Mediet autoklaveres ved 121°C i femten minutter. Organismen er i stand til å utnytte de følgende karbonkilder i et grunnmedium inneholdende (NH4)2S04 som en kilde for nitrogen: glukose, mannitol, inositol, xylose, arabinose og fruktose. Veksten understøttes ikke av sorbitol, ramnose og laktose.
Organismen virker proteolytisk på melk og gelatin, hydrolyserer stivelse, er H2s-ne9"ativ og reduserer ikke nitrat.
Det følgende er en beskrivelse av kolonier av mikroorganismen inkubert i forskjellige medier: Tomatpasta-havremel-agar, luftmyceliet er svakt grønt. Baksiden er artemisia-grønn M+P-15E5;
Gjær-maltekstrakt-agar, luftmyceliet er svakt grønt, baksiden er fløtegul. Ingen oppløselige pigmenter dannes.
En syntetisk salt-stivelsesagar, luftmyceliet er gressgrønt, og blir mørkt, gråaktig grønt etter hvert, og bak-

siden er rødbrun. Ingen oppløselige pigmenter dannes.
De generelle kjennetegn for mikroorganismen er: grønn sporefarge, sporoforer i kroker eller primitive spiraler, og ikke-kromogene i proteinholdige medier. Disse kjennetegn plas-serer organismen i Waksmans viridus-serie V.
Det følgende er en beskrivelse av kolonier av mikroorganismen Streptomyces hirsutus inkubert på forskjellige medier: Syntetisk agar: Veksten tilfeldig fargeløs; først er luftmyceliet sparsomt og melkehvitt, deretter blir det svakt grønt.
Syntetisk opløsning: Sediment og fnug hvitgrå; overflatevekst punktformig med hvitgrått luftmycelium.
Dekstroseagar: Vekst punktformig, fargeløs, intet luftmycelium. Dekstrose-asparagin-agar: Vekst punktformet, fargeløs, intet luftmycelium
De alminnelige karakteristika for Streptomyces hirsutus er: Sporer med stive spisser opp til o,6,u lange og likeledes smale ved grunnen. Luftmycelium mørkegrønt. Kjeder ay sporer i korte, åpne, ensartede spiraler med ca. 3 snoniger i de fleste tilfeller; monopodiale forgreninger. Ingen melamindannelse.
Det følgende er en beskrivelse av kolonier av mikroorganismen Streptomyces prasino-pilosus inkubert på forskjellige medier: Syntetisk agar: Vekst tilfeldig, murstensrød eller rødbrun; substrat lyserødt; luftmycelium fløyelsaktig, svakt grønt med hvite flekker.
Syntetisk oppløsning: Sediment, fnug lyserøde; substrat lyserødt; hinner med svakt grønt luftmyceliifim.
Dekstroseagar: Vekst meget sparsom, tilfeldig, murstensrød. Dekstrose-asparagin-agar: Vekst meget sparsom, murstensrød:, substrat murstensrødt;luftmycelium fløyelsaktig hvitt til svakt grønt.
De alminnelige karakteristika for Streptomyces prasino-pilosus er: Sporer med bøyelige, skjøre, meget fine filamenter opp til ^u i lengde. Luftmycelium mørkegrønt. Kjeder av sporer i korte, åpne, ensartede spiraler med kun 1-3 snoninger i de fleste tilfeller; monopodiale forgreninger. Ingen melamindannelse.
Mikroorganismene Streptomyces prasinus, Streptomyces hirsutus og Streptomyces prasino-pilosus er under de ovenfor an-førte deponeringsnumre deponert i og tilgjengelige hos American Type Culture Collection, Rockville, Maryland.

Prasinomycin (og alle dets komponenter) har en bred aktivitet mot gram-positive bakterier og Mycobacterium tubercu-losis, og begrenset aktivitet mot gram-negative bakterier.
Dobbelte rørfortynningsprøver ble utført med flere mikroorganismer. Det prasinomycin som ble anvendt ved dette forsøk, er av samme renhet som syrefraksjonen beskrevet i nedenstående eksempel 2.
Mus injiseres intraperitonialt med 1000 LD5Q doser av Streptococcus pyogenes C203, og fem timer etter injiseringen får de den antibiotiske blanding subcutant, med de følgende resultater:
En prasinomycinblanding bestående av 50% rent anti-biotikumkompleks og 50% indifferent materiale har begrenset aktivitet over for gramnegative bakterier som følger:
Prasinomycins in vitro-spektrum, bestemt ved en 2 gangers reagensglassfortynningsprøve, fremgår for prasinomycin A, B og C av nedenstående tabell:
Prasinomycin (fremstilt som beskrevet i nedenstående eksempel 2) hydratiseres lett når det utsettes for luft og opptar opp til 7,2% vann basert på sin vekt, hvilket vann går tapt ved tørking ved 80°C ved 0,1 mm Hg trykk.

Sterk syrehydrolyse av prasinomycin frigjør fosforsyre og ninhydrin-positive stoffer inklusive D-glukosamin og 6-desoksy-D-glukosamin.
Prasinomycin er. negativt når det prøves med ninhydrin eller sølvnitrat, men gir en blå farge ved behandling med 2% t-butylhypokloritt oppløst i cykloheksan fult av stivelsekalium-jodid-oppløsning.
Når bariumsaltet (fremstilt som i nedenstående eksempel 2) utsettes for luft, hydratiserer dét i en utstrekning av 9,7% vann, basert på sin vekt, hvilket vann går tapt ved tørking ved 80°C ved 0,1 mm Hg trykk.

Oppfinnelsen skal illustreres ytterligere ved de følgende eksempler: EKSEMPEL 1
Tomatpasta-havremel-agar-næringsmedier podes med Streptomyces prasinus (ATCC 15.825) . De inkuberes i syv til ti dager og anvendes deretter til å inokulere 100 ml vandig soyamel-medium som befinner seg i 500 ml Erlenmeyer-kolber. Sammensetningen av spiringsmediet er:
Mediet steriliseres i 20" minutter ved 121°C og ved 7 kg damptrykk. Spiringskolbene inkuberes ved 2 5°C i 72 til 96 timer i en roterende ryster.
En 10% overføring foretas fra spiringskolben til 500 ml Erlenmeyer-kolber inneholdende 100- ml av det følgende medium:
Soyamel 60 g
Glukose 50 g
CaC03 10 g
Destillert vann 1 liter
Fermenteringskolbene inkuberes og rystes i likhet med spirings-flaskene. Det uttas prøver etter 3,5 og 7 dagers forløp. Prøv-ene fremstilles ved at myceliet blir sentrifugert fra, ekstra-hert med samme volum metanol som væsken på toppen, og undersøkt ved en agar-diffusjonsmetode. Resultatene er som følger:
Prøveorganismen er Staphylococcus aureus 209P.

EKSEMPEL 2
En 570 1 porsjon av Streptomyces prasinus (ATCC 15.825) fermenteres som følger: (A) Fremstilling av inokulum iriokulum-kilde: Kultur av Streptomyces prasinus (ATCC 15.825) oppbevart ved lyofilisering i melk. 125 ml av mediet i en 500 ml kolbe inkuberes i 96 timer ved 25°C på den roterende ryster. 125 ml av den ovennevnte kultur settes til 1000 ml av det ovennevnte medium, bortsett framat agaren utelates, i en 4000 ml kolbe og inkuberes i 48 timer ved 25°C på en frem- og tilbakegående ryster. 1000 ml av det resulterende materiale settes deretter til 22 7 1 av et medium med følgende sammensetning og inkuberes i 72 timer:
Under inkuberingen luftes buljongen i en grad av 30 cm pr. min. overflate-lufthastighet og omrøres i de første 12 timer, og om-røres deretter ved en overflate-lufthastighet på 90 cm pr. min.
34 1 av det resulterende materiale settes til 570 1 av et medium med den følgende sammensetning og inkuberes i 168 timer, ledsaget av lufting og rysting.
Fermenteringsbuljongen ansyres til pH 1,5 - 3,0 med saltsyre,
6 Kg filterhjelpemiddel tilsettes, og alt det uoppløselige materiale fjernes ved filtrering.
Ti kg av den erholdte filterkaken ekstraheres to
ganger med metanol, under anvendelse av 15 1 porsjoner metanol
i 1 time. Kaken filtreres mellom ekstraheringene. Det samlede filtrat justeres til pH 6 - 7 med natriumhydroksyd, og oppløs-ningen konsentreres i vakuum for å fjerne metanolen, og man får tilbake en vandig suspensjon på ca. 1 liter.
Den vandige suspensjon, erholdt ved konsentrering av
den metanoliske ekstrakt, justeres til pH 1,5 med HCl og ekstraheres tre ganger med kloroform, under anvendelse av 0,25 volum-
del kloroform hver gang. pH-verdien bestemmes etter hver ekstrahering og justeres til pH 1,5 på nytt hvis det er nødvendig. De samlede kloroformekstrakter konsentreres i vakuum til en tung sirup.
Fire liter aceton settes til den tunge sirup, blandingen rystes i 1 time og filtreres. Kaken tørkes under vakuum ved romtemperatur. Fra 555 g av den tunge sirup får man 61,1 g av et aceton-uoppløselig, lysebrunt, amorft, fast stoff med en aktivitet på 725 enheter/mg.
Seksti g av det dannede aceton-uoppløselige produkt males til et fint pulver ved hjelp av morter og støter, suspenderes i 3,2 1 vann (pH 3,4) og justeres til pH 8,6 ved tilsetning av 2,5 NaOH (25 ml) under omrøring. Det gjenværende spor av uopp-løselig materiale ignoreres. Den basiske, vandige oppløsning ekstraheres tre ganger med 800 ml porsjoner av n-butanol. Den resulterende, mørkerøde, vandige fase ansyres til pH 2 med 5%
HCl (82 ml) og'ekstraheres tre ganger med 800 ml porsjoner av n-butanol. De samlede butanolekstrakter konsentreres under vakuum for å gi et mørkebrunt, amorft, surt produkt, 29,0 g, aktivitet 2.200 enheter/mg.
Videre rensning kan oppnåes ved en motstrømsfordeling
av den sure fraksjon i systemet kloroform, metanol, vann (5,4,2 volum/volum/volum). Den aktive fraksjon fordeles deretter igjen med et oppløsningsmiddelsystem av n-butanol, etylacetat, vann
2,1,3 volum/volum/volum). Rørene inneholdende den aktive fraksjon kombineres, inndampes til tørrhet under vakuum, residuet oppløses i metanol, og det fargeløse produkt utfelles ved tilsetning av etylacetat. Man får prasinomycin med en aktivitet på 2.655 enheteer/ mg, sm.p. (med spaltning) 165-165°C, og med det infrarøde spektrum som vist på fig. 1.

Alternativt kan ti g av det aceton-uoppløselige pulver erholdt ved den ovenfor beskrevne fremgangsmåte males til et fint pulver med morter og støter, suspenderes i 100 ml vann (pH 3,4)
og justeres til pH 9 ved tilsetning av 2,5 N NaOH (v,3 ml) under omrøring. Det gjenværende spor av uoppløselig materiale ignoreres. Den basiske, vandige oppløsning ekstraheres tre ganger med 75 ml porsjoner av n-butanol. Den resulterende mørkerøde, vandige fase justeres til pH 5 med 6N HCl (0,35 ml) og vaskes to ganger med 75 ml porsjoner av n-butanol. Den vandige fase ansyres til pH 1,5 - 2,0 med 6 N HCl (0,6 ml) og ekstraheres to ganger med 75 ml porsjoner av n-butanol. De kombinerte butanolekstrakter fra den siste ekstrahering justeres til pH 7 med NaOH, og blandingen konsentreres under vakuum for å gi det amorfe natriumsalt, 2 g, aktivitet 3.440 enheter/mg.
For å fremstille den frie syre oppløses 1 g av natriumsaltet i 25 ml destillert vann og syren frigjøres ved tilsetning av sterkt sur ionebytterharpiks, inntil pH-vérdien av oppløsningen er ca. 2,5. Harpiksen filtreres fra, og den frie syre utvinnes fra det vandige filtrat ved frysetrøking, og man får 0,7 g av et lysebrunt pulver med aktivitet 3.200 enheter/mg.
Metallsalter av prasinomycin
Prasinomycin suspenderes i vann (30 mg/ml). Barium-hydroksydoppløsning tilsettes til pH 8, og oppløsningen omrøres for å fullføre oppløsning av det antibiotiske stoff. Fem volumdeler etanol tilsettes, og bunnfallet av bariumsalt filtreres fra og vaskes med etanol.
På samme måte, men ved å anvende ekvivalente mengder natriumhydroksyd eller kaliumhydroksyd istedenfor bariumhydroksyd, får man de respektive natrium- og kaliumsalter.
EKSEMPEL 3
Ved å følge fremgangsmåten fra eksempel 2, men ved å anvende Streptomyces hirsutus (ATCC 19.091) istedenfor Streptomyces prasinus, får man 0,5 g prasinomycin med samme sammensetning som det ifølge eksempel 1 fremstilte produkt.

Ved å følge fremgangsmåten fra eksempel 2, men ved å anvende Streptomyces prasino-pilosus (ATCC 19.092) istedenfor Streptomyces prasinus, får man prasinomycin.

EKSEMPEL 5
Prasinomycin A, prasinomycin B, prasinomycin C, prasinomycin D og prasinomycin E isoleres ved motstrømsfordeling av materialet erholdt i eksempel 2, under anvendelse av oppløsnings-middelsystemet: n-propanol, n-butanol, 0,2M N-etylmorfolin (2,3,6 etter volum). Syren (4,4 g) oppløses i 200 ml av den laveste fase, justeres til pH 9 med N-etylmorfolin og underkastes 1500 overføringer i et motstrømsfordelingsapparat i en atmosfære av nitrogengass.
Materialet i rørene svarende til de enkelte aktive komponenter, kombineres, og oppløsningsmidlet fjernes under vakuum. De resulterende N-etylmorfolinsalter opptaes i vann, og de frie syrer frigjøres ved behandling med ionebytterharpiks. Harpiksen filtreres fra, og filtratet konsentreres til tørrhet under vakuum. Residuet utfelles fra metanoloppløsning ved tilsetning av etylacetat for å få de fargeløse, amorfe antibiotiske stoffer. Ved denne fremgangsmåte fikk man 600 mg prasinomycin A, 506 mg, prasinomycin B og 412 mg prasinomycin C.
De andre komponenter, prasinomycin D og E, kan, selv
om de ennu ikke er isolert, utskilles ved papir-fordelingskromatografi og påvises ved bioautografi. 0,1 g av materialet erholdt i eksempel 2 anbringes på dobbelt syrevasket filtrer-
papir (f.eks., "Munktells nr. S-302"), 56 cm langt, og filtrer-papirarket dyppes gjennom en oppløsning av aceton, nedre fase av oppløsningsmiddelsystemet (2:1 etter volum). Arket lufttørkes ved romtemperatur (2-3 minutter) for å fjerne acetonet, og frem-kalles deretter med den øvre fase av et oppløsningsmiddelsystem bestående av n-butanol, pyridin, vann (4:1:4 etter volum) i 16 timer ved romtemperatur. Oppløsningsmidlene fjernes ved luft-tørking, og arket bioautograferes på agar podet med Staphylococcus aureus 209P. Prasinomycin A kjennetegnes ved en Rf-verdi på 0,23, prasinomycin B ved Rf 0,28, prasinomycin C ved Rf 0,36
og prasinomycin D ved Rf 0,38, og prasinomycin E ved Rf 0,45.
Claims (1)
- Fremgangsmåte til fremstilling av antibiotisk stoffblanding kalt prasinomycin eller dens bestanddeler prasinomycin A, B, C, D og E eller salter derav, karakterisert ved at en av mikroorganismene Streptomyces prasinus ATCC 15.825, Streptomyces hirsutus ATCC 19.091 eller Streptomyces prasino-pilosus ATCC 19.092 dyrkes i et vandig næringsmedium inneholdende assimilerbare kilder for karbon og nitrogen under aerobe betingelser, hvoretter den antibiotiske blanding utvinnes fra dyrkningsmediet og om ønsket oppdeles i sine bestanddeler eller omdannes til et salt eller begge deler.
