Description
Foreliggende oppfinnelse angår fremstilling av nye terapeutisk virksomme 16-fenoksy- og 16-(o, m eller p)-substituerte fenoksyderivater av (dl)-9-keto-lla,15a-dihydroksy-17,18, 19,20-tetranorprosta-4,3,13-trans-triensyre.
Prostaglandiner er klassisk blitt beskrevet som kjemisk nærstående 20 karbonkjedede hydroksyfettsyrer med et basis-skjellett slik man finner det i prostanoinsyre:
Prostaglandiner som har en hydroksylgruppe i C-ll-stillingen
og en ketogruppe i C-9-stillingen, er kjent som PGE-seriene, mens de som har en hydroksylgruppe istedet for ketogruppen,
er kjent som PGF-seriene, og blir ytterligere betegnet med et a- eller 3-suffiks for å angi konfigurasjonen på hydrok-sylgruppen i nevnte stilling. De naturlige forbindelser er a-hydroksysubstituerte forbindelser.
Det kan være forskjellige grader av umettethet i molekylet,
da spesielt ved C-5, C-13 og C-17, og umettetneten blir også angitt ved hjelp av et suffiks. Således vil f.eks.
PGFj- og PGE^-seriene referere seg til prostanoinsyrer med
en trans-olefinbinding ved C-13-stillingen, mens PGF2- og PGE2-seriene refererer seg til prostadiensyrer med en cis-olefinbinding i C-5-stillingen og en trans-olefin-

Fremstillingen av derivater av prostansyre har vist seg å
bli stadig viktigere ettersom man har kunnet påvise at
naturlige prostaglandiner har en rekke interessante biologiske og farmakologiske aktiviteter.
Størstedelen av undersøkelsene er blitt fokusert på modi-fikasjonen av de to sidekjedene, eller modifikasjoner av substituentene knyttet til cyklopentangruppen (se f.eks.
U. Axen et al., "Synthesis" Vol. 1, John Wiley and Sons Inc., New York, N.Y. 1973 og P.H. Bently, "Chem. Soc. Reviews"
2, 29 (1973)). Det er bl.a. beskrevet syntesen av prostaglandinanaloger med en dietylenisk (allenisk) umettethet i karboksylsyrekjeden, se f.eks. US-patent nr. 3.879.438. Syntesen av flere prostaglandinanaloger hvor alkylkjeden knyttet til C-15-stillingen i de naturlige forbindelsene er blitt erstattet med en aryloksymetylengruppe, er bl.a. angitt i US-patentene nr. 3864.387, 3.954.881 (9-keto-16-fenoksy-5,13-prostadienforbindelser), 3.985.791 (9a-hydroksy-16-fenoksy-4,5,13-prostatrienforbindelser) og belgisk patent nr. 806.995.
Ifølge foreliggende oppfinnelse fremstilles nye 16-fenoksy-og 16-substituerte fenoksy-9-keto-prostaglandinanaloger med formelen:
hvor R^ er en alkylgruppe med 1-4 karbonatomer, og X er hydrogen, o-, m- eller p-halogen (fluor, klor eller brom), o-, m- eller p-metyl eller o-, m- eller p-metoksy.
Linjene som er vist i ovennevnte formel og i de ettefølgende formler som "" indikerer at substituentene er i en ot-konfigurasjon, dvs. under planet i cyklopentanringen. Dobbeltbindingen ved C-13 i forbindelser som fremstilles ifølge foreliggende oppfinnelse har samme konfigurasjon som i naturlige prostaglandiner av PGE- og PGF-seriene, dvs. i trans-konfigurasjon.
De nye forbindelsene har assymetriske sentra og kan således fremstilles som racemiske (dl) blandinger eller som individuelle 8R-antimerer. De racemiske blandinger kan, om ønsket, spaltes på passende syntesetrinn ved hjelp av i og for seg kjente fremgangsmåter for oppnåelse av de respektive individuelle antimerer.
Alkylgruppen omfatter rette eller forgrenete alkylgrupper som, som nevnt, har opptil 4 karbonatomer, dvs. metyl, etyl, n-propyl, isopropyl, n-butyl og lignende.
I US-patent nr. 3.985.791 beskrives beslektede prostatrien-syrederivater og hovedforskjellen mellom disse og de som fremstilles ifølge oppfinnelsen finnes ved 9-stillingen, idet forbindelsene med formel (4) har en ketogruppe, mens forbindelsene i nevnte US-patent har en hydroksylgruppe. Videre anvendes forbindelsene med formel (4) hovedsakelig for behandling og hindring av sår i magesekken og i tolvfingertarmen, mens forbindelsene i nevnte US-patent er særlig egnet som luteolytiske midler hos pattedyr av hunn-kjønn.
Forbindelsene med formel (4) fremstilles ifølge foreliggende oppfinnelse ved at man hydrolyserer en forbindelse med formelen:
hvor R^ og X har de ovenfor angitte betydninger.
Hele reaksjonssekvensen inklusive oppnåelsen av utgangs-materialet kan illustreres som følger:

Utgangsforbindelsene med formel (1) kan fremstilles ved hjelp av de fremgangsmåter som er angitt i US-patent nr. 3.985.791.
Forbindelser med formel (1), fortrinnsvis de hvor R-^ er metyl, behandles med T-butyldimetylsilylhalogenoid, fortrinnsvis t-butyldimetylsilylklorid, i nærvær av imidazol, N-metylimidazol og lignende, fortrinnsvis imidazol, i nærvær av et organisk oppløsningsmiddel, f.eks. dimetylformamid, dimetylacetamid, pyridin og lignende, fortrinnsvis dimetylformamid, eller blandinger av disse, ved temperaturer fra -40°C til -10°C, fortrinnsvis fra -30° til -20°C, i tidsrom fra 5 til 24 timer, fortrinnsvis fra 15-20 timer, hvorved man får fremstilt 9-keto-lla,ISa-bis-t-butyldimetyl-silyloksyf orbindelser med formel (2). Eventuelle mono- og/ eller trisilyloksyforbindelser som eventuelt blir dannet, kan hydrolyseres slik det mer detaljert er beskrevet i det etterfølgende, hvorved man får fremstilt forbindelser med formel (1) som så kan resirkulere som utgangsforbindelser.
Forbindelsene med formel (2) blir så behandlet med et oksyda-sjonsmiddel, f.eks. kromtrioksyd, pyridindikromat og lignende, fortrinnsvis kromtrioksyd, i nærvær av pyridin, heksametylfosfortriamid, 3,5-dimetylpyrazol og lignende, fortrinnsvis pyridin, eller pyridiniumklorkromat med natriumacetat, og et organisk oppløsningsmiddel, f.eks. diklormetan, dikloretan og lignende, fortrinnsvis diklormetan, eller blandinger av disse, ved temperaturer fra -10° til +30°C, fortrinnsvis fra +15° til +25°C, i tidsrom fra 30 min. til 2 timer, fortrinnsvis fra 15. min. til 45 min., hvorved man får fremstilt 9-keto-lla,15a-bis-t-butyldimetylsilyloksy-forbindelser med formel (3). Fordelaktig kan denne reak-sjonen utføres under vannfrie betingelser i en inert atmos-fære, f.eks. under nitrogengass.
Forbindelser med formel (4) fremstilles ved hydrolyse av forbindelser med formel (3), fortrinnsvis syrehydrolyse med en organisk syre eller mineralsyre, f.eks. eddiksyre, monokloreddiksyre, propionsyre og lignende, eller blandinger av slike syrer, fortrinnsvis eddiksyre, ved temperaturer fra 0 til 35°C, fortrinnsvis fra 15° til 25°C, i tidsrom fra 10 til 24 timer, fortrinnsvis fra 15 til 20 timer.
Forbindelser fremstilt ifølge foreliggende oppfinnelse har prostaglandin-lignende biologiske aktiviteter, og kan således brukes ved behandlingen av pattedyr, hvor det er ønskelig med bruk av prostaglandiner. Således kan forbind-eiser fremstilt ifølge foreliggende oppfinnelse brukes for kontroll av astma-anfall, fordi de er bronkodilatorer og fordi at de dessuten viser anti-allergiske egenskaper ved at de hemmer en mediatorfrigjøring. I tillegg til dette kan de brukes for behandling av bronkierkramper hos pattedyr, eller alle de steder hvor det er ønskelig med bronkodilatorer. Forbindelsene viser også vasodilaterende egenskaper og kan derfor brukes for å kontrollere eller å lindre for høyt blodtrykk hos pattedyr, foruten at de har en sentralnervesystemdepressiv aktivitet hos pattedyr, og kan således brukes som beroligende midler.
Mer spesielt og overraskende har det vist seg at de fremstilte 9-keto-16-fenoksy-4,5,13-prostatrienforbindelser har vist seg å være sterkere hemmere for magesaftutskillelse og følgelig sterkere midler mot magesårdannelse enn de tilsvarende 9-keto-16--f enoksy-5 ,13-prostandienforbindelser . Således er de ifølge oppfinnelsen fremstilte forbindelser verdifulle ved behandling og hindring av magesår både i magesekken og i tolvfingertarmen.
De fremstilte forbindelser kan tilføres i en rekke forskjellige doseringsformer, enten alene eller i kombinasjon med andre farmasøytisk forenelige preparater, og kan brukes i form av farmasøytiske preparater beregnet for oral eller parenteral tilførsel eller for innhalering i forbindelse med bronkodilatorer. Typisk vil de tilføres som farma-søytiske preparater inneholdende i alt vesentlig den aktive forbindelsen og en farmasøytisk bærer. Den farmasøytiske bæreren kan enten være et fast materiale, en væske eller en aerosol, hvor forbindelsen er oppløst, dispergert eller suspendert, og kan dessuten inneholde mindre mengder av konserverende midler og/eller buffermidler for justering av pH. Som egnede konserveringsmidler kan man f.eks. bruke benzylalkohol og lignende. Egnede buffere innbefatter f.eks. natriumacetat og farmasøytiske fosfatsalter og lignende.
De flytende preparatene kan f.eks. være i form av oppløs-ninger, emulsjoner, suspensjoner, siruper og lignende. De faste preparatene kan være i form av tabletter, pulvere, kapsler, piller og lignende, fortrinnsvis i enhetsdoserings-form for enkel tilførsel eller mer nøyaktig dosering.
Egnede faste bærestoffer innbefatter f.eks. farmasøytisk kvalitet av stivelse, laktose, natriumsakkarin, talkum, natriumbisulfitt og lignende.
For innhalering kan forbindelsene med formel (4) tilføres
som en aerosol som innbefatter forbindelsene i et inert drivmiddel sammen et kooppløsningsmiddel, f.eks. metanol, sammen med eventuelle konserveringsmidler og buffere. Før ytterligere informasjon angående innhalgeringstilførsel av aerosoler, vises det til US-patentene nr. 2.868.691 og 3.095.355.
Forbindelsene fremstilt ifølge foreliggende oppfinnelse vil typisk tilføres i doser på 1-100 yg pr. kg kroppsvekt. Den presise effektive dose vil selvsagt være avhengig av til-førselsmåten, den tilstand som skal behandles, samt pasienten. For f.eks. å oppnå en bronkodilatering vil man måtte tilføre 1-10 yg pr. kg kroppsvekt i en aerosol, og for å hemme magesyreutskillelse må man tilføre 1-50 yg pr. kg kroppsvekt oralt.
Følgende eksempel illustrerer oppfinnelsen. Det er under-forstått at isolering av de her beskrevne forbindelser kan utføres på mange egnede måter hva angår utseparering og rensning, f.eks. ekstraksjon, fordampning, filtrering, destillasjon, krystallisering, tynnsjiktkromatografi, høytrykks-væskekromatografi eller kolonnekromatografi eller ved en kombinasjon av disse fremgangsmåter. Illustrasjoner på egnet separasjons- og isolasjonsteknikk fremgår av de fremstillinger og eksempler som er beskrevet nedenfor.
Man kan selvsagt også bruke andre fremgangsmåter for separasjon og rensning.
Eksempel
a) En oppløsning av 0,50 g (dl)-9a,lia,15a-trihydroksy-16-fenoksy-17,18,19,20-tetranorprosta-4,5,13-trans-triensyremetylester (1) med følgende fysikalske konstanter: MeOH U.V. %l£g 220, 265, 271, 278 nm (log e 3,99, 3,11, 3,23,

Dietyleteroppløsningen ble tørket over vannfritt natriumsulfat og fordampet under redusert trykk, hvorved man fikk (dl)-9a-hydroksy-lla,15a-bis-t-butyldimetylsilyloksy-16-fenoksy-17,18,19,20-tetranorprosta-4,5,13-trans-triensyremetylester (2) og en mindre mengde av tri- og mono-t-butyldimetylsilyloksyanalogene tilsvarende denne forbindelse.
Den urene reaksjonsblandingen ble kromatografert på en kolonne inneholdende 100 g silisiumdioksygel og eluert med etylacetatheksan (15:85), hvorved man fikk (dl)-9a-hydroksy-lla,15a-bis-t-butyldimetylsilyloksy-16-fenoksy-17 , 18 , 19,20-tetranorprosta-4,5,13-trans-triensyremetylester (2)
med følgende fysikalske konstanter:

De mer polare monosilylerte og de mindre polare trisilyl-erte kromatografiske fraksjoner kan omdannes til utgangs-forbindelsen med formel (1) for deretter å omdanne dette igjen til forbindelse med formel (2), ved f.eks. å bruke følgende hydrolysefremgangsmåte: En oppløsning av 0,2-1,0 g av nevnte mono- og trisilylderi-vatene som ellers tilsvarer de forbindelser som er beskrevet ovenfor, i 250 ml eddiksyre-vann (65/35; vol./vol.) ble ved romtemperatur omrørt i 15-20 timer. Eddiksyre-vannet ble fjernet under redusert trykk fulgt av en azeotrop vakuum-destillasjon, idet man brukte 100 ml toluen. Den således oppnådde regenererte triol (1) kan så direkte omdannes til sitt lia,15a-bis-t-butyldimetylsilyloksyderivat (2) slik dette er beskrevet ovenfor.
b) 0,77 g vannfritt kromtrioksyd ble tilsatt en omrørt oppløsning av 1,5 ml tørr pyridin i 20 ml tørr diklormetan
og omrørt under en tørr nitrogenatmosfære ved 20°C i 15 minutter, hvoretter man tilsatte en oppløsning av 305 mg (dl)-9a-hydroksy-lla,15a-bis-t-butyl-dimetylsilyloksy-16-fenoksy-17,18,19,20-tetranorprosta-4,5,13-trans-triensyremetylester (2) i 10 ml tørr diklormetan, hvoretter reaksjonsblandingen ble omrørt i en halv time ved 20°C. Opp-
løsningen ble avhelt fra resten og denne ble vasket med to 200 ml porsjoner av dietyleter. De organiske oppløsninger ble kombinert, vasket suksessivt med tre 50 ml porsjoner med vann og så tørket over vannfritt natriumsulfat. Fordampning under redusert trykk gir en oljeaktig rest som ble kromatografert på en silisiumdioksydgelkolonne, idet man ut-førte en eluering med etylacetat-heksan (15:85), hvorved man fikk (dl)-9-keto-lla,15a-bis-t-butyldimetylsilyloksy-16-fenoksy-17,18,19,20-tetranorprosta-4,5,13-trans-triensyremetylester (3) med de følgende fysikalske konstanter:

c) En oppløsning av 230 mg (dl)-9-keto-lla,15a-bis-t-butyldimetylsilyloksy-16-fenoksy-17,18,19,20-tetranor-prosta-4,5,13-trans-triensyremetylester (3) i 260 ml eddiksyre inneholdende 140 ml vann ble omrørt ved 20-25°C i 15-20 timer.
Eddiksyrevannet ble fjernet under redusert trykk fulgt av
en azeotrop destillasjon ved redusert trykk, idet man brukte 100 ml toluen. Den oljeaktige rest ble kromatografert på en silisiumdioksydgelkolonne og eluert med etylacetat-
heksan (25/75:100/0), hvorved man fikk (dl)-9-keto-lla,15a-dihydroksy-16-fenoksy-17,18,19,20-tetranorprosta-4,5,13-trans-triensyremetylester (4) med følgende fysikalske konstanter : U.V. xMe?H 220, 265, 271, 277 nm (log e 4,01,

Histaminindusert magesyresekresjonsprøve
Forsøk_A
Sprague-Dawley (Hilltop) hannrotter ble brukt i dette for-søket. Dyrene hadde en sirkelformet plastkrage festet om-kring nakken for å hindre adgang til mat eller avføring og for å sikre en tømming av magen i løpet av 48 timers sulteperiode. Prøveforbindelsene ble tilført oralt ved hjelp av et rør den morgenen eksperimentet skulle utføres, og 30 minutter før operasjonen. Under denne fremgangsmåten ble dyrene eterbedøvet, og en ombinding ble plassert på tolvfingertarmen nær den pyloriske ringmuskelen, og en annen på esofagusposterior til larynks. Laparotomien ble lukket med treklyper, og 2,5 mg pr. kg kroppsvekt av histamindi-fosfat ble injisert subkutant, og ved 15 minutters inter-valler deretter i 3 timer for stimulering av magesyresekre-sjonen. Ved slutten av 3 timers-perioden ble-rottene obdu-sert, og magesyreinnholdet i magen tatt ut og dets volum ble målt. En mengde av magesyren ble titrert med 0,02 N NaOH til pH = 7,0 ± 0,1 sluttpunkt på et pH-meter. Mage-syre utskilt ble så beregnet som milliekvivalenter pr.
100 g kroppsvekt. Behandlede grupper ble så sammenlignet
statistisk for kontroll. I denne prøven ble (dl)-9-keto-lla , 15a-dihydroksy-16-fenoksy-17,18,19, 20-tetranorprosta-4,5,13(t)-triensyremetylester gitt i doser på 7,5-1000 pg/ kg, og man fant at forbindelsen hadde ID^q på 21 ug/kg.
Forsøk_B
Simonsen hannmus (18-24 g) ble oppdelt i to grupper på 8 dyr. Hver dose ble gitt til 8 mus. Injeksjoner ble utført via halevenen. Prøvede mus ble observert i løpet av de neste 48 timer.
Resultat:
I denne prøven fant man at (dl)-9-keto-lla,15a-dihydroksy-16-fenoksy-17,18,19,2 0-tetranorprosta-4,5,13(t)-triensyremetylester hadde en LD^q på 1,45 mg/kg kroppsvekt.
