Die Prostaglandine sind eine Gruppe langkettiger, ungesättigter
Hydroxy- und Hydroxyketocarbonsäuren mit einem Cyclopentanring. Chemisch gesehen sind die Prostaglandine Derivate
der Prostansäure, die folgende Struktur hat:
Es gibt eine Reihe natürlich vorkommender Prostaglandine. Das Prostaglandin PGE^ hat z.B. die folgende Struktur

entspricht.
Die Prostaglandine kommen in vielen Säugetiergeweben in unterschiedlichen
Mengenverhältnissen vor und entsprechend verschiedenartig sind auch die physiologischen Eigenschaften dieser Substanzklasse.
So sind von den einzelnen Prostaglandinen eine
Reihe von recht interessanten pharmakologischen Wirkungen bekannt. Neben einer Beeinflussung der Herzfrequenz und einer
blutdrucksenkenden Wirkung verhindern die Prostaglandine auch 'die Blutplättchen-Adhäsivität und Aggregation, die den ersten Schritt zur Thrombose darstellen.
Reihe von recht interessanten pharmakologischen Wirkungen bekannt. Neben einer Beeinflussung der Herzfrequenz und einer
blutdrucksenkenden Wirkung verhindern die Prostaglandine auch 'die Blutplättchen-Adhäsivität und Aggregation, die den ersten Schritt zur Thrombose darstellen.
Trotz dieses recht wertvollen Wirkungsspektrums sind die
Prostaglandine bisher jedoch für pharmazeutische Zwecke kaum
zu verwenden, da ihre Halbwertszeit im Blut nur ca. 10 Minuten beträgt. Daher hat es in der Vergangenheit nicht an Versuchen

gefehlt, diesem Mangel durch geeignete Prostaglandin-Derivate abzuhelfen. So sind in den US-Patentschriften 3 632 627 und
3 746 728 Verfahren zur Veresterung von Prostaglandinen mit
Glycerin oder Glycerinphosphorsäureestern beschrieben, wobei Esterverbindungen erhalten werden, die der Formel

Bei diesen Verbindungen soll die physiologische Wirksamkeit, d.h. die Wirkungsdauer der Prostaglandine durch die Derivatisierung
schon verlängert worden sein.

Es wurde nun festgestellt, dass Ester»-oder Amidderivate
der Prostaglandine mit Phospholipiden, die noch über reaktionsfähige Gruppen im Molekül verfügen, wie z.B. Phosphatidyläthano1-amin,
Phosphatidylinositid, Phosphatidylserin und Phosphatidylglykol,
nicht nur einen gewissen Depoteffekt, d.h. eine verzögerte Freisetzung des Wirkungsprinzips und eine wesentlich gesteigerte
Wirkungsdauer, sondern überraschenderweise auch noch eine starke Organspezifität bezüglich der beobachteten Wirkung
aufweisen.
So zeigt z.B. das Prostaglandin Fp-Derivat des Phosphatidyläthanolamins
nach i.v.-Verabreichung praktisch keine Aktivität im Blut und nur wenig Aktivität in der Leber und im Magen, dagegen
aber eine starke Wirksamkeit in der Milz und in der Bauchspeicheldrüse. Auch die anderen Prostaglandine besitzen in Form
der erfindungsgemässen Phospholipidverbindungen ähnliche
spezifische Wirksamkeiten, wodurch diese neue Gruppe von Verbindungen zu einer wertvollen Bereicherung des Arzneimittelschatzes
führt.
Die erfindungsgemässen Phospholipid-Derivate der Prostaglandine

oder deren optische Isomere ist.
Die für die neuen Derivate benötigten Prostaglandine können entweder voll synthetisch oder biochemisch nach literaturbekannten
Verfahren hergestellt werden. Zur Umwandlung in die erfindungsgemässen Phospholipid-Derivate können die Prostaglandine
erfindungsgemäss amid- oder esterartig mit den entsprechenden
Phospholipiden in einem geeigneten Lösungsmittel zur Umsetzung
gebracht werden. Dazu löst man die Phosphatide in einem inerten Lösungsmittel und lässt die Reaktion unter Zusatz von Dicyclohexylcarbodiimid
unter dem Schutz eines inerten Gases vorzugsweise bei Temperaturen unter 500C ablaufen.
Als inerte organische Lösungsmittel kommen zum Zwecke der Erfindung z.B. chlorierte Kohlenwasserstoffe (wie Tetrachlorkohlenstoff,
Chloroform, Dichloräthan u.a.), Pyridin, aliphatische, aromatische sowie cyclische Kohlenwasserstoffe in Frage.
Die Reaktion ist im allgemeinen innerhalb von 48 Stunden beendet. Das Reaktionsprodukt kann durch allgemein übliche
Methoden, vorzugsweise jedoch durch Säulenchromatographie auf Kieselsäure oder durch präparative Dünnschichtchromatographie
gereinigt werden.

Phosphatidyläthanolamin wurde aus Sojarohphosphatid durch Säulenchromatographie
gereinigt. 15o( s150C-Dihydroxy-9-keto-5,13-prostadiensäure (PGE ) wurde biochemisch
dargestellt.
250 mg (ca. 0,33 mMol) Phosphatidyläthanolamin wurden in 20 ml trockenem Chloroform
gelöst. Es wurden 100 mg PGE (0,28 mMol) in 20 ml trockenem Chloroform
gelöst und anschließend 64· mg (0,31 mMol) Dicyclohexylcarbodiimid in 10 ml
Chloroform zugesetzt. Das Reaktionsgemisch wurde bei Raumtemperatur über Nacht gerührt. Es wurde im Vakuum eingeengt. Das Produkt wurde über Säulenchromatographie
mit Kieselsäure und Chloroform/Methanol als Eluent in Form eines gelben
Öles erhalten.
Die Ausbeute betrug 206 mg (0,21 mMol) oder 75 % d.Th. Das Infrarotspektrum
(KBr-Preßling) zeigt die Haupt-Absorptionen bei 31+50, 3050, 2925, 2860, 1740

und 1650 cm
in der gleichen Weise wurde erhalten:
PGE--Derivat
9,ll,15-Trihydroxy-13-prostensäure (PGF ) wurde nach Literatur-Verfahren
(E.J. Corey et al. (1970) J. Am. Oil Chem. Soc. 92_, 397 und 2586) synthetisiert.
Phosphatidylglykol wurde nach der Deutschen Patentschrift 1 949 399 aus Phosphatidyläthanolamin
hergestellt und säulenchromatographisch gereinigt. 115 mg PGF1 (0,3 mMol) wurden in 5 ml trockenem Pyridin gelöst und mit 300 mg
Trichloräthoxycarbonylchlorid in 5 ml trockenem Pyridin versetzt. Das Reaktionsgemisch wurde über Nacht bei Raumtemperatur gerührt, im Vakuum eingeengt und
durch Kieselsäuresäulenchromatographie gereinigt.
Die so gewonnene 9 ,lljlS-Tris-trichloräthoxycarbonyloxy-lS-prostensäure wurde
in 10 ml trockenem Pyridin gelöst und mit 375 mg (ca. 0,5 mMol) Phosphatidyl-

glykol und 125 mg (0,6 mMol) Dicyclohexylcarbodiimid in 10 ml trockenem
Pyridin versetzt. Das Reaktionsgemisch wurde über Nacht gerührt, dann
im Vakuum eingeengt und mit Zink und Eisessig von der Schutzgruppe befreit. Das Produkt wurde mit Hilfe von Kieselsäuresäulenchromatographie als ein
gelbes Öl erhalten. IR: 3450, 3050, 2920, 2850 und 1740 cm"1.
In gleicher Weise wurde das PGF -Derivat des Phosphatidylinositids
-C 9^11 jl5-dihy_droxv-5_jl3-p_rostadienYl)-inosit
als gelbes Öl erhalten.

- 2. Verfahren zur Herstellung der Verbindungen nach Anspruch 1, dadurch gekennzeichnet , dass ein Phospholipid r in Gegenwart von DicyclohexylcarlDodiimid mit einem Prostaglandin umgesetzt wird.
- 3. Verfahren nach Anspruch 2, dadurch gekennzeichnet, dass ein durch eine Schutzgruppe geschütztes Prostaglandin verwendet und nach Beendigung der Reaktion die Schutzgruppe unter milden Bedingungen abgespalten wird.
- 4. Arzneimittel, dadurch gekennzeichnet , dass es als Wirkstoff die Verbindungen nach Anspruch 1 enthalte709382/0076
