Den foreliggende opfindelse angår en analogifremgangsmåde til fremstilling af hidtil ukendte organiske amidforbindelser, der er terapeutisk anvendelige i farmaceutiske præparater. Fremgangsmåden er ejendommelig ved det i krav l's kendetegnende del angivne. Disse 5 hidtil ukendte organiske amider består primært af en carboxylsyredel og en nitrogenholdig lipiddel.
Adskillige forbindelser vides at være virksomme in vitro og alligevel have kun ringe eller ingen virkning in vivo. Især vides mange carboxylsyrer at være vigtige for funktionen af det periferale nervesy-10 stem og centralnervesystem og er virksomme in vitro, men er enten uvirksomme eller kun i ringe grad virksomme in vivo. Det er kendt, at fx gamma-aminosmørsyre (GABA) indvirker på det centrale nervesystem in vitro og er blevet foreslået som en mulig inhibitionsformidler (Louis S. Goodman og Alfred Gilman, The Pharmacological Basis of 15 Therapeutics, 4. udgave, s. 429 (1970)). Det har imidlertid vist sig, at GABA er ineffektiv, når den administreres in vivo, således som det er blevet målt ved konvulsionstests hos mus.
Det har vist sig, at forbindelser med den almene formel I, der fremstilles ved at kombinere en carboxylsyre såsom GABA med et nitroso genholdigt lipid såsom et phospholipid eller sphingosin, fører til amidforbindelser, der er virksomme in vivo, idet de har en virkning, der langt overstiger virkningen af de tilsvarende carboxylsyre- eller lipidforbindelser, når disse administreres hver for sig. Fx giver amidet af GABA med en sphingosin en forbindelse, der er langt mere 25 virksom in vivo, end GABA alene. Tilsvarende er lysergsyre, dihy-drolysergsyre og isolysergsyre i det væsentlige uvirksomme, når de administreres alene, men når de kombineres med en sphingosin, fører dette til forbindelser med den almene formel I, der har en signifikant virkning in vivo som målt ved deres hypoprolactinæmiske virkning 30 på rotter.
De væsentligt forbedrede farmakologiske egenskaber ved forbindelserne med den almene formel I formodes at hidrøre fra disse forbindelsers evne til at gennemtrænge den hæmatoencephaliske barriere og/eller nå de perifere organer i langt højere grad end carboxylsyreforbindelser-35 ne alene. 2 fremgangsmåden ifølge opfindelsen, fremmer interaktionerne mellem den biologisk aktive forbindelse såsom carboxylsyren og stedet for de specifikke interaktioner, der findes i membranerne.
Forbindelserne, der fremstilles ved fremgangsmåden ifølge opfindel-5 sen, er således nyttige til at forbedre eller forøge den biologiske virkning in vivo af carboxylsyrer, som er biologisk virksomme in vitro, samt til at stimulere den biologiske virkning in vivo af carboxylsyrer, som er biologisk virksomme in vitro, men som kun har en ringe eller ingen virkning in vivo.

De ovennævnte R^-COOH-syrer, der fører til R^-CO-resten, er primært sådanne syrer, der er fundamentalt vigtige for det perifere nervesystem og centralnervesystemet. Disse syrer har den fælles farmaceuti-30 ske egenskab, at de er virksomme in vitro, men uvirksomme eller kun i ringe grad virksomme in vivo.

7 fremgangsmåder, der har vist sig at være særlig velegnede, er følgende de mest foretrukne.
1. Reaktionen mellem R^CO^-aziderne (svarende til R^-COOH-syre) og de nitrogenholdige lipidderivater. Fremstilling af R1CON3- 5 aziderne kan udføres på kendt måde.
2. Acylimidazolfremstillingsmetoden, der omfatter omsætning af R^COOH-syren med N, n'-carboxyldiimidazol, efterfulgt af omsætning af den således fremstillede acylimidazol med det nitrogenholdige lipid.
10 3. Blandet arihydrid-fremstillingsmetoden, som omfatter omsætning af R^COOH-syren og trifluoreddikesyre til dannelse af et blandet anhydrid, efterfulgt af omsætning af det blandede arihydrid med det nitrogenholdige lipid.
4. Fremstilling af chloridet af R^COOH-syren efterfulgt af omsæt- 15 ning af chloridet med det nitrogenholdige lipid.
5. Direkte omsætning mellem Rj_C00H-syren og det nitrogenholdige lipid i nærværelse af et carbodiimid (fx dicyclohexylcarbodi-imid, benzylisopropylcarbodiimid eller benzylethylcarbodiimid) eller et andet stof, der svarer til 1-hydroxybenzotriazol.
20 6. Direkte kondensation efter opvarmning af R^COOH-syren sammen med de nitrogenholdige lipidderivater.
7. Direkte omsætning mellem methylesteren af R^COOH-syren og det nitrogenholdige lipidderivat; denne omsætning fremmes ved opvarmning .
25 8. Fremstilling af en ester ved omsætning mellem en R^COOH-syre og en phenol (fx paranitrophenol) efterfulgt af omsætning af esteren med det nitrogenholdige lipid. Fremstillingen af en ester af syren og en phenol kan udføres på kendt måde.

Ved fremstillingen af de produkter med den almene formel I, der er afledt af gamma-aminosmørsyre, foretrækkes det at anvende den fremgangsmåde, der består af den indledningsvise fremstilling af et gamma- amino smør syrederivat, hvor amingruppen er knyttet til en beskyt-5 telsesgruppe, fx en phthaloyl- eller benzyloxycarbonylgruppe. Det således fremstillede derivat kondenseres derefter yderligere med det nitrogenholdige lipid under anvendelse af de ovenstående reaktioner. Beskyttelsesgruppen fjernes derefter ved en hensigtsmæssig reaktion, hvorved fås produktet med den almene formel I. Hvis beskyttelses-10 gruppen fx er phthaloyl, kan denne gruppe fjernes ved hydrazinolyse.
De forbindelser med den almene formel I, hvor R^COOH-syren indeholder en basisk gruppe (fx gamma-aminosmørsyre, nicotinsyre, lysergsyre eller dihydrolysergsyre), kan omdannes til et salt med terapeutisk anvendelige syrer såsom saltsyre, brombrintesyre, methansulfonsyre og 15 æblesyre.
Som anført ovenfor kan adskillelige forbindelser, især carboxylsyrer, kombineres med nitrogenholdige lipider til dannelse af amidforbindelser, der er farmaceutisk virksomme in vivo.

Farmakologiske egenskaber

Gamma-aminosmørsyre (GABA), ΝΙ^ίΟΗβ^-ΟΟΟΗ, er et endogent stof og er biologisk virksomt som følge af interaktion med en specifik receptor, men er i sig selv ude af stand til at gennemtrænge den encephaliske 5 barriere. GABA har således vist sig at være virksomt in vitro, men relativt uvirksomt in vivo. De forbindelser, der fremstilles ved fremgangsmåden ifølge opfindelsen, er derimod virksomme såvel in vitro som in vivo. For at vise denne aktivitet af forbindelserne fremstillet ved fremgangsmåden ifølge opfindelsen blev der udført in 10 vitro-tests, ved hvilke bindingsniveauerne af radioaktivt gamma-ami-nosmørsyre, %-GABA, til synapsismembraner i rotters cortex blev målt i henhold til den af Enna og Snyder (S.H., Mol. Pharmacol. 13, s. 442-353 (1977)) beskrevne metode.
I tabel 1 vises resultaterne af disse tests, udtrykt i % af det 15 bundne virksomme produkt, idet in vitro-aktiviteten af produkt 1 sammenlignes med aktiviteten af de enkelte bestanddele i produkt 1. Resultaterne viser, at sphingosin alene Ikke har nogen biologisk virkning, medens GABA alene har en virkning, der kan sammenlignes med virkningen af produkt 1.

De organiske amider, der er fremstillet ved fremgangsmåden ifølge opfindelsen ud fra nitrogenholdige lipider, kan anvendes som lægemidler til forskellige terapeutiske anvendelser, især sådanne anven-35 delser, som svarer til virkningen af de aktive syrer, ud fra hvilke amiderne fremstilles. Fx er derivater af lysergsyre, isolysergsyre, dihydrolysergsyre, 2-brom-lysergsyre, 2-brom-dihydrolysergsyre, 1-

41 methyl-lysergsyre, 1-methyl-dihydrolysergsyre, 1-methyl-2-brom-ly s ergsyre, 1-methyl-2-brom-dihydrolysergsyre, gamma-aminosmørsyre, valproinsyre, trimethoxybenzoesyre og nicotinsyre egnede til anvendelse som lægemidler, der kan udøve farmakologisk virkning på cen-5 tralnervesysternet. Derivaterne af lysergsyre, 2-brom-lysergsyre, 1-methyl-lysergsyre og l-methyl-2-brom-lysergsyre har også en signifikant virkning på uterus. Specifikt kan de forbindelser, der er fremstillet ved fremgangsmåden ifølge opfindelsen, og som i forsøg har vist sig at være virksomme mod isoniazid-konvulsioner og binding af 10 GABA in vitro, og de farmaceutiske præparater, der indeholder disse forbindelser, være terapeutisk nyttige til behandling af lidelser, som er forbundet med ændringer i funktionen af GABA-sys ternet, eftersom disse produkter er i stand til at forøge niveauerne af GABA i centralnervesystemet og i de specifikke cerebrale områder, hvorved 15 det GABA, der bundet til naturlige aminoalkoholer, sættes i stand til at gennemtrænge blod-hjernebarrieren. 25 De forbindelser, der har vist sig at være virksomme til irihibering af serumniveauerne af prolactin og til binding in vitro af den dopami-nerge ligand i hypofysen, og de farmaceutiske præparater, der fremstilles ud fra disse forbindelser, hvilket er eksemplificeret i de ovenfor anførte in vivo- og in vitro-data, kan med fordel anvendes 30 til behandling af lidelser, der viser sig som ændringer i afgivelsen af neuropeptider fra hypofysen såsom prolactin, som følge af forandringer i reguleringen af neurotransmittersystemerne ledsaget af tab af tonisk inhibering af det dopaminerge system eller i almindelighed af de hypothalamiske baner såsom ved hyperprolactinæmi forårsaget af 35 neuroleptica såsom sulpirid-chlorpromazin, etc.

42 Således kan lægemidler, der fremstilles ud fra de her omhandlede forbindelser, anvendes i behandlingen af adfærdsændringer, der er en følge af modifikationer af neuropeptidhormoner fra hypofysen såsom hyperprolactinæmiske syndromer med tab af libido og impotens og 5 hypopituitarismus med personlighedsændringer, apati, ligegyldighed, asteni, tab af libido og forvirring, præmenstruelle syndromer med depression og stemningssvingninger og klimaterielle syndromer med stemningssvingninger, irritabilitet, angst, nervøsitet og depression.
Forbindelserne, der fremstilles ved fremgangsmåden ifølge opfindel-10 sen, kan administreres i form af farmaceutiske præparater, der som den aktive bestanddel indeholder ét eller flere af amiderne sammen med én eller flere kompatible og farmaceutisk acceptable excipienser. Præparaterne kan administreres ad forskellig vej, som inj icerbare opløsninger, tabletter, gelatinekapsler og suppositorier. Den admini-15 strerede dosis varierer afhængig af den ønskede virkning og administrationsvej , men fx kan doserne ved oral administration være på mellem 10 og 300 mg aktivt stof om dagen, idet en enkelt dosis er på 10-100 mg.

15. Fremgangsmåde ifølge krav 1, kendetegnet ved, at der fremstilles 3,4,5-trimethoxyben-zoylphosphatidinylethanolaminamider med den almene formel DK 159204 B ch2-o-r I c CH-O-RI ’ I 9 . yOCH3 ch2-o-p-oh /z=\
0-CH2-CH2-NH-CO^--ά \-OCH3 ' ^OCH3 hvor R og R' betegner mættede eller umættede fedtsyrer.
16. Fremgangsmåde ifølge krav 15, kendetegnet ved, at R og R' er oleoyl-, stearoyl-, palmi-5 toyl-, linoleoyl- eller arachinoylradikaler.
