Abstract
Se desarrolló una familia de receptores de antígeno quimérico (CARs) que contiene scFv específico CD123 para dirigir diferentes epítopos en CD123. En algunas modalidades, tal gen del receptor de antígeno quimérico CD123 (CD123CAR) incluye una región scFv anti-CD123 fusionada en el marco a una región de bisagra IgG4 modificada que comprende una sustitución S228P, una sustitución L235E, y opcionalmente una sustitución N297Q; un dominio de señalización coestimulante; y un dominio de señalización de cadena zeta del receptor de célula T (TCR) . Cuando se expresan en células T de donador saludable (CD4/CD8), los CD123CARs redirigen la especificidad de la célula T y median la actividad efectora potente contra las líneas de célula CD123+ así como muestras de paciente AML primarias. Además, las células T obtenidas de pacientes con AMIJ activo, pueden modificarse para expresar genes CD123CAR y son capaces de usar blastos AML autólogos in vitro. Finalmente, una dosis sencilla de 5.0 x 106 células T CAR123 resulta en un progreso leucémico significativamente retardado en ratones.
