본 발명은 2',3'-디데옥시노신, 2',3'-디데옥시-2',3'-디데히드로티미딘 및 2',3'-디데옥시-2'-플루오로이노신의 높은 수용성을 가지고 안정한 항비루스(항역비루스를 포함) 결정체염, 특히 그의 나트륨염에 관한 것이다.
감염성 비루스 질병은 중요한 의학적 문제로서 인식되어 왔다. 감염성 비루스 질병에 대한 증진은 정상세포주에 양성으로 유지되는 반면에 선택적 항비루스 활성을 가진 약물의 개발을 필요로 한다. 현재 연구중, 약간의 선택성을 소유한 듯한 일련의 항비루스제는 뉴클레오시드 동족체이다. 일반적으로 이들 화합물은 자연발생 뉴클레오시드의 구조적 동족체이다. 푸린 또는 피리미딘 염기핵 및/또는 사카라이드 성분에서 구조적 변형은 비루스 핵산 형성과정에 삽입될때 비루스 핵산의 추가 합성을 저해토록 작용하는 합성적 변형 뉴클레오시드 유도체를 만든다.
이들 항비루스제의 효능은 숙주 효소에 의해서가 아닌 비루스 효소에 의하여, 그후 삼인산염으로 전환되고 비루스 핵산에 삽입으로 이어지는 상응하는 뉴클레오티드 동족체로의 선택적 전환에 의존한다. 이와 같은 항비루스 전략으로써 문제는 비루스 균주의 효소가 뉴클레오시드 동족체의 인산화반응을 촉진시키지 못하는 어떠한 비루스 균주의 출현인바 있다. 이와 같은 문제를 해결하기 위하여, 완전한 뉴클레오티드 동족체가 비루스 핵산에 삽입을 위한 항비루스로서 강력하게 아주 유용한듯하다.
미쯔야와 브로더에 대한 PCT 출원 WO87/01284호에서는 AIDS에 대한 용도로서 2',3'-디데옥시노신의 사용을 기재하고 있다.
린과 프루소프의 EP 273,277호에서는 역비루스로 감염된 환자를 치료하는데 2',3'-디데옥시-2',3'-디데히드로티미딘의 사용을 기재하고 있다.
Potential Drugs for the Treatment of AIDS, Erik De Clercq, Journal of Antimicrobial Chemotherapy, 23, Suppl. A, 35-46, (1989)에서는 사람의 면역결 핍 비루스(HIV)의 감염 및 세포변성 효과를 억제하는 디데옥시뉴클레오시드 동족체의 사용을 기재하고 있다.
EP 287,313호에서는 2',3'-디데옥시-2'-플루오로이노신(F-ddI) 및 HIV에 대한 그의 활성을 개시하고 있다.
본 발명은 다음 일반식(I)의 2',3'-디데옥시-2',3'-디데히드로티미딘(D4T''), 일반식(II)의 2',3'-디데옥시이노신(DDI) 및 일반식(III)의 2',3'-디데옥시-2'-플루오로이노신(F-ddI)의 수용성이 매우 높고 안정한, 결정체염에 관한 것이다.
(상기 식에서 X는 양이온, 예를들어, Na, K 또는 Mg이다).
본 발명은 또한 이와 같은 염의 항비루스 유효량 및 고체, 액체 및 기체상의 생리적으로 허용되는 희석제로 이루어진 약제 조성물에 관한 것이다.
또한 추가로, 본 발명은 온혈동물, 예를들어, 사람에게 이전에 언급된 염 한가지의 항비루스 유효량을 투여하는 것으로 이루어진 이와 같은 온혈동물의 치료방법에 관한 것이다.
상기에 제시된 것처럼, 본 발명은 예를들어, Na+, Li+, K+, Ca++및 Mg++를 함유한 2',3'-디데옥시이노신, 2',3'-디데옥시-2'3'-디데히드로티미딘 및 2',3'-디데옥시-2'-플루오로이노신의 약리적으로 허용되는 비독성염에 속한다. 이와 같은 염은 유기 양이온과 또한 알칼리 또는 알칼리토금속 이온 또는 암모늄 또는 사차 아미노 이온과 같은 적합한 양이온을 함유한 것들을 들수 있다. 보다 바람직한 염은 나트륨염이다. 수산화 금속을 2',3'-디데옥시이노신, 2',3'-디데옥시-2',3'-디데히드로티미딘 또는 2',3'-디데옥시-2'-플루오로이신과 반응시켜 금속염을 제조할 수 있다. 용해도가 적은 금속염은 용해도가 큰 염용액으로부터 적합한 금속이온의 첨가에 의하여 침전될 수 있다.
본 발명의 화합물은 바람직한 항비루스 활성을 가지고 있다. 그들은 비루스, 예를들어 헤르페스 심플렉스 비루스 I, 헤르페스 심플렉스 비루스 Ⅱ, 시토메탈로 비루스, 바리셀라 조스터 비루스, 인플루엔자 비루스, 백시니아, 소아마비, 풍진, 천연두, 우두, 엡스타인-바르 비루스, 홍역 비루스, 사람의 호흡기 비루스, 유두종 비루스 및 신비스 비루스, 바로 언급된 여러가지에 대하여 그리고 또한 역비루스, 예를들어, 사람의 면역결핍 비루스(HIV)에 대한 활성을 나타낸다.
상기에 언급된 것처럼, 본 발명의 화합물은 역비루스에 의하여 야기된 질병의 치료 및 예방을 위한 인체 및 수의 의약에 유용한 활성성분이다. 역비루스에 관한 인체 의약에서 지정분야의 실예는 다음과 같다 :
(1) 사람의 역비루스 감염의 치료 또는 예방 ;
(2) HIV(사람의 면역결핍 비루스 ; 이전에 HTLVIII/LAV 또는 AIDS로 지칭됨)에 의하여 야기된 질병과 ARC(AIDS 관련 복합체) 및 LAC(림프선증 증후군)와 같이 그들과 수반된 단계 및 이와 같은 역비루스에 의하여 야기된 면역약화 및 뇌질환의 치료 또는 예방 ;
(3) HTLV I 감염 또는 HTLVII 감염의 치료 또는 예방 ;
(4) AIDS 운반상태(AIDS 전이상태)의 치료 또는 예방 ;
(5) 간염 B비루스에 의하여 야기된 질병의 치료 또는 예방.
수의 의약에서 지정실에는 다음과 같다 :
(1) 마에디비스나(Maedivisna) (양과 염소에서),
(2) 진행 폐렴 비루스(PPV) (양과 염소에서),
(3) 카프린 아르트리티드 앤세팔리티스 비루스(양과 염소에서),
(4) 즈뵈게르지크테(Zwoegerziekte) 비루스(양에서),
(5) 빈혈증의 감염성 비루스(말에서) 및
(6) 고양이의 백혈병 비루스에 의하여 야기된 감염.
비루스 가염에 대한 용도로서 본 발명의 화합물은 약제조제로 배합될 수 있다. 이와 같은 조제는 약리적으로 허용되는 담체와 수반하여 본 발명의 화합물 한가지 또는 그 이상으로 구성된다. 참고문헌(Remington's Pharmaceutical Sciences, 17판,A.R. Gennaro, editor, 편집자(Mack Publishing Company, 1985)에서는 전형적인 담체 및 제조방법을 기재하고 있다.
본 발명의 화합물은 온혈동물, 예를들어 사람에게 전신적으로 투여된다. 전신투여란 경구, 직장 및 비경구적(즉, 근육내, 정맥내, 피하 및 비강)경로를 뜻한다. 일반적으로, 본 발명의 화합물이 경구투여될때, 비경구로 제공된 적은 양과 동일한 효과를 나타내기 위하여 다량의 활성제를 필요로 할 수 있다. 양호한 임상적 실제에 따라, 어떠한 유해하거나 난처한 부작용을 야기시킴이 없이 유효 항비루스 효과를 나타낼 농도 수준에서 본 화합물을 투여하는 것이 바람직하다.
치료 및 예방적으로 본 화합물은 통상 상기에 언급된 바와 같이 본 발명에 따른 화합물의 항비루스 유효량과 한가지 또는 그 이상의 약리적으로 허용되는 담체로 구성된 약제조성물로서 제공된다. 이와 같은 처리에 효과가 있는 약제조성물은 약제담체와 조합하여 본 발명의 화합물 적어도 한가지를 다량 또는 소량, 예를들어 100-0.5% 함유할 것이며 담체는 한가지 또는 그 이상의 고체, 반고체, 또는 액체 희석제, 충전제 및 비독성, 불활성 및 약리적으로 허용되는 배합 부형제로 이루어진다.
이와 같은 약제조성물은 투여량 단위형태, 즉, 필요로 한 치료반응을 나타내도록 계산된 투여량의 분율 또는 배수에 상응하는 약물의 미리 결정된 양을 함유하는 물리적으로 분리되는 단위가 바람직하다. 다른 치료제가 또한 존재할 수 있다.
단위투여량당 활성성분 약 50mg-2g을 제공하는 약제조성물이 바람직하며 통상적으로 정제, 당의정, 캡슐, 분제, 입제, 수용성 또는 유성 현탁액, 시럽, 엘릭시르 및 수용액으로서 제조된다. 바람직한 경구 조성물은 정제 또는 캡슐의 형태이며 결합제(예, 시럼, 아카시아, 젤라틴, 소르비톨, 트라가간트 또는 폴리비닐피롤리돈), 충전제(예, 락토스, 당, 옥수수전분, 인산칼슘, 소르비톨, 또는 글리신), 윤활제(예, 스테아린산 마그네슘, 탈크, 폴리에틸렌 글리콜 또는 실리카, 방부제(예, 전분), 완충제(무기 또는 유기) 및 습윤제(예, 황산 라우릴나트륨)와 같은 종래의 부형제를 함유할 수 있다. 종래의 약제담체와 함께 발명 화합물의 용액 또는 현탁액은 정맥내 주상용 수용액 또는 근육내 주사용 유성 현탁액과 같은 비경구 조성물에 대하여 사용된다. 비경구적 사용을 위한 필요로한 명도, 안정성 및 적합성을 가진 이와 같은 조성물은 물 또는 글리세린, 프로필렌 글리콜 및 폴리에틸렌 글리콜 또는 이들의 혼합물과 같은 지방족 다가 알코올로 이루어진 담체에 활성인 발명 화합물 0.1중량%-35중량%를 용해시켜 얻어진다. 폴리에틸렌 글리콜은 물 및 유기용액 두가지 모두에 용해되는 비휘발성의 통상적으로 액체인 폴리에틸렌 글리콜의 혼합물로 이루어지며 약 200-1500의 분자량을 가진다.
발명 화합물은 산 가수분해 경향이 있다. 최적의 생이용성을 위하여 경구투여에 필요한 정제, 캡슐 및 입제는 장피복, 위산에 대한 강한 완충, 또는 두가지 모두가 되어야 한다.
본 특허출원의 염은 안정하고 결정체이며 물에 매우 높은 용해도(300mg/ml)를 가지며 고농도의 저부피 IV-IM 주사제에 이상적으로 적합하다. 가장 중요한 것으로서 이들 새롭고 신규의 염중 고알칼리성(pH 9.5-11)은 위산에 대한 자체-완충을 허용한다. 따라서, 경구투여 형태에서 완충이 덜 될 수 있다.
본 발명의 염은 엔올 형태로 가역적으로 고정되며 따라서 엔을 에스테르(원약물) 및 엔올 에테르와 같이 변형된 강력한 생활성 유도체의 합성용 중간체로서 고려될 수 있다. 에스테르 및 에테르는 또한 다른 작용에 대한 합성작업을 허용하는 보호(차단)기 또는 그 분자의 성분으로서 작용할 수 있다.
본 발명의 화합물이 가지고 있는 생물활성을 고찰하면, 이들 화합물은 특히 그 사용이 비루스 감염을 퇴치하는데 적합항 항비루스 성질을 가진다는 것을 알 수 있다. 따라서, 본 발명의 또다른 일예는 포유루에 발명 화합물의 유효 투여량을 전신 투여하는 것으로 이루어진 이와 같은 치료를 필요로한 포유류에서 비루스(역비루스 포함) 감염의 치료방법에 관한 것이다.
시험을 기초로하여 유효투여량은 약 0.1-약 5mg/kg 체중, 바람직한 투여량 범위 약 1-4mg/kg 체중인 것으로 예상될 수 있다. 임상적 항비루스 응용에 대하여 본 발명의 화합물은 참조약물 지도루딘(AZT)에 대한 것처럼 동일방식으로 투여될 것이다. 그러나 임상적 응용에 대하여, 시술자의 연령, 체중 및 상황, 투여경로 및 병의 성격 및 정도에 대한 건전한 직업적 판단 및 고찰을 이요하여 각 경우에 투여량 및 투여요범이 주의깊게 조절되어야 한다. 일반적으로, 일간 경구투여량은 1일 1-3회 투여되는 발명화합물 약 150mg-약 5g, 바람직하게는 50-1500mg으로 구성될 것이다. 여러가지 일예에 있어서, 보다 낮은 투여량에서 충분한 치료효과를 얻을 수 있지만 다른 것에서 보다 큰 투여량이 필요할 것이다.
본 발명을 구성한 화합물 및 그들의 제조방법은 예시목적으로만 제공된 다음의 실시예의 고찰로부터 보다 상세하게 나타날 것이며 본 발명의 영역 또는 범위를 국한하는 것으로서 해석되지 않는다. 모든 온도는 특정화되지 않을때 도 C로 이해된다. 모든 화합물은 만족할만한 성분분석을 제공하였다.
실시예 1
나트륨-DDI 모노히드레이트의 제조
MW : DDI=236.14
MW : NaOH=40
1. DDI 1g을 5분간격에 걸쳐 10-25℃에서 정상 교반하면서 1N-NaOH 수용액 4.5ml(1.05몰 당량)에 첨가한다.
2. 필요하다면, 정제하기 위하여 0.2-0.45마이크론 겔만 HT-터프린 여과기의 등가물로 용액을 통과시킨다. 단계 1과 2는 1.5시간내에 완료된다.
3. 10-15분간 간격에 걸쳐 Na-DDI 용액을 매우 세게 교반된 이소프로판올(달리, 아세톤이 사용될 수 있다) 50-70ml에 첨가시킨다. 결정체를 형성한다. 2시간동안 센교반(폐쇄계)을 계속한다.
4. 적합한 진공여과 방법을 통하여 결정체를 수집한다. 과량의 공기를 필터-케이크로 통과시키지 않는다. 결정체 필터-케이트를 죄어 형성할 수 있는 어떠한 균열 또는 파열을 제거한다. 과량 공기를 케이크에 통과시키지 않는다.
5. 필터-케이크를 세개의 분리된 이소프로판을 15ml 부분으로 그후 세개의 분리된 아세톤 20ml 부분으로 세척한다. 세척간에 과량의 공기를 필터-케이크로 통과시키지 않는다. 죔으로서 세척간에 형성될 수 있는 어떠한 균열 또는 파쇄를 제거한다.
6. 결정체를 24-35℃에서 24시간 고진공 건조시킨다. 예상수율 : 1-1.1g.
7. 필요하다면, 바람직한 이소프로판올에 대하여 전체적으로 아세톤을 대체시킬 수 있으며 첨가형태를 전환시키고, 필요에 따라 즉, 반시간 간격에 걸쳐 DDI의 교반수용액에 용매를 첨가시킨다.
KOH 및/또는 다른 유기 및 무기 강염기를 상응하는 염을 형성하도록 NaOH에 대하여 치환시킬 수 있다.
Na-DDI H2O에 대하여 얻어진 화학적 및 물리적 특성

1. 15-25oC 에서 5분간격에 걸쳐 정상 교반하여 D4T 1g을 1N-NaOH 수용액 4.7ml(1.05몰 당량)에 첨가시킨다. 용액(pH 9.5-11)또는 유사용액을 얻는다.
2. Na-DDI H2O의 제조에 대한 실시예 1에서 기재된 단계 2-7을 반복하고 따른다.

실시예 3
나트륨 F-ddI 헤미히드레이트의 제조

MW : NaOH=40
1. FDDI(426mg)을 1N NaOH 1.8ml(256mg/ml NaFDDI)에서 슬러리화시켰다. 2분에 pH 9.9용액을 얻었다.
2. 이소프로판을 2ml를 첨가하였다. 용액은 깨끗하게 유지되었다.
3. 슬러이화하면서 이소프로판올을 천천히 첨가하였다. 이스프로판을 15ml가 첨가되었을때 중침상 결정화가 시작되었다. 혼합물을 10분간 슬러리화 하였다.
4. 아세톤 15ml를 첨가하고 혼합물을 추가 15분 슬러리화 하였다.
5. 결정체를 진공여과에 의하여 제거하고, 아세톤 15ml 및 에테르 20ml로 세척한 다음 진공건조시켰다 (P2O550℃에서 2시간 그후 24oC 에서 24시간). 수율 :430 mg.

- 제1항에 있어서, X가 Na인 것을 특징으로 하는 2',3'-디데옥시-2',3'-디데히드로티미딘의 염.
- 제1항에 따른 염의 항비루스 유효량과 생리적으로 허용되는 고체 또는 액체 희석제로 이루어지는 약제조성물.
- 제1항에 따른 염의 항비루스 유효량 단독으로 또는 희석제와 혼합하여 또는 약물형태로 투여되는 것을 특징으로 하는 비루스로 감염된 인간을 제외한 온혈동물의 치료방법.
- 제3항에 있어서, 상기의 염이 경구투여를 위한 정제, 캡슐 또는 분제형태인 약물이고 장용성 제피로 피복되며 위산에 대하여 완충작용하는 것을 특징으로 하는 약제조성물.
- 제6항에 있어서, X가 나트륨인 것을 특징으로 하는 2',3'-디데옥시이노신의 염.
- 2',3'-디데옥시이노신의 나트륨염 항비루스 유효량과 생리적으로 허용되는 고체 또는 액체 희석제로 이루어지는 약제조성물.
