(1) Thr-Leu-Arg-Gln-His-Tyr-Ser-Ser-Leu-Ile-Pro-Asn-Leu-Phe-Ile-Ala-Ala-Val-Gly-Asn-Ile-Asn-Glu-Leu-Asn-Asn-Ala-Asp-Gln-Ala-Ala-Arg-Glu-Leu-Phe-Leu-Asp-Trp-Asp-Thr, 및
(2) Asn-Lys-Thr-Asp-Phe-Ala-Glu-Ile-Glu-Leu-Tyr-Asp-Val-Leu-Tyr-Thr-Asp-Ala-Asp-Ile-Ser-Gly-Asp-Ala-Pro-Leu-Leu-Ala-Pro-Ala-Tyr-Lys.
을 갖고, 분자량이 약 63,000달톤인 브레비박테리움 락토페르멘툼(Brevibacterium lactofermentum)으로부터 유도되는 신규한 세포 표증 단백질의 암호화 유전자를 포함하는 DNA 단편을 코리네포름 세균에 도입시켜 형질전환체를 제조한다. 새로이 제조된, 신규한 세포 표층 단백질이 부족한 이러한 형질전환체 또는 균주는 L-아미노산 발효법에 의해 L-아민노산을 포함한 유용물질을 제조하는데 사용된다.

Claims (11)

  1. 분자내 2개의 서열
    (1) Thr-Leu-Arg-Gln-His-Tyr-Ser-Ser-Leu-Ile-Pro-Asn-Leu-Phe-Ile-Ala-Ala-Val-Gly-Asn-Ile-Asn-Glu-Leu-Asn-Asn-Ala-Asp-Gln-Ala-Ala-Arg-Glu-Leu-Phe-Leu-Asp-Trp-Asp-Thr, 및
    (2) Asn-Lys-Thr-Asp-Phe-Ala-Glu-Ile-Glu-Leu-Tyr-Asp-Val-Leu-Tyr-Thr-Asp-Ala-Asp-Ile-Ser-Gly-Asp-Ala-Pro-Leu-Leu-Ala-Pro-Ala-Tyr-Lys.

  2. 제1항에 따른 세포 표층 단백질의 암호화 유전자를 포함하는 DNA 단편.
  3. 제2항에 다른 DNA 단편을 코리네포름(Coryneform) 세균 세포에서 자동 복제할 수 있는 벡터와 결합시켜 수득된 재조합 DNA.
  4. 제2항에 따른 DNA 단편을 코리네포름 세균 세포에서 증폭시켜 수득된 형질전환체.
  5. 유용 물질 생성 활성을 갖는 제4항에 다른 형질전환체를 배양 배지에서 배양하고, 배양 배지에 유용물질을 생성 및 축척시킨 후, 배양 배지로부터 유용물질을 수집하는 단계를 포함하여 발효 과정에 의해 유용물질을 제조하는 방법.
  6. 제5항에 있어서, 유용물질이 L-아미노산인 방법.
  7. 제1항의 세포 표층 단백질 또는 이 세포 표층 단백질과 실질적으로 동일한 단백질이 부족하고 세포 응집 특성이 있음을 특징으로 하는 코리네포름 세균.
  8. 제7항에 있어서, 제1항에 따른 세포 표층 단백질의 암호화 유전자 또는 이 세포 표층 단백질과 실질적으로 동일하고 코리네포름 세균의 세포 표층에 존재하는 단백질의 암호화 유전자가, 세포 표층 단백질의 암호화 유전자 일부분 이상을 포함하는 DNA 단편과 이 DNA 단편과 동일하거나 상동성인 염색체상의 DNA 서열과의 상동 재조합(homologous recombination)에 의해 파괴됨을 특징으로 하는 코리네포름 세균.
  9. 제7항 또는 제8항에 있어서, 코리네포름 세균이 브레비박테리움 락토페르멘툼인 코리네포름 세균.
  10. 유용물질을 생성하는 활성을 갖는, 제7항 내지 제9항중 어느 한 항에 따른 코리네포름 세균을 배양하고, 유용물질을 배양 배지에 생성 및 축적시킨후, 유용물질을 배양 배지로부터 수집하며, 추가로 배양 완료후 배양 배지를 정치시켜 코리네포름 세균의 세포를 침전시키는 단계를 포함하여 유용물질을 제조하는 방법.
  11. 제10항에 있어서, 유용물질이 L-아미노산인 방법.
