Abstract

본 발명은 유전자 재조합 기술에 의해 재조합된 L-페닐알라닌의 생선성이 향상된 새로운 플라스미드에 의해 형질 전환된 숙주 세포를 배양 요지에서 배양하여 L-페닐알라닌을 생산, 축적시키고, 배양액중으로부터 L-페닐알라닌을 회수함을 특징으로 하는 L-페닐알라닌을 고수율로 제조하는 방법에 관한 것이다.

Description

유전자 조작 미생물 발효에 의한 L-페닐알라닌의 제조방법
본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 에쉬리시아 콜리(Escherichia coli)에 있어서 방향족 아미노산의 생합성 경로 및 조절 기작을 나타낸 것이며,
제2도는 본 발명의 재조합 플라스미드 pMW16의 제조 공정도 및 제한효소 부위도를 나타낸 것이다.

Claims (6)

  1. 코리스메이트 뮤타아제 p-프리페네이트 디히드라타제를 코딩하는 pheA 유전자, β-데옥시-D-아라비노헵툴로소네이트-7-포스페이트 신타아제를 코딩하는 AroF, 이들 두 유전자의 발현을 위한 두개의 프로모터 및 온도 감수성 리프레서 유전자를 함유하는 플라스미드에 의해 형질전환된 숙주 세포를 배지에서 배양하여 L-페닐알라닌을 생산 축적하고, 배양액으로부터 L-페닐알라닌을 회수함을 특징으로 하는 L-페닐알라닌의 제조방법.
  2. 제1항에 형질전환된 숙주 세포는 에쉬리시아 콜리(Escherichia coli)MWPWJ304(KFCC 10738)임을 특징으로 하는 제조방법.
  3. 제1항에 있어서, 프로모터는 PL프로모터임을 특징으로 하는 제조방법.
  4. 제1항에 있어서, 온도 감수성 리프레서는 C1867리프레서임을 특징으로 하는 방법.
  5. 코리스메이트 뮤타아제 p-프리페네이트 디히드라타제를 코딩하는 pheA 유전자, α-데옥시-D-아라비노헵룰로소네이트-7-포스페이트 신타아제를 코딩하는 AroF, 이들 두 유전자의 발현을 위한 두개의 프로모터 및 온도 감수성 리프레서 유전자를 함유함을 특징으로 하는 플라스미드 pMW16.
  6. 플라스미드 pMW16에 의해 형질전환된 것임을 특징으로 하는 숙주 세포.
