Abstract
La presente invención se refiere a un método para aumentar la actividad catalítica de una proteína que requiere grupo Fe-S en una célula hospedera de levadura recombinante, caracterizado porque comprende: (a) proporcionar una célula hospedera de levadura recombinante que comprende al menos un polinucleótido heterólogo que codifica para una proteína que requiere grupo Fe-S, en donde la proteína que requiere grupo Fe-S es una dihidroxi-ácido deshidratasa (DHAD); (b) eliminar la expresión de un polipéptido que afecta la biosíntesis de grupo Fe-S en la célula hospedera de lavadura recombinante, en donde el polipéptido que afecta la biosíntesis de grupo Fe-S se selecciona de FRA2, GRX3 y CCCI; (c) cultivar la célula hospedera de levadura recombinante obtenida de la etapa (b) bajo condiciones en las que se aumente la actividad catalítica de la DHAD.
