Abstract

Esta invención proporciona un proceso en un solo recipiente para preparar pergolida que comprende reaccionar 8, 9-dihidroelimoclavina con 1-yodopropano a elevada temperatura en presencia de un bicarbonato o carbonato de calcio, magnesio o alcalino metálico y un disolvente seleccionado N-metilpirrolidinona, hexametilfosforamida, dimetilpropilenourea y dimetiletilenourea hasta que la 8, 9-dihidroelimoclavina se consume substancialmente para preparar una primera mezcla intermedia; diluir la primera mezcla intermedia con un volumen grande de un disolvente básico seleccionado de piridona, picolina y lutidina, enfriar la mezcla diluida a una temperatura baja, combinarla con un haluro de tolueno o de Cl-C3 alcanosulfonilo, y dejar que la reacción continúe a baja temperatura hasta que el primer intermedio se ha consumido substancialmente para preparar una segunda mezcla intermedia; combinar la segunda mezcla intermedia fría con una solución fría de tiometóxido alcalino metálico y dejar que la reacción continúe a elevada temperatura hasta que el segundo intermedio se consuma substancialmente para preparar una mezcla de pergolida bruta; diluir la mezcla de pergolida bruta con agua para obtener pergolida sólida y lavar la pergolida con agua para obtener pergolida de al menos 90 por ciento de pureza.
